Population dynamics for voles (Cricetidae), Tengmalm's owl (Aegolius funereus (L.)), red fox (Vulpes vulpes (L.)) willow grouse (Lagopus lagopus (L.)), black grouse (Lyrurus tetrix (L.)), capercaillie (Tetrao urogallus L.), hazel hen (Tetrastes bonasia (L.)), mountain hare (Lepus timidus L.) and tularemia (Francisella tularensis (McCoy & Chapin)) and game bird recruitment were studied by index methods in northern Sweden. In addition contemporary temperature records and spruce (Picea abies (L.) Karst.) and pine (Pinus silvestris L.) cone crops (as indices for plant production) and the occurrence of forest damage, caused by voles eating bark, were studied.During 1970–80 two synchronous 4-year cycles were observed for voles, predators (Tengmalm's owl and red fox) and their alternative prey species (grouse and mountain hare). In grouse the change of numbers was correlated with that of recruitment. Autumn vole numbers peaked about a year before the other species and extensive forest damage occurred at winter peak densities of voles. These population fluctuations are consistent with a predator-prey model for their regulation. In short the model suggests that vole-food plant interactions trigger the cycle of voles, that voles generate the cycle of predators and that these in turn synchronize alternative prey populations to the others at vole declines.For voles, grouse and red fox the amplitude was higher in the first cycle compared to the second one whilst the opposite was true for the mountain hare. Although temperature and cone crops showed large interannual variations they still implied that herbivore food conditions were better during the former cycle. Hence, the reduction of the amplitude of the vole cycle may be explained by inter-cyclic differences in plant food conditions, implying food shortage (as indicated by bark-eating) at different population levels. The similar decrease of grouse and red fox populations may also be explained by deteriorated food conditions and/or for the fox by an outbreak of sarcoptic mange (Sarcoptes scabiae var. vulpes). The increased amplitude of the mountain hare cycle was part of a long-term rise in numbers after a tularemia epidemic in 1967. This is interpreted as a recovery, probably towards the generally higher pre-epidemic population level.  相似文献

In order to test the hypothesis that enhanced fibrinolytic activity is a factor which prevents the blood of diving seals from clotting, we instrumented two female grey seals (Halichoerus grypus) with subcutaneous electrodes for measurements of heart rate (HR) and an extradural intravertebral venous catheter for collection of blood samples before, during and after simulated dives of 10 min duration. Blood samples were used for in vitro determination of clot lysis time (CLT), which is a measure of the level of fibrinolytic activity, and for analyses of plasma levels of cortisol, noradrenaline and adrenaline (A). The seals displayed profound diving bradycardia indicative of a substantial reduction in blood flow rates (pre-dive HR: 78 (63–98) bpm; dive HR: 8 (7–10) bpm; (median (range); n=2)) and elevated catecholamine levels (pre-dive A: 121 (98–184) pg·ml−1; peak dive/post-dive A: 3510 (447–6181) pg·ml−1), both of which are factors which promote blood coagulation. Nevertheless, we found that CLT always increased in connection with diving (pre-dive CLT: 436 (356–568) min; peak CLT during diving: 1380 (640–1800) min), which implies a reduced, rather than enhanced, fibrinolytic activity in this situation. These results show that enhanced fibrinolytic activity is not part of the defence system which prevents fatal clotting from occurring in diving grey seals.  相似文献

In the shoot apices of the haploid Pelargonium cultivar Kleine Liebling, all mitoses are haploid (n = 9); however, ca. 20% of the interphase nuclei have DNA contents greater than 2C (up to 4C), indicating a tendency to chromosome endoreduplication in this material. — In internodes in vivo, the few mitoses present are haploid (quite probably, cambium cells); in addition to haploid interphases (1C to 2C DNA contents), endoreduplicated (endopolyploid) nuclei and nuclei in the course of endoreduplication occur with a frequency of ca. 40–50% (DNA contents up to 8C). — When internodes are cultured in vitro, differentiated cells are stimulated to divide, thus forming a population of diploid and tetraploid mitoses in addition to the preexistent meristem (haploid) cell population. In the process of time, diploid and tetraploid mitoses continue to be present in the callus, whilst haploid mitoses may decrease in number and eventually disappear. All mitoses analyzed had euploid chromosome numbers (9, 18 and 36) and their DNA contents were correspondingly 2C, 4C and 8C. Since no extensive chromosome counts were made, aneuploidy in the cultured material cannot be excluded; but, if occurring, it should be rather rare. — Under the experimental conditions used, prolonged culture in vitro leads to the production of nuclei with DNA contents (16C and 32C) greater than those occurring in vivo (8C), due to one and two additional DNA replications respectively beyond the limits attained in vivo. Even in these cultures, however, a population of the meristematic haploid cell line (DNA values 1C to 2C) is still present. — The present results are discussed in their relations with previous works on nuclear conditions in vivo and in vitro and on regeneration processes in cultured tissues in plants.  相似文献

We investigated whether acute iron intoxication causes oxidative DNA damage, measured in terms of 7-hydro-8-oxo-2′-deoxyguanosine, 8-oxodG, in nuclear DNA in testes and epididymal sperm cells in vivo and in vitro in rats. In addition, we investigated levels of the modified nucleoside in liver and kidney and measured its urinary excretion.

Sperm cells were isolated from the epididymides and the testes cells were isolated after homogenisation. In vitro, the sperm and testes cells were incubated with increasing concentrations of FeCl2 ranging from 0 to 600 μM. The median (range) levels of 8-oxodG/105 dG in the epididymal sperm cells increased from 0.48 (0.42–0.90) to 15.1 (11.4–17.6) (p < 0.05), whereas the level rose from 0.63 (0.22–0.81) to 8.8 (4.5–11.6) (p < 0.05) at 0 and 600 μM, respectively, in the testicular cells.

In vivo groups of 7–8 rats received 0, 200 or 400 mg iron/kg as dextran i.p. After 24h, epididymal sperm cells, testes, kidneys and liver were collected for analysis. Kidney and sperm DNA showed a significant increase in 8-oxodG in the iron-treated animals. The median (range) values of the 8-oxodG/105 dG in the epididymal sperm cells rose from 0.66 (0.38–1.09) to 1.12 (0.84–5.88) (p < 0.05) at 0 and 400 mg iron/kg, respectively, whereas the values in the testes and liver showed no significant change. In the kidneys the 8-oxodG/105 dG median (range) values were 0.98 (0.73–1.24), 1.21 (1.13–1.69) and 1.34 (1.12–1.66) after 0, 200 and 400 mg iron/kg, respectively (p < 0.05).

The 8-oxodG-excretion rate was measured in 24 h urine before and after iron treatment. The rate of urinary 8-oxodG excretion increased from 129 (104–179) pmol/24 h before treatment to 147 (110–239) pmol/24h after treatment in the group receiving 400 mg iron/kg (p < 0.05).

The results indicate that acute iron intoxication may increase oxidative damage to sperm and kidney DNA.  相似文献

In order to investigate the prevalence of tick-borne infectious agents among ticks, ticks comprising five species from two genera (Hemaphysalis spp. and Ixodes spp.) were screened using molecular techniques. Ticks (3,135) were collected from small wild-caught mammals or by dragging/flagging in the Republic of Korea (ROK) and were pooled into a total of 1,638 samples (1 to 27 ticks per pool). From the 1,638 tick samples, species-specific fragments of Anaplasma phagocytophilum (1 sample), Anaplasma platys (52 samples), Ehrlichia chaffeensis (29 samples), Ehrlichia ewingii (2 samples), Ehrlichia canis (18 samples), and Rickettsia rickettsii (28 samples) were amplified by PCR assay. Twenty-one pooled and individual tick samples had mixed infections of two (15 samples) or three (6 samples) pathogens. In addition, 424 spleen samples from small captured mammals (389 rodents, 33 insectivores, and 2 weasels) were screened for selected zoonotic pathogens. Species-specific DNA fragments of A. phagocytophilum (110 samples), A. platys (68 samples), E. chaffeensis (8 samples), E. ewingii (26 samples), E. canis (51 samples), and Rickettsia sp. (22 samples) were amplified by PCR assay. One hundred thirty small mammals had single infections, while 4, 14, and 21 striped field mice (Apodemus agrarius) had mixed infections of four, three, and two pathogens, respectively. Phylogenetic analysis based on nucleotide sequence comparison also revealed that Korean strains of E. chaffeensis clustered closely with those from China and the United States, while the Rickettsia (rOmpA) sequences clustered within a clade together with a Chinese strain. These results suggest that these agents should be considered in differential diagnosis while examining cases of acute febrile illnesses in humans as well as animals in the ROK.  相似文献

The study was designed to examine the gender-related differences in maximum mechanical power output in various short-burst activities during growth. The subject sample consisted of four subgroups: 9 boys (14.11 +/- 0.6 yr), 9 boys (10.67 +/- 0.71 yr), 7 girls (14.29 +/- 0.49 yr), 7 girls (10.57 +/- 0.54 yr). We measured peak power (PP), mean power (MP), fatigue index (FI) during 30-s WAnT, squat jump height (SJH) and power (SJP), and counter movement jump height (CMJH) and power (CMJP), maximum speed over 20-metre distance (S20). Lactation concentration was measured in the 3rd and 5th minutes after the WAnT Ratio normalisation and ANCOVA were used to remove the influence of the differences in muscle (MM) and body mass (BM). Male adolescents had higher absolute values of PP (P < 0.05), MP (P < 0.05) than female. Ratio normalisation showed that boys had higher PP/BM (P < 0.05), PP/MM (P < 0.05), MP/BM (P < 0.05), MP/MM (P < 0.06) than girls. The ANCOVA adjustment for MM showed differences between genders in PP (P < 0.001), MP (P < 0.001), SJH (P < 0.05), SJP (P < 0.05) and CMJP (P < 0.001), whereas the ANCOVA adjustment for BM showed differences only in PP (P < 0.001), MP (P < 0.001). Prepubertal boys had higher absolute values only in SJP (P < 0.05). We concluded that variations in body composition could not be the only key to gender-related differences in power output in short-burst activities.  相似文献

Isolated chromoplasts from Narcissus pseudonarcissus flowers contain: a fatty acid synthesizing system; acyl-CoA synthetase (EC 6.2.1.3); glycero-phosphate acyltransferase (EC 2.3.1.15); acylglycero-phosphate acyltransferase; phosphatidate phosphatase (EC 3.1.3.4); diacylglycerol galactosyltransferase (EC 2.4.1.46); and diacylgalactosylglycerol galactosyltransferase, i.e. all enzymatic activities necessary for the synthesis of diacylgalactosylglycerol and diacylgalabiosylglycerol from acetate, HCO - 3 , sn-glycerol 3-phosphate, and UDP-d-galactose. Diacylgalactosylglycerol and diacylgalabiosylglycerol, however, are synthesized from these precursors to only a very low extent in an in vitro system. This is attributed to a specificity of diacylglycerol galactosyltransferase for highly unsaturated diacylglycerols. Specificities of acyltransferase reactions were also found.  相似文献

The cell-mediated immune responses of 12 healthy, coccidioidin skin-test positive subjects (Group I) were compared with those of 15 healthy, coccidioidin skin-test positive persons who had primary asymptomatic coccidiodomycosis, (Group II), 12 patients with active, pulmonary coccidioidomycosis (Group III), four patients with disseminated disease (Group IV), and five patients who had been in clinical remission for 1 year or longer (Group V). Lymphocytes from healthy subjects in Groups I and II responded in vitro to Coccidioides immitis antigen by undergoing an increased DNA synthesis (lymphocyte transformation) and/or by producing macrophage migration inhibitory factor (MIF). In contrast, patients in Groups III and IV failed to respond to Coccidioides antigens in vivo (skin tests) or in vitro (lymphocyte transformation and production of MIF). The responses of subjects in Group V with inactive disease fell in between those of healthy donors in Groups I and II and patients in Groups III and IV. The cellular immune defect, in terms of antigen recognition, appeared to be specific for C. immitis in all but one patient.  相似文献

The age-dependent variability of ecosystem carbon (C) fluxes was assessed by measuring the net ecosystem exchange of C (NEE) in five managed forest stands in northern Wisconsin, USA. The study sites ranged in age from 3-year-old clearcut to mature stands (65 years). All stands, except the clearcut, accumulated C over the study period from May to October 2002. Seasonal NEE estimates were −655 ± 17.5 g C m–2 in the mature hardwood (MHW), −648 ± 16.8 in the mature red pine (MRP), −195 ± 15.6 in the pine barrens (PB), +128 ± 17.1 in the young hardwood clearcut (YHW), and −313 ± 14.6 in the young red pine (YRP). The age-dependent differences were similar in the hardwood and conifer forests. Even though PB was not part of either the hardwood or conifer chronosequence, and had a different disturbance agent, it still fits the same general age relationship. Higher ecosystem respiration (ER) in the young than in the mature stands was the combined result of earlier soil warming in spring, and higher temperature and greater biological activity in summer, as indicated by temperature-normalized respiration rates. The fire-generated PB had lower ER than the harvest-generated YHW and YRP, where high ER was sustained partly on account of logging residue. During the main growing season, the equivalent of 31 (MHW), 48 (MRP), 68 (PB), 114 (YHW) and 71% (YRP) of daily gross ecosystem production (GEP) was released in ER during the same day. The lower ER:GEP ratio in the mature stands was driven by greater age-dependent changes in ER than GEP. The magnitude of the increase in ER:GEP ratio in spring and fall was interpreted as the extent of the decoupling of ER and GEP. Decoupling (sustained high ER despite decreasing GEP) was observed in YHW, PB and MHW, whereas in coniferous stands (MRP and YRP) the stable ER:GEP ratio suggested preferential use of new photosynthates in ER. The results indicate that a great part of the variation in landscape-level C fluxes can be accounted for by mean stand age and associated parameters, which highlights the need to consider this source of heterogeneity in regional C balance estimates.  相似文献

In vitro activity of milk leukocytes (viz. neutrophils, lymphocytes and macrophages) was evaluated in forty-eight (48) clinically healthy high-yielding cross-bred cows of mid-lactation stage (100–200 days of lactation), divided into four groups namely 1st parity (n = 12), 2nd parity (n = 12), 3rd parity (n = 12) and 4th and above parity (n = 12). Milk samples were taken (250 ml/cow) were taken. Milk somatic cell counts (SCC) and differential leukocyte counts (DLC) were performed microscopically. In vitro phagocytic index (PI) of milk neutrophils and macrophages was evaluated by colorimetric nitro blue tetrazolium reductive assay. Mitogen-induced milk lymphocyte blastogenic response was measured by colorimetric MTT (tetrazolium) assay after isolation of the milk leukocytes by density gradient centrifugation. Milk SCC differed significantly (p < 0.01) between different parity. Cows of 4 and above parity showed significantly (p < 0.01) higher milk SCC compared to primiparous cows. There was no significant difference in milk DLC during different parities in high-yielding cross-bred cows. There was a significant (p < 0.01) variation in lymphocyte blastogenesis amongst parity. The highest value of lymphocyte blastogenesis was seen at 3rd parity, whereas lowest value was obtained in the cows of both 1st and 4th or above parity. PI of milk neutrophils did not differ significantly between parity. PI of milk macrophages was significantly (p < 0.01) higher in 3rd parity and lower (p < 0.01) in 1st and 4th parities. The study indicated that depressed activity of milk lymphocytes and macropages was lower and SCC was higher in the cows of 4th and above parity indicating more mammary stress and hence susceptible to udder infection and mastitis. Therefore, better care and managemental interventions should be taken around these periods.  相似文献

The rapid shift to high-grain (HG) diets in ruminants can affect the function of the rumen epithelium, but the dynamic changes in the composition of the epithelium-associated (epimural) bacterial community in sheep still needs further investigation. Twenty male lambs were randomly allocated to four groups (n = 5). Animals of the first group received hay diet and represented a control group (CON). Simultaneously, animals in the other three groups (HG groups) were rapidly shifted to an HG diet (60% concentrate)which continued for 7 (HG7), 14 (HG14) and 28 (HG28) days, correspondingly. Results showed that ruminal pH dramatically decreased due to the rapid shift to the HG diet (P <0.001), while, the concentrations of butyrate (P <0.001), lactate (P = 0.001), valerate (P = 0.008) and total volatile fatty acids (P = 0.001) increased. Diversity estimators showed a dramatic decrease after the shift without recovering as the HG feeding continued. The principal coordinates analysis showed that CON group clustered separately from all HG groups with the presence of significant difference only between HG7 and HG28 (P = 0.034). The non-parametric multivariate analysis (npmv R-package) deduced that the primary significant differences in phyla and phylogenetic investigation of communities by reconstruction of unobserved states (PICRUSt)-predicted Kyoto Encyclopedia of Genes and Genomes (KEGGs) was attributed mainly to the diet composition (P <0.001, P = 0.001) compared to its application period (P = 0.140, 0.545) which showed a significant effect only on the genus (P = 0.001) and the operational taxonomic units (OTUs) level (P = 0.011). The Kruskal–Wallis test deduced that six phyla showed a significant effect due to the shift in diet composition. At the genus level, HG feeding altered the abundance of 12 taxa, four of which showed a significant variation due to the duration of the HG diet application. Similarly, we found that 21 OTUs showed significant variations due to the duration of the HG diet application. Furthermore, the genes abundance predicted by PICRUSt revealed that the HG feeding significantly affected seven metabolic pathways identified in the KEGG. Particularly, the abundance of gene families associated with carbohydrates metabolism were significantly higher in HG feeding groups (P = 0.027). Collectively, these results revealed that the rapid transition to an HG diet causes dramatic alterations in ruminal fermentation and the composition and function of ruminal epithelium-associated microbiome in sheep, while, the duration of the HG diet application causes drastic alterations to the abundance of some species.  相似文献

A total of 404 samples of commercial swine feed from Portugal feed mills were analysed by HPLC methods for the presence of mycotoxins: 277 samples of feed for fattening pigs were analysed for ochratoxin A (OTA), zearalenone (ZEA), and deoxynivalenol (DON), and 127 samples of feed for sows were analysed for ZEA and fumonisins (FB1 + FB2). Concerning feed for fattening pigs, 21 (7.6%) samples were positive for OTA, (2–6.8 μg/kg), 69 (24.9%) were positive for ZEA (5–73 μg/kg), and 47 (16.9%) were positive for DON (100–864 μg/kg). In feed for sows, the results showed 29.9% of positive samples for ZEA (5–57.7 μg/kg) and 8.7% positive samples for FB1 and FB2 (50–391.4 μg/kg). Co-occurrence of DON/ZEA was found most frequently, but simultaneous contamination with OTA/ZEA and OTA/DON was also found.  相似文献

Purpose of Review

This study aimed to isolate and characterize filamentous fungi onychomycosis agents in a military population assisted at a hospital outpatient clinic.

Recent Findings

In onychomycosis, the fungi colonize the subungual region causing thickening, discoloration, or cracking of the nail bed. Samples were collected from patients with clinical sights of onychomycosis.

Summary

Among 80 samples collected, 50 (62.5%) had positive culture. Isolated dermatophytes (86%) were Trichophyton rubrum (21; 42%), T. mentagrophytes var. interdigitale (19; 38%), and Microsporum gypseum (3; 6%) and non-dermatophyte molds were Fusarium spp. (1; 2%), Scytalidium spp. (1; 2%), and Chaetomium globosum (5; 10%). Minimal inhibitory concentrations (mg/L) of terbinafine, itraconazole, and fluconazole necessary to inhibit 50/90% of the isolates were respectively 0.015/0.06, 0.06/0.12, and 32/32. Etiological agents of onychomycosis in a military hospital are similar as reported in studies for the general population. High prevalence of non-dermatophytic agents was observed, especially for Chaetomium globosum.

The qualitative and relative quantitative anthocyanin content of 19 species belonging to the genera Sambucus, Lonicera and Viburnum in the family Caprifoliaceae has been determined. Altogether 12 anthocyanins were identified; the 3-O-glucoside (2), 3-O-galactoside (5), 3-O-(6″-O-arabinosylglucoside) (7), 3-O-(6″-O-rhamnosylglucoside) (9), 3-O-(2″-O-xylosyl-6″-O-rhamnosylglucoside) (10), 3-O-(2″-O-xylosylgalactoside) (11), 3-O-(2″-O-xylosylglucoside) (12), 3-O-(2″-O-xylosylglucoside)-5-O-glucoside (14), 3-O-(2″-O-xylosyl-6″-O-Z-p-coumaroylglucoside)-5-O-glucoside (15) and 3-O-(2″-O-xylosyl-6″-O-E-p-coumaroylglucoside)-5-O-glucoside (16) of cyanidin, in addition to the 3-O-glucosides of pelargonidin and delphinidin (1 and 3). Pigment 7 is the first complete identification of the disaccharide vicianose, 6″-O-α-arabinopyranosyl-β-glucopyranose, linked to an anthocyanidin.  相似文献

A survey of aflatoxins in sesame in Iran   总被引:1,自引:0,他引:1  
A study of the occurrence of aflatoxins (AF) in sesame seeds was conducted in the Khorasan province of Iran between September 2009 and August 2010. Samples (n = 182) were analyzed by liquid chromatography (LC), and detection limits for AFB1, AFB2, AFG1 and AFG2, were 0.45, 0.19, 0.61, and 0.22 ng/g, respectively. AFB1 was detected in 33 samples (18.1%), at a mean level of 1.62 ± 1.32 ng/g, and a maximum level of 5.54 ng/g. AFB1 levels exceeded the European Union (EU) maximum tolerated level (MTL, 2 ng/g) in 9 samples, and the Iran MTL (5 ng/g) in 1 sample. Regarding total aflatoxins (AFT), the mean level was 0.92 ± 1.36 ng/g, and the maximum level was 5.54 ng/g. No sesame sample exceeded the Iran MTL (15 ng/g), but two samples exceeded the EU MTL (4 ng/g) for AFT. It is concluded that low levels of AFs occur frequently in sesame from Iran.  相似文献

Needle traces have been used for revealing historical changes in needle longevity of Pinus sylvestris L. and Picea abies (L.) H. Karst. In this paper we compared the needle-trace diameter of five conifer species [Abies sibirica Ledeb., Picea abies, Pinus sylvestris, Pseudotsuga menziesii (Mirb.) Franco and Taxus baccata L.] in respect of needle size. In case of P. sylvestris, we studied how needle-trace diameter varied among different growing sites and among different shoots within a site, and related that variation to needle morphology and structure. Pinus sylvestris had the greatest diameter of needle traces (mean±SD =233±30 m) followed by T. baccata (141±19 m) and P. menziesii (121±30 m). The smallest needle-trace diameter was in A. sibirica (85±21 m) and P. abies (80±18 m). Among species, the needle-trace diameter was strongly correlated with needle length (r =0.93, P <0.05). Within P. sylvestris, the needle-trace diameter was significantly affected by location (P <0.001) and varied significantly among shoots within a location (P <0.001). The shoots of P. sylvestris which had greater average diameter of needle traces, tended to have longer (r =0.36, P <0.05), wider (r =0.55, P <0.05) and heavier (r =0.64, P <0.05) needles with greater stomatal density (r =0.55, P <0.05). Although our results do not allow the use of needle-trace diameter to predict properties of needle structure at the present stage, they nevertheless indicate general trends and point to a need for further studies.  相似文献

The northern range of Atlantic cod (Gadus morhua), overlaps the southern range of the Greenland cod (Gadus ogac), in the coastal waters of Western Greenland. The availability of a temperate water species (G. morhua) in the same area and oceanographic conditions as a polar species (G. ogac) presented us with the ideal circumstances to test the hypothesis of metabolic cold adaptation (MCA) since many of the problems associated with MCA studies (adaptation of the animals beyond their normal temperature range or mathematical extrapolation of data to common temperatures) could thus be avoided. We therefore used a swim tunnel to measure oxygen consumption in fish at 4°C over a range of swimming speeds and following exhaustion, monitored the size of the oxygen debt and time of oxygen debt repayment. There were no significant differences in standard (60–72 mg O2 kg–1· hr–1), routine (76 mg O2 kg–1·hr–1), active (137mg O2 kg–1·hr–1), or maximal (157 mg O2 kg–1·hr–1) metabolic rate, metabolic scope (2.5) or critical swimming speed (2.2 BL·s–1) between the two species. Following exhaustive swimming, however, the half-time for oxygen debt repayment in G. ogac (43 min) was almost twice that of G. morhua (25 min). Despite its circumpolar distribution, therefore, there was no evidence of MCA in G. ogac.  相似文献
