The behavior of individual allocyclic chromosomes has been analyzed in lymphocytes of a sister and a brother with Bloom's syndrome. Of 4,633 diploid cells, 115 showed allocyclic chromosomes, and 74 of these had 44, 45 or 46 normal metaphase chromosomes accompanied by one or two allocyclic chromosomes. Of 56 tetraploid cells, 9 contained such chromosomes. The allocyclic chromosomes appeared pulverized or extended corresponding to S or G2 PCC. We have proposed the hypothesis that individual allocyclic chromosomes do not, as a rule, come from micronuclei, as has often been assumed, but have been left behind in their cycle. This would be caused by a mutation or deletion of a hypothetical coiling center situated near the centromere of each chromosome arm. The following observations agree with our explanation but less well or not at all with the idea of micronuclei: (1) In only 9.6% of the cells does the allocyclic chromosome lie at the edge of the metaphase plate. (2) In 24 cells a part of a chromosome is pulverized while the rest is in metaphase. (3) Both a pulverized and an extended chromosome were present in the same cell. (4) A pulverized acrocentric is often nose-to-nose with a normal D or G chromosome. (5) No allocyclic chromosomes corresponding to G1 PCC have been found in our material. (6) When a ring is replaced by an allocyclic chromosome, it is usually a member of a 46-chromosome complement. Furthermore, the occurrence of allocyclic chromosomes is correlated with that of other chromosome anomalies which do not follow a Poisson distribution. A novel approach was used to compare male and female recombination rates in wheat. Doubled haploid lines were developed from an F1 using two distinct approaches: the anther-culture technique and the Hordeum bulbosum system, from which sets of lines were developed from male and female meioses, respectively. The genotype of the lines was established at RFLP and isozyme markers polymorphic on chromosomes of homoeologous groups 6 and 7, and male and female linkage maps were calculated using this information. The markers in one segment of chromosome 6B exhibited disturbed segregation frequencies in the anther-culture population. The male and female maps differed significantly in recombination frequency between some markers on two chromosomes, and these were consistent in direction within chromosomes and inconsistent in direction between chromosomes. In two of the four chromosomes studied the male map was much longer than the female map. These results suggest that significant differences may exist in male and female recombination frequencies in bread wheat which are specific to certain chromosomal segments but are inconsistent in direction between chromosomes. Other factors, such as environmental influences, may also be important in creating differences.  相似文献

Summary The GCTT polymorphism recently described at positions — 1106 and — 1105 in the 5 flanking region of the G globin gene for the Bantu S haplotype was analysed by denaturing gradient gel electrophoresis. We studied 108 S chromosome and 122 A chromosomes. The TT sequence was found as follows: in all of 80 chromosomes bearing the Bantu S haplotype with the 6-bp deletion -400 nt from the G gene in 3 out of 5 Bantu S chromosomes without the deletion, in 1 out of 122 A chromosomes from different ethnic origins but in none of 23 S chromosomes bearing the Senegal, Benin or Cameroon haplotypes. These results confirm the heterogeneity of the Bantu S haplotype and allow a tentative evolutionary sequence for the different alleles at this locus to be presented.  相似文献

Change in chromosome size in root tip meristems of rye and Allium cepa are induced by growing the plants in solutions differing in phosphorus content. The chromosomes are 50% larger by volume in a high phosphate as compared with a no phosphate solution. Alteration of other elements supplied in culture also induces change in the size of chromosomes. — The size variation is a reflection of change in the chromosome dry mass. In part at least this change in mass is attributable to alteration in the amount of protein. The DNA component of the chromosomes remains unchanged. — A consistent pattern of change in chromosome size, quite independent of that induced by varying the culture solution, is related to age and development. For example, the root tip chromosomes double in size during the first three weeks of growth in rye seedlings. Thereafter the size decreases. As with the induced chromosome changes the protein content alters, the DNA amount remains constant. — Variation in the non-permanent component of the chromosomes in meristems appears to be closely correlated with the rate of cell metabolism.  相似文献

Marker chromosome are small supernumerary chromosomes that are sometimes associated with developmental abnormalities. Hence, the genes involved in such cases provide an interesting approach to understanding developmental abnormalities in man. As a first step towards isolating such sequences, marker chromosomes need complete characterization. By combining chromosome isolation by flow sorting and the degenerate oligonucleotide primed — polymerase chain reaction, we have constructed a DNA library specific for a marker chromosome found in a child with severe developmental abnormalities. We used fluorescent in situ hybridization of the library onto normal metaphase spreads (reverse chromosome painting) and were thus able to determine that the marker consists of the centromeric part of chromosome 7, the telomeric region of the long arm of chromosome 5 and the telomeric region of the short arm of the X-chromosome. Subsequently, we hybridized normal chromosome-specific libraries of the relevant chromosomes onto metaphases containing the marker chromosome (forward chromosome painting) and could in this manner establish the precise location of the different chromosome regions on the marker chromosome itself. This is a general approach suitable for outlining marker chromosomes in detail, and will aid the identification of the genes involved.  相似文献

Summary Detailed restriction enzyme analysis of the DNA from a Chinese female showed that one of her chromosomes had a >17.5 kb deletion of DNA, including the , 2, and 1 globin genes, which is present in many Southeast Asians with an -thalassemia-1 chromosome. Her normal chromosome had the expected cluster of -like globin genes (5----2-1-3), but the segment of DNA between the two globin genes was elongated by some 0.5–0.7 kb. Analyses of various restriction sites suggested that this normal variant of the human globin gene complex is due to a crossover between a normal chromosome with () and a chromosome with an -thalassemia-2 (–3.7) and an -21-hybrid gene.  相似文献

Relative DNA values and percent lengths of chromosome arms have been studied for six species of the super-family Dasyuroidea, five species of Phalangeroidea and four species of Perameloidea. By multiplying by relative DNA values all percent lengths have been expressed in the same units. Two species are said to share a chromosome if neither of the arms differs at the 5% level of probability. It is argued that if species share at least two two-armed chromosomes, this can be taken as evidence of relationship. A standard dasyurid karyotype has been defined and individual species of Dasyuridae show only small deviations from it. The nature and significance of the bi-modal distribution of chromosome numbers in marsupials is discussed.  相似文献

Summary A series of man-Chinese hamster and man-mouse somatic cell hybrids was investigated to study the localization of the genes coding for the human lysosomal enzyme -galactosidase (EC 3.2.1.23) and for its protective protein. Using a monoclonal antibody, raised against human placental -galactosidase, it was observed that the structural locus for the -galactosidase polypeptide is located on chromosome 3. The nature of the involvement of chromosome 22 in the expression of human -galactosidase was elucidated by metabolic labelling of the hybrids with radioactive amino acids, immunoprecipitation with monoclonal and polyclonal antibodies against -galactosidase, followed by analysis via gel electrophoresis and fluorography.The data show that the presence of chromosome 22 coincides with the presence of a 32 kd protein. This polypeptide, the protective protein was previously shown to be intimately associated with human -galactosidase. In addition, the protective protein was found to be essential for the in vivo stability of -galactosidase by aggregating -galactosidase monomers into high molecular weight multimes. Both chromosome 3 and 22 are therefore necessary to obtain normal levels og -galactosidase activity in human cells.  相似文献

In male hybrids of the cross Chironomus thummi thummi (stock Hl) x Ch. th. piger (stock E) , but not the reciprocal cross, rudimentary testes develop at a growth temperature of 21 ° C. Within the dysgenic testes of these hybrids a number of abnormalities are observed which are restricted to the germ line. Approximately 60% of the hybrid males show allocyclic chromosome behaviour in spermatogonia and spermatocyte I nuclei. Within these nuclei two groups of four chromosomes are formed which differ from one another in their state of condensation. Each chromosome group consists of three long and one short chromosome. In cases where allocycly is very pronounced, the chromosomes of both groups disintegrate into numerous unequally sized fragments at meiotic prometaphase I, and gametes are not produced. In individuals, in which the allocycly is less pronounced or absent the nuclear divisions appear to be normal but chromosome and chromatid aberrations are frequent, and the number of viable sperm is reduced. In these males, the chiasma frequency is also decreased more than 12-fold in comparison with the reciprocal, unaffected piger x thummi hybrids.  相似文献

Summary The first pollen grain mitosis in Scilla sibirica takes place within three weeks after the completion of meiosis. Within one anther the duration of the first pollen grain mitotic cycle varies substantially. The duration of the mitotic cycle affects the length of chromosomes at metaphase of the first pollen grain mitosis. In grains which divide early the chromosomes at metaphase are longer, up to twice the length, of the chromosomes in grains dividing late. The diminution in length with increase in the mitotic cycle is due to more intensive coiling which, in turn, is explained by a lengthening of G2 and of prophase. The relationship between the duration of the mitotic cycle and chromosome length at metaphase would account, at least largely, for the variation in chromosome length between different tissues within organisms. It explains also why the chromosome at metaphase of mitosis are shorter in polyploids than in their diploid ancestors.  相似文献

A standard pachytene karyotype of chickpea (Cicer arietinum L.) is presented for the first time. Individual pachytene chromosomes were identified and described in detail. An idiogram was prepared on the basis of chromosome length, arm ratio, and distribution of heterochromatin and euchromatin. Chickpea pachytene chromosomes belong to the differentiated type with darker staining heterochromatin proximal to and lighter staining euchromatin distal to the centromeres. Chromosomes were numbered from 1 to 8 following a descending order of length. The total length of the chromosome complement at pachytene was 335.33 , and chromosome size ranged from 58.05 to 30.53 .  相似文献

The karyotype of three species of South American grasshoppers are studied in this paper. Leiotettix sanguineus has two chromosome races, one of them with 2n=23 and an XO sex mechanism and the other, as far as we know limited to the Cerro Chato population, with 2n=22 and an XY sex mechanism. Leiotettix politus has two kinds of individuals, one with 2n=14 and XY sex chromosomes and the other 2n=13 and an X1X2Y mechanism. Dichroplus dubius presents 2n=21 and an X1X2Y sex chromosomes. One of the three specimens studied shows aberrant behaviour in the meiotic process.  相似文献

Summary A complete set of isogenic wheat/rye D-genome substitutions were produced by crossing an inbred line of spring rye Secale cereale L. cv. Prolific to a tetraploid wheat, the A-and B-genomes of which had previously been extracted from hexaploid wheat, Triticum aestivum L. em Thell. cv. Thatcher. After chromosome doubling, the derived hexaploid triticale (x Triticosecale Wittmack) was backcrossed to 6x Thatcher and selection for wheat/rye substitution lines was carried out in BCF3 to BCF6 families by using Giemsa C-banding. Five fertile disomic wheat/rye D-genome substitution lines were obtained and their chromosomal constitution was determined to be 1D/1R, 2D/2R, 7D/4R, 6D/6R, 7D/7R. The two remaining 3R and 5R substitutions are at the moment in a monosomic condition. Another 1D/7R substitution was detected but this plant was very weak and sterile, indicating that only substitutions between homoeologous chromosomes result in fertile, vigorous plants. Furthermore, many rye telocentrics as well as rye-rye and rye-wheat translocations were selected. Since all lines selected in this program share the same genetic background of Thatcher wheat, genetic heterogeneity is excluded. The material is very useful, therefore, for analyzing the effects of different rye chromosomes or chromosome segments in an otherwise homozygous background.Contribution No. 797  相似文献

Observations on morphology, sterility and cytology of some interchangetrisomics obtained from the progeny of radiation induced interchangeheterozygotes in a 6-rowed barley (Hordeum vulgare L.) var. K12 are described. The different interchange trisomics show distinct phenotypic expression and have been classified as bushy, slender, robust, semi-erect and pseudonormal on the basis of their gross morphology, leaf characteristics, awn and spike lengths. All interchange trisomics show pollen and ovule sterility which varies between plants. Meiotic behaviour is described. The five different interchange trisomics appear to have different extra chromosomes.  相似文献

Cytological silver-staining procedures reveal the presence of a core running along the chromatid axes of isolated HeLa mitotic chromosomes. In this communication we examine the relationship between this core and the nonhistone chromosome scaffolding, isolated and characterized in previous publications from this laboratory. When chromosomes on coverslips were subjected to the steps used for scaffold isolation in vitro and subsequently stained with silver, the characteristic core staining was unaffected. Control experiments suggested that the core does not contain large amounts of DNA. When scaffolds were isolated in vitro, centrifuged onto electron microscope grids, and stained with silver, they were found to stain selectively under conditions where specific core staining was observed in intact chromosomes. These results suggest that the nonhistone scaffolding is the principal target of the silver stain in chromosomes.  相似文献

The chromosomes of ten species of Cyrtonus and the genome sizes of six are surveyed. Among the total of 15 chromosomally studied species, 11 have 2n=28 chromosomes and a 13+Xyp male meioformula, three have 2n=40 and 19+Xyp and one 2n=46 and 22+Xyp. All but one species with 28 chromosomes show only metacentric or submetacentric chromosomes, whereas the species with 40 and 46 chromosomes display some telocentrics or subtelocentrics, that are probably derived from the former by centric fissions. However, since the number of major chromosome arms is strikingly higher in these latter species (NF=70 and 78) than in the 28-chromosome species (mostly NF=56), other chromosomal rearrangements such as pericentric inversions or heterochromatin accretions could also be involved. The genome sizes display a narrow range, from 1C=0.6–1.22pg, and they are not significantly correlated with the chromosome numbers. Some possible factors implied in the rough chromosomal evolution of Cyrtonus are discussed in relation to a few other genera of the subfamily Chrysomelinae.  相似文献

180 rainbow trouts (Salmo gairdneriRich.), aged from 1 to 3 years, were examined for fluctuations, caused by age and season, by means of colour index (CI), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC).CI and MCH behave similarly. Both are increasing until the 2nd year and stay relatively constant thereafter. If the gender is not considered — there are no significant differences in the values of males and females — the CI increases from 1,4 in the first year over 1,6 to 1,7 in the age of 3 years, and the MCH increases from 44,4 over 52,6 , 56,8 , 58,1 to 55,5 .A seasonal periodicity of both indices could not be indicated on not-matured animals (F2) which were two summers of age. Only, the january values appeared increased — CI: 2, MCH: 68,3 — otherwise the CI varies between 1,8 and 1,7 and the MCH between 53,3 and 59,1 .The MCHC-values of the age groups examined vary between 24,4% and 27,3%. The values of the yearlings form an exception (19,8%). These values certainly are inexact and too low because of the small number of individuals checked (3).

Summary During normal human development, a switch is classically observed in the relative expression of the two globin genes, the G/A ratio varying from 70/30 at birth to 40/60 by the end of the first year. An exception to this developmental pattern is linked to the presence of an XmnI restriction site at a position — 158 to the Cap site of the G gene. Another exception is observed in individuals homozygous for two easily detectable variations of the A gene: the presence of a threonine residue at codon 75 and a HindIII site within the second intron. A 4-bp deletion has been described around position — 225 in some thalassemic patients presenting with these variations. In this study, we find this deletion to be haplotypelinked in a series of 156 individuals of various ethnic origins and presenting with various normal and pathological phenotypes. In sickle cell patients heterozygous for this 4-bp deletion, the relative expression of the A genes on the two chromosomes can be measured by estimating the AT and AI chains, the former always being synthesized at a lower rate. These results suggest a functional role for the deleted sequence.  相似文献

The acridine grasshopper, Caledia captiva exists as two chromosomal races in south-east Queensland. One of these, the Moreton race inhabits the coastal region to the east of the Great Dividing Range. All chromosomes of the complement (2n=11II+XO/XX) have been involved in centromeric rearrangement, which transforms the acro- and telocentric chromosomes into submeta- and metacentric elements. The second, or Torresian race is widely distributed through southern Papua, Arnhem Land, Cape York Peninsula and down the east coast of Australia as far south as Brisbane. This race, which is characterised by a completely acro- and telocentric chromosome complement, approaches the Moreton race in south-east Queensland where the two races are separated by less than 1 km, along a front of at least 150 km. Evidence is presented to show that chromosome introgression is occurring across the contact zone and this takes place in one direction only, namely the Torresian chromosomes are infiltrating into the Moreton race but not reciprocally. Furthermore, the introgression of chromosomes across the zone is limited to certain members of the Torresian complement and even then these successful chromosomes show highly variable degrees of penetrance into the Moreton race. It is proposed that a tension zone exists between these two races which is maintained by the interaction of (a) ecological tolerance differences on either side of the zone and (b) by partial competitive exclusion due to the interracial differences in phenology. This case of parapatric association with limited hybridisation is unique in its clarity due to the marked differences in the appearance of the chromosome complements of these races which permits direct assessment of the behaviour of most members of the genome in hybrids and their derivatives.  相似文献

Eggs from crosses of 40 adult male R. prolixus irradiated with 6K rad -rays with normal females had a mean fertility of 23.9%, only 2 crosses being completely sterile. The 86 F1 progeny of both sexes, when outcrossed with normal mates, had a mean egg fertility of 12.6%, and 43 of these matings were completely sterile. Twenty-eight F2 bugs reared from F1 x normal crosses were mated with normal partners and had a mean fertility of 44.6%, 6 of them being fully fertile, a reversal towards normal fertility. Cytogenetic examination of F1 F2 and F3 males showed that these changes in fertility correlated well with the degree of chromosomal abnormality found. The very high recovery rate of translocations in F1 generation males can be related to the holocentric chromosomes of these bugs which precludes the formation of dicentric chromosomes which are inviable in monocentric species. In F1 and F2 males the majority of translocations were associated as chains of III+I or as chains of IV. Only one bug was found with a ring of IV chromosome association and it is suggested that chromosome morphology, combined with a low chiasma frequency, favours chain association. Most chain multivalents showed linear orientation which may lead to duplication deficiencies and zygotic death. However parallel, indifferent and the more stable convergent modes of chain orientation were also all observed indicating that survival of some translocations in this species may be possible. The survival to the F2 generation of chromosomal fragments confirmed the holocentric nature of triatomine chromosomes. It is suggested that semi-sterile males would prove more effective than releases of completely sterile males for reducing wild populations of R. prolixus, because of the delayed effects of sterilizing radiation consequent upon the holocentric structure of triatomine chromosomes.  相似文献
