Pharyngeal muscle of the planarian Dugesia tigrina was studied by electron microscopy after osmium tetroxide fixation. The muscle cell was observed to contain one myofibril or bundle of myofilaments parallel to its longitudinal axis. The myofilaments were of two types, different in size and distribution. No Z lines or myofilament organization into cross or helical striations were seen. Dense bodies were seen as projections from an invagination of the plasma membrane and as dense lines parallel to the myofilaments. The muscle cells are surrounded by a plasma membrane which is structurally associated with dense body projections, with vesicles and cisternae of sarcoplasmic reticulum, and with synaptic nerve endings. The cell has sarcoplasmic projections perpendicular to its long axis; these projections are seen to contain the nucleus or mitochondria and granules. Mitochondria and granules are also seen in a sarcoplasm rim around the fibril. The dense bodies may serve as attachment for thin myofilaments and function in transmission of stimuli from plasma membrane to the interior of the fibril.  相似文献

In March through April when the oocyte growth in the ovaries of the wall lizard (Hemidactylus) is very rapid, the yolk nucleus continues to persist through various stages of previtellogenesis. This persisting yolk nucleus and associated cell components have been studied with histochemical techniques. The spherical and dense yolk nucleus stains for protein, lipoprotein and RNA. It does not form any close morphological association with the other cell components such as the mitochondria, lipid bodies (L2), spaces or canals, diffuse sudanophilic substance and dense bodies, which are arranged into three zones round the yolk nucleus proper. The mitochondria stain for lipoprotein; the L2 bodies consist of phospholipid; the spaces do not contain any material demonstrable with histochemical techniques; and the ooplasm containing the diffuse sudanophilic substance and dense bodies shows lipoprotein, protein and RNA. Eventually, the yolk nucleus disintegrates, and its substance as well as the other cell components are distributed in the cortical ooplasm of oocytes which are ready to form the yolk bodies. Concepts of the origin, morphology, cytochemistry and function of the yolk nucleus in the oocytes of invertebrates and vertebrates, which have come about recently through the application of cytochemical and submicroscopical techniques, are discussed.  相似文献

Fine structure of gametocytes and oocyst formation of Sarcocystis sp. from Quiscalus quiscula Linnaeus grown in cultured embryonic bovine kidney cells was studied. Microgametocytes measured up to ~5 μm diameter. During nuclear division of the microgametocyte, dense plaques were found adjacent to the nucleus just beneath the pellicle; occasionally microtubules were present within these plaques. These microtubules subsequently formed 2 basal bodies with a bundle of 4 microtubules between them. Microgametocytes also contained numerous mitochondria, micropores, granules, vacuoles, and free ribosomes. Each microgamete was covered by a single membrane and consisted of 2 basal bodies, 2 flagella, a bundle of 4 microtubules, a perforatorium, a mitochondrion, and a long dense nucleus which extended anteriorly and posteriorly beyond the mitochondrion. The bundle of 4 microtubules is thought to be the rudiment of a 3rd flagellum. Macrogametes were covered by a double membrane pellicle, and contained a large nucleus (~2.5 μm), vacuoles, and a dilated nuclear envelope connected with the rough endoplasmic reticulum (ER). In young macrogametes (~4 μm), the ER was arranged in concentric rows in the cortical region, and several sizes of dense granules were found in the cytoplasm. However, in later stages (~8 μm) the ER was irregularly arranged and was dilated with numerous cisternae; only large dark granules remained and a few scattered polysaccharide granules were found. No Golgi apparatus or micropores were observed. After the disappearance of dark granules 5 concentric membranes appeared. Four of these fused to form an oocyst wall composed of a dense outer layer (~66 nm thick) and a thin inner layer (~7 nm). The 5th or innermost membrane surrounded the cytoplasmic mass which was covered by a 2-layered pellicle and contained a nucleus, small amounts of ER, large vacuoles, and mitochondria. The localization of atrial natriuretic factor (ANF)-like immunoreactivity in the central nervous system of the frog Rana ridibunda was examined by the indirect immunofluorescence technique, using an antiserum against synthetic ANF (Arg101-Tyr126). Immunoreactive cell bodies were principally found in the dorsal and medial pallium, the medial septal nucleus, the ventrolateral and anteroventral areas of the thalamus, the lateral forebrain bundle, the posterolateral thalamic nuclei, the preoptic nucleus, the dorsal infundibular nucleus, and the anteroventral tegmentum nucleus of the mesencephalon. Numerous cell bodies and a very dense fiber bundle were visualized in the interpeduncular nucleus. All the areas mentioned above contained a high density of immunoreactive fibers. In addition, the amygdala, the infundibular nucleus, the median eminence, and most of the areas of the mesencephalon contained a moderate number of ANF-positive nerve processes. In the frog pituitary, fibers and nerve terminals were found in the peripheral zone of the neural lobe. The intermediate and anterior lobes of the frog pituitary were totally devoid of ANF immunoreactivity. These results indicate that ANF-like material is widely distributed in the frog brain and that ANF may be involved in various brain functions including neuroendocrine regulations.  相似文献

Sperm ultrastructure of four acotylean (Idioplana atlantica, Armatoplana leptalea, Styloplanocera fasciata, Melloplana ferruginea) and three cotylean polyclads (Pseudoceros bicolor, Phrikoceros mopsus, Enchiridium evelinae) was investigated. All spermatozoa are biflagellate, exhibiting a 9+"1" axoneme pattern. All acotylean axonemes originate and extend within the sperm shaft, and once exiting the shaft, remain attached to it. The flagella of all cotylean spermatozoa exit the shaft immediately and remain free. Structures shared by all species include: an elongated nucleus, in acotyleans located only in the posterior part of the shaft, whereas in cotyleans it extends along the entire sperm body; mitochondria along with small and large dense bodies arranged in a specific pattern; and a ring of microtubules that extends along the entire sperm shaft just beneath the cell membrane. A unique spermatozoon has been found in E. evelinae, where round vesicle-like structures fill the anterior part of the nucleus, and a different type of large dense bodies is present. The spermatozoa of all studied species exhibit numerous characters (axoneme/flagella position, distribution and position of large and small dense bodies, of mitochondria, presence of nuclear vesicles) that may be of phylogenetic value at the family and higher taxonomic levels.  相似文献

SYNOPSIS. Trypanosoma cruzi in tissue cultures was studied with the electron microscope after double fixation in glutaraldehyde and osmium tetroxide, and embedding in Epon. Previous findings on its fine structure were confirmed, and some new structures were found in the flagellum and kinetoplast-chondriome. In the flagellum, an intraflagellar body was found, similar to that observed in other trypanosomes, beginning at the base of the flagellum and running along the axial fibre bundle thruout its length. The axial fibre bundle is formed by interconnecting tubules, the outer ones apparently smooth, the inner ones with a helical substructure. Lateral extensions from the outer tubules in the flagellar bundle seem to enter the intraflagellar body. The kinetopiast in the leishmania bodies has the same electrondense structure described before. In the trypanosome form it has assumed a large spherical shape, in which the formerly short, compressed fibres have grown in length, are more dispersed and have an irregular shape. They are oriented in the direction of the body's length in parallel array. The whole formation is continuous with a long mitochondrion which begins in the region of the nucleus and extends up almost to the tip of the trypanosome. The matrix of the kinetoplast in these forms is electron-transparent; the matrix of the mitochondria is rather dense. In a few extracellular trypanosomes, a special structure was found in which the kinetoplast is composed of electron-transparent formations, arranged in orderly horizontal lines quite similar to the mitochondrial cristae of the parasite. Summary The ultrastructure and composition of cotton (Gossypium hirsutum) pollen, exclusive of the wall, was examined immediately before and after germination. The pollen grain before germination consists of two parts: the outer layer and a central core. The outer layer contains large numbers of mitochondria and dictyosomes as well as endoplasmic reticulum (ER). The core contains units made of spherical pockets of ER which are lined with lipid droplets and filled with small vesicles; the ER is rich in protein and may contain carbohydrate while the vesicles are filled with carbohydrate. Starch-containing plastids are also present in the core as are small vacuoles. The cytoplasm of the pore regions contains many 0.5 spherical bodies containing carbohydrate. After germination the ER pockets open and the lipid droplets and small vesicles mix with the other portions of the cytoplasm. With germination the pore region becomes filled with mitochondria and small vesicles. The vegetative nucleus is large, extremely dense and contains invaginations filled with coils of ER. A greatly reduced nucleolus is present in the generative cell which is surrounded by a carbohydrate wall. The cytoplasm of the generative cell is dense and contains many ribosomes, a few dictyosomes and mitochondria, many vesicles of several sizes, and some ER. No plastids were identified. The generative nucleus is also dense with masses of DNA clumped near the nuclear membrane. An unusual tubular structure of unknown origin or function was observed in the generative cell.  相似文献

The three-dimensional arrangement of vimentin intermediate filaments (IF) was studied in 3Y1, rat fibroblastic cell line, to elucidate its biological role in the cell. While actin filaments were observed exclusively in the superficial part of the cell, vimentin IF were found to be abundantly present in the inside of the cell where microtubules were occasionally discovered. By whole-mount immunoelectron microscopy and computer-graphic reconstruction of serial thin sections, it was observed in more detail that vimentin IF are located very close to the nucleus, endoplasmic reticulum, and mitochondria. Vimentin IF were observed to be attached to these organelles laterally or terminally. Thus, we can reasonably assume that vimentin IF are major cytoskeletal structures deep inside the cell and that they play an important role in supporting the location of the organelles. This is the first report which has visualized the three-dimensional relationship between vimentin IF and the organelles of the cell.  相似文献

Summary

Within the unpaired testis, spermatogonia, spermatocytes, spermatids and spermatozoa were found. In early spermatids, mitochondria take perinuclear positions and centrioles a diplosomal arrangement. Rootlet-like striated differentiations occur in slightly more advanced stages. Then a conical cytoplasmic projection develops, supported by a single row of closely spaced microtubules. At this stage of maturation, giant Golgi stacks occur within the cytoplasm of the cytophore which is rich in different elongate structures and oval dense bodies. With progressive differentiation, the nucleus elongates and its chromatin condenses into twisted lamellae. Two centrioles, which change their diplosomal configuration and come to lie in line to each other, and rootlet-like structures remain near the tip of the median cytoplasmic outgrowth. Mitochondria start to fuse into a single long cylindrical mitochondrial rod extending beside the lengthening nucleus. Bone-shaped rods, smaller dense sticks and dense bodies migrate into the outgrowth. Spermatozoa are totally ensheathed by cortical microtubules. These tubules show different arrangements along the cell body. The thread-like nucleus extends along the cell, the first quarter excepted, whereas the single mitochondrion extends over two thirds of the cell. Two strings with linearly arranged oval dense bodies run in the median to post-median cell segment; four rows of bone-shaped rods and two rows of smaller electron-dense sticks extend from the frontal end up to the beginning of the last third of the cell. All the different longitudinal cords run in the gaps between 4 sets of microtubules. Ciliary axonemes or lateral bristles were not observed. The present findings substantiate the hypotheses, that spermatozoa in the Macrostomida are aciliate and that Myozona takes an isolated position within the Macrostomidae. The occurrence of two centrioles, which come to lie in line to each other and which stay in the tip of the cytoplasmic outgrowth in spermatids, may indicate that biciliate spermatozoa are characteristic for the Rhabditophora (= Macrostomorpha+Trepaxonemata) and not an evolutionary novelty of the Trepaxonemata.  相似文献

A quantitative study regarding the age-related changes occurring in the somatic organelles of the neocerebellar Purkinje cell is carried out, using female rats aged 2 to 24 months. Standard manual morphometric techniques are used to calculate the following parameters: somatic volume, volumetric fractions and absolute volumes of the intracellular structures as well as the membrane profile concentration, the membrane surface concentration and the mean surface of the rough endoplasmic reticulum cisternae per cell (RER-S). From a statistical point of view, all the cell components significantly modify their volumetric fractions (except the multivesicular bodies and nucleolus; the latter in relation to the nucleus) and their absolute volumes (except the mitochondria and the multivesicular bodies); the parameters regarding the reticulum are also modified during ageing. There is a linear trend between the age and either the somatic volume of the RER-S or the absolute volumes of the following structures: mitochondria, dense bodies, ground substance and total cytoplasm. A linear correlation is also observed between the cell volume and either the RER-S or the absolute volume of intracellular structures (the Golgi apparatus, the multivesicular bodies and nucleolus being excluded). Anatomophysiological considerations about the findings are discussed. The role of the ground substance as the major modulator of the volumetric plasticity of the Purkinje cell during ageing, is emphasized as a conclusion.  相似文献

The later stages of sporulation in oocysts of Eimeria brunetti were examined in samples which had been allowed to sporulate at 27 degrees C for 24, 36 and 48 hours. It was observed that the sporoblasts became ellipsoidal and the nucleus underwent the final division. A nucleus with associated Golgi bodies was not observed at either end of the organism. The cytoplasm was limited by two unit membranes and contained rough endoplasmic reticulum, dense bodies, electron translucent vacuoles and mitochondria. The first evidence of sporozoite formation was the appearance of a dense plaque at either end of the organism. This appeared in the vicinity of the nuclei, and adjacent to the limiting membrane of the soroblast. At this stage the sporocyst wall was still unformed. Then the two sporozoites were formed from opposite ends of the organism by growth of the dense plaques and invaginations of the plasmalemma which thus formed the pellicles of the developing sporozoites. A conoid and subpellicular microtubules were observed at this stage as development continued, a number of vacuoles were found between the nucleus and the conoid. These vacuoles constituted the precursors of the rhoptries and micronemes. At the same stage a large dense body had appeared within the forming sporozoite. As the sporozoite developed, this body, anterior refractile body, is followed by the nucleus and another dense body which formed the posterior refractile body. During this period, the thin sporocyst wall was formed and Stieda and sub-Stieda bodies were now present at one end of the sporocyst. Each mature sporocyst contained two sporozoites.  相似文献

Mature sperm has two axonemes of the 9 + '1' pattern incorporated in the sperm body, a row of peripheral microtubules interrupted along part of the sperm by the axonemes, some microtubules in the interior of the sperm and a long lateral extension (lobe) of the sperm body, an elongate nucleus and mitochondrion, and many dense rod-like structures. A supporting rod extends underneath a specialized region consisting of alternating thin and thick transverse rows of irregular dense patches, and with surface ridges around (all or) most of the surface of the sperm. Primary spermatocytes in the prophase of the first meiotic division have synaptonemal complex(es), and are rich in mitochondria. In early spermiogenesis, mitochondria are arranged around the surface of the nucleus, a dense layer appears at one pole of the nucleus, close to an apposed dense layer at the cell membrane in which a row of microtubules develops. The intercentriolar (= central) body develops close to the nucleus. The fully developed intercentriolar body has a regular striation and is located perpendicular and close to the surface of the nucleus. Two flagella extend into the space surrounding the outgoing median process, their basal bodies are located perpendicular to the intercentriolar body and their cross-striated rootlets extend along the surface of the rounded nucleus. At a later stage, rootlets and flagella become more parallel with the intercentriolar body, the nucleus and the fused mitochondria migrate into the median process, and the flagella become incorporated into the median process (= sperm body). The outgrowing spermatozoa are connected to the cytoplasm of the cytophore by dense arching membranes. Finally, rootlets of flagella are resorbed and the spermatozoa are pinched off close to the basal bodies. Two species (Lobatostoma and Multicotyle) of the same family differ strongly in the type of spermiogenesis, although their mature sperm is of the same basic type, i.e. spermiogenesis is not necessarily more useful for phylogenetic considerations than sperm structure.  相似文献

The localization of human GRF1-44-immunoreactive neurons was studied in the rat brain. A dense accumulation of GRF-containing fibers was noted in the external layer of the median eminence. Cell bodies were observed in colchicine-treated rats. The most intensely fluorescent cluster of cells was contained in the arcuate nucleus. Other cells were seen on the base of the hypothalamus, within the median forebrain bundle, dorsal and ventral aspects of the ventromedial nucleus, zona incerta and dorsal part of the dorsomedial nucleus. These cells may influence the pulsatile release of pituitary growth hormone.  相似文献

Summary A light and electron microscopic study was made of the epiplexus (Kolmer) cells of the cat choroid plexus. These polymorphic, motile cells were typically found juxtaposed to the ventricular surface of the choroidal epithelium. They have many ultrastructural features in common with free macrophages of other systems, namely, an indented nucleus with condensed chromatin, sparse mitochondria and endoplasmic reticulum, free ribosomes, multiple Golgi elements, microtubules, coated surface invaginations and microvesicles, and numerous membrane-limited vacuoles and lysosomal dense bodies. A unique feature of epiplexus cells is the manner in which they are anchored to the choroidal epithelium by the invagination of their surfaces by epithelial cell microvilli and cilia.Electron dense tracer particles (biological India ink, Thorotrast, ferritin) injected into the cerebral ventricles were ingested rapidly by epiplexus cells. Uptake of the particles was by way of coated surface invaginations which produced coated cytoplasmic microvesicles. Particle-containing microvesicles subsequently fused with each other and presumably also with pre-existent cytoplasmic vacuoles and lysosomal dense bodies to form storage vacuoles (phagosomes phagolysosomes and residual bodies).Present evidence suggests that epiplexus cells are of hematogenous origin. Under certain conditions these cells may detach from the surface of the choroid plexus to become free-floating cells in the various cerebrospinal fluid compartments of the brain.This investigation was supported by USPHS research grants 1-K04 HD20871, 5 R01 HD 02616 and NB-04456.  相似文献

This is the first study investigating spermatogenesis and spermatozoan ultrastructure in the polyclad flatworm Prosthiostomum siphunculus. The testes are numerous and scattered as follicles ventrally between the digestive ramifications. Each follicle contains the different stages of sperm differentiation. Spermatocytes and spermatids derive from a spermatogonium and the spermatids remain connected by intercellular bridges. Chromatoid bodies are present in the cytoplasm of spermatogonia up to spermatids. During early spermiogenesis, a differentiation zone appears in the distal part of spermatids. A ring of microtubules extends along the entire sperm shaft just beneath the cell membrane. An intercentriolar body is present and gives rise to two axonemes, each with a 9 + “1” micro‐tubular pattern. Development of the spermatid leads to cell elongation and formation of a filiform, mature spermatozoon with two free flagella and with cortical microtubules along the sperm shaft. The flagella exit the sperm shaft at different levels, a finding common for acotyleans, but so far unique for cotylean polyclads. The Golgi complex produces numerous electron‐dense bodies of two types and of different sizes. These bodies are located around a perinuclear row of mitochondria. The elongated nucleus extends almost along the entire sperm body. The nucleus is wide in the proximal part and becomes narrow going towards the distal end. Thread‐like chromatin mixed with electron‐dense intranuclear spindle‐shaped bodies are present throughout nucleus. The general sperm ultrastructure, the presence of intranuclear bodies and a second type of cytoplasmic electron‐dense bodies may provide characters useful for phylogenetic analysis.  相似文献

Ultrastructural observations of the ovary and uterus of Dirofilaria immitis reveal some characteristics of oogonia, oocytes, and uterine sperm. Oogonia are confined to the distal portion of the ovary including a blind tip, where a morphologically distinct terminal cap cell was not observed. These cells contain a nucleus with a nucleolus, numerous dense bodies, scanty ribosomes, lipid droplets, and an occasional mitochondrion. Endoplasmic reticulum is lacking and Golgi complexes were observed only in fully grown oogonia. Primary oocytes located in the middle portion of the ovary are large, elongate, and have a complete set of organelles including many small mitochondria, fragmentary endoplasmic reticulum, ribosomes, Golgi complexes, and very few dense bodies. These cells are arranged into many rosettes about central cytoplasmic masses, the rachises, to which they maintain cytoplasmic continuity by pseudopodlike processes. The rachises contain no organelle except a few dense granules and are bound by winding membranes. Oocytes from the proximal portion differ from those of the middle portion of the ovary in their larger size, round shape, absence of many organelles, presence of small dense granules, and lacking a rachis. Dense bodies are specific to the oogonia and exhibit DNase susceptibility and a positive reaction for a mitochondrial enzyme. These findings together with their decreased number and a concomitant increase of mitochondria in the oocytes suggest a relationship between these bodies and mitochondria.Uterine sperm of D. immitis are of the amoeboid type and contain several chromatin masses without a nuclear envelope, many mitochondria, and specialized membranous organelles referred to as mesosomelike vesicles. The vesicles are probably originated from the sperm plasma membrane. Upon fertilization, the entire spermatozoon penetrates the oocyte and its contents are gradually dissolved in the ooplasm with a simultaneous appearance of large numbers of ribosomes at the site of dissolution. Ribosomes were later found in the nucleus. A pronucleus was not observed. These findings are basically in agreement with those described for Ascaris but differ in the morphologic features and number of rachises, presence of dense bodies, absence of refringent granules in the oocytes and the absence of a refringent body and presence of several chromatin masses in the sperm.  相似文献

The ultrastructure of spermatocytes, spermatids and spermatozoa of the free-living marine nematode Leptosomatides marinae was studied by transmission electron microscopy. In early spermatids, the number of mitochondria, cisterns of the rough endoplasmic reticulum (RER) and dictyosomes increased; the number of membranous organelles (MOs) was insignificant. Later, dictyosomes and MOs filled the cytoplasm. The cytoplasm became distinctly segregated in late spermatids and the MOs concentrated around the nucleus; the mitochondria and organelles of synthesis settled on the cell periphery. Later, a densely packed conglomerate was formed from the central nucleus and a mass of MOs surrounded by an extensive zone of the cytoplasm containing mitochondria and organelles of synthesis. Early spermatozoa had an elongated nucleus surrounded by a layer of cytoplasm containing mitochondria, polarized MOs and bundles of filamentous material, which can be interpreted as fibrous bodies (FBs). The formed spermatozoa had elongated nuclei surrounded by a transparent halo; the cell periphery was a dense matrix, in which MOs and sparse mitochondria were submerged; no FBs were revealed in that phase. In general, the spermatozoa of L. marinae have the main attributes common to the Enoplida spermatozoa, availability of nuclear environment and development of the specific organelles, MOs and FBs, which are not united in complexes.  相似文献

At maturity, spermatozoids of the green algaChara vulgaris are biflagellated, contain little cytoplasm, and coil for approximately 2 1/2 gyres within the mother cell wall. The anterior of the cell contains an ovoid headpiece anchoring two slightly staggered basal bodies that are positioned above and directly in front of approximately 30 linearly arranged mitochondria. An elongated stellate pattern occupies the transition zone between the BBs and axonemes. Flagella emerge from the cell just in front of the nucleus and encircle the full length of the spermatozoid. The spline comprises a maximum of 38 microtubules surrounding the anterior mitochondria and gradually decreases posteriorly to a minimum of 11. The dense nucleus is narrow, cylindrical, and occupies the central revolution of the cell. Six starch-laden plastids and associated mitochondria are linearly arranged at the cell posterior. Phylogenetic analyses of charalean taxa and archegoniates based on spermatogenesis strongly support the orderCharales, withNitella as the sister group toChara. Diagnostic features ofChara spermatozoids include absence of a lamellar strip and axonemes embedded in the cell for almost the entire length of the anterior mitochondria. Potential relationships amongCharales, Coleochaetales and archegoniates are evaluated in regards to the probable course of evolution of streamlined biflagellated gametes.  相似文献
