The diffusion equation according to Fick’s law is solved for a spherical cell, surrounded by an infinite medium with different diffusion properties. The method of Laplace transform is used to obtain the formal solution, however, no inversion can be found for all times and an expansion suitable for small times is performed. The final expression found is expanded further to be more suited for the determination of the diffusion coefficient from an experimental curve. Application to a biological problem is dissussed.  相似文献

The research describe the big amount of ticks of the Amblyomma genus, found on bovines through monthly samplings carried out in ten farms in eight ecological zones (EZ) of Costa Rica. Ticks larger than 4 mm were picked up from the right side of the animals during the visit. The study compiled meteorological information for some farms located in the experiment, showing that the most fluctuant variable is rainfall. The most important Amblyomma species found was A. cajennense. Amblyomma nymphs were found only from January to May, which coincides with the lower humidity season in the rain seasonality area; as for it is expected only one generation per year. In the lab work Amblyomma nymphs are kept to measure the moulting season and the surviving time under controlled conditions, but no major differences were found between both sexes. The surviving periods show that it is not possible to do a grazing land handling, in order to control this genus species. Adults of the genus Amblyomma are present through all the year, not showing any specific preference for a season. The research divided the investigation areas in rain seasonability and not-seasonality systems. The highest amount of Amblyomma is found given in the rain seasonality system or of pacific influence. A. maculatum is present only in the EZ of Tropical Humid Forest transition to pre-mountainous. Likewise, Ixodes boliviensis is found in the EZ of low mountainous Very Humid Forest.  相似文献

Malate dehydrogenase from the thermoacidophilic archaebacterium Thermoplasma acidophilum is purified 50-fold to electrophoretic homogeneity. The purified enzyme crystallizes readily. Native malate dehydrogenase shows a relative molecular mass of 144 000. It is a tetramer of identical subunits with a relative molecular mass of 36 600. Malate dehydrogenase from Thermoplasma uses both NADH and NADPH as coenzyme to reduce oxaloacetate. The enzyme shows A-side (pro-R) stereospecificity for both coenzymes. The pH optimum for the reduction of oxaloacetate in the presence of NADH is found to be at pH 8.1. At pH 7.4 the Km value for oxaloacetate is found to be 5.6 microM while for NADH a value of 11.7 microM is found. The homogeneous enzyme shows a turnover number of kcat = 108 s-1.  相似文献

Intraspecific diversity and molecular relations among regional populations were studied for 16 moss species in three European regions, Central Europe, Southern Scandinavia, and Northern Scandinavia, based on internal transcribed spacer and rpl16 G2 intron. The range of nuclear diversity values found is mainly similar to that of other organisms, and to that found in bryophytes from other regions, but higher diversity was found in Isothecium alopecuroides (Dubois) Isov. No correlations were found between diversity values or number of haplotypes unique to a region and morphological diversity, geographical distribution range, or regional frequency, possibly since this study did not include sufficiently rare species to reflect the factors affecting such species. Finally, no general differences in diversity levels were found among the three studied regions. When haplotype composition is considered, differences were found among the regions for some species, but again no general inter-regional pattern of intraspecific relationships exists. While it is clear that intraspecific variation is crucial to consider in biodiversity conservation contexts since a high proportion of the total diversity is found below the species level, it is also evident that it is necessary to investigate each individual species rather than to rely on what has been found for other taxa.  相似文献

For a single genetic locus with multiple alleles, Littler and Fackerall (1975, Biometrics 31, 117–123) found the transition density of the allelic proportions in the case of no mutation and no selection. Their method is found to be suitable for a model in which symmetric mutation is allowed between alleles. The sampling probabilities and moments are found. Watterson's (1977, Genetics 85, 789–814) test for selection is found to be non-robust for non-stationary populations.  相似文献

The common excision skin pattern is either a fusiform ellipse or another pattern with dissimilar length and width. The purpose of this study was to define the most advantageous skin pattern regarding skin waste, vertex angle, and scar length. Five skin excision patterns used traditionally for closure of round lesions were analyzed: fusiform ellipse, fusiform circle, rhomboid, mosque, and S-shaped. In the analysis, the pattern characteristics were formulated by geometric principles, from which the results were compared. The smallest skin waste was found in rhomboid and mosque patterns, whereas the largest skin waste was found in the fusiform circle and ellipse. The vertex angle was found to decrease monotonously with the excision length-to-width ratio for all patterns except the mosque shape, which is zero per definition. The paradigm stating that a vertex angle of 30 degrees or less is maintained for length-to-width ratios below 4 in the surgical ellipse was found incorrect. It holds only for rhomboid and S-shaped excisions. The scar length was found almost independent of the pattern, with a variance of 3 percent. The authors conclude that the most advantageous surgical skin patterns are the rhomboid and mosque excisions.  相似文献

Extrafloral nectaries (EFNs) are found in many species of Fabaceae. The aim of this work is to describe the internal morphology of the EFNs from species of Fabaceae found in areas of dry‐seasonal forest in north‐eastern Brazil. All species of Fabaceae with EFNs found were collected and samples were submitted to conventional techniques for anatomical and scanning electronic microscopy analysis. EFNs were found in 35 species, of which 32 were examined anatomically. All types have epidermal cells, secretory tissues and vascular bundles in the EFNs. Sclerenchymatous cells were found between the secretory tissues and the vascular tissues, with a few exceptions. The function of these cells is not clear; however, a role in the transportation of the sap in the nectary or with the support of the secretory tissue is possible. The nectar is released through glandular trichomes, secretory pores or even by breaking the epidermal cells and cuticle. The internal patterns found in the EFNs from different species and genera can provide important information for taxonomic and evolutionary studies in the family. © 2010 The Linnean Society of London, Botanical Journal of the Linnean Society, 2010, 163 , 87–98.  相似文献

The method for purification of biomolecules by a combination of affinity interactions and membrane filtration for separation of unwanted material has been found to be of interest for large-scale work. This study examines the suitability of silica nanoparticles as carriers in the process. Alcohol dehydrogenase and lactate dehydrogenases were chosen as target molecules to be purified. The binding capacity was found to be comparative to what is obtained for high-performance liquid chromatography (HPLC) packing material. Both binding and desorption of the enzymes were found to be effective. The limiting factor of the process was the filtration flow rate.  相似文献

The disulfide-sulfhydryl (SS/SH) ratios of subcellular fractions of rat hepatic tissue were found to vary diurnally with the ratio lowest in the early morning and highest in the early evening. These changes were found in the nuclear, microsomal and cytosol fractions. The primary reaction is the reversible formation of mixed disulfides of glutathione with proteins. This formation is controlled by the activity of thiol transferase and the level of oxidized glutathione (GSSG) as substrate. Several enzymes including mitochondrial and microsomal oxidases, glutathione reductase and peroxidase and glucose-6-phosphate dehydrogenase were found to control the levels of GSSG. An NADPH-dependent microsomal oxidase system, inhibited by GSSG, was found to produce activated oxygen which served as substrate for flutathione peroxidase. Evidence is presented for the concept that the formation of mixed disulfides of proteins with glutathione is a mechanism for maintenance of a disulfide-sulfhydryl ratio such that the integrity of particulate membranes is maintaine during oxidative and reductive stresses on the hepatic cells.  相似文献

The human endocrine cells reacting with the monoclonal antibody HISL-19 were identified with hormone antisera of proven specificity using a double immunostaining procedure. The epitope for HISL-19 was found in all types of pituitary cells except ACTH cells, in thyroid C cells, in all types of adrenal medullary and pancreatic islet cells and in somatostatin and pancreatic polypeptide cells of the gastrointestinal mucosa. No staining was found in parathyroid cells and in most gastrointestinal endocrine cells. Either paranuclear focal accumulation or diffuse cytoplasmic distribution of immunoreactive material were found. The spectrum of HISL-19 immunoreactive cells was found to be only in part complementary to that of cells immunoreactive for chromogranin A. Thus, it is concluded that the monoclonal antibody HISL-19 is a useful addition to other immunohistochemical markers for the detection of cells showing neuroendocrine features.  相似文献

Abstract: Rat brain was found to enzymatically methylate phospholipids to form phosphatidylcholine with S -adenosyl- l -methionine serving as the methyl donor. Methyltransferase activity was localized in the microsomes and synaptosomes. In synaptosomes, at least two enzymes were found to be involved in the formation of phosphatidylcholine. The first methyltransferase which catalyzes the methylation of phosphatidylethanolamine to form phosphatidyl- N -monomethylethanolamine was found to have a pH optimum of 7.5, a low Km for 5-adenosyl- l -methionine and a partial requirement for Mg2. Methyltransferase I is tightly bound to membranes. The second methyltransferase (II) catalyzes the successive methylations of phosphatidyl- N -monomethylethanolamine to phosphatidyl- N , N -dimethylethanolamine and then to phosphatidylcholine. In contrast to methyltransferase I, methyltransferase II has a pH optimum of 10.5, a high apparent Km for S -adenosyl- l -methionine and no requirement for Mg2. Methyltransferase II is easily solubilized by sonication. The highest specific activity for both enzymes was found in the synaptosomal plasma membrane.  相似文献

1. Values for liver weight, in growing and adult male and female mammals, both terrestrial and aquatic, as well as values for hepatic blood flow, blood volume and oxygen consumption are submitted to linear (log-log) regression analysis. 2. The slope of the regression line for liver weight on adult body weight in adult mammals was found to be 0.886. No statistically significant difference was found between male and female, nor between terrestrial and aquatic mammals (at the 1% confidence level). 3. Over about four orders of magnitude there is (on present evidence) a tendency for the mammalian liver to grow as about the 0.94 power of body weight (pre- and post-natal). 4. The slopes of the regression lines for hepatic blood flow, blood volume and oxygen consumption were found to be 0.91, 0.86 and 0.69, respectively. 5. The mean hepatocyte size in fixed tissue of rats was found to be 7400 micrometers 3. 6. It is argued that the slope of the regression line for hepatic oxygen consumption in mammals generally is likely to fall in the range of 0.67-0.77.  相似文献

The base-stacking patterns in over 70 published crystal structures of nucleic acid constituents and polynucleotides were examined. Several recurring stacking patterns were found. Base stacking in the solid state apparently is very specific, with particular modes of interaction persisting in various crystalline environments. The vertical stacking of purities and pyrimidines in polynucleotides is similar to that observed in crystals of nucleic acid constituents. Only partial base overlap was found in the majority of the structures examined. Usually, the base overlap is accomplished by positioning polar substituents over the ring system of an adjacent base. The stacking interactions are similar to those found in the crystal structures of other polar aromatic compounds, but are considerably different from the ring–ring interactions in nonpolar aromatic compounds. Apparently, dipole-induced dipole forces are largely responsible for solid-state base stacking. It is found that halogen substituents affect base-stacking patterns. In general, the presence of a halogen substituent results in a stacking pattern which permits intimate contact between the halogen atom and adjacent purine or pyrimidine rings. Considering differences in the stacking patterns found for halogenated and nonhalogenated pyrimidines, a model is proposed to account for the mutagenic effects of halogenated pyrimidines.  相似文献

The present paper describes the development of a simple and sensitive analytical method for quantification of Peptide T (PT) in rabbit plasma, using standard analytical equipment and on-line column enrichment, without prior extraction, clean-up or derivatization. The analytical procedure was found to be accurate, precise and linear. The accuracy was 100% (range 97–103%) and the mean precision was 8% (range 3–14%) for all (n=6) concentrations (0, 15, 50, 100 and 200 ng/ml). The total recovery was found to be approximately 80%, and it was found to be dependent upon the injection rate onto the extraction column. The correlation between added and found concentrations was 0.9982, and the limit of detection was estimated to be around 5 ng/ml. The method is therefore found to be suitable for bioavailability studies, involving Peptide T, in rabbits.  相似文献

Spleen cells from nonimmunized BALB/c mice were fused with two nonsecreting myeloma lines. The hybrids were selected in HAT medium and screened for Ig production and for antibody activity against actin, tubulin, myosin, thyroglobulin, myoglobin, spectrin, dsDNA, fetuin, and transferrin. Among 161 hybrids secreting Ig, three were found to react with DNA, one with thyroglobulin, and one mainly with myosin. Two of these hybrids could be propagated and further characterized. On the basis of inhibition experiments, one was found to be directed against dsDNA; the other was directed mainly against myosin but at the same time reacted significantly with actin, tubulin, spectrin, and dsDNA. Reactivity with myosin seemed to be concentrated in the light meromyosin subfragment, known to be rich in alpha-helical structure. These results indicate: 1) There are reactive B cell clones directed against self antigens. 2) The antibody specificities found for these antibodies are very similar to those found for natural antibodies in normal human serum and for human monoclonal Ig. 3) The widespread reactivity found for the clone mainly reacting with myosin raises the possibility that the determinant recognized by this antibody is a conformational structure that possibly is associated with alpha-helical structures.  相似文献

The effect of competition between species is considered in terms of a mathematical model. It is found that, except for special relations among the parameters, only one species should remain, as is found experimentally. It is also found that unless new factors arise in the competition, one can calculate an index to predict which of the two species is most likely to become extinct. The index involves quantities measured from isolated populations. A preliminary extimate yields satisfactory values when compared with the experimental results of competition.  相似文献

4-aminobutyrate-2-oxoglutarate aminotransferase (GABA-T) has been found in adrenal medulla. The enzyme from this tissue is very similar to those found in other tissues in respect to their mitochondrial localization, optima pH and responses to cofactor. The enzyme from medulla has substrate km values similar to those for the brain enzyme, while it differs from those found for other tissues such as kidney, liver and platelets.  相似文献
