Water contents of various body compartments were estimated within 9 h of birth in eight baboon neonates and at a mean of 29 days (27-32 days) in seven baboons. All animals were normally grown and delivered spontaneously at term. There was no difference in mean antipyrine space, corrected bromide space, intracellular water, interstitial water, and plasma and blood volumes observed on day 1 and day 29. Mean estimate of red cell volume was lower on day 29 than on day 1.  相似文献

Body water content and distribution were estimated before, during, or after 32 pregnancies in baboons. Water content of the various compartments (in liters) correlated with body weight in both nonpregnant and pregnant baboons, and with length of gestation in pregnant baboons. In proportion to body weight (in ml/kg), most water compartments did not change significantly with length of gestation. Mean plasma volume and blood volume were higher during pregnancy than before or after.  相似文献

Water contents of the various body water compartments were estimated in 16 nonpregnant adult female baboons and compared to human data. Total body water, extracellular water, and plasma volumes were determined by dilution studies. Intracellular water, interstitial water, blood volume, and red cell volume were calculated. The data were compatible with the finding that baboon females probably have less body fat than their human counterparts. Our data establish a basis for longitudinal studies of the changes in body water content and distribution during normal and pathological pregnancies.  相似文献

Background

Obesity and overweight are increasing in prevalence in developed countries as a result of changing dietary habits and a lack of physical activity. The purpose of the present study was to evaluate the changes in body composition during short-term overfeeding using the three-component model, which is composed of fat mass (FM), total body water (TBW), and fat-free dry solids (FFDS).

Methods

Ten healthy men completed 3 days of overfeeding during which they consumed 1,500 kcal/day more energy than consumed in their normal diets. Body composition was evaluated at three time points: the day before and after their normal diets and the day after the 3-day overfeeding diet.

Results

Before and after their normal diets, there were no significant differences in body weight and composition, but after 3 days of overfeeding, body weight, TBW, and FFDS increased 0.7, 0.7, and 0.2 kg, respectively (P <0.0001). There was no significant difference in FM between the normal and overfeeding diets.

Conclusion

This study suggests that TBW gain contributes to weight gain following a short-term overfeeding.  相似文献

In some chronic disease studies, distinctions have been made regarding the importance of body mass index (BMI) as a risk factor in younger versus older men and women. In order to determine the significance of these differences in BMI-disease associations, we determined the extent of age-dependent variations in the relation of BMIs to body composition in large probability samples of U.S. men and women from the First and Second U.S. National Health and Nutrition Examination Surveys (NHANES I and II). BMIs are more highly correlated with estimates of body fat in younger than in older men and women, and with muscle mass in older than in younger adults. Caution should be exercised in interpreting the significance of BMI as a risk factor for chronic disease, particularly in comparison of age groups.  相似文献

Genetic trends for body composition and blood plasma parameters of newborn piglets were estimated through the comparison of two groups of pigs (G77 and G98, respectively) produced by inseminating Large White (LW) sows with semen from LW boars born either in 1977 or in 1998. Random samples of 18 G77 and 19 G98 newborn piglets were used for whole carcass and tissue sampling. Plasma concentrations of glucose, albumin and IGF-1 were determined on 75 G77 and 90 G98 piglets from 18 litters. The G98 piglets had less carcass dry matter, protein and energy (P < 0.01) than their G77 counterparts. When expressed in g/kg birth weight, livers were lighter (P < 0.001) and contained less glycogen (P < 0.01) in G98 piglets, with no difference in the activity of the hepatic glucose-6-phosphatase between G98 and G77 piglets. Concentrations of protein, DNA, RNA in longissimus dorsi muscle were unaffected by selection. Plasma concentrations of glucose (P < 0.05) and IGF-1 (P < 0.01) were lower in G98 than in G77 piglets. On the whole, the results suggest that the improvement in lean growth rate and in sow prolificacy from 1977 to 1998 has resulted in a lower maturity of piglets at birth.  相似文献

Measurement of intracellular volume in monolayers of cultured cells   总被引:1,自引:0,他引:1  
Summary Methods have been developed for measuring the intracellular water space (ICS) in cultured diploid fibroblast monolayers. The values obtained have been compared to similar measurements of ICS in suspended fibroblasts using an oil spin method. Markers commonly used for measurement of total water space (TWS) ([3H]H2O, [14C]urea) and extracellular space (ECS) ([3H]inulin, [14C]l-glucose, [3H]sucrose, [3H]d-mannitol) were investigated for use in cell monolayers. Monolayer incubations were terminated by rapidly rinsing the culture dishes three times with ice cold buffer. The distribution volume of the TWS marker [14C]urea plateaued at 10 to 15 min and was independent of urea concentration. [3H]H2O was not a suitable marker for measuring total water space in cell monolayers because of rapid loss of intracellular label during rinsing. Intracellular space was calculated by subtracting [3H]sucrose space (5 min) from [14C]urea space (20 min) after incubation of fibroblasts with both markers. Values obtained for ICS (mean ± SE;μl/106 cells) of fibroblasts from two cell lines measured in monolayer (1.74±0.11 and 1.60±0.10) and in suspension (1.88±0.07 and 1.78±0.11) was approximately 10% smaller than the values for cell size (2.01 and 2.22) obtained from Coulter Counter sizing. Thus, the methods developed for measurement of ICS in monolayer fibroblasts yield data comparable to those obtained with the more standard oil spin method. Furthermore, the methods can be applied to measurement of ICS in other types of adherent cells. Measurement of total energy expenditure using [2H,18O]water requires both accurate and precise determination of the rates of disappearance of 2H and 18O from body water over time and determination of the 2H and 18O pool sizes. However, the impact of the isotopic determination of body water upon the determination of energy expenditure is often overlooked. For measurement of total body water per se, the delay after administration before sampling body fluids becomes important, and saliva sampling can be used to resolve the timing of early samples for body water determination. For energy expenditure measurement per se, linear regression can be used to define the initial dilution. Because the hydrogen tracer dilutes into a pool significantly larger than body water pool per se due to the presence of labile hydrogens, a correction to the isotope pool size must be applied. The theoretical calculations of the exchangeable hydrogen pool presented here suggest that the hydrogen pool size is <3% greater than the body water pool and data are provided to support this idea. Finally, the two approaches used to define the body water pool space contribution to the calculation of energy expenditure using 2H218O are reviewed. Using a pool size based upon the average of the two pool spaces limits the effect of pool size error in the calculation of energy expenditure.  相似文献

本实验采用脑片膜片钳全细胞记录和生物胞素(biocytin)组化染色相结合的技术,研究出生后(postnatalday,P)3~30日龄大鼠(P3~30)内侧膝状体腹侧部(ventralpartitionofmedialgeniculatebody,MGBv)神经元的电生理和形态学特性的发育变化。结果显示:(1)在P3~30的发育过程中,MGBv神经元的静息膜电位自?40mV降至?67mV(P<0.01);输入阻抗由1832M?降至806M?(P<0.01);时间常数由2.55ms降至0.96ms(P<0.01)。同时,动作电位的幅度、阈值和时程也表现出显著差异(P<0.01);(2)K+通道阻断剂4-AP使P6的MGBv神经元诱发动作电位数目减少,幅度降低,时程变宽,并使P16的动作电位幅度逐渐降低至去极化脉冲终末达到平台电位,而Ca2+通道阻断剂CdCl2仅引起P16的MGBv神经元动作电位的幅度降低,时程延长;(3)在用biocytin标记的MGBv神经元观察到,幼稚MGBv蓬丛样神经元(tuftedneuron)胞体呈圆形或椭圆形,而随着出生后日龄的增长,胞体逐渐变成梭形。轴突出现较早,树突的发育相对较晚,但其发育变化更为显著和复杂。以上结果提示,大鼠出生后MGBv神经元电生理和形态学特性仍有显著的发育变化,且两者明显相关。  相似文献

Summary The concentration and distribution of glycogen in relation to postnatal differentiation of the mouse Leydig cell are studied by biochemical and ultrastructural methods. Glycogen decreases to less than one third in the first twelve days after birth. This decrease is accompanied by modifications of its distribution in the cytoplasm. In the newborn it is abundant and arranged in clusters of beta particles; in the mature Leydig cell, glycogen is found scattered in extremely low concentration interspersed among elements of the endoplasmic reticulum.The role of glycogen during Leydig cell differentiation can be interpreted as a source of energy and/or as a source of building material in the biogenesis of membranous components.This work was supported by Grant M 63,121 from the Population Council, U.S.A.Fellow Consejo Nacional de Investigaciones Cientificas y Técnicas, Argentina.  相似文献

Summary This study establishes a method for determining the concentration of Na and K in single red blood cells from electron probe microanalysis of a cell's Na and K content. To this end, red blood cells were separated into subpopulations according to their buoyant density by means of bovine serum density gradient centrifugation. Cell water and Na+K contents were then determined in each fraction by conventional analytic methods with cell volume estimated from measurements of hematocrits and cell number. It was found that an inverse relationship obtains between the mean cell volume and buoyant cell density since cells increased in size as density decreased. Although the amount of hemoglobin per cell was found to slightly increase as cell density decreased, hemoglobin concentration showed the inverse relationship, indicating that buoyant cell density differences are primarily the result of differences in hemoglobin concentration. In confirmation of Funder and Wieth (Funder, J., Wieth, J.O. 1966.Scand. J. Lab. Invest. 18:167–180) cell water and cell volume was found to vary directly with the summed content of Na+K. Finally, by means of electron probe microanalysis of single cells, the cellular concentration of hemoglobin was found to vary inversely with the Na+K content, providing a quantitative basis for directly estimating cell volume, and thus ionic concentration, with this technique.  相似文献

The present study was conducted to evaluate and compare the physiological responses of sheep and goats to water restriction using the deuterium dilution technique (D2O) to predict the total water intake (TWI) in both species. In two consecutive trials, 10 non-lactating Boer goats and 10 non-lactating German black-head mutton ewes were each randomly allocated into a treatment and a control group. In the control groups (n = 5, for each species), water was offered ad libitum, whereas the treatment groups (n = 5, for each species) received water 3 h/day on experimental days 8 to 14 and 6 h every 2 days on experimental days 15 to 22. The respiratory rate, rectal temperature, body mass and drinking behaviour were also recorded. The TWI was estimated by D2O for each animal. Water restriction for 21 h/day or 42 h/2 days had no significant (P > 0.05) effect on water intake (WI), feed intake, WI to dry matter intake ratio or body mass in both species. The absence of differences between species in their WIs was also confirmed using D2O. However, sheep had higher respiratory rates and rectal temperatures than goats in both control and treatment groups. Both species showed the ability to tolerate a moderate water shortage by activating several physiological mechanisms and behavioural strategies.  相似文献

Purpose: In a recent small sample study, red blood cell distribution width (RDW) was suggested as a predictor of homocysteine levels. The current study was aimed to reexamine this association in a large scale sample.

Methods: A retrospective cross-sectional study of healthy adults, conducted at Rabin Medical Center, during 2000–2014. Data were retrieved from the medical charts and a logistic regression controlling for interfering factors was carried out. Sensitivity analysis was implemented by exclusion of individuals with anaemia.

Results: Five thousand, five hundred fifty-four healthy individuals were included. Mean serum homocysteine level was 10.10 (SD 2.72) μmol/L. 34.4% of the study population had a homocysteine level higher than the upper limit of normal (10.8?μmol/L). Homocysteine showed no association with RDW (OR 1.00; 95% CI 0.97–1.03), but increased with age (OR 1.05; 95% CI 1.04–1.06) and decreased with a rise in haemoglobin (OR 0.77; 95% CI 0.71–0.83), and in the mean corpuscular volume (OR 0.86; 95% CI 0.85–0.88). Exclusion of individuals with anaemia did not reveal an association between homocysteine and RDW but found a somewhat smaller association between haemoglobin and RDW [OR 0.82; 95% CI 0.73–0.91].

Conclusions: In our large scale sample we did not find an association between RDW and serum homocysteine.  相似文献

The plasma membrane of red blood cells permits sugars to be loaded into the cytoplasm simply by incubation in a suitable buffer solution containing the sugar. This may provide some hope for the freeze-drying of human red blood cells. However, the effect of the loading process on red blood cells has not been fully investigated. The exposure of phosphatidylserine (PS) on the surface of the cell can be recognized by macrophages and result in shortened circulation in vivo. This study evaluates the effects of the concentration, the incubation time, and the temperature of exposure of human red blood cells to extracellular trehalose or glucose. Exposure of PS was demonstrated by annexin V labeling. It was shown that the efficiency of loading of glucose was significantly greater than that of trehalose. The loading efficiency of both sugars increased with increase in extracellular sugar concentration, prolongation of incubation time, and increase of incubation temperature. The percentages of cells with exposed PS and of damaged cells were dependent on the extracellular sugar concentration, the incubation time, and the temperature. With an extracellular glucose concentration of 0.8M, the percentage of cells with exposed PS was more than 80% and significantly higher than that of red blood cells loaded with trehalose (approximate 20%, P<0.01). As the incubation time was prolonged, the percentage of PS exposure and of damaged cells also increased. After incubation for 5h, the percentage of red cells with exposed PS following loading with glucose was more than 80% and significantly higher than that of cells loaded with trehalose (40%, P<0.01). In addition, the incubation temperature had a major effect on PS exposure. The percentage of cells with PS exposure and the proportion of damaged cells increased with increase of incubation temperature. At 37 degrees C, the percentage of cells with exposed PS and of damaged cells after loading with glucose was more than 80% and significantly higher than that of cells loaded with trehalose (P<0.01). However, when the temperature was below 25 degrees C, the percentage of cells with exposed PS and of damaged cells after loading with glucose or trehalose were both less than 10%. In conclusion, the loading efficiency for glucose was higher than that for trehalose, but the lesser effect of trehalose on exposure of PS suggests that it can maintain the asymmetrical distribution of membrane phospholipids and the intracellular trehalose can increase the osmotic tolerance of cells.  相似文献

This experiment evaluated different strategies for allocating first-cut grass silages to dry dairy cows that had low body-condition score (BCS) at drying off. A total of 48 moderately yielding Holstein-Friesian cows were used, receiving one of three dietary treatments in the dry period and a single lactation diet based on a flat-rate of concentrates and grass silage ad libitum. Throughout the dry period, one group received a low-digestibility silage (harvested 15 June 1998; LL; metabolisable energy (ME) = 10.3 MJ/kg dry matter (DM)) and a second group received a high-digestibility silage (harvested 9 May 1998, HH; ME = 11.7 MJ/kg DM). A third strategy (LH) offered the low-digestibility silage in the early dry period and the high-digestibility silage in the final 3 weeks before calving. The silages had very different crude protein concentrations (144 and 201 g/kg DM) and intakes were widely divergent (10.1 v. 13.5 kg DM/day) across the dry period. No concentrates were fed during the dry period. Silage quality had a very large effect on liveweight change, with treatment means of 0.32 and 1.75 kg/day for LL and HH, respectively. BCS changes followed a similar pattern, though no cows became over-conditioned and blood metabolites were within normal ranges. Increased silage digestibility in the late dry period led to a substantial increase in milk fat concentration and a smaller increase in milk protein concentration, the latter confined to the first full week of lactation. Depression of milk fat appears related to low blood glucose when dry cows in low body condition are fed at a low level. The LH strategy avoided the tendency for lower milk yields and fat concentration that resulted from feeding the low-digestibility silage until calving. This strategy also avoided the higher calf weights that resulted from feeding the high-digestibility silage in the early dry period.  相似文献

Body water content was determined on groups of Cyprinodon variegatus after sequential acclimation to ambient salinities in the range from fresh water to 100%. A multiple linear regression model applied to the data included body mass ( M ) of individuals and acclimation salinity ( S ) as independent variables, and body water content ( W ) as the dependent variable. The model that described the relationship was W (g) = 0·059 + 0·723 M (g)–0001 S (%). Converting values of body water content to percent of wet body mass produced a range of values from 74·5% in fresh water to 72·0% at 100%, only a 2·5% difference in body water content over this wide range of ambient salinities.  相似文献

Purpose

The purpose of this study was to investigate the effectiveness of a composite weight-loss dietary supplement on body composition and blood lipid changes in middle-aged women.

Methods

Thirty seven middle-aged women living in the Kyunggi area participated in this study and they were randomly divided into 2 groups (Dietary supplement ingestion group; DG, n = 20 and Placebo group; PG, n = 17). Blood draw and dual energy x-ray (DEXA) measurements were conducted to examine changes in body composition and blood lipids.

Results

There were no significant changes in weight and BMI in both groups. There was an interaction between the composite weight-loss dietary supplement intake and lean body mass in DG and there was a significant decrease in percent body fat in DG. Blood lipid changes in the study results showed that there was no significant difference in TC, TG, and LDL in both groups; however, there was a significant interaction between the composite weight-loss dietary supplement intake and HDL-C as well as an increase in the HDL-C of DG.

Conclusion

In conclusion, it seems that 4-week ingestion of the composite weight-loss dietary supplement decreased body fat, increased lean body mass, and increased HDL-C. Therefore, the composite weight-loss dietary supplement is expected to prevent obesity and induce health improvements in middle-aged women.  相似文献

Abstract. Anopheles stephensi mosquitoes which had fed upon mice infected with Plasmodium yoelii nigeriensis malaria parasites produced significantly fewer eggs than mosquitoes fed on an uninfected mouse. Fecundity reduction was more pronounced when the bloodmeal contained malaria gametocytes and the mosquitoes developed oocysts. Egg production and haematin excretion were correlated for uninfected bloodfed mosquitoes; the presence of P.y. nigeriensis in the blood affected this relationship. Reduced fecundity was associated with a significant reduction of bloodmeal size (measured by haematin excretion) in mosquitoes which ingested gametocytaemic blood. The bloodmeal size in mosquitoes fed on parasitaemic blood without gametocytes was not significantly reduced. The use of haematin assays for determination of bloodmeal size in mosquitoes is discussed.  相似文献
