The mathematical method of experimental design was used to develop a new enzymic medium for cultivation of Bacillus thuringiensis H14 266/2-1, a bactoculicide producer. The optimized medium based on corn flour enzyme lysate as a carbon source and fodder yeast enzyme lysate as a source of nitrogen amine made it possible to increase twice the titre biomass yield for 24 h cultivation as compared to the initial medium. The above medium does not yield to the initial production medium in the insecticide activity.  相似文献

好氧反硝化作用的发现打破了反硝化只能在严格厌氧条件下进行的传统认知,为生物脱氮提供了一条新的途径,已成为近些年的研究热点。碳源可为好氧反硝化过程提供能量和电子供体,其代谢难易程度直接影响着好氧反硝化细菌的脱氮效率,因此有必要明确碳源在好氧反硝化脱氮过程中的代谢机理。基于此,本文阐述了好氧反硝化细菌的种类及其对硝态氮与亚硝态氮的代谢途径;系统分析了不同好氧反硝化细菌对碳氮源代谢的差异与代谢机理;综合分析了碳代谢差异对好氧反硝化脱氮过程的影响,并对未来的研究方向进行了展望,旨在深入理解好氧反硝化细菌同时去除碳氮的机理,为提高废水生物脱氮除碳效率提供理论依据。  相似文献

Polyhexamethylene biguanide (PHMB), a biocide used in a wide variety of disinfection and preservation applications, is a polydisperse mixture in which the end-groups may be any combination of amine, guanidine and cyanoguanidine. Using PHMB model compounds (1,6-diaminohexane; 1,6-diguanidinohexane; 1,6-di(cyanoguanidino)hexane; 4-guanidinobutyric acid), we have determined the biodegradation characteristics of each end-group in several strains of bacteria isolated for their ability to utilise PHMB as a sole source of nitrogen. Bacteria were screened for growth at the expense of each model compound (at non-inhibitory concentrations) as sole nitrogen source. None of the isolated bacteria was capable of utilising a cyanoguanidine end-group as growth substrate, whereas several bacteria were shown to utilise amine or guanidine end-groups. In particular, a strain of Pseudomonas putida was capable of extensive growth with 1,6-diguanidinohexane as a sole nitrogen source, with complete removal of guanidine groups from culture medium within 2 days, and with concomitant formation of unsubstituted urea, which in turn was also utilised by the organism. We conclude that whilst amine and guanidine end-groups in PHMB are likely to be susceptible to biodegradation, cyanoguanidine end-groups are likely to be recalcitrant.  相似文献

Klebsiella pneumoniae RS-13 and Acetobacter liquefaciens S-1, both methyl red (MR)-degrading bacterial strains, degraded N,N'-dimethyl- p -phenylenediamine (DMPD) under aerobic conditions. DMPD, a toxic and mutagenic aromatic amine, is formed during the reductive cleavage of azo dyes such as MR. The effects of physical parameters, such as temperature and aeration, and chemical parameters, such as pH and concentrations of glucose, ethanol and ammonium sulphate in the culture medium, on the degradation of DMPD by these bacteria were determined. Klebsiella pneumoniae RS-13 degraded DMDP more efficiently than A. liquefaciens S-1 under all physicochemical conditions, except in the presence of ethanol as carbon and energy sources. In addition, Kl. pneumoniae RS-13 degraded DMDP at low levels of carbon and nitrogen at pH 6–8. These results indicate that it is feasible to use Kl. pneumoniae RS-13 to completely degrade the detoxify MR under aerobic conditions.  相似文献

Various bacteria, actinomycetes, and filamentous fungi decomposed methionine, but only certain aerobic bacteria isolated from soil decomposed it in the absence of other organic substrates. These bacteria could grow on methionine as the only organic substrate and source of nitrogen and sulfur. Methionine was first deaminated and then demethiolated with production of methanethiol, part of which was oxidized to dimethyl disulfide. The amount of methanethiol that was oxidized varied with different cultures. A bacterial culture initially unable to grow on methionine developed capacity to do this in a medium which contained methionine and other growth substrates. The two sulfur products, methanethiol and dimethyl disulfide, are volatile and escaped from the media, resulting in a decrease in the sulfur content proportional to the amount of methionine decomposed.  相似文献

An ability to synthesize and accumulate in the growth medium for extracellular amino acids of 108 strains of 13 aerobic sporeforming bacteria species at deep cultivation on the simple synthetic glucose-mineral liquid nutrient medium optimized for the representatives of this genus. Has been studied the bacillus strains studied in the present conditions synthesize 19 amino acids and 2 amino carbons in different quantities and combinations. Amino acids being synthesized the most and least by these microorganisms have been determined in quantitative expression. Extracellular amino acid accumulation was proved to be a strain property not of genus. In general, the genus studied was evaluated as active one in amino acid production. The most active strains may present an interest as initials for further selective obtaining of producers of separate amino acids. The outlook for aerobic bacilli strains having marked amino-synthesizing properties and complex of other biological activities is being discussed for construction of medicinal and prophylactic of live microbial culture preparations.  相似文献

Abstract: Methanobacterium formicicum utilized molecular nitrogen as the sole nitrogen source for growth as monitored by methane production and culture turbidity measurements. The rate of methane production by the bacteria was correlated to nitrogen gas concentrations. In the absence of nitrogen gas or any other nitrogen source, the bacteria completely stopped growing. The presence of selenium and molybdenum in the culture medium was vital for the growth of the bacteria under nitrogen fixing conditions.  相似文献

Aerobic production of rhamnolipid by Pseudomonas aeruginosa was extensively studied. But effect of medium composition on anaerobic production of rhamnolipid by P. aeruginosa was unknown. A simplifying medium facilitating anaerobic production of rhamnolipid is urgently needed for in situ microbial enhanced oil recovery (MEOR). Medium factors affecting anaerobic production of rhamnolipid were investigated using P. aeruginosa SG (Genbank accession number KJ995745). Medium composition for anaerobic production of rhamnolipid by P. aeruginosa is different from that for aerobic production of rhamnolipid. Both hydrophobic substrate and organic nitrogen inhibited rhamnolipid production under anaerobic conditions. Glycerol and nitrate were the best carbon and nitrogen source. The commonly used N limitation under aerobic conditions was not conducive to rhamnolipid production under anaerobic conditions because the initial cell growth demanded enough nitrate for anaerobic respiration. But rhamnolipid was also fast accumulated under nitrogen starvation conditions. Sufficient phosphate was needed for anaerobic production of rhamnolipid. SO4 2? and Mg2+ are required for anaerobic production of rhamnolipid. Results will contribute to isolation bacteria strains which can anaerobically produce rhamnolipid and medium optimization for anaerobic production of rhamnolipid. Based on medium optimization by response surface methodology and ions composition of reservoir formation water, a simplifying medium containing 70.3 g/l glycerol, 5.25 g/l NaNO3, 5.49 g/l KH2PO4, 6.9 g/l K2HPO4·3H2O and 0.40 g/l MgSO4 was designed. Using the simplifying medium, 630 mg/l of rhamnolipid was produced by SG, and the anaerobic culture emulsified crude oil to EI24 = 82.5 %. The simplifying medium was promising for in situ MEOR applications.  相似文献

To enhance the production of micrococcin GO5, a bacteriocin produced by Micrococcus sp. GO5, cultivation conditions and medium composition were optimized. The optimal initial pH and temperature for bacteriocin production were 7.0-9.0 and 37 degrees C, respectively. Micrococcus sp. GO5 displayed the highest micrococcin GO5 activity when grown in modified MRS medium that contained lactose or sucrose, rather than glucose, as a carbon source. The maximum bacteriocin activity was obtained in modified MRS medium containing 0.5% tryptone and 1.0% yeast extract as nitrogen sources instead of the other nitrogen sources present in MRS medium. Bacteriocin production was greatly affected by the concentration of K(2)HPO(4); strain GO5 produced eight-fold more bacteriocin in medium containing 2.0-2.5% K(2)HPO(4) than in medium containing 0.2% K(2)HPO(4). The optimal concentration of MgSO(4).7H(2)O for bacteriocin production was 0.5%. The production of micrococcin GO5 was increased 32-fold in shake flask culture and 16-fold in a bioreactor using the optimized medium (TY medium), compared with culturing in MRS medium.  相似文献

研究了国产及进口LB培养基对产嗜热性木聚糖酶基因工程菌1020生长及产酶的影响,并以国产原料为基准,优化了培养基。结果表明,国产的酵母膏及蛋白胨和进口原料相比,缺少某些成分,单纯增加用量不能弥补这种差别;乳糖可以代替昂贵的IPTG起到有效的诱导作用;10g·L-1的乳糖可兼起能源及诱导剂的双重效果,菌的生物量和产酶量达到最佳;发酵8h后补加5g·L-1乳糖,生物量及酶活皆有提高。复合氮源优于单一的无机或有机氮源。优化后的酶活从进口LB培养基的270U~290U提高到1700U~1800U。  相似文献

Herein we describe a novel and effective screening method for aerobic denitrifying bacteria. For this procedure, we utilized KCN to inhibit the electron transference from Cytaa3 to oxygen in the bacteria respiratory chain. We employed a 3-h aeration operation cycle and intermittent rotations. The resultant bacterial suspensions were plated on a KCN-screening medium and incubated aerobically. Single colonies were selected and incubated in an aerobic culture medium. Culture nitrate and nitrite levels were determined over time, and ultimately four bacterial strains that performed denitrifying under aerobic conditions were identified by this method. Of these, strain Y2-1-1 demonstrated the best aerobic denitrifying ability. In a 5-day test, the NO3--N of the aerobic culture medium was reduced from 282.0+/-8.3 mg L(-1) to 149.2+/-17.1 mg L(-1), with little nitrite or N2O production. The morphological, physiological and biochemical characteristics and the 16S rRNA gene sequence homology comparison data for this strain were consistent with the classification of the genus Pseudomonas. We named this strain Pseudomonas sp. Y2-1-1.  相似文献

The Rhizobium sp. isolated from healthy and mature root nodules of a leguminous tree, Dalbergia lanceolaria Linn. f., preferred mannitol and KNO3 for growth as carbon and nitrogen sources, respectively. The bacterium produced a high amount (22.3 microg/ml) of indole acetic acid (IAA) from L-tryptophan supplemented basal medium. Growth and IAA production started simultaneously. IAA production was maximum at 20 hr when the bacteria reached the stationary phase of growth. Cultural requirements were optimized for maximum growth and IAA production. The IAA production by the Rhizobium sp. was increased by 270.8% over control when the medium was supplemented with mannitol (1%,w/v), SDS (1 microg/ml), L-asparagine (0.02%,w/v) and biotin (1 microg/ml) in addition to L-tryptophan (2.5 mg/ml). The possible role of IAA production in the symbiosis is discussed.  相似文献

The fungus Fusarium oxysporum 11dn1 was found to be able to grow and produce nitrous oxide on nitrate-containing medium in anaerobic conditions. The rate of nitrous oxide formation was three to six orders of magnitude lower than the rates of molecular nitrogen production by common denitrifying bacteria. Acetylene and ammonia did not affect the release of nitrous oxide release. It was shown that under anaerobic conditions fast increase of nitrate reductase activity occurred, caused by the synthesis of enzyme de novo and protein dephosphorylation. Reverse transfer of the mycelium to aerobic conditions led to a decline in nitrate reductase activity and stopped nitrous oxide production. The presence of two nitrate reductases was shown, which differed in molecular mass, location, temperature optima, and activity in nitrate- and ammonium-containing media. Two enzymes represent assimilatory and dissimilatory nitrate reductases, which are active in aerobic and anaerobic conditions, respectively. 【背景】好氧反硝化是指在有氧条件下进行反硝化作用,使得硝化和反硝化过程能够在同一反应器中同时发生,是废水脱氮最具竞争力的技术。红树林湿地中蕴藏着丰富的微生物资源,分布着大量好氧反硝化微生物。【目的】了解耐盐微生物的脱氮机制,为含盐废水生物脱氮的工程实践提供理论依据,对一株分离于红树林湿地中的耐盐好氧细菌A63的硝酸盐异化还原能力进行分析。【方法】利用形态学特征及16S rRNA基因序列测定分析,对其种属进行了鉴定,采用单因子实验测定该菌在不同环境因子下的硝酸盐还原能力,并对其反硝化脱氮条件进行了优化。【结果】初步判定该菌株为卓贝儿氏菌(Zobellellasp.),其能在盐度0%-10%、pH5.0-10.0、温度20-40°C范围内进行反硝化脱氮和硝酸盐异化还原为氨(dissimilatorynitratereductiontoammonium,DNRA)作用。菌株A63最适生长碳源为柠檬酸钠(1.2 g/L),适宜脱氮盐度为3%、pH 7.0-7.5、温度30-35°C,且C/N为10。在最适脱氮条件下,该菌株12h内能将培养基中208.8mg/L硝态氮降至0,且仅有少量铵态氮生成,无亚硝态氮积累,脱氮率高达99%。此外,该菌株在高盐度、低C/N比、弱酸性和低温等不利生境中DNRA作用显著。【结论】细菌A63生长范围宽,脱氮效率显著,适用于海水养殖废水处理。研究为今后开发高效含盐废水生物脱氮工艺奠定了基础,对于加深氮素转化规律的认识、丰富生物脱氮理论有着重要意义。  相似文献

As part of an investigation into the use of biological nitrogen fixation for fertilizer ammonia production, continuous culture studies of respiration and nitrogen fixation in the aerobic bacteria Azotobacter vinelandii under oxygen-limited conditions were conducted. Respiration and growth rates followed Monod forms with respect to dissolved oxygen concentration. However, specific nitrogen fixation rate and nitrogenase activity exhibited maximum values at dissolved oxygen concentrations of ca. 0.02 mM (10% of air saturation). These results suggest careful control of oxygen in the environment is necessary to optimize fixed nitrogen production by this organism.  相似文献

Nitrogen-fixing activity associated with different wetland rice varieties was measured at various growth stages by an in situ acetylene reduction method after the activities of blue-green algae (cyanobacteria) in the flood water and on the lower portion of the rice stem were eliminated. Nitrogen-fixing activities associated with rice varieties differed with plant growth stages. The activities increased with plant age, and the maximum was about at heading stage. The nitrogen fixed during the whole cropping period was estimated at 5.9 kg of N per ha for variety IR26 (7 days) and 4.8 kg of N per ha for variety IR36 (95 days). The population of aerobic heterotrophic N2-fixing bacteria associated with rice roots and stems was determined by the most-probable-number method, using semisolid glucose-yeast extract and semisolid malate-yeast extract media. The addition of yeast extract to the glucose medium increased the number and activity of aerobic heterotrophic N2-fixing bacteria. The glucose-yeast extract medium gave higher counts of aerobic N2-fixing bacteria associated with rice roots than did the malate-yeast extract medium, on which Spirillum-like bacteria were usually observed. The lower portion of the rice stem was also inhabited by N2-fixing bacteria and was an active site of N2 fixation.  相似文献

【目的】生物除氮中反硝化菌具有重要的作用,需氧反硝化菌研究较少,有着很好的应用潜力,本研究主要从环境样品中分离具有高效去除铵氮和亚硝酸盐氮活性的好氧反硝化菌,并对其分类及除氮特性进行研究。【方法】以高效去除铵氮、除亚硝酸盐氮和好氧反硝化能力为主要指标,从富营养化的池塘淤泥水和工厂污泥样品中进行菌株分离筛选。通过生理生化特点以及16S rRNA序列分析对活性最好的菌株进行初步鉴定。在好氧条件下,分别以NO-3-N、NH+4-N和NO-2-N作为唯一氮源,考察菌株的好氧反硝化特性、去除铵氮和亚硝酸盐氮特性,以及不同初始pH值、温度、碳源、摇床转速对该菌去除铵氮和亚硝酸盐氮特性的影响。【结果】得到的细菌中,以菌株C-4的活性最好,其16S rRNA序列与不动杆菌的同源性达99%,结合生理生化特点,初步确定菌株C-4属于不动杆菌属(Acinetobacter sp.)。以柠檬酸钠作为碳源,30℃、120 r/min振荡培养,种龄为18 h,用初始pH为8.5的200 mg/L NH +4-N培养基和初始pH为7.5的100 mg/L NO -2-N培养基进行测定,分别培养15 h与12 h,净除氮率分别达到65.8%和47.8%。【结论】从鱼塘水样中分离到一株好氧反硝化菌C-4,初步鉴定为不动杆菌属的一个种(Acinetobacter sp.),具有较高的反硝化特性和高效去除铵氮与亚硝酸盐氮的能力,在处理实际池塘污水时中,净除氮率可达73.04%以上。  相似文献

Influence of nitrogen source on proteinases synthesis in aerobic alkalotolerant and halotolerant bacteria from soda-salt lakes of Transbaikalia was studied. Maximal accumulation of proteinases was revealed on medium with peptones. Introduction of various sources of nitrogen in the medium did not result in increase of enzyme activity in cultural liquid. It was indicated that secreting proteinases of the studied bacteria strains possess narrow substrate specificity, hydrolyze proteins and n-nitroanilide substrates have maximal activity during GlpAALpNA hydrolysis. Data of inhibitory analysis and substrate specificity of studied extracellular enzymes indicate that they belong to a class of serine proteinases of subtilisin-like type.  相似文献

The influence of medium composition and culture conditions on Streptococcus pneumoniae serotype 23F cultivation was investigated in order to develop an industrial method for polysaccharide (PS) production. Acid-hydrolyzed casein (AHC) and dialyzed enzymatically hydrolyzed soybean meal (EHS) were investigated as nitrogen sources, and the vitamin solution of Hoeprich's medium and dialyzed yeast extract as vitamin sources. The influence of initial glucose concentration was also evaluated. In flask experiments, the best nitrogen source for PS production was AHC; EHS yielded small amounts of PS without interfering with bacterial growth. Dialyzed yeast extract provided an approximately 2-fold increase in PS production when compared to Hoeprich's vitamin solution. In a 5-l bioreactor, it was observed that the pneumococcus did not grow under aerobic conditions, CO(2) did not increase PS yield, glucose was inhibitory above 30 g l(-1), and the main glucose catabolism product was lactate, which had an inhibitory effect on cell growth. When anaerobic cultivation was performed under N(2) flow using the optimized medium, 240 mg l(-1) of soluble PS was obtained, which represents a 3-fold increase in yield as compared to that described in the published patent [Yavordios and Cousin (1983) European Patent 0 071515 A1]. Application of these results would considerably simplify upstream and downstream processes for PS production.  相似文献
