Pine wilt disease seriously endangers pine tree growth and can lead to wilting and death. In this study, Masson pine (Pinus massoniana) was used as the donor plant to study miRNA expression by high-throughput sequencing in needle leaves of trees during the first 3 days after infection by Bursaphelenchus xylophilus. The results showed that the number of differentially expressed miRNAs first increased and then decreased in samples within 3 days of infection, with the highest expression in samples collected on day 2. Analysis of miRNA target genes in needles on different days after infection revealed enrichment in two pathways: plant hormone signal transduction and RNA transport. Further comparison of the samples indicated differential expression of 10 miRNAs on different days after infection, and the corresponding target genes, such as those participating in plant hormone signal transduction, were also significantly enriched. In addition, the concentrations of zeatin and indole acetic acid in needle tissues were significantly lower after infection than in the control sample. The above results indicated that B. xylophilus infection influenced the expression of miRNAs related to plant hormone signal transduction in P. massoniana, decreasing plant hormone synthesis and ultimately affecting the growth of Masson pine. 相似文献

Tuber huidongense and T. liyuanum are common commercial white truffles in China that belong to the Rufum and Puberulum groups of the genus Tuber, respectively. Their mycorrhizae were successfully synthesized with two native trees—Castanea mollissima and Pinus armandii—under greenhouse conditions. The identities of the mycorrhizae were confirmed through internal transcribed spacer (ITS) sequence analyses, and their morphological characteristics were described. All of the obtained mycorrhizae have an interlocking pseudoparenchymatous mantle, which is a typical feature of truffle mycorrhizae. The mycorrhizae of T. huidongense on the two trees have hyaline branched emanating hyphae, similar to the documented mycorrhizae of the Rufum group. The unramified, spiky, and hyaline cystidia on the mycorrhizae of T. liyuanum with both C. mollissima and P. armandii further confirmed that this characteristic is constant for the mycorrhizae of the Puberulum group. The successful mycorrhizal syntheses on the two nut-producing trees will be of economic importance in the cultivation of the two truffles. 相似文献

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes. 相似文献

of 2- isopenthenyladenine (2iP) and indole-3-acetic acid (IAA), either singly or in combinations, as potential medium for
shoot multiplication by nodal segments was tested. In the following experiment, equal molar concentrations of four cytokinins
[2iP, kinetin, zeatin, and N
6-benzyladenine (BA)] in combination with equal molar concentrations of three auxins [IAA, α-naphthaleneacetic acid (NAA),
and indole-3-butyric acid (IBA)] were tested for ability to induce axillary shoot development from single-node stem segments.
The highest rate of axillary shoot proliferation was induced on MS agar medium supplemented with 1.96μM 2iP and 1.14μM IAA
after 6 wk in culture. Different auxins (IAA, IBA, and NAA) were tested to determine the optimum conditions for in vitro rooting of microshoots. The best results were accomplished with IAA at 5.71μM (89% rooting) and with IBA at 2.85 or 5.71μM
(86% and 86% rooting, respectively). Seventy and 90 percent of the microshoots were rooted ex vitro in bottom-heated bench (22 ± 2°C) after 2 and 4 wk, respectively. In vitro and ex vitro rooted plantlets were successfully established in soil. 相似文献

This study aimed to investigate physiological and protein expression alterations of mycorrhizal Pinus massoniana Lamb. inoculated with Lactarius insulsus in response to drought stress. The P. massoniana seedlings were inoculated with L. insulsus (Li group) and ectomycorrhized fungal-free filtrate (control, CK group), respectively. After two and a half years, all the plants were exposed to a simulate drought condition without water for 21 days. The soil relative water content (SRWC), wilting degree (WD) and wilting rate (WR) of the plants were measured and root proteome was analyzed based on two-dimensional gel electrophoresis (2-DE), respectively at four time points as 0, 7, 14 and 21 days during the whole drought period. Finally, the electrospray ionization mass spectrometry (ESI-MS) was used to identify the differentially expressed proteins (DEPs) between Li and CK groups. The SRWC was higher, while WR and WD were lower in Li group, compared with that in CK group. Based on 2-DE and ESI-MS, 22 DEPs were identified between Li and CK groups during drought stress. Among them, four proteins had the annotated information in relevant databases, including 1,4-benzoquinone reductase, PSCHI4, ribosomal protein L16 (RPL16) and AINTEGUMENTA-like (AIL) protein. Mycorrhizal P. massoniana inoculated with L. insulsus achieved an enhanced drought resistance as compared to the non-mycorrhizal, and the altered protein expressions such as 1,4-benzoquinone reductase, PSCHI4, RPL16, and AIL might contribute to the improved resistance under drought stress. 相似文献

0.5 to 2.5 mg dm−3 and 100 % rooting and optimal subsequent acclimatization was achieved on 1.0 mg dm−3 IBA. After 4 weeks of culture, the rooted shoots (>5 cm) were planted in pots containing peat, vermiculite and bark (2:1:1),
covered with plastic domes and maintained at 25 °C for 2 weeks before being transferred to a glasshouse. Plant survival was

The ecology and physiology of ectomycorrhizal (EcM) symbiosis with conifer trees are well documented. In comparison, however,
very little is known about the molecular regulation of these associations. In an earlier study, we identified three EcM-regulated
Pinus expressed sequence tags (EST), two of which were identified as homologous to the Medicago truncatula nodulin MtN21. The third EST was a homologue to the receptor-like kinase Clavata1. We have characterized the expression patterns of these genes and of auxin- and mycorrhiza-regulated genes after induction
with indole-3-butyric acid in Pinus sylvestris and in a time course experiment during ectomycorrhizal initiation with the co-inoculation of 2,3,5-triiodobenzoic acid, an
auxin transport inhibitor. Our results suggest that different P. sylvestris nodulin homologues are associated with diverse processes in the root. The results also suggest a potential role of the Clv1-like gene in lateral root initiation by the ectomycorrhizal fungus. 相似文献

A genetic transformation system has been developed for callus cells of Crataegus
aronia using Agrobacterium
tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with
5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different
types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red
colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Studying Pneumocystis has proven to be a challenge from the perspective of propagating a significant amount of the pathogen in a facile manner.
The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate
larvae Galleria

The maT clade of transposons is a group of transposable elements intermediate in sequence and predicted protein structure to mariner and Tc transposons, with a distribution thus far limited to a few invertebrate species. We present evidence, based on searches of publicly available databases, that the nematode Caenorhabditis briggsae has several maT-like transposons, which we have designated as CbmaT elements, dispersed throughout its genome. We also describe two additional transposon sequences that probably share their evolutionary history with the CbmaT transposons. One resembles a fold back variant of a CbmaT element, with long (380-bp) inverted terminal repeats (ITRs) that show a high degree (71%) of identity to CbmaT1. The other, which shares only the 26-bp ITR sequences with one of the CbmaT variants, is present in eight nearly identical copies, but does not have a transposase gene and may therefore be cross mobilised by a CbmaT transposase. Using PCR-based mobility assays, we show that CbmaT1 transposons are capable of excising from the C. briggsae genome. CbmaT1 excised approximately 500 times less frequently than Tcb1 in the reference strain AF16, but both CbmaT1 and Tcb1 excised at extremely high frequencies in the HK105 strain. The HK105 strain also exhibited a high frequency of spontaneous induction of unc-22 mutants, suggesting that it may be a mutator strain of C. briggsae. 相似文献

Summary An efficient and reproducible procedure for the large-scale propagation of Pseudoxytenanthera stocksii is described. High-frequency multiple shoot induction was achieved from nodal shoot segments collected from superior/elite
genotypes on Murashige and Skoog (MS) liquid medium supplemented with 1-naphthaleneacetic acid (NAA; 2.68 μM) and 6-benzylaminopurine (BA; 4.40 μM) at 28±1°C and 60 μmol m−2 s−1 light intensity under 12h photoperiod. In vitro-differentiated shoots were multiplied on MS liquid medium fortified with NAA (2.68 μM), BA (2.21 μM) and additives: ascorbic acid (283.93 μM), citric acid (118.10 μM), cysteine (104.04 μM), and glutamine (342.24 μM). Subculturing was carried out every 2wk on fresh shoot multiplication medium. About 125–150 shoots per culture flask were
harvested within 45–50d. In vitro-differentiated shoot clumps (three or four shoots) were successfully rooted on half-strength MS basal liquid medium with
indole-3-butyric acid (4.90 μM), BA (0.44 μM), and additives. This is the first report where in vitro- and in vivo-(through tillers) raised clonal plants were acclimatized and established in the field, where they exhibited normal growth. 相似文献

Bacterial biofilms are associated with chronic infections due to their resistance to antimicrobial agents. Staphylococcus
aureus is a versatile human pathogen and can form biofilms on human tissues and diverse medical devices. To identify novel biofilm
inhibitors of S. aureus, the supernatants from a library of 458 Actinomycetes strains were screened. The culture supernatants (1% v/v) of more than 10 Actinomycetes strains inhibited S. aureus biofilm formation by more than 80% without affecting the growth. The culture supernatants of these biofilm-reducing Actinomycetes strains contained a protease (equivalent to 0.1 μg proteinase K ml−1), which both inhibited S. aureus biofilm formation and detached pre-existing S. aureus biofilms. This study suggests that protease treatment could be a feasible tool to reduce and eradicate S. aureus biofilms. 相似文献
