This study explored the possibility of using biofeedback to arrest spontaneous declines in tonic electrodermal levels. For 3 days, 10 subjects (S) received 20 minutes of training in increasing skin potential level (SPL) negativity (arrested declines), followed by 20 minutes of training in decreasing SPL negativity (facilitating declines), with reverse order for half of the Ss. Simultaneous analogue and binary feedback of SPL were used. Training in the direction of SPL increases resulted in arrest of spontaneous declines in SPL negativity but did not result in large-magnitude increases above baseline. Training in the direction of SPL decreases facilitated declines in SPL. The absolute mean difference between the increase condition and the decrease condition on the 3rd day was 8.28 mV. Correlations suggested that considerable intraindividual variability may have been independently related to the Locus of Control personality dimension and to a Law of Initial Values limitation on SPL change. It was concluded that SPL was controllable to the extent of facilitating or arresting spontaneous declines.  相似文献

Two groups of rhesus (Macaca mulatta),two groups of cynomolgus (M. fascicularis),and one group of stumptailed monkeys (M. arctoides)were adapted to testing in the Wisconsin General Test Apparatus using a standardized procedure that produced an objective measure of the speed of adaptation (pretraining). Four of the groups were then given standard object-discrimination learning-set training. The fifth group, one of the groups of rhesus monkeys, was given delayed response training before the learning-set training and delayed matching-to-sample learning-set training after. Only performance on the delayed response task was correlated with speed of pretraining. The results suggest that differences in pretraining performance reflect primarily emotional and/or temperamental differences, whereas differences in learning-set performance reflect primarily differences in cognitive functioning.  相似文献

Background. The skin neurogenic inflammation is mainly related to Substance P (SP) and Calcitonin Gene-related Peptide (CGRP). There is no data on their availability in the dynamics of skin nerve endings, concerning their release and replenishment after a nociceptive stimulus, so this was investigated. Materials and methods. 25 rats were randomly distributed in 5 groups. The animals of the control group (CG) determined the baseline levels of neuropeptides in the skin. The groups S0 and S30 did not receive any cutaneous stimulus at 30 and 60 minutes, respectively. In the group S1, an “incision stimulus” was made at 30 minutes. In the group S31, a nociceptive stimulus was performed by subdermal scratching at 30 minutes and, at 60 minutes, the “incision stimulus” was carried out in the same location (“nociceptive hyperstimulation”). The skin samples of the other animals were harvested from the back 1 minute after their death. SP, pro-CGRP and CGRP were quantified by Western Blotting. Results. The “incision stimulus” released SP, S1 compared to S0 (p <0.05) detected in the first minute, and the replenishment time was more than 30 minutes. Also, it cleaved pro-CGRP, S1 compared to S31 (p <0.05) in the first minute, and its replenishment time less than 30 minutes. Release of CGRP was not detected. Conclusion. The incision released SP already detected in the first minute; its replenishment time is more than 30 minutes. The incision decreased pro-CGRP, also detected in the first minute; and its replenishment time is less than 30 minutes.  相似文献

Introduction

Physical fitness is an important prognostic factor in heart failure (HF). To improve fitness, different types of exercise have been explored, with recent focus on high-intensity interval training (HIT). We comprehensively compared effects of HIT versus continuous training (CT) in HF patients NYHA II-III on physical fitness, cardiovascular function and structure, and quality of life, and hypothesize that HIT leads to superior improvements compared to CT.

Methods

Twenty HF patients (male:female 19:1, 64±8 yrs, ejection fraction 38±6%) were allocated to 12-weeks of HIT (10*1-minute at 90% maximal workload—alternated by 2.5 minutes at 30% maximal workload) or CT (30 minutes at 60–75% of maximal workload). Before and after intervention, we examined physical fitness (incremental cycling test), cardiac function and structure (echocardiography), vascular function and structure (ultrasound) and quality of life (SF-36, Minnesota living with HF questionnaire (MLHFQ)).

Results

Training improved maximal workload, peak oxygen uptake (VO2peak) related to the predicted VO2peak, oxygen uptake at the anaerobic threshold, and maximal oxygen pulse (all P<0.05), whilst no differences were present between HIT and CT (N.S.). We found no major changes in resting cardiovascular function and structure. SF-36 physical function score improved after training (P<0.05), whilst SF-36 total score and MLHFQ did not change after training (N.S.).

Conclusion

Training induced significant improvements in parameters of physical fitness, although no evidence for superiority of HIT over CT was demonstrated. No major effect of training was found on cardiovascular structure and function or quality of life in HF patients NYHA II-III.

In the previous paper (Rejneket al., 1967) we described the fractionation of light chains (L) by Zn ions resulting in an accumulation of antigenic determinants of the heavy chain (H) in the Zn precipitate. Peptide maps of the obtained fractions of the L chains differ considerably from each other. Peptides of the L chains, the position of which corresponds within the experimental error to peptides of the H chain may be detected by comparing them with the peptide map of the H chains. The number of such peptides increases with qualitatively assayed accumulation of the component precipitated with anti-H serum during fractionation. The concentration of N-terminal glutamic acid, characteristic for the H chains increases at the same time.  相似文献

Systemic lupus erythematosus (SLE) is an autoimmune chronic inflammatory disease that presents several clinical manifestations, affecting multiple organs and systems. Immunological, environmental, hormonal and genetic factors may contribute to disease. Genes and proteins involved in metabolism and detoxification of xenobiotics are often used as susceptibility markers to diseases with environmental risk factors. Cytochrome P450 (CYP) enzymes activate the xenobiotic making it more reactive, while the Glutathione S-transferases (GST) enzymes conjugate the reduced glutathione with electrophilic compounds, facilitating the toxic products excretion. CYP and GST polymorphisms can alter the expression and catalytic activity of enzymes. This study aimed to investigate the role of genetic variants of CYP and GST in susceptibility and clinical expression of SLE, through the analysis of GSTM1 null, GSTT1 null, GSTP1*Ile105Val, CYP1A1*2C and CYP2E1*5B polymorphisms. 371 SLE patients from Hospital de Clínicas de Porto Alegre and 522 healthy blood donors from southern Brazil were evaluated. GSTP1 and CYP variants were genotyped using PCR–RFLP and GSTT1 and GSTM1 variants were analyzed by multiplex PCR. Among European-derived individuals, a lower frequency of GSTP1*Val heterozygous genotypes was found in SLE patients when compared to controls (p = 0.005). In African-derived SLE patients, the CYP2E1*5B allelic frequency was higher in relation to controls (p = 0.054). We did not observe any clinical implication of the CYP and GST polymorphisms in patients with SLE. Our data suggest a protective role of the GSTP1*Ile/Val heterozygous genotype against the SLE in European-derived and a possible influence of the CYP2E1*5B allele in SLE susceptibility among African-derived individuals.  相似文献

The cytogenetic effect of two radiolytic cytosine products, i.e. of isobarbituric acid and of dialuric acid has been studied on a system of resting meristem ofVicia faba L. on chromosomal level. Both compounds produced in a concentration 10-3 and 10-4 M chromosomal aberrations with a relatively low frequency, about 4 aberrations per 100 anaphases after 12 h of treatment. The present report is focused on the study of participation of exogenous DNA in the process of postirradiation reparation of meristematic cells ofVicia faba primary roots. It is aimed at comparison of the positive reparative effect of isologous DNA with postirradiation action of heterologous DNA in its native, thermally denatured and DNAase-degraded forms, or DNA degraded by ultrasound, and with the effect of other biologically important macromolecules (RNA, histone, heparin, and dextran sulphate). For this purpose, the roots ofVicia faba irradiated by 150 r exposure were cultivated in solutions containing the above substances for an appropriate time interval. In squash slides both mitotic activity of the investigated cell population and frequency of postmetaphase chromosomal aberrations induced by radiation were evaluated. It was shown that a stimulation of cell division and reparation of chromosome damages were supported exclusively by isologous DNA. On the contrary, exogenously applied heterologous DNA increased postirradiation frequency of aberrations; maintenance of native structure of applied DNA was an essential condition for the above effect. Other macromolecules investigated on the course of postirradiation reparation ofVicia faba meristematic cells were without effect.  相似文献

The thermophilic autotrophMethanobacterium thermoautotrophicum assimilates CO2 via a novel pathway rather than via the Calvin cycle. The central intermediate of this pathway is acetyl CoA which is reductively carboxylated to pyruvate. Cell extracts of the organism contained phosphoenolpyruvate synthetase with a specific activity of 100 nmol min-1 mg-1 protein (65°C). Pyruvate kinase and pyruvate, phosphate dikinase were not detected. Phosphoenolpyruvate synthetase was partially purified (50-fold) and the following reaction stoichiometry was established: $${\text{Pyruvate + ATP + H}}_{\text{2}} {\text{O }} \to {\text{ Phosphoenolpyruvate + AMP + P}}_{\text{i}} $$ The enzyme activity was depedent on free Mg2+ ions, NH 4 + or K+ ions, and SH-groups. Mn2+, but not Ca2+, could partially substitute for Mg2+; Na+ could not substitute for K+ or NH 4 + . The pH-optima,V max-values and the apparentK M-values for the substrates of the enzyme in both directions were determined. Thermodynamic, kinetic and regulatory features indicate that, in vivo, the enzyme functions in the direction of phosphoenolpyruvate synthesis from pyruvate. Not only is the synthesis of phosphoenolpyruvate via the PEP synthetase reaction energetically favorable; the enzyme also catalyzed this synthesis 100 times faster than the reverse reaction, the apparentK M value for pyruvate (40 μM) being low and the apparentK M value for phosphate (100 mM) being high. Furthermore, AMP, ADP, PP and α-ketoglutarate were inhibitors of PEP synthesis, indicating that the enzyme activity may be controlled in vivo. The role of phosphoenolpyruvate synthetase in autotrophic CO2 assimilation pathway ofMethanobacterium, as expected from previous labelling studies, is confirmed.  相似文献

Previously, we have reported the role of MAPKs (mitogen-activated protein kinases) under cadmium stress. This work continue to explore the relationship between MAPKs, H2O2, auxin signaling, and OsHMA and OsZIP gene expression in rice (Oryza sativa L.) roots under combined cadmium (Cd) and zinc (Zn) stress. Compared with Cd, Cd+Zn reduced Cd levels but increased Zn accumulation in the roots. Three OsMAPK genes were negatively regulated, while two OsHMA and two OsZIP genes were positively regulated by MAPK pathways under Cd+Zn stress. Transgenic rice expressing DR5-GUS exhibited enhanced GUS activity in H2O2-, PD (MAPKK inhibitor PD98059)-, or (Cd+Zn)-treated roots, which also exhibited increased H2O2 concentrations, whereas GUS staining decreased in roots in response to Cd+Zn+PD, DMTU (N,N′-dimethylthiourea, a H2O2 scavenger), or Cd+Zn+DMTU treatment, with reduced H2O2 levels. GUS levels were consistent with H2O2 levels, suggesting that MAPK pathway-mediated auxin redistribution occurs via H2O2, and H2O2 functions downstream of MAPK but upstream of auxin signaling pathways. Furthermore, MAPK pathways serve specific functions in regulating the expression of some key genes of auxin signaling (OsYUCCA, OsPIN, OsARF, and OsIAA) under Cd+Zn stress. Overall, MAPK cascades function in the integration of metal transport, H2O2 generation, and auxin signaling in rice seedlings grown under Cd+Zn stress.  相似文献

Many tropical regions lack models predicting the biological and environmental conditions expected in any given area, thus precluding the implementation of reference condition-based water policies. We focused on streams of French Guiana, and tested two predictions: geomorphology determines ecological sub-regions that have typical invertebrate communities, and diversity declines as anthropogenic pressure increases. Sixty-five stream sites were sampled for benthic invertebrates and physical–chemical variables across various watersheds. We used the Self-Organizing Map algorithm (neural network) to model relationships between invertebrate communities and environmental variables. Sites characterized by invertebrate communities clustered into two major subsets matching French Guiana’s hydro-ecoregions: the coastal alluvial plain characterized by recent sediment and low elevations, and the Guiana Shield characterized by an eroded rocky substrate and dense rainforests. Changes in community composition, and to a lesser extent taxonomic richness within each sub-region revealed ecological impacts of gold mining and logging, further clustering hydro-ecoregions into subsets of reference and impaired sites. Further analyses would, however, be needed to identify tipping points between natural and disturbed states, especially in remote headwater streams where gold mining had the harsher impact upon freshwater diversity, making upstream communities resembling the most downstream impacted ones.  相似文献

Numerous studies have investigated association of interleukin-13 (IL-13) G+2044A polymorphism with COPD susceptibility; however, the results were inconsistent and inconclusive. To evaluate the association between the IL-13 G+2044A polymorphism and susceptibility to COPD, a meta-analysis of published case–control studies was performed. Based on PubMed and Chinese database, this research selected studies that examined the association of the IL-13 G+2044A polymorphism with COPD. A genetic model-free approach was used to assess whether the combined data showed this association. Then a subgroup analysis was also performed, with stratifications for race, study design, and sample size. Six studies (total 1,213 COPD patients and 801 control subjects) for the IL-13 G+2044A polymorphism with COPD were included in the meta-analysis (G- vs A-allele: OR 1.12, 95 % CI 0.96–1.32, P = 0.15; genotypes GG+GA vs genotype AA: OR 0.99, 95 % CI 0.49–2.00, P = 0.98; genotype GG vs genotypes GA+AA: OR 1.18, 95 % CI 0.97–1.44, P = 0.09; genotype GA vs genotypes GG+AA: OR 0.85, 95 % CI 0.70–1.04, P = 0.11). This meta-analysis demonstrates that the IL-13 G+2044A polymorphism does not confer susceptibility to COPD. More detailed data about individual and environment, larger sample sizes with unbiased genotyping methods and matched controls in different populations are required.  相似文献

This study aimed to evaluate the biological activity of rat dorsal root ganglion cell (DRGC) and human vascular endothelial cell (HMVEC) in mono- and co-culture. Expression levels of vascular endothelial growth factor (VEGF) and nerve growth factor (NGF) mRNA were measured by quantitative real-time RT-PCR (qRT-PCR). Western blot analysis was used to identify VEGF and NGF protein expressions. Cell injury was assessed by measuring cell viability with methylthiazol tetrazolium (MTT) assay. The results showed that VEGF and NGF mRNA levels in the HMVEC+DRGC group were significantly higher than those in the DRGC and HMVEC groups (all p < 0.05). There were also greater increases in both VEGF and NGF protein expressions in the HMVEC+DRGC group than those in the DRGC and HMVEC groups (all p < 0.05). The results of MTT analysis revealed significant differences in cell viability among the HMVEC+DRGC group and the DRGC and HMVEC groups (all p < 0.05). In summary, our findings provide evidence that DRGC and HMVEC in co-culture may exhibit greater biological activity than DRGC in mono-culture.  相似文献

  1. Growth requirements of blue-green algae containing only the c-phycocyanin + chlorophyll a pigment system have been studied under blue light (380–540 nm) which approximates light conditions existing in subsurface waters in nature.

While a few species were capable of very slow photosynthetic growth on minimal medium with NO3 - as nitrogen source, most species were dependent on organic compounds for comparable growth under this condition. Some organisms did quite well with only Casamino Acids as a supplement, others did well with only glucose. One species, Agmenellum quadruplicatum strain PR-6, grew only when glucose and Casamino Acids were supplied simultaneously.
  3. Inhibitory effects of blue light on CO2 fixation and nitrogen metabolism are noted as possible explanations of these responses.

PprA, a pleiotropic protein involved in radioresistance of Deinococcus radiodurans was detected in multiprotein DNA processing complex identified from this bacterium. pprA mutant expressing GFP-PprA could restore its wild type resistance of γ radiation. Under normal conditions, GFP-PprA expressing cells showed PprA localization on both septum trapped nucleoids (STN) and nucleoids located elsewhere (MCN). Cell exposed to 4 kGy γ radiation showed nearly 2 h growth lag and during this growth arrest phase, the majority of the cells had GFP-PprA located on MCN. While in late phase (~120 min) PIR cells, when cells are nearly out of growth arrest, PprA was maximally found with STN. These cells when treated with nalidixic acid showed diffused localization of PprA across the septum. gyrA disruption mutant of D. radiodurans showed growth inhibition, which increased further in gyrA pprA mutant. Interestingly, gyrA mutant showed ~20-fold less resistance to γ radiation as compared to wild type, which did increase further in gyrA pprA mutant. These results suggested that PprA localization undergoes a dynamic change during PIR, and its localization on nucleoid near septum and functional interaction with gyrase A might suggest a mechanism that could explain PprA role in genome segregation possibly through topoisomerase II.  相似文献

Background

Integrating cardiopulmonary resuscitation (CPR) training in secondary schools will increase the number of potential CPR providers. However, currently too few certified instructors are available for this purpose. Training medical students and physical education student teachers to become CPR instructors could decrease this shortage.

Aim

Examine whether medical students and physical education student teachers can provide CPR training for secondary school pupils as well as (i.?e., non-inferior to) registered nurses.

Methods

A total of 144 secondary school pupils were randomly assigned to CPR training by a registered nurse (n = 12), a  medical student (n = 17) or a physical education student teacher (n = 15). CPR performance was assessed after training and after eight weeks in a simulated cardiac arrest scenario on a resuscitation manikin, using manikin software and video recordings.

Results

No significant differences were found between the groups on the overall Cardiff Test scores and the correctness of the CPR techniques during the post-training and retention test. All pupils showed sufficient CPR competence, even after eight weeks.

Conclusion

Training by medical students or physical education student teachers is non-inferior to training by a registered nurse, suggesting that school teachers, student teachers and medical students can be recruited for CPR training in secondary schools.

  1. The optomotor response of tethered flying houseflies (Musca domestica) has been studied at the level of the neural output which controls the activities of some non-fibrillar flight muscles (N-muscles).-a) During visually induced turning responses in a given direction some N-muscles on the right side of the thorax are synergistically active together with other N-muscles on the left side of the thorax. The same muscles are inactive during turning reactions in the opposite direction while the corresponding antagonists are now active (synopsis in Table 1).-b) The response activities of the N-mussles show a considerable variation during the course of time in spite of constant visual input.-c) There is a strong tendency for N-muscle spikes to be phase-locked with respect to the wingbeat period.-d) The findings obtained fromMusca are in accordance with the corresponding results obtained fromCalliphora (Heide, 1971b).
  2. TheN-muscle activities have also been investigated in tethered flying blowflies (Calliphora erythrocephala) which tried to yaw spontaneously with both wings beating. In spontaneous left (right) turn reactions the features of the observed neural output are nearly identical with the features of the motor output showing up during visually induced left (right) turn reactions.-A different motor output pattern has been found in flies with only one wing beating.
  3. The wingbeat synchronous rhythm observed in spike trains from activeN-muscles is produced in the thorax without the participation of higher stages of the fly's CNS. On the other hand no distinct rhythms can be found in spike trains fromN-muscles of non-flying flies when their motoneurons are artificially activated by non-rhythmic stimuli. Afferent information from thoracic sense organs seems to be essential for the production of the rhythm observed during flight.
  4. The results about the production of the wingbeat synchronous rhythm in spike trains fromN-muscles suggest that the information derived from the motion detectors only acts to gate the output needed to achieve yaw-turn reactions. The strength of the influence of signals from the motion detectors on the output producing system can be modified by the animals “state of excitement”.
  5. A model is presented which summarizes some features of information processing in the output systems supplying theN-muscles of flies. Available physiological data are discussed in relation to the model.
