Summary A physical map of safflower (Carthamus tinctorius L.) chloroplast DNA has been generated using SalI, PstI, KpnI and HindIII restriction endonucleases. The circular plastid genome (151 kbp) has the usual inverted repeat. Heterologous probes containing psbA, rbcL, atpA or rrnA structural genes mapped colinearly with spinach and other chloroplast genomes.  相似文献

Summary Nicotiana tabacum chloroplast DNA was digested with several restriction endonucleases chosen for their potential usefulness in distinguishing between species of the genus, Nicotiana. The resulting fragments were ordered into a circular configuration of about 160 kilobase pairs, equalling about 100x106 daltons. The physical map features an inverted, repeated unit of about 24 kilobase pairs separated by a unique sequence region with a mean size of 13 kilobase pairs on the short side. The cistrons coding for chloroplast ribosomal RNA are contained within the inverted repeat and have the arrangement: 16S, spacer, 23S, 4.5S/5S. Restriction endonuclease maps obtained with the enzymes PvuII, XhoI, and BglI are presented.  相似文献

The dynamics of a population of the stoloniferous Potentilla anserina was studied during four years. The mortality of established ramets followed a log-linear pattern superimposed on a seasonal fluctuation with an increased mortality during the period of vegetative spread. The major external mortality factors were unfavourable weather conditions such as drought and freezing. The turnover rate of the population was slow (31.6 yr). Vegetative spread dominated the recruitment to the population. Seedlings occurred but were exposed to a much higher mortality than vegetative daughter ramets. The juvenile period of seedlings is at least 4 yr in the field. This contrast to vegetative daughter ramets which are potentially flowering and stoloniferous the year after they are established, even if age affects both reproduction and vegetative spread. The density of most of the population is below the level where intraspecific regulative effects operate. However, in very dense stands both stolon and flower production were negatively density dependent.Nomenclature follows Lid (1974), Norsk og Svensk Flora, 2nd ed. Total tRNA of Chlamydomonas reinhardii was fractionated by 2-dimensional gel electrophoresis. Sixteen tRNAs specific for eleven amino acids could be identified by aminoacylation with Escherichia coli tRNA synthetases. Hybridization of these tRNAs with chloroplast restriction fragments allowed for the localization of the genes of tRNATyr, tRNAPro, tRNAPhe (2 genes), tRNAIle (2 genes) and tRNAHis (2 genes) on the chloroplast genome of C. reinhardii. The genes for tRNAAla (2 genes), tRNAAsn and tRNALeu were mapped by using individual chloroplast tRNAs from higher plants as probes.  相似文献

The closed circular DNA of pea chloroplast has been digested with restriction endonucleases SalI, SmaI, BamHI, XbaI, XhoI, HindIII, and EcoRI. A physical restriction map of pea ctDNA has been constructed by mapping the SalI and SmaI sites. The pea ctDNA has been found to contain one set of ribosomal RNA genes by Southern hybridization of restriction endonuclease digest, R-loop studies, and DNA-DNA heteroduplex mapping. The 23 S and 16 S RNA genes are confined to a DNA region of 3.0 and 1.5 kbp, respectively. The two rRNA chains are separated by a spacer region of 2.2 kbp.  相似文献

以Hoagland溶液培养的三角叶滨藜幼苗为材料,分别采用电导率法、原子吸收分光光度计法和压力室法测定了不同浓度盐胁迫下三角叶滨藜根的质膜透性、离子吸收和根系反射系数,分析其抗盐特点和机制.结果表明:随着盐胁迫强度的增加,三角叶滨藜根细胞质膜透性增大、根系反射系数减小;盐胁迫导致三角叶滨藜根系对K+的总吸收量减少、对Na+的总吸收量增多,但对Na+的相对吸收量减少、对K+的相对吸收量增加.盐胁迫条件下,三角叶滨藜根系对离子吸收有较强的调节能力;而根系反射系数的减小有利于根系用较小的负压力吸收水分,减小木质部空化的危险.说明三角叶滨藜具有较高的抗盐能力.  相似文献

Ribosomal RNA (5S) from Euglena gracilis chloroplasts was isolated by preparative electrophoresis, labeled in vitro with 125I, and hybridized to restriction nuclease fragments from chloroplast DNA or cloned chloroplast DNA segments. Euglena chloroplast 5S rRNA is encoded in the chloroplast genome. The coding region of 5S rRNA has been positioned within the 5.6 kilobase pair (kbp) repeat which also codes for 16S and 23S rRNA. There are three 5S rRNA genes on the 130-kbp genome. The order of RNAs within a single repeat is 16S-23S-5S. The organization and size of the Euglena chloroplast ribosomal repeat is very similar to the ribosomal RNA operons of Escherichia coli.  相似文献

Summary A complete physical map of the Codium fragile chloroplast genome was constructed and the locations of a number of chloroplast genes were determined. Several features of this circular genome are unusual. At 89 kb in size, it is the smallest chloroplast genome known. Unlike most chloroplast genomes it lacks any large repeat elements. The 8 kb spacer region between the 16 S and 23 S rRNA genes is the largest such spacer characterized to date in chloroplast DNA. This spacer region is also unusual in that it contains the rps12 gene or at least a portion thereof. Three regions polymorphic for size are present in the Codium chloroplast genome. The psbA and psbC genes map closely to one of these regions, another region is in the spacer between the 16 S and 23 S rRNA genes and the third is very close to or possibly within the 16 S rRNA gene. The gene order in the Codium genome bears no marked resemblance to either the consensus vascular plant order or to that of any green algal or bryophyte genome. Summary DNA was isolated and purified from chloroplasts of safflower (Carthamus tinctorius L.), digested with HindIII restriction endonuclease, and ligated into the HindIII site of the plasmid pUC9. Recombinant DNAs were isolated from ampicillin resistant white colonies which grew in the presence of the appropriate indicator, digested with HindIII, and then identified by comparison of agarose gel electrophoretic mobilities. HindIII digests of chloroplast DNA were used as a standard. Such recombinants were radiolabeled and hybridized with Southern blots of PstI, SalI, KpnI, and HindIII single and double digests of safflower chloroplast DNA. A physical map was subsequently generated showing the location of each recombinant on the circular plastid genome. Recombinants containing heterologous chloroplast gene markers from spinach or Euglena were also radiolabeled and mapped. The relative mapping positions of these genes are in good agreement with those which have previously been published for spinach and several other higher plants.  相似文献

Two hypervariable regions of the alfalfa (Medicago sativa L) chloroplast genome were used to describe levels of genetic relatedness among populations. PCR primers were developed to amplify the hypervariable regions. The frequency of occurrence of fragments of like size between populations was used to develop a measure of genetic relatedness. Relationships among 135 alfalfa accessions were investigated with principal component and cluster analyses, based on the genetic distance measures. Distinct clusters were taken as an indication of genetically distinct lineages. The populations investigated represented collections from world regions defined as the centers of origin of specific alfalfa germplasm sources, or else represented collections of introduced, and naturally adapted, accessions from agriculturally advanced regions. In general, this analysis indicated that the accessions from regions of origin of germplasm sources were largely homogeneous, while accessions from areas of introduction were much more diverse. In some cases, the accessions from a region of origin formed distinct clusters, suggesting that divergence has resulted in genetically distinct lines persisting in the original region of origin. Investigation of the stability of the marker fragments through vegetatively, and sexually, propagated plants indicated stable transmission through the sexual phase. However, one of the two regions underwent a deletion of 145 bp of one copy of a tandemly repeated 146 bp region in the equivalent of 80 years of vegetative growth. 以溶液培养的三角叶滨藜植株为材料研究了不同光照条件下其叶片光合作用对盐(NaCl)激胁迫的即刻反应及变化规律.结果表明,三角叶滨藜光合作用对盐激胁迫的响应有8 min左右的滞后期.在光照强度为100umol·m-2·-1和100 mmol·L-1浓度NaCl共同作用下,三角叶滨藜叶片净光合速率略有上升;但随NaCl浓度和光照强度进一步增加,其净光合速率呈下降趋势,且光照越强,盐胁迫导致的净光合速率下降幅度越大.同时,弱光下或强光低浓度NaCl胁迫下,盐激胁迫导致的净光合速率下降主要是气孔限制引起的;而强光下,高浓度的NaCl胁迫导致的净光合速率下降在盐激胁迫处理的前30-40 min主要由气孔限制引起.40 min后则主要由非气孔限制引起.可见,不同光照强度和NaCl浓度胁迫下三角叶滨藜叶片光合作用响应规律不同,引起净光合速率下降机制各异.  相似文献

Abstract The mitochondrial DNA was isolated from Aspergillus niger WU-2223L, a citric acid-production strain, and characterized by restriction-endonuclease mapping. Cloned fragments which covered the total range of the mitochondrial DNA were assembled and utilized to construct the restriction-endonuclease map for nine restriction enzymes. This map showed that the mitochondrial DNA was a circular molecule of 32.6 kb.  相似文献

Summary Medicago sativa L. cv Regen S is heteroplasmic for chloroplast DNA (cpDNA). Previous analyses of regenerated plants have shown a predominance of one of the cpDNAs which we have designated type A (the other we have designated type B). Studies of the replication of the two cpDNAs in tissue culture were carried out using leaflet expiants with defined cpDNA types and a distinguishing probe. The explants obtained showed a bias toward type A cpDNA during tissue culture. The data suggest that chloroplasts with different DNAs in a common nuclear background can multiply at different rates.  相似文献
