Indoor cultures of the green alga Scenedesmusacutus were tested for sensitivity to varying concentrations of seven insecticides and three fungicides. The growth rates of the organisms were considerably reduced by BHC and DDT analogues even at low concentrations of 500 ppb and 1 ppm. Lindane above 5 ppm and Technical BHC and DDT at 100 ppm were lethal to this alga. Of the three fungicides, TMTD (Tetra Methyl Thiurum Disulfide) was most toxic resulting in death of the culture at 10 and 100 ppm. Blitox (Copper-oxy chloride) and Zineb (Zinc Ethylene-Bis dithiocarbamate) considerably retarded growth at all concentrations. This alga was more sensitive to Lindane, BHC (Technical), DDT and TMTD. The growth rate appeared to be influenced by the concentration of pesticide present in the algal culture. As Scenedesmusacutus is being considered as a source of Single Cell Protein (SCP) for human consumption, knowledge on the sensitivity of this alga to pesticide contamination will be useful.  相似文献

The presence of an electron transport component with an EPR spectrum similar to that of a ferredoxin has been demonstrated in the blue-green alga Anabaenacylindrica, the green alga Euglenagracilis, and in chloroplasts from sorghum (Sorghumbicolour) and beans (Phaseolusvulgaris). The component is photoreduced at 77°K and is very similar to that previously reported in spinach. It seems likely that this component is a primary electron acceptor in photosynthesis in all of these organisms.  相似文献

D-Ribulose 1, 5-diphosphate carboxylase has been purified to a state of homogeneity from the marine blue-green alga Agmenellumquadruplicatum strain PR-6. The enzyme has been found to be easily separated from the bulk soluble protein by means of centrifugation into a sucrose gradient. RuDP carboxylase from Agmenellum, upon chromatography using a calibrated Sephadex G-200 column, exhibits a molecular weight of 456,000 daltons, considerably smaller than the protein from eucaryotic algae. Only one polypeptide of approximately 56,000 daltons was obtained upon dissociation in sodium dodecylsulfate.  相似文献

We report the isolation and some properties of a flavodoxin from a eukaryotic organism, the naturally occurring red alga Chondrus crispus. Unlike the situation with most other organisms the flavodoxin, under normal growth conditions, is the predominantly formed low-potential electron carrier, an accompanying ferredoxin occurring in only very small amounts. The flavodoxin is of molecular weight 21000 and one mole of FMN is present per mole of protein. Reduction of the flavoprotein proceeds via a blue flavosemiquinone radical. The flavodoxin is active both in photosynthetic NADP reduction by broken chloroplasts, and in phosphoroclastic cleavage of pyruvate by cell-free extracts of Clostridium pasteurianum.  相似文献

The heterotrophically grown, P-700-free mutant No. 8 of Scenedesmus obliquus is unable to carry out photosynthesis. Yet, chloroplast particles isolated from the alga reduced ferricyanide. They also reduced methyl viologen in the presence of the artificial donor reduced 2,6-dichlorophenol indophenol with a low yield but an appreciable saturation rate. NADP reduction or P-700 turn-over could not be detected.When grown mixotrophically, the mutant showed increasing P-700 activity with a concomitant increase in the rate of photosynthesis. Both activities were lost again when the algae were returned to darkness. Isolated chloroplast particles showed a good P-700 turn-over and reasonable rates of NADP reduction.The data suggest that the mutation occurred at a site preceding the formation of the pigment. The results on the photochemical activities are discussed in the light of reports concerning the involvement of P-700 in linear electron transport.  相似文献

Isolated heterocysts of the N2-fixing blue-green alga Anabaena cylindrica contain the Photosystem I components P-700, bound and soluble ferredoxins and ferredoxin-NADP reductase. They also show Photosystem I activity being able to photoreduce both methylviologen and NADP when ascorbate+dichlorophenol-indophenol acts as reductant. They photophosphorylate (64 μmol ATP produced/mg chlorophyll ah) and carry out oxidative phosphorylation (8.7 μmol ATP produced/mg chlorophyll ah). Ninety per cent of the total cell-free extract nitrogenase activity is located in the heterocyst fraction of aerobic cultures.  相似文献

Cytochalasin B (CB) (100 μg/ml) reversibly blocked cell division and cuased the formation of abnormal cytoplasmic bodies in the alga Cricosphaera carterae. Concentrations of 20 μg/ml and 40 μg/ml CB were without effects. In the presence of CB, calcified bodies (coccoliths) which form in Golgi vesicles and are normally extruded through the plasma membrane were not extruded and accumulated within the cell. CB appeared to alter the membranes of Golgi vesicles containing coccoliths. DMSO (10% vv), the solvent for CB, was without effect on cell division and coccolith extrusion. A concentration of 20% vv DMSO inhibited cell division irreversibly.  相似文献

A method to separate phycoerythrin 545, isolated from the cryptomonad alga, Rhodomonaslens, into two subunits has been developed. The method uses no denaturants (urea, guanidine, detergent) but relies on dissociation of the dimeric protein and subsequent aggregation of the β subunit at pH 3.0. The absorption spectra and amino acid composition of the subunits are presented. The spectra of the α subunit was red-shifted relative to β in both pH 3.0 buffer and in acidic 8.0 M urea.  相似文献

Using heat and urea treatment, the degradation products of denatured 25S cytoplasmic ribosomal RNA from jack bean (a higher plant) and Chlamydomonas reinhardi (an alga) were compared. Several specific modes of 25S RNA breakdown are postulated. The results suggest that (1) both higher plant algal 25S RNA may contain homologous endonuclease susceptible regions as judged by the stoichiometry of degradation and (2) that similar secondary structural properties of 25S cytoplasmic RNA have been conserved by evolution in the plant kingdom.  相似文献

Fractions enriched in either Photosystem I or Photosystem II activity have been isolated from the blue-green alga, Synechococcus cedrorum after digitonin treatment. Sedimentation of this homogenate on a 10–30% sucrose gradient yielded three green bands: the upper band was enriched in Photosystem II, the lowest band was enriched in Photosystem I, while the middle band contained both activities. Large quantities of both particles were isolated by zonal centrifugation, and the material was then further purified by chromatography on DEAE-cellulose.The resulting Photosystem II particles carried out light-induced electron transport from semicarbizide to ferricyanide of over 2000 μmol/mg Chlorophyll per h (which was sensitive to 3-(3,4-dichlorophenyl)-1,1-dimethylurea), and was nearly devoid of Photosystem I activity. This particle contains β-carotene, very little phycocyanin, has a chlorophyll absorption maximum at 675 nm, and a liquid N2 fluorescence maximum at 685 nm. The purest Photosystem II particles have a chlorophyll to cytochrome b-559 ratio of 50 : 1. The Photosystem I particle is highly enriched in P-700, with a chlorophyll to P-700 ratio of 40 : 1. The physical structure of the two Photosystem particles has also been studied by gel electrophoresis and electron microscopy. These results indicate that the size and protein composition of the two particles are distinctly different.  相似文献

The amino acid composition of stalk and cap cell wall polypeptides of the unicellular alga Polyphysa (A.) cliftonii has been investigated. In spite of chemical and physical differences between stalk and can cell wall polysaccharides, the amino acid composition of the cell wall polypeptides appeared qualitatively similar in both structures. however, quantitative differences have been observed. The results are discussed on the basis of a possible role of the polypeptides in the growth of the cell wall.  相似文献

Reelectrophoresis of the oligomer form (CP II1) of the chlorophyll ab light-harvesting complex (LHC) from the green alga Acetabularia yields two green bands which run at the position typical of the monomer (CP II). The upper green band (CP II1) is enriched in the 27 kDa polypeptide of the LHC, while the lower is enriched in the 26 kDa polypeptide. The fact that both bands have both chlorophyll (Chl) a and b, and in the same ratio, implies that the LHC is made up of two Chl ab proteins. Neither of these bands can be attributed to the Chl ab complex ‘CP 29’ (Camm, E.L. and Green, B.R. (1980) Plant Physiol. 66, 428–432). Resolution of CP II1 and CP II2 of spinach can be obtained if sucrose gradient fractions of an octylglucoside extract are subjected to SDS-polyacrylamide gel electrophoresis. CP II1 and CP II2 are interpreted as being fundamental subunits of the light-harvesting complex as it is defined on SDS-polyacrylamide gels.  相似文献

Eight chlorophyll-protein complexes were isolated from thylakoid membranes of a Codium species, a marine green alga, by mild SDS-polyacrylamide gel electrophoresis. CP 1a1, CP 1a2, CP 1a3 and CP 1a4 were partially dissociated Photosystem (PS) I complexes, which in addition to the core reaction centre complex, CP 1, possessed PS I light-harvesting complexes containing chlorophyll (Chl) a, Chl b and siphonaxanthin. LHCP1 and LHCP3 are orange-brown green chlorophyll ab-proteins (Chl aChl b ratios of 0.66) that contain siphonaxanthin and its esterified form, siphonein. CP a and CP 1, the core reaction centre complexes of PS II and PS I, respectively, had similar spectral properties to those isolated from other algae or higher plants. These P-680- or P-700-Chl a-proteins are universally distributed among algae and terrestrial plants; they appear to be highly conserved and have undergone little evolutionary adaptation. Siphonaxanthin and siphonein which are present in the Codium light-harvesting complexes of PS II and PS I are responsible for enhanced absorption in the green region (518 and 538 nm). Efficient energy transfer from both xanthophylls and Chl b to only Chl a in Codium light-harvesting complexes, which have identical fluorescence emission spectra at 77 K to those of the lutein-Chl ab-proteins (Chl aChl b ratios of 1.2) of most green algae and all higher plants, proved that the molecular arrangement of these light-harvesting pigments was maintained in the isolated Codium complexes. The siphonaxanthin-Chl ab-proteins allow enhanced absorption of blue-green and green light, the predominant light available in deep ocean waters or shaded subtidal marine habitats. Since there is a variable distribution of lutein, siphonaxanthin and siphonein in marine green algae and siphonaxanthin is found in very ancient algae, these novel siphonein-siphonaxanthin-Chl ab-proteins may be ancient light-harvesting complexes which were evolved in deep water algae.  相似文献

Alternate exposure of the nitrogen-fixing blue-green alga Anabaena L-31 to acetylene in air and ambient atmosphere resulted in substantial enhancement in the rate of acetylene reduction activity. The stimulation occurred even when protein synthesis was inhibited in the dark or in the presence of rifampicin and chloramphenicol. Residual nitrogenase of ammonium chloride treated cultures also showed the stimulation effect. The results indicate that the stimulation is due to a substrate (acetylene) mediated alteration of the enzyme molecule and not due to fresh synthesis.  相似文献

The fraction of the chloroplast DNA transcribed in the single celled alga Euglena has been determined by RNA-DNA hybridization. A vast excess of total cell RNA from cells which were rapidly dividing in the light was hybridized in liquid to [125I] — chloroplast DNA, and the resulting duplexes separated on hydroxyapatite columns. The contribution of DNA-DNA duplex formation was determined separately and was used to calculate that portion of the duplex which was actually a RNA-DNA hybrid. Sixteen percent of the single stranded chloroplast DNA forms a duplex with this RNA suggesting that 32 percent of the double stranded DNA molecule is being transcribed into RNA under these conditions of cell growth.  相似文献

An anaerobic incubation period of varying duration is required to induce hydrogenase activity in C., reinhardtii. Inclusion of sodium acetate, a metabolizable carbonaceous substrate, in the medium during anaerobic incubation accelerates the activation process. Thus, in the presence of sodium acetate, hydrogen photoproduction is detected within 7 to 15 minutes after the onset of anaerobiosis. On the contrary, if an uncoupler of phosphorylation, such as CCCP or sodium arsenate, is present during anaerobic incubation, little activation of the hydrogenase is observed even after hours of anaerobic adaptation. Since the uncouplers had no inhibitory effect on hydrogen photoproduction by the alga when added to previously activated cells, they are not inhibitors of activated hydrogenase. The uncouplers interfere, most likely, with the activation of hydrogenase. Similar effects of uncouplers on the hydrogenase activation process were obtained using a cell-free assay of hydrogenase activity. These observations provide strong evidence that anaerobic activation of the hydrogenase is an energy requiring process.  相似文献

(1) Five minor chlorophyll-protein complexes were isolated from thylakoid membranes of the green alga Acetabularia by SDS-polyacrylamide gel electrophoresis, after SDS or octylglucoside solubilization. None of them were related to CP I (Photosystem I reaction center core) or CP II (chlorophyll ab light-harvesting complex). (2) Two complexes (CPa-1 and CPa-2) contained only chlorophyll (Chl) a, with absorption maxima of 673 and 671 nm, and fluorescence emission maxima of 683 nm compared to 676 nm for CP II. The complexes had apparent molecular masses of 43–47 and 38–40 kDa, and contained a single polypeptide of 41 and 37 kDa, respectively. They each account for about 3% of the total chlorophyll. (3) Three complexes had identical spectra, with Chl ab ratios of 3–4 compared to 2 for thylakoid membranes, and a pronounced shoulder around 485 nm indicating enrichment in carotenoids. One of them was the complex ‘CP 29’ (Camm, E.L. and Green, B.R. (1980) Plant Physiol. 66, 428–432) and the other two were slightly different oligomeric forms of CP 29. They could be formed from CP 29 during reelectrophoresis; but about half the complex was isolated originally in an oligomeric form. Together they account for at least 7% of the total chlorophyll. The metabolism of drobuline has been examined in the dog, rabbit, rat, guinea pig and hamster. In the dog, unlike the other species, glucuronide conjugation is the major route of metabolism. The structure of the conjugate has been established as an O-glucuronide by isolation using HPLC following by field desorption mass spectral analysis. When the separate d- and l-isomers of drobuline were administered to a series of dogs the l-isomer reached plasma levels approximately three time higher than those of the d-isomer. Deuterium labeled drobuline was synthesized and resolved by multiple crystallizations of the malate salts. Racemic mixtures containing d6-d and h6-l drobuline and d6-l and h6-d drobuline were prepared and analyzed by GC-MS as the pentafluoropropionate derivatives. When either racemic mixture was administered to dogs (10 mg/kg, p.o.) the plasma levels of the l-isomer were found to be approximately three times those of the d-isomer. Using these deuterium labeled mixtures the disposition of the two isomers has been examined in the isolated perfused dog liver, in hepatocytes and isolated microsomes. The results indicate that the difference in plasma levels of the d- and l-isomers is not dependent upon stereospecific absorption or excretion but rather it is caused by metabolism of the d-isomer at a faster rate than that of the l-isomer.  相似文献

The replication defective transducing phage λplac5O29P3 carries a portion of the E.colilac operon in the b2 region of the lambda phage. This lac operon segment contains the lac promoter, the lac operator, and the β-galactosidase z gene, but does not contain the lac repressor i gene. The z gene can be expressed from both the inserted lac promoter and the phage promoter. When E.coli strain 594 (z?, i+) or JC6256 (Δlac) is infected by λplac5O29P3 in the absence of additional cyclic AMP, β-galactosidase synthesis is shown to be expressed from the phage promoter. When 594 (λ+) or JC6256 (λ+) is infected by λplac5O29P3 in the presence of additional cyclic AMP and IPTG, β-galactosidase synthesis is shown to be expressed from the inserted lac promoter.The ability to separate the phage promoter from the inserted lac promoter for β-galactosidase expression will simplify the interpretation whenever λplac5 is used.  相似文献
