Seedlings of Meloidogyne incognita-resistant (N.C. 95) and -susceptible (McNair 30) tobacco cultivars were grown aseptically for 55 days inside isolator chambers in autoclaved soil infested with 0 or 3,000 axenized eggs of M. im ognita per 500 cc of soil. Healthy and infected plants were compared. Dry root weights of infected plants of resistant and susceptible cultivars were 16% and 84%, respectively, less than the controls. Sixteen amino acids, including those precursors for nicotine, and nicotine, increased significantly in infected roots of both cultivars. Increases in amino acids in infected roots ranged from 28% for valine to 103% for tyrosine in the resistant N.C. 95, and from 30% for leucine to 148% for tyrosine in lhe susceptible McNair 30. Nicotine content (dry weight basis) increased 42% and 62% in infected roots of resistant and susceptible cultivars, respectively. Nematode infection increased nicotine by 112% in leaves of N.C. 95, and decreased it by 56% in leaves of McNair 30. Root damage by M. incognita probably decreased nicotine movement into leaves of McNair 30. In N.C. 95, nicotine movement into leaves apparently was not adversel b affected due to lack of significant root damage.  相似文献

A GUS-transformed strain of B. sorokiniana was used to study the relationship between fungal growth, lesion development and natural root cortical cell death (RCD) in roots of different cereals. Roots of 10-day-old seedlings, grown on filter paper, were inoculated with the fungus and at different time intervals lesion size and GUS-activity in the roots were determined. A significant, positive correlation was found between GUS-activity and ergosterol content in barley roots infected with this transformed strain and these results indicate that GUS can be used as a marker to study fungal growth in plant tissue. The fungus had a slower growth rate in resistant barley varieties, i.e., those producing smaller lesions, than in more susceptible varieties.
In wheat and triticale, the fungal growth was faster than in barley, despite the smaller and lighter coloured lesions in these species. This may be explained by the fact that wheat and triticale have a faster rate of root cortical senescence than barley. Presumably, dying cortex cells cannot respond to the fungal invasion by producing phenolic compounds that cause browning of the tissue. Among seven investigated cereal species, there was a positive correlation between the degree of RCD and fungal biomass increase in roots after inoculation.  相似文献

The morphological and physiological responses of barley to moderate Pi deficiency and the ability of barley to grow on phytate were investigated. Barley cultivars (Hordeum vulgare L., Promyk, Skald and Stratus) were grown for 1–3 weeks on different nutrient media with contrasting phosphorus source: KH2PO4 (control), phytic acid (PA) and without phosphate (−P). The growth on −P medium strongly decreased Pi concentration in the tissues; culture on PA medium generally had no effect on Pi level. Decreased content of Pi reduced shoot and root mass but root elongation was not affected; Pi deficit had slightly greater impact on growth of barley cv. Promyk than other varieties. Barley varieties cultured on PA medium showed similar growth to control. Extracellular acid phosphatase activities (APases) in −P roots were similar to control, but in PA plants were lower. Histochemical visualization indicated for high APases activity mainly in the vascular tissues of roots and in rhizodermis. Pi deficiency increased internal APase activities mainly in shoot of barley cv. Stratus and roots of cv Promyk; growth on PA medium had no effect or decreased APase activity. Protein extracts from roots and shoots were run on native discontinuous PAGE to determine which isoforms may be affected by Pi deficiency or growth on PA medium; two of four isoforms in roots were strongly induced by conditions of Pi deficit, especially in barley cv. Promyk. In conclusion, barley cultivars grew equally well both on medium with Pi and where the Pi was replaced with phytate and only slightly differed in terms of acclimation to moderate deficiency of phosphate; they generally used similar pools of acid phosphatases to acquire Pi from external or internal sources.  相似文献

Charcoal rot, caused by Macrophomina phaseolina, is the most common yield reducing constraint in sorghum. In the present study, eight cultivars of Sorghum bicolor were screened against M. phaseolina to determine its effects on several growth parameters as well as the content of phenols, salicylic acid (SA), total protein and peroxidase activity and were examined for their relationship, with disease resistance. Out of the eight cultivars tested, PJ-1430 was the most resistant and SU-1080, the most susceptible. The roots of plants were more affected by the pathogen than the shoots. The cultivar PJ-1430 produced a higher plant root and shoot dry mass and was associated with higher polyphenols, SA, peroxidases and thus sustained less oxidative damage whereas SU-1080 experienced maximum oxidative damage, so was considered susceptible to charcoal rot. A positive relationship was observed among various biochemical parameters and disease resistance of cultivars PJ-1430 and SU-1080.  相似文献

Summary Significant differences in total dry matter yields of shoots and roots were found between 11 ryegrass (Lolium) cultivars grown in a glasshouse. Although shoot yield varied significantly between individual cultivars there was no overall difference between the annual and perennial cultivars; whereas for roots, the yields of the perennial plants were much smaller than those of the annual types. Water use (g H2O g total DM–1) also varied significantly between cultivars. However, there was no relationship between efficient water use and dry matter production.No significant differences were found in shoot composition between the cultviars for nitrogen, phosphorus, and potassium; however, concentrations of sulphur, magnesium, calcium, and sodium varied significantly. Sodium concentrations were generally higher in the annual compared to the perennial cultivars. For roots only nitrogen, phosphorus, and sulphur differed significantly between cultivars. Of the elements only calcium in the shoots was shown to be related to water use. Thus cultivars which were low users of water also had significantly lower calcium concentrations in their shoots. Water use appeared to affect the absorption of calcium by the root to a far greater extent than the transport from roots to shoot. An apparent relationship between magnesium concentration in the shoots and water use was shown to be due to the close association of magnesium with calcium in the plant.  相似文献

Ethylene production was determined in excised tomato (Lycopersicon esculentum) root cultures of Meloidogyne javanica susceptible and resistant cultivars infected with M. javanica. Uninfected cultivars produced very low amounts of ethylene. Relatively high amounts of ethylene were produced by the infected susceptible cultivars. Peak production of 1.6 n moles * g root⁻¹ * h¹⁻ occurred between 9 and 16 days after inoculation (DAI). The period of high ethylene production coincided with that of rapid increase in gall weight. Low amounts of ethylene were also released by the infected resistant cultivar between 9 and 12 DAI, which follows the hypersensitivity reaction. Ethylene production in infected intact plants during the period of rapid gall growth was twice as much as in uninfected plants during the same time. Exposing excised root cultures to 0.5 or l0 ppm ethylene accelerated the rate of increase in gall weight of M. javanica infected roots. In contrast, overall root growth was inhibited by these treatments, compared to infected roots which were not exposed to ethylene.  相似文献

The development of cereal cyst nematode (CCN; Heterodera avenae ) induced syncytia in the host roots of infected resistant bread wheat ( Triticum aestivum cv. AUS10894), diploid wheat ( Aegilops tauschii ), barley ( Hordeum vulgare cv. Chebec and cv. Galleon) and in the susceptible wheat cv. Meering and barley cv. Clipper were studied over a period of 13 d. The resistance to CCN in these cereal plants is conferred by the resistance genes Cre1 in the wheat cv. AUS10894, Cre3 in A. tauschii , Ha2 in barley cv. Chebec and Ha4 in barley cv. Galleon. Anatomical observations were made on the development of the syncytia in CCN-infected wheat and barley roots, which carry each of these four sources of resistance genes. Accelerated development of the syncytia in resistant plants, especially in the barley cultivars, was observed. The sites of syncytia development in susceptible wheat and barley were also closely associated with the vascular tissues in the stele, but less so in the resistant plants. The syncytia in the infected susceptible wheat and barley were also metabolically active at day 13. By contrast, the syncytia of resistant wheat plants carrying the Cre1 or Cre3 genes remained extensively vacuolated and less metabolically active. In barley plants with the Ha2 or Ha4 genes, the syncytia appeared non-functional and in early stages of degeneration by day 13 after inoculation.  相似文献

Drought stress responses and sensitivity of dioecious plants, such as Populus cathayana Rehd., are determined by different mechanisms in each sex. In general, males tend to be more resistant while females are more sensitive. Here, we used reciprocal grafting between males and females to determine the relative importance of roots and shoots when plants are exposed to drought stress. Total dry matter accumulation (DMA), photosynthetic capacity, long‐term water‐use efficiency (Δ), water potential and ultrastructure of mesophyll cells were evaluated to determine the different roles of root and shoot in sex‐related drought responses. Plants with male roots were found to be more resistant and less sensitive to water stress than those with female roots under drought conditions. On the contrary, plants with female shoots grew better than those with male shoots under well‐watered conditions. These results indicated that the sensitivity of males and females to water stress is primarily influenced by root processes, while under well‐watered conditions sexual differences in growth are primarily driven by shoot processes. Furthermore, grafting female shoot scion onto male rootstock was proved to be an effective mean to improve resistance to water stress in P. cathayana females.  相似文献

Effects of different soil moisture (soil drought and waterlogging) and soil compaction (1.33 and 1.50 g·cm−3) on the growth and morphological traits of the root system were studied in four breeding forms and seven cultivars of triticale. Morphological changes, including the restriction of root extension, expansion and proliferation of laterals roots, occur in plants grown in different soil moisture and in compact soil. The investigations comprised quantitative and qualitative analyses of a developed plant root system through determining the number, length and dry matter of the particular components of the root system. Obtained results have demonstrated a relatively broad variation in the habit of the triticale root system. Plants grown under compact soil and low or high soil water content showed a smaller number and less dry matter of lateral branching than plants grown in control conditions. The harmful effects of compact soil and drought conditions on the growth of roots was greater when compared with that of plants exposed to waterlogging. The observed effects of all treatments were more distinct in a drought sensitive strains. The drought resistant forms were a more characterize with extensive rooting and by smaller alterations in the root morphology under the stress conditions compared with drought sensitive one. Results confirm that the breeding forms (CHD-12 and CHD-173) of a high drought susceptibility was found to be also more sensitive to periodical soil water excess. A more efficient water use and a lower shoot to root (S/R) ratio were found to be major reasons for a higher stress resistance of the breeding forms (CHD-220 and CHD-247). The reasons for a different response of the examined breeding forms and cultivars to the conditions of drought or waterlogging may be a more economical water balance and more favourable relations between the shoot and root dimensions in the drought resistant forms and cultivars. The results suggest that the morphological traits of the triticale root system may be used in practice as direct or indirect selection criteria in maize breeding.  相似文献

Environmental stresses (soil compaction, drought, waterlogging) cause changes in plants’ root system structure, also affecting the growth of above-ground parts. The aim of this study was to estimate phenotypic variation among maize and triticale genotypes in root penetration ability through petrolatum-wax-layer (RPA). Also, the effect of shortage or excess of soil water on dry matter of shoots and roots and morphological changes in root system structure in sensitive and resistant maize and triticale genotypes grown in low or high soil compaction level was evaluated. To estimate RPA index, the petrolatum-wax-layer method (PWL) was used. The strength of three petrolatum-wax concentrations 60, 50 and 40 % was 0.52, 1.07 and 1.58 MPa, respectively. High coefficients of variation (CV) were observed in 0.52 and 1.07 MPa and for maize were 19.2 and 21.7 %, and for triticale, 12.5 and 18.3 %, respectively. The data indicate that the use of PWL technique is an effective screening method, and makes it possible to divide the genotypes into resistant and sensitive groups. The second part of this study investigated a multistress effect of soil compaction combined with drought or waterlogging on root and shoot growth and morphological changes in root system structure of maize and triticale genotypes differing in susceptibility to environmental stresses. Seedlings were grown for 4 weeks in root-boxes under conditions of low (LSC 1.1 g cm?3) or severe (SSC 1.6 g cm?3) soil compaction. Drought or waterlogging stresses were applied for 2 weeks from 14th to 28th day. In comparison to LSC treatment, in SSC treatment the decrease in dry matter of shoots and roots was greater for sensitive genotypes of maize and triticale (Ancora, CHD-147). Soil drought or waterlogging caused greater decrease of dry matter of shoots and roots in seedlings grown in SSC in comparison to LSC. The root penetration index (RPI) was estimated as a ratio of root dry matter in 15–40 cm root-box layer to total root dry matter. On the basis of RPI it was possible to group the genotypes according to their ability to distribute roots in soil profile. In comparison to LSC, SSC exerted a strong influence on the length of seminal and seminal adventitious roots, as well as the number and length of L- and S-type lateral roots developed on seminal and nodal roots. In both species the restriction effect of soil compaction on number and length of roots was more severe in sensitive (Ankora, CHD-147) than in resistant (Tina, CHD-247) genotypes. The restriction in roots propagation was greater in triticale than in maize. Exposure to drought or waterlogging in the case of genotypes grown in LSC and SSC treatments caused a decrease in number and length of particular components of root system structure. In both species the decrease of root number and length in plants grown under waterlogging was greater than under drought. The observed changes in root system were greater in sensitive (Ankora, CHD147) than in resistant (Tina, CHD-247) genotypes. Statistically significant correlations were found between RPA and RPI and also between these indexes and soil compaction, drought and waterlogging susceptibility indexes. This indicates that genotypes resistant to soil compaction were resistant to drought or waterlogging and also that genotypes resistant to drought were resistant to waterlogging.  相似文献

Changes in root- and leaf-soluble proteins were investigated in tomato after invasion by the root-knot nematode Meloidogyne javanica, or in barley and wheat after invasion by the cereal cyst nematode Heterodera avenae. Infection of susceptible tomato plants by M. javanica did not cause any change in the soluble-protein composition of leaves or roots compared with uninoculated plants at an early infection stage. No pathogenesis-related proteins (chitinase, glucanase, or P-14) were induced in the leaf apoplast. Changes in leaf proteins were not observed after invasion of wheat cultivars by H. avenae, whereas, in barley, a few changes in intercellular leaf proteins were recorded in resistant cultivars. These changes, however, were not the same among different H. avenae-resistant cultivars. Protein changes were found at an early stage of infection in barley and wheat roots infected with H. avenae, but no difference was found between resistant and susceptible cultivars.  相似文献

Plantago major L. ssp. major , a grassland species from a relatively nutrient-rich habitat, was grown in nutrient-rich and nutrient-poor culture solutions. Half of the plants were transferred from high to low or from low to high nutrient conditions. The rate of dry matter accumulation in both shoots and roots decreased slowly upon transfer of plants to low nutrient conditions and the shoot to root ratio was unaffected. The rate of structural growth of both roots and shoots increased upon transfer from low to high nutrient conditions and the shoot to root ratio, if calculated from non-structural-carbohydrate-free dry weights, increased.
Photosynthesis was largely independent of the nutrient supply. Root respiration, particularly the activity of the alternative oxidative pathway, decreased with increasing age. This decrease was ascribed to a decreased shoot to root ratio, which reduced the relative amount of carbohydrates translocated to the roots and thus the amount available for the alternative pathway. It is calculated that in young as well as in old plants grown in full nutrient solution 48% of the daily produced photosynthates was translocated to the roots.
This is at variance with data on P. lanceolata , where a decreasing proportion of the daily produced photosynthates was translocated to the roots when the plants grew older. It is concluded that shoot growth plus shoot respiration consumed a constant amount of the daily produced photosynthates in P. major and that the rest was left for translocation. It is further calculated that in P. major plants grown in full nutrient solution c . 25% and c . 2% of the daily produced photosynthates in young and old plants, respectively, was respired in a way that is not involved in production of energy that is utilized in growth and maintenance ('inefficient root respiration').
The results are discussed in comparison with those of P. lanceolata , a species from a relatively nutrient-poor habitat.  相似文献

The concentration of dry matter and the content of esterified fatty acids in total lipids of roots and etiolated shoots of 3- to 10-day-old seedlings of wheat (Triticum aestivum L.) infected with Bipolaris sorokiniana (Sacc.) Schoemaker, the agent of helminthosporic root rot, were determined in the course of germination. At the onset of germination, fungal infection caused a considerable increase in the dry matter concentration in both roots and shoots due to the enhanced mobilization of seed reserves. However, after the 7th day of germination, dry matter concentration fell below the level of noninfected control seedlings as a result of infection. The content of total lipids rose immediately after infection and always exceeded the control index up to the end of germination, in spite of a continuous decrease in this index in both control and infected seedling. It is concluded that an increase in the content of cellular lipids is a characteristic response of both shoots and roots to the root rot infection of wheat seedlings.  相似文献

Soybean cultivars varied in their response and tolerance to low initial Rotylenchulus reniforrnis populations of 10,000 nematodes/3.8 liters of soil, but a high initial population of 25,000 consistently reduced yields on resistant and susceptible cultivars by an average of 33.1%. At the 10,000 nematode inoculum level, dry seed yields of Hood decreased while those of Pickett increased significantly. Generally, total phosphorus decreased 11.1 and 11.5% and potassium increased 5.9 and 4.5% in seeds harvested from plants receiving initial inoculum levels of 5000 and 10,000 nematodes/pot, respectively. Little change in the total nitrogen content in seed was noted. Leucine content of seeds from infected plants was slightly less than from noninfected plants.  相似文献

The mechanism of resistance toB toxicity in barley and wheat was studied in a solution culture experiment using several cultivars displaying a large range of sensitivity to excessB supply. Plants were cultured for 35 d atB concentrations ranging from normal to excessive (15 to 5000 M, respectively) then examined for dry matter production and theB distribution between roots and shoots.In both species, increasedB supply was accompanied by increased tissueB concentrations, development ofB toxicity symptoms and depressed growth. At each level ofB supply, however, resistant cultivars accumulated considerably lessB than did sensitive cultivars, in both roots and shoots. Even at the lowestB supply, at which noB toxicity symptoms developed and growth was not affected, resistant cultivars maintained relatively low tissueB concentrations. No cultivar displayed an ability to tolerate high tissueB concentrations.These results indicate that sensitivity toB toxicity in barley and wheat is governed by the ability of cultivars to excludeB. If theB concentrations of tissues is used to indicate resistance toB toxicity, then cultivars have the same ranking whether cultured at a normal or excessB supply.  相似文献

Vanadium bioaccumulation calculated as the ratio of its content in plant biomass to that in the substrate (Vbi-index) was studied in pea. Vbi was on average 11.275, 11.770 and 13.153 in the roots, and 0.809, 0.467 and 0.749 in the shoots of the cultivars Opal, Laser and Ramir, respectively. This indicates cultivar differences in vanadium uptake, and low translocation rates from roots to shoots. Vanadium (3 to 30 mg 1-1) decreased shoot and root fresh and dry masses of the three cultivars. Seedlings of the cv. Opal were the most susceptible to higher concentrations of vanadium (20 to 30 mg 1-1), whereas seedlings of cv. Laser were the most resistant.  相似文献

Abstract: We tested the hypothesis that cultivar selection during the process of domestication in cereal plants led to a change in dry mass allocation, e.g., less root mass and more leaf mass or more leaf area per unit leaf mass. We divided 24 varieties of diploid, tetraploid and hexaploid winter wheat and two-rowed winter barley into three categories of domestication levels (wild species, old landraces and modern cultivars) and compared the patterns of dry matter fractionation at the time of anthesis under standardized outdoor growth conditions. In both cereals, total biomass per individual increased significantly with domestication level but, to our surprise, we found no significant change in dry matter investment between domestication levels: neither the dry mass fraction of leaves increased, nor was there a trend of reduced investment in stems and roots, contrary to what we expected. Specific leaf area (SLA) and leaf area ratio (LAR) of modern wheat and barley cultivars were significantly lower compared to wild varieties. Major differences in both cereals were of a purely morphological nature, namely a decrease in the number of stems and ears from wild species to domesticated varieties, along with more synchronous tiller development and therefore similar tiller size. Fertilizer increased total biomass in all domestication levels in both cereals, but influenced the dry matter fractionation only in barley. Tissue nitrogen concentration was unresponsive to both domestication and fertilization. The expected shift in functional traits, conventionally considered to determine plant growth, was not found. Indeed, dry matter fractionation among the major plant biomass components seems to be very conservative.  相似文献

Potato Responds to Salt Stress by Increased Activity of Antioxidant Enzymes   总被引:1,自引:0,他引:1  
To understand the response of potato to salt stress, antioxidant enzyme activities and ion content were analyzed for a sensitive and a tolerant cultivar. Nodal cuttings of the tolerant cultivar, Kennebec, and the sensitive cultivar, Concord, were exposed to media without or with 30, 60, 90 or 120 mmol/L NaCl for 4 weeks. On exposure to NaCl, the length and fresh and dry weight of both shoots and roots of Concord showed greater decrease than those of Kennebec. The decrease in shoot growth was more severe than that of the root for both cultivars. The K+ content of shoots and roots of both cultivars was reduced in a dose-dependent manner by exposure to NaCl; the Na+ content Increased. Activities of ascorbate peroxidase, catalase and glutathione reductase were increased in NaCl-exposed shoots of Kennebec; the corresponding activities inNaCl-exposed shoots of Concord were decreased. Roots of both cultivars showed similar changes in the activities of these enzymes on exposure to NaCl. These studies established that enzyme activities In Concord shoots are inversely related to the NaCl concentration, whereas those in Kennebec do not show a dose dependency, which is also the case for the roots of both cultivars. Our findings suggest that an Increase in activity of antioxidant enzymes, such as ascorbate peroxidase,cetalase and glutathione reductase, can contribute to salt tolerance in Kennebec, a salt resistant cultivar of potato.  相似文献

Continuous measurements of CO2-evolution and dry matter accumulation were carried out on shoots and roots separately of intact Helianthus annuus L. cv. Autumn Beauty plants grown in nutrient solution at different root temperatures. The data were used to distinguish between growth and maintenance components of respiration. The maintenance and growth coefficients were higher in the root system than in the shoots. The overall efficiency of assimilate utilization was within the range reported in the literature. An increase in root temperature increased the maintenance part of root respiration and, to a lesser degree, also shoot maintenance respiration. Neither root nor shoot growth respiration coefficients were affected by root temperature. It is concluded that the study of whole-plant respiration masks differences in energy utilization between shoots and roots.  相似文献

The histo- and cytopathological effects in resistant (ILC-195) and susceptible (Canitez-87) chickpea cultivars were examined by light, transmission and scanning electron microscopy 3, 5 and 7 days after inoculation (d.a.i) of seedlings with Ascochyta rabiei. The fungus produced typical appressoria that penetrated both cuticle and stomata. The resistant plants had physical barriers and a cuticle layer against fungal penetration 3 d.a.i. The fungus spread intercellularly and subepidermally in the leaves and stems of susceptible plants 3 d.a.i., and was followed 5 d.a.i. by cell plasmolysis, degeneration of organelles and of cellulose, but not lignified, walls. Pycnidia formation occurred between 5 and 7 d.a.i. 7 d.a.i., organelle degeneration, pycnidia formation and symptom severity increased. Tracheidal elements, including lignified elements, were almost intact in both resistant and susceptible cultivars. In the susceptible plants, lignin cell walls were slightly degraded after 7 days. There was less cell degeneration and pycnidia formation in resistant plants. Some electron-dense large bodies and lipid granules were observed within intracellular fungal hyphae in infected cells of resistant plants 7 d.a.i.  相似文献
