A series of experiments was conducted to study the decline in reproduction associated with age in the male Japanese quail. In Experiment 1, eggs were collected from pairs that were 28, 56, 107, and 149 weeks of age. Pairs that were 56 weeks of age or older showed a sharp drop in fertility and hatchability. Subsequent experiments were designed to study the endocrine and behavioral basis for this decline in the male. In Experiment 2, males that were between 23 and 70 weeks of age were tested for mating behavior, plasma testosterone was measured, and testes wet weight was determined. There were no significant differences between the age groups. However, samples taken from an additional group of males 168 weeks of age showed significantly (P < 0.05) lower serum testosterone and testes weight. Therefore, the third experiment was designed to include age groups of males between 4 and 126 weeks of age in order to further examine this decline in males older than 70 weeks of age. There was a significant (P < 0.05) drop in this number of males showing sexual behavior and of those males showing sexual behavior; the quantity of mating activity also declined. Plasma testosterone was reduced in the older groups; however, the change was not significant. In addition, the size of the cloacal gland, an androgen-dependent organ, declined with age (P < 0.05). These results argue that the age-related reproductive decline may have behavioral as well as endocrine basis. Possibly, the behavioral changes may result from altered receptor sensitivity at the level of areas of the brain, which control reproductive behavior, and at the level of the testes, which affect hormone production and spermatogenesis.  相似文献

In aging quail, an increasing proportion of males show no sexual behavior. A decrease in the mean size of the tests, cloacal gland, and sternotracheal muscles is also observed. In both sexually active and inactive males, plasma testosterone decreases with age but more so in inactive birds. The behavioral and morphological changes observed during aging are correlated with shifts in the intracellular testosterone metabolism resulting in a change in the ratio of active versus inactive metabolites. In the hypothalamus there is a steady decrease with age of 5 beta-reductase activity in all birds and an increase in 5 alpha-reductase activity only in the birds which remain sexually active. In the cloacal gland, the 5 beta-reductase activity markedly increases with age but more so in the birds which become sexually inactive. These data support the notion that the effects of testosterone are controlled by enzymatic shifts which could modulate the sensitivity to the hormone at the cellular level.  相似文献

In the male Japanese quail, testosterone is required for the activation of sexual behavior. This steroid dependent process may rely heavily on mediation via monoaminergic neurons. These experiments were conducted to study the relationship between reproductive state (hormonal and behavioral components) and levels of monoamines in selected areas of the brain in Japanese quail. In Experiment 1, monoamine levels in a number of brain areas were compared in castrates, testosterone-implanted castrates, and intact males. Monoamine levels were comparable to those previously measured in Japanese quail, and there were no significant differences due to treatment. Plasma luteinizing hormone (LH) levels and recovery of cloacal gland area in implanted castrates confirmed the afficacy of treatments. In Experiment 2, the disappearance of dopamine (DA) and norepinephrine (NE) following administration of a-methyl-para-tyrosine (aMPT) was used as an indicator of turnover rate. Male and female quail were gonadectomized at 3 weeks of age. At the age of five weeks, some gonadectomized males and females were given implants containing testosterone. Only intact males and testosterone-implanted castrated males showed reproductive behavior. Plasma gonadotropin levels were elevated in gonadectomized birds and reduced in steroid-implanted gonadectomized birds. The aMPT treatment significantly reduced the levels of DA and NE in the telecephalon and the level of DA in the hypothalamus. After aMPT treatment, the disappearance of NE in the telecephalon and of DA in the hypothalamus were significantly different according to the sex or treatment of the birds or both. Significant interactions between these two factors were observed. Disappearance rate of NE in the telecephalon was decreased by castration of males and increased by ovariectomy of females. Both effects were counteracted by testosterone. Reverse effects were observed for DA disappearance in the hypothalamus (increase with castration in males and decrease with ovariectomy in females). These results give evidence for altered aminergic function in specific areas of the brain relative to altered reproductive state.  相似文献

Fertilized eggs of chicken and quail were incubated under the simulated microgravity condition provided by a clinostat. The number of Primordial Germ Cells (PGCs) was counted in early embryogenesis, and the reproductive capacity of quail hatched following the simulated microgravity was investigated.Simulated microgravity caused significant decline of PGCs in the blood of early chicken embryos and in the gonads. The numbers of spermatogonia in the hatchling testis were also fewer than those in the control groups. Therefore, simulated microgravity may retard gonadial development and reduce the reproductive capacity.  相似文献

Several drugs have been shown to cross the placental barrier and affect the fetal testis causing a reduction in testosterone with a resultant impairment of sexual differentiation and an ultimate problem in adult sexual function. In this study, pregnant female rats were treated with 25 mg/kg of the pesticide 1,2-dibromo-3-chloropropane (DBCP). Treatment began on Days 14.5, 16.5, or 18.5 and continued through Day 19.5 of gestation. Some animals were killed on Day 20.5 of intrauterine life and fetal intratesticular testosterone was measured. All other animals were allowed to deliver, and the males were raised to adulthood. At adulthood, body, testis, prostate glands and seminal vesicle weights were recorded. Intratesticular testosterone and luteinizing hormone (LH) receptors were measured. Male and female sexual behavior was quantified and the volume of the sexually dimorphic nucleus of the preoptic area of the hypothalamus was calculated. The histological appearance of the testis was also examined. Treatment for 6 days during fetal life with DBCP decreased intratesticular testosterone by 50% compared to controls at 20.5 days of gestation. At adulthood, all male rats treated during fetal life had a reduced body weight that was correlated with the duration of exposure. Adult testis weight was reduced to 75% of controls as a result of 2 days of fetal exposure to DBCP, whereas 4 and 6 days of exposure during fetal life reduced testis weight by greater than 90%. A study was conducted to investigate endocrine and testicular changes in the male chicken associated with the ingestion of 10 or 20 ppm aflatoxin at 3 different stages of development. Weekly body weight gain, absolute and relative combined testes weights, and plasma testosterone concentrations were reduced in aflatoxin-fed males as compared to controls, with the greatest differences seen at 12 wk of age. The effect of dietary aflatoxin on levels of plasma luteinizing hormone (LH) was dependent on age at exposure. Concentrations of plasma LH in 6-wk-old control males were significantly higher than in aflatoxin-fed birds, whereas no treatment differences were observed in older males. Additionally, few changes were observed in static levels of monoamines in any of the brain regions assayed, regardless of age at exposure. The delay in peak levels of LH, as well as the suppression of plasma testosterone and testicular weight, indicate a possible delay in the onset of sexual maturation associated with aflatoxicosis in this species.  相似文献

Effects of yolk androgens on postnatal growth of offspring have been widely studied but their physiological role in the growth control is not fully understood due to an inconsistency in obtained results. We investigated androgen-mediated maternal effects on postnatal growth in relation to endocrine control mechanisms using two lines of Japanese quail divergently selected for high (HET) and low (LET) egg testosterone (T) content. Embryonic growth did not differ between the lines. During the growth period HET quail were heavier and displayed longer tarsi as compared with LET quail, with more pronounced line differences in males than females. HET males were heavier than LET males from the age of 2 weeks, reached the age of maximum growth rate earlier, and displayed higher asymptotic body weight than LET males. Accelerated growth in HET males was not accompanied by increased postembryonic plasma T concentrations. Plasma triiodothyronine levels did not differ between lines while plasma thyroxine levels were decreased in HET as compared with LET female chicks. Line differences in body weight disappeared in adult quail suggesting that yolk androgens, increased in a physiological way, resulted in stimulation of juvenile growth rate in precocial Japanese quail under stable social and environmental conditions.  相似文献

Three experiments were performed to analyze the time course of demasculinization in the Japanese quail and to test the activating and organizing effects of estradiol (E2) in adult sexually active birds. In Experiment 1, males and females were castrated at the age of 1 day or 1, 2, 4, and 6 weeks and treated as adults with testosterone (T). The age of castration had no effect on behavior and morphology in males. Plasma gonadotrophins (LH and FSH) were, however, higher in males castrated at or before than in those castrated after 2 weeks of age. This suggests that postnatal testicular secretions have organizing effects on the pituitary activity. Females which were castrated before 1 week of age were less sensitive to the activating effects of T than males, but were not fully demasculinized. The demasculinization of different reproductive characteristics such as male sexual behavior, cloacal gland size, and weight of the syringeal muscles is achieved in females at different times posthatching. In Experiment 2, castration of male and female quail at the ages of 4 days or 4 weeks confirmed that postnatal ovarian secretions contribute to the full behavioral and morphological demasculinization of females. It is easier to elicit mounting in T-treated females when they are tested in their home cage instead of a test arena. This difference was not observed in males. During Experiment 3, it was impossible to demasculinize sexually active adult males or females by treatment with Silastic implants of E2. E2 did not maintain sexual behavior in ovariectomized females showing male sexual behavior when treated with T but maintained the behavior in males.  相似文献

通过声谱分析,研究了5-120日龄雌、雄白腰文鸟(Lonchura striata swinhoei)的声谱变化,及该时段3个主要发声控制核团)HVC、RA、Area X)体积、睾丸(睾酮)的相应改变。结果如下:①45日龄以前,雌雄鸟只能发出简单鸣叫(call),鸣声基本不会鸣唱。②雄性HVC,RA,AreaX体积均比雌性大2-6部。3个核团的大小发育不完全一致。各核团的快速生长期与鸣唱学习的主要时段(60-120日龄)不同步,说明核团的个体发育可能不完全受发声行为的影响。③睾丸的充分发育(120日龄后)及血液中具有较高的睾酮水平是雄鸟发出成熟鸣唱语句的重要条件。  相似文献

This experiment was designed to determine the contribution, if any, of posthatching gonadal hormones to sexual differentiation of behavior in Japanese quail (Coturnix coturnix japonica). Males and females were gonadectomized or sham-operated (controls) prior to age 7 days posthatching. At age 4-9 weeks controls were gonadectomized. All birds were then given 2 weeks of testosterone propionate injections and tested for sexual behavior with female partners. Neonatally gonadectomized females exhibited more male-typical copulatory behavior than control females, but this effect was not statistically significant. Neonatal gonadectomy had no effect on males, and neonatally gonadectomized males exhibited significantly more male-typical copulatory behavior than neonatally gonadectomized females. Although the process of sexual differentiation may extend to a minor degree into the posthatching period in females, nonetheless it is largely complete at hatching in this species.  相似文献

An attempt has been made to investigate the relationship of cloacal gland with testes, testosterone and fertility in different lines of Japanese quail (Coturnix coturnix japonica). For this study, three lines of healthy adult male Japanese quails (<12 weeks) viz., heavy body weight (HB), white breasted (WB), and white egg shell (WES) were taken. They were housed in individual cages under uniform husbandry conditions and were provided with normal quail breeder ration and water ad libitum, with 14 h light/day. The experimental birds were selected from each of these three lines and categorized into different groups (15 birds/group) based on the increasing order of the area of cloacal gland. At the end of this experiment (24 weeks) the data indicated that size of the cloacal gland was directly proportional to foam discharge, foam weight, testicular weight, fertility and testosterone concentration in plasma. From this study it may be concluded that area of cloacal gland in Japanese quail is positively correlated with testicular weight, level of testosterone in plasma and fertility. Visual examination of cloacal gland and cloacal foam may provide a valuable non-invasive tool for predicting the fertilizing ability of individual male bird.  相似文献

Male rats given 250 mug oestradiol benzoate by subcutaneous injection on Day 4 of postnatal life showed a marked delay in the onset of the pubertal increase in the weight of the testes and seminal vesicles and in spermatogenesis but not a complete failure of sexual development. The increase in plasma testosterone concentration at puberty was also delayed in oestrogen-treated males but the eventual increase in seminal vesicle weight was closely related in time to the delayed increase in plasma testosterone concentration. Both plasma LH and FSH concentrations were reduced for about 10 days after oestrogen administration as compared to control values. After 22 days of age, plasma LH concentration did not differ significantly from the control values. The plasma FSH concentration of the oestrogen-treated males showed a delayed rise to values equal to or higher than those of controls of the same age. The delayed rise in plasma FSH concentration in the oestrogen treated males preceded the delayed rise in plasma testosterone in these animals. The decrease in plasma FSH concentration from the high prepubertal values to the lower values in adults occurred at different ages in the control and in oestrogen-treated rats but in both groups the decrease occurred as plasma testosterone levels were increasing and the first wave of spermatogenesis was reaching completion. The increase in plasma FSH concentration after castration was reduced in oestrogen-treated males during the period throughout which FSH levels in the intact animals were subnormal but the levels in oestrogen-treated males castrated after the delayed rise in FSH had occurred did not differ from control values. It is suggested that the delayed sexual maturation of male rats treated with high doses of oestrogen in the neonatal period is related principally to abnormalities in the secretion of FSH.  相似文献

The reproductive tracts of male sub-Antarctic fur seals Arctocephalus tropicalis ( n = 123), taken at Gough Island (40° 20'S, 9° 54'W) between November 1977 and October 1978, were examined. The presence of spermatozoa in the epididymal tubules showed that all males ≥4 years old had reached puberty, and the marked slower increase in mean testis weight and baculum length suggested that sexual (social) maturity was approached by 8-year-olds. Full adulthood was attained at 10–11 years of age based on the peak in mean testis and prostate weights, and mean baculum length. Secondary sexual characteristics were only fully developed in males ≥9 years old. A significant decline in mean testosterone concentration, and in testis, epididymis and prostate weights showed that adult males were reproductively quiescent during winter from May to July when seminiferous and epididymal tubules had lowest mean diameters with no spermatozoa present. Both the mean plasma testosterone concentration and mean testis weight peaked twice during the austral summer. The first peak coincided with the breeding season, and the second peak with the moulting period when adult males were impotent. Photoperiodic cueing might explain this seasonal trend.  相似文献

Experiments were designed to examine the influence of either one or both parents on the sexual maturation of male mice. A pregnant female and an adult vasectomized male were housed together and, at delivery, the litter was reduced to 6 pups, 3 of each sex. After weaning, mother, foster-father or both, stayed in cohabitation with their pups until sacrifice. The results showed that: (1) the permanent presence of one or both parents did not affect either testicular growth or the age at first fertile matings. (2) in maturing males (30-40 days), seminal vesicle growth was accelerated by the presence of the mother, inhibited by the presence of the foster-father. (3) in maturing males, the presence of one or both parents lowered plasma testosterone levels. On the opposite, in 90-day-old males, the same conditions increased testosterone levels. These results indicate that only the endocrine function of the testis is affected by the parental environment. The effects are age-dependent.  相似文献

Summary The influence of growth on the extent of heterosis for juvenile body weight and gonad development was studied in a diallel cross among two lines of Japanese quail differing in adult body size. A total of 1,096 birds (563 males and 533 females) was slaughtered between 25 and 49 days of age. Reciprocal cross differences were non-significant. Heterosis showed a curvilinear course with age peaking during early growth (body weight) and during sexual maturity (gonad percentage). Overall advanced physiological development of the crossbreds probably begins as early as during the embryonic stages and results in earlier sexual maturity. In females, heterosis for percentage gonads was biased strongly by the presence of a hard-shelled egg in the uterus.  相似文献

Eight sexually experienced long-term ovariectomized female rhesus monkeys were given tests of sexual behavior following treatment with 19-hydroxytestosterone (19-OH-T, 1 mg/day for 13 days), and their performance was compared with that following treatment with estradiol benzoate (EB, 10 μg/day for 13 days). Each female was tested for 10 min with each of nine adult males. Blood samples were taken on the last day of treatment with EB, at the end of the intervening 3-month period of no treatment, and on the last day of treatment with 19-OH-T. Blood levels of testosterone and estradiol were quantified by radioimmunoassay. Mean rate of presenting at a distance (proceptive behavior) was significantly higher (P < 0.05) when they were treated with 19-OH-T, but the ratio of presents to male contacts (receptive behavior) was significantly higher (P <0.05) when they were treated with EB. All other components of female sexual behavior were the same. Males displayed fewer annoyance responses (rejecting jerk, P < 0.05) when the females were treated with 19-OH-T than when they were treated with EB. All other male responses occurred with the same frequencies under the two female treatment conditions. Injection of 19-OH-T and EB both resulted in plasma testosterone and estradiol levels higher than those found in the untreated condition. Testosterone levels did not differ under the two treatments (P > 0.05), but estradiol levels were higher under EB treatment than under 19-OH-T (P < 0.05). This study suggests that both testosterone and estradiol are essential for maximum sexual performance and that various components of sexual behavior may be differentially influenced by the ratio of testosterone to estradiol in plasma.  相似文献

Tanyang is one of the most important sheep breeds in the northwest of China. However, there is no detailed data on sexual development and timing of puberty/sexual maturity of this breed. This experiment was designed to study the relationships between growth rate and sexual development by measuring body weight, testis diameter, and plasma testosterone concentrations, and observing sexual behavior through puberty from 3 to 8 months old of entire Tanyang ram lambs. At same time, to assess the potential efficacy and welfare benefits of non-surgical castration, the effect of active immunization against gonadotrophin-releasing hormone (GnRH) on sexual development and growth rate on ram lambs was evaluated, and compared with surgically castrated and entire lambs. The results showed that body weight increased at a constant rate from 3 to 8 months old of age. Testis diameter increased rapidly between 3 and 5 months, followed by a period of slow growth. Plasma testosterone concentrations increased sharply from 3 to 6 months of age (P<0.05), after which there were no significant changes from 6 to 8 months of age. Sexual behavior started from 5 months of age. After ram lambs were immunized against GnRH at 3 months of age, there was no increase in testis diameter in the first 4 months after immunization. Plasma testosterone concentrations remained at similar levels to surgically castrated lambs until 6 months of age, and the onset of sexual behavior was delayed, compared with entire ram lambs. GnRH immunization and surgical castration had no significant effect on the body weight by the end of this experiment. These results suggest that 4-5 months of age is a critical period for pubertal changes and testosterone plays an important role in the initiation of puberty and sexual maturity of Tanyang ram lambs in China. In addition, a single shot GnRH immunization at 3 months, acting as an alternative to surgical castration, is a practical method for Chinese Tanyang ram lambs, and provides animal welfare benefits.  相似文献

Inhibins and activins are implicated as endocrine regulators of follicle-stimulating hormone production and of testicular steroidogenesis and spermatogenesis in mammals. The potential involvement of these proteins in cockerels was investigated by measurement of circulating inhibin A, inhibin B, total inhibin alpha-subunit immunoreactivity (ir-alpha), activin A, LH, FSH, and testosterone from the juvenile state through to sexual maturity. Plasma inhibin A remained low between 6 to 12 wk of age and increased approximately threefold (P < 0.05) to a prepubertal peak between Weeks 14 to 18, followed by a gradual decline to the end of the study (Week 24). Although plasma FSH levels were not correlated to inhibin A before Week 16 (r = -0.17), they were negatively correlated from Week 18 (r = -0.49; P < 0.005). Inhibin B levels were below the assay detection limit until 16 wk of age but thereafter rose steadily in parallel with FSH (r = 0.27; P < 0.02) and testosterone (r = 0.35; P < 0.005). Thus, inhibins A and B showed divergent profiles during sexual maturation. Plasma ir-alpha levels were much higher than dimeric inhibin levels throughout, although the relative difference varied with age. Plasma activin A levels were below the assay detection at all times. Juvenile cockerels were actively immunized against a synthetic chicken inhibin alpha-subunit peptide conjugate to determine effects on plasma hormones and on testicular weight, morphology, and activin A content. Immunization generated circulating antibodies that bound (125)I-bovine 32-kDa inhibin but did not affect plasma FSH or testosterone levels at any stage of development. However, immunization reduced postpubertal plasma LH levels (P < 0.05) and promoted increased testicular weight (24%; P < 0.01) and total testicular activin A content (42%; P < 0.001) at 24 wk. Testis weight of immunized birds was positively correlated with inhibin antibody titer (r = 0.61; P < 0.05). Live weight gain was not affected by immunization. Morphometric analysis of testis sections showed that inhibin immunization had no effect on the fractional volume of the seminiferous tubule wall, seminiferous tubule lumen, or interstitial tissue area. Likewise, seminiferous tubule surface area and surface area:volume ratios were not different from controls. These findings support differential roles for inhibins A and B in regulating the pituitary-testicular axis during sexual maturation in the cockerel but highlight the need for more detailed studies to distinguish between potential endocrine and local intragonadal roles of inhibin-related peptides and to elucidate the mechanism by which immunization against inhibin alpha-subunit promotes testis enlargement without raising plasma FSH.  相似文献

This series of experiments sought to determine whether conversion of androgen to estrogen is important in the activation of male sexual behavior in quail by seeing if an antiestrogen will block androgen stimulated copulation in this species. Experiment I compared the ability of two antiestrogens, MER-25 (5 mg/day) and CI-628 (2 mg/day), to block estrogen stimulated characteristics in female quail. Both treatments greatly reduced oviduct growth in “photically castrated” females given estradiol benzoate (EB, 50 μg/day), but only CI-628 reduced receptivity in these birds. In Experiment II surgically castrated males given 50 μg/day EB together with 2 mg/day CI-628 were much less receptive than castrated males given EB alone, and in addition copulated in fewer tests. In Experiments III, IV, and V, castrated males given testosterone propionate (TP) together with CI-628 were compared with males given TP alone. The ability of CI-628 to suppress TP-stimulated copulation increased with increasing CI/TP dosage ratio, and at the highest ratio (4:1), CI-628 effectively blocked copulation in five out of seven birds. Those birds that did copulate did so in fewer tests and performed fewer cloacal contact movements. CI-628 had no antiandrogenic effects in these experiments. These results suggest that estrogens may be important active metabolites of testosterone with respect to quail copulation.  相似文献

An experiment was carried out to study the contribution of endogenous steroids during postnatal life to the sexual differentiation in Japanese quail. Male and female quail were gonadectomized at one week of age and when adult received testosterone silastic capsules. Seventy seven percent of these females showed male-type sexual behaviour and 46 percent even copulated. All males showed sexual behaviour including cloacal contact movements. Males and females grew cloacal glands of nearly identical sizes in response to the testosterone treatment. It is concluded that sexual differentiation in quail is at least in part a postnatal process and more research is needed to elucidate its exact mechanisms and time course.  相似文献
