The bronze (bz), locus encodes a flavanoid glucosyltransferase (UFGT) which is required for anthocyanin biosynthesis. Enzyme accumulation in the primary root and coleoptile were compared for the wild-type Bz-McC allele and the regulatory mutant bz-m4 Derivative 6856. UFGT levels are quantitatively reduced in bz-m4 plant tissues as compared to that of Bz-McC, however both alleles had similar developmental profiles.  相似文献

Summary The maize mutable allele bz-m2 (Ac), which arose from insertion of the 4.6 kb Ac element in the bz (bronze) locus, gives rise to stable bz (bz-s) derivatives that retain an active Ac element closely linked to bz. In the derivative bz-s:2114 (Ac), the Ac element is recombinationally inseparable from bz and transposes to unlinked sites at a frequency similar to that in the progenitor allele bzm2 (Ac). Both alleles have been cloned and sequenced. The bz-s:2114 (Ac) mutation retains Ac at the original site of insertion, but has lost a 789 pb upstream bz sequence adjacent to the insertion, hence the stable phenotype. The 8 bp target site direct repeat flanking the Ac insertion in the bz-m2 (Ac) allele is deleted in bz-s: 2114 (Ac), yet the Ac element is not impaired in its ability to transpose. The only functional Ac element in bz-s:2114 (Ac) is the one at the bz locus: in second-cycle derivatives without Ac activity, the loss of Ac activity correlated with the physical loss of the Ac element from the bz locus. The deletion endpoint in bz-s: 2114 (Ac) corresponds exactly with the site of insertion of a Ds element in a different bz mutation, which suggests that there may be preferred integration sites in the genome and that the deletion originated as the consequence of an abortive transposition event. Finally, we report two errors in the published Ac sequence.  相似文献

Summary A study was made of the effects of pre-meiotic x-irradiation on the bronze locus in chromosome 9 of maize. Plants of Sh Bz Wx/Sh Bz Wx constitution were treated with ca. 1000r and pollen from these individuals was applied to silks of sh bz wx tester plants. In the F1 progeny, three Sh Wx kernels having a bz aleurone or showing Bz-bz variegation (the bz-x3, bz-x4 and bz-x5 mutants) were selected as possible mutations at the bz locus. One kernel of sh bz wx phenotype as well as one exhibiting sh, bz and Wx (sh-bz-x3) were also selected for more intensive study. Progeny tests of the sh bz wx individual along with cytological observations indicated that a ring chromosome was the probable cause of the mutant phenotype although an alternative hypothesis is not ruled out. The behavior of sh-bz-x3 can be interpreted as the result of either a minute deficiency involving the Sh and Bz loci or a simultaneous suppression of the two dominant alleles. Progeny of the bz-x mutants exhibited genetic instability of bronze. It is hypothesized that this behavior is due to the activation or alteration by x-rays of gene control mechanisms which affect the bronze gene.Dedicated with appreciation and affection to Dr. M. M. The pattern of transposition of Ac from the mutable allele bz-m2(Ac) has been investigated. Stable (bz-s) and finely spotted (bz-m(F)) exceptions were selected from coarsely spotted bz-m2(Ac) ears. The presence or absence of a transposed Ac (trAc) in the genome was determined and, when present, the location of the trAc was mapped relative to the flanking markers sh and wx. The salient general features of Ac transposition to sites linked to bz are that the receptor sites tend to be clustered on either side of the bz donor site and that transposition is bidirectional and nonpolar. Thus, the symmetrical clustering in the distribution of receptor sites adjacent to bz differs from the asymmetrical clustering reported in 1984 for the P locus by I. M. GREENBLATT. Though Ac tends to transpose preferentially to closely linked sites, an appreciable fraction of Ac transpositions from bz-m2(Ac) is to unlinked sites: 41% among bz-s derivatives and 59% among bz-m(F) derivatives. Many transposition events among the bz-m(F) selections result in kernels carrying a genetically noncorresponding embryo. These can be interpreted as twin sectors arising at one of the megagametophytic mitoses. The bz locus data fit the earlier (1962) model of I. M. GREENBLATT and R. A. BRINK in which transposition takes place from a replicated donor site to either an unreplicated or replicated receptor site.  相似文献

Aberrant Ratio (AR) stocks of maize were tested for transposition activity. Lines exhibiting AR and homozygous for the dominant alleles at the Sh Bz and Wx loci in the short arm of chromosome 9 were crossed as males to a sh bz wx tester. Among a population of 346,201 kernels, eight mutations of sh and two of bz were recovered. Eight of the ten mutations survived and none was as vigorous as its normal sibs. At least five of the sh mutants appear to be unstable in F2 and subsequent generations. An unexpected observation was the high incidence of somatic loss of chromosome 9 markers (Sh Bz and Wx), indicating chromosome breakage or nondisjunction. Southern blot hybridization analysis of the sh alterations indicate that all but one mutant are associated with structural DNA rearrangements at the shrunken locus. Possible mechanisms by which these alterations arose are discussed.  相似文献

The major UDPG: flavonol glucosyltransferase (UFGT) in maize is an enzyme of strict positional specificity known to be coded by the Bz locus. In bz mature endosperms, no UFGT can be detected. However, bz embryos possess a residual flavonol glucosyltransferase activity which is independent of Bz locus control. The bronze (bz) locus in maize, located in the short arm of chromosome 9 (9S), is the structural gene for the anthocyanin biosynthetic enzyme UFGT. The gene has been cloned and its physical map has been oriented relative to the centromere of 9S. We report here the genetic fine structure mapping of several biochemically characterized EMS-induced bz-E mutations, derived from the Bz-W22 isoallele, and Ds insertion bz-m mutations, derived from the Bz-McC isoallele. Two UFGT(-), CRM(+ ) mutants (bz-E2 and bz-E5), which genetically identify coding sequences in the gene, and three UFGT(-), CRM(- )bz-E mutants were mapped against the Ds insertion mutants bz-m1 and bz-m2(DI) by selecting Bz intragenic recombinants from heterozygotes of the type bz-E/bz-m . The exclusive occurrence of one recombinant outside marker class allowed the unambiguous placement of the mutants in a genetic fine structure map. Peculiarly, the two CRM(+)bz-E mutants lie upstream of the three CRM(-)bz-E mutants and at a considerable genetic distance. The UFGT allozymes encoded by the progenitor alleles Bz-W22 and Bz-McC differ in two properties, thermal stability and activity. The sites responsible for these properties were mapped as unselected markers among the Bz intragenic recombinants. The thermal stability site, which also identifies a coding region of the gene, mapped very close to the CRM(+)bz-E mutant sites. The site responsible for variation in activity, which probably identifies a region involved in regulation of expression of the bz locus, mapped at the 5' or proximal end of the locus. It was found to be inseparable from the Ds insertion in bz-m1 that lies very close to the 5' end of the transcribed region.-Evidence was obtained that the insertion of Ds within the bz gene has a suppressing effect on intragenic recombination. Additional data are also presented supporting our observation that Ds affects the pattern of intragenic recombination at bz.-Based on the total genetic length of the bz gene and on the physical size of the transcribed region, we estimate that one unit of recombination at bronze corresponds to 14 kb of DNA. This estimate is more than 100 times smaller than the average value for the whole genome and implies that there may be regions, such as bronze, that serve as hotspots for recombination.  相似文献

A case of somatic instability affecting aleurone colour in a strain of maize from India with flint background was analysed. The somatic instability is localized to theC 1 (Inhibitor) allele ofC locus on the short arm of chromosome 9. Molecular tests indicated thatAc is not present in the Indian stock and the evidence is consistent with the involvement of theEn (Spm) transposable element in the instability. The presence of theEn (Spm)-like element in the stock would suggest that these elements have been present in the maize genome for a long time. A new allele ofshrunken (sh1) gene with a somewhat unorthodox breeding behaviour is also described.  相似文献

Summary Several stable germinal derivatives, with varied phenotypic expression have been recovered from testcrosses of four different a2 mutable alleles (a2m-7 8018, a2m-6 8144, a2m-6 8140, and a2m-1 1511) under the control of the En controlling-element system. These are colored, pale, and colorless aleurone types; the former two represent mutations of a2-mA2 type. The phenotypic variation among colored and pale derivatives has been confirmed by quantitative determination of anthocyanin content in the aleurone tissue of the kernels.The data suggest that significant variation exists between phenotypically similar colored derivatives arising from different mutable alleles as well as from the same source allele. Every tested colored derivative is significantly different from the pale, colorless, and colored control (W22 color converted line). Genetic analysis of these colored derivatives suggests that all of them contain one or more En and, hence, they are characterized as permanent changes of a nonresponsive (nr) type. Their frequencies are variable among the testcrosses.Similarly, the pale derivatives, which are clearly different from colored and colorless, also show significant differences in terms of anthocyanin content. Significant differences exist between pales arising from the same allele as well as between pales arising from different alleles. All the tested pales are significantly different from colorless and colored derivatives. Thus, the pales, as well as colored derivatives, represent a differential impairment of anthocyanin synthesis in the aleurone tissue. The genetic analysis confirmed that all the pales, except the pales of a2m-1 1511, lack the capacity to respond to En. These pale derivatives can be with or without En.Qualitative differences among the colored, pale, and colorless types have been investigated by thin-layer chromatographic and spectroscopic techniques. The results suggest that there are no qualitative differences, in terms of anthocyanin pigments, between colored, pale, and colored control. Summary Phenotypic revertants from several kinds of mutations, including deletions, have been detected by pollen analysis at the wx and Adh loci in maize. Mutations in these genes give phenotypic revertants with median frequencies of 0.7 and 0.5×10–5, respectively. However, the nature of such revertants can only be analyzed following their recovery from conventional matings. In the current study large seed populations derived from crosses involving several bz (bronze) locus mutations in maize were examined for reversion to a Bz (purple) expression. Deletion, insertion and point mutations were included in the study. Principally, over 2 million gametes of the bz-R mutation, which is shown here to be associated with a 340 base pair deletion within the transcribed region of the gene, have been screened for reversion. No revertants from it or any of the other bz mutations have been recovered, even though a total of almost 5 million gametes from homoallelic crosses have been examined to date. Results from seed analysis are discussed in reference to those from pollen analysis in maize.  相似文献

We produced and studied for 3?years two synthetic populations of maize differing in their constitution only for the selected alleles present at the red color 1 (r1) locus (R-sc vs. r?Cr). r1 is a regulatory gene conferring anthocyanin pigmentation in different tissues: the R-sc allele confers pigmentation only in the aleurone seed layer, while the r?Cr allele confers pigmentation in several tissues such as root, silk and anther but the seed is colourless. The colourless population (r?Cr/r?Cr) was characterized by improved agronomic features, such as ear weight and plant height, compared with the R-sc/R-sc coloured population. This finding was confirmed by studying single F4 R/r families where the presence of the r?Cr allele conferred positive features, acting as a dominant trait. Quantitative trait locus (QTL) analysis performed using molecular markers on the long arm of chromosome 10 (bin 10.06), where the r1 gene maps, identified a QTL map position for plant height tightly associated to the r1 gene. Thus the r1 gene may represent a major QTL or it could be closely linked to another gene involved in the agronomic performance of the two populations studied.  相似文献

Summary The regulatory mutation bronze mutable 4 Derivative 6856 (bz-m4 D6856) contains a complex 6.7 kb Dissociation (Ds) element tagged with a duplication of low copy bz 3 flanking sequences (Klein et al. 1988). This creates a unique opportunity to study the transposition of a single member of the repetitive family of Ds elements. Eighteen full purple revertants (Bz alleles) of bz-m4 were characterized enzymatically and by genomic mapping. For 17 of the Bz alleles, reversion to a wild-type phenotype was caused by excision of the 6.7 kb Ds transposon. Nine of these Bz alleles retained the transposon somewhere in their genome. In this study we show that like Ac (Schwartz 1989; Dooner and Belachew 1989), the 6.7 kb Ds element can transpose within a short physical distance, both proximal and distal to its original position. Additional bz sequences have been mapped immediately distal to the mutant locus in bz-m4 D6856; genetic evidence suggests these are flanked by two additional Ds elements. The remaining Bz revertant, Bz :107, arose from excision of a more complex 13 kb Ds element.  相似文献

The Enhancer (En) controlling element system in maize includes numerous alleles, one of which, a2m(r-pa-pu), was recovered as a derivative of the a2m(1 1511) allele. The behavior and composition of this allele is described. In the presence of the independently inherited regulatory element, En, the a2m(r-pa-pu) allele mutates and expresses in the aleurone purple, pale and colorless sectors within a colorless background. In the absence of En, the pigmentation is uniformly pale. Colored and colorless germinal mutations of the a2m(r-pa-pu) allele were recovered, although the derivative alleles tested did not respond to En. Uniformly colored derivatives, designated A2', arise only from states of a2m(r-pa-pu) that respond to an active En at relatively early stages in plant development, suggesting that somatic and germinal mutations may arise through similar events. Mutations to A2-expressing alleles appear to occur relatively late in plant development, that is, at or near meiosis, since recognizable mutant sectors do not appear on testcross ears. Kernels exhibiting such mutants are randomly distributed over the ear. The resulting A2' alleles express pigmentation ranging in a continual sequential series from dark pale to intense purple.  相似文献

The regulation of excision of Mu elements of the Mutator transposable element family of maize is not well understood. We have used somatic instability of Mu receptor elements from the Bronze 1 and Bronze 2 loci to monitor the frequency and the timing of excision of Mu elements in several tissues. We show that spot size in the aleurone of a bz2::mu1 stock varies between one to approximately 256 cells. This indicates that excision events begin eight divisions prior to full aleurone differentiation and end after the last division of the aleurone. We show that excision is equally biased for late events in all other tissues studied. A locus on chromosome 5 has been identified that affects spot size, possibly by altering the timing of Mu excision. Using somatic excision as an assay of Mutator activity, we found that activity can change in small sectors of the tassel; however, there are no overall activity changes in the tassel during the period of pollen shedding. We also report the recovery of germinal revertants for the bz1::mu1 and bz2::mu1 alleles. One of these revertant alleles was characterized by Southern blot analysis and found to be similar to the progenitor of the mutable allele.  相似文献
