Copper nutrition inhibited lipid peroxidation in root nodules of yellow lupin plants at the early growth stages by about 50 %. The antioxidative activity of copper in the process of lipid peroxidation could be associated with Cu taking part in oxidative reaction of nodule catechol-like siderophores and its effect on iron accumulation and reactivity. The obtained results, for the first time, suggest that the ability of copper to inhibit lipid peroxidation in nodules could be considered as a major function for Cu requirements by symbiotic N2 fixation in grain legume nodules.  相似文献

The effect of copper nutrition on symbiotic N2 fixation in Lupinus luteus L. was studied. Copper nutrition increased the yield, total nitrogen content and dry weight of nodules. The control plants did not produce pods. Copper deficiency limited iron uptake and its translocation to the nodules. Nodules of copper-deficient plants contained less than half the leghaemoglobin concentration of copper-adequate plants and about one third the polyphenol oxidase activity, tested with catechol as a substrate.  相似文献

The effect of different rates and methods of fertilizer nitrogen application and potassium nutrition on the root nodule formation of faba bean plants var. Nadwiślański and on their nitrogenase activity was studied. It was found that fertilizer N depressed the nodule formation and nitrogenase activity, but inhibitory effect of N was smaller when it was supplied to the leaves instead to the soil. Plants growing at higher K level were in a position allowing on better development of nodules and consequently higher N2- fixation.  相似文献

Faba bean (Vicia faba) plants were inoculated with rhizobia and then their sap was infected with broad bean mottle bromovirus (BBMV) or bean yellow mosaic potyvirus (BYMV) in a field experiment. Both viral infections significantly decreased shoot and root dry weight, number of nodules, nodule dry weight, numbers of flowers and pods/plant, total plant N, grain yield and N2 fixation. However, inoculation withRhizobium leguminosarum significantly increased all these parameters, both in healthy and virus-infected plants. Fifteen genotypes of faba bean (Vicia faba L.) were inoculated with salt-tolerant Rhizobium leguminosarum biovar. viciae strain GRA 19 in solution culture with 0 (control) and 75 mM NaCl added immediately after transplanting. Genotypes varied in their tolerance of high levels of NaCl. Physiological parameters (dry weight of shoot and root, number and dry weight of nodules) were not affected by salinity in lines VF46, VF64 and VF112. Faba bean line VF60 was sensitive to salt stress. Host tolearance appeared to be a major requisite for nodulation and N2 fixation under salt stress. Tolerant line VF112 sustained nitrogen fixation under saline conditions. Activity of the ammonium assimilation enzymes glutamine synthetase and glutamate synthase, and soluble protein content, were reduced by salinity in all genotypes tested. Summary Experiments were done to test whether N fixation is more sensitive to high soil temperatures in common bean than in cowpea or soybean. Greenhouse experiments compared nodulation, nitrogenase activity, growth and nitrogen accumulation of several host/strain combinations of common bean with the other grain legumes and with N-fertilization, at various root temperatures. Field experiments compared relative N-accumulation (in symbiotic relative to N-fertilized plants) of common bean with cowpea under different soil thermal regimes. N-fertilized beans were unaffected by the higher temperatures, but nitrogen accumulation by symbiotic beans was always more sensitive to high root temperatures (33°C, 33/28°C, 34/28°C compared with 28°C) than were cowpea and soybean symbiosis. Healthy bean nodules that had developed at low temperatures functioned normally in acetylene reduction tests done at 35°C. High temperatures caused little or no suppression of nodule number. However, bean nodules produced at high temperatures were small and had low specific activity. ForP. vulgaris some tolerance to high temperature was observed among rhizobium strains (e.g., CIAT 899 was tolerant) but not among host cultivars. Heat tolerance ofP. acutifolius andP. lunatus symbioses was similar to that of cowpea and soybean. In the field, high surface soil temperatures did not reduce N accumulation in symbiotic beans more than in cowpea, probably because of compensatory nodulation in the deeper and cooler parts of the soil.  相似文献

The relationship between ureide N and N2 fixation was evaluated in greenhouse-grown soybean (Glycine max L. Merr.) and lima bean (Phaseolus lunatus L.) and in field studies with soybean. In the greenhouse, plant N accumulation from N2 fixation in soybean and lima bean correlated with ureide N. In soybean, N2 fixation, ureide N, acetylene reduction, and nodule mass were correlated when N2 fixation was inhibited by applying KNO3 solutions to the plants. The ureide-N concentrations of different plant tissues and of total plant ureide N varied according to the effectiveness of the strain of Bradyrhizobium japonicum used to inoculate plants. The ureide-N concentrations in the different plant tissues correlated with N2 fixation. Ureide N determinations in field studies with soybean correlated with N2 fixation, aboveground N accumulation, nodule weight, and acetylene reduction. N2 fixation was estimated by 15N isotope dilution with nine and ten soybean genotypes in 1979 and 1980, respectively, at the V9, R2, and R5 growth stages. In 1981, we investigated the relationship between ureide N, aboveground N accumulation, acetylene reduction, and nodule mass using four soybean genotypes harvested at the V4, V6, R2, R4, R5, and R6 growth stages. Ureide N concentrations of young stem tissues or plants or aboveground ureide N content of the four soybean genotypes varied throughout growth correlating with acetylene reduction, nodule mass, and aboveground N accumulation. The ureide-N concentrations of young stem tissues or plants or aboveground ureide-N content in three soybean genotypes varied across inoculation treatments of 14 and 13 strains of Bradyrhizobium japonicum in 1981 and 1982, respectively, and correlated with nodule mass and acetylene reduction. In the greenhouse, results correlating nodule mass with N2 fixation and ureide N across strains were variable. Acetylene reduction in soybean across host-strain combinations did not correlate with N2 fixation and ureide N. N2 fixation, ureide N, acetylene reduction, and nodule mass correlated across inoculation treatments with strains of Bradyrhizobium spp. varying in effectiveness on lima beans. Our data indicate that ureide-N determinations may be used as an additional method to acetylene reduction in studies of the physiology of N2 fixation in soybean. Ureide-N measurements also may be useful to rank strains of B. japonicum for effectiveness of N2 fixation.  相似文献

Nodulated and denodulated roots of adzuki bean (Vigna angularis), soybean (Glycine max), and alfalfa (Medicago sativa) were exposed to 14CO2 to investigate the contribution of nodule CO2 fixation to assimilation and transport of fixed nitrogen. The distribution of radioactivity in xylem sap and partitioning of carbon fixed by nodules to the whole plant were measured. Radioactivity in the xylem sap of nodulated soybean and adzuki bean was located primarily (70 to 87%) in the acid fraction while the basic (amino acid) fraction contained 10 to 22%. In contrast, radioactivity in the xylem sap of nodulated alfalfa was primarily in amino acids with about 20% in organic acids. Total ureide concentration was 8.1, 4.7, and 0.0 micromoles per milliliter xylem sap for soybean, adzuki bean, and alfalfa, respectively. While the major nitrogen transport products in soybeans and adzuki beans are ureides, this class of metabolites contained less than 20% of the total radioactivity. When nodules of plants were removed, radioactivity in xylem sap decreased by 90% or more. Pulse-chase experiments indicated that CO2 fixed by nodules was rapidly transported to shoots and incorporated into acid stable constituents. The data are consistent with a role for nodule CO2 fixation providing carbon for the assimilation and transport of fixed nitrogen in amide-based legumes. In contrast, CO2 fixation by nodules of ureide transporting legumes appears to contribute little to assimilation and transport of fixed nitrogen.  相似文献

Summary Mineral nitrogen did not increase grain yield and seed protein levels ofVicia faba L. andLupinus luteus L. in field trials and pot experiments. Fixed N2 was substituted by mineral nitrogen in these cases because of inhibition of N2 fixation by mineral nitrogen. Contrary to these results mineral nitrogen increased grain yields and seed protein amounts ofLupinus albus L.,Pisum sativum L., andGlycine max. (L.) Merr. The nitrogen effect was caused at an early stage by saving energy due to inhibition of N2 fixation (measurement of gas exchange by means of IRGA). In case of the N application after flowering grain, yields and seed protein levels increased because the mineral N was an additional nitrogen source for plants. At this stage the plants had ceased fixing atmospheric nitrogen. The high sink activity of growing fruits induced a lack of assimilates in nodules (determined by means of14CO2 application). The N effect was therefore the consequence of the lower assimilate pool for supplying root nodules in these plants in comparison withVicia faba L. andLupinus luteus L. Hence it follows that response to mineral nitrogen can be a criterion for discovering more effective Rhizobium-host combinations.  相似文献

Faba bean (Vicia faba L. var. minor cv. Alborea) and pea (Pisum sativum L. cv. Lincoln) plants, inoculated with Rhizobium leguminosarum biovar. viciae strain GRA19, were treated with salt (100 mM NaCl) and/or nitrate (8 mM KNO3) to test whether plants grown with inorganic-nitrogen are more tolerant to salinity than plants entirely reliant upon fixed nitrogen. According to the growth inhibition recorded, pea plants dependent on dinitrogen fixation proved more tolerant to salt stress than those N-fertilized, in contrast to results obtained for faba bean plants. This study therefore confirms that plants dependent on nitrogen fixation are not always more sensitive to salinity than are N-fertilized plants. Nitrate addition did not reduce the specific nitrogenase activity in pea, but did in faba bean. However, nodulation was inhibited in both legumes. The specific nitrogenase activity was more affected by salt treatment in N-fertilized plants for both legumes. The activity of the enzymes mediating ammonium assimilation in nodules (GS, NADH-GOGAT) was inhibited by salt stress both in N-fixing and in N-fertilized pea and faba bean plants.  相似文献

The adzuki bean (Vigna angularis (Wild.) Ohwi and Ohashi) and common bean (Phaseolus vulgaris L.) have a high physiological demand for N. A 2-year field study was conducted to investigate the seasonal change of available soil N and symbiotic N2 fixation usage. The beans were seeded at two densities, 22.2 plants m–2 with a row spacing of 0.3 m and 11.1 plants m–2 with a row spacing of 0.6 m. The amount of fixed N2 in the shoot was calculated using the 15N natural abundance method. The common bean demonstrated low N2 fixation and the ability to accumulate high levels of soil N. Soil nitrate under the common bean was continually absorbed. The adzuki bean, on the other hand, had a remarkable peak of N accumulation in the early reproductive stage. This was mainly due to N2 fixation, though the soil nitrate level was high. Narrowing the plant row spacing increased the dry matter yield of both species, but the origin of the increased N differed between the species. For the first 77 DAP in 1999 (73 DAP in 2000) the N increase for both beans was due to both soil and atmospheric N2. At harvest, though, the increase of N in common bean was mainly due to soil N, while that in adzuki bean was mainly due to atmospheric N2. It can be concluded that the low symbiotic N2 fixation ability of common bean was due to its high soil N uptake ability and constant N accumulation, which enabled an efficient soil N absorption. Adzuki bean absorbed N mainly for a short period and depended more on symbiotically fixed N2 and, in contrast to common bean, left a high level of NO3-N remaining in the soil after cropping.  相似文献

In order to assess symbiotic activity (the nodules integrity and the iron use efficiency) in common bean (Phaseolus vulgaris L.) under low iron availability, the growth of plants and nodules, the concentration of leghaemoglobin and malondialdehyde, and activity of nitrogenase, catalase, peroxidase and superoxide dismutase were analysed in two (contrasting) common bean varieties subjected to iron deficiency. Results show that nitrogen fixation and leghaemoglobin accumulation decreased at limiting iron availability while malondialdehyde concentration increased under these conditions. The tolerant variety to iron deficiency, ARA14, was clearly less affected than the sensitive one, Coco blanc. A significant stimulation of peroxidase (POD) activity was observed in ARA14 under iron deficiency. At the same conditions, SOD and CAT activities in ARA14 plants were maintained at high level. It was also found that the iron use efficiency for leghaemoglobin accumulation, SOD, CAT and POD activities were critical for the protection of symbiotic system against oxidative burst and for the maintaining of an optimal functioning of N2 fixing system.  相似文献

Iron is an essential element for plants as well as all living organisms, functioning in various physiological and biochemical processes such as photosynthesis, respiration, DNA synthesis, and N2 fixation. In the soil, Fe bioavailability is extremely low, especially under aerobic conditions and at high pH ranges. In contrast, plants with nodules on their roots that fix atmospheric nitrogen need much more iron. To highlight the physiological traits underlying the tolerance of N2-fixing common bean to iron deficiency, two genotypes were hydroponically cultivated in a greenhouse: Coco nain (CN) and Coco blanc (CB). Plants were inoculated with an efficient strain of Rhizobium tropici, CIAT899, and received a nutrient solution added with 0 µM Fe (severe Fe deficiency, SFeD), 5 µM Fe (moderate Fe deficiency, MFeD) or 45 µM Fe (control, C). Several physiological parameters related to photosynthesis and symbiotic nitrogen fixation were then analyzed. Iron deficiency significantly reduced whole plant and nodule growth, chlorophyll biosynthesis, photosynthesis, leghemoglobin (LgHb), nitrogenase (N2ase) activity, nitrogen, and Fe nutrition, with some genotypic differences. As compared to CB, CN maintained better Fe allocation to shoots and nodules, allowing it to preserve the integrity of its photosynthetic and symbiotic apparatus, thus maintaining the key functional traits of the plant metabolism (chlorophyll biosynthesis and photosynthesis in shoots, leghemoglobin accumulation, and nitrogenase activity in root nodules). Plant growth depends on photosynthesis, which needs to be supplied with sufficient iron and nitrogen. Fe deficiency stress index (FeD-SI) and Fe use efficiency (FeUE) are two physiological traits of tolerance that discriminated the studied genotypes.  相似文献

Background and Aims

Drought is the principal constraint on world production of legume crops. There is considerable variability among genotypes in sensitivity of nitrogen fixation to drought, which has been related to accumulation of ureides in soybean. The aim of this study was to search for genotypic differences in drought sensitivity and ureide accumulation in common bean (Phaseolus vulgaris) germplasm that may be useful in the improvement of tolerance to water deficit in common bean.

Methods

Changes in response to water deficit of nitrogen fixation rates, ureide content and the expression and activity of key enzymes for ureide metabolism were measured in four P. vulgaris genotypes differing in drought tolerance.

Key Results

A variable degree of drought-induced nitrogen fixation inhibition was found among the bean genotypes. In addition to inhibition of nitrogen fixation, there was accumulation of ureides in stems and leaves of sensitive and tolerant genotypes, although this was higher in the leaves of the most sensitive ones. In contrast, there was no accumulation of ureides in the nodules or roots of stressed plants. In addition, the level of ureides in the most sensitive genotype increased after inhibition of nitrogen fixation, suggesting that ureides originate in vegetative tissues as a response to water stress, probably mediated by the induction of allantoinase.

Conclusions

Variability of drought-induced inhibition of nitrogen fixation among the P. vulgaris genotypes was accompanied by subsequent accumulation of ureides in stems and leaves, but not in nodules. The results indicate that shoot ureide accumulation after prolonged exposure to drought could not be the cause of inhibition of nitrogen fixation, as has been suggested in soybean. Instead, ureides seem to be produced as part of a general response to stress, and therefore higher accumulation might correspond to higher sensitivity to the stressful conditions.  相似文献

Summary The total amount of nitrogen derived from symbiotic nitrogen fixation in two pea and one field bean cultivar, supplied with 50 kg N ha−1 at sowing (‘starter’-N), was estimated to 165, 136, and 186 kg N ha−1, respectively (three-year means). However, estimates varied considerably between the three years. At the full bloom/flat pod growth stage from 30 to 59 per cent of total N2 fixation had taken place. The proportion of total N derived from N2 fixation at maturity was higher in seeds than in vegetative plant parts and amounted to 59.5, 51.3 and 66.3 per cent of total above-ground plant N in the two pea cultivars and field bean, respectively (three-year means). The recovery of fertilizer N was 62.2, 70.2, 52.1, and 69.5 per cent in the two pea cultivars, field bean and barley, respectively. Growth analysis indicated that barley did not meet the claims for an ideal reference crop in the15N fertilizer dilution technique for estimating N2 fixation in pea and field bean. ‘Starter’-N neither increased the seed yield nor the N content of the grain legumes.  相似文献

The combined effect of Vesicular Arbuscular Mycorrhizae (VAM) and Rhizobium on the cold season legumes, lentil and faba bean, as well as on summer legume, soybean, were studied in soils with low indeginous VA mycorrhizal spores. Inoculation of the plant with VA mycorrhizal fungi increased the level of mycorrhizal root infection of lentil, faba bean and soybean. The inoculation with Rhizobium had no significant effect on VA mycorrhizal infection percent, but VA mycorrhizal inoculation increased nodulation of the three legumes. The inoculation with Rhizobium alone significantly increased plant dry weight and N content of lentil and faba bean as well as seed yield of soybean. VA mycorrhizal inoculation also significantly increased plant dry weight and phosphorus content of the plants as did fertilization with superphosphate. Rock phosphate fertilization, however, had no significant effect on plant growth or phosphorus uptake. The addition of rock phosphate in combination with VA mycorrhizal inoculation significantly increased plant dry weight and P uptake of the plants. The dual inoculation with both rhizobia and mycorrhizae induced more significant increases in plant dry weight, N and P content of lentil and faba bean as well as seed yield of soybean than inoculation with either VA mycorrhizae or Rhizobium alone.  相似文献

A field experiment was carried out to quantify biological nitrogen fixation (BNF) using the 15N isotope natural abundance method in maize (Zea mays L.)/faba bean (Vicia faba L.) and wheat (Triticum aestivum L.)/faba bean intercropping systems. Faba bean was yielding more in the maize/faba bean intercropping, but not in the wheat/faba bean intercropping. Biomass, grain yield and N acquisition of faba bean were significantly increased when intercropped with maize, and decreased significantly with wheat, irrespective of N-fertilizer application, indicating that the legume could gain or lose productivity in an intercropping situation. There was yield advantage of maize/faba bean intercropping, but no in wheat/faba bean intercropping. The grain yield of the faba bean intercropped with maize was greater than that of faba bean monoculture due to increases of the stems per plant and the pods per stem of faba bean. N fertilization inhibited N fixation of faba bean in maize/faba bean and wheat/faba bean intercropping and faba bean monoculture. The responses of different cropping systems to N-fertilizer application, however, were not identical, with competitive intercropping (wheat/faba bean) being more sensitive than facilitative intercropping (maize/faba bean). Intercropping increased the percentage of N derived from air (%Ndfa) of the wheat/faba bean system, but not that of the maize/faba bean system when no N fertilizer was applied. When receiving 120 kg N/ha, however, intercropping did not significantly increase %Ndfa either in the wheat/faba bean system or in the maize/faba bean system in comparison with faba bean in monoculture. The amount of shoot N derived from air (Ndfa), however, increased significantly when intercropped with maize, irrespective of N-fertilizer application. Ndfa decreased when intercropped with wheat, albeit not significantly at 120 kg N/ha. Ndfa was correlated more closely with dry matter yield, grain yield and competitive ratio, than with %Ndfa. This indicates that that total dry matter yield (sink strength), not %Ndfa, was more critical for the legume to increase Ndfa. The results suggested that N fixation could be improved by yield maximization in an intercropping system.  相似文献

Faba bean (Vicia faba L. cv. Fiord) plants were raised in agrowth room for 5 weeks and then transplanted to a hydroponicsystem. After 48 h for acclimation, nine plants were removed(day 0) for the measurement of nitrogenase activity by acetylenereduction (AR), for determination of nodule number and noduleweight, volume of the active N2 fixing region (VAR), and volumeof the senescent N2, fixing region (VSR). Half the nodule populationon a further 18 plants was excised, and nine of these plantswere assayed for AR. The nine plants from which the noduleshad been removed (treated plants) and nine control plants witha full complement of nodules, were left to grow for 5 d, afterwhich they were all harvested and assayed. The average weight of nodules and VAR remained constant in thecontrol plants between day 0 and day 5, whereas the nodulesleft on the treated plants increased in weight by 1.2 timesand VAR by 2.2 times. By day 5, VAR per plant was the same inthe control plants as in the treated, whereas VSR of controland treated plants increased by 4.6 and 2.2 times, respectively.Removal of half of the nodules at day 0 halved the AR activityper plant, but specific activity remained the same. After 5d, however, the nodules of the treated plants showed the sametotal activity as those of the control. Thus the specific activityof the nodules left on the treated plants doubled after 5 din response to excision. The indeterminate nodule of faba bean appears to be able toincrease its specific activity substantially in response toincrease in the demand for fixed N. N2 fixation per nodule wasresponsive to substrate supply, in that halving the number ofnodules on a plant induced the remaining nodules to increaseactivity, presumably because they could use the assimilate previouslydistributed over a larger number of nodules. Improvement of dinitrogen fixation in beans (Phaseolus vulgaris L.) will depend on the selection of superior plant genotypes and the presence of efficient rhizobial strains. This study was conducted to evaluate diverse bean lines for N2 fixation potential using the15N-depleted dilution technique under field conditions in Wisconsin, USA. Plants of 21 bean lines and three non-nodulating isolines of soybean received appliin Wisconsin, USA. Plants of 21 bean lines and three non-nodulating isolines of soybean received applications of15N-depleted ammonium sulphate. Shoots harvested at the V6, R3 and R7 stages and dry seeds were analyzed for total N using the Kjeldahl procedure, and the ratio of15N to14N was determind on a MAT 250 mass spectrometer. Nodule occupancy of the applied strain ofR. leguminosarum biovarphaseoli, CIAT 899, was determined in five of the bean lines. Total shoot N content showed a pattern of accumulation similar to shoot dry weight and fixed N2 in the shoot. Based on shoot total N, N2 fixed in the shoot and shoot dry weight Riz 30 and Preto Cariri were identified as being as good fixers as Puebla 152 and Cargamanto appear to begin N2 fixation early. Furthermore, some bean lines that fixed considerable N2 did not translocate a large amount of N to the grains. Preto Cariri accumulated 21.2 kg N ha−1 in the seeds compared to Puebla 152 which accumulated 43.8 kg N ha−1 of the fixed N2 into the grains. At the early sampling, Puebla 152 and 22–27 had a considerable higher percentage of their crown nodules formed by the inoculant strain CIAT 899, than did Rio Tibagi which has been considered a poor N2 fixer.  相似文献

Flavonoids produced by legume roots are signal molecules acting as nod gene inducers for the symbiotic rhizobium partner. Nevertheless, the changes of flavonoids in root exudates in intercropping system are still unknown. Based on pot experiment of faba bean and wheat intercropping, here we showed that faba bean and wheat intercropping increased the nodules number and dry weight, dry weight per nodule of faba bean compared with those found in monocropping, and the increase of faba bean nodulation was likely caused by the enhancement with flavonol, isoflavone, chalcone and hesperetin in its root exudates. It also promoted exudation of five types of flavonoids by wheat compared with monocropping. Our findings suggest that the flavonoids in root exudates have a positive effect on the nodulation and nitrogen fixation of faba bean in faba bean and wheat intercropping.  相似文献
