The poor growth of young Eucalyptus regnans seedlings in undried soil from the mature forest of E. regnans can be overcome by previously air-drying the soil or by adding sufficient amounts of complete soluble fertilizer or equivalent concentrations of P (as NaH2PO4) and N (as NaNO3). A factorial pot experiment in which phosphate and nitrate were added to undried soil indicated that P was the primary deficiency for young seedlings and that response to N did not occur until this lack was satisfied. In dried soil, seedlings also responded to additions of complete fertilizer but most of this effect was due to N rather than P. Field trials in the mature forest also indicated greater growth in dried soil than undried soil and confirmed a response of young seedlings to superphosphate. In pot experiments, the concentration of P and N per g plant dry weight after four months was relatively constant irrespective of the final size of the plant. Seedlings in dried soil extracted up to 15 times more P than did those grown in undried soil. In general, chemical analysis of soil indicated more extractable P and N from dried soil although this was not always consistently so. Soil desiccation resulted in an increase in soil surface area due to the fragmentation of larger peds and to an increase in the number of microfractures which remained in the soil crumbs after rewetting. Mycorrhiza are likely to be important since the differentiation of the growth response of seedlings in dried and undried soil, which occurred at 5–6 weeks, corresponded with the establishment of full ectomycorrhizal development (80% root tips). Lupins, canola, ryegrass and wheat fertilized with Na2 35SO4 and either 15NH4Cl or K15NO3(N:S=10:1), were grown in the field in unconfined microplots, and the sources of N and S (fertilizer, soil, atmosphere, seed) in plant tops during crop development were estimated. Modelled estimates of the proportion of lupin N derived from the atmosphere, which were obtained independently of reference plants, were used to calculate the proportion of lupin N derived from the soil. Total uptake of N and S and uptake of labelled N and S increased during crop development. Total uptake of S by canola was higher than lupins, but labelled S uptake by lupins exceeded uptake by canola. The form of N applied had no effect on uptake of labelled and unlabelled forms of N or S. Ratios of labelled to unlabelled S and ratios of labelled to unlabelled N derived from soil sources decreased during growth, and were less for S than for N for each crop at each sampling time. Although ratios of labelled to unlabelled soil-derived N were similar between crops at 155, 176 and 190 days after sowing, ratios of labelled to unlabelled S for lupins were higher than for the reference crops and declined during this period. The ratios of labelled to unlabelled S in lupins and the reference plants therefore bore no relationship either to ratios of labelled to unlabelled soil-derived N in the plants, or to total S uptake by the plants. Therefore the hypothesis that equal ratios of labelled N to unlabelled soil-derived N in legumes (Rleg) and reference plants (Rref) would be indicated by equal ratios of labelled to unlabelled S was not supported by the data. Abstract. The growth rate of Eucalyptus regnans seedlings in their first year can be much increased if the soil is first dried and then rewetted. The ratio of growth on predried soil to growth on undried soil (the Growth Ratio or GR) reaches a maximum at air-dryness (pF 6.0–6.4). In E. regnans forest soil, GR is greatest in humus-rich topsoil and declines with depth. The effect of air-drying persists for several months after rewetting when soil is stored under glasshouse conditions. It is largely unaffected by repeated drying and wetting, by the rate of drying or by the season of collection. The mixing of dried and undried soil or the placement of a layer of dried soil above undried soil produces an enhancement of growth proportional to the amount of dried soil added. Firing of a litter layer above soil at wilting point increases subsequent seedling growth to that in air-dried soil. The addition of ash from a litter fire to undried soil produces an increase in growth approximately equal to that caused by air-drying The drying effect is most pronounced in soils from mature E. regnans forest and nearby brackenland and is less in dense younger forest, frost-hollow grasslands and old grassy gaps in the mature forest. The effect is restored by the inoculation of E. regnans mycorrhizal roots from both dried and undried soil. The effect varies along an gradient from 500 to 1500 m a.s.l. and is a maximum in the wet E. regnans climatic zone and a minimum in zones or local aspects where forests are normally subject to frequent drying. The stimulatory effect on seedling growth in soils of the E. regnans zone may have an effect on the outcome of competition during regeneration in large gaps. Part of the growth responses previously ascribed to the ‘ash-bed’ effect may be due to the desiccation effect in these soils.  相似文献

In an unburnt, mature forest the germination of E. regnans on undisturbed bare ground is very poor in spite of adequate seed fall, and the presence of conditions suitable for germination over much of the year. No seed storage occurs in the top soil in spite of some temporary seed dormancy and the disturbance of the surface soil by earthworms and lyre birds. Seed is removed from the top soil by several ant species– Prolasius pallidus, P. brunneus, P. flavicornis and Chelaner leae –and is taken into nests. Removal of seed is more rapid and complete in summer than winter and is more vigorous in young forest than old. Observations on artificial nests in the field and laboratory indicate that seed is eaten and not stored for any length of time. The testae may be left intact, fragmented or moulded into crumbs with other material. Certain sugar-like substances, which are extractable in low concentrations from fresh seed, may be involved in the attractivity of seed to ants. The numbers of ant species in the forests vary with the micro-climate of the site, and in the mature understorey of Pomaderris aspera the activities of the three dominant species Iridomyrmex biconvexus, Prolasius pallidus and P, hrunneus are largely separated diurnally, seasonally, and spatially in the foraging areas of the litter layer. The density of the nests of seed-harvesting ants is high, particularly in bare areas of the forest floor. The total number of ants probably exceed 5–6 million/ha and is probably sufficient to effectively remove 60% of the sporadic seed fall of normal years. It is suggested that the success of germination of E. regnans after wild fires is not due to any specific stimulation but rather to a temporary interference of ant foraging activity and then to the saturation of their food requirements by a massive seed fall from canopy-stored capsules.  相似文献

Abstract We examined the effects of adding fertilizers on nutrient concentrations in foliage, and on species composition and density of the understorey in a 10 year old Mountain Ash (Eucalyptus regnans F. Muell.) forest. Nutrient concentrations in foliage of three woody understorey species (Acacia dealbata, Pomaderris aspera and Olearia argophylla) showed no significant response to the addition of phosphorus and nitrogen either alone, in combination or combined with other nutrients. The phosphorus concentration in foliage and stems of two herbaceous species (Australina pusilla and Urtica incisa) was significantly increased by applying phosphorus. The addition of fertilizers had no significant effect on species composition and density of the understorey.  相似文献

王桉 (Eucalyptus regnans F. Muell.) 是澳大利亚桉树中最重要的商业用材和人工造林树种之一.研究王桉的施肥与其体内氨基酸的积累和转化及与食叶虫害之间的相关性具有重要的经济和生态意义.在温室内利用2种不同来源的土壤对王桉幼苗进行了不同磷施用量(100 kg hm~(-2)和 200 kg hm~(-2))处理.结果显示,不同土壤和不同磷施用量对苗木生长影响显著,但均未显著影响苗木各部分的氮和磷含量水平.苗木木质部渗出液中的氨基酸含量以谷氨酰胺为主,并与苗木生长和磷施用量呈反相关.不同土壤和磷施用量对苗木组织中游离氨基酸组分和含量的影响不显著,但游离氨基酸的组分和相对水平随叶龄变化明显,尤其是精氨酸在嫩叶氨基酸总量中只占2%~3%,但在老叶中占到20%多;精氨酸在老叶中的积累极有可能是某些蛋白质降解而精氨酸即时合成所致,因为精氨酸一般不在韧皮部转运.谷氨酰胺在树液中含量最高并与苗木生长呈反相关或许可以作为预测桉树发生食叶昆虫危害的一个有用指标.  相似文献

This study was conducted to compare the effects on the growth of Eucalyptus regnans seedlings of unheated soil and soil heated to different extents (as indicated by soil colour–bright red or black) in burnt logging coupes, and to separate the effects of heating of the soil on direct nutrient availability and on morphotypes and effectiveness of ectomycorrhizae. Burnt soils were collected from three logging coupes burnt 2, 14 and 25 months previously and unbumt soil from adjacent regrowth forests. Compared to unburnt soil, the early seedling growth was stimulated in black burnt soil from all coupes (burnt 2, 14 and 25 months previously). Seedling growth was generally poor in red burnt soil, especially in soil collected 2 months after burning. However, the concentration of extractable P was extremely high in red burnt soil, especially in soil collected 2 months after burning. In black burnt soil, extractable P was increased in soil 2 months after burning, but not in the soils collected 14 or 25 months after burning. However, both total P content and concentration in seedlings were increased in all collections of black burnt soil. Frequency of ectomycorrhizae was high in seedlings grown in all black burnt soils, but the mycorrhizal mantles were poorly developed in seedlings in black burnt soil collected 2 months after burning. Seedlings were also ectomycorrhizal in red burnt soil, except in soil collected 2 months after burning. Fine root inocula from seedlings grown in black burnt soils collected 14 and 25 months after burning significantly stimulated both seedling growth and P uptake compared with the uninoculated control, whereas the fine root inocula from the seedlings grown in all the other soils did not. These results suggest that, in black burnt soil, both direct nutritional changes and changes in the ectomycorrhizae may contribute to seedling growth promotion after regeneration burns. The generally poor seedling growth in red burnt soils is likely to have been due to N deficiency as the seedlings in these soils were yellow-green and the tissue concentrations of N were significantly lower than in other treatments. Relatively little is known about changes in leaf attributes over the lifespan of woody plants. Knowledge of such changes may be useful in interpreting physiological changes with age. This study investigated changes in leaf morphology and anatomy with tree age and height in the broadleaved evergreen species, Eucalyptus regnans. Fully expanded leaves were sampled from the upper canopy of tree ages ranging from 6 to 240 years, and tree heights ranging from about 10–80 m. There were significant changes in leaf form with increasing tree age and height. Leaf size and specific leaf area (SLA; leaf area/leaf mass) decreased, leaf thickness increased, and leaves became narrower relative to their length, with increasing tree age and height. Cuticle thickness and leaf waxiness, including wax occlusion of the stomatal antechamber, increased with increasing age and height. By comparison, there were no clear trends in stomatal frequency or stomatal length with tree age, although there were curvilinear relationships between an index of total stomatal pore area per leaf lamina and both tree age and tree height. The results support the hypothesis that leaves of E. regnans become more xeromorphic with tree age and height. The results are discussed in relation to their significance for changes in water relations in the canopy with age.  相似文献

Spindles of CUBA 87-51 sugarcane were cultured in Murashige and Skoog (MS) basal medium and supplemented with different nutrients. Embryogenic and non-embryogenic callus obtained were comparatively studied by scanning electron microscopy (SEM). Samples of embryogenic callus cultured in regeneration medium (MS without 2.4 dichlorophenoxyacetic acid) were taken at different times for analyzing the sequential process. Distinctive features of two types of callus are shown by SEM: cells organized in embryos are noted in embryogenic callus; while elongated, disorganized cells can be seen in non-embryogenic callus. The characteristics of the embryos during plant regeneration are described. Sugarcane embryoid stages are: globular, globular with lateral notch and scutellum. In this process also appear shoot meristems, leaf and root primordia and finally, true leaves and roots. It is concluded that callus plant regeneration from young leaf segments of sugarcane mainly occur via somatic embryogenesis.  相似文献

Seed morphology in the genus Aeschynanthus (Gesneriaceae) is important in sectional classification. The number and type of appendages at the hilar end of the seed, papilla structure and testa cell orientation are all significant. SEM studies of seed and appendage development, at seven-day intervals from pre-pollination to maturity, were carried out in order to investigate possible sectional and seed type relationships. Fifteen species, representing six sections, were examined. Results show that there are no significant differences pre-pollination but two patterns were evident post-pollination, for both seed and hilar appendage development. Pattern I occurred in sects Microtrichium and Haplotrichium s.s . Pattern II was seen in sects. Aeschynanthus , Diplotrichium and Polytrichium . Patterns were less clear in species belonging to a group not yet circumscribed and here referred to as sect. X , but most closely resembled Pattern II. This study shows that the orientation of the testa cells is a developmental feature, defining the two major clades in the genus. It also shows that, in the two sections possessing more than one hilar appendage, there is initial development of one appendage relative to the coma in sect. Polytrichium and of one appendage relative to the second in sect. Diplotrichium . Water movement between a root and the soil depends on the hydraulic conductances of the soil, the root, and the intervening root-soil air gap (Lgap) created as roots shrink during soil drying. To measure Lgap, segments of young cylindrical roots of Agave deserti about 3 mm in diameter were placed concentrically or eccentrically within tubes of moistened filter paper at a known water potential. As the width of the air gap between the filter paper and a concentrically located root was made smaller, measured Lgap increased less than did predicted Lgap based on isothermal conditions. For gaps of the size expected in the soil during water loss from roots (e.g., 10% of the root radius), the underprediction was about 70% and was primarily caused by a lowering of the root surface temperature accompanying water evaporation. As a root segment was eccentrically moved toward the filter paper, the measured Lgap increased. For the most eccentric case of touching the filter paper, the measured Lgap was 2.4-fold greater than for the concentric case, compared with an infinite Lgap predicted if the water potential were constant around the root surface. When a root touched soil with a water potential of -1.0MPa, Lgap estimated using a graphical method increased about 2.3-fold and the overall conductance of the root-soil system increased by 31% compared with the concentric case. For markedly eccentric locations of roots in air gaps, Lgap, which can be the principal conductance initially limiting water loss from roots to a drying soil, can be about 60% of the value predicted for the concentric isothermal case.  相似文献

Shikimic acid was determined in leaves, at different stagesof growth, from 1- and 4-year-old specimens of Eucalyptus sieberianaF. Muell. A linear relationship was found between the shikimicacid concentration (mg./g.) and the weight of the leaf up tothe point where the leaf had reached full size. Cells from 25 cases of acute lymphoblastic leukemia (ALL) were studied under the scanning electron microscope (SEM). In 24 of the cases, the vast majority of circulating leukaemic cells had few microvilli. Villous cells were rarely encountered and prominent ridge-like profiles and ruffled membranes were not seen. Only six cases were studied by immunological techniques and four of the cases were of the null type while in two the cells bore detectable T-markers. It seems that ALL is almost always associated with the presence of cells with few microvilli in the peripheral circulation, differing in this respect from most cases of CLL. Although circulating leukaemic lymphocytes with few microvilli are sometimes seen in CLL, the most frequent cell type encountered is a more villous lymphocyte. Differences between leukaemic cells from patients with ALL, CLL and non-lymphoblastic leukaemias are discussed. It appears that SEM may help to distinguish lymphoblastic and nonlymphoblastic leukaemic cells in many instances and can be used as a useful adjunct to other modes of microscopy in the diagnosis of acute leukaemia.  相似文献

The extracellular matrix of unfixed, unstained rat corneal stroma, visualized with high-resolution scanning electron microscopy and atomic force microscopy after minimal preliminary treatment, appears composed of straight, parallel, uniform collagen fibrils regularly spaced by a three-dimensional, irregular network of thin, delicate proteoglycan filaments. Rat tail tendon, observed under identical conditions, appears instead made of heterogeneous, closely packed fibrils interwoven with orthogonal proteoglycan filaments. Pre-treatment with cupromeronic blue just thickens the filaments without affecting their spatial layout. Digestion with chondroitinase ABC rids the tendon matrix of all its interconnecting filaments while the corneal stroma architecture remains virtually unaffected, its fibrils always being separated by an evident interfibrillar spacing which is never observed in tendon. Our observations indicate that matrix proteoglycans are responsible for both the highly regular interfibrillar spacing which is distinctive of corneal stroma, and the strong interfibrillar binding observed in tendon. These opposite interaction patterns appear to be distinctive of different proteoglycan species. The molecular details of proteoglycan interactions are still incompletely understood and are the subject of ongoing research.  相似文献
