We investigated the effect of competition from grass roots (as controlled by herbicide application) on the depth distribution of white roots in cherry trees, grown with varying rates and frequency of application of N in an agroforestry system. Statistical summaries of distribution, namely mean and skewness, produced a concise and interpretable analysis of the data.There was a large increase in tree root numbers in the surface horizons after the herbicide had reduced grass root competition. Where the surrounding grass had not been reduced by herbicide, the average depth of tree roots increased with time, contrasting with a marked shift in the mode of distribution of root numbers to shallow depths when grass competition was removed. These findings are important in the understanding of plant root competition and for prescribing best practise for tree establishment in agroforestry systems.  相似文献

Levels of endogenous growth substances (abscisic acid: ABA; indole-3-acetic acid: IAA) and applied benzyladenine (BA) were quantified during the eight first days of in vitro propagation of Wild Cherry (Prunus avium L.). Axillary buds from the middle part of the explants started to grow at day 2, thus were released from apical dominance. Hormone levels were measured in the apical, median and basal parts of the explants using an avidin-biotin based enzyme-linked immunosorbent assay (ELISA) after a purification of the extracts by high performance liquid chromatography (HPLC). All hormones showed rapid and considerable changes during the first eight days of growth. Exogenous IBA was probably transformed into IAA mainly in the basal part of the explant, and BA penetrated quickly. ABA levels were transiently enhanced in the apical part of the explants bearing young leaves. These phenomena are discussed in connection with the axillary bud reactivation.  相似文献

Regeneration of adventitious shoots from leaves and, for the first time, from internode sections were compared and optimized for five economically important sweet cherry cultivars, i.e. Schneiders, Sweetheart, Starking Hardy Giant, Kordia and Regina (Prunus avium L.). The influence of basal media, carbon source, combination and dosage of phytohormones, ethylene inhibitor such as silver thiosulfate and a 16 h:8 h light:dark photoperiod versus complete darkness were evaluated. Both, DKW/WPM (1:1) and Quoirin/Lepoivre (QL) basal media stimulated organogenesis more than QL/WPM (1:1), Chee and Pool (CP), Murashige Skoog (MS), Driver and Kuniyuki (DKW) or woody plant (WPM) media did. An induction phase in darkness resulted in lower or zero regeneration rates. The best regeneration efficiencies were generally obtained with thidiazuron in combination with indole-3-butyric-acid. The addition of silver thiosulfate resulted in a similar or reduced regeneration efficiency. Significant genotypic variability in adventitious bud formation was evident for both explant sources, leaf and internode section. Adventitious shoots were obtained from 11% of leaf explants and 50% of internode sections indicating that shoot regeneration from internodes was significantly more efficient than from leaves.  相似文献

Shoot regeneration was achieved from leaves of in vitro cultures of Prunus avium L. cv. 'Lapins' and 'Sweetheart' using woody plant medium (WPM) supplemented with 1-naphthalene-acetic acid (NAA) and thidiazuron (TDZ) or benzyladenine (BA). Percent regeneration was influenced by plant growth regulators and by explant type, orientation and wounding. Optimal regeneration was observed with whole-leaf explants wounded by transverse cuts along the midrib and incubated abaxial surfaces uppermost, on media supplemented with 2.27 or 4.54 µM TDZ plus 0.27 µM NAA. The percent regeneration of the two cultivars was not significantly different. Optimum conditions for regeneration resulted in 71.4% of 'Lapins' and 54% of 'Sweetheart' explants producing one or more shoots per explant.  相似文献

Growth and nutrient utilization of alfalfa (Medicago sativa L. cv. Arc) and common bean (Phaseolus vulgaris L. cv. Carioca) were studied in an acid soil adjusted to eight levels of soil acidity by lime addition. Application of lime significantly (P<0.05) increased shoot and root growth for both species. However, common bean was far less sensitive to soil acidity than alfalfa. Maximum alfalfa growth was obtained at a soil pH of 5.8 and maximum bean growth was achieved at pH 5.0. Root and shoot growth of both legumes was positively correlated (P<0.01) with soil pH, exchangeable Ca and exchangeable Mg and negatively correlated (P<0.01) with soil exchangeable Al. Common bean had a lower internal P requirement for maximum growth and was more efficient than alfalfa in taking up Ca and Mg. These characteristics would contribute to the favorable growth of common bean in acid-infertile soils.  相似文献

Antibodies to phase related proteins in juvenile and mature Prunus avium   总被引:1,自引:1,他引:0  
In the search for biochemical and molecular markers of juvenility in trees proteins have been identified which are preferentially or differentially expressed in either the juvenile or the mature phases of Prunus avium cv. Stella. These fall into two classes: those which are phase-related and those which may be affected by root-shoot distance. N-terminal amino acid sequence data from some of these proteins were used to produce polyclonal antibodies to corresponding synthetic peptides in order to determine if they could be used as markers of phase state in woody plants. Western blot analysis was performed on proteins extracted from three sources; juvenile trees, mature trees and rooted cuttings from mature trees. The results showed that the antibodies recognised differentially-expressed proteins. In particular, one antibody to a juvenile specific protein cross-reacted with a polypeptide of approx. 28 kDa which was present in greater amounts in shoot tips of juvenile P. avium cv. Stella seedlings compared with rooted cuttings of mature plants placed in the same growth environment.  相似文献

The contribution of indigenous arbuscular mycorrhizal fungi (AMF) to growth and phosphorus (P) uptake by oilseed flax (Linum usitatissimum L.) was examined in two field experiments covering soil P levels from 20–86 mg kg-1 NaHCO3-extractable P. The fumigant dazomet was applied to the soil in half of the plots to obtain control plants with reduced mycorrhiza formation. An extensive AMF colonization of up to 48% of the root length was established in untreated soil of both experiments, although P fertilization reduced colonization to 28–39% at the latest harvests. Fumigation markedly decreased or totally prevented AMF colonization throughout the experiments. Root growth responded to fumigation by increased total and specific root length. Shoot P uptake was decreased by fumigation at soil P levels lower than ca. 50 mg kg-1 whereas shoot growth was reduced by fumigation at soil P levels lower than ca. 40 mg kg-1. The effects of fumigation were ascribed to the suppression of mycorrhiza formation. The effect of the AMF increased with decreasing soil P levels. Phosphorus inflow through roots (based on shoot P uptake) was reduced more strongly by fumigation than total P uptake. The P inflow through fungal tissue in roots was estimated to 4 × 10-14 mol P cm-1 s-1. We conclude that AMF are essential to flax growth at soil P levels below ca. 40 mg P kg-1, which is representative of the conditions under which most flax is grown.  相似文献

A study of the polyamine profile was carried out during zygotic embryo development in Prunus avium. Zygotic embryos were collected from 2 donor trees and sorted into 3 size classes: C1 [2.5 to 3.5 mm]; C2 [3.6 to 4.5 mm] and C3 [5.5 to 7 mm]. Evolution of the various polyamines was similar for the two donor trees. Changes in the relative amount of the various free polyamines were observed during zygotic embryo development. Agmatine and spermine levels increased from C1 to C3. Spermidine, the predominant polyamine, showed a two-fold decrease in C3 compared with C1 and C2; the evolution of putrescine was opposed, showing an increase in the last developmental stage. The putrescine/spermidine ratio could be a marker for these 3 developmental stages with a higher ratio in C3 compared with C1 and C2. Polyamine changes in cotyledons from class C1 were investigated during in vitro culture. A 10-day induction on a medium containing 2,4-dichlorophenoxyacetic acid (2,4-D) and kinetin caused a strong decline in free spermidine levels and a dramatic increase in free putrescine. When applied to spurs of mature Prunus avium before floral initiation, gibberellins GA1, GA4 and GA3 inhibited floral initiation by 9–17%, GA7 by 43%, GA3 by 65–71% and 2,2-dimethyl GA4 by 78%. GA9 and GA20 were inactive. Thus activity only of the GAs with a C-3 hydroxyl was increased markedly by a double bond in the C-1,2 or C-2,3 position, and activity increased with increasing hydroxylation. None of the GAs affected the total number of buds (vegetative and floral) surviving in the spur. Measured by the threshold dose required for activity, seedling shoot growth responses to GA3, GA7, GA1 or GA4 resembled those of floral initiation, but di-methylation of GA4 at C-2 had no effect, and GA9 was as active as GA7. Mature shoots, including those on rooted cuttings, were less responsive to GA treatment than were juvenile shoots, with terminal shoots on mature trees more responsive than spur shoots. Spur shoot growth on mature trees responded to GA3 and to a lesser extent GA7, but not to GA1 or GA4. However, all these GAs promoted the growth of terminal shoots on mature trees to similar extents, whereas 2,2-dimethyl GA4 was less active than GA4 The differences between juvenile and mature shoot growth in sensitivity to a C-1,2 or C-2,3 double bond, and between mature shoot growth and floral initiation in GA-structure requirements, indicate that phase change alters the GA complement and/or GA receptor/transduction mechanisms of P. avium. The difference in sensitivity to 2,2-dimethyl GA4 indicates that floral initiation and growth have different requirements for GA transport and/or action.  相似文献

The objective of this study was to investigate the effect of cyclic soil wetting and drying on maize (Zea mays L.) root hair growth. Three soils, Chalmers silty clay loam (Typic Haplaquolls), Raub silt loam (Aquic Argiudolls) and Aubbeenaubbee sandy loam (Aric Ochraqualfs) and two soil moisture contents, −175 (M0) and −7.5 kPa (M1), were used to study root hair growth in a controlled-climate chamber. Increasing soil moisture after 7d from M0 and M1 resulted in a cessation of root hair growth behind the root cap while drying the soil after 7d from M1 and M0 promoted root hair growth on new but not old or existing roots. By maintaining liquid continuity under cyclic wetting and drying of a soil, root hairs may be of far greater significance to the nutrition of the plant than originally thought. The effects of decomposing peach root residues in soil on peach growth were determined in two pot experiments. In the first, peach root residues, despite their high C:N ratio and lignin content, largely decomposed under experimental conditions, leading to an immobilization of inorganic N. Shoot growth of peach seedlings was depressed by the addition of peach root residues, an effect that depended on their size and concentration: fine-textured root fragments (0.45–1.00 mm) resulted in more severe effects than medium sized ones (2–8 mm), while growth depression occurred only at concentrations higher than 0.35%. Peach root growth was depressed by root residues regardless of their size and amount. In the second experiment, where nitrogen was added to all pots to minimize the effects of immobilization of N during decomposition of root fragments, the growth of peach roots in residue-supplemented soil almost stopped. Pre-planting phosphate enrichment was very effective in stimulating growth of peach in virgin soil but did not offset the depression caused by peach root residues. It thus appears that besides mineral deficiencies, there exist alternative explantations of poor growth of peach in replant soils, including growth-inhibiting substances from decomposing root residues.  相似文献

The effects of nitrogen form and phosphorus source on the growth, nutrient uptake and rhizosphere soil property of tea (Camellia sinensis L.) were investigated in a pot experiment. The experiment was performed with a compartmental cropping device, which enables the collection of rhizosphere soil at defined distances from the root of tea plant. Nitrogen was supplied as nitrate or ammonium in combination with soluble phosphorus as Ca(H2PO4)2 or insoluble P as rock phosphate. The leaf dry matter production of tea was significantly greater in the treatments with NH4 + than NO3 -, whereas dry matter production of root and stem was not significantly affected. Addition of phosphorus as either source did not influence the dry matter production. The concentrations of K in root, Mg and Ca in both the shoot and root supplied with NO3 - were significantly higher than in NH4 + and influence of P sources was minor. On the contrary, Al and Mn concentrations were significantly larger in NH4 --fed plants which could be attributed to remarkably increased availability of Al and Mn caused by acidification of the rhizosphere soil (the first 1-mm soil section from the root surface) with NH4–N nutrition. The concentration of N in shoot was also significantly higher in NH4- than in NO3-fed plants, indicating higher use efficiency of NH4–N. Whatever the phosphate source, rhizosphere pH declined in ammonium compared to in nitrate treatment. The pH decrease was much larger when no P or soluble P were applied and reached 0.85–1.30 units which extended to 3–5 mm away from the root surface. Exchangeable acidity, content of exchangeable Al and Mn were also considerably higher in the rhizosphere soils of NH4 + fed tea plants. Significant amounts of P dissolved from rock phosphate accumulated in rhizosphere of NH4 +, not NO3 -, suggesting that the dissolution of rock phosphate was induced by the proton excreted by tea root fed with ammonium. With soluble P addition, shoot and root P concentrations were greater in NH4 + than in NO3 - treatment and it appeared that this difference could not be sufficiently explained by the available P content in soil which was only slightly higher in NH4 + treatment. With rock phosphate addition, the shoot and root P concentrations were hardly affected by nitrogen form, although the available P content was much higher and accumulated in the rhizosphere soil supplied with ammonium. Field and growth chamber studies were used to determine the effect of in-furrow application of PGR-IV on root and shoot development, and yield of cotton. In the field study, an in-furrow application of PGR-IV @ 73 mL ha–1 at planting increased yield by 18% compared to the untreated control, and by 11% compared to 2-foliar applications of 292 mL/ha–1 each at pinhead square stage of flower development and at first flower appearance. Growth chamber studies revealed that the in-furrow applications of PGR-IV @ 1.131L/plant dramatically increased root length (+47%), root dry weight (+29%), number of lateral roots per plant (+75%), and nutrient uptake one week after planting. These differences were still apparent five weeks later at pinhead square but to a lesser degree. The yield enhancement from the foliar applications was associated with increases in leaf growth, nutrient uptake, and boll number, whereas the yield enhancement from the soil application was associated with enhanced root growth and nutrient uptake. The positive effect of PGR-IV on root growth and accelerated early-season growth could have very substantial benefits in cotton production.  相似文献

Antiserum to a protein fraction of an extract of mature styles of P. avium cv. Lambert (S 3 S 4) was raised in rabbits. Two major antigenic components of the style extracts were detected by immunoelectrophoresis and immunodiffusion. The presence of one antigen (S-antigen) correlated with a particular S-genotype (S 3 S 4). This antigen is restricted to mature styles of P. avium. The second antigen (P-antigen) was detected in styles of all Prunus species examined, but not in styles of other species of the Rosaceae. The S-antigen is positively charged and the P-antigen negatively charged at pH 8.8.  相似文献

Soil compaction is a widespread cause of reduced plant productivity. If the effects of soil compaction on plant growth are to be reproduced in simulation models, then the processes through which compaction reduces root elongation must be expressed mathematically and then tested against experimental data. The mathematical theory by which these processes may be represented is given in the accompanying article. In this article, the behavior of a simulation model based on this theory is tested against data for root growth and soil gas concentration recorded from soil columns of which the middle layers were compacted to different bulk densities. The model was able to reproduce the failure of the root system to penetrate the compacted middle layer within the period of the experiment when bulk density exceeded 1.55 Mg m-3. The model also reproduced decreases in O2 concentrations, and increases in CO2 concentrations, in the atmospheres of the compacted layer and of the uncompacted layer below it as bulk density of the compacted layer increased. The simulated time course of O2 and nutrient uptake and of O2 concentrations in the compacted layer at different depths is presented and its consistency with experimental findings is examined. As part of a larger ecosystem model, this model will be useful in estimating site-specific effects of soil compaction on carbon cycling in agroecosystems.  相似文献

Hairy roots were obtained following inoculation of the stems of Lobelia inflata L. with Agrobacterium rhizogenes strain ATCC 15834. These hairy roots contained agropine and mannopine. In addition, lobeline was detected by HPLC and confirmed by mass spectrometry. Various media were tested for the growth of hairy roots as well as for the content of lobeline in hairy roots. The growth rate of hairy roots cultured in Nitsch and Nitsch's medium was approximately one third of those cultured in other media. The lobeline content of hairy roots (18–54 g/g dry weight) cultured in these media was the same order of magnitude compared with that of roots of L. inflata (24 g/g dry weight) cultivated in pots. The growth of carrot (Daucus carota L.) seedlings in clear polystyrene vials (7.5 × 2.5 cm diameter) which were either perforated or unperforated and suspended in a large container containing nutrient solution was compared with that of similar seedlings grown directly in the main container. After 28 days plants grown in the perforated, aerated vials had smaller shoots, tap roots (c 20 percent reduction) and fibrous roots (c 60 percent reduction) than unrestricted plants. In the absence of adequate aeration, the effects of root restriction were greater. Foliar applications of ethephon to the leaves of unrestricted plants reduced shoot weights but had little effect on either tap root or fibrous root weights, or even increased tap root weight at lower concentrations. Silver thiosulphate, aminovinylglycine and a highly mobile cytokinin (SD8339) all slightly enhanced shoot growth of restricted plants but had no effect on either tap roots or fibrous root development. One-year old nectarine trees [Prunus persica, Batsch var. nectarina (Ait.) Maxim.], cv Nectaross grafted on P.S.B2 peach seedlings [Prunus persica (L.) Batsch] were grown for five months in 4-litre pots filled with two alkaline soils, one of which was also calcareous. Soils were regularly subjected to fertigation with either ammonium sulphate or calcium nitrate providing a total of 550 mg N/tree. Trees were also grown in such soils receiving only deionized water, as controls. Rhizosphere pH, measured by the use of a microelectrode inserted in agar sheet containing a bromocresol purple as pH indicator and placed on selected roots, was decreased by about 2–3 units compared to the bulk soil pH in all treatments. This decrease was slightly less marked when plants were supplied with calcium nitrate rather than ammonium sulphate or control. Measurements conducted during the course of the experiment indicated that ammonium concentration was similar in the solution of soils receiving the two N fertilizers. During the experiment, soil solution nitrate-N averaged 115 mg L–1 in soil fertilized with calcium nitrate, 68 mg L–1 in those receiving ammonium sulphate and 1 mg L–1 in control soils. At the end of the experiment nitrate concentrations were similar in soils receiving the two N sources and bulk soil pH was decreased by about 0.4 units by ammonium sulphate fertigation: these evidences suggest a rapid soil nitriflcation activity of added ammonium. Symptoms of interveinal chlorosis in apical leaves appeared during the course of the experiment in trees planted in the alkaline-calcareous soil when calcium nitrate was added. The slightly higher rhizosphere pH for calcium nitrate-fed plants may have contributed to this. The findings suggest that using ammonium sulphate in a liquid form (e.g. by fertigation) in high-pH soils leads to their acidification and the micronutrient availability may be improved.  相似文献

Cores of repacked soil were consolidated with a compressive strength testing machine, after peas had been planted in the centre of the core. The number that emerged were counted and root and shoot lengths and diameters were measured. Consolidation had no effect on emergence, root length or root diameter of the peas grown in a loamy sand, whereas emergence, root length and root diameter were affected by a small increase in load in a clay loam.  相似文献
