目的:应用随机引物扩增多态性DNA技术( random amplified polymorphic DNA , RAPD)对大耳白黑眼兔( white hair black eyes rabbit , WHBE rabbit )、日本大耳白兔( Japanese white rabbit , JW rabbit )和新西兰兔(New Zealand white rabbit, NZW rabbit)3个实验兔品系进行遗传分析。方法选用90只实验兔的皮肤组织样品提取基因组DNA,用60个随机引物对实验兔基因组DNA进行PCR扩增,根据电泳结果筛选出多态性较高的引物进行RAPD-PCR分析,再利用Popgene 3.2统计软件对3个品系的扩增条带进行遗传分析,获得实验数据。结果分析结果表明:(1)60个随机引物中筛选出25个多态性较高的引物,3个品系实验兔共检测到493个扩增片段,长度在100~1800 bp之间,筛选的25个引物中,其中16个引物既可扩增出3个品系共同的DNA条带,也可扩增出WHBE兔特有的特征条带;(2) WHBE兔位点数为234个,其中多态位点数166个,多态位点比为70.94%,JW兔位点数为228个,其中多态位点数122个,多态位点比为53.51%,NZW兔位点数为231个,其中多态位点数94个,多态位点比为40.69%;(3)三个群体的Shannon多样性指数分别为0.3385,0.2222和0.1905;(4) JW兔和NZW兔的遗传相似系数最高,为0.8443,其次为WHBE兔和JW兔的遗传相似系数,为0.8204,WHBE兔和NZW兔的遗传相似系数最低,为0.7862。结论结果表明WHBE兔与JW兔和NZW兔之间有遗传的相似性,也存在着遗传差异,应用RAPD技术可以很好地检测实验兔不同品系之间以及同一品系不同个体之间的亲缘关系。  相似文献

Between weaning and adulthood, the length and height of the facial skull of the New Zealand rabbit (Oryctolagus cuniculus) double, whereas much less growth occurs in the width of the face and in the neurocranium. There is a five-fold increase in mass of the masticatory muscles, caused mainly by growth in cross-sectional area. The share of the superficial masseter in the total mass increases at the cost of the jaw openers. There are changes in the direction of the working lines of a few muscles. A 3-dimensional mechanical model was used to predict bite forces at different mandibular positions. It shows that young rabbits are able to generate large bite forces at a wider range of mandibular positions than adults and that the forces are directed more vertically. In young and adult animals, the masticatory muscles differ from each other with respect to the degree of gape at which optimum sarcomere length is reached. Consequently, bite force can be maintained over a range of gapes, larger than predicted on basis of individual length-tension curves. Despite the considerable changes in skull shape and concurrent changes in the jaw muscles, the direction of the resultant force of the closing muscles and its mechanical advantage remain stable during growth. Observed phenomena suggest that during development the possibilities for generation of large bite forces are increased at the cost of a restriction of the range of jaw excursion.  相似文献

Normative data of visual evoked potentials were obtained from occipital screw electrodes referenced to a nasal electrode in 10 white New Zealand rabbits. Two time-bases were used (200 and 400 ms) in order to reliably identify both early components (of retinal and cortical origin), as well as late components. The putative origin of the single components was established by simultaneous recording of ERG and by stereotaxic recordings from the lateral geniculate body. Test-retest variability was measured repeating the recordings after 24 hours.  相似文献

Efficiency of the New Zealand white rabbit as a host for infecting larval Ixodes dammini, I. pacificus, and I. ricinus with Lyme disease spirochetes was evaluated. Rabbits inoculated with infected midgut suspensions of I. dammini from Shelter Island, New York, or fed upon by infected ticks from the same area, responded with spirochetemias of sufficient concentrations to infect as many as 30 percent of the ticks. When infected ticks were used as indicators, it appeared that spirochetemias persisting for up to ten days occurred as early as the tenth day after inoculation or feeding of ticks.  相似文献

The effect of castration and testosterone propionate (TP) treatment on the motor copulatory pattern was studied by an accelerometric technique in five sexually experienced New Zealand white male rabbits. This technique permits the oscillographic recording of thrusting frequency, rhythmicity, and amplitude of the pelvic movements occurring during copulation. Castration resulted in a marked decrease in sexual activity in all rabbits. Mounting, including occasional intromissions, was retained by four of the rabbits for periods ranging from 2 to 15 weeks. Castration did not affect mount duration, but decreased strength and frequency of pelvic thrusting. Diminution in the frequency of pelvic thrusting was mainly due to intercalation of pauses within the mounting trains. The change in the rhythm of pelvic thrusting was related to the failure of most mounts performed by castrated rabbits to stimulate lordosis in the female. Testosterone propionate (TP, 10 mg daily for 15 days) restored mounting activity and increased strength and frequency of pelvic thrusting in all rabbits. It is concluded that TP, besides stimulating sexual motivation, regulates the vigor and rhythm of pelvic movements during copulation in the rabbit.  相似文献

In conscious animals, the response to hemorrhage is biphasic. During phase 1, arterial pressure is maintained. Phase 2 is characterized by profound hypotension. Despite allied roles, less is known about the integrated cardiovascular and respiratory response to blood loss in conscious animals. We evaluated cardiorespiratory changes during hemorrhage to test the hypotheses that 1) respiratory rate (RR) and blood gases do not change during phase 1; 2) RR increases during phase 2; and 3) RR and blood gas changes during hemorrhage are similar in males and females. We measured mean arterial pressure, RR, and blood gases during hemorrhage in 16 conscious, chronically prepared, male and female New Zealand white rabbits. We removed venous blood until mean arterial pressure was < or =40 mmHg. Sex did not affect mean arterial pressure, heart rate, Pa(O(2)), Pa(CO(2)), or pH during hemorrhage or the blood loss required to induce phase 2. Pa(CO(2)) decreased significantly from 37 +/- 1 to 33 +/- 1 and 29 +/- 1 mmHg (P < 0.001) during phase 1 and 2, respectively. Before hemorrhage, Pa(O(2)) was 87 +/- 2 mmHg. Pa(O(2)) was unchanged in phase 1 (92 +/- 2 mmHg) but increased in phase 2 (101 +/- 2 mmHg; P < 0.001). Body temperature, Pv(CO(2)) (thoracic vena cava), and ventilation-perfusion mismatch (A-a gradient) were unchanged during phases 1 and 2. Neither sex increased RR during phase 1. While males doubled RR during phase 2, RR in females did not change (P < 0.001). Thus, while Pa(CO(2)) decreases in phase 1 and phase 2, the decreases are achieved in different ways across the two phases and in the two sexes.  相似文献

Large trees are a significant structural component of old-growth forests and are important as habitat for epiphytic biodiversity; as substantial stores of biomass, carbon and nutrient; as seed trees; and as engineers of large gap sites for regeneration. Their low density across the landscape is an impediment to accurately measuring growth and mortality, especially as infrequent tree deaths are rarely captured without long periods of monitoring. Here we present large-tree (≥ 30 cm in diameter at breast height) growth and mortality rates for six common New Zealand tree species over a 42-year period from 28 large permanent plots (0.4?0.8?ha) in the central North Island. Our goal was to examine how rates of growth and mortality varied with tree size and species. In total we sampled 1933 large trees across 11.6 ha, corresponding to a large-tree density of 167 trees?ha?1, of which we used 1542 as our six study species. Mean annual mortality rates varied more than 10-fold among species being least in Dacrydium cupressinum (0.16%) and greatest in Weinmannia racemosa (2.21%). Diameter growth rates were less variable among species and ranged from 1.8 mm?yr?1 in Ixerba brexioides to 3.3?mm?yr?1 in D.?cupressinum. Tree size influenced the rate of mortality in Beilschmiedia tawa, I. brexioides and W.?racemosa but there was no support for including tree size in models of the remaining three species. Likewise, tree size influenced growth rates in I.?brexioides and Nothofagus menziesii but not the remaining four species. These data provide robust size- and species-specific estimates of large-tree demographic rates that can be used as baselines for monitoring the impacts of management and global change in old-growth forests.  相似文献

Off the eastern coast of New Zealand, warm, saline, nutrient-poorSubtropical Waters (STW) are separated from cool, fresher, relativelynutrient-rich Sub-Antarctic Waters (SAW) by the SubtropicalConvergence (STC). The Chatham Rise, a submarine rise, restrictsthe latitudinal movement of the STC as well as mixing of STWand SAW. Due to this restriction, this sector of the STC ischaracterized by sharp gradients in temperature, macro- (nitrate,silicate and phosphate) and micro- (iron) nutrient concentrations.Shipboard incubations were conducted during austral spring 2000and 2001 to test the hypothesis that these gradients affectthe taxonomic composition and/or growth rates of phytoplanktonon either side of and at the STC. Maximum chlorophyll a concentrationsduring 2000 were 0.39 µg L–1, but were an orderof magnitude higher in 2001. During both years, STC phytoplanktonwere dominated by diatoms (77% of the total chlorophyll a duringaustral spring 2000 and 70% during spring 2001), whereas cryptophytesand prasinophytes dominated STW assemblages (27 and 36% during2000, and 63 and 17% during 2001). Chlorophyll in the SAW wasdominated by procaryotes and photosynthetic nanoflagellatesduring 2000 (17% procaryotes, 68% nanoflagellates), and by diatomsduring the austral spring 2001 cruise (53%). Growth rates ofthe phytoplankton assemblage were determined by 14C-labelingof chlorophyll a and photosynthetic pigments. During 2000, temperature-normalizedgrowth rates were near maximal at the STC, and decreased onaverage to less than half of the maximum north and south ofthat front, whereas in 2001 both absolute and relative growthrates were low at all stations. Growth rates did not closelyparallel biomass of the various taxa, suggesting that nutrientlimitation and/or grazing were significantly impacting standingstocks. It appeared that growth was strongly influenced by nutrientsand light, but that biomass was more strongly influenced bygrazing. The STC is a globally important region of enhancedbiomass and productivity; however, the phytoplankton assemblagereflects control by both top–down and bottom–upprocesses that makes a predictive understanding of the area'sbiogeochemical cycles extremely difficult.  相似文献

In 2 replicated factorial experiments, 7-h climate chamber exposures were used to study the responses of adult NZW rabbits to a range of elevated temperatures and humidities. At 18 mm Hg water vapour pressure, 23.8° C was well tolerated, rectal temperature (RT) and respiration rate (RR) averaging 38.6±0.3° C and 82.9±15.5 breaths/min, respectively. Both parameters were elevated (P<0.001) at 32.2°, 37.8° and 43.3° C. RT and RR reached plateau levels of 39.5–40.1° C and 410–460/min at 32.2° C, which was tolerated for the full 7-h test period. Test temperatures of 37.8° and 43.3° C, on the other hand, could be tolerated for only 80 and 40 min respectively, before RT reached the safe upper limit of 41.7° C. Final RR values at 37.8° and 43.3° C were 701.6±42.7 and 812±55.1/min, respectively. In a 34.5° C atmosphere a humidity of 21 mm Hg water vapour pressure was classified as dry, and was tolerated for 323±123 min. RT and RR increased by 0.6° C and 316/min during the first 20 min of exposure (P<0.05). Thereafter both parameters increased progressively, but with no significant differences between successive recording periods, until RR reached 550.3±88.8/min at 41.7° C RT. Humidities of 25, 29 and 33 mm Hg water vapour pressure were, on the other hand, classified as wet and were tolerated for only 92±22, 81±16 and 119±50 min, respectively. RR at the times that RT reached 41.7° C at these 3 humidities was 732±26, 789±30 and 764±23/min, respectively. The results point to the likelihood that thermal stress will adversely affect the productivity and welfare of NZW rabbits in the tropics unless adequate housing environments are provided. Significant between-individual phenotypic differences in heat tolerance suggest the need for genetic studies of the possibility of selecting for improved heat tolerance.  相似文献

The pathological examination of the rabbit knee joint with antigen-induced arthritis produced by heat-killed Streptococcus faecalis (Str. faec.) as antigen was carried out. Macroscopically, there were findings of acute inflammation about five hours after the injection. Histopathologically, very remarkable acute exudative inflammation was seen 48 hours later. This supported the picture of Arthus reaction. The Arthus reaction disappeared with time, and this supported the view of delayed-type hypersensitivity three weeks later. After that, an obvious chronic inflammation was admitted in 10 weeks. This resembled the histopathological feature of rheumatoid arthritis (RA) when these findings are summarized. It was suggested that arthritis produced by Str. faec. progresses from an acute condition to a one with time. As mentioned above, it thought that Arthus reaction of both immediate hypersensitivity and delayed-type hypersensitivity are necessary in the occurrence of this arthritis.  相似文献

The motor mounting patterns of male and female New Zealand white rabbits were analyzed by means of an accelerometric technique and frequency analysis. Clear behavioral dimorphism was noted in the motor mounting pattern. Pelvic thrusting by males was periodic while that performed by females lacked rhythmicity. Thrusting in males was more vigorous than in females. Ovariectomy markedly decreased the incidence of mounting behavior. Testosterone propionate (TP, 5 mg daily for 1 month), restored mounting in all ovariectomized rabbits. TP stimulated the vigor of thrusting and induced a rhythmic mounting pattern in many cases similar to that displayed by intact male rabbits, i.e., thrusting frequency 13-16 per second. Estradiol benzoate (10 micrograms daily for 1 month), elicited mounting in three of the seven rabbits tested. Pelvic thrusting in these rabbits was often highly synchronous showing a frequency higher (18 to 21 thrusts per second) than that displayed by male rabbits. The results suggest the following conclusions: (a) the behavioral dimorphism in mounting observed in rabbits is due to variations in the secretion of sex steroids by the adult gonads rather than to differences in the organization of the neural substrate of mounting; (b) gonadal steroids influence directly or indirectly the neural structures involved in some characteristics of pelvic thrusting, i.e., rhythmicity and vigor.  相似文献

Atrial fibrillation (AF) is characterized by multiple rapid and irregular atrial depolarization, leading to rapid ventricular responses exceeding 100 beats per minute (bpm). We hypothesized that rapid and irregular pacing reduced intravascular shear stress (ISS) with implication to modulating endothelial responses. To simulate AF, we paced the left atrial appendage of New Zealand White rabbits (n = 4) at rapid and irregular intervals. Surface electrical cardiograms were recorded for atrial and ventricular rhythm, and intravascular convective heat transfer was measured by microthermal sensors, from which ISS was inferred. Rapid and irregular pacing decreased arterial systolic and diastolic pressures (baseline, 99/75 mmHg; rapid regular pacing, 92/73; rapid irregular pacing, 90/68; p < 0.001, n = 4), temporal gradients ( ${\partial\tau/\partial t}$ from 1,275 ± 80 to 1,056 ± 180 dyne/cm2 s), and reduced ISS (from baseline at 32.0 ± 2.4 to 22.7 ± 3.5 dyne/cm2). Computational fluid dynamics code demonstrated that experimentally inferred ISS provided a close approximation to the computed wall shear stress at a given catheter to vessel diameter ratio, shear stress range, and catheter position. In an in vitro flow system in which time-averaged shear stress was maintained at ${{\tau_{\rm avg}} = 23 \pm 4\, {\rm dyn}\, {\rm cm}^{-2} {\rm s}^{-1}}$ , we further demonstrated that rapid pulse rates at 150 bpm down-regulated endothelial nitric oxide, promoted superoxide (O 2 .? ) production, and increased monocyte binding to endothelial cells. These findings suggest that rapid pacing reduces ISS and ${\partial\tau/\partial t}$ , and rapid pulse rates modulate endothelial responses.  相似文献

Mangrove forest coverage is increasing in the estuaries of the North Island of New Zealand, causing changes in estuarine ecosystem structure and function. Sedimentation and associated nutrient enrichment have been proposed to be factors leading to increases in mangrove cover, but the relative importance of each of these factors is unknown. We conducted a fertilization study in estuaries with different sedimentation histories in order to determine the role of nutrient enrichment in stimulating mangrove growth and forest development. We expected that if mangroves were nutrient-limited, nutrient enrichment would lead to increases in mangrove growth and forest structure and that nutrient enrichment of trees in our site with low sedimentation would give rise to trees and sediments that converged in terms of functional characteristics on control sites in our high sedimentation site. The effects of fertilizing with nitrogen (N) varied among sites and across the intertidal zone, with enhancements in growth, photosynthetic carbon gain, N resorption prior to leaf senescence and the leaf area index of canopies being significantly greater at the high sedimentation sites than at the low sedimentation sites, and in landward dwarf trees compared to seaward fringing trees. Sediment respiration (CO2 efflux) was higher at the high sedimentation site than at the low one sedimentation site, but it was not significantly affected by fertilization, suggesting that the high sedimentation site supported greater bacterial mineralization of sediment carbon. Nutrient enrichment of the coastal zone has a role in facilitating the expansion of mangroves in estuaries of the North Island of New Zealand, but this effect is secondary to that of sedimentation, which increases habitat area and stimulates growth. In estuaries with high sediment loads, enrichment with N will cause greater mangrove growth and further changes in ecosystem function.  相似文献

Induction and evaluation of atherosclerosis in New Zealand white rabbits   总被引:2,自引:0,他引:2  
Atherosclerosis was experimentally induced in New Zealand white rabbits by feeding a high cholesterol diet for 12 weeks for screening of drugs against atherosclerosis. After 12 weeks, blood was collected from ear vein for evaluation of total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) levels, then the animals were sacrificed to collect the livers for estimation of cholesterol, and aorta for gross and histopathological evaluations. The elevated levels of serum and liver parameters accompanied by gross and histopathological changes like accumulation of foam cells, atheromatous plaque formation and replacement fibrosis supported the successful induction of atherosclerosis in New Zealand white rabbits.  相似文献
