Using an EA rosetting system, it was observed that Fc receptors (FcR) were present on the surface of T cells as well as B cells, and that functional differences existed between FcR-positive (FcR+) and FcR-negative (FcR?) cells in both T and B cells in in vivo humoral immune responses. Approximately 15% of splenic T cells obtained by nylon wool passage are FcR+. The number of surface immunoglobulinbearing cells as detected by immunofluorescent staining accounted for less than 10% of these FcR+ cells. FcR+ and FcR? T+B-cell populations obtained from spleens contain 60 and 20% of surface immunoglobulin-positive cells, respectively. In the adoptive primary response in which horse RBC and dinitrophenyl-conjugated dextran (DNP-DE) were used as T-dependent and T-independent antigens, respectively, the majority of precursor B cells were FcR?. In the secondary response using hapten-primed B cells and carrier-primed T cells, the majority of memory B cells for a haptenic determinant were also FcR?. Furthermore, the majority of functional cells exerting helper activity in the same hapten-carrier system are FcR? cells, and FcR+ T cells collaborate much less effectively with either memory B cells or helper FcR? In in vitro plaque-forming cell (PFC) response to particulate as well as to soluble antigen, the functional difference between Fc receptor-bearing (FcR+) and nonbearing (FcR?) murine splenic lymphocytes was analyzed using the EA rosetting method. In the secondary anti-horse red blood cell (HRBC) response of C3H mice, FcR? cells showed higher IgM and IgG responses than did FcR+ cells. When nylon wool (NW)-purified T cells primed with keyhole limpet hemocyanin (KLH) were fractionated into FcR? and FcR+ T cells, helper activity was proven in the former subset in the cooperation with syngeneic spleen cells primed with dinitrophenylated ascaris extract (DNP-Asc). FcR+ T cells showed essentially no helper activity. When FcR? cells were cultured, neogenesis of FcR+ cells was observed on Days 3 to 5. The conversion from FcR? to FcR+ cells was prominent in B cells (40 to 50%), whereas NW-purified nonadherent FcR? T cells converted poorly (15 to 20%). The converting process was accelerated slightly by mitogens, but was least affected by antigens. To examine the possible contribution of neogeneic FcR+ T cells in the helper activity, KLH-primed FcR? T cells were precultured for 7 days with homologous antigen. The specific helper activity of the cultured T cells proved to be unaffected by the depletion of neogeneic FcR+ T cells by EA rosetting. The neogeneic FcR+ T cells had no helper activity. It was thus suggested that helper T cells remain in the FcR? cell fraction and do not convert to the FcR+ state during the cooperating process.  相似文献

By separating FcR+ and FcR? cells from stimulator spleens using an EA rosetting procedure, it was found that EA-rosetting (FcR+) cells stimulate mixed-lymphocyte culture reaction (MLR) far more effectively than do non-EA-rosetting (FcR?) cells. The difference in stimulatory activity is observed in MLR of both H-2 and M-locus different combinations and cannot be explained by the proportion of B cells and macrophages contained in each population. The finding that FcR+ cells can stimulate allogeneic responding T cells more effectively than FcR? cells suggests a close association of FcR with Ia and Mls antigens on the cell surface.  相似文献

EA, i.e., antigen-antibody complexes are able to induce an antigen-nonspecific suppressive factor(s) from FcR+ B cells by binding on FcR. This factor, termed “suppressive B-cell factor (SBF)” was only effective on H-2 compatible, but not on H-2 incompatible spleen cells in an adoptive cell transfer system. Furthermore, SBF, prepared from B10.A (H-2a) splenic FcR+ B cells, suppressed the adoptive primary response of B10.D2 mice (H-2d), in addition to A/J mice (H-2a) against DNP-DE, by the pretreatment of cells with SBF in vitro. Absorption with affinity columns demonstrated that active components) of SBF from C3H/He mice (H-2k) was eliminated by both B6 anti-CBA (H-2b anti-H-2k) and B10.D2 anti-B10.BR (H-2d anti-H-2k), but not B10 anti-B10.A (H-2b anti-H-2a). In contrast, the suppressive activity of SBF was eliminated neither by anti-mouse Ig nor by a heat-aggregated human γ-globulin column. These results indicate that SBF contains a product coded by the right-hand side of H-2 gene complex, but does not contain Ig determinants nor FcR. Thus, it is conceivable that a compatibility of the right-hand side of H-2 gene complex is required for inducing effective suppression of spleen cells by SBF. SBF was considered to be a trypsin-resistant and heat-labile substance with a molecular weight of 30,000–63,000. The target cells for SBF were FcR? B precursors, but not helper T cells.  相似文献

Soluble antigen-antibody complexes composed of 3 M KCl-extracted L1210 antigens and alloantibody to L1210 given to C3H mice caused immunosuppression in the mice. This was reflected in part by the inhibition of cytophilic antibody receptors on macrophages which could be used as a measure of the suppression. Thymocytes or splenic T cells from mice treated with immune complexes could adoptively transfer the suppression to normal syngeneic mice. These cells, which we have termed suppressor inducers, were found to be Ia positive: specifically, I-A+, I-J?. Thus, treatment of the inducers with anti-la or anti-I-A antibodies and complement in vitro abrogated their ability to transfer the suppression to normal mice. In contrast treatment with anti-I-J serum and complement had no effect. Through a similar approach, the cooperating (acceptor) T cells were found to be I-A+, I-J?. Pretreatment of mice with anti-Ia or anti-I-A serum before the administration of antigen-antibody complexes prevented the inhibition of macrophages. This was due at least in part to steric hindrance of adjacent Fc receptors on the FcR+ T cells with which the complexes interacted. Early interaction of immune complexes with FcR+ T cells was in fact demonstrated directly by the inability of the complexes to induce suppression when FcR+ T cells were depleted. The thymocytes or splenic T cells from anti-Ia-pretreated mice failed to transfer the suppression to recipient mice. In contrast, treatment with either anti-Ia or anti-I-A after the immune complexes did not abrogate the generation of suppressor inducers. Treatment of normal recipient mice with anti-Ia serum in vivo before they received the suppressor inducer cells did not prevent cooperation between the two types of cells. By the same token, blocking of Ia antigens of the inducers in vitro with anti-Ia serum (without complement) also did not impair the cooperative interaction. These results indicate that antigen-antibody complexes generate I-A-positive, I-J-negative T-suppressor inducer cells from FcR+ naive T cells. These in turn interact with Ia-positive (I-A+ and I-J?) normal thymocytes or spleen T cells. This interaction most likely generates the ultimate suppressor T cells that suppress cytophilic antibody receptors on macrophages in vivo. However, the I-region determined antigens did not appear to be directly involved in the T-T interaction of suppressor inducer and acceptor cells.  相似文献

The expression of cell surface alloantigens and receptors on purified populations of murine macrophages from short-term bone marrow cultures and peritoneal exudates was analyzed by rosetting techniques, by microcytotoxicity, and by absorption. Staphylococcal enterotoxin B (SEB) is a potent mitogen for both human and murine T lymphocytes. We report here studies which demonstrate that a suppressor cell population, capable of suppressing the primary immune response of normal syngeneic mouse splenocytes to heterologous sheep erythrocytes (SRBC), is activated by SEB. Enterotoxin concentrations ranging from 0.05 to 5.0 Mg ml?1 are capable of activating this suppressor cell population, and significant suppression can be detected with relatively small numbers of SEB-primed spleen cells (SEB-PSC) in culture. Elimination of macrophages before or after priming splenocytes with SEB does not reduce the suppression of plaque-forming cell (PFC) responses when SEB-PSC are added to normal cells in Mishell-Dutton cultures. Treatment of cells with anti-Thy-1 serum plus complement, after priming with SEB, effectively eliminates the activity of the suppressor cell population. Enrichment for T cells before priming with SEB results in greater suppression of PFC responses than do SEB-PSC generated in cultures of nonfractionated splenocytes. Activation of suppressor cells with SEB in vitro appears to occur through the induction of the T-cell subpopulation expressing the Lyt-1?,2+,3+ cell surface phenotype, since selective depletion of this T-cell subpopulation with monoclonal rat anti-mouse Lyt-2 antisera after priming cells with SEB virtually eliminates the suppressor activity.  相似文献

ADCC of lymphocyte suspensions from the spleen, lymph node, and blood in the rat, rabbit, and guinea pig, was directly correlated with the presence of Fc+ C3? and indirectly with the presence of Fc+ C3+ lymphocyte subpopulations in these preparations. Rabbit lymphocytes, from all three lymphoid compartments, were deficient in the Fc+ C3? subpopulation but enriched in the Fc+ C3+ subpopulation and lacked K-cell activity. A similar lymphocyte profile and lack of K-cell activity was found in the lymph node of the rat and guinea pig. Lymphocytes from the blood and spleen, on the other hand, had a substantial population of Fc+ C3? lymphocytes and a prominent K-cell activity. The Fc+ C3+ lymphocyte, although unable to lyse IgG-coated target cells, is able to compete in vitro with the Fc+ C3? (K) cell and significantly reduce ADCC. Phagocytic cells lyse very effectively erythroid target cells and contamination by such cells may explain previously reported K-cell activity of rabbit lymphocytes.  相似文献

Antisera against sheep red blood cells (SRBC) specifically suppressed the direct anti-SRBC plaque-forming cell (PFC) response in mice when passively administered with the antigen. The suppressive activity of mouse and rabbit anti-SRBC sera was found to correlate with anti-SRBC opsonic activity but not with hemagglutination or hemolysin titers. Macrophage depletion of mice, using carrageenan treatment, inhibited antibody-mediated immune suppression. When mice immunized with SRBC were given 125I-labeled Udr, radiolabeled spleen lymphocytes were obtained which specifically formed rosettes with SRBC. These radiolabeled antigen-reactive cells (1ARC) were specifically opsonized in mice treated with antigen-antibody complexes but not in mice treated with antigen or antibody alone. These results suggest that antibody-mediated immune suppression may be due to specific opsonization (and subsequent destruction) of ARC in the presence of antigen-antibody complexes.  相似文献

Mouse aggregated IgG, when continuously present in cultures of mouse spleen cells immunized with sheep erythrocytes, causes a dose dependent inhibition of the generation of plaque forming cells with a maximum of about 90% at 400 μg IgG/culture. Unaggregated IgG induces a similar inhibition, whereas treatment with mouse albumin or F (ab1)2, under the same conditions, does not affect the generation of plaque forming cells.It has been reported that unaggregated IgG binds poorly to Fc receptors of B lymphocytes and thus should not be expected to inhibit PFC generation if the effect is at the level of B lymphocyte Fc receptors. Competitive binding experiments were carried out and showed that aggregated and unaggregated IgG compete similarly with 125I-labeled aggregated IgG for binding to Fc receptors of mouse spleen cells.The same inhibition of PFC can be induced by aggregated IgG in cultures of B lymphocytes immunized with the T-independent antigen DNP-Ficoll. When IgG is absorbed extensively with sheep erythrocytes and added to cultures immunized with sheep erythrocytes, PFC generation is inhibited to a level comparable to that of nonabsorbed IgG.These results suggest that IgG binding to Fc receptors leads to a severe inhibition of the induction of PFC by both T-dependent and T-independent antigens. This and other work from our laboratory indicate that this effect may be at the level of B lymphocyte Fc receptors.Taken together with reports from several laboratories, the data presented here suggest that Fc receptors may have a regulatory role on the activation of B lymphocytes by antigens or mitogens.  相似文献

The functional role of complement receptor (CR+) lymphocytes in antibody responses was investigated. Initially it was found that in the spleens of 6–8-week-old CBA/H mice only approximately 40% of the B cells were CR+. The CR+ and CR? splenocytes were then separated by a recently described fractionation procedure (Parish, C. R., et al., Eur. J. Immunol.4, 808, 1974) and assayed alone or in combination for their ability to transfer a range of antibody responses to irradiated recipients. All of the antigens studied, irrespective of their structure or T-cell dependence, were capable of activating CR+ B cells to synthesize antibody. However, only repeating determinant antigens, such as horse red blood cells (HRBC) and dinitrophenyl-polymerized flagellin (DNP-POL), were capable of activating CR? B cells, the soluble antigen DNP-flagellin (DNP-MON) being unable to trigger these cells. Repeating determinant nature rather than T-cell dependence appeared to be the factor that determined whether an antigen could provoke the CR? B cells to synthesize antibody, as HRBC and DNP-POL differ widely in their T-cell dependence. The same phenomenon was observed with direct and indirect PFC responses and also with primary and secondary antibody responses. In addition, there was no evidence for collaboration between CR+ and CR? B cells in the induction of antibody responses to the T-dependent antigens, HRBC and DNP-MON. Furthermore, no CR+ helper T cells were detected.It is postulated that complement receptors facilitate T-B interaction by stabilizing the union of soluble antigens with antigen-specific receptors on CR+ B cells. In contrast, repeating determinant antigens can avidly bind to antigen-specific receptors on B lymphocytes without involvement of the complement receptors.  相似文献

A Sephadex G-10 column coated with antigen-antibody complexes and complement retains complement receptor-bearing (CR+) mouse spleen cells. The effluent is rich in thymus-derived cells (T cells), and contains bone marrow-derived cells (B cells) which carry surface immunoglobulin (Ig), Ir-associated antigen (Ia), and Fc receptors, but no complement receptors (CR-). Although both unfractionated and CR- B cell populations are capable of producing antibody to red cell antigens, they differ in their requirements for the initiation of the response. Unfractionated B cells cooperate with primed as well as unprimed helper T cells; macrophages are required for this cooperation but can be replaced by 2-mercaptoethanol. CR- B cells cooperate with primed but not with unprimed T cells provided macrophages are added to cultures. After addition of culture supernatant from BCG-activated macrophages CR- B cells cooperate with both unprimed and primed T helper cells.  相似文献

T lymphocytes are stimulated to release T-cell-replacing factors in response to Fc fragments of human IgG. Lyt 1+23? T cells are directly triggered to factor production by Fc subfragments, derived from intact Fc fragments by macrophage-dependent enzymatic cleavage. The characteristics of suppressor cells induced by 2,4-dinitrophenyl (DNP)-coupled syngeneic lymphocytes (syninduced suppressor cells) were studied. 2,4-dinitro-1-fluorobenzene (DNFB) contact hypersensitivity was completely suppressed when the syninduced suppressor cells were transferred intravenously. These syninduced suppressor cells had surface markers of Thy-1, FcR? and Lyt-2+, 3+ antigens, as well as I-J gene products on their cell surfaces. The suppression of DNFB contact hypersensitivity was abrogated when these suppressor T cells were incubated in Petri dishes coated with the DNP-syngeneic lymphoid cell membrane, which suggests that these suppressor T cells had the specific antigen-binding receptors on their cell surfaces.  相似文献

To determine the mechanisms in the triggering of thymus-independent lymphocytes (B cells) for development into antibody-forming cells (AFC), genesis of IgM AFC elicited polyclonally by nonspecific stimulation with B-cell mitogen, such as nystatin and bacterial lipopolysaccharide, was compared with that of IgM AFC specifically elicited by antigenic stimulation, using mouse spleen cell cultures as an experimental system and sheep erythrocytes (SRBC) as a test antigen. Considering that differentiation and proliferation are necessary cellular events for precursor B cells to develop into AFC, the effect of different antimetabolic agents on the generation of each type of AFC in spleen cell cultures was examined. The generation of anti-SRBC IgM hemolysin plaque-forming cells (PFC) in B-cell mitogen-stimulated spleen cell cultures was found to be less susceptible to X-irradiation or mitomycin C than that in the SRBC-stimulated cultures. These apparently paradoxical results were affiirmed using colcemid as an inhibitor of cell mitosis and hydroxyurea (HU) as an inhibitor of cellular DNA synthesis. Thus, when spleen cell cultures responding to either SRBC or B-cell mitogen were exposed to colcemid or HU during a period from 2 days to 3 days after the stimulation, the exponential generation of anti-SRBC IgM PFC in the cultures responding to SRBC was completely halted, whereas that in the cultures responding to B-cell mitogen was not. Furthermore, N6, O2′ -dibutyryl adenosine 3′, 5′ -cyclic monophosphoric acid was found to halt the exponential generation of antigen-induced anti-SRBC IgM PFC but not that of the B-cell mitogen-induced anti-SRBC IgM PFC. From these results it was suggested that B-cell mitogen might stimulate precursor Bμ cells at a late stage in the differentiative pathway to develop into AFC without cell division, and that antigenic stimulation might stimulate relatively primitive precursor Bμ cells to proliferate and then differentiate into AFC. Based on this idea, mechanisms in the triggering of B-cell activation are discussed.  相似文献

Supernatants of peritoneal macrophages cocultivated with spleen cells or thymocytes contain a factor suppressing the 3H-thymidine (3H-TdR) incorporation of PHA-stimulated lymphocytes. The suppressing activity is not due to cytotoxicity and does not affect the rate of cell transformation or cell proliferation. The factor reduces only the 3H-TdR incorporation and not the DNA synthesis of lymphocytes. It does not show target cell specificity. The factor is dialysable, heat stable, and generated by macrophages.  相似文献

Using a culture system of bone marrow progenitor cells with GM-CSF and TGF-β1, a study was performed to analyze the effect of TGF-β1 on the development of dendritic cells (DC) and to elucidate the regulatory role of macrophages co-developing with dendritic cells. The results demonstrate that DC generated in the presence of TGF-β1 were immature with respect to the expression of CD86, nonspecific esterase activity and cell shape. Such inhibitory effects of TGF-β1 were dependent on FcR+ macrophages, which were depleted by panning. TGF-β1 did not appear to inhibit the commitment of progenitor cells to the DC lineage. In addition, TGF-β1 also acted directly on the intermediate stage of DC to prevent their over-maturation, which results in a preferential decrease in MHC class II, but not in CD86, in the presence of TNF-α. FcR+ suppressive macrophages were also shown to facilitate DC maturation when stimulated via FcR-mediated signals even in the presence of TGF-β1. These results indicate that TGF-β1 indirectly and directly regulate the development of DC and that co-developing macrophages have a regulatory role in DC maturation.  相似文献

Purified human C-reactive protein (CRP) inhibited the in vitro anti-hapten antibody plaque-forming cells (PFC) response of both carrier keyhole limpet hemocyanin (KLH)-primed and unimmunized Balb/c spleen cells to TNP-KLH. The inhibitory effect was neutralized by the CRP-substrate, C-polysaccharide. The response to the T-independent antigens, TNP-T4 and DNP-lys-Ficoll, was not inhibited by CRP. A cell population that was suppressive for the in vitro PFC response was generated by incubating normal spleen cells with CRP. These cells suppressed the PFC response of syngeneic KLH-primed cells to TNP-KLH in proportion to the number of added lymphoid cells with bound CRP. Selective depletion of B cells, T cells or macrophages before incubation with CRP revealed that T cells were required for the induction of suppressive cells. Treatment of spleen cells after incubation with CRP, with T cell-specific antisera and C abolished suppressor-cell activity. Mitomycin-C treatment of the CRP-binding cells did not alter their suppressive activity. These results indicated that CRP mediates suppression of antibody induction to T-dependent antigens by interacting with T cells and generating a suppressive T-cell population.  相似文献

The emergence of novel T cells during mammary tumorigenesis has been previously described. T cells with surface markers usually associated with B cells, i.e. complement receptors (CR), appear in the spleens from tumor bearing mice. We now report on the appearance of Fc receptor (FcR) positive T cells in the spleens from the same animals. The kinetics of appearance of the two kinds of cells are similar.Based on evidence from double and triple label assays, it was concluded that FcR and CR are not coexpressed on the same T cell and that the two kinds of T cells which emerge do so in an independent fashion. Furthermore, they appear to represent a branch in the differentiation process influenced by tumor growth. The development of CR+ T cells represents an irreversible process as evidenced by the lack of change in the cells' representation following surgical procedures. In contrast the development of FcR+ T cells appears to be quite flexible in nature since mere surgical trauma as well as tumor mass removal can effect a decrease in the proportion of such cells.  相似文献

In vitro culture techniques have been used to compare the direct (IgM) plaqueforming cell (PFC) response to heterologous erythrocytes (RBC) by normal mouse spleen cells and spleen cells from mice injected intravenously with 5 × 104 RBC ten days previously [low dose primed (LDP)]. Although LDP mice fail to undergo a significant primary PFC response, their spleen cells are capable of a secondary or enhanced PFC response in vitro. The secondary PFC response is shown to be a function of: (A) an increase in the frequency of immunocompetent cells or units (IU) due to in vivo priming, and (B) an increased number of PFC generated per IU subsequent to in vitro stimulation. The latter increase is shown to be mediated through a shorter PFC doubling-time during logarithmic expansion of the PFC population. Analysis of nonadherent spleen cell dose response experiments indicate that two nonadherent cell types interact in the secondary response. Subsequent cocultivation experiments suggested that both of these cell types must be “primed” to allow induction of a secondary response. Although adherent cells are required for the secondary response, normal splenic adherent cells serve as equivalent substitutes for LDP adherent cells.  相似文献
