The emission of reduced volatile sulfur compounds from twigs of Norway spruce (Picea abies (L.) Karst.) was measured in the field by cryosampling and gaschromatographic analysis. Trees were growing in the Erzgebirge (E-Germany) at Oberbärenburg and at the Kahleberg and at a third stand in NW-Bavaria (S-Germany). Emission rates were also measured for Scotch pine (Pinus sylvestris L.) and Blue spruce (Picea pungens Engelm.) at the Kahleberg. Twigs still attached to the trees were enclosed in a flow-through gas exchange cuvette. H2S was detected as the predominant reduced sulfur compound emitted from the twigs. The mean H2S emission rate from twigs of Norway spruce varied between 0.04 pmol kg-1 dw s-1 at Würzburg and 6.21 pmol kg-1 dw s-1 at the Kahleberg. Comparing different species at the Kahleberg, the mean H2S emission rate was almost the same from twigs of Norway spruce (6.2 pmol kg-1 dw s-1) and Blue Spruce trees (5.9 pmol kg-1 dw s-1) but it was approximately 18 times higher for Scotch pine (110 pmol kg-1 dw s-1). The percentage of SO2-exclusion via H2S-emission of the tree species investigated at the Kahleberg is calculated on the basis of data on SO2 fluxes. It is very small for Norway spruce and Blue spruce. However, for Scotch pine, H2S emission contributes about 10% to the detoxification of SO2.  相似文献

Background

Somatic embryogenesis in conifer species has great potential for the forestry industry. Hence, a number of methods have been developed for their efficient and rapid propagation through somatic embryogenesis. Although information is available regarding the previous process-mediated generation of embryogenic cells to form somatic embryos, there is a dearth of information in the literature on the detailed structure of these clusters.

Methodology/Principal Findings

The main aim of this study was to provide a more detailed structure of the embryogenic tissue clusters obtained through the in vitro propagation of the Norway spruce (Picea abies (L.) Karst.). We primarily focused on the growth of early somatic embryos (ESEs). The data on ESE growth suggested that there may be clear distinctions between their inner and outer regions. Therefore, we selected ESEs collected on the 56th day after sub-cultivation to dissect the homogeneity of the ESE clusters. Two colourimetric assays (acetocarmine and fluorescein diacetate/propidium iodide staining) and one metabolic assay based on the use of 2,3,5-triphenyltetrazolium chloride uncovered large differences in the metabolic activity inside the cluster. Next, we performed nuclear magnetic resonance measurements. The ESE cluster seemed to be compactly aggregated during the first four weeks of cultivation; thereafter, the difference between the 1H nuclei concentration in the inner and outer clusters was more evident. There were clear differences in the visual appearance of embryos from the outer and inner regions. Finally, a cluster was divided into six parts (three each from the inner and the outer regions of the embryo) to determine their growth and viability. The innermost embryos (centripetally towards the cluster centre) could grow after sub-cultivation but exhibited the slowest rate and required the longest time to reach the common growth rate. To confirm our hypothesis on the organisation of the ESE cluster, we investigated the effect of cluster orientation on the cultivation medium and the influence of the change of the cluster’s three-dimensional orientation on its development. Maintaining the same position when transferring ESEs into new cultivation medium seemed to be necessary because changes in the orientation significantly affected ESE growth.

Conclusions and Significance

This work illustrated the possible inner organisation of ESEs. The outer layer of ESEs is formed by individual somatic embryos with high metabolic activity (and with high demands for nutrients, oxygen and water), while an embryonal group is directed outside of the ESE cluster. Somatic embryos with depressed metabolic activity were localised in the inner regions, where these embryonic tissues probably have a very important transport function.  相似文献

The mode of inheritance of chloroplast and mitochondrial DNA (mtDNA) in rye × triticale intergeneric hybrids has been studied with the use of specific PCR markers for loci 18S/5S and 3rbcL in organelle DNA. In rye × triticale BC1, mtDNA copies of two types, paternal and maternal, have been found; in BC2 plants, only paternal mtDNA and chloroplast DNA (cpDNA) have been detected. Summary Resistance of Pteridium aquilinum to insect attack was studied by incorporating air dried bracken leaf meal and extracts of bracken leaf meal into an artificial diet for Trichoplusia ni (Lepidoptera: Noctuidae). When larvae are reared on diets containing 6% bracken leaf meal, they do not mature past the second instar and after 16 days the average weight is approximately 1 mg compared to 70 mg for control larvae. Feeding initiation studies indicate that a feeding deterrent is present in bracken fern but feeding rates and food utilization efficiency studies suggest that either the deterrent or another compound also functions as a toxin. This toxin does not affect growth, feeding rates, or utilization efficiency for the first 4 days after third instar larvae are transferred to a diet containing the water extract of bracken leaf meal; thereafter growth is terminated and feeding is greatly reduced. The active factor is water soluble, heat labile, and non-volatile and this partial characterization indicates that neither the bracken ecdysones or the anti-thiamine factor of bracken is involved in the resistance of this fern to insect attack by T. ni.  相似文献

1.白僵菌(Beauveria bassiana(Bals.)Vuill.)是大豆食心虫幼虫的寄生性真菌。自然寄生率5—10%,有时可达30%以上,是造成大豆食心虫幼虫自然死亡的有力因素之一。 2.白僵菌的发育及孢子发芽温度是18—28℃,其中以25—28℃最为适宜。适宜的发芽湿度要在99%以上。在土壤中如果温度适宜(25—28℃)土壤含水量10%以上就可以引起幼虫显著寄生。 3.幼虫体沾着一定数量的病菌孢子可以引起大量死亡,如果幼虫体上沾着孢子数4944个以上则可致100%寄生,不足此数的,寄生率成比例地降低。菌粉在太阳光照射下6天后寄生力受到影啊,30天以后寄生作用已极小。病菌对于带菌的幼虫虽可同样寄生,但寄生能力要比直接使幼虫接触孢子要小得多。 在室温下的菌粉中,病菌的生活力(致病力)可以保持一年左右。 4.利用白僵菌防治大豆食心虫的效果是:室内不论菌粉浓度及用量多少,寄生率均达100%。小区试验可致幼虫寄生70.19—100%,田间防治可以提高幼虫寄生30.15—36.11%,降低成虫羽化49.83—69.8%。 5.作者认为:利用白僵菌防治大豆食心虫是有希望的。今后是对于大量繁殖病菌的方法,施用药剂时期及与农药混用以提高效果有进一步研究之必要。  相似文献

A method for micropropagation of mature trees of Capparis decidua was developed. Multiple shoots were obtained from nodal explants on Murashige and Skoog's (1962) medium+0.1mgl–1 NAA+5.0mgl–1BAP+additives (50mgl–1 ascorbic acid and25 mgl–1 each of adenine sulphate, L-arginine and citric acid) at 28 ± 2°C, 12 h/dphotoperiod and 35–40 mol m-2s–1 photon flux density. The shoots were multiplied by (i) subculture of nodal shoot segments onto MS +0.1 mgl-–1 IAA+1.0mgl–1 BAPH+additives, and (ii) repeated transfer of original explant onto MS+ 0.1mgl–1 IAA+mg l–1 BAP+additives, at intervals of 3 weeks. Sixty to 70% of the shoots rooted when pulse treated with 100 mg l–1 IBA in half strength MS liquid medium for 4h, and then transferred onto hormone-free half-strength agar-gelled MS basal saltmedium. Incubation in dark at 33 ± 2°C for 6d favoured root induction. Isolated ovules from the maize homozygous recessive brown midrib (bm3) were in vitro pollinated by pollen carrying the dominant allele — and the purple embryo marker (PEM). The colour characters of the embryos and kernels corresponded to the results of control pollination and confirmed the process of double fertilization.  Rice is the only cereal in which germination and coleoptile elongation occur in hypoxia or anoxia. Little is known of the molecular basis directly underlying coleoptile cell extension. In this paper, we describe the expression of α-expansin genes in embryos during seed development and young seedlings grown under various oxygen concentrations. The genes Os-EXP2 and Os-EXP1 were predominantly expressed in the developing seeds, mainly in newly developed leaves, coleoptiles, and seminal roots. These expansins expressed in the developing seeds may give cells the potential to expand after seed imbibition begins. In coleoptiles, Os-EXP4 and Os-EXP2 mRNAs were greatly induced by submergence, while they were weakly detected in aerobic or anoxic conditions. Under submerged soil conditions, the signals hybridized with probes Os-EXP4 and Os-EXP2 in coleoptiles were strongest when coleoptiles elongated in the water layer. These data show that expansin gene expression is highly correlated with coleoptile elongation in response to oxygen concentrations. The Os-EXP4 gene was also expressed in leaves, mesocotyls, and coleorhizas of young seedlings. The growth of these tissues was also correlated with the presence of expansins. Therefore, the evidence derived from this study clearly demonstrates that expansins are indispensable for the growing tissues of rice seedlings. The results of studying of the induced apomixis in pollen of sea-buckthorn irradiated by the 60Co gamma-Radiation are considered. Was established that the most effective dose for pollination of the experimental plants is 50 k Gy. In total, from 46 seedlings 19 cases of apomictic origin were revealed, 7 individuals were found to be haploid (n = = 12) and 19 ones were diploid (2n = 24) of maternal origin. Was supposed that apomictic plants (19 seedlings) have parthenogenetic origin. The reason for such conclusion is that the irradiated anomalous pollen tubes despite not having spermia, are entering embryo sac and stimulate the development of apomictic embryo from non-fertilized female gametes. Apparently, pollen tubes cause the induction of DNA replication in the ovules and the development of parthenogenetic plants. Consequently, the described method can be used for the regulation of parthenogenesis in sea-buckthorn to change natural ratio (1 : 1) of male to female plants in desirable quantity.  相似文献

. In vitro-grown shoot tips excised from preconditioned stock shoots of 'Troyer' citrange were successfully cryopreserved by encapsulation-dehydration. Optimal survival of cryopreserved shoot tips was achieved when encapsulated shoot tips were dehydrated to 17.1% water content. The sucrose concentration in the preconditioning medium significantly influenced the growth and dry matter percentage of the stock shoots as well as subsequent survival of the cryopreserved shoot tips. Maximal growth of stock shoots was obtained in sucrose concentrations in the range of 0.15 M to 0.29 M, while the dry matter percentage increased as sucrose concentration increased up to 0.44 M. The survival of cryopreserved shoot tips increased from 40% to approximately 80% as the sucrose concentration for stock shoots increased from 0.09 M to 0.22 M or 0.29 M. The benzyladenine concentration in the post-culture medium significantly affected the survival and regrowth of the cryopreserved shoot tips. Survival of the shoot tips was lowest when they were post-cultured on benzyladenine-free medium. However, high benzyladenine concentrations (3-4 µM) induced callus formation. Optimal recovery was obtained in post-culture medium containing 2 µM benzyladenine and 0.05 µM !-naphthalene acetic acid. The extraction of shoot tips from alginate beads greatly improved the regrowth of cryopreserved shoot tips.  相似文献

Ontogeny of the species, proceeding analogously to that of the unicellular forms, was studied; however, the cells in the coenobium ripen successively. Abstract. Since the 18th century, the medicinal leech Hirudo medicinalis has been thought to comprise a single species with several different color morphs, but recently some of these color morphs have been assigned to separate species based on morphology, geographical distribution, and molecular sequence data. This research was aimed at testing the ability of three of these species, H. medicinalis, Hirudo verbana, and Hirudo orientalis, to interbreed. We found that in the laboratory, all three species were able to mate with each other and produce hybrid offspring. This suggests that the reproductive isolation is not strong among these species of the genus Hirudo. However, fewer offspring were produced from interspecific crosses compared with intraspecific crosses. This decrease of fecundity (and in some cases, offspring viability) indicates some degree of reproductive isolation between H. medicinalis, H. verbana, and H. orientalis.  相似文献

Nucleotide polymorphisms at genomic regions including 17 nuclear loci, two chloroplast and one mitochondrial DNA fragments were used to study the speciation history of three pine species: dwarf mountain pine (Pinus mugo), peat‐bog pine (P. uliginosa) and Scots pine (P. sylvestris). We set out to investigate three specific speciation scenarios: (I) P. uliginosa is a homoploid hybrid between the other two, (II) the species have evolved without gene flow after divergence and (III) there has been substantial gene flow between the species since their divergence. Overall, the genetic data suggest that P. mugo and P. uliginosa share the same gene pool (average net divergence of 0.0001) and that the phenotypic differences (e.g. growth form) are most likely due to very limited areas of the genome. P. mugo and P. uliginosa are more diverged from P. sylvestris than from each other (average net divergence of 0.0027 and 0.0026, respectively). The nucleotide patterns can best be explained by the divergence with migration speciation scenario, although the hybrid speciation scenario with small genomic contribution from P. sylvestris cannot be completely ruled out. We suggest that the large amount of shared polymorphisms between the pine taxa and the lack of monophyly at all loci studied between P. sylvestris and P. mugoP. uliginosa can largely be explained by relatively recent speciation history and large effective population sizes but also by interspecific gene flow. These closely related pine taxa form an excellent system for searching for loci involved in adaptive variation as they are differentiated in phenotype and ecology but have very similar genetic background.  相似文献

Aegle marmelos is well documented for antihyperglycemic effect and PPAR-γ activation has been suggested to be the molecular mechanism of its action. Also, the plant has been used in Ayurveda as a brain tonic and has been postulated to have antidepressant activities. The present study was designed to investigate the anticonvulsant effects of A. marmelos leaf extract (AME) in pentylenetetrazole and maximal electroshock induced convulsions; involvement of PPAR-γ, nitric oxide pathway and effect of chronic AME treatment on post-ictal depression. AME was administered at doses of 50, 100 and 200 mg kg?1 in PTZ and MES model. Severity of convulsions was noted in both the models. Pretreatment with bisphenol A diglycidyl ether (BADGE) was used to study the involvement of PPAR-γ and l-arginine and N-nitro-l-arginine methyl ester hydrochloride (l-NAME) to study the involvement of nitric oxide (NO). Chronic treatment with AME interspersed with sub maximal doses of PTZ (50 mg kg?1) on every fifth day up to 15 days was given to study post-ictal depression using forced swimming and actophotometer. AME showed significant increase in the onset time and decrease in the duration of convulsions in PTZ and MES models dose dependently. In MES a dose of 100 mg kg?1 had effect comparable to phenytoin. Pretreatment with BADGE and l-arginine reversed the protective effect while l-NAME did not alter the protective effect, thereby indicating possible involvement of PPAR-γ and inhibition of NO. Chronic AME treatment ameliorated the post-seizure depression significantly as evidenced by increase in the locomotor activity and decrease in the immobility time.  相似文献

The endogenous gibberellins (GAs) in leaf tissues of two day-neutral cultivars (Rapella and Selva) of strawberry (Fragaria × ananassa Duch.) were analysed using combined gas chromatography -- mass spectrometry (GC-MS). Seven of the later members of the 13-hydroxylation GA biosynthetic pathway were identified, by comparison of Kovats retention indices and mass spectral data obtained for methyl ester trimethylsilyl ether derivatives, either with data obtained from authentic compounds or literature values. GA1, GA3, GA8, GA17, GA19, GA20 and GA29 were detected in extracts of both cultivars.  相似文献
