This paper aims to assess the marine sediment quality along the Tunisian coast using clam Ruditapes decussatus embryos and larval bioassays tests. Elutriate samples prepared with sediment from four sites (Monastir Lagoon, Chebba, Mahres, and Zarat) were used for chemical analysis and clam bioassays tests (embryogenesis, larval growth. and metamorphosis). For embryogenesis test, five elutriates concentrations (12%, 25%, 50%, 75%, and 100%) were prepared. Metal concentrations were highest at the Mahres and Monastir Lagoon sites. Clam embryogenesis success was significantly (p < 0.05) reduced in all elutriates preparations at concentrations up to 50% except Chebba, where inhibition was evident at only the highest concentration (100%). Compared to the control (9.4 μm day?1), larval growth rate was significantly (p < 0.05) reduced only in two sites, Monastir Lagoon (6.8 μm day?1) and Zarat (5.6 μm day?1). However, larval survival was lower (42.9%) in elutriates prepared from Mahres sediment compared to other studied sites and control treatment. We did not note any significant (p > 0.05) effect of elutriate preparations on clam metamorphosis; conversely, survival in this stage was significantly affected in three sites (Monastir: 59.0 ± 6.7%; Zarat: 52.1 ± 6.3%, and Mahres: 64.1 ± 5.4%) compared to the control (83.2 ± 4.7%). This work shows that both clam embryos and larvae are sensitive to contaminants and can be used to evaluate sediment contamination and monitor pollution.  相似文献

Planktic foraminiferal faunas from different environments in the Arabian Sea were size fractionated using 14 sieves with meshes between 100 and 710 μm, to assess the effect of the sieve mesh size cut off level on the faunal composition and to determine the size frequency distribution of individual species. Nine samples from a plankton pump and a towed net, a sediment trap, a box-core and a piston core were selected, to cover living and settling flux faunas as well as fossil faunas from the sediment. In living faunas, most species show an exponential size frequency distribution, with highest numbers in the finest interval of the size spectrum. In sediment trap and core samples, individual species size frequency distributions may consist of: (1) an exponential distribution of relatively small pre-adult specimens; (2) a Gaussian-shaped distribution of larger specimens, which may be classified as adult or terminal; or (3) a combination of both. The distributions are separated using a best fit technique. The composition of the total planktic foraminiferal fauna strongly changes along the size spectrum. Dominant taxa in >355 μm fractions are Orbulina universa, Globorotalia menardii, Globorotalia tumida, Globigerinella siphonifera and Globigerinoides sacculifer, in 125–355 μm fractions Globigerina bulloides, Globigerinoides ruber, Neogloboquadrina dutertrei and Globigerinita glutinata, and in <125 μm fractions Dentigloborotalia anfracta, Tenuitella compressa, Tenuitella iota, Turborotalita quinqueloba and the immature specimens of larger species. Consequently, the choice of the sieve mesh size strongly determines the percent composition of the assemblage and in turn the paleoceanographic interpretations based on these counts. Species richness and the Shannon diversity increase with decreasing sieve mesh size, while equitability generally decreases with decreasing size. In the water column approximately 60% of the fauna (>100 μm) is present in the 100–125 μm fraction and 1–6% is larger than 250 μm. In samples representing a settling flux (sediment trap and sediment samples) 29–57% of the fauna is present in the 100–125 μm fraction, while 6–23% is larger than 250 μm. Size frequency distributions of the dextral Neogloboquadrina complex (= Neogloboquadrina dutertrei and Neogloboquadrina pachyderma + P–D intergrades) show a bimodal pattern; a smaller peak reflecting dextral Neogloboquadrina pachyderma, and a larger peak of adult Neogloboquadrina dutertrei. By applying a best fit technique to the data, the two species may be separated from each other. In size fractions larger than 150 μm most species have reached the adult stage of ontogeny and we recommend this mesh size for standard faunal analysis. In addition, sieve mesh sizes of 125 and 250 μm have to be used to obtain a reliable estimate of the abundance of small and large species, respectively.  相似文献

Giardia lamblia trophozoites frequently are associated with mucus in vivo. We investigated the effects of human intestinal mucus on parasite attachment and survival in vitro. All samples of mucus from the duodenum and ileum (from four humans and two rabbits) enhanced attachment at 100 μm/ml. Attachment increased with mucus concentrations from 1 to 1000 μg/ml but declined toward the unstimulated level at concentrations above 1000 μg/ml. Mucus from the small intestine also promoted the survival of the parasites during the 2-h incubation. In contrast, colonic mucus promoted survival, but inhibited attachment. Fractionation of mucus from the human small intestine by cesium chloride equilibrium density gradient ultracentrifugation revealed that both attachment- and survival-promoting activities were in the low density, protein-rich fraction. The high density fractions containing the mucins were devoid of activity. Thus, a non-mucin fraction of mucus from the human small intestine may promote colonization by G. lamblia.  相似文献

Soil aggregates, with complex spatial and nutritional heterogeneity, are clearly important for regulating microbial community ecology and biogeochemistry in soils. However, how the taxonomic composition and functional attributes of N-cycling-microbes within different soil particle-size fractions under a long-term fertilization treatment remains largely unknown. Here, we examined the composition and metabolic potential for urease activity, nitrification, N2O production and reduction of the microbial communities attached to different sized soil particles (2000–250, 250–53 and <53 μm) using a functional gene microarray (GeoChip) and functional assays. We found that urease activity and nitrification were higher in <53 μm fractions, whereas N2O production and reduction rates were greater in 2000–250 and 250–53 μm across different fertilizer regimes. The abundance of key N-cycling genes involved in anammox, ammonification, assimilatory and dissimilatory N reduction, denitrification, nitrification and N2-fixation detected by GeoChip increased as soil aggregate size decreased; and the particular key genes abundance (e.g., ureC, amoA, narG, nirS/K) and their corresponding activity were uncoupled. Aggregate fraction exerted significant impacts on N-cycling microbial taxonomic composition, which was significantly shaped by soil nutrition. Taken together, these findings indicate the important roles of soil aggregates in differentiating N-cycling metabolic potential and taxonomic composition, and provide empirical evidence that nitrogen metabolism potential and community are uncoupled due to aggregate heterogeneity.  相似文献

Wheat straw was pretreated with saturated lithium chloride in 4 m hydrochloric acid at 50°C for 1 h, then hydrolysed at 100°C for 1 min, to give 84% conversion to monosaccharides. Particle sizes, 150–355 mesh, were easily hydrolysed. Samples pretreated with saturated lithium chloride in 1 m hydrochloric acid at 27°C for 24 h were hydrolysed by Trichoderma viride cellulase (MVA 1284) [1,4-(1,3;1,4)-β-d-glucan 4-glucanohydrolase, EC 3.2.1.4] to give 20–23% monosaccharides for particle sizes of 150–250 mesh, and 82–95.4% for particle sizes of 250–355 mesh.  相似文献

Oxidative stress negatively affects the in vitro maturation (IVM) of oocytes. Procyanidin B1 (PB1) is a natural polyphenolic compound that has antioxidant properties. In this study, we investigated the effect of PB1 supplementation during IVM of porcine oocytes. Treatment with 100 μM PB1 significantly increased the MII oocytes rate (p <0.05), the parthenogenetic (PA) blastocyst rate (p <0.01) and the total cell number in the PA blastocyst (p < 0.01) which were cultured in regular in vitro culture (IVC) medium. The PA blastocyst rate of regular MII oocytes activated and cultured in IVC medium supplemented with 100 and 150 μM PB1 significantly increased compared with control (p < 0.01 and p < 0.05). We also evaluated the reactive oxygen species (ROS) levels, mitochondrial membrane potential (Δψm) levels, glutathione (GSH) levels, and apoptotic levels in MII oocytes and cumulus cells following 100 μM PB1 treatment. The results showed that the PB1 supplementation decreased ROS production and apoptotic levels. In addition, PB1 was found to increase Δψm levels and GSH levels. In conclusion, PB1 inhibited apoptosis of oocytes and cumulus cells by reducing oxidative stress. Moreover, PB1 improved the quality of oocytes and promoted PA embryo development. Taken together, our results suggest that PB1 is a promising antioxidant additive for IVM of oocytes.  相似文献

This investigation assessed the effect of soil particle size on soil ingestion estimates of children residing at a superfund site. Earlier research indicated that wide intertracer variability in soil ingestion estimates are based on soil concentrations with a soil particle size of 0 to 2?µm was markedly reduced when the estimates were based on soil tracer concentrations for a soil particle size of 0-250?µm. The reduced intertracer variation was principally attributed to changes in soil concentrations of only three of the soil tracers (i.e., Ce, La, Nd) which became concentrated in the finer particle size by approximately 2.5 to 4.0-fold. It was hypothesized that the intertracer agreement in soil ingestion estimates may continue to improve if the estimates are based on concentrations of tracers at finer particle sizes assuming that children ingest finer particles and that the above three tracers would continue to be further concentrated in the finer sized soil particles. The principal findings indicate: 1. The soil concentrations of Al, Si, and Ti do not increase at the two finer particle size ranges measured. 2. The soil concentrations of Ce, La, and Nd increased by a factor 2.5 to 4.0 in the 100 to 250?µm particle size range when compared with the 0 to 2?µm particle size range. No further substantial increase in concentration was observed in the 53 to 100 |jm particle size range. 3. The soil ingestion estimates are consistently and markedly changed only between the estimates based in 0 to 2?µm and 100 to 250?µm for Ce, La, and Nd. These changes reduced the intertracer variability in estimating soil ingestion, suggesting that the children eat finer soil particle sizes. 4. Because the particle sizes for all tracers (except Zr) were only modestly affected at the 53 to 100?µm range, it was not possible to confidently resolve the particle size of soil ingested by the children. 5. Residual intertracer variability in soil ingestion estimates based on Ce, La, Nd are likely to be significantly affected by non-food, non-soil sources of these tracers (i.e., source error). 6. Soil ingestion estimates of this study will be more reliable when derived from the finer-sized particles.  相似文献

The diterpenoid furanolactone (columbin) from Aristolochia albida inhibited growth of culture forms of Trypanosoma brucei. In vitro analysis of the compound at 5–250 μg/ml showed complete lysis of the parasites within 10–20 minutes post incubation. At 50 μg/ml, columbin killed about 50% of the parasites which initially appeared swollen under phase contrast microscopy. Also the total amount of cholesterol diminished dose-dependently in the presence of 10–100 μg/ml of columbin after a 3-day incubation period.

In vivo analysis of the compound in T. brucei-infected mice revealed that 25 mg/kg administered for 3 consecutive days, completely cleared the parasites from the peripheral circulation. However, columbin could not clear parasites in the cerebrospinal fluid.  相似文献

Investigations were undertaken to study the nature and behaviour of P. clandestina in soil. The pathogen was recovered only from soil sievings 250–499 μm and 500 μm–0.99 mm, containing small root fragments. In soil the introduced inoculum of the fungus was incapable of saprophytically and competitively colonizing the dead cotyledons of subterranean clover used as bait material. Exposure of the inoculum to increasing numbers of microbes by adding greater proportions of nonsterile fields, oil to the growth medium of the plant had no significant effect on survival rate and fresh shoot weight of subterranean clover. However, microbes present in the field soil reduced the severity of root rot of subterranean clover. P. clandestina was, able to spread between 15–30 mm through pasteurized soil within a period of 20 days.  相似文献

Protocols are now available for seed harvest, storage and germination of several mesohaline and polyhaline species; however, low seedling survival rates point to the need for an increased understanding of factors affecting seedling establishment. Depth of seed burial in sediments and initial seedling growth rates are shown to be limiting factors for photosynthetic competency of Ruppia maritima and Potamogeton perfoliatus. Seedling emergence is inversely proportional to planting depth on sediments ranging in grain size from coarse sands (850 μm) to silt (63 μm). Less than 6% of the seeds of either species emerged when buried to a depth of 3 cm in test sediments. Germination was greatest for seeds placed on the surface of sediments; however, these seedlings were subject to displacement because of the weak and fragile roots produced during early growth. Fine sediments may be more favorable for R. maritima seedling establishment, because seedling emergence and height decreased with increasing sediment grain size. Potamogeton perfoliatus seedlings seem to be more tolerant of a wider range of sediment grain sizes than R. maritima as indicated by the lack of an effect of sediment grain size on P. perfoliatus seed emergence, seedling height, and biomass. Increasing nutrients stimulated seedlings of both species; however, even at the highest concentrations tested, growth, as determined by shoot elongation and leaf and root formation, slowed within 7–10 days. This suggests factors other than mineral nutrients and light limit growth or that growth shifts from aboveground biomass production to belowground vegetative spread.  相似文献

为了明确草方格人工固沙造林植被恢复过程中土壤颗粒组成、分形维数及对土壤理化性质的影响,以腾格里沙漠东南缘2016年(1 a)、2013年(4 a)和1987年(30 a)草方格固沙林为研究样地,以周围流动沙地为对照(CK),研究了草方格固沙造林后植被恢复过程中土壤颗粒组成、分形维数及与土壤理化性质的作用关系.结果表明: 100~250、250~500 μm土壤颗粒含量较高,分别为42.5%~80.1%、12.5%~42.2%;50~100 μm土壤颗粒含量居中,为0.2%~20.8%;<2和2~50 μm的土壤颗粒含量次之,分别在0~1.3%和0~22.7%;而500~1000 μm的土壤颗粒含量较低,在0.3%以下.<2和2~50 μm土壤颗粒仅在30 a固沙林有分布;50~100 μm土壤颗粒分布为30 a最高,4 a和1 a居中,而CK最低;100~250 μm土壤颗粒分布依次为4 a>1 a>CK>30 a;250~500 μm土壤颗粒分布为CK>1 a>4 a>30 a;但500~1000 μm土壤颗粒在各样地分布均较少,且不同样地之间无显著差异.研究区土壤颗粒分形维数为0.54~2.59,并且不同样地间存在显著差异,表现为30 a最高,4 a与1 a居中,而CK最低.土壤颗粒分形维数与土壤黏粒、粉粒、极细砂粒含量呈极显著正相关,而与土壤中砂粒呈极显著负相关.土壤颗粒分形维数与土壤电导率、有机碳、全氮和碳氮比均呈极显著正相关,而与土壤pH和含水量无相关性.土壤中<2、2~50、50~100 μm颗粒与土壤电导率、有机碳、全氮和碳氮比均呈极显著正相关,而250~500 μm土壤颗粒与上述4个土壤指标和土壤含水量呈显著负相关.500~1000 μm土壤颗粒与土壤含水量亦呈极显著负相关.在腾格里沙漠东南缘地区利用草方格进行人工固沙植被建设,可有效促进土壤颗粒细粒化,长期演变导致土壤黏粒和粉粒及土壤分形维数显著增加,促使土壤有机碳和全氮含量提高,有利于土壤理化性质改善和促进沙漠化治理.  相似文献

The allelopathic effects of fresh tissue, dry powder and aqueous extracts of three macroalgae, Ulva pertusa, Corallina pilulifera and Sargassum thunbergii, on the growth of the dinoflagellates Heterosigma akashiwo and Alexandrium tamarense were evaluated using coexistence culture systems in which concentrations of the three macroalga were varied. The results of the coexistence assay showed that the growth of the two microalgae was strongly inhibited by using fresh tissue, dry powder and aqueous extracts of the three macroalga; the allelochemicals were lethal to H. akashiwo at relatively higher concentrations of the three macroalga. The macroalgae showing the most allelopathic effect on H. akashiwo and A. tamarense using fresh tissue were U. pertusa and S. thunbergii, using dry powder were S. thunbergii and U. pertusa, and using aqueous extracts were U. pertusa and C. pilulifera. We also examined the potential allelopathic effect on the two microalgae of culture filtrate of the three macroalga; culture medium filtrate initially exhibited no inhibitory effects when first added but inhibitory effects became apparent under semi-continuous addition, which suggested that continuous release of small quantities of rapidly degradable allelochemicals from the fresh macroalgal tissue were essential to effectively inhibit the growth of the two microalgae.  相似文献

1. Wind‐induced sediment resuspension in shallow lakes affects many physical and biological processes, including food gathering by zooplankton. The effects of suspended sediment on clearance rate were determined for a dominant cladoceran, Daphnia carinata, and calanoid copepod, Boeckella hamata, in Lake Waihola, New Zealand. 2. Animals were incubated at multiple densities for 4 days in lake water containing different amounts of suspended lake sediment. Rates of harvest of major food organisms were determined for each sediment level (turbidity) from changes in net growth rate with grazer density. 3. Daphnia cleared all food organisms 7–40 μm in length at similar rates, but was less efficient in its removal of free bacteria, phytoplankton <7 μm, and large cyanobacterial filaments. Elevation of sediment turbidity from 2 to 10 nephelometric turbidity units (NTU) (63 mg DW L?1 added sediment) reduced Daphnia clearance of phytoplankton, heterotrophic flagellates and ciliates by 72–100%, and of amoebae and attached bacteria by 21–44%. Further inhibition occurred at higher turbidity. 4. Boeckella hamata removed microzooplankton primarily, rather than phytoplankton. The rate at which it cleared rotifers was reduced by 56% when turbidity was increased from 2.5 to 100 NTU. 5. In the absence of macrozooplankton, algal growth increased with sediment turbidity, suggesting that sediment also inhibits rotifer grazing. 6. As mid‐day turbidity in Lake Waihola is ≥10 NTU about 40% of the time, sediment resuspension may play a major role in moderating energy flow and structuring pelagic communities in this lake.  相似文献

Lauric Acid Exhibits Antifungal Activity Against Plant Pathogenic Fungi   总被引:3,自引:0,他引:3  
This study aimed at examining the effects of the saturated fatty acid lauric acid on mycelial growth of Rhizoctonia solani and Pythium ultimum and on infection of barley seedlings with Blumeria graminis f. sp. hordei. Mycelial growth of R. solani and P. ultimum in agar culture was significantly reduced by lauric acid at concentrations of 100 μm and above, while no fungal growth occurred in liquid culture at concentrations above 50 μm . Application of lauric acid at concentrations ranging from 250 to 1000 μm to barley leaves before or after inoculation with B. graminis f. sp. hordei led to significant reductions in infection. This study provides the first report of the activity of lauric acid against plant pathogenic fungi and indicates the need for investigation of its mechanism of action.  相似文献

Fritillaria ussuriensis (FU, derived from the bulbs of various species of the genus Fritillaria, including Fritillaria thunbergii Miq.) is used in herbal medicine to treat conditions such as eczema, skin burns, and frostbite. In this study, we investigated the mechanism of the anti-allergy effect of FU. FU extract (80 mg/kg), orally administered to Sprague-Dawley (SD) rats, significantly inhibited the passive cutaneous anaphylaxis (PCA) reaction. It inhibited the compound 48/80-induced release of histamine from rat peritoneal mast cells in a concentration-dependent manner. Significant inhibitory effects of the FU extract on IL-6, IL-8, and TNF-α (1, 10, and 100 μg/mL) were observed in HMC-1 cells. Treatment with FU attenuated PMA plus A23187-induced phosphorylation of all three MAPKs, especially at concentrations of 10 and 100 μg/mL. Further, it (80 mg/kg) led to significant inhibition of mast-cell accumulation in ear tissue at the chronic phase. These results indicate that it inhibits allergic reactions.  相似文献

Although well known for delivering various pharmaceutical agents, liposomes can be prepared to entrap gas rather than aqueous media and have the potential to be used as pressure probes in magnetic resonance imaging (MRI). Using these gas-filled liposomes (GFL) as tracers, MRI imaging of pressure regions of a fluid flowing through a porous medium could be established. This knowledge can be exploited to enhance recovery of oil from the porous rock regions within oil fields. In the preliminary studies, we have optimized the lipid composition of GFL prepared using a simple homogenization technique and investigated key physico-chemical characteristics (size and the physical stability) and their efficacy as pressure probes. In contrast to the liposomes possessing an aqueous core which are prepared at temperatures above their phase transition temperature (Tc), homogenization of the phospholipids such as 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) or 1,2-distearoyl-sn-glycero-3-phosphocoline (DSPC) in aqueous medium below their Tc was found to be crucial in formation of stable GFL. DSPC based preparations yielded a GFL volume of more than five times compared to their DPPC counter part. Although the initial vesicle sizes of both DSPC and DPPC based GFL were about 10 μm, after 7 days storage at 25°C, the vesicle sizes of both formulations significantly (p < 0.05) increased to 28.3 ± 0.3 μm and 12.3 ± 1.0 μm, respectively. When the DPPC preparation was supplemented with cholesterol at a 1:0.5 or 1:1 molar ratio, significantly (p < 0.05) larger vesicles were formed (12–13 μm), however, compared to DPPC only vesicles, both cholesterol supplemented formulations displayed enhanced stability on storage indicating a stabilizing effect of cholesterol on these gas-filled vesicles. In order to induce surface charge on the GFL, DPPC and cholesterol (1: 0.5 molar ratio) liposomes were supplemented with a cationic surfactant, stearylamine, at a molar ratio of 0.25 or 0.125. Interestingly, the ζ potential values remained around neutrality at both stearylamine ratios suggesting the cationic surfactant was not incorporated within the bilayers of the GFL. Microscopic analysis of GFL confirmed the presence of spherical structures with a size distribution between 1–8 μm. This study has identified that DSPC based GFL in aqueous medium dispersed in 2% w/v methyl cellulose although yielded higher vesicle sizes over time were most stable under high pressures exerted in MRI.  相似文献

The ontogenetic diet shifts and age specific ability of the two cladoceran species Moina macrocopa and Ceriodaphnia cornuta to derive energy from ciliated protists have been investigated in laboratory. The postembryonic developmental rates and life table demography (longevity, age and size at first reproduction, fecundity and intrinsic rate of natural increase) of the cladocerans have been elucidated on algae (Chlorella vulgaris) and the ciliated protists (Tetrahymena pyriformis, Colpoda (c.f.) steini) as food. For either of the cladoceran, the somatic growth rate and average body size at first reproduction were higher with algal diet. During initial stages of development (0–5 days), either cladoceran realized higher rate of somatic growth on algal diet, subsequently ciliated protists supported significantly higher growth rate than the alga. Algal and ciliate diets did not differ in maximum body size (C. cornuta: 539–554 μm; M. macrocopa: 1274.8–1309 μm) reached by either of the cladocerans. The maximum body sizes were larger than size at first reproduction with either of the ciliated protists, however, with algal diet the maximum body sizes did not differ from the size at first reproduction in each case. In case of C. cornuta the generation time (20.5 ± 0.3 days on ciliate; 15.6 ± 0.17 days on algal diet), reproductive rates (net reproductive rate: 20.05 ± 3.2 on ciliate; 15.5 ± 1.2 on algal diet), and average life expectancy at hatching (27 ± 0.8 days on ciliate; 22.7 ± 0.71 days on alga) were higher, whereas the size at first reproduction (482 μm on ciliate; 521 μm on alga) was smaller with the ciliate than with an algal diet. The algal and the ciliate diets did not differ in survival (life expectancy at hatching: 9.2 ± 0.7 days) and fecundity (NRR: 23.6 ± 2.4) for M. macrocopa. The two ciliates used in the experiment did not differ in their performance as food source for either cladoceran species. Our results suggest that both the cladoceran species are able to utilize smaller ciliate (e.g., T. pyriformis, C. Seedling establishment ofCastanopsis cuspidata var.sieboldii Nakai andPersea thunbergii Kosterm. (Machilus thunbergii Sieb. et Zucc.) in early succession was studied on the lava and the scoria of the 1962 eruption on Miyake-jima Island of the Izu Islands. While tall individuals (> 1 m) ofP. thunbergii were found on both the lava and the scoria, those ofC. cuspidata (> 1 m) were found only on the lava. The height growth rate ofC. cuspidata was very low on the scoria, and there was no individual ofC. cuspidata taller than 0.4 m on the scoria. The proportion of yellow leaved individuals was much higher (37.1%) inC. cuspidata than inP. thunbergii (4.0%) on the scoria, while it was similar between the two species on the lava.Persea thunbergii seemed to be able to establish on both the lava and the scoria, butC. cuspidata established only on the lava. The differences in growth rate and vigor between the two species on the lava and the scoria in early succession may influence later stages of succession.  相似文献

The influence of fine sediment (<63 m diameter) upon the assimilation rates of the snail Potamopyrgus antipodarum, and the mayfly Deleatidium sp. were determined by allowing individuals to feed upon 14C radiolabelled periphyton which had been contaminated with varying quantities of sediment (sediment:food ratios of: 0:1, 1:1, 5:1, 10:1, 50:1, 100:1 [dry weight]). For both grazers, the assimilation rate falls in direct proportion to the sediment fraction in the (sediment+food) matrix. In a second experiment the growth of P. antipodarum was monitored over 3 months when fed organic matter that had been contaminated with fine sediment at ratios of 0:1, 1:1, 5:1, 10:1, 50:1, 100:1, 500:1. In contrast to the monotonic relation between sediment and short-term assimilation, growth rates (mm shell height d–1) were highest at intermediate levels of sediment contamination (5:1 and 10:1 by dry weight) and lowest in the treatment with no sediment added. Growth rates were significantly lower, and mortality high, at sediment:food contamination ratios above 50:1. The reasons for the contrast between the results from the short-term and the long-term experiments are unknown at present, but the fact that snail growth was greatest at intermediate levels of sediment contamination might indicate that they derive trace nutrients from ingested sediment.  相似文献

The role of mannose receptors from hepatic sinusoidal endothelium (HSE) in liver colonization by B16 melanoma (B16M) cells was studied. The expression of high mannose-type oligosaccharides on the surface of B16M cells was enhanced by in vitro treatment with 1-deoximannojirimycin (1-DMM). There was a significant (P < 0.01) enhancement of hepatic metastasis when B16M cells were 1-DMM-treated before being intrasplenically injected into C57BL/6J mice. Intraperitoneal administration of 5 mg/kg recombinant human interleukin-1 receptor antagonist (rHuIL-1Ra) inhibited the 1-DMM-induced enhancement of metastasis. Expression of high mannose-type oligosaccharides on the surface of 1-DMM-treated B16M cells and their in vitro adhesion to the HSE was significantly correlated (R = 0.82). The addition of either 100 μg/ml mannan or paraformaldehyde (PFA)-fixed 1-DMM-treated B16M cells to cultured HSE for a period of 12 h significantly (P < 0.01) increased the release of IL-1β from the HSE compared to that liberated by the HSE incubated with either basal medium or PFA-fixed untreated B16M cells. The same HSE treatments also significantly (P < 0.01) increased the degree of adhesion of other B16M cells to HSE, being abrogated by anti-mouse vascular cell adhesion molecule-1 (VCAM-1) antibodies. The conditioned media from HSE cultures, activated by PFA-fixed, 1-DMM-treated B16M cells significantly (P < 0.01) increased B16M cell proliferation when compared to conditioned media from HSE cultures incubated with PFA-fixed, untreated B16M cells. Thus, 1-DMM treatment of B16M cells enhanced the development of hepatic metastasis by IL-1-dependent mechanisms. The mechanism is consistent with in vitro mannose receptor-mediated melanoma cell attachment to the HSE, which subsequently upregulates IL-1β release, VCAM-1-dependent adherence, and melanoma growth factor(s) release by HSE.
