Following a 7.5-h transport haul, juvenile pallid sturgeon (Scaphirhynchus albus) showed a small but significant increase in plasma cortisol to 4.7 ng ml(-1) but similar increases did not occur after fish were handled in a net held in the air for 30 s. Subsequent experiments on yearling pallid sturgeon and hybrid pallidxshovelnose (S. albusxplatorynchus) sturgeon using the same 30-s handling stressor failed to evoke increases in plasma cortisol, lactate or glucose. Plasma cortisol increased significantly from about 2 to 13-14 ng ml(-1) in both pallid and hybrid sturgeon during a 6-h severe confinement stressor with handling. Plasma cortisol in 2-year-old pallid sturgeon subjected to the same stressor demonstrated a linear pattern of increase during the initial 1 h. Plasma lactate increased from 1.11 to about 2.11 mmol l(-1) in hybrid sturgeon during the first hour of severe confinement but did not change throughout the entire confinement period in pallid sturgeon. A significant increase in plasma cortisol to 5.4 ng ml(-1) in 2-year-old pallid sturgeon 1 h after being subjected to 30 s handling at 19:00 h but not at 07:00 or 13:00 h suggests that a small diurnal variation in their stress response may exist. Although both pallid and hybrid sturgeons were responsive to stress, they exhibited very low physiological responses compared with those following equivalent stressors in most teleostean fishes or another chondrostean, the paddlefish (Polyodon spathula). Reasons for the apparent low responses to handling and confinement in scaphirhynchid sturgeons are not known but may relate to their evolutionary history, neuroendocrine mechanisms involved in their corticosteroid responses, or anatomy of their interrenal tissue structure.  相似文献

The study objectives were to determine the predominant manatee glucocorticoid; validate assays to measure this glucocorticoid and adrenocorticotropic hormone (ACTH); determine diagnostic thresholds to distinguish physiological vs. pathological concentrations; identify differences associated with sex, age class, female reproductive status, capture time, and lactate; and determine the best methods for manatee biologists and clinicians to diagnose stress. Cortisol is the predominant manatee glucocorticoid. IMMULITE 1000 assays for cortisol and ACTH were validated. Precision yielded intra‐ and inter‐assay coefficients of variation for serum cortisol: ≤23.5 and ≤16.7%; and ACTH: ≤6.9 and ≤8.5%. Accuracy resulted in a mean adjusted R2≥0.87 for serum cortisol and ≥0.96 for ACTH. Assay analytical sensitivities for cortisol (0.1 µg/dl) and ACTH (10.0 pg/ml) were verified. Methods were highly correlated with another IMMULITE 1000 for serum cortisol (r=0.97) and ACTH (r=0.98). There was no significant variation in cortisol or ACTH with sex or age class and no correlation with female progesterone concentrations. Cortisol concentrations were highest in unhealthy manatees, chronically stressed by disease or injury. ACTH was greatest in healthy free‐ranging or short‐term rehabilitating individuals, peracutely stressed by capture and handling. Cortisol concentrations ≥1.0 µg/dl were diagnostic of chronic stress; ACTH concentrations ≥87.5 pg/ml were diagnostic of peracute stress. In healthy long‐term captive manatees, cortisol (0.4±0.2 µg/dl) and ACTH (47.7±15.9 pg/ml) concentrations were lower than healthy free‐ranging, short‐term rehabilitated or unhealthy manatees. Capture time was not significantly correlated with cortisol; ACTH correlation was borderline significant. Cortisol and ACTH were positively correlated with lactate. In vivo experiments were conducted to examine the haematology of juveniles from two relic bony fishes, Atlantic sturgeon Acipenser oxyrhinchus and shortnose sturgeon Acipenser brevirostrum . Oxygen transport characteristics (haematocrit, haemoglobin and mean erythrocytic haemoglobin concentration), ionic composition (Na+, Cl, K+ and osmolality), metabolite concentration (lactate, cortisol and glucose) and protein content in blood were measured or calculated at rest and during recovery from forced activity. Under resting conditions, plasma osmolality and concentrations of Na+, Cl, lactate, cortisol and total protein were significantly different between Atlantic and shortnose sturgeon. All other resting variables were not different between species. Following forced activity, plasma lactate levels were significantly higher in both species than at rest. Plasma cortisol levels in both species were only significantly higher 1 h following forced activity compared to resting values. Plasma lactate levels were significantly higher in Atlantic sturgeon than in shortnose sturgeon, but these levels returned to resting levels by 1 h in both species. Cortisol increases were greater in shortnose sturgeon than in Atlantic sturgeon. In general, oxygen transport characteristics, blood glucose, plasma protein and plasma osmolality were not altered by forced activity in either sturgeon species. Overall, both species had reduced responses ( i.e . the magnitude of changes in measured variables) to forced activity compared with teleosts.  相似文献

Data on the blood chemistry of a chondrostean fish, the Adriatic sturgeon ( Acipenser naccarii ), are reported as measured with different sampling procedures, and as related to rearing conditions and age. Serum Cortisol, glucose, osmolality, Na+, Cl-, Ca2+ and total protein concentrations were measured. Reference values for the blood chemistry of farmed sturgeon were measured on samples from resting undisturbed animals collected via a chronic indwelling catheter in the dorsal aorta that was implanted under anaesthesia. Following 24h recovery from catheterization, serum Cortisol, glucose and osmolality levels were 9.4 ng/ml, 58.8 mg/dl and 261.4 mOsm/kg, respectively. Furthermore, blood samples collected with the chronic indwelling catheters indicated that the surgical procedure of cannulation caused a stress response, with physiological changes that followed a pattern like that described in teleosts. Cortisol, glucose and osmolality were more sensitive to stress than the other variables measured. Sampling by cardiac puncture tended to be associated with elevated serum Cortisol levels in older, larger sturgeon, but not in young fish. Greater capture, confinement and handling stress in older, larger, sturgeon may have been responsible for this and other age-related differences in blood chemistry values measured following cardiac puncture. Within the same age class, both rearing conditions and temperature affected Cortisol, sodium and total protein concentrations significantly. Anaesthesia did not appear to reduce the degree of stress associated with cardiac puncture but altered serum ion concentrations.  相似文献

Data on the concentrations of some blood constituents of captive Adriatic sturgeon, Acipenser naccarii, a primitive bony fish, are reported. Serum osmolality, Na+, K+, Cl, Ca2+, cortisol, glucose and total protein concentrations were measured. The effects of anaesthesia, temperature, crowding and prolonged handling stress were tested on a group of 12 4-year-old sturgeons sampled repeatedly. The anaesthetic dose of MS 222 (140 mg l−1) induced significant osmolality elevation in the sturgeon. After exposure to colder temperature (17 versus 25°C), cortisol and Cl concentrations significantly decreased. The cultured sturgeon did not seem susceptible to crowding and prolonged handling stress, since neither the serum cortisol and glucose levels nor the other blood parameters were affected by these stressors. Results are compared with the few available data on other chondrostean fish and with those on teleosts.  相似文献

The effects of acute stressors on physiological responses of juvenile great sturgeon or beluga Huso huso L. were investigated in two experiments. In the first experiment, fish were handled by placing them in containers at either low density (LD, one fish l?1) or high density (HD, four fish l?1) for 60 s. Concentrations of plasma cortisol, glucose and lactate were determined from blood collected at 0, 1, 3, 6 and 12 h after application of the stressor. Plasma cortisol concentrations increased after the disturbance in H. huso from both handling treatments, but changes were not significant. Plasma glucose rose significantly by 22·9 and 31·6% in LD and HD handling treatments, respectively, after 3 h. Significant increases in plasma lactate occurred within 1 h in both treatment groups, but that of the HD group was much higher. In the second experiment, fish were held at two different densities, LD (2 kg m?2 tank bottom surface area) and HD (7 kg m?2), for 8 weeks and then subjected to an aerial emersion handling stressor in a net for 60 s; blood samples were taken before handling (resting, 0 h) and at 1, 3, 6 and 9 h after handling. Plasma cortisol increased significantly in fish from the HD treatment from 8·8 ± 0·3 to 19·2 ± 2·4 ng ml?1 (mean ±s.e. ) by 1 h after stress, but post‐handling changes in the LD group were not significant. Significant increases in both plasma glucose and lactate were observed by 1 h in both treatment groups, with peak levels of plasma glucose evident at 3 h [69·4 ± 2·9 and 60·9 ± 1·7 mg dl?1 (mean ±s.e .) in LD and HD groups, respectively]. Plasma glucose levels were significantly higher in the LD group than in the HD group at 3 and 6 h. Post‐handling haemoglobin content increased by 1 h and white blood cell numbers were reduced by 3 and 6 h in the HD treatment group compared with resting values, but changes in these blood features in the LD group were not significant. Acute handling did not affect haematocrit in either treatment. The results suggest that H. huso is relatively resistant to handling and confinement, and could tolerate normal hatchery practices associated with aquaculture. Because changes in cortisol concentrations were relatively low compared with those in most teleosts, glucose and lactate concentrations may be more useful as stress indicators in juvenile H. huso. This study also demonstrated that prior exposure to a chronic stressor, specifically high stocking density, could alter the physiological response to subsequent acute handling in H. huso.  相似文献

The effects of time of day and water temperature on the acute physiological stress response were investigated in young-of-the-year green sturgeon (Acipenser medirostris). The response to a 1-min air-emersion stressor was assessed during the day (08.00 h) and at night (20.00 h), as well as after acclimation to either 11 degrees C or 19 degrees C. Blood samples were collected prior to stress and at several times after exposure to the stressor, and plasma concentrations of cortisol, lactate, and glucose were determined. The magnitudes of cortisol (19.1 ng ml(-1) vs. 4.9 ng ml(-1)) and lactate (190.6 mg l(-1) vs. 166.7 mg l(-1)) were significantly higher in fish stressed at night when compared with the day. There were no significant differences in glucose levels between time periods. Although, acclimation temperature did not affect peak cortisol concentrations (56.7 and 50.3 ng ml(-1) at 11 degrees C and 19 degrees C, respectively), the duration of the response was significantly extended at 11 degrees C. Post-stressor lactate increases were similar between temperature groups, but at 11 degrees C post-stressor glucose levels were significantly increased through 6 h, suggesting stressor-induced glycogenolysis and gluconeogenesis or decreased glucose utilization. These data demonstrate that the physiological stress response in green sturgeon is modified by both time of day and temperature.  相似文献

Cortisol and lactate are classic indicators of stress in fishes and their interactive effects on metabolism during recovery from stress have recently become a subject of more intense study. We examined how stressing green sturgeon through acid infusion affected the cortisol response and lactate metabolism in green sturgeon (Acipenser medirostris). Both lactic acid (0.3 M) and HCl (0.3 N) infusion (infusion volumes 1.5 ml kg(-1)) elicited an immediate cortisol response (21.61+/-4.61 ng ml(-1) and 17.50+/-3.00 ng ml(-1), respectively). Lactic acid prolonged the cortisol response compared to HCl (90 min vs. 25 min). Neutralized lactate (0.23 M; with 1 N NaOH; final pH 7.8) and NaCl (0.9%) infusion (infusion volumes 1.5 ml kg(-1)) did not affect plasma cortisol. Sturgeon infused with lactic acid showed a faster rate of lactate disappearance from plasma than those with neutralized lactic acid. We relate these findings to lactate metabolism following exercise, acid-infusion and air immersion stress in fishes.  相似文献

The effect of body size on various hematological variables was examined in juvenile shortnose sturgeon (Acipenser brevirostrum) ranging in mass from 38 to 730 g. The blood was examined for differences in plasma ionic composition (Na+, K+, Cl?), blood oxygen carrying capacities (hemoglobin, hematocrit), and plasma metabolite concentrations (lactate, glucose), before and following a standard 5‐min chasing stress. All measured resting hematological variables were size independent in shortnose sturgeon. After exercise, levels of plasma lactate, potassium, and hemoglobin increased in all fish. Only post‐exercise levels of chloride and hemoglobin changed in a size‐dependent manner; however, the relationships were weak. The general lack of a relationship between body size and hematological variables might reflect the narrow range of fish sizes used in the present study. From a practical perspective, the results suggest that when examining the hematological stress response in juvenile shortnose sturgeon, a range of fish sizes could be used. This is important considering the variability in the growth rates of juvenile shortnose sturgeon under laboratory conditions.  相似文献

Sexually immature rainbow trout were acclimated to small-volume (1 m3) holding tanks and then exposed to short-term stress to examine the relationship between feeding, stress, plasma ghrelin levels and other plasma stress parameters. Plasma ghrelin levels showed an increase 24 h after a single feed, plasma lactate and glucose levels decreased over the same period and plasma cortisol levels were low and constant. One hour of confinement stress resulted in elevations of plasma cortisol, glucose and lactate and depression of plasma ghrelin levels. In a separate experiment, 2 h of confinement stress also depressed feeding immediately after stress, concomitant with increases in plasma cortisol, lactate and glucose; however, in this case there was no change in plasma ghrelin concentrations. A repeat of the 2-h confinement experiment using fish that had not been acclimated to small-volume holding tanks produced a more marked elevation in plasma cortisol and a stronger suppression of feeding post-stress but in this case also, there was no change in plasma ghrelin levels. The results of this study confirm that feeding in rainbow trout is suppressed by confinement stress although the effect is transitory in this domesticated stock. Similar to that in other fishes, plasma ghrelin levels appear to be modulated by feeding status and may be influenced by stress, suggesting an orexigenic role for ghrelin in rainbow trout.  相似文献

This study compared the influence of feeding methods on growth parameters of young‐of‐year Beluga sturgeon Huso huso in a 6‐week trial. Fish with an average weight 150.3 ± 0.8 g (±SE) were stocked into nine circular concrete tanks (30 fish per tank) in an open circular system with water temperature of 18.9°C. All fish were fed by three different feeding methods: (a) hand‐fed (HF), (b) continuously available (automated feeder; AF), (c) half of daily feed provided by hand, and another half by automated feeder (combined feeding). For the hand‐feeding method, fish were fed at 09:00, 14:00, 19:00, and 24:00. The entire automatic feeding groups were fed with the same amount of feed. The mean final body weight was the highest in fish fed by AF compared to fish fed by HF. Body weight increase, condition factor, specific growth rate, and feed conversion ratio did not differ among the feeding groups. Fish fed by AF revealed higher swimming activity than the HF group. No significant changes were found in hematocrit, glucose and total protein concentrations among treatments. The results showed less dependence of growth and physiology of Beluga sturgeon on feeding method, but automated‐feeding was shown to be suitable for sturgeon rearing because of further low labour costs in rearing systems.  相似文献

This study was carried out to investigate the effect of an acute stressor on the variation of some physiological and immunological parameters of Siberian sturgeon (Acipenser baerii) juveniles. Fish, reared in 3 tanks for 10 weeks, were used for this study. The acute stress of fish consisted of 2 min of air exposure stress. Plasma levels of cortisol, glucose, and lactate as well as lysozyme activity in plasma were measured before stress and 1 hr, 3 hr, 6 hr, 9 hr, 12 hr, and 24 hr after stress. The plasma cortisol significantly increased in the highest level 1 hr after stress, yet it gradually declined after 3 hr. The glucose significantly increased only 1 hr after stress. There was no significant difference between plasma lactate prestress and poststress. Moreover, lysozyme activity was enhanced by stress, thus reaching the highest level 9 hr after stress. The results of this study indicate that Siberian sturgeon not only have a rapid response to acute stress, but also a great capacity for recovery from stress, thus returning physiological parameters to prestress levels after 6 hr.  相似文献

Tropical labrids Hemigymnus melapterus sampled underwater had low plasma levels of cortisol, glucose, and lactate. Plasma cortisol levels were elevated by capture stress within 5–6 min, while glucose and lactate levels were not. Plasmalevels of cortisol and glucose increased after 2–4 h of handling and transport to the laboratory. Levels of cortisol and glucose fell with laboratory acclimation back to values similar to those found in wild fish. Parasitism by gnathiid isopods across an order of magnitude of isopod numbers had no effect on plasma levels of cortisol or glucose. Thus, H. melapterus has a stress response similar to that shown by temperate species, and relatively high parasite loads are not apparently stressful to fish in the wild. This may be related to the counterproductive effects of physiological stress responses on the immune system or behaviour-modulated processes that counter parasitic invasion.  相似文献

To evaluate whether stress-response indicators in blood plasma (BP) are similarly reflected in the peritoneal fluid (PF) white sturgeon Acipenser transmontanus were stressed by a 30 min air exposure and pH, PCO2, osmolality, cortisol, glucose and lactate levels measured. Changes in certain stress indicators in the BP (pH, PCO2, osmolality and glucose) also occurred in the PF, while stressor-induced changes in cortisol and lactate were restricted to the BP. Data suggest that PF is a modified ultrafiltrate of the blood and potentially a useful indicator of animal stress.  相似文献

Circadian rhythms of plasma insulin, Cortisol, and glucose concentrations were examined in scotosensitive (reproductively sensitive to inhibitory effects of short daylengths) and scotorefractory male and female Syrian hamsters (Mesocricetus auratus) maintained on short (LD 10:14) and long (LD 14:10) daylengths. The baseline concentration (mean of all values obtained every 4 hr six times of day) of insulin was much greater in female than in male scotosensitive hamsters kept on short daylengths. These differences in insulin concentration may account for the observed heavy fat stores in female and low fat stores in male scotosensitive hamsters kept on short daylengths. The baseline concentrations of Cortisol were approximately equal in both scotosensitive and scotorefractory males held on short and long daylengths, but were relatively low in females held on short daylengths and especially high in scotorefractory females held on long daylengths.

The plasma concentrations of both cortisol and insulin varied throughout the day in many of the groups tested. However, the variations were not equivalent. The circadian variations of cortisol were similar irrespective of sex, seasonal condition and daylength. Peak concentrations generally occurred about 12 hr after light onset. In contrast, the circadian variations of insulin differed markedly. For example in male hamsters, robust daily variations were found in scotosensitive hamsters held on short daylengths but not on long daylengths and in scotorefractory hamsters held on long daylengths but not on short daylengths. Furthermore, the daily peak occurred during the light in the scotosensitive hamsters and during the dark in the scotorefractory animals. Neither the daily feeding pattern (about 60% consumed during dark) nor the daily variations of glucose concentration varied appreciably with seasonal condition or daylength. They do not appear to determine nor directly reflect the variations in cortisol and glucose concentrations. It is postulated that the daily rhythms of cortisol and insulin are regulated by different neural pacemaker systems and that changes in the phase relations of circadian systems account in part for seasonal changes in body fat stores.  相似文献

Lake sturgeon, Acipenser fulvescens, are considered threatened or endangered throughout most of their North American Range. Current hatchery rearing for re-stocking programs utilise conventional methods with little to no understanding of the relationship between rearing conditions and the development of the hypothalamic-pituitary-interrenal (HPI) stress axis. In the present study we examined the effects of substrate type and temperature on the development of the HPI stress axis in prolarval and larval lake sturgeon. Lake sturgeon raised over either gravel or no substrate did not consistently show an increase in whole body cortisol at the prolarval stage. However, after the onset of exogenous feeding a consistent increase in whole body cortisol following a stress was evident. Lake sturgeon larvae raised in gravel substrate demonstrated a sustained increase in whole body cortisol for at least 240 min post stress whereas whole body cortisol in larvae raised in no substrate returned to baseline within 240 min post stress. Lake sturgeon larvae raised at 9, 12 and 15°C exhibited markedly different cortisol responses with baseline whole body cortisol being, 38.6 ± 3, 5.67 ± 0.41 and 25.38 ± 2.84 ng.g−1 respectively. Furthermore, the chase induced increases in whole body cortisol at the larval stage were significantly different for each temperature treatment. These experiments demonstrate that physical environment has a significant impact on the development of the HPI stress axis in lake sturgeon.  相似文献

The response of the brown trout to a single, short ( c . 1 min) incidence of handling stress was monitored for a period of 1 month post-stress. Significant changes were found in feeding behaviour, in the levels of plasma cortisol, glucose and lactate, in the concentration of circulating lymphocytes and in the degree of epidermal mucification. No changes were detected in the growth rate and coefficient of condition, in the levels of plasma thyroxine, in the concentrations of circulating erythrocytes, neutrophils and thrombocytes or in the thickness of the epidermis. The time-course for recovery of each parameter was examined and from this it was concluded that a minimum of 2 weeks was required for complete recovery and a return to normality.  相似文献

This study investigated the effect of high‐dose vitamin C supplementation on growth, tissue ascorbic acid concentrations and physiological response to transportation stress in juvenile silver pomfret, Pampus argenteus (initial average weight 6.2 ± 0.2 g). Three practical diets were formulated to contain 100 (control), 450 and 800 mg ascorbic acid/kg diet, respectively, supplied as l‐ascorbyl‐2‐polyphosphate. Each diet was fed to triplicate groups of fish in circular tanks (3 m ø, 1.5 m depth) (60 fish/tank) for 9 weeks. Growth did not change significantly with dietary vitamin C levels, although an improvement tendency with an increase in vitamin C supplementation was observed. Ascorbic acid concentrations in the liver and muscle of fish fed diets containing graded levels of vitamin C were positively correlated with dietary levels of this vitamin. Tissue ascorbic acid concentrations significantly increased with increasing vitamin C supplementation. After 9 weeks, the fish were subjected to transportation stress for 4 h to determine the influence of high vitamin C supplementation on the physiological response to this stressor. Serum cortisol, glucose and lactate levels significantly increased in stressed fish. Serum cortisol and glucose concentrations after stress were significantly higher in fish fed the control diet (7.91 μg L?1 and 0.80 mm , respectively) than in the other groups. However, there were no significant differences in serum cortisol and glucose levels after stress between the 450 and 800 mg kg?1 diets. No significant change could be found in serum lactate levels after stress among the different treatments. In conclusion, the dietary administration of high doses of vitamin C could reduce stress in silver pomfret and increase the survival of fish under stress conditions.  相似文献

The anaethetic effects of 2‐phenoxyethanol (2‐PE) on possible primary (cortisol level) and secondary (hematological indices and glucose level) stress responses were studied in Persian sturgeon (Acipenser persicus). Fish were first exposed to 0.1, 0.3, 0.5, 0.7, 0.9 and 1.1 ml L?1 2‐PE, and the time to induction (deep anaethesia) and recovery were measured. At a concentration of 0.1 ml L?1, 2‐PE failed to induce deep anaethesia in fish, whereas at concentrations of 0.7, 0.9 and 1.1 ml L?1 all fish were anaethetized within 3 min of exposure. For assessing possible stress effects caused by effective concentrations of 2‐PE, the hematological indices, serum cortisol and glucose were determined in the deeply anaethetized fish as stress indicators. The 2‐PE exposure resulted in significant increases in red blood cell (RBC) values at 0.3 and 0.5 ml L?1; parallel increases in hemoglobin values were also observed at these concentrations (P < 0.01). Moreover, a lower concentration of 2‐PE (0.3 ml L?1) caused a significant increase in hematocrit values (P < 0.05). Among the hematological indices, mean corpuscular volume (MCV) values decreased at 0.5 ml L?1 when compared with the control and other groups (P < 0.05). Serum cortisol level was elevated at 0.3, 0.5 and 0.7 concentrations of 2‐PE (P < 0.01). The glucose level followed a trend similar to that observed for cortisol. The outcome of these experiments shows that 2‐PE at a concentration of 0.9 ml L?1 is a suitable anaesthetic for Persian sturgeon. This study demonstrates that rapid induction of deep anaethesia with a relatively high concentration of 2‐PE (0.9 and 1.1 ml L?1) was associated with the lowest effects on signs of physiological stress in Persian sturgeon.  相似文献

The effects of confinement and exercise on the stress response of the spiny damselfish Acanthochromis polyacanthus were investigated in a laboratory stock of fish. Cultured spiny damselfish had basal plasma cortisol values (<16 ng ml−1) similar to those found in wild fish, and basal plasma glucose and lactate levels that were similar to those found in other teleosts. Plasma cortisol concentrations increased in response to stress with a latency period of 5–10 min. Removal of the stressor resulted in partial recovery of cortisol levels by 24 h. Plasma glucose levels increased in response to stress in all experiments with significant increases occurring within 15 min of the imposition of stress. Elevations in plasma glucose concentrations were not initially reflected in changes in liver or muscle glycogen content, with significant reductions in liver glycogen concentrations only occurring in response to extended periods of stress.
