The geographical extent of Arctic char ( Salvelinus alpinus ) mitochondrial DNA introgression into brook char ( Salvelinus fontinalis ) populations found in eastern Québec was determined by analysing a total of 598 fish from 29 lakes. The nuclear genome was analysed by protein electrophoresis, whereas the ND-5,6 portion of the mitochondrial genome was analysed by restriction fragment length polymorphism. This survey revealed that introgressed S. fontinalis populations are restricted to only one river subdrainage of the Portneuf basin, where Arctic char is completely absent. Elsewhere, nonintrogressed pure S. fontinalis populations populate the lakes. These findings suggest that the initial hybridization event between the species is ancient and probably occurred shortly after recolonization of the area. At that time, the species would have been in contact and the chances of reproductive isolation mechanisms breaking down would have been high. We discuss the possibility that a combination of biogeographical conditions coupled with positive selection for mtDNA introgression led to the present-day distribution of introgressed S. fontinalis in northeastern North America.  相似文献

Competition for females generally results in some males adopting alternative reproductive tactics to acquire matings. For fish, the ecological and evolutionary consequences of these tactics are not well understood because of an inability to link directly the interactions of individuals on the breeding grounds with genetic data. This study combines behavioural observations with genetic estimates of male reproductive success within an intensively studied wild population of lacustrine brook trout (Salvelinus fontinalis). Male brook trout exhibit a conditional reproductive strategy with small males adopting a peripheral position to that of larger dominant males in their proximity to spawning females. Parentage analysis of eggs collected from wild redds confirmed the reproductive success of individual males. Males relegated to peripheral positions during spawning participated frequently in spawning events, but in most cases the first male to spawn was the sole contributor, and no more than two males contributed successfully to a single brood. While behavioural observations of salmonines suggests that reproduction is partitioned among males in a manner dependent upon body size and proximity to spawning females, the genetic evidence from this study suggests a more limited distribution of reproductive success in the field. The genetic contributions of male brook trout are highly skewed towards larger males for this population. A review of the salmonine literature suggests little difference in individual reproductive success for males exhibiting size-related tactics within a conditional mating strategy vs. precocial maturation. Collectively, these genetic studies provide new insights on the evolution of alternative life histories among salmonines.  相似文献

Salmonid fishes rank among species being most severely affected by introgressive hybridization as a result of a long tradition of stocking with hatchery-reared conspecifics. The objectives of this study were (i) to evaluate the genetic consequences of stocking and resulting introgression rates on the genetic integrity of natural populations of brook charr, (ii) to identify genomic regions potentially associated with adaptation to natural and artificial rearing environments, and (iii) to test the null hypothesis that introgression from domesticated brook charr into wild populations is homogeneous among loci. A total of 336 individuals were sampled from nine lakes, which were stocked at different intensities with domestic fish. Individuals were genotyped at 280 SNPs located in transcribed regions and developed by means of next-generation sequencing. As previously reported with microsatellites, we observed a positive relationship between stocking intensity and genetic diversity among stocking groups, and a decrease in population differentiation. Individual admixture proportions also increased with stocking intensity. Moreover, genomic cline analysis revealed 27 SNPs, seven of which were also identified as outliers in a genome scan, which showed an introgression rate either more restricted or enhanced relative to neutral expectations. This indicated that selection, mainly for growth-related biological processes, has favored or hampered the introgression of genomic blocks into the introgressed wild populations. Overall, this study highlights the usefulness of investigating the impact of stocking on the dynamics of introgression of potentially adaptive genetic variation to better understand the consequences of such practice on the genomic integrity of wild populations.  相似文献

Previous studies have found lower levels ofgenetic variation in lake than streampopulations of brook trout (Salvelinusfontinalis). We test the generality of thisobservation by examining whether brook troutgenetic variation at 10 allozyme loci differedwithin and among 9 pairs of lake and adjacentstream populations. With one exception, wefound that lake populations had lowerheterozygosity than their adjacent streampopulations. Although the lakes in this studyare small and some have had documented fishmortality events, no association was foundbetween lake size characteristics and thedegree of difference in heterozygosity betweenlakes and their adjacent stream populations. There were, however, negative associationsbetween metrics of fishing mortality and thedifference in heterozygosity between lakes andtheir adjacent stream populations. Thegreater the estimated fishing pressure onlake-dwelling trout, the greater the reductionin heterozygosity in those populationsrelative to their adjacent stream populations. We interpret our findings to suggest thatintensive fishing pressure can significantlyreduce genetic variation. Managers shouldtherefore prevent human-induced mortality atany indication of a large natural mortalityevent to allow populations to increase in sizeas rapidly as possible following a decline.  相似文献

Carotenoid pigmentation in Salvelinus alpinus has been connected to stress responsiveness in earlier studies. This has, however, only been tested with time-consuming image analysis from photos. Here, we used quick visual categorization of carotenoid pigmentation to investigate the stress responsiveness of the extreme groups. The visually selected charr were then exposed to a net restraint stressor. Arctic charr with few spots also had a lower stress responsiveness compared to charr with many spots. Thus, visual selection could be used as a simple method within aquaculture.  相似文献

Understanding the drivers of successful species invasions is important for conserving native biodiversity and for mitigating the economic impacts of introduced species. However, whole‐genome resolution investigations of the underlying contributions of neutral and adaptive genetic variation in successful introductions are rare. Increased propagule pressure should result in greater neutral genetic variation, while environmental differences should elicit selective pressures on introduced populations, leading to adaptive differentiation. We investigated neutral and adaptive variation among nine introduced brook trout (Salvelinus fontinalis) populations using whole‐genome pooled sequencing. The populations inhabit isolated alpine lakes in western Canada and descend from a common source, with an average of ~19 (range of 7–41) generations since introduction. We found some evidence of bottlenecks without recovery, no strong evidence of purifying selection, and little support that varying propagule pressure or differences in local environments shaped observed neutral genetic variation differences. Putative adaptive loci analysis revealed nonconvergent patterns of adaptive differentiation among lakes with minimal putatively adaptive loci (0.001%–0.15%) that did not correspond with tested environmental variables. Our results suggest that (i) introduction success is not always strongly influenced by genetic load; (ii) observed differentiation among introduced populations can be idiosyncratic, population‐specific, or stochastic; and (iii) conservatively, in some introduced species, colonization barriers may be overcome by support through one aspect of propagule pressure or benign environmental conditions.  相似文献

The karyotypes and cytogenetic characteristics of flatfishes species Paralichthys orbignyanus , Paralichthys patagonicus , Citarichthys spilopterus and Etropus crossotus (Paralichthyidae), Bothus ocellatus (Bothidae) and Symphurus tessellatus (Cynoglossidae) were investigated by conventional [Giemsa staining, C-banding, Ag- and chromomycin (CMA3)-stainings] and molecular [ in situ hybridization (ISH)] cytogenetic techniques. The results showed 2n = 46 and FN = 48 (2msm + 46sta) in P. orbignyanus , 2n = 46 and FN = 46 (46sta) in P. patagonicus , 2n = 26 and FN = 44 (18msm + 8sta) in C. spilopterus , 2n = 38 and FN = 64 (26msm + 12sta) in E. crossotus , 2n = 32 and FN = 50 (18msm + 14sta) in B. ocellatus , and 2n = 46 and FN = 62 (46msm + 62sta) in S. tessellatus . All species exhibited weak C-band positive segments in terminal and centromeric positions of some chromosome pairs. Silver staining of the nucleolus organizer regions (Ag-NOR) technique showed a single Ag-NOR-bearing chromosome pair in all species except E. crossotus . All these sites were CMA3 positive and showed clear ISH signals after probing with a 18S rRNA probe. Etropus crossotus presented until seven chromosomes with Ag-NORs and CMA3 positively stained segments in five chromosome pairs. Conversely only one chromosome pair was identified with the ISH experiments in this species. The available results show that the fishes of the order Pleuronectiformes experienced a marked chromosome evolution that included reduction in diploid number, mainly due to Robertsonian rearrangements, and several chromosome inversions.  相似文献

Habitat utilization and the life history of browntrout Salmo trutta and Arctic charr Salvelinus alpinus were investigated in fivesympatric populations and five allopatric brown troutpopulations in Høylandet catchment, a atmosphaericlow deposition area in Mid Norway. There was asignificant inverse correlation in abundance ofepibenthic Arctic charr and brown trout in theselakes, indicating that the latter species is dominant.The largest numbers of sympatric brown trout andArctic charr were caught in epibenthic habitat. In twolakes, brown trout to some extent also occurredpelagically, while pelagic individuals of Arctic charrwere found in all five lakes. The main food items forboth epibenthic and pelagic brown trout wereterrestrial surface insects and chironomid pupae.Zooplankton was the primary food item for Arctic charrin both habitats. Although the age distribution wasvery different in the populations, neither speciesseem to suffer from recruitment failure. There was nosignificant difference in survival rates betweensympatric populations of brown trout and Arctic charr.We found a significant inverse correlation betweenepibenthic catches of brown trout and the mean weightof 4+ fish, the most abundant age group. However, ifusing weight data for three-year-old fish, no suchrelationship was found for Arctic charr. Brown troutand Arctic charr reached asymptotic lengths of197–364 mm and 259–321 mm, respectively. Both speciestypically reached sexual maturity at age 2–3, and nomaturation-induced mortality was evident. We concludethat fish populations in Høylandet lakes areregulated throughout their lifes by inter- andintraspecific competition.  相似文献

The stonefly component of a naturally reproducing population of brook trout ( Salvelinus fontinalis) was studied by analyzing 216 stomach pump samples collected between May 15 and August 10, 1991 and 1992, from Libby Creek, an alpine stream in the Medicine Bow National Forest in Wyoming. Stoneflies constituted 10.3% of the total items recovered and were the second most abundant order in the samples. Nineteen species of Plecoptera representing five families were identified. Approximately 43% of all stoneflies were Sweltsa lamba; the next most prevalent species was Zapada haysi (12%). The other prevalent insect orders in the samples were Diptera (57.2%), Ephemeroptera (8.4%) and Trichoptera (6.1%). Terrestrial Coleoptera, Hymenoptera, and Hemiptera were most common in July and August. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

The North Pacific Ocean has been of great significance to understanding biogeography and speciation in temperate faunas, including for two species of char (Salmonidae: Salvelinus) whose evolutionary relationship has been controversial. We examined the morphology and genetics (microsatellite and mitochondrial DNA) of Arctic char (Salvelinus alpinus) and Dolly Varden char (Salvelinus malma) in lake systems in western Alaska, the eastern and western Arctic, and south of the Alaskan Peninsula. Morphologically, each lake system contained two forms: one (Arctic char) largely confined to lake habitats and characterized by greater numbers of pyloric caeca, gill rakers, and shallower bodies, and another (Dolly Varden) predominated in adjacent stream habitats and was characterized by fewer pyloric caeca, gill rakers, and deeper bodies. MtDNA partial (550 bp) d-loop sequences of both taxa were interspersed with each other within a single 'Bering' clade and demographic inferences suggested historical gene flow from Dolly Varden to Arctic char had occurred. By contrast, the taxa were strongly differentiated in sympatry across nine microsatellite loci in both lakes. Our data show that the two taxa are highly genetically distinct in sympatry, supporting their status as valid biological species, despite occasional hybridization. The interaction between these species highlights the importance of the North Pacific, and Beringia in particular, as an evolutionary wellspring of biodiversity.  相似文献

Until now the supplementation of cryopreservation extenders with antioxidants has not been examined in teleost fish. Therefore, the present study investigated whether addition of antioxidants (catalase, superoxide dismutase, peroxidase, reduced glutathione, reduced methione, mixtures of reduced and oxidized glutathione or methionine) to the cryopreservation extenders could increase the quality of frozen-thawed semen of brook trout, Salvelinus fontinalis, and rainbow trout, Oncorhynchus mykiss. In brook trout and rainbow trout semen post-thaw fertility and motility were evaluated and in brook trout additionally the membrane integrity, DNA integrity, and sperm lipid peroxidation were evaluated. The tested antioxidants affected the motility parameters, DNA integrity, and fertility of cryopreserved semen, but not the membrane integrity. Most of the observed effects were negative and only minor positive effects were found. In brook trout 1.5 mmol/l reduced methionine and a mixture of 1.5 mmol/l oxidized and reduced glutathione increased the swimming velocity of frozen-thawed semen. One hundred U/l catalase, 1.5 mmol/l reduced glutathione, and 1.5 mmol/l reduced methionine slightly, but not statistically significantly increased the semen post-thaw fertility. However, these effects were not detectable in rainbow trout. Antioxidative stress or damage seems to play no role during cryopreservation, as also in the lipid peroxidation test no differences were obtained between fresh and cryopreserved semen. Therefore, for routine cryopreservation extender supplementation with antioxidants is not recommended in brook trout and rainbow trout.  相似文献

Arctic charr (Salvelinus alpinus L.) from Lake Thingvallavatn, Iceland occur as four distinct morphs: large benthivorous (LB), dwarf benthivorous (DB), piscivorous (PI) and planktonivorous (PL). The morphs differ with respect to body size, head morphology, growth rate, and life history. The aim of this study was to investigate the paired box protein 7 (Pax7) gene as a candidate for such polymorphisms due to its importance in cranio-facial, skeletal muscle, and central nervous system development. No variation in coding and intronic sequences was found between morphs. We identified 10 alternate Pax7 isoforms with insertions/deletions: a four-residue (GNRT) deletion, a GEASS insertion truncated by the first serine residue (GEAS), and a thirteen-residue insertion (GQYA/TGPEYVYCGT). The latter insertion with a threonine (T) contains a putative casein kinase II (CK-2) phosphorylation site. Pax7 spatial expression patterns were identical in embryos of DB-, LB-, and PL-morphs, and were similar to those described for zebrafish Pax7c, but a difference in temporal expression for segmentation was observed between DB and LB morphs. At the end of segmentation, novel expression was observed in the mandibular region as two bilateral domains. The potential role of multiple alternative splicing of the Pax7 gene for the generation of different Arctic charr morphs is briefly discussed.  相似文献

Despite geographical isolation and widespread phenotypic polymorphism, previous population genetic studies of Arctic charr, Salvelinus alpinus , have detected low levels of intra- and interpopulation variation. In this study, two approaches were used to test the generality of low genetic diversity among 15 Arctic charr populations from three major drainages of the central Alpine region of Europe. First, a representative subsample of each drainage was screened by PCR–RFLP analysis of mtDNA using 31 restriction enzymes. All individuals but one shared an identical haplotype. In contrast, microsatellite DNA variation revealed high levels of genetic diversity within and among populations. The number of alleles per locus ranged from six to 49, resulting in an overall expected heterozygosity from 0.72 ± 0.09 to 0.87 ± 0.04 depending on the locus. Despite evidence for fish transfers among Alpine charr populations over centuries, genetic diversity was substantially structured, as revealed by hierarchical Φ statistics. Eighteen per cent of total genetic variance was apportioned to substructuring among Rhône, Rhine, and Danube river systems, whereas 19% was due to partitioning among populations within each drainage. Cluster analyses corroborated these results by drainage-specific grouping of nonstocked populations, but also revealed damaging effects of stocking practices in others. However, these results suggest that long-term stocking practices did not generally alter natural genetic partitioning, and stress the importance of considering genetic diversity of Arctic charr in the Alpine region for sound management. The results also refute the general view of Arctic charr being a genetically depauperate species and show the potential usefulness of microsatellite DNAs in addressing evolutionary and conservation issues in this species.  相似文献

Karyological and genomic in situ hybridization (GISH) approaches provided evidence of the parentage and origin of the hybrid species Narcissus obsoletus. Here, we demonstrate that the putative parental species, N. serotinus L. and N. elegans (Haw.) Spach, recently proposed because of their intermediate morphological traits, have participated in the hybridization process forming this taxon. Karyotype characterization of parental genomes in populations from S Spain and N Morocco has revealed differences in chromosome length and karyotype asymmetry, highlighting their diploid nature. Multicolour GISH on metaphase plates of N. obsoletus, with N. serotinus and N. elegans DNA used as probes, showed differential fluorescent staining of 10 and 20 chromosomes from parental genomes, respectively. Both parental genomes were detected in the allopolyploid, albeit in a duplicated manner. Secondary hybridization between N. obsoletus and N. serotinus was also detected karyologically. 1. Field observations indicate that the ability to feed at different light intensities may differ between brown trout and Arctic charr, and this is the first study to test this experimentally. To establish a background level of feeding in daylight at midday, trout and charr in two size groups were kept in tanks (one fish per tank) at three constant temperatures (5.0, 10.8 and 13.0 °C) and each fish was offered, one at a time, 50 freshly killed shrimps (Gammarus pulex), the number eaten being recorded. Shrimps could only be taken in the water column because a metal mesh prevented access to dead shrimps on the tank bottom. In a first series of experiments, individual fish were kept at one of 10 natural light intensities (range 0.001–50 lx). In a second series, conditions were similar except that the fish tank was covered in black polyethylene and had a light‐tight lid with a shutter so that light levels could be kept constant, using artificial illumination. In a third series, the fish were fed in total darkness, but the false bottom was removed, allowing access to dead shrimps on the tank bottom as well as in the water column. 2. The results of the first and second series differed interspecifically but were very similar intraspecifically, with no significant differences between the food intake for the two size groups or in the experiments at 10.8 and 13.0 °C. Food intake remained fairly constant at light intensities between 50 lx (dusk or dawn) and 0.03 lx and was similar to that of fish feeding at midday. At 10.8 and 13.0 °C, food intake between 0.03 and 50 lx was higher for trout than for charr, mean values for shrimps eaten per fish being 39.9 for trout (range 36–44, n = 100 fish) and 32.0 for charr (range 28–38, n = 100), but at 5.0 °C, the situation was reversed with mean values of 15.1 for trout (range 11–18, n = 50 fish) and 19.8 for charr (range 17–22, n = 50). 3. As light intensity decreased from 0.04 to 0.001 lx, feeding rate decreased exponentially but was always higher for charr than for trout, with a mean number of shrimps eaten at 0.001 lx of 9.3 for trout (range 5–13, n = 20 fish) and 13.6 for charr (range 9–20, n = 20) at 10.8 and 13.0 °C, and 2.0 for trout (range 1–4, n = 10 fish) and 5.5 for charr (range 2–8, n = 10) at 5.0 °C. In total darkness (false bottom fitted), none of the 50 shrimps was taken by either species. When the false bottom was removed in the third series, the mean number of shrimps consumed over 24 h was eight for trout (range 3–11, n = 20 fish) and 14.9 for charr (range 9–20, n = 20) at 10.8 and 13.0 °C, and two for trout (range 0–4, n = 10 fish) and five for charr (range 3–8, n = 10) at 5.0 °C. 4. Therefore, the feeding ability of trout was superior to that of charr when using photopic vision in daylight and mesotopic vision at dusk and dawn, but inferior to that of charr when using scotopic vision at low light intensity. Charr were also superior at low temperatures and when foraging for food in total darkness. Therefore, as light intensity decreases after dusk in their natural habitat, the advantage in feeding will shift from trout to charr, with the reverse occurring as light intensity increases after dawn.  相似文献

Nine karyotypes are described within a single species of common New Zealand tree weta. Their diploid numbers range from 11 to 25. The distribution of the karyotypes suggests that each had a single origin except the 17-karyotype which was the most common karyotype and had a disjunct distribution. The overall level of allozyme diversity observed is similar to that seen within many widespread taxa. The distribution of allozyme alleles did not coincide with the distribution of karyotypes within this species and the Neighbour-Joining tree was not concordant with the chromosome based sub-divisions of the species. Thus, no evidence was found to suggest that chromosomal differentiation has been acting as a barrier to the flow of alleles within H. thoracica. The lack of concordance of genetic markers is thought to result from rapid chromosome radiation and reticulate evolution. Northland peninsula of North Island, New Zealand is a region of high chromosomal and allozymic diversity in H. thoracica. This may have resulted from geographic isolation during the Pliocene when Northland formed an archipelago of many small low-lying islands.  相似文献
