Most monocotyledons like cereals accumulate antifreeze proteins in the apoplast during cold acclimation, but it is still uncertain whether dicotyledons do. Here we report the isolation and characterisation of a 33-kD apoplastic chitinase extracted from the corolla of wintersweet (Chinmonanthus praecox communis L.), which was purified using successive column chromatography on Phenyl-Sepharose, DEAE-Sepharose, and CM-Sepharose. Antifreezing activity of chitinase was confirmed in terms of the formation of bipyramidal ice crystals and high thermal-hysteresis values. Interestingly, chitinase was also found to affect germination of fungal spores of four major plant pathogens. From these data, we hypothesize that, under natural conditions, wintersweet as one of the overwintering dicotyledons also accumulates apoplastic antifreeze proteins like monocotyledons. To our knowledge, this is the first report on the isolation of dicotyledon apoplastic chitinase with high-level antifreeze and antifungal activities.  相似文献

The chemistry of the lemon-scented oil gland secretion ofCollohmannia gigantea, a middle-derivative mixonomatanoribatid mite, was investigated by gas chromatography – massspectrometry.Gas chromatographic profiles of whole body extracts of C.gigantea revealed two distinct chromatographic zones, the firstcontaining a set of six volatile compounds, comprising the lemon-scentedmonoterpene aldehydes neral and geranial, the scented monoterpene ester nerylformate, a distinctly scented aromatic aldehyde(2-hydroxy-6-methyl-benzaldehyde= 2,6-HMBD), and the two non-scented hydrocarbons, tridecane and pentadecane.All six components appeared to be present in steady relative proportions inscenting mites only, indicating their unity within the scented secretion. Incontrast, the components of the second chromatographic zone were less volatileand found in both, scenting and non-scenting mites. Chemically, they representaset of fatty acids of already known cuticular origin.The secretion bouquet ofthe first chromatographic zone was linked with oil glands by histochemicalmeans: large amounts of aldehydes were present only in oil gland reservoirs,notin any other region of the mite body. While chemical profiles of oil glandsecretions of several dozen astigmatid mites are known, only one other oribatidoil gland composition, from a desmonomatan species, has been elucidated, beingalmost the same as that of C. gigantea. Moreover, allcomponents of these two secretions are widely distributed amongst astigmatidmite species and may also be common in a restricted set of middle-derivativeoribatids. These findings are consistent with the idea of astigmatid miteoriginfrom a mixonomatan-desmonomatan group.  相似文献

Rose‐scented geranium (Pelargonium spp.) is one of the most important aromatic plants and is well known for its diverse perfumery uses. Its economic importance is due to presence of fragrance rich essential oil in its foliage. The essential oil is a mixture of various volatile phytochemicals which are mainly terpenes (isoprenoids) in nature. In this study, on the geranium foliage genes related to isoprenoid biosynthesis (DXS, DXR and HMGR) were isolated, cloned and confirmed by sequencing. Further, the first gene of 2‐C‐methyl‐d ‐erythritol‐4‐phosphate (MEP) pathway, 1‐deoxy‐d ‐xylulose‐5‐phosphate synthase (GrDXS), was made full length by using rapid amplification of cDNA ends strategy. GrDXS contained a 2157 bp open reading frame that encoded a polypeptide of 792 amino acids having calculated molecular weight 77.5 kDa. This study is first report on heterologous expression and kinetic characterization of any gene from this economically important plant. Expression analysis of these genes was performed in different tissues as well as at different developmental stages of leaves. In response to external elicitors, such as methyl jasmonate, salicylic acid, light and wounding, all the three genes showed differential expression profiles. Further GrDXS was over expressed in the homologous (rose‐scented geranium) as well as in heterologous (Withania somnifera) plant systems through genetic transformation approach. The over‐expression of GrDXS led to enhanced secondary metabolites production (i.e. essential oil in rose‐scented geranium and withanolides in W. somnifera). To the best of our knowledge, this is the first report showing the expression profile of the three genes related to isoprenoid biosynthesis pathways operated in rose‐scented geranium as well as functional characterization study of any gene from rose‐scented geranium through a genetic transformation system.  相似文献

The shrub Chimonanthus praecox L. (wintersweet) which is native to Chinese montane forests produces its flowers in the midst of winter. This indicates that the floral organs of this species are adapted to growth and development under freezing temperatures. Here, we report the isolation and preliminary characterisation of a 33 kDa apoplastic antifreeze chitinase (CpCHT1) from the petals and its corresponding cDNA. The chitinase activity of CpCHT1 was confirmed by activity staining. Antifreeze activity was validated in terms of the formation of bipyramidal ice crystals and high thermal-hysteresis values. CpCHT1 was also found to affect the germination of fungal spores of four major plant pathogens. In addition, the gene and protein are expressed constitutively not only in flowers, but also in leaves, bark and root tissues. From these data we hypothesize that this protein is multifunctional and may protect wintersweet from freezing injury and provide nonspecific disease resistance.  相似文献

Most flower visitors innately prefer a particular color and scent, and use them as cues for flower recognition and selection. However, in most cases, since color and scent serve as a combined signal, not only does the preference for an individual cue, but also the preference hierarchy among different cues, influence their flower visitation. In the present study, we attempted to reveal (1) the chromatic and (2) the olfactory cues that stimulate flower visiting, and (3) the preference hierarchy between these cues, using the naïve adult butterfly Vanessa indica. When we offered 12 different-colored (six chromatic and six achromatic) paper flower models, V. indica showed a color preference for yellow and blue. When we examined the proboscis extension reflex (PER) of V. indica towards 16 individual compounds identified in the floral scents from two nectar plants belonging to the family Compositae, Taraxacum officinale and Cirsium japonicum, six compounds were found to have relatively high PER-eliciting activities, including benzaldehyde, acetophenone, and (E+Z)-nerolidol. When we combined color and scent cues in two-choice bioassays, where butterflies were offered flower models that were purple (a relatively unattractive color), the models scented with these active compounds were significantly more attractive than the odorless controls. In addition, synthetic blends mimicking the floral scents of T. officinale and C. japonicum (at doses equivalent to that of ten flowers) enhanced the number of visits to the scented models. However, the effect of odorizing was not conspicuous in parallel bioassays when yellow flower models were used, and the butterflies also significantly preferred odorless yellow models to scented purple models. These results demonstrate that V. indica depends primarily on color and secondarily on scent during flower visitation.  相似文献

Wintersweet (Chimonanthus praecox L.) is a traditional winter-flowering plant in China and a popular cut flower in winter. Its unique flowering characteristics under cold stress may involve the regulation of a large number of proteins. Protein post-translational modification is an important regulating type for the gene function. However, little is known about the post-translational modification in wintersweet. SUMOylation is an important post-translational modification in eukaryotes. Small ubiquitin-like modifier (SUMO) E3 ligases perform multiple functional regulatory activities in plants via SUMOylation. Here, we cloned and identified a SIZ/PIAS-type SUMO E3 ligase, CpSIZ1, from wintersweet. CpSIZ1 shared high similarity with other SIZ1 proteins. Quantitative real-time PCR (qRT-PCR) indicated that CpSIZ1 was expressed in all tissues tested, with the highest expression in flower wither period of stage 6, and followed by mature leaves except for different flower development stages. The ectopic expression of CpSIZ1 in Arabidopsis, including the CpSIZ1 overexpression in siz1-2 mutant (HB line) and CpSIZ1 overexpression in WT (OE line), not only promoted vegetative growth, delayed flowering and accelerated leaf senescence, but also improve the cold tolerance in Arabidopsis. Therefore, our studies have enrich the understanding of function of SIZ1 gene in woody plant, and provide a good foundation for further research on the post-translational modification regulation mechanism in this winter-flowering plant.

The ability of scented geraniums (Pelargonium sp. Frensham) to tolerate metal stress was assessed using chlorophyll a fluorescence kinetics. The effects of various concentrations of cadmium and nickel in the culture solution on photosynthetic efficiency in scented geranium was evaluated in comparison to two well-established metal accumulators, the Indian mustard (Brassica juncea), and the sunflower (Helianthus annuus), under greenhouse conditions. The efficiency of the photosynthetic apparatus was affected to varying degrees at all metal concentrations for the plants tested. High concentrations of cadmium (1000 mg L-1) did not significantly affect the efficiency of photosystem II activity, expressed as the ratio of variable fluorescence to maximal fluorescence (Fv/Fm), which remained high (0.738) in scented geraniums, but decreased significantly (P < 0.05) in Indian mustard (0.089) and sunflower (0.026) plants following 4 days of metal exposure. Similar trends were observed for nickel treatments. Also, the number and size of active photosynthetic reaction centers, as measured by the Fv/Fo ratio, was not significantly affected by metal exposure in scented geranium plants, while the ratio significantly decreased in Indian mustard and sunflower seedlings. The results suggest that scented geranium plants were able to overcome metal stress through (1) maintaining an efficient photosystem II activity, which is required for plant metabolism and physiological functions, as well as to overcome metal ion mediated stress, and (2) restricting damage to the photosynthetic apparatus (reaction centers) by metal ions.  相似文献

Tetragonisca angustula stingless bees are considered as solitary foragers that lack specific communication strategies. In their orientation towards a food source, these social bees use chemical cues left by co-specifics and the information obtained in previous foraging trips by the association of visual stimuli with the food reward. Here, we investigated their ability to learn the association between odors and reward (sugar solution) and the effect on learning of previous encounters with scented food either inside the hive or during foraging. During food choice experiments, when the odor associated with the food was encountered at the feeding site, the bees’ choice is biased to the same odor afterwards. The same was not the case when scented food was placed inside the nest. We also performed a differential olfactory conditioning of proboscis extension response with this species for the first time. Inexperienced bees did not show significant discrimination levels. However, when they had had already interacted with scented food inside the hive, they were able to learn the association with a specific odor. Possible olfactory information circulation inside the hive and its use in their foraging strategies is discussed.  相似文献

The ability of scented geraniums (Pelargonium sp. ‘Frensham’) plants to tolerate, uptake, and accumulate lead was assessed compared with two well-established metal accumulators, Indian mustard (Brassica juncea) and sunflower (Helianthus annuus), under greenhouse conditions. The efficiency of the photosynthetic apparatus and the number and size of active photosynthetic reaction centers (expressed as chlorophyll a fluorescence ratios of variable fluorescence to maximal fluorescence [Fv/Fm] and variable fluorescence to unquenchable portion of fluorescence [Fv/Fo], respectively) were affected to varying degrees at all metal concentrations in all the plants tested. Lead exposure did not significantly affect the efficiency of photosystem II activity or the number and size of the photosynthetic reaction centers in scented geraniums, but the ratios decreased significantly in Indian mustard and sunflower plants following lead exposure. In addition to tolerating high levels of lead, the scented geraniums accumulated in excess of 3000 mg of lead per kg DW of shoot and above 60,000 mg of lead per kg DW of root tissue. Additionally, scented geraniums exposed to a mixture of metals (lead + cadmium + nickel) had the ability to uptake in excess of 4.72% Pb + 0.44% Cd + 0.52% Ni per kg of root DW, as well as 0.17% Pb + 0.07% Cd + 0.14% Ni per kg of shoot DW within 14 d, indicated the potential for existence of more than one functional tolerance and metal uptake mechanism(s). The cellular localization of lead was assessed using transmission electron microscopy coupled with an X-ray microanalyzer. Lead accumulation was observed in the apoplasm and in the cytoplasm, vacuoles, and as distinct globules (potentially as lead-lignin or lead-phosphate complexes) on the cell membrane and cell wall. We conclude that the ability of scented geraniums to tolerate high lead accumulation in its biomass is due, in part, to limiting damage to photosynthetic apparatus and metal detoxification by formation of metal complexes.  相似文献

The hawkmoth, Manduca sexta, uses both colour and odour to find flowers when foraging for nectar. In the present study we investigated how vision and olfaction interact during learning. Manduca sexta were equally attracted to a scented blue coloured feeding target (multimodal stimulus) as to one that does not carry any scent (unimodal stimulus; visual) or to an invisible scented target (unimodal stimulus; odour). This naive attraction to multimodal as well as to unimodal stimuli could be manipulated through training. Moths trained to feed from a blue, scented multimodal feeding target will, when tested in a set-up containing all three feeding targets, select the multimodal target as well as the scented, unimodal target, but ignore the visual target. Interestingly, moths trained to feed from a blue, unimodal visual feeding target will select the visual target as well as the scented, multimodal target, but ignore the unimodal odour target. Our results indicate that a multimodal target is perceived as two separate modalities, colour and odour, rather than as a unique fused target. These findings differ from earlier studies of desert ants that perceive combined visual and odour signals as a unique fused stimulus following learning trials.  相似文献

Drones (Apis mellifera ) were attracted to a lure scented with queen pheromone suspended at a height of 11–18m. An unscented test object, usually a black sphere, was lowered on a vertical string at a horizontal distance of 50 cm or 103 cm from the scented lure. Drones often made a rapid, direct, displacement from the scented lure to the test object. Drones responded to objects that subtended an angle as small as 0.41° which is less than the acceptance angle of a single ommatidium. We calculate that a drone can respond to a stimulus corresponding to a reduction in the light incident on one ommatidium at a time of 8%; the brevity of the stimulus on each ommatidium would seem to be a factor in the design of the eye.  相似文献

When a predators attack prey, damaged prey tissue releases chemical information that reliably indicates an actively foraging predator. Prey use these semiochemicals to cue anti-predator behaviour and reduce their probability of predation. Here, we test central mudminnows, Umbra limi (Kirtland 1840), for anti-predator behavioural responses to chemical cues in conspecific skin extract. In a field experiment, traps scented with mudminnow skin extract (alarm cue) caught fewer mudminnows than traps scented with water (control). Under controlled laboratory conditions, mudminnows showed a significant reduction in activity and movement to the bottom in response to alarm cues relative to water controls. Reduced activity and increased time on the bottom of the tank are both known components of an anti-predator response. Thus, based on field and lab data, mudminnows exhibited anti-predator behavioural responses to chemical alarm cues released by damaged epidermal tissue. Histological preparations of epidermal tissue did not reveal the presence of specialised “alarm substance” cells for the production of chemical alarm cues. This is the first report of an alarm reaction in an esociform, an order with a long evolutionary history of piscivory.  相似文献

In order to elucidate the relationship between androgens and the function of the muskrat (Ondatra zibethicus) scented glands during the breeding season, we investigated immunolocalization of steroidogenic enzymes P450scc, 3βHSD and P450c17 in the muskrat testes and scented glands. Nine adult muskrats were obtained in March (n=3), May (n=3) and July (n=3) 2010. Steroidogenic enzymes were immunolocalized using polyclonal antisera raised against bovine adrenal P450scc, human placental 3βHSD and porcine testicular P450c17. Histologically, all types of spermatogenic cells including mature-phase spermatozoa in seminiferous tubules were observed in all testes. Glandular cells, interstitial cells, epithelial cells and excretory tubules were identified in scented glands during the breeding season. P450scc, 3βHSD and P450c17 were only identified in Leydig cells during the breeding season; P450scc and P450c17 were observed in glandular cells of scented glands, however, 3βHSD was not found in scented glands during the breeding season. These novel findings provide the first evidence showing that scented glands of the muskrats are capable of locally synthesizing androgens and androgens acting via an endocrine, autocrine or paracrine manner may play an important role in scented gland function during the breeding season.  相似文献

In the ornamental cut flower plant Gerbera hybrida the spatial distribution of regulatory molecules characteristic of differentiation of the composite inflorescence is visualized as the various patterns of anthocyanin pigmentation of different varieties. In order to identify genes that the plant can regulate according to these anatomical patterns, we have analysed gene expression affecting two enzymatic steps, chalcone synthase (CHS) and dihydroflavonol-4-reductase (DFR), in five gerbera varieties with spatially restricted anthocyanin pigmentation patterns. The dfr expression profiles vary at the levels of floral organ, flower type and region within corolla during inflorescence development according to the anthocyanin pigmentation of the cultivars. In contrast, chs expression, although regulated in a tissue-specific manner during inflorescence development, varies only occasionally. The variation in the dfr expression profiles between the varieties reveals spatially specific gene regulation that senses the differentiation events characteristic of the composite inflorescence.  相似文献

The localization and timing of production and emission of scent was studied in different Rosa × hybrida cultivars, focusing on three particular topics. First, it was found that petals represent the major source of scent in R. × hybrida. In heavily scented cultivars, the spectrum and levels of volatiles emitted by the flower broadly correlated with the spectrum and levels of volatiles contained within the petal, throughout petal development. Secondly, analysis of rose cultivars that lacked a detectable scent indicated that the absence of fragrance was due to a reduction in both the biosynthesis and emission of scent volatiles. A cytological study, conducted on scented and non-scented rose cultivars showed that no major difference was visible in the anatomy of the petals either at small magnification in optical sections or in ultrathin sections observed by TEM. In particular, the cuticle of epidermal cells was not thicker in scentless cultivars. Thirdly, using two different techniques, solid/liquid phase extraction and headspace collection of volatiles, we showed that in roses, both epidermal layers are capable of producing and emitting scent volatiles, despite the different morphologies of the cells of these two tissues. Moreover, OOMT, an enzyme involved in scent molecule biosynthesis was localized in both epidermal layers.  相似文献

Here we have explored the effect of neoagarotetraose (NAT) on liver injury caused by intense exercise. Our results showed that NAT treatment obviously decreased liver weight (p < 0.01), improved the liver morphological structure, decreased ALT level (p < 0.05) and endotoxin (LPS) (p < 0.01). In addition, NAT could regulate bile acid profiles in feces and serum of mice, which indicated the potential of liver function, suggesting that NAT was effective to relieve intense exercise-induced liver injury. NAT could regulate the expression of colon genes. NAT tended to alter the microbial composition of mice under intense exercise. We uncovered the network interactions between liver traits and microbial communities in NAT treatment mice. Interestingly, our data indicated that intense exercise-induced liver injury may be related to Clostridiales. In summary, these results demonstrated that NAT relieved liver injury induced by intense exercise may be related to gut microbiota.  相似文献
