The adaptation of barley (Hordeum vulgare L. cv. Akcent) plants to low (LI, 50 µmol m–2 s–1) and high (HI, 1000 µmol m–2 s–1) growth irradiances was studied using the simultaneous measurements of the photosynthetic oxygen evolution and chlorophyll a (Chl a) fluorescence at room temperature. If measured under ambient CO2 concentration, neither increase of the oxygen evolution rate (P) nor enhancement of non-radiative dissipation of the absorbed excitation energy within photosystem 2 (PS2) (determined as non-photochemical quenching of Chl a fluorescence, NPQ) were observed for HI plants compared with LI plants. Nevertheless, the HI plants exhibited a significantly higher proportion of QA in oxidised state (estimated from photochemical quenching of Chl a fluorescence, qP), by 49–102 % at irradiances above 200 µmol m–2 s–1 and an about 1.5 fold increase of irradiance-saturated PS2 electron transport rate (ETR) as compared to LI plants. At high CO2 concentration the degree of P stimulation was approximately three times higher for HI than for LI plants, and the irradiance-saturated P values at irradiances of 2 440 and 2 900 µmol m–2 s–1 were by 130 and 150 % higher for HI plants than for LI plants. We suggest that non-assimilatory electron transport dominates in the adaptation of the photosynthetic apparatus of barley grown at high irradiances under ambient CO2 rather than an increased NPQ or an enhancement of irradiance-saturated photosynthesis.  相似文献

The aim of this study was to investigate the physiological basis of increased root exudation from Festuca rubra, in response to defoliation. The hypothesis, that assimilate supply to roots is a key determinant of the response of root exudation to defoliation was tested by imposing CO2-deplete (< 50 mol mol–1) atmospheres to F. rubra. This was done as a non-destructive means of preventing supply of new assimilate to roots of intact and defoliated plants. F. rubra was grown in axenic sand systems, with defoliation and CO2-depletion treatments applied to plants at 14 and 35 days after planting. Root exudation and NO3 uptake were quantified throughout, and post-treatment uptake and allocation of N were determined from the distribution of 15N label, supplied as 15NO3 . Defoliation of F. rubra resulted in significantly (P <0.01) increased root exudation, CO2-depletion did not result in increased exudation from plants of either age. When treatments were applied to F. rubra after 14 days, defoliation and CO2-depletion each reduced NO3 uptake significantly (P <0.05). However, in older plants, uptake of NO3 was less sensitive to defoliation and CO2-depletion. The results indicate that increased root exudation following defoliation is not related directly to reduced assimilate supply to roots. This was evident from the lack of effect of CO2-depletion on root exudation, and the absence of correlation between root-C efflux and the rate of NO3 uptake. The physiological basis of increased exudation following defoliation remains uncertain, but may be dependent on physical damage, either directly or as a consequence of systemic responses to wounding.  相似文献

In C4 grasses belonging to the NADP-malic enzyme-type subgroup, malate is considered to be the predominant C4 acid metabolized during C4 photosynthesis, and the bundle sheath cell chloroplasts contain very little photosystem-II (PSII) activity. The present studies showed that Flaveria bidentis (L.), an NADP-malic enzyme-type C4 dicotyledon, had substantial PSII activity in bundle sheath cells and that malate and aspartate apparently contributed about equally to the transfer of CO2 to bundle sheath cells. Preparations of bundle sheath cells and chloroplasts isolated from these cells evolved O2 at rates between 1.5 and 2 mol · min–1 · mg–1 chlorophyll (Chl) in the light in response to adding either 3-phosphoglycerate plus HCO 3 or aspartate plus 2-oxoglutarate. Rates of more than 2 mol O2 · min–1 · mg–1 Chl were recorded for cells provided with both sets of these substrates. With bundle sheath cell preparations the maximum rates of light-dependent CO2 fixation and malate decarboxylation to pyruvate recorded were about 1.7 mol · min–1 · mg–1 Chl. Compared with NADP-malic enzyme-type grass species, F. bidentis bundle sheath cells contained much higher activities of NADP-malate dehydrogenase and of aspartate and alanine aminotransferases. Time-course and pulse-chase studies following the kinetics of radiolabelling of the C-4 carboxyl of C4 acids from 14CO2 indicated that the photosynthetically active pool of malate was about twice the size of the aspartate pool. However, there was strong evidence for a rapid flux of carbon through both these pools. The long-term effects of exogenous sucrose (3 percnt;) on growth, photosynthesis and carbon metabolism ofin vitro tomato plantlets were investigated under two sets of growth conditions that respectively favor source- or sink-limitations of photosynthesis: 1) low photosynthetic photon flux (PPF) (50 μmol m−2 · s−1) and low CO2 concentration (400 μmol mol−1) and 2) high PPF (500 μmol m−2 · s−1 and high CO2 concentration (4000 μmol mol−1). The supply of sucrose under source-limitation conditions increased the growth, the maximal photosynthetic rate, the chl content, the maximal quantum yield of Photosystem II estimated by the Fv/Fm chl fluorescence ratio as well as the soluble sugars (hexoses, sucrose) and starch contents in roots, young and mature leaves when compared to those of photo-autotrophic plantlets. Also, sucrose feeding under these conditions strongly increased the activity of sucrose synthase (SS) (EC 2.4.1.13) in roots and young leaves whereas the activities of sucrose phosphate synthase (SPS) (EC 2.4.1.14), acid invertase (INV) (EC 3.2.1.26) and ADP-glucose pyrophosphorylase (ADPGppase) (EC 2.7.7.27) were highly stimulated in roots and mature leaves. Contrary to these observations, the supply of sucrose to plantlets developed under high PPF and CO2 concentration decreased growth and led to a somewhat lower maximal photosynthetic rate relative to photo-autotrophic plantlets. These negative responses to exogenous sucrose were accompanied by stronger accumulations of hexose and starch, larger stimulation of INV in mature leaves developed under conditions of sink limitation than those from source limitation conditions. Moreover, under high PPF and high CO2 concentration, exogenous sucrose led to a marked repression of the SPS activity and caused much lower stimulations of ADPGppase in mature leaves than those observed at low PPF and low CO2 concentration. We therefore conclude that under our experimental conditions, the interactive effects of exogenous sucrose and environmental conditions on growth and photosynthesis could be rationalized by the source-sink equilibrium of thein vitro tomato plantlets.  相似文献

Leaves developed at high irradiance (I) often have higher photosynthetic capacity than those developed at low I, while leaves developed at elevated CO2 concentration [CO2] often have reduced photosynthetic capacity compared with leaves developed at lower [CO2]. Because both high I and elevated [CO2] stimulate photosynthesis of developing leaves, their contrasting effects on photosynthetic capacity at maturity suggest that the extra photosynthate may be utilized differently depending on whether I or [CO2] stimulates photosynthesis. These experiments were designed to test whether relationships between photosynthetic income and the net accumulation of soluble protein in developing leaves, or relationships between soluble protein and photosynthetic capacity at full expansion differed depending on whether I or [CO2] was varied during leaf development. Soybean plants were grown initially with a photosynthetic photon flux density (PPFD) of 950 µmol m–2 s–1 and 350 µmol [CO2] mol–1, then exposed to [CO2] ranging from 135 to 1400 µmol mol–1 for the last 3 d of expansion of third trifoliolate leaves. These results were compared with experiments in which I was varied at a constant [CO2] of 350 µmol mol–1 over the same developmental period. Increases in area and dry mass over the 3 d were determined along with daily photosynthesis and respiration. Photosynthetic CO2 exchange characteristics and soluble protein content of leaves were determined at the end of the treatment periods. The increase in leaflet mass was about 28 % of the dry mass income from photosynthesis minus respiration, regardless of whether [CO2] or I was varied, except that very low I or [CO2] increased this percentage. Leaflet soluble protein per unit of area at full expansion had the same positive linear relationship to photosynthetic income whether [CO2] or I was varied. For variation in I, photosynthetic capacity varied directly with soluble protein per unit area. This was not the case for variation in [CO2]. Increasing [CO2] reduced photosynthetic capacity per unit of soluble protein by up to a factor of 2.5, and photosynthetic capacity exhibited an optimum with respect to growth [CO2]. Thus CO2 did not alter the relationship between photosynthetic income and the utilization of photosynthate in the net accumulation of soluble protein, but did alter the relationship between soluble protein content and photosynthetic characteristics in this species.  相似文献

Adjustment in leaf area : mass ratio called leaf area ratio (LAR) is one of the strategies to optimize photon harvesting. LAR was recorded for 10 genotypes of Hevea brasiliensis under high irradiance and low temperature and the genotypes were categorized into two groups, i.e. high LAR and low LAR types. Simultaneously, the growth during summer as well as winter periods, photosynthetic characteristics, and in-vitro oxidative damage were studied. Low LAR (19.86±0.52 m2 kg–1) types, recorded an average of 18.0 % chlorophyll (Chl) degradation under high irradiance and 7.1 % Chl degradation under low temperature. These genotypes maintained significantly higher net photosynthetic rate (P N) of 10.4 mol(CO2) m–2 s–1 during winter season. On the contrary, the high LAR (24.33±0.27 m2 kg–1) types recorded significantly lower P N of 4 mol(CO2) m–2 s–1 and greater Chl degradation of 37.7 and 13.9 % under high irradiance and low temperature stress, respectively. Thus LAR may be one of the physiological traits, which are possibly involved in plant acclimation process under both stresses studied.  相似文献

We studied the export of inorganic carbon and nitrous oxide (N2O) from a Danish freshwater wetland. The wetland is situated in an agricultural catchment area and is recharged by groundwater enriched with nitrate (NO3 ) (1000 M). NO3 in recharging groundwater was reduced (57.5 mol NO3 m–2 yr) within a narrow zone of the wetland. Congruently, the annual efflux of carbon dioxide (CO2) from the sediment was 19.1 mol C m–2 when estimated from monthly in situ measurements. In comparison the CO2 efflux was 4.8 mol C m–2 yr–1 further out in the wetland, where no NO3 reduction occurred. Annual exports of inorganic carbon in groundwater and surface water was 78.4 mol C m–2 and 6.1 mol C m–2 at the two sites, respectively. N2O efflux from the sedimenst was detectable on five out of twelve sampling dates and was significantly (P < 0.0001) higher in the NO3 reduction zone (0.35–9.40 mol m–2 h–1, range of monthly means) than in the zone without NO3 reduction (0.21–0.41 mol m–2 h–1). No loss of dissolved N2O could be measured. Total annual export of N2O was not estimated. The reduction of oxygen (O2) in groundwater was minor throughout the wetland and did not exceed 0.2 mol 02 m–2yr–1. Sulfate (SO4 ––) was reduced in groundwater (2.1 mol SO4 –– m–2 yr–1) in the zone without NO3 reduction. Although the NO3 in our wetland can be reduced along several pathways our results strongly suggest that NO3 loading of freshwater wetlands disturb the carbon balance of such areas, resulting in an accelerated loss of inorganic carbon in gaseous and dissolved forms.  相似文献

In the present paper we report partial pressureof CO2 (pCO2) data obtained off theBelgian coast during 24 cruises. The temporaland spatial resolution of this data set allowsus to discuss satisfactorily seasonal andinter-annual variability of pCO2 in thestudy area. The dynamics of pCO2 aredescribed using two approaches: fixed referencestations and area survey cruises. The air-waterfluxes of CO2 in the Scheldt estuarineplume and in the outer-plume region areestimated quantitatively, showing that theseareas correspond respectively to a net annualsource and sink of atmospheric CO2. Theannually integrated air-water fluxes for theScheldt estuarine plume range between +1.1 and+1.9 mol m–2 year–1 as a function ofthe formulation of the exchange coefficient ofCO2. The annual net emission of CO2from the estuarine plume to the atmosphere isestimated to be between +2.3 to +4.0 Gmolyear–1 which represents 17 to 29% of theestimate reported in the literature for the Scheldtinner estuary.  相似文献

Net O2 evolution, gross CO2 uptake and net HCO inf3 su– uptake during steady-state photosynthesis were investigated by a recently developed mass-spectrometric technique for disequilibrium flux analysis with cells of the marine cyanobacterium Synechococcus PCC7002 grown at different CO2 concentrations. Regardless of the CO2 concentration during growth, all cells had the capacity to transport both CO2 and HCO inf3 su– ; however, the activity of HCO inf3 su– transport was more than twofold higher than CO2 transport even in cyanobacteria grown at high concentration of inorganic carbon (Ci = CO2 + HCO inf3 su– ). In low-Ci cells, the affinities of CO2 and HCO inf3 su– transport for their substrates were about 5 (CO2 uptake) and 10 (HCO inf3 su– uptake) times higher than in high-Ci cells, while air-grown cells formed an intermediate state. For the same cells, the intracellular accumulated Ci pool reached 18, 32 and 55 mM in high-Ci, air-grown and low-Ci cells, respectively, when measured at 1 mM external Ci. Photosynthetic O2 evolution, maximal CO2 and HCO inf3 su– transport activities, and consequently their relative contribution to photosynthesis, were largely unaffected by the CO2 provided during growth. When the cells were adapted to freshwater medium, results similar to those for artificial seawater were obtained for all CO2 concentrations. Transport studies with high-Ci cells revealed that CO2 and HCO inf3 su– uptake were equally inhibited when CO2 fixation was reduced by the addition of glycolaldehyde. In contrast, in low-Ci cells steady-state CO2 transport was preferably reduced by the same inhibitor. The inhibitor of carbonic anhydrase ethoxyzolamide inhibited both CO2 and HCO inf3 su– uptake as well as O2 evolution in both cell types. In high-Ci cells, the degree of inhibition was similar for HCO inf3 su– transport and O2 evolution with 50% inhibition occurring at around 1 mM ethoxyzolamide. However, the uptake of CO2 was much more sensitive to the inhibitor than HCO inf3 su– transport, with an apparent I50 value of around 250 M ethoxyzolamide for CO2 uptake. Summary Analyses of carbon-assimilation patterns in response to intercellular CO2 concentrations, and the photosynthetic water-and nitrogen-use efficiencies, were conducted for a C3, a C4, and three C3–C4 species in the genus Flaveria in order to determine some of the advantages and disadvantages of C3–C4 intermediate photosynthesis. Operational intercellular CO2 partial pressures (pi), determined when the atmospheric CO2 partial pressure (pa) was approximately 330 bar, in the C3–C4 species were generally equal to, or greater than, those observed in the C3 species under well-watered or water-stressed conditions. This reflects equal, or lower, water-use efficiencies (WUEs) in the C3–C4 species. The only case in which higher WUEs were observed in the C3–C4 species, compared to the C3 species, was when photosynthesis rates were limited by available nitrogen and were less than 12.5 mol CO2 m-2s-1. At higher photosynthesis rates, the C3–C4 species exhibited lower values of photosynthesis rate for equal values of stomatal conductance (lower WUE), compared to the C3 species. Comparing slopes for the linear regions of the relationship between leaf nitrogen content and net photosynthesis rate (taken as an index of photosynthetic nitrogen-use efficiency, NUE), the C4 species exhibited the highest NUE, followed by the C3–C4 species, F. ramosissima, with the other two C3–C4 species and the C3 species being equal and exhibiting the lowest NUEs. The lack of consistent advantages in NUE and WUE in the C3–C4 species F. pubescens and F. floridana suggest that in some C3–C4 Flaveria species C4-like anatomy and biochemistry do not provide the same gas exchange advantages that we typically attribute to the CO2-concentrating mechanism of fully-expressed C4 plants.  相似文献

The chemical compositions of ground water and organic matter in sediments were investigated at a sandy shore of Tokyo Bay, Japan to determine the fate of ground water NO3 . On the basis of Cl distribution in ground water, the beach was classified into freshwater (FR)-, transition (TR)-, and seawater (SW)-zones from the land toward the shoreline. The NO3 and N2O did not behave conservatively with respect to Cl during subsurface mixing of freshwater and seawater, suggesting NO3 consumption and N2O production in the TR-zone. Absence of beach vegetation indicated that NO3 assimilation by higher plants was not as important as NO3 sink. Low NH4 + concentrations in ground water revealed little reduction of NO3 to NH4 +. These facts implied that microbial denitrification and assimilation were the likely sinks for ground water NO3 . The potential activity and number of denitrifiers in water-saturated sediment were highest in the low-chlorinity part of the TR-zone. The location of the highest potential denitrification activity (DN-zone) overlapped with that of the highest NO3 concentration. The C/N ratio and carbon isotope ratio (13C) of organic matter in sediment (< 100 -m) varied from 12.0 to 22.5 and from –22.5 to –25.5, respectively. The 13C value was inversely related to the C/N ratio (r 2 = 0.968, n = 11), which was explained by the mixing of organic matters of terrestrial and marine origins. In the DN-zone, the fine sediments were rich in organic matters with high C/N ratios and low 13C values, implying that dissolved organic matters of terrestrial origin might have been immobilized under slightly saline conditions. A concurrent supply of NO3 and organic matter to the TR-zone by ground water discharge probably generates favorable conditions for denitrifiers. Ground water NO3 discharged to the beach is thus partially denitrified and fixed as microbial biomass before it enters the sea. Further studies are necessary to determine the relative contribution of these processes for NO3 removal.  相似文献

One and a half year-old Ginkgo saplings were grown for 2 years in 7 litre pots with medium fertile soil at ambient air CO2 concentration and at 700 μmol mol−1 CO2 in temperature and humidity-controlled cabinets standing in the field. In the middle of the 2nd season of CO2 enrichment, CO2 exchange and transpiration in response to CO2 concentration was measured with a mini-cuvette system. In addition, the same measurements were conducted in the crown of one 60-year-old tree in the field. Number of leaves/tree was enhanced by elevated CO2 and specific leaf area decreased significantly.CO2 compensation points were reached at 75–84 μmol mol−1 CO2. Gas exchange of Ginkgo saplings reacted more intensively upon CO2 than those of the adult Ginkgo. On an average, stomatal conductance decreased by 30% as CO2 concentration increased from 30 to 1000 μmol mol−1 CO2. Water use efficiency of net photosynthesis was positively correlated with CO2 concentration levels. Saturation of net photosynthesis and lowest level of stomatal conductance was reached by the leaves of Ginkgo saplings at >1000 μmol mol−1 CO2. Acclimation of leaf net CO2 assimilation to the elevated CO2 concentration at growth occurred after 2 years of exposure. Maximum of net CO2 assimilation was 56% higher at ambient air CO2 concentration than at 700 μmol mol−1 CO2.  相似文献

Gas exchange and chlorophyll (Chl) fluorescence were measured on young mature leaves of rose plants (Rosa hybrida cvs. First Red and Twingo) grown in two near-to-tight greenhouses, one under control ambient CO2 concentration, AC (355 µmol mol–1) and one under CO2 enrichment, EC (700 µmol mol–1), during four flushes from late June to early November. Supply of water and mineral elements was non-limiting while temperature was allowed to rise freely during daytime. Leaf diffusive conductance was not significantly reduced at EC but net photosynthetic rate increased by more than 100 %. Although the concentration of total non-structural saccharides was substantially higher in the leaves from the greenhouse with EC, PS2 (quantum efficiency of radiation use) around noon was not significantly reduced at EC indicating that there was no down-regulation of electron transport. Moreover, CO2 enrichment did not cause any increase in the risk of photo-damage, as estimated by the 1 – qP parameter. Non-photochemical quenching was even higher in the greenhouse with EC during the two summer flushes, when temperature and photosynthetic photon flux density (PPFD) were the highest. Hence rose photosynthesis benefits strongly from high concentrations of atmospheric CO2 at both high and moderate temperatures and PPFD.  相似文献

We investigated the influence of CO2/HCO3 -depletion and of the presence of acetate and formate on the in vivo photosynthetic electron transport in the two green algae Chlamydobotrys stellata and Chlamydomonas reinhardtii by means of thermoluminescence technique and mathematical glow curve analysis. The main effects of the removal of CO2 from the algal cultures was: (1) A shift of the glow curve peak position to lower temperatures resulting from a decrease of the B band and an increase of the Q band. (2) Treatment of CO2-deficient Chl. stellata with DCMU yielded two thermoluminescence bands in the Q band region peaking at around +12°C and +5°C; in case of Chl. reinhardtii DCMU treatment induced only one band with an emission maximum at +5°C. The presence of acetate or formate in CO2-depleted algal cultures lowered the intensities of all of the individual TL bands but that of a HT band (TL+37). The influence of elevated CO2 concentrations on growth and photosynthesis ofGracilaria sp. andG. chilensis was investigated in order to procure information on the effective utilization of CO2. Growth of both was enhanced by CO2 enrichment (air + 650 ppm CO2, air + 1250 ppm CO2, the enhancement being greater inGracilaria sp. Both species increased uptake of NO3 with CO2 enrichment. Photosynthetic inorganic carbon uptake was depressed inG. chilensis by pre-culture (15 days) with CO2 enrichment, but little affected inGracilaria sp. Mass spectrometric analysis showed that O2 uptake was higher in the light than in the dark for both species and in both cases was higher inGracilaria sp. The higher growth enhancement inGracilaria sp. was attributed to greater depression of photorespiration by the enrichment of CO2 in culture.  相似文献

Summary In order to document the natural CO2 environment of the moss Hylocomium splendens, and ascertain whether or not the moss was adapted to this, and its interactions with other microenvironmental factors, two studies were carried out. Firstly, the seasonal variations of CO2 concentration, photosynthetically active radiation (PAR), tissue water content and temperature were measured in the natural microenvironment of H. splendens in a subarctic forest during the summer period (July–September). Secondly, the photosynthetic responses of the species to controlled CO2 concentrations, PAR, temperature, and hydration were measured in the laboratory. CO2 concentrations around the upper parts of the plant, when PAR was above the compensation point (30 mol m–2 s–1), were mostly between 400 and 450 ppm. They occasionally increased up to 1143 ppm for short periods. PAR flux densities below saturating light levels for photosynthesis (100 mol m–2 s–1), occurred during 65% (July), 76% (August) and 96% (September) of the hours of the summer period. The temperature optimum of photosynthesis was 20° C: this temperature coincided with PAR above the compensation point during 5%, 6% and 0% of the time in July, August and September, respectively. Optimal hydration of tissues was infrequent. Hence PAR, temperature and water limit CO2 uptake for most of the growing season. Our data suggest that the higher than normal ambient CO2 concentration in the immediate environment of the plant counteracts some of the limitations in PAR supply that it experiences in its habitat. On reaching the respiratory compensation point (RCP) during rapidly increasing incremental exercise, the ratio of minute ventilation (VE) to CO2 output (VCO2) rises, which coincides with changes of arterial partial pressure of carbon dioxide (P aCO2). Since P aCO2 changes can be monitored by transcutaneous partial pressure of carbon dioxide (PCO2,tc) RCP may be estimated by PCO2,tc measurement. Few available studies, however, have dealt with comparisons between PCO2,tc threshold (T AT) and lactic, ventilatory or gas exchange threshold (V AT), and the results have been conflicting. This study was designed to examine whether this threshold represents RCP rather than V AT. A group of 11 male athletes performed incremental excercise (25 W · min–1) on a cycle ergometer. The PCO2,tc at (44°C) was continuously measured. Gas exchange was computed breath-by-breath, and hyperaemized capillary blood for lactate concentration ([la]b) and P aCO2 measurements was sampled each 2 min. The T AT was determined at the deflection point of PCO2,tc curve where PCO2,tc began to decrease continuously. The V AT and RCP were evaluated with VCO2 compared with oxygen uptake (VO2) and VE compared with the VCO2 method, respectively. The PCO2,tc correlated with P aCO2 and end-tidal PCO2. At T AT, power output [P, 294 (SD 40) W], VO2 [4.18 (SD 0.57)l · min–1] and [la] [4.40 (SD 0.64) mmol · l–1] were significantly higher than those at V AT[P 242 (SD 26) W, VO2 3.56 (SD 0.53) l · min–1 and [la]b 3.52 (SD 0.75), mmol · l–1 respectively], but close to those at RCP [P 289 (SD 37) W; VO2 3.97 (SD 0.43) l · min and [la]b 4.19 (SD 0.62) mmol · l–1, respectively]. Accordingly, linear correlation and regression analyses showed that P, VO2 and [la]b at T AT were closer to those at RCP than at V AT. In conclusion, the T AT reflected the RCP rather than V AT during rapidly increasing incremental exercise.  相似文献

Single leaf photosynthesis (Pn) and stomatal conductance (Cg) of drought stressed and nonstressed pearl millet [Pennisetum americanum (L.) Leeke] were measured across growth stages to determine if a pattern exists in Pn and Cg during the growing season and to evaluate the influence of air vapor pressure deficit (VPDa) on the seasonal variations of Pn and Cg. Leaf photosynthesis and Cg were measured independently on pearl millet plants grown at the driest (drought stressed) and wettest (nonstressed) ends of a line-source irrigation gradient system. Well defined and predictable variations in both Pn and Cg were found across two growing seasons. Leaf photosynthesis of the nonstressed plants declined from a maximumof 25.8 mol m–2 s–1 at the flag leaf emergence (48 days after planting, DAP) to a minimum of 14.5 mol m–2 s–1 at physiological maturity. Stomatal conductance of the nonstressed plants peaked at the flowering and early grain fill stages and declined as plants approached maturity. In contrast, Pn and Cg of the stressed plants declined from a maximum at flag leaf emergence to a minimum at flowering and increased as plants approached maturity. High VPDa during the flowering and grain fill stages induced stomatal closure and decreased Pn in the stressed plants. High mid-season VPDa did not induce stomatal closure and did not reduce leaf photosynthesis in nonstressed plants. Summary Cells ofChara corallina grown under high CO2 culture conditions were able to utilize exogenous HCO3 to give appreciable rates of net photosynthesis. Since these rates of photosynthesis could be detected within 10 min of being transferred from high-CO2 to normal HCO3 (pH 8.2) culture conditions, it would appear that the HCO3 -accumulating system ofChara is not fully repressed under these high CO2 culture conditions. The membrane potential of these cells also responded to light/dark treatments in a manner consistent with the operation of a HCO3 acquisition system. With prolonged exposure (2–6 days) to CPW/B, net photosynthesis continued to increase towards the expected control rate and, in parallel, the electrical responses elicited by light/dark treatments converged towards those obtained on control (CPW/B-grown)Chara cells. Charasomes were absent in CPW/CO2-grownChara, but redeveloped in mature cells once the culture was returned to CPW/B conditions; a minimum period of 7 days in CPW/B was required before charasomes were detected in tissue examined in the transmission electron microscope. Onion (Allium cepa L.) plants were examined to determine the photosynthetic role of CO2 that accumulates within their leaf cavities. Leaf cavity CO2 concentrations ranged from 2250 L L–1 near the leaf base to below atmospheric (<350 L L–1) near the leaf tip at midday. There was a daily fluctuation in the leaf cavity CO2 concentrations with minimum values near midday and maximum values at night. Conductance to CO2 from the leaf cavity ranged from 24 to 202 mol m–2 s–1 and was even lower for membranes of bulb scales. The capacity for onion leaves to recycle leaf cavity CO2 was poor, only 0.2 to 2.2% of leaf photosynthesis based either on measured CO2 concentrations and conductance values or as measured directly by 14CO2 labeling experiments. The photosynthetic responses to CO2 and O2 were measured to determine whether onion leaves exhibited a typical C3-type response. A linear increase in CO2 uptake was observed in intact leaves up to 315 L L–1 of external CO2 and, at this external CO2 concentration, uptake was inhibited 35.4±0.9% by 210 mL L–1 O2 compared to 20 mL L–1 O2. Scanning electron micrographs of the leaf cavity wall revealed degenerated tissue covered by a membrane.
