In this study, we show that 5α‐reductase derived from rat fresh liver was inhibited by certain aliphatic free fatty acids. The influences of chain length, unsaturation, oxidation, and esterification on the potency to inhibit 5α‐reductase activity were studied. Among the fatty acids we tested, inhibitory saturated fatty acids had C12–C16 chains, and the presence of a C?C bond enhanced the inhibitory activity. Esterification and hydroxy compounds were totally inactive. Finally, we tested the prostate cancer cell proliferation effect of free fatty acids. In keeping with the results of the 5α‐reductase assay, saturated fatty acids with a C12 chain (lauric acid) and unsaturated fatty acids (oleic acid and α‐linolenic acid) showed a proliferation inhibitory effect on lymph‐node carcinoma of the prostate (LNCaP) cells. At the same time, the testosterone‐induced prostate‐specific antigen (PSA) mRNA expression was down‐regulated. These results suggested that fatty acids with 5α‐reductase inhibitory activity block the conversion of testosterone to 5α‐dihydrotestosterone (DHT) and then inhibit the proliferation of prostate cancer cells.  相似文献

The fatty acids composition of the polar and non-polar lipid fractions of wheat leaves was affected due to progressive brown rust infection during early stages of pathogenesis,i.e. 0, 12, 24, 36, 48 and 72 h after inoculation. The three races ofPuccinia recondita differentially affected the composition of saturated and unsaturated fatty acids and their relative occurrence in wheat leaves. The infection of wheat by race 77 resulted in a relative decrease in fatty acid chain length as measured through C16∶C18 fatty acid ratio. An increase in the relative degree of unsaturation (18∶2/18∶3 acids ratio) was recorded in both lipid fractions. Such changes may be taken as one of the earliest characteristics of disease development.  相似文献

The monocarboxylic fatty acids and hydroxy fatty acids of three species of freshwater microalgae—Vischeria punctata Vischer, Vischeria helvetica (Vischer et Pascher) Taylor, and Eustigmatos vischeri (Hulbert) Taylor, all from the class Eustigmatophyceae— were examined. Each species displayed a very similar distribution of fatty acids, the most abundant of which were 20:5n-3, 16:0, and 16:1n-7; C18 polyunsaturated fatty acids were minor components. These fatty acid distributions closely resemble those found in marine eustigmatophytes but are quite distinct from those found in most other algal classes. These microalgae also contain long-chain saturated and unsaturated monohydroxy fatty acids. Two distinct types of hydroxy fatty acids were found: a series of saturated α-hydroxy acids ranging from C24 to C30 with a shorter series of monounsaturated α-hydroxy acids ranging from C26 to C30 together with a series of saturated β-hydroxy acids ranging from C26 to C30. The latter have not previously been reported in either marine or freshwater microalgae, although C30 to C34 midchain (ω-18)-hydroxy fatty acids have been identified in hydrolyzed extracts from marine eustigmatophytes of the genus Nannochloropsis, and C22 to C26 saturated and monounsaturated α-hydroxy fatty acids have been found in three marine chlorophytes. These findings have provided a more complete picture of the lipid distributions within this little studied group of microalgae as well as a range of unusual compounds that might prove useful chemotaxonomic markers. The functions of the hydroxy fatty acids are not known, but a link to the formation of the lipid precursors of highly aliphatic biopolymers is suggested.  相似文献

Previous work from our laboratory has shown dinoflagellates, which possess the carotenoid peridinin, have been divided into two clusters based on plastid galactolipid fatty acid composition. In one cluster major forms of monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG), lipids that comprise the majority of photosynthetic membranes, were C18/C18 (sn‐1/sn‐2), with octadecapentaenoic [18:5(n‐3)] and octadecatetraenoic [18:4(n‐3)] acid as principal fatty acids. The other cluster contained C20/C18 major forms, with eicosapentaenoic acid [20:5(n‐3)] being the predominant sn‐1 fatty acid. In this study, we have found that Symbiodinium microadriaticum isolated from the jellyfish, Cassiopea xamachana, when grown at 30°C, produced MGDG and DGDG with a more saturated fatty acid, 18:4(n‐3), at the sn‐2 carbon than when grown at 20°C where 18:5(n‐3) predominates. This modulation of the sn‐2 fatty acid's level of saturation is mechanistically similar to what has been observed in Pyrocystis, a C20/C18 dinoflagellate. We have also examined the effect of growth temperature on the betaine lipid, diacylglycerylcarboxyhydroxymethylcholine (DGCC), which has been observed by others to be the predominant non plastidial polar lipid in dinoflagellates. Temperature effects on it were minimal, with very few modulations in fatty acid unsaturation as observed in MGDG and DGDG. Rather, the primary difference seen at the two growth temperatures was the alteration of the amount of minor forms of DGCC, as well as a second betaine lipid, diacylglyceryl‐N,N,N‐trimethylhomoserine.  相似文献

Seasonal changes in the fatty acid composition of the total lipid extracted from the whole body of Cydia pomonella L. larvae were determined by gas chromatography. The six most abundant fatty acids in both non-diapause and diapause larvae of codling moth were oleic (35%-39%), palmitic (23%-33%), linoleic (16%-30%), palmitoleic (5%-10%), stearic (1.5%-3.0%) and linolenic acids (1.0%-2.5%). This represents a typical complement of Lepidopteran fatty acids. The fatty acid composition of total lipid of C. pomonella larvae was related to diapause. In similarity to most other reports, the proportion of unsaturated fatty acids increased in diapause initiation state. The total lipid of diapause larvae contained more linoleic acid (25.8% vs. 16.1%) and less palmitic acid (24.7% vs. 33.4%), than that of non-diapause larvae. The weight percentage of linoleic acid (C 18:2) increased from 16% to 26% from early-August through early-September during transition to diapause, while palmitic acid (C16:0) decreased from 33% to 25% at the same time. These changes resulted in an increase in the ratio of unsaturated to saturated fatty acids (UFA/SFA) from 1.72 in non-diapause larvae to 2.63 in diapause larvae.  相似文献

The development of renewable alternatives to diesel and jet fuels is highly desirable for the heavy transportation sector, and would offer benefits over the production and use of short‐chain alcohols for personal transportation. Here, we report the development of a metabolically engineered strain of Escherichia coli that overproduces medium‐chain length fatty acids via three basic modifications: elimination of β‐oxidation, overexpression of the four subunits of acetyl‐CoA carboxylase, and expression of a plant acyl–acyl carrier protein (ACP) thioesterase from Umbellularia californica (BTE). The expression level of BTE was optimized by comparing fatty acid production from strains harboring BTE on plasmids with four different copy numbers. Expression of BTE from low copy number plasmids resulted in the highest fatty acid production. Up to a seven‐fold increase in total fatty acid production was observed in engineered strains over a negative control strain (lacking β‐oxidation), with a composition dominated by C12 and C14 saturated and unsaturated fatty acids. Next, a strategy for producing undecane via a combination of biotechnology and heterogeneous catalysis is demonstrated. Fatty acids were extracted from a culture of an overproducing strain into an alkane phase and fed to a Pd/C plug flow reactor, where the extracted fatty acids were decarboxylated into saturated alkanes. The result is an enriched alkane stream that can be recycled for continuous extractions. Complete conversion of C12 fatty acids extracted from culture to alkanes has been demonstrated yielding a concentration of 0.44 g L?1 (culture volume) undecane. Biotechnol. Strains ofRhodotorula sp.,Candida spp. andLangermania sp. cultivated on polyunsaturated oil preferentially incorporated more unsaturated fatty acids. These fatty acids were used mainly for growth needs whereas the saturated ones accumulated in the microbial cell. The cellular oil and the remaining oil in the culture had a lower degree of unsaturation as compared to the initial oil, and a modified fatty acid composition.Candida lipolytica, in a chemostat continuous culture, incorporated C18 fatty acids in the order of C18:3>C18:2>C18:1>C18:0, and accumulated mostly the saturated ones. The specific productivity of the cellular oil and of the oil remaining in the culture medium was 0.036 and 0.487 gg−1 h−1, respectively, at dilution rateD=0.2/h.  相似文献

Ultrastructure and lipid chemistry of specialized epidermal structure of Indian porcupines and hedgehog. —Acta Zoologica (Stockholm) 92 : 134–140. In the present study, we investigated the ultrastructural variations of specialized epidermal structure of Indian porcupines (Hystrix indica and Atherurus macrourus) and hedgehog (Hemiechinus collaris) as well as the variation in the fatty acid composition of total lipid fraction. Scanning electron microscope images reveal the usual scaly structure in surface view and network of channels in cross‐section but with different orientation of partition walls. The lipid profile reveals the presence of free sterol, long‐chain alcohol, free fatty acids, wax ester and sterol ester in all the three cases and trace amount of triglyceride, diglyceride and monoglyceride. Gas chromatography–mass spectrometry analysis of fatty acid methyl ester of total lipid fraction indicates the presence of C8‐C22 fatty acids in Hystrix indica, C8‐C18 in Atherurus macrourus and C8‐C20 fatty acids in Hemiechinus collaris. It is interesting to note that the total lipid fraction of hedgehog shows no branched‐chain, unsaturated and odd‐carbon fatty acids. Odd‐carbon fatty acid and branched‐chain fatty acids detected in the adult H. indica but were absent in juvenile H. indica as well as in A. macrourus. With the exception of C18:1, the other unsaturated fatty acids were also absent in both juvenile H. indica and A. macrourus.  相似文献

Changes in fatty acid composition, cholesterol and fat‐soluble vitamins were studied during development (fertilized eggs, yolk‐sac larvae, and after yolk resorption of shabbout, Barbus grypus). Significant differences were found in the monounsaturated fatty acids (MUFAs), polyunsaturated fatty acids (PUFA), PUFA/saturated fatty acids (SFA), ∑n‐3 and eicosapentaenoic acid (EPA)/docosahexaenoic acid (DHA) ratios between eggs and larvae (P < 0.05). Significant differences were also observed in the C14:0, C16:1n‐7, C18:1n‐9, C18:3n‐6, C20:0, C20:4n‐6, C24:0, C24:1, C22:6n‐3 fatty acids between eggs and larvae after yolk‐sac resorption (P < 0.05). Vitamin α‐ Tocopherol and retinol content increased during embryogenesis, but changes were insignificant in retinol acetate, δ‐Tocopherol, K1, K2 and cholesterol content between eggs and larvae after yolk resorption (P > 0.05).  相似文献

An improved method is described for separating saturated fatty acids by reversed-phase paper chromatography. The 2,4-dinitrophenylhydrazides prepared from saturated fatty acids from C2 to C22 are run upward on paper impregnated with tetralin, using 90% methanol—acetic acid—tetralin, 80% ethanol—acetic acid—tetralin, or 90% methanol—tetralin as the moving solvent. The simultaneous separations of all even-carbon acids from C6 to C22, all odd-carbon acids from C7 to C19, and also all odd- and even-carbon acids from C7 to C19 can be successfully performed by means of this paper chromatography. The method is useful for the detection of component saturated fatty acids in natural fats.  相似文献

The Chlorarachniophyceae are unicellular eukaryotic algae characterized by an amoeboid morphology that may be the result of secondary endosymbiosis of a green alga by a nonphotosynthetic amoeba or amoeboflagellate. Whereas much is known about the phylogeny of chlorarachniophytes, little is known about their physiology, particularly that of their lipids. In an initial effort to characterize the lipids of this algal class, four organisms from three genera were examined for their fatty acid and sterol composition. Fatty acids from lipid fractions containing chloroplast‐associated glycolipids, storage triglycerides, and cytoplasmic membrane‐associated polar lipids were characterized. Glycolipid‐associated fatty acids were of limited composition, principally eicosapentaenoic acid [20:5(n‐3)] and hexadecanoic acid (16:0). Triglyceride‐associated fatty acids, although minor, were found to be similar in composition. The polar lipid fraction was dominated by lipids that did not contain phosphorus and had a more variable fatty acid composition with 16:0 and docosapentaenoic acid [22:5(n‐3)] dominant along with a number of minor C18 and C20 fatty acids. Crinosterol and one of the epimeric pair poriferasterol/stigmasterol were the sole sterols. Several genes required for synthesis of these sterols were computationally identified in Bigelowiella natans Moestrup. One sterol biosynthesis gene showed the greatest similarity to SMT1 of the green alga, Chlamydomonas reinhardtii. However, homologues to other species, mostly green plant species, were also found. Further, the method used for identification suggested that the sequences were transferred to a genetic compartment other than the likely original location, the nucleomorph nucleus.  相似文献

Using the paper disc-agar plate method, a number of fatty and related acids have been tested for tested activity for inhibiting the growth of Chlorella pyrenoidosa Chick. Of the saturated acids, a peak in growth inhibiting activity wax observed in the C7–C12 range, where inhibition wax observed when solutions down to 0.02 M were applied to the discs. Most of the unsaturated acids tested showed greater inhibition than did the corresponding saturated acids. Acrylic acid showed detectable inhibition at 0.001 M concentration.  相似文献

Aims: This study provides a first approach to observing the alterations of the cell membrane lipids in the adaptation response of Listeria monocytogenes to the sanitizer benzalkonium chloride. Methods and Results: A thorough investigation of the composition of polar and neutral lipids from L. monocytogenes grown when exposed to benzalkonium chloride is compared to cells optimally grown. The adaptation mechanism of L. monocytogenes in the presence of benzalkonium chloride caused (i) an increase in saturated‐chain fatty acids (mainly C16:0 and C18:0) and unsaturated fatty acids (mainly C16:1 and C18:1) at the expense of branched‐chain fatty acids (mainly Ca‐15:0 and Ca‐17:0) mainly because of neutral fatty acids; (ii) no alteration in the percentage of neutral and polar lipid content among total lipids; (iii) a decrease in lipid phosphorus and (iv) an obvious increase in the anionic phospholipids and a decrease in the amphiphilic phosphoaminolipid. Conclusions: These lipid changes could lead to decreased membrane fluidity and also to modifications of physicochemical properties of cell surface and thus changes in bacterial adhesion to abiotic surfaces. Significance and Impact of the Study: The adaptation and resistance of L. monocytogenes to disinfectants is able to change its physiology to allow growth in food‐processing plants. Understanding microbial stress response mechanisms would improve the effective use of disinfectants.  相似文献

An analytical method for the determination of saturated very long chain (VLC) fatty acids in the serum has been devised. Free fatty acids obtained after hydrolysis of total lipid extracts were converted intop-bromophenacyl esters. The derivatives were purified in two sequential steps by clean-up on C18 reversed-phase cartridge and fractionation by reversed-phase thin-layer chromatography (TLC), and then quantitated by high performance liquid chromatography (HPLC) analysis. This technique provides a reliable and alternative method for the biochemical identification of patients and carriers of an inherited metabolic disease characterized by the accumulation of saturated VLC fatty acids (C24–C26) such as Adrenoleukodystrophy (ALD). In four cases of diagnosed ALD the fatty acid composition of serum total lipids was dramatically enriched in saturated VLC fatty acids compared to controls. The ratio of hexacosanoic acid (C260) to docosanoic acid (C220) in ALD patients was approximately six-fold higher than that of healthy controls or patients affected by metabolic or neurological disorders other than ALD.  相似文献

The fatty acid composition of five strains ofCorallococcus coralloides and three reference species ofMyxococcus were determined by gas-liquid chromatography. Methyl esters of fatty acid containing from 12 to 22 carbon atoms were identified. The major fatty acids present were C15 and C17 saturated branched chain, and both C16 saturated and unsaturated straight chain acids. The C17 saturated branched and straight chain acids, which were in valuable concentration in species ofMyxococcus, were not, however, detected in all strains ofC. coralloides. The application of these results in the distinction ofC. coralloides from the genusMyxococcus is discussed.  相似文献

Zusammenfassung Die physikalischen und chemischen Kennzahlen und Daten des durch Ätherextraktion gewonnenen Rohlipids von Beauveria tenella wurden bestimmt und spektrale Absorptionskurven aufgenommen.Die Methylester der Fettsäuren wurden gaschromatographisch analysiert. Die einzelnen Fettsäuren wurden identifiziert und ihr mengenmäßiger Anteil ermittelt.Etwa 90% der Fettsäuren haben eine Kettenlänge von 16–18 Kohlenstoffatomen. Nahezu 60% der Fettsäuren sind ungesättigt; der prozentuale Anteil dieser Säuren nimmt mit steigender Zahl der Doppelbindungen ab.Octadecensäure hatte mit 25,6% den größten Anteil an der Gesamtmenge.An seltenen Fettsäuren konnte unter anderem eine gesättigte C17- und C24-Fettsäure nachgewiesen und das Vorkommen einer gesättigten C19-und C21-Fettsäure wahrscheinlich gemacht werden.
Summary The physical and chemical constants and the spectral absorption curves of crude lipids of Beauveria tenella, obtained by means of etherextraction, were determined.The methyl esters of the fatty acids were analyzed using gas-liquid chromatography. The individual fatty acids were identified and the amount of each in the complete sample determined.Approximately 90% of the fatty acids had a 16–18 carbon-chain length, while almost 60% of the fatty acids were unsaturated. The percentage of fatty acids with unsaturated bonds was reciprocally proportional to the number of double bonds present.Octadecenoic acid comprised 25,6% of the total fatty acids and represented the largest single amount of a specific fatty acid present.With respect to unusual fatty acids, a saturated C17 and a C24 fatty acid were identified, while the presence of a C19 and a C21 fatty acid was indicated.

The phospholipid fatty acid composition and poly--hydroxybutyrate (PHB) content of initially adhered and free-living cells of a Pseudomonas sp. isolated from the rape plant Brassica napus were examined with gas chromatography (GC). Five different adhesion experiments were made including variations in surface charge (hydrophilic and lipophilic), temperature, media composition and time of adhesion. Lipids and poly--hydroxybutyrate (PHB) were extracted with a chloroform-methanol-water mixture, hydrolyzed and esterified with pentafluorobenzyl bromide. Analysis was performed with capillary gas chromatography and flame ionization detection. A pronounced difference in both the ratio saturated/unsaturated fatty acids and in PHB content between free-living and adhered bacteria were found. The free-living bacteria has a significantly smaller ratio of saturated/unsaturated C16 and C18 fatty acids and also a smaller ratio of total C18/total C16 fatty acids. Bacteria adhered to the lipophilic surface had a higher ratio of saturated to unsaturated C16 fatty acids than at the hydrophilic surface. There were no major differences between the treatments regarding the amount of bacteria adhered to the surface or their lipid composition.  相似文献

A feeding trial was conducted to determine the effect of replacing costly cod liver oil with corn oil as a source of dietary lipid on the growth and fatty acid composition of the larval freshwater prawn, Macrobrachium rosenbergii de Man. Prawn larvae were weaned to artificial diets containing cod liver oil and corn oil either singly or in various combinations (2 : 1, 1 : 1, 1 : 2, w/w). Weaning to artificial diets from Artemia nauplii commenced at larval stage III with complete substitution by stage X. The reference group was reared solely on Artemia nauplii during the entire experiment. Incorporation of corn oil at 33–67% of dietary supplemental oil did not have significant effects on the post‐larval production. However, larvae fed with corn oil alone revealed a significantly lower post‐larval production compared to other experimental diets as well as to the reference group. No significant differences (P > 0.05) were observed in dry weight, protein and lipid concentration among larvae fed on various dietary treatments. Palmitic (16 : 0) and oleic/vaccenic (18 : 1) acids were the dominant saturated and monounsaturated fatty acids in larval tissues, respectively, whereas the polyunsaturated fraction was dominated by eicosapentaenoic (20 : 5n‐3) acid. The polyunsaturated fatty acid composition was dominated by n‐3 acids rather than n‐6 fatty acids. The fatty acid composition of the prawn in general reflected that of the diet. Larvae on diets containing higher concentrations of corn oil rich in linoleic (18 : 2n‐6) acid showed a higher concentration of this acid in their tissues. No evidence of de novo synthesis of linoleic (18 : 2n‐6) acid was found. Higher levels of stearic (18 : 0), arachidonic (20 : 4n‐6) and eicosapentaenoic (20 : 5n‐3) acids found in larvae as compared with those fed Artemia and artificial diets strongly indicated the larval ability in chain elongation and desaturation of palmitic (16 : 0), linoleic (18 : 2n‐6) or linolenic (18 : 3n‐3) acids, respectively. Despite a large variation of n‐3 to n‐6 ratios of the live and artificial diets, larval n‐3 to n‐6 ratios were relatively stable among different dietary treatments, possibly indicative of the importance of such a ratio in the larval fatty acid metabolism.  相似文献

The orange wheat blossom midge, Sitodiplosis mosellana (Géhin), can significantly reduce wheat yield. Growing resistant wheat cultivars is an effective way of managing this pest. The assessment of cultivar resistance in field trials is difficult because of unequal pressure of S. mosellana caused by differences in cultivar heading dates relative to the flight period of S. mosellana adult females and huge variations of egg laying conditions from 1 day to another. To overcome these hurdles and to expose all cultivars homogeneously to the pest, an assessment method of cultivar resistance was developed under semi‐field conditions. In 2015, the resistance of 64 winter wheat cultivars to S. mosellana was assessed. Few or no larvae developed in the ears of resistant cultivars, but in susceptible cultivars, large numbers of larvae developed. Seventeen cultivars proved to be resistant, whereas 47 were susceptible. The identification of new resistant cultivars offers more opportunities to manage S. mosellana. The phenotyping method is easy, cheap, efficient and reliable. It can be used to guide the breeding of new resistant wheat cultivars. Using specific midge populations, this method could also be used in research on new resistance mechanisms in winter wheat or in other cereal species.  相似文献

Qualitative and quantitative profiles of phospholipids, neutral lipids, and fatty acid composition in Cr. neoformans during the growth phase were investigated in relation to pyrophosphatidic acid. A marked increase of the total lipid content, which depended on the accumulation of triglyceride in yeast cells with the growth, was observed. The total phospholipid contents in yeast cells remained almostly constant during the exponential phase and slightly decreased in the stationary phase. The major phospholipids of this yeast were phosphatidylcholine, phosphatidylethanolamine, phosphatidylinositol, phosphatidylserine, and cardiolipin, the next groups being pyrophosphatidic acid, phosphatidic acid, lysophos-phatidylcholine, and unidentified components. The amounts of phosphatidylcholine, phosphatidylinositol, and cardiolipin were fairly constant throughout the growth phase, but the amount of phosphatidylethanolamine increased and that of phosphatidylserine decreased with progressive growth. The pyrophosphatidic acid contents were 0.9~0.7% for total phospholipid during the growth phase. The major fatty acids of pyrophosphatidic acid were C16:0, C18:1, and C18:2 acids. The changing patterns of fatty acid composition in pyrophosphatidic acid through the growth phase closely resembled that of phosphatidic acid, which contained larger amounts of C18:1 acid (35~45%) than C16:0 acid (30~25%) and C18:2 acid (30~25%). Phosphatidylserine and phosphatidylinositol contained considerable amounts of saturated fatty acid (C16:0 acid, more than 55%). On the other hand, phosphatidylcholine, phosphatidylethanolamine, and cardiolipin contained extremely large amounts of unsaturated fatty acid (C18:1 and C18:2 acid, 85ç90%).  相似文献
