The cellular long-chain fatty acids of 36 strains representing 18 genera of the Saccharomycetaceae, Endomycetaceae, Metchnikowiaceae, Saccharomycodaceae, Schizosaccharomycetaceae and Dipodascaceae were extracted and analyzed as methyl esters by gas chromatography. On the basis of their fatty acid content the set of strains was divided into 6 groups, coinciding with the above families. The members of the Saccharomycetaceae (group I) had a high percentage of oleic acid while the strains classified under the Endomycetaceae (group II) and Metchnikowiaceae (group III) were characterized by oleic acid and linoleic acid as major fatty acids. The Saccharomycodaceae (group IV) had the highest percentage of palmitoleic acid. The Schizosaccharomycetaceae (group V) had the highest percentage of oleic acid, while the Dipodascaceae (group VI) were characterized by a high percentage of linoleic acid.This article originally appeared in an incorrect form in Antonie van Leeuwenhoek, Vol. 52, No. 1.  相似文献

Abstract The cellular long-chain fatty acids of 32 strains representing 10 species of the genus Kluyveromyces were extracted by saponification and analyzed as methyl esters by gas chromatography. The Kluyveromyces strains were characterized by the presence of palmitate, palmitoleate, oleate, and linoleic acid as the major fatty acids. These strains were divided into 3 groups on the basis of fatty acid content. 1. Three groups of female rats (8-12 weeks old) were maintained respectively on a linoleic acid-rich diet, a linoleic acid-poor predominantly saturated-fatty acid diet and a normal diet. Changes in the fatty acid compositions of serum, brain, brain mitochondria-rich fraction and myelin were observed. 2. Of the serum fatty acids, linoleic acid showed the greatest change in the percentage of the total acids in response to diet; the change in the proportion of oleic acid was considerable. The percentages of arachidonic acid in serum fatty acids in the groups on the linoleic acid-rich and linoleic acid-poor diets were similar, but higher than those in the normal group. 3. Changes in the proportions of linoleic acid, arachidonic acid and docosahexaenoic acid occurred in brain fatty acids that to some extent paralleled those occurring in the serum. Changes in the proportions of most other acids in the serum fatty acids were not accompanied by corresponding changes in the brain fatty acids. 4. The percentage fatty acid compositions of a mitochondria-rich fraction and myelin are given, and changes in the relative proportions of linoleic acid, arachidonic acid and possibly some docosapolyenoic acids were demonstrated to occur as a result of diet. 5. The results are discussed in relation to the possible aetiology of multiple sclerosis.  相似文献

The fatty acid methyl ester (FAME) analysis of the 12 heterocystous cyanobacterial strains showed different fatty acid profiling based on the presence/absence and the percentage of 13 different types of fatty acids. The major fatty acids viz. palmitic acid (16:0), hexadecadienoic acid (16:2), stearic acid (18:0), oleic acid (18:1), linoleic (18:2), and linolenic acid (18:3) were present among all the strains except Cylindrospermum musicola where oleic acid (18:1) was absent. All the strains showed high levels of polyunsaturated fatty acid (PUFAs; 41-68.35%) followed by saturated fatty acid (SAFAs; 1.82-40.66%) and monounsaturated fatty acid (0.85-24.98%). Highest percentage of PUFAs and essential fatty acid (linolenic acid; 18:3) was reported in Scytonema bohnerii which can be used as fatty acid supplement in medical and biotechnological purpose. The cluster analysis based on FAME profiling suggests the presence of two distinct clusters with Euclidean distance ranging from 0 to 25. S. bohnerii of cluster I was distantly related to the other strains of cluster II. The genotypes of cluster II were further divided into two subclusters, i.e., IIa with C. musicola showing great divergence with the other genotypes of IIb which was further subdivided into two groups. Subsubcluster IIb(1) was represented by a genotype, Anabaena sp. whereas subsubcluster IIb(2) was distinguished by two groups, i.e., one group having significant similarity among their three genotypes showed distant relation with the other group having closely related six genotypes. To test the validity of the fatty acid profiles as a marker, cluster analysis has also been generated on the basis of morphological attributes. Our results suggest that FAME profiling might be used as species markers in the study of polyphasic approach based taxonomy and phylogenetic relationship.  相似文献

We investigated the changes in adiposity, cardiovascular and liver structure and function, and tissue fatty acid compositions in response to oleic acid-rich macadamia oil, linoleic acid-rich safflower oil and α-linolenic acid-rich flaxseed oil (C18 unsaturated fatty acids) in rats fed either a diet high in simple sugars and mainly saturated fats or a diet high in polysaccharides (cornstarch) and low in fat. The fatty acids induced lipid redistribution away from the abdomen, more pronounced with increasing unsaturation; only oleic acid increased whole-body adiposity. Oleic acid decreased plasma total cholesterol without changing triglycerides and nonesterified fatty acids, whereas linoleic and α-linolenic acids decreased plasma triglycerides and nonesterified fatty acids but not cholesterol. α-Linolenic acid improved left ventricular structure and function, diastolic stiffness and systolic blood pressure. Neither oleic nor linoleic acid changed the left ventricular remodeling induced by high-carbohydrate, high-fat diet, but both induced dilation of the left ventricle and functional deterioration in low fat-diet-fed rats. α-Linolenic acid improved glucose tolerance, while oleic and linoleic acids increased basal plasma glucose concentrations. Oleic and α-linolenic acids, but not linoleic acid, normalized systolic blood pressure. Only oleic acid reduced plasma markers of liver damage. The C18 unsaturated fatty acids reduced trans fatty acids in the heart, liver and skeletal muscle with lowered stearoyl-CoA desaturase-1 activity index; linoleic and α-linolenic acids increased accumulation of their C22 elongated products. These results demonstrate different physiological and biochemical responses to primary C18 unsaturated fatty acids in a rat model of human metabolic syndrome.  相似文献

Fatty acid composition and stability of vegetable oils have taken more attention as an essential source of biologically active compounds in a good balanced diet. The purpose of the study was to determine peroxide value, free fatty acids, unsaponifiable matter, total carotenoid content, iodine value and fatty acid composition of sunflower, rapeseed, mustard, peanut and olive oils. Rapeseed and peanut oils had the highest peroxide values, while sunflower oil had the lowest peroxide values. The free fatty acid value of the tested oils varied between 0.43 and 1.36% oleic. The peanut oil had the highest free acid value and the mustard oil had the lowest one. Total carotenoid contents of mustard and rape seed oil were higher than those of the other oils tested. Palmitic acid (C16:0), oleic acid (C18:1) and stearic acid (C18:0) were the common main fatty acid components of the vegetable oils tested. Followed by linoleic acid, the amount of oleic acid was the highest among other fatty acid components. Mustard oil had the highest erucic acid (C22:1) with the amount of 11.38%, indicating that it cannot be used for human consumption. Among the oils investigated, sunflower and mustard oils were more stable than rapeseed, peanut and olive oils.  相似文献

AIMS: To determine susceptibility of Clostridium perfringens strains CCM 4435(T) and CNCTC 5459 to C(2)-C(18) fatty acids, and evaluate influence of pH in cultures grown on glucose. Straw particles were added to cultures to simulate the presence of solid phase of the digestive tract milieu. METHODS AND RESULTS: Antimicrobial activity of fatty acids was expressed as a concentration at which only 50% of the initial glucose was utilized. Lauric acid showed the highest antimicrobial activity, followed by myristic, capric, oleic and caprylic acid. Only strain CNCTC 5459 was susceptible to linoleic acid. Neither caproic acid and acids with a shorter carbon chain nor palmitic and stearic acid influenced substrate utilization. The antimicrobial activity of myristic, oleic and linoleic acid decreased when clostridia were grown in the presence of straw particles. In cultures of both strains treated with capric and lauric acid at pH 5.0-5.3, the number of viable cells was <10(2) ml(-1). Only lauric acid reduced number of viable cells of both strains below 10(2) ml(-1) at pH > 6. Transmission electron microscopy revealed separation of inner and outer membranes and cytoplasma disorganization in cells treated with lauric acid. CONCLUSIONS: Lauric acid had the highest activity towards C. perfringens among fatty acid tested. Its activity was not influenced by the presence of solid particles and did not cease at pH > 6. SIGNIFICANCE AND IMPACT OF THE STUDY: Lauric acid might be a means for control of clostridial infections in farm animals.  相似文献

The fatty acid composition of trigylcerides from oranges, grapefruit, lemons and limes, was determined by GLC. Each species possessed its own intrinsic fatty acid pattern which might be used to differentiate it from the other species. The five major acids in all species were palmitic, palmitoleic, oleic, linoleic and linolenic. Collectively these acids comprised greater than 92% of the total acid content. Lemons were distinguished from all other species by their higher 16/16:1 ratios while grapefruit showed the highest total percentage of 16 and 16:1 acids. Lemons and limes contained higher percentages of branched-chain acids than oranges and grapefruit.  相似文献

The fatty acid composition of oil of the zero erucic acid commercial Brassica napus L. is typical for this species. It is rich in oleic acid and contains moderate levels of linoleic and linolenic acid. For human nutrition, it is advantageous primarily to obtain the highest possible content of oleic acid and to maintain the 2:1 ratio of linoleic to linolenic acid, while preserving the average total content of saturated acids. Uni- and multivariate analyses of variance were used for evaluation of doubled haploid lines of winter oilseed rape in respect of five fatty acids: palmitic (C16:0), stearic (C18:0) oleic (C18:1), linoleic (C18:2) and linolenic (C18:3). Some proposals of studying doubled haploid (DH) lines with the use of canonical transformation were also given. In MANOVA, the five original variables (individual fatty acids) were replaced by three 'new' variables (combinations of these acids) and used to evaluate DH lines with respect to the requirements concerning the nutritional role of fatty acids. The first variable was the total content of the saturated acids (C16:0 + C18:0), the second (unchanging) was the content of the monounsaturated acid C18:1, and the third was the difference between polyunsaturated acids, i.e. between linoleic acid, and the doubled content of linolenic acid (C18:2 - 2 x C18:3).  相似文献

The fatty acid composition of 64 strains from 15 species of dermatophyte fungi was studied by gas-liquid chromatography. The percentage amounts of palmitic, palmitoleic, stearic, oleic and linoleic acids were measured. These proved to be very similar for all species, with considerable intraspecific variation. As a group, Epidermophyton floccosum strains could be distinguished but individual strains could not be assigned reliably to any species or genus. No significant changes due to age or formaldehyde treatment were noted.  相似文献

Summary The cellular long-chain fatty acids of 69 strains of yeasts, representing 29 species associated with the brewing industry, were extracted by saponification and analyzed asmethyl esters by gas chromatography. The strains were characterized by the presence of palmitic, palmitoleic, stearic, oleic, linoleic and linolenic acid as the major fatty acids. The strains were divided into six groups on the basis of their fatty acid content. Rapeseed-mustard is one of the most economically important oilseed crops in India. Speciality oils having high amounts of a specific fatty acid are of immense importance for both nutritional and industrial purposes. Oil high in oleic acid has demand in commercial food-service applications due to a long shelf-life and cholesterol-reducing properties. Both linoleic and linolenic acids are essential fatty acids; however, less than 3% linolenic acid is preferred for oil stability. High erucic acid content is beneficial for the polymer industry, whereas low erucic acid is recommended for food purposes. Therefore, it is important to undertake systematic characterization of the available gene pool for its variable fatty acid profile to be utilized for specific purposes. In the present study the Indian rapeseed-mustard germplasm and some newly developed low-erucic-acid strains were analysed by GLC to study the fatty acid composition in these lines. The GLC analysis revealed that the rapeseed-mustard varieties being commonly grown in India are characterized by high erucic acid content (30-51%) in the oil with low levels of oleic acid (13-23%). However, from among the recently developed low-erucic-acid strains, several lines were identified with comparatively high oleic acid (60-70%), moderate to high linoleic acid (13-40%) and low linolenic acid (< 10%) contents. Work is in progress at TERI (New Delhi, India) to utilize these lines for development of strains with particular fatty acid compositions for specific purposes.  相似文献

Microorganisms with high activity of linoleic acid delta12-cis,delta11-trans-isomerase were isolated from the digestive tract of ruminants and characterized. The isolate with the highest isomerase activity was identified as Pseudobutyrivibrio ruminis. The susceptibility of this strain to 3 fatty acids added to the grow medium was determined. A significant inhibition of bacterial growth (during a 3-d period) by linoleic acid (0.1 %) and oleic acid (5 ppm) was observed; no inhibition was found in the presence of stearic acid.  相似文献

Fatty acid desaturases play vital roles in the synthesis of unsaturated fatty acids. In this study, Δ12 and Δ12/Δ15 fatty acid desaturases of the oleaginous yeast Lipomyces starkeyi, termed LsFad2 and LsFad3, respectively, were identified and characterized. Saccharomyces cerevisiae expressing LsFAD2 converted oleic acid (C18:1) to linoleic acid (C18:2), while a strain of LsFAD3-expressing S. cerevisiae converted oleic acid to linoleic acid, and linoleic acid to α-linolenic acid (C18:3), indicating that LsFad2 and LsFad3 were Δ12 and bifunctional Δ12/Δ15 fatty acid desaturases, respectively. The overexpression of LsFAD2 in L. starkeyi caused an accumulation of linoleic acid and a reduction in oleic acid levels. In contrast, overexpression of LsFAD3 induced the production of α-linolenic acid. Deletion of LsFAD2 and LsFAD3 induced the accumulation of oleic acid and linoleic acid, respectively. Our findings are significant for the commercial production of polyunsaturated fatty acids, such as ω-3 polyunsaturated fatty acids, in L. starkeyi.

1. Changes produced by a cholesterol-biosynthesis inhibitor, trans-1,4-bis-(2-chlorobenzylaminomethyl)cyclohexane dihydrochloride (AY-9944), in the total fatty acids in the liver and brain, and in phospholipids in the serum, liver, heart, brain and lungs from male rats, have been studied. 2. Treatment with AY-9944 produced the following changes in the fatty acid composition: (a) a marked decrease in the percentage of linoleic acid and an increase in oleic acid in the total fatty acids in the liver; (b) in the serum, an overall decrease in the percentage of linoleic acid in neutral lipids and phospholipids; (c) an increased content of linoleic acid in the beta-acyl chain of phosphatidylcholines in the liver and in sphingomyelins in the brain and lungs; (d) an increased content of palmitic acid and oleic acid in the beta-acyl chain of phosphatidylcholine in the liver, heart and lungs; (e) an increased content of phosphatidylcholines and sphingomyelins, together with an increased percentage of saturated fatty acids in these phosphatides in the lungs. 3. Changes in the phosphatides and the production of foam cells in the lungs suggest that AY-9944 may be of use in the study of the alveolar membrane.  相似文献

以采自11个种源的山桐子为材料,测定其果实脂肪酸的组成及其变异情况,结果表明:山桐子果实中不饱和脂肪酸含量较高,尤以亚油酸含量最高,11个种源的平均值为63.58%,且种源间差异显著,分宜、宜昌种源亚油酸相对含量明显高于其他9个种源;饱和脂肪酸以棕榈酸为主,11个种源山桐子果实棕榈酸差异显著,且以平武种源最高;其余脂肪酸含量均较低,变异幅度较大;种子中棕榈酸和棕榈烯酸含量明显高于果肉,而亚油酸、亚麻酸及硬脂酸含量明显低于果肉,油酸含量相近:果实不饱和脂肪酸含量依次为果肉〉全果〉种子,且变异系数及相对极差均较小,尤以果肉中最小;除硬脂酸外,山桐子果实中其他4种主要脂肪酸组分受海拔等地理环境的影响均较小。  相似文献

贵州地方芝麻种质资源品质性状的分析与评价   总被引:1,自引:0,他引:1  
为探究贵州芝麻种质资源的品质特征,并对地方芝麻资源进行初步鉴定与评价,本研究对73份贵州芝麻种质资源的8个品质性状进行测试分析。结果表明:(1)贵州芝麻种质资源含油量介于41.45%~52.12%之间,平均含量为49.69%。在脂肪酸组成中,油酸、亚油酸等不饱和脂肪酸的平均含量分别为35.65%和50.66%;而棕榈酸、硬脂酸等饱和脂肪酸的平均含量仅为8.40%和4.79%。此外,贵州芝麻资源中芝麻素、芝麻林素和木质素的平均含量分别为5.03 mg/g、2.63 mg/g和4.79 mg/g。8个品质性状的变异系数介于3.69%~32.62%范围内,其中芝麻素含量变异系数最大,含油量变异系数最小。而芝麻素含量、芝麻林素含量及硬脂酸含量的变异系数均大于10%,表明这3个性状在芝麻样本间存在较大差异。(2)相关性分析结果显示:含油量与油酸、芝麻素含量呈极显著正相关,与亚油酸含量呈极显著负相关;油酸含量与芝麻素含量呈极显著正相关,与亚油酸含量呈极显负相关;亚油酸含量与芝麻素含量呈极显著负相关。表明品质性状间相关性大、关联程度较高,性状间相互影响较大。(3)主成分分析将8个品质性状综合为3个主成分,分别为油酸因子、含油量因子和芝麻素因子,3个主成分因子包含了贵州芝麻种质资源品质性状的绝大部分信息,累计贡献率达96%以上。(4)在欧氏距离D=9.75处将73份贵州芝麻资源划分为6个类群:第Ⅰ类群包含2份资源、第Ⅱ类群有7份、第Ⅲ类群有12份、第Ⅳ类群有5份、第Ⅴ类群有16份、第Ⅵ类群有31份。其中第Ⅵ类群油酸含量最高,且含油量、芝麻素含量较高。本研究探明了贵州芝麻品质的特征特性,可为芝麻种质资源的利用和创新提供依据,为芝麻品种选育和遗传改良提供参考。  相似文献

The fatty acid constituents of mustard oil are palmitic, stearic, oleic, linoleic, linolenic and erucic acids. With the objective of mapping loci influencing the content of these fatty acids, a population of F6 generation recombinant inbred lines (RILs) derived from an inter-varietal cross of mustard was analyzed. Transgressive variation was evident for all the six fatty acids analysed irrespective of the levels of differences between the parents. The frequency distribution was normal for the linolenic acid, linoleic acid and stearic acid contents, while deviation from normality was observed for the other three fatty acids. The content of erucic acid was negatively correlated with the contents of all other fatty acids, which were positively correlated. Based on single marker analysis and interval mapping, two loci each for linoleic, linolenic and erucic acids were mapped to marker intervals on three linkage groups. Position of log of odds ratio (LOD) peaks suggested presence of common, linked and independently segregating loci for the fatty acid contents. The percentage of phenotypic variance explained by individual quantitative trait loci (QTLs) ranged from 10.5 to 19.5%, whereas the cumulative action of loci detected for different traits accounted for 16.3 to 27.6% of the variance. The additive effect for an individual locus ranged from 1.09 to 4.33. Presence of the favourable alleles at both the contributing loci in most of the RILs with a high linolenic acid content and of the unfavourable alleles in the lines with a low linolenic acid content indicated the possibility of pyramiding useful genes from phenotypically similar parental lines.  相似文献

Bacillus sphaericus species are mesophilic round-spored organisms that readily utilize fatty acid-based surfactants during growth, but their ability to modify fatty acids is unknown. Among 57 B. sphaericus-like strains tested for fatty acid transformation activity in Wallen fermentation (WF) medium, ten converted oleic acid to a new product determined by gas chromatography – mass spectrometry (GC-MS) to be 10-ketostearic acid (10-KSA). Additionally, a few other strains converted ricinoleic acid and linoleic acid to new products that remain to be characterized. Unlike most microbial hydrations of oleic acid, which produce a mixture of 10-KSA and 10-hydroxystearic acid, the conversion of oleic acid by B. sphaericus strains was unique in that 10-KSA was the sole reaction product. By replacing dextrose with sodium pyruvate in WF and adjusting to pH 6.5, conversion of oleic acid to 10-KSA by strain NRRL NRS-732 was improved from about 11% to more than 60%. Using the defined optimal conditions, the conversion reaction was scaled up in a stirred-batch reactor by using technical-grade oleic acid as substrate. 【目的】工蚁死亡后易受病原菌的侵染,进而危害蚁巢的健康。为了避免病菌横向传播,活工蚁根据尸体体内特定的化学物质的变化来识别尸体并将其搬运到弃尸堆。本实验旨在研究6种脂肪酸(油酸、亚油酸、棕榈油酸、棕榈酸、硬脂酸和肉豆蔻酸)对红火蚁Solenopsis invicta工蚁搬尸行为的影响,明确不同脂肪酸在其搬尸行为中发挥的作用。【方法】本实验利用GC-MS分析了工蚁活体和尸体提取物的脂肪酸成分,并在室内用滤纸片法测定了红火蚁对6种脂肪酸(10 μg/μL)、不同浓度(0.75和3 μg/μL)的单组分油酸或亚油酸以及不同浓度(0.01, 0.1, 1和10 μg/μL)的这两种酸的混合液的反应。【结果】结果表明,红火蚁尸体仅含有亚油酸和油酸两种脂肪酸;另外4种脂肪酸(棕榈油酸、棕榈酸、硬脂酸和肉豆蔻酸)对红火蚁的搬尸行为无显著影响;单组分的油酸和亚油酸均能促进红火蚁的搬尸行为,浓度越高,搬尸行为越显著。10 μg/μL的油酸和亚油酸混合液对工蚁的搬尸行为也具有显著的促进效果,低浓度的混合液对其无明显的促进作用(P>0.05)。【结论】结果提示油酸和亚油酸可调控红火蚁的搬尸行为,其他脂肪酸对红火蚁搬尸行为不产生影响。  相似文献
