Based on the theories of tissue optics and artificial neural network, the relationship between the optical properties and biological parameters was studied, and a new experimental method was derived. The properties of the organism were obtained indirectly by a black-box model derived by self-study of the artificial neural network between optical parameters and thermo-physical properties without using the heat transfer equation. In this method, the energy of light in diffuse radiation, diffuse transmission and collimated transmission was absorbed by a dual-integrating sphere experimental system of a spectrometer, and the spectrogram of the energy was obtained. Combining these spectral data of the energy, the diffuse-reflecting power, the diffuse transmissivity and the collimated transmissivity were calculated. The calculated results were taken as the input parameters of a black-box model. The experimental results show that there are apparent differences between the spectrogram of the energy on the diffuse radiation, the diffuse transmission and the collimated transmission of different matters, while there is a little difference in the same matter. Each spectrogram has its own characteristic. The values of the four thermal properties including the density, the constant pressure specific heat, the thermal diffusivity and the viscosity were calculated using the black-box model. Compared with the real values the calculated one has an average relative error between −5% and 5%. The conductivity of the tongue is 0.68 W/(m K) that calculated from the value of the density, the constant pressure specific heat and the thermal diffusivity. The results also show that there is a little difference on the conductivities in the longitudinal cross-section and the transverse section, but the effect of temperature on the conductivity of the tongue is not apparent. The difference implies the anisotropy of the properties of the organism, which cannot be easily obtained by a conventional experimental method.  相似文献

The thermal limitations inherent with the use of invasive thermistor probes in the measurement of thermal properties of biomaterials have been investigated. An electronic temperature controller has been developed which provides a nearly instantaneous step rise in average probe resistance (temperature). The method of experimentally determining the heat rate required to maintain the average probe temperature constant and incorporation of that heat rate into the general heat diffusion equation provides a solution which allows the determination of both thermal conductivity and diffusivity values with improved accuracy. The method is general to all media which wet the surface of the probe; the need for calibrating media is avoided. The solution also predicts the minimum required sample size.  相似文献

A method for measuring hydraulic conductivity and embolism in xylem   总被引:28,自引:3,他引:28  
Abstract Hydraulic conductivity of the xylem is computed as the quotient of mass flow rate and pressure gradient. Measurements on excised plant stems can be difficult to interpret because of time-dependent reductions in flow rate, and because of variable degrees of embolism. Using Acer saccharum Marsh. stems, we found that certain perfusing solutions including dilute fixatives (e.g. 0.05% formaldehyde) and acids with pH below 3 (e.g. 10 mol m?3 oxalic) prevent long-term decline in conductivity. Xylem embolism can be quantified by expressing the initial conductivity as a percentage of the maximum obtained after flow-impeding air emboli have been removed by repeated high-pressure (175 kPa) flushes. Correlation between microbial contamination and declining conductivity suggests that long-term (> 4h) declines are caused by microbial growth within the vessels. Unpredictable trends in short-term (< 4h) measurements may be caused by movements of air emboli in vessels and/or participate matter.  相似文献

We tested the accuracy, sensitivity, and reproducibility of a new lung water computer, based on the thermal conductivity technique, in 22 anesthetized closed-chest ventilated sheep with different treatments: 1) controls (n = 8), 2) 0.05 ml/kg of oleic acid + 100 ml/kg of lactated Ringer solution (n = 6), and 3) airway instillation of saline [3.1 +/- 1.3 (SD) g/kg, n = 8]. After 4 h, we determined the extravascular lung water gravimetrically. We found a significant overall correlation between the final extravascular lung thermal volume and the gravimetric extravascular lung mass (P < 0.001). Although the average ratio of extravascular lung thermal volume to extravascular lung mass was 0.97 +/- 0.25 ml/g for all groups, the computer overestimated extravascular lung mass in controls by 10% (17 g) and underestimated it in sheep with oleic acid by 15% (95 g) and in sheep with airway instillation by 8% (37 g). The computer also underestimated the small quantities of saline placed via the airway in the alveolar space by 75% (61 g). Reproducibility of three consecutive measurements was 4.3% (SE). We conclude that the thermal conductivity technique has an ability to detect the baseline extravascular lung mass but has a poor ability to detect an accurate increment of the extravascular lung water under poor tissue perfusion in anesthetized ventilated sheep.  相似文献

A facile radiometric technique for measuring ATP in samples of biologic origin is presented. The d-glucose-6-phosphate produced by the phosphorylation of excess tritiated d-glucose with crystalline hexokinase is stoichiometrically equivalent to the ATP present. Product is selectively precipitated with ethanolic barium acetate, washed with ethanol and the tritium label counted in a scintillation spectrometer. The technique is especially suitable for the measurement of ATP in large numbers of samples (50–100) and offers acceptable sensitivity down to 62 fmoles.  相似文献

1. HeLa cells were cultured in the presence of heterologous immunoglobulin G and guinea-pig serum together with [(32)P]phosphate. 2. Incorporation of [(32)P]phosphate was significantly stimulated by anti-HeLa immunoglobulin G and complement-sufficient serum compared with immunoglobulin G from unimmunized rabbits and complement. Within 2.5h heat-inactivated guinea-pig serum and anti-HeLa immunoglobulin G stimulated [(32)P]phosphate incorporation to the same extent as heat-inactivated complement and immunoglobulin G from unimmunized rabbits. 3. Compared with cells exposed to immunoglobulin G from unimmunized rabbits together with complement, anti-HeLa immunoglobulin G with complement increased the phospholipid content of HeLa cells twofold within 5h of incubation. 4. Exposure of HeLa cells to anti-HeLa immunoglobulin G and complement for 5-22h resulted in a twofold increase in the net accumulation of [(32)P]phosphate in sphingomyelin and phosphatidylcholine and a 50% increase in the net accumulation of [(32)P]phosphate in phosphatidylethanolamine, compared with cultures exposed to immunoglobulin G from unimmunized rabbits and complement. 5. A transient accumulation of (32)P-labelled lysophosphoglycerides in HeLa cells exposed to antibody and complement was detected, confirming previous findings (Güttler & Clausen, 1969b). 6. The stimulation of [(32)P]phosphate turnover occurred in cells filling up their cytoplasma with vacuoles. This supports the suggestion that the accumulation of phospholipid in these cells may be concerned with the synthesis and function of cytomembranes.  相似文献

[2-3H]Inosinic acid ([2-3H]IMP) has been biosynthesized in good yield from [2-3H]hypoxanthine and PRPP via the action of a partially purified preparation of hypoxanthine/guanine phosphoribosyl transferase from mouse brain. The product was purified in one step by ascending paper chromatography, and used to assess the activity of IMP dehydrogenase. To conduct the assay, tritiated substrate is admixed with enzyme in a final volume of 10 microliters; NAD is present to serve as cofactor for the reaction, and allopurinol to inhibit the oxidation of any hypoxanthine generated as a consequence of side reactions. After an appropriate period of incubation, the 3H2O arising from the oxidation of tritiated IMP via [3H]NAD is isolated by quantitative microdistillation. Performed as described, the assay is facile, sensitive, and accurate, with the capability of detecting the dehydrogenation of as little as 1 pmol of [3H]IMP. Using it, measurements have been made of IMP dehydrogenase in a comprehensive array of mouse organs. Of these, pancreas contained the enzyme at the highest specific activity.  相似文献

Water encounters various resistances in moving along a path of decreasing potential energy from the soil through the plant to the atmosphere. The reported relative magnitudes of these pathway resistances vary widely and often these results are conflicting. One reason for such inconsistency is the difficulty in measuring the potential drop across various segments of the soil-plant-atmosphere continuum. Summary The technique described provides a rapid method for screening thermoplastic polyurethanes against deteriogenic micro-organisms using thin films (0.4–0.5 mm thick) of plastic prepared in an electric plantens press. The films are inoculated with a spore suspension of Gliocladium roseum and can be viewed directly under the light microscope for evaluation of surface effects; selective staining can be used to reveal fungal mycelium. Results can be obtained within a week which correlate with longer term tests using commercial samples. The technique can also be used to isolate potential polyurethane deteriorating micro-organisms from the environment and to confirm their biodeteriogenic activities.  相似文献

An automated 36 place valve to provide continuous soil respiration measurements was constructed. The valve is fully computer controlled and can sample and purge the soil atmosphere as frequently as every 75 minutes. The concentrations, automatically measured by the valve, are essentially identical to those measured manually by gas chromatography in the concentration range of 0.1 to 1% CO2, and are kept in this range by adjusting the mass of soil and the sampling frequency. Data are transferred automatically to a computer spreadsheet program for data handling and plotting on either a rate or cumulative basis. The system has proved reliable over many thousands of analyses and has made detailed analysis of microbial activity on a continuous basis possible.  相似文献

We describe a method for measuring nociception in cattle using a CO(2) laser aimed at the caudal aspect of the metatarsi. In Experiment 1, infrared thermography showed that calves responded by lifting their legs when skin temperatures reached 45-55 degrees C. In Experiment 2a, the validity of the method was tested by comparing the response latencies of 14 calves to two power settings (2.25 W vs. 4.5 W) with each setting being applied six times. We found that both leg-lift latencies and tail-flick latencies were lower at the higher power setting, and the calves were more likely to respond by kicking than by simply moving the leg. The standard deviations between and within calves were smaller at the higher power setting, and the large within-calf variation means that at least three tests were required to obtain reliable measures that could differentiate between calves. In Experiment 2b, application of the laser at a range of power settings (2.0, 3.0, 4.0, 4.5, 5.0 and 5.5 W) on 16 calves showed that response latencies decreased as power increased up to 4.5 W, after which no further change occurred. In Experiment 3, the repeatability of the method was evaluated on nine measures with the high power setting (4.5 W). The coefficient of variation associated with repetition of the measures was 36%. In general, we found little change in response latencies with repeated use of the laser, except that responses on the second test tended to be shorter. Experiment 4 showed that ambient temperatures between 16 degrees C and 27 degrees C did not affect response latencies, but these were longer at temperatures of 7 degrees C. We suggest that the method is a useful way of measuring cattle's sensitivity to nociception as the animals need not be restrained and the distance to the animal need not be closely controlled. However, to obtain accurate, valid and reliable measures it is necessary to use a high power setting (4.5 W) and take at least three consecutive measures of the response latency.  相似文献

2. 2. The device measures temperature differences across the sample and across a polyethylene plate with known conductivity which is placed in series with the sample.

3. 3. The conductivity of the polyethylene was determined from the steady state temperature difference and heat flux through the wall of a polyethylene pipe with a central heat source.

5. 5. The thermal conductivity of harp seal (Phoca groenlandica) and minke whale (Balaenoptera acutorostrata) blubber, as determined by use of this device, is very close to previously reported values.
