New direct syntheses of [Pt(trpy)(NCCH3)](CF3SO3)22 (where trpy = 2,2′:6′,2′′-terpyridine) and [Pt(tBu3-trpy)(NCCH3)](CF3SO3)23 (where tBu3-trpy = 4,4′,4″-tri-tert-butyl-2,2′:6′,2″-terpyridine) via the displacement of acetonitrile from [Pt(NCCH3)4](CF3SO3)2 have been developed. The synthetic utility of 2 was investigated in reactions with triphenylphosphine (PPh3), 2,6-dimethylphenyl isocyanide (CN-Xyl), 2,5-dimethyl-2,5-diisocyanohexane (TM4), and tert-butyl isocyanide (CN-tBu). Whereas the expected substitution products were observed for reactions with PPh3, CN-Xyl, and CN-tBu, dealkylation of TM4 occurred to afford [Pt(trpy)(CN)](CF3SO3) 6. The structures of [Pt(trpy)L]2+ dications show little intermolecular interactions in the solid state, with the exception of the tBu3-trpy complex 3 which exists as head-to-tail dimers with a Pt-Pt distance of 3.29 Å. The cyano product 6 was found to stack in infinite chains of cations with a Pt-Pt distance of 3.45 Å.  相似文献

Bifunctional polynuclear platinum compounds represent a novel class of metal-based antitumor drugs which are currently undergoing preclinical development. A typical agent is [(trans-PtCl(NH(3))(2))(2)H(2)N(CH(2))(4)NH(2)]Cl(2) (1,1/t,t), which coordinates to bases in DNA and forms various types of covalent crosslinks. It also forms a 1,2-d(GpG) intrastrand adduct, the equivalent of the major DNA lesion of 'classical' cisplatin. In the present study differential scanning calorimetry and spectroscopic techniques were employed to characterize the influence of this crosslink on the thermal stability and energetics of 20 bp DNA duplexes site-specifically modified by 1,1/t,t. Thermal denaturation data revealed that the crosslink of 1,1/t,t reduced thermal and thermodynamical stability of the duplex noticeably more than that of 'classical' cisplatin. The energetic consequences of the intrastrand crosslink at the d(GG) site are discussed in relation to the structural distortions induced by this adduct in DNA and to its recognition and binding by HMG domain proteins. It has been suggested that the results of the present work are consistent with different DNA binding modes of cisplatin and polynuclear bifunctional DNA-binding drugs, which might be relevant to their distinct biological effectiveness.  相似文献

We show that UVA irradiation (365 nm) of the PtIV complex trans,trans,trans-[PtIVCl2(OH)2(dimethylamine)(isopropylamine)] (1), induces reduction to PtII photoproducts. For the mixed amine PtII complex, trans-[PtIICl2(isopropylamine)(methylamine)] (2), irradiation at 365 nm increases the rate and extent of hydrolysis, triggering the formation of diaqua species. Additionally, irradiation increases the extent of reaction of complex 2 with guanosine-5′-monophosphate and affords mainly the bis-adduct, while reactions with adenosine-5′-monophosphate and cytidine-5′-monophosphate give rise only to mono-nucleotide adducts. Density Functional Theory calculations have been used to obtain insights into the electronic structure of complexes 1 and 2, and their photophysical and photochemical properties. UVA-irradiation can contribute to enhanced cytotoxic effects of diamine platinum drugs with trans geometry.  相似文献

The reaction of Pt(COD)Cl2, where COD is 1,5-cyclooctadiene, with one equivalent of a diamidato-bis(phosphino) Trost ligand ((R,R)-2 = N,N′-bis(2-diphenylphosphino-1-benzoyl)-(1R,2R)-1,2-diaminocyclohexane, (R,R)-N,N′-bis(2-diphenylphosphino-1-naphthoyl)-(1R,2R)-1,2-diaminocyclohexane, or (±)-N,N′-bis(2-diphenylphosphino-1-benzoyl)-1,2-bis(aminobenzene)) in the presence of base afforded square planar diamidato-bis(phosphino) platinum(II) complexes (R,R)-2-Pt, (R,R)-3-Pt, (±)-4-Pt. Characterization of all complexes included the solution and solid state structure determination of each complex based on multinuclear NMR and X-ray analyses, respectively. Stability of the complexes in acid was examined on addition of HCl to (R,R)-2-Pt in chloroform and compared to the unreactive nature of the similar diamidato-bis(phosphino) complex 1-Pt (= 1,2-bis-N-[2′-(diphenylphosphino)benzoyl]diamino-benzene) in the presence of acid. Protonation of the bound amidato nitrogen atoms of (R,R)-2-Pt was observed along with decoordination of the nitrogen atoms from the platinum(II) center producing (R,R)-2-PtCl2 in quantitative yield by NMR analysis. Confirmation of the product was made on comparison of the NMR spectra to that of authentic (R,R)-2-PtCl2 prepared on reaction of Pt(COD)Cl2 with (R,R)-2 in CH2Cl2 and characterized by single-crystal X-ray diffraction analysis and NMR spectroscopy. Results add to the knowledge of rich coordination chemistry of bis(phosphino) ligands with late transition metals, metal-amidato chemistry, and has implications in catalysis.  相似文献

The reaction of dimeric precursor [Ir(CO)2Cl]2 with two molar equivalent of the pyridine-ester ligands (L) like methyl picolinate (a), ethyl picolinate (b), methyl nicotinate (c), ethyl nicotinate (d), methyl isonicotinate (e) and ethyl isonicotinate (f) affords the tetra coordinated neutral complexes of the type [Ir(CO)2ClL] (1a-f). The single crystal X-ray structure of 1d reveals that the Ir atom occupies the centre of an approximately square planar geometry with two CO groups cis- to each other. Intermolecular C-H?O and Ir?C interactions greatly stabilize the supramolecular structure of 1d in the solid state. The oxidative addition (OA) reactions of 1a-f with different electrophiles such as CH3I, C2H5I and I2 undergo decarbonylation of one CO group to generate the oxidized products of the type [Ir(CO)RClIL] where R = -CH3 (2a-f); -C2H5 (3a-f) and [Ir(CO)ClI2L] (4a-f). Kinetic study of the reaction of 1c-f with CH3I indicates a first order reaction which follow the order 1d > 1c > 1f > 1e. All the synthesized complexes were characterized by elemental analyses, IR, and multinuclear NMR spectroscopy.  相似文献

Bis(platinum) complexes [[cis-PtCl2(NH3)]2H2N(CH2)nNH2] are a novel series of potential anticancer agents in which two cis-diamine(platinum) groups are linked by an alkyldiamine of variable length. These complexes are potentially tetrafunctional, a unique feature in comparison with known anticancer agents. Studies of DNA interactions of bis(platinum) complexes in comparison with cisplatin demonstrate significant differences. Investigations of interstrand crosslink formation in which crosslinking of a short DNA fragment is detected by gel electrophoresis under denaturing conditions demonstrate that interstrand crosslinks are 250 fold more frequent among bis(platinum) adducts than among cisplatin-derived adducts under the conditions examined. These investigations indicate that bis(platinum) adducts contain a high frequency of structurally novel interstrand crosslinks formed through binding of the two platinum centers to opposite DNA strands. Unlike cisplatin, bis(platinum) complex binding does not unwind supercoiled DNA. Studies with the E. coli UvrABC nuclease complex demonstrate that both linear and supercoiled DNA containing bis(platinum) adducts are subject to incision by the repair enzyme complex. Initial studies using UvrABC nuclease as a probe to define the base and sequence specificity for bis(platinum) complex binding suggest that the specificity of the bis(platinum)s is similar, but not identical, to that of cisplatin.  相似文献

The distorted square-planar complexes [Pd(PNHP)Cl]Cl (1) (PNHP = bis[2-(diphenylphosphino)ethyl]amine), [M(P3)Cl]Cl [P3 = bis[2-(diphenylphosphino)ethyl]phenylphosphine; M = Pd (2), Pt (3)] and [Pt(NP3)Cl]Cl (5) (NP3 = tris[2-(diphenylphosphino)ethyl]amine), coexisting in the later case with a square-pyramidal arrangement, react with one equivalent of CuCl to give the mononuclear heteroionic systems [M(L)Cl](CuCl2) [L = PNHP, M = Pd (1a); L = P3, M = Pd (2a), Pt (3a); L = NP3, M = Pt (5a)]. The crystal structure of 3a confirms that Pt(II) retains the distorted square-planar geometry of 3 in the cation with P3 acting as tridentate chelating ligand, the central P atom being trans to one chloride. The counter anion is a nearly linear dichlorocuprate(I) ion. However, the five-coordinate complexes [Pd(NP3)Cl]Cl (4), [M(PP3)Cl]Cl (M = Pd (6), Pt (7); PP3 = tris[2-(diphenylphosphino)ethyl] phosphine) containing three fused five-membered chelate rings undergo a ring-opening by interaction with one (4, 6, 7) and two (6, 7) equivalents of CuCl with formation of neutral MCu(L)Cl3 [L = NP3, M = Pd (4a); L = PP3, M = Pd (6a), Pt (7a)] and ionic [MCu(PP3)Cl2](CuCl2) [M = Pd (6b), Pt (7b)] compounds, respectively. The heteronuclear systems were shown by 31P NMR to have structures where the phosphines are acting as tridentate chelating ligands to M(II) and monodentate bridging to Cu(I). Further additions of CuCl to the neutral species 6a and 7a in a 1:1 ratio resulted in the achievement of the ionic complexes 6b and 7b with ions as counter anions. It was demonstrated that the formation of heterobimetallic or just mononuclear mixed salt complexes was clearly influenced by the polyphosphine arrangement with the tripodal ligands giving the former compounds. However, complexes [M(NP3)Cl]Cl constitute one exception and the type of reaction undergone versus CuCl is a function of the d8 metal centre.  相似文献

The reaction between the antitumor octahedral complex trans-RuCl2(DMSO)4 and d(GpG) leads to the formation of a stable compound characterized by a covalent bifunctional coordination of the bases to the metal center. The structure of the compound has been fully characterized by NMR and molecular modeling studies, showing the presence of two N7-coordinated guanine moieties in a head to head conformation, two dimethyl sulfoxide molecules, and one halogen atom in the coordination sphere of the ruthenium. The glycosidic chi angles are essentially in the anti range, the sugar puckering of the 5'G is 3'-endo (100% N), whereas that of the 3'G is more flexible but mainly in 2'-endo conformation (85% S), the two bases are strongly destacked. The compound shows structural features which are surprisingly similar to those exhibited by the corresponding cisplatin complex, indicating that such a way of interaction with DNA is not exclusive to Pt or to metals with square planar coordination geometries.  相似文献

The reaction of the antitumor active agent cis-[Pt(NH3)2(4-mepy)Cl]Cl (4-mepy stands for 4-methylpyridine) with d(GpG) has been investigated by 1H magnetic resonance spectroscopy. Initially, two mononuclear complexes cis-Pt(NH3)2(4-mepy)[d(GpG)-N7(1)] 1 and cis-Pt(NH3)2(4-mepy)[d(GpG)-N7(2)] 2 are formed in an unexpected ratio 65:35, as determined by 1H NMR and enzymatic digestion techniques. Both products react further with a second equivalent of cis-[Pt(NH3)2(4-mepy)Cl]Cl forming the dinuclear platinum complex [cis-Pt(NH3)2(4-mepy)]2[mu-d(GpG)- N7(1),N7(2)] 3. With [Pt(dien)Cl]Cl and [Pt(NH3)3Cl]Cl similar complexes are formed. No evidence was found for the formation of chelates cis-Pt(NH3)(4-mepy) [d(GpG)-N7(1),N7(2)], which would be formed upon ammonia release from the mononuclear complexes 1 and 2. Even addition of strong nucleophiles, like sodium diethyldithiocarbamate, thiourea, cysteine, or methionine, before or after reaction, do not induce the formation of a chelate. Under all conditions the N-donor ligands remain coordinated to Pt in 1,2 and 3. In addition, the results of bacterial survival and mutagenesis experiments with E. coli strains show that the in vivo formation of bifunctional adducts in DNA, comparable to those induced by cis-Pt(NH3)2Cl2, by treatment of cells with cis-[Pt(NH3)2(4-mepy)Cl]Cl is unlikely. Also, a mechanism of binding and intercalation is not supported by experimental data. All experiments suggest that the mechanism of action of this new class of antitumor agents must be different from that of cis-Pt(NH3)2Cl2.  相似文献

The preparation and molecular structure of [(diethylenetriamine) (7,9-dimethylhypoxanthine) platinum(II)] (PF6)2·1.5H2O and [(ethylenediamine) (7,9-dimethylhypoxanthine)2platinum(II)] (PF6)2, are reported. These complexes represent the first structurally characterized N(1)-bound Pt(II) 6-oxopurine complexes. In each case, the Pt(II)N(1) bond length [2.051(6)A in the diethylenetriamine complex and 2.021(8)A in the ethylenediamine complex] indicates a strong metal-to-base binding. Both complexes contain interligand hydrogen bonds, with the ammine ligand acting as the donor and the O(6) atom of the base acting as the acceptor. These N(1)-bound complexes are compared with N(7)-bound 6-oxopurine and N(3)-bound cytosine complexes of Pt(II) anti-tumor agents.  相似文献

A conformational study of the double-stranded decanucleotide d(GCCG*G*ATCGC).d(GCGATCCGGC), with the G* guanines chelating a cis-Pt(NH3)2 moiety, has been accomplished using 1H and 31P NMR, and molecular mechanics. Correlation of the NMR data with molecular models has disclosed an equilibrium between several kinked conformations and has ruled out an unkinked structure. The deformation is localized at the CG*G*.CCG trinucleotide where the helix is kinked by approximately 60 degrees towards the major groove and unwound by 12-19 degrees. The models revealed an unexpected mobility of the cytosine complementary to the 5'-G*. This cytosine can stack on either branch of the kinked complementary strand. The energy barrier between the two positions has been calculated to be less than or equal to 12 kJ/mol. The NMR data are in support of rapid flip-flopping of this cytosine. An explanation for the strong downfield shift observed in the 31P resonance of the G*pG* phosphate is given.  相似文献

HMG-domain proteins bind strongly to bent DNA structures, including cruciform and cisplatin-modified duplexes. Such protein-platinated DNA complexes, formed where the DNA is modified by the active cis but not the inactive trans isomer of diamminedichloroplatinum(II), are implicated in the cytotoxic mechanism of the drug. A series of oligonucleotide duplexes with deoxyguanosine nucleosides flanking a cis-[Pt(NH(3))(2)?d(GpG)-N7(1),-N7(2)?] cross-link have been synthesized. These probes were used to determine the flanking sequence dependence of the affinity of the individual HMG domains of HMG1 toward cisplatin-modified DNA. Nine related sequences, where N(1) and N(2) are not dG and GG is the 1,2-intrastrand cisplatin adduct in N(1)GGN(2), were previously investigated [Dunham, S. U., and Lippard, S. J. (1997) Biochemistry 36, 11428-11436]. Three of the seven remaining possible sequences for which N(1) and/or N(2) was dG were prepared here by using normal deoxyguanosine, but the rest, where N(1) is dG and N(2) is dA, dC, T, or dG, could not be isolated in pure form. These sequences were accessed by using the synthetic bases 7-deazaadenine and 7-deazaguanine, which lack the nucleophilic N7 atom in the purine ring. Deaza nucleotides accurately mimic the properties of the natural bases, allowing the interaction of the HMG-domain proteins with cisplatin-modified DNA to be examined. These experiments reveal that the flexibility of A.T versus G.C flanking base pairs, rather than base-specific contacts, determines HMG1domA protein selectivity. This conclusion was supported by use of mutant HMG1domA and HMG1domB proteins, which exhibit identical flanking sequence selectivity. The methods and results obtained here not only improve our understanding of how proteins might mediate cisplatin genotoxicity but also should apply more generally in the investigation of how other proteins interact with damaged DNA.  相似文献

From the reaction between dihydroxoplatinum(II) and l-ascorbic acid, two types of platinum(II) ascorbate complexes were obtained and structurally characterized with ethylenediamine (en), N,N-dimethylethylenediamine (dmen) and N,N,N′-trimethylethylenediamine (trimen) as stabilizing ligands. In [Pt(en)(asc-C,O)] (1), [Pt(dmen)(asc-C,O)] (2) and [Pt(trimen)(asc-C,O)] (4), the ascorbate dianion forms a five-membered chelate ring, coordinating to the Pt(II) ion at the 2-carbon and the 5-oxygen atoms (C,O-chelate). From the same mother solution, crystals of [Pt(trimen)(asc-O,O′)] (3) were obtained during the precipitation of 4; in 3 the ascorbate is bound to the Pt at the 2- and 3-oxygen atoms (O,O′-chelate). Compounds 3 and 4 are the first well-characterized linkage isomers among the transition-metal ascorbate complexes. The O,O′-chelated 3 slowly changes to the C,O-chelated 4 in an aqueous solution. Bulkiness of the stabilizing ligand, i.e. en, dmen and trimen has an influence on the formation of the C,O-chelated species, 1, 2 and 4.  相似文献

The binding of platinum (II)-terpyridine complexes to DNA was studied by using equilibrium dialysis. Optical absorption methods were used to measure the ability of the ligands to aggregate in aqueous buffer. Scatchard plots for the binding of the monomeric [Pt(terpy)SC4H9]+ cation to DNA at I0.01 are curvilinear, concave upwards, suggesting two modes of binding. The association constant decreases at higher ionic strengths, consistent with polyelectrolyte theory, and 1.1 cations are released per bound ligand molecule. The association constants of the binuclear ligands [Pt(terpy)S[CH2]4S(terpy)Pt]2+ and [Pt(terpy)S[CH2]6S(terpy)Pt]2+ are 8 and 23 times larger respectively than the affinity of the monomer. For the latter binuclear derivative the increase may be ascribed to bifunctional reaction. Differential dialysis experiments with DNAs of differing base composition show that [Pt(terpy)SC4H9]+ has a requirement for a single G X C base-pair at the highest-affinity site. However, in the binuclear ligands chromophore specificity is severely compromised. Similar experiments indicate that 9-aminoacridine and selected methylene-linked diacridines show no significant sequence selectivity.  相似文献

Reaction of either cis or trans Pt(NH3)2Cl2 with poly(A) in dilute aqueous solution leads to quantitative precipitation of the polymer at Pt/nucleotide ratios above 0.5. It is proposed that at ratios less than this, intramolecular binding of one Pt to two bases is favored; at higher ratios, intermolecular cross-linking becomes important and precipitation results. The absence of isomer selectivity in precipitation implies that the biological specificity of the cis form results from a process other than cross-linking of polynucleotide strands. Other observations suggest that the coordinated ammonia of nucleotide-platinum(II) ammine complexes may be unusually labile.  相似文献

The reaction of H2[PtCl6] · 6H2O and (H3O)[PtCl5(H2O)] · 2(18C6) · 6H2O (18C6 = 18-crown-6) with 9-methylguanine (MeGua) proceeded with the protonation of MeGua forming 9-methylguaninium hexachloroplatinate(IV) dihydrate (MeGuaH)2[PtCl6] · 2H2O (1).The same compound was obtained from the reaction of Na2[PtCl6] with (MeGuaH)Cl.On the other hand, the reaction of guanosine (Guo) with (H3O)[PtCl5(H2O)] · 2(18C6) · 6H2O in methanol at 60 °C proceeded with the cleavage of the glycosidic linkage and with ligand substitution to give a guaninium complex of platinum(IV), [PtCl5(GuaH)] · 1.5(18C6) · H2O (2).Within several weeks in aqueous solution a slow reduction took place yielding the analogous guaninium platinum(II) complex, [PtCl3(GuaH)] · (18C6) · 2Me2CO (3).H2[PtCl6] · 6H2O and guanosine was found to react in water, yielding (GuoH)2[PtCl6] (4) and in ethanol at 50 °C, yielding [PtCl5(GuoH)] · 3H2O (5).Dissolution of complexes 2 and 5 in DMSO resulted in the substitution of the guaninium and guanosinium ligands, respectively, by DMSO forming [PtCl5(DMSO)].Reactions of 1-methylcytosine (MeCyt) and cytidine (Cyd) with H2[PtCl6] · 6H2O and(H3O)[PtCl5(H2O)] · 2(18C6) · 6H2O resulted in the formation of hexachloroplatinates with N3 protonated pyrimidine bases as cation (MeCytH)2[PtCl6] · 2H2O (6) and (CydH)2[PtCl6] (7), respectively. Identities of all complexes were confirmed by 1H, 13C and 195Pt NMR spectroscopic investigations, revealing coordination of GuoH+ in complex 5 through N7 whereas GuaH+ in complex 3 may be coordinated through N7 or through N9. Solid state structure of hexachloroplatinate 1 exhibited base pairing of the cations yielding (MeGuaH+)2, whereas in complex 6 non-base-paired MeCytH+ cations were found. In both complexes, a network of hydrogen bonds including the water molecules was found. X-ray diffraction analysis of complex 3 exhibited a guaninium ligand that is coordinated through N9 to platinum and protonated at N1, N3 and N7. In the crystal, these NH groups form hydrogen bonds N–HO to oxygen atoms of crown ether molecules.  相似文献

A series of di- and monosubstituted cis-platinum(II) silanolate complexes, Pt(OSiR3)2(dppe) (R=Et, 1; R=Me, 2) and Pt(OSiR3)Cl(dppe) (R=Et, 3; R=iPr, 4) where dppe is 1,2-bis(diphenylphosphino)ethane, have been isolated and characterised spectroscopically. Complex 1 does not react with CO and H2 under anhydrous conditions, but the complexes Pt{C(O)OCH3)}2(dppe) (6) and Pt(CO3)(dppe) (7) have been isolated bubbling CO in methanol and CO2 in moist benzene solutions of 1, respectively. The behaviour of 1 towards water or methanol is discussed on the basis of 1H, and 31P{1H} NMR spectroscopic data. The new complex Pt{S2C(OSiEt3)2}(dppe) (8) has been isolated by reaction of 1 with CS2 in benzene solution. This reactivity would suggest a high sensitivity towards water, but not towards H2 or CO, of the bonding of slightly oxidised platinum particles with silanol groups of silica surface.  相似文献

Both cisplatin and the estrogen receptor (ER) are known to bend DNA. The influence of the bending of sequences by the d(GpG)cisPt adduct binding of ER to estrogen response element (ERE)-like sequences was examined. Three ERE-like oligonucleotides with different affinities for ER and which include a GG in the linker sequence were designed in order to form a single central d(GpG)cisPt adduct. Using electrophoretic mobility shift assay and Scatchard analysis, it was shown that the presence of a single d(GpG)cisPt adduct in the linker sequence decreases the ER affinity for DNA. These results do not support a critical role of a DNA bend in the initial recognition of ERE by ER. Then, the platination of DNA outside of the ERE half-sites decreases the interaction of ER with ERE.  相似文献

Reactions of cis-diaminediaqua palladium and platinum dinitrates and of trans-diaminediaqua platinum dinitrate give complexes of the type Pd(tmeda)(OH)(C4O4)Pd(tmeda)(C4O4H) (tmeda = tetramethylethylenediamine) (1), (en)M(C4O4)2M(en) (en = ethylenediamine (M = Pd, Pt) and trans-[Pt- (NH3)2C4O4]n, respectively.
