The densities of amorphous maltose-water, maltose-glycerol and starch-sorbitol-water mixtures were measured using a vibrating-tube density meter and pycnometry. The volumetric change on mixing was investigated through the calculation of the quantity DeltaV/V, the difference between (experimental) volume of the mixture and the linear composition weighted pure constituent volumes (ideal mixing). For all of the systems studied the quantity DeltaV/V was negative and approached a minimum of -0.03 and -0.015 at mass fractions of maltose in the region of 0.75 and 0.85 for the maltose-water and maltose-glycerol mixtures, respectively. Results are discussed in the context of volume change due to structural relaxation of vitreous materials and related to the phenomenon of antiplasticisation.  相似文献

Data at 298.2 K for aqueous NaCl solutions containing either D-phenylglycin, D-(p-hydroxy)phenylglycine, 6-amino penicillanic acid (6-APA), amoxicillin or ampicillin, are reported. The mean ionic activity coefficients of NaCl were determined from measurements of the responses of a sodium and a chloride ion-selective electrode. The activity coefficients of the precursors or the antibiotics were calculated from the values of the mean ionic activity coefficients using the exact cross differential relation between them. The correlation of solubility data using the activity coefficients measured in this work shows the same puzzling results previously observed in systems containing amino acids.  相似文献

As a contribution to the understanding of the thermodynamic state of single salts in living systems, the activity coefficients of KCl were determined in concentrated bovine serum albumin (BSA) solutions. The concentration range studied was 0.01 to 0.5 M KCl and zero to 18% wt BSA, thus amply covering physiological conditions. The activity coefficients of the salt were measured using the EMF method with ion exchange membrane electrodes. Keeping the salt concentration constant, the activity coefficients of the salt decrease linearly with protein concentration, the effect being more pronounced for low salt content. The maximal deviations of the activity coefficients with respect to those in pure salt solution amount to ca. 40% for 0.01 M KCl and 18% wt BSA. The results were interpreted on the assumption of the superposition of three effects i.e. water bound to BSA molecules as non-solvent water, specific Cl ion binding and the electrostatic interactions of the polyions with the salt ions. In view of the results it can be concluded that only a small portion of simple intracellular ions are bound, based on the assumption that the cytoplasm of living cells may be regarded as a concentrated protein-salt solution.  相似文献

The activity coefficients at 298.15 K of glycine in water + NaNO(3) + glycine system and dl-methionine in water + NaCl + dl-methionine system are reported. The measurements were performed in an electrochemical cell with two ion selective electrodes, a cation and an anion ion selective electrode, each versus a double junction reference electrode. The concentrations of the electrolytes and the amino acids studied covered up to 1.0 molality electrolyte, 2.4 molality glycine and 0.2 molality dl-methionine. The results of the activity coefficients of glycine are compared with the activity coefficients of glycine in water + NaCl + glycine and water + KCl + glycine systems, obtained from the previous studies. The results show that the nature of both the cation and the anion of an electrolyte have significant effects on the activity coefficient of glycine in aqueous electrolyte solutions. The results also show that there are attractive interactions between the molecules of glycine and NaNO(3) and repulsive interactions between the molecules of dl-methionine and NaCl.  相似文献

GSK3/shaggy-like protein kinases have been shown to play diverse roles in development and signal transduction pathways in various organisms. An Arabidopsis homologue of GSK3/shaggy-like kinase, AtGSK1, has been shown to be involved in NaCl stress responses. In order to further clarify the role of AtGSK1 in NaCl stress responses in plants, we generated transgenic Arabidopsis plants that over-expressed AtGSK1 mRNA. These plants showed enhanced resistance to NaCl stress when assayed either as whole plants or by measurement of root growth on NaCl plates. In addition, AtGSK1 transgenic plants in the absence of NaCl stress showed phenotypic changes, such as accumulation of anthocyanin, that were similar to those observed in wild-type plants under NaCl stress. Transgenic plants accumulated 30-50% more Na+ than did wild-type plants when subjected to NaCl stress, and Ca2+ content was increased by 15-30% in the transgenic plants regardless of the NaCl stress level. Northern blotting revealed that AtGSK1 over-expression induced expression of the NaCl stress-responsive genes AtCP1, RD29A and CHS1 in the absence of NaCl stress. In addition, AtCBL1 and AtCP1 were super-induced in the NaCl-stressed transgenic plants. Taken together, these results suggest that AtGSK1 is involved in the signal transduction pathway(s) of NaCl stress responses in Arabidopsis.  相似文献

Anabaena 7120 cells were exposed to NaCl (25-175 mM) stress. Maximum growth was recorded in media containing 150mM NaCl. Short-term exposure (48h) of the cyanobacterial biomass to 150mM NaCl, induced highest trehalose level (37mM). Control cells lacking NaCl did not show any trace of trehalose as ascertained by NMR and HPLC analysis. Trehalose biosynthesis observed with NaCl plus high temperature (40 degrees C) indicated that its production was specifically triggered by NaCl, not temperature. The increase in trehalose level during NaCl stress was the result of overexpression of the trehalose-forming enzymes maltooligosyltrehalose synthase (MTSase), EC 5.4.99.15 (114kDa) and maltooligosyltrehalose trehalohydrolase (MTHase), EC 3.2.1.141 (68 kDa) as evidenced by SDS-PAGE analysis. To our knowledge this is the first report of induced trehalose biosynthesis in Anabaena 7120 during salt-stress, accompanied by identification of MTSase and MTHase enzymes on gel. It is suggested that Anabaena 7120 cells synthesize the osmolyte trehalose to withstand osmotic fluctuations.  相似文献

在含NaCl营养液中培养的小麦幼苗较之无NaCl营养液中的幼苗。其脯氨酸氧化酶活性降低,而游离脯氨酸含量则升高;培养液的渗透势越低,培养时间越长,则脯氨酸氧化酶的活性越低,且游离脯氨酸的含量越高。去除胁迫后酶活性恢复,脯氯酸含量下降。不同渗透剂对氧化酶活性抑制强弱顺序为MgCl_2>NaCl>甘露醇,引起脯氨酸累积效应的强度顺序为MgCl_2>NaCl>甘露醇。超微结构显示,高NaCl浓度下部分线粒体结构受损伤,膜和嵴部分消失。  相似文献

The activity coefficients of glycylglycine in four aqueous electrolyte solutions (+NaCl, +NaBr, +KCl and +KBr) were obtained at 298.2 K. The mean ionic activity coefficient of the electrolyte in aqueous solutions containing the peptide was determined from measurements of the potential differences of a cation and an anion ion-selective-electrode, each vs. a double junction reference electrode. The results show that the nature of the anion has a major effect on the activity coefficients of glycylglycine. Comparison of activity coefficient data for glycylglycine with literature data for glycine, both in aqueous NaCl solutions, indicates that the effect of the electrolyte is larger for the peptide than for the amino acid. For the peptide, in all cases, the effect of the electrolyte is more important at low molalities of the electrolyte. The Wilson equation was used to correlate the activity coefficient data obtained. The correlation results were satisfactory for the region of concentrated electrolyte.  相似文献

在温室盆栽条件下,用NaCl处理新疆长果阿月浑子与美国品种Kerman,处理浓度为50、150、250和500 mmol·L-1,盐处理后5、10、20 d分别取叶、茎和根,测定各器官中Na+、K+含量及Na+/K+变化规律.研究结果表明,随着土壤盐浓度的增加,长果阿月浑子和Kerman的叶、茎和根中Na+离子含量在盐处理5、10、20 d后每个处理都表现出升高趋势;而两个品种的叶、根和茎中的K+含量在盐处理后其变化规律都不稳定.长果阿月浑子叶片、茎和根系中Na+/K+的比值在盐处理5 d和10 d都升高.Kerman的叶和根系中Na+/K+比值在处理5、10、20 d后升高;茎中Na+/K+处理10 d后升高,但20 d后没有明显变化.实验结果表明NaCl胁迫后,长果阿月浑子的叶片中Na+含量增加幅度均大于Kerman,Kerman根和茎中的Na+含量增加幅度大于长果阿月浑子;NaCl胁迫5 d和10 d后,长果阿月浑子叶片中的Na+/K+的比值高于Kerman,Kerman茎和根部中的Na+/K+的比值高于长果阿月浑子,表明了长果阿月浑子和Kerman都具有一定的耐盐性,但Kerman的耐盐性强于长果阿月浑子.由于叶片和根中Na+和Na+/K+比值的变化很稳定,可以确定为阿月浑子主要抗盐指标,茎中Na+/K+的变化规律较稳定,可以作为抗盐性参考指标.  相似文献

采用蛭石栽培,在100mmol·L-1NaCl胁迫下,对耐盐性不同的2个菜用大豆[Glycinemax(L.)Merr.]品种结荚期干物质积累、单株产量及叶片游离态多胺(PAs)水平的变化进行了研究。结果表明:NaCl胁迫显著降低了菜用大豆植株干重及单株产量,但耐盐品种"绿领特早"的降幅低于盐敏感品种"理想高产95-1";与"理想高产95-1"相比,"绿领特早"叶片在整个NaCl胁迫期间均维持了相对较低的H2O2含量、游离态腐胺(Put)含量及较高的游离态亚精胺(Spd)含量,在胁迫6~15d期间维持了相对较高的游离态精胺(Spm)含量、(Spd+Spm)/Put值及较低的Put/PAs值。说明耐盐品种"绿领特早"叶片具有较强的由游离态Put向游离态Spd和Spm转化的能力,维持了较低的游离态Put含量和较高的游离态Spd及Spm含量,进而抑制了活性氧过量积累。  相似文献

The grand-canonical Monte Carlo (GCMC) technique, which is primarily used to calculate the concentration of a solute for a given activity coefficient, can be inverted and applied to calculate the activity coefficients corresponding to a given concentration of a solute. The changes needed to be introduced in the GCMC algorithm are discussed in the paper. The new method called the inverse GCMC (IGCMC) technique is applied to calculate the mean activity coefficient of 1:1, 2:2, 2:1, 3:1 salts with equal and unequal ion sizes and over a wide range of the electrolyte concentrations. The results for equal ion sizes are compared with the corresponding data obtained from the GCMC of Valleau and Cohen. It is shown that the IGCMC technique, after some extensions, can be used to compute the individual ionic activity coefficients. A comparison is made with the Sloth and Sørensen individual activity results and with the theoretical predictions for the 1:1 electrolyte with unequal ion sizes.  相似文献

The enthalpies of dissolving glycine and DL-alanine in water solutions of D-glucose, D-maltose, and sucrose at 298.15 K were determined by calorimetry. From the results obtained, the coefficients of enthalpy for pairwise interactions hxy of the amino acids and saccharides in water were calculated. It was found that the hxy values for glycine in solutions of all saccharides studied are negative; in the case of DL-alanine, the hxy values are positive for all saccharides except for sucrose solution. It was shown that the hxy values reflect the sum effect of interactions between the amino acids and saccharides in aqueous solutions and the contribution of hydration of the solutes.  相似文献

以烤烟品种K326为材料,研究不同浓度NaCl胁迫处理下种子萌发及幼苗生理生化指标的变化。结果表明,低浓度NaCl处理对幼苗生长具有一定的促进作用,而高浓度NaCl胁迫处理具有显著或极显著抑制作用;随着NaCl处理浓度的增加,发芽势、发芽指数、简单活力指数、形态指标、钾离子含量、单株干鲜重均呈明显下降趋势,而钠离子含量则相反;随着NaCl处理浓度的增加以及胁迫时间的延长,K326幼苗叶片中丙二醛、游离脯氨酸、可溶性糖含量及电解质渗透率均呈上升趋势,而可溶性蛋白、叶绿素含量及根系活力变化则相反。  相似文献

Isothermal-isobaric molecular dynamics simulations are used to calculate the specific volume of models of trehalose and three amorphous trehalose-water mixtures (2.9%, 4.5% and 5.3% (w/w) water, respectively) as a function of temperature. Plots of specific volume versus temperature exhibit a characteristic change in slope when the amorphous systems change from the glassy to the rubbery state and the intersection of the two regression lines provides an estimate of the glass transition temperature T(g). A comparison of the calculated and experimental T(g) values, as obtained from differential scanning calorimetry, shows that despite the predicted values being systematically higher (about 21-26K), the trend and the incremental differences between the T(g) values have been computed correctly: T(g)(5.3%(w/w))相似文献

以中国南瓜360-3×112-2杂交种和黑籽南瓜为材料,在水培条件下研究了成株期NaCl胁迫对其植株生长和根系生理生化特征的影响.结果表明:(1)NaCl胁迫对2材料的根系鲜重、地上部鲜重、主蔓长度和功能叶单叶面积都有不同程度抑制,而黑籽南瓜所受抑制更强,并以功能叶单叶面积受抑制程度最大.(2)NaCl处理能显著提高杂交种的根系活力,却对黑籽南瓜植株根系活力有显著抑制作用.(3)随NaCl胁迫延续,2材料根系中可溶性糖和脯氨酸含量均呈现先上升后下降的规律,且杂交种植株的上升幅度较大而下降的幅度较小并始终显著高于同期黑籽南瓜.(4)2材料根系O2·-产生速率随NaCl胁迫延续而倍增,但杂交种上升幅度逐渐减小,而黑籽南瓜上升幅度持续增加;MDA含量在杂交种根系中先升后降,而在黑籽南瓜中持续上升,且同期黑籽南瓜增加幅度明显大于杂交种;黑籽南瓜根系的O2·-产生速率和MDA含量始终显著高于同期其它处理.(5)NaCl胁迫使2材料根系抗氧化酶活性均有不同程度增加但变化趋势有异,且杂交种的酶活性增幅均大于同时期黑籽南瓜.研究发现,成株期的中国南瓜杂交种360-3×112-2在NaCl胁迫下生长抑制率较低,根系活力、渗透调节能力和活性氧平衡调节能力较强,耐盐性优于黑籽南瓜,可望作为耐盐瓜类砧木使用.  相似文献

Botrytis cinerea, the causal agent of grey mould in a broad range of crops, is considered a high‐risk plant pathogen for fungicide resistance development. The use of fungicide mixtures, particularly combinations with synergistic activity, can be a useful tactic to counteract resistance build‐up in pathogen populations. The present study aimed to investigate the effects of different ratios of two‐way mixtures of carbendazim, iprodione, kresoxim‐methyl, tebuconazole and penconazole on four B. cinerea isolates that were sensitive or resistant to benzimidazoles, dicarboximides and strobilurins. The isolates that were resistant to benzimidazoles and strobilurins had E198A and G143A mutations in β‐tubulin and cytochrome b genes, respectively. The mixtures had different effects on each of the isolates in vitro but, in 13 combinations, the synergistic effect was observed against all or three isolates. In greenhouse experiments, 11 fungicide combinations used in decreased (EC75) concentrations showed the maximum control efficiency. The two follow‐up greenhouse experiments using six selected combinations revealed they were highly effective against additional isolates with various fungicide resistance profiles. The identified mixtures‐ratios have potential for use in grey mould management programs in the greenhouse.  相似文献

NaCl胁迫初期 ,Na 主要在根和叶鞘中积累。相应地 ,根和叶鞘液泡膜ATP酶和焦磷酸酶水解活性、依赖ATP和PPi的质子泵活性及Na /H 逆向转运活性均明显增加 ,根和叶鞘的生长没有受到抑制。NaCl胁迫后期 ,Na 开始向地上部分运输并在叶片中积累。此时 ,叶片液泡膜质子泵和Na /H 逆向转运活性开始增加 ,根和叶鞘的Na/K比增加 ,其液泡膜ATP酶和焦磷酸酶水解活性、质子泵活性和Na /H 逆向转运活性下降。相应地 ,根和叶鞘的生长也下降。当保温介质中Na/K比超过 1时 ,液泡膜微囊ATP酶和焦磷酸酶活性均随Na/K比的增加而下降。表明非盐生植物液泡膜质子泵在盐胁迫的初期对Na 在液泡内的积累及其耐盐性起重要作用  相似文献

培养在含NaCl营养液中的幼苗,根生长被抑制,线粒体超微结构受损伤。用扫描电镜、X-射线能谱仪和电感耦合等离子直读光谱(ICP)分析发现,培养在含NaCl营养液中的根,积累大量Na+和Cl-,而K+和Ca~(2+)的含量降低,无机离子总量和Na+/K+比值提高。  相似文献
