Seasonal changes of some water relations parameters of Norway spruce shoots ( Picea abies [L.] Karst.) were studied during two experiments using the pressure-volume analysis. For each experiment only shoots of a single tree were used.
During the first study, the course of the turgor loss point (as bulk osmotic pressure when turgor first reaches zero, πp) of shoots developed in late 1986 vegetation period, were measured in 1987. The turgor loss point decreased temporarily from –2.5 MPa at the beginning of the year to –3.3 MPa at the end of March, but then increased to the original level for the rest of the year.
During the second study, water relations parameters were measured in late summer 1987 and in late winter 1988. Winter shoots at full water saturation contained up to 20% less water than in late summer. Accordingly, the bulk osmotic pressure at full water saturation (πp) decreased from –1.7 MPa in late summer to –1.9 MPa in winter, πp decreased also from –2.2 MPa to –2.8 MPa. However, the amount of osmotically active substances (mOsmol, N) remained unchanged. The relative amount of apoplastic water in the total shoot water content appeared to drop insignificantly from 17% to 15%.
The results show that the decrease in πo and πp in late winter is not due to an accumulation of osmotically active substances in the vacuoles but is due to a decrease in tissue water content. The temporary reduction of the symplastic volume by deposition of osmotically inert substances seems to be the most probable cause of this phenomenon.  相似文献

Bean plantlets ( Phaseolus vulgaris L. cv. Topcrop) were stressed at the age of 16–18 days by gradual (2–8%) or abrupt addition of 6% (w/v) polyethylene glycol Mw 6000 (PEG 6000) to Hoagland solution. Leaf conductance, photosynthesis, internal CO2 partial pressure (Ci), relative water content (RWC), water content/dry weight (H2O/DW), apoplastic PEG concentrations and weight of leaves, stems and roots were determined. Leaf conductance, photosynthesis and Ci were determined on non-detached primary leaves, and leaf potentials (water, osmotic and turgor potentials) were investigated in freshly detached (non-rehydrated) primary leaves, both in treated and control plants; RWC and osmotic potential were also assessed at the null turgor point. Low PEG 6000 concentrations induced early and evident decrease in leaf conductance and photosynthesis, whereas Ci decreased only moderately and tended to recover during advanced stress. There were moderate though significant decreases in RWC and H2O/DW, no change or increases in water potential, no significant changes in osmotic potential and a moderate but significant increase in turgor potential. Even when referred to null turgor point, RWC significantly decreased and osmotic potential was unchanged. It was concluded that apoplastic PEG 6000 accumulation at evaporating sites would account for the early decrease in conductance which would also justify the unchanged or the prevalent increase in water potential and turgor potential. The subsequent PEG diffusion and concentration in the leaf apoplastic water would have induced the RWC and H2O/DW decrease and the final turgor flexion documented.  相似文献

Genetic variation in the drought response of leaf and root tissue water relations of seedlings of eight sources of black walnut ( Juglans nigra L.) was investigated using the pressure-volume technique. Tissue water relations were characterized at three stages of a drying cycle during which well-watered plants were allowed to desiccate and then were reirrigated.
Sources varied both in the capacity for and degree of leaf and root osmotic adjustment, and in the mechanism by which it was achieved. A decrease in osmotic potential at the turgor loss point (ψπp) of 0.4 MPa was attributable to increased leaf tissue elasticity in seedlings of four sources, while seedlings of an Ontario source exhibited a 0.7–0.8 MPa decline in ψπp as a result of both increased solute content and increased leaf tissue elasticity. Seedlings of a New York source showed no detectable osmotic adjustment.
In roots, decreased ψπp (osmotic potential at full hydration) and ψπp were observed under drought. Sources that exhibited significant leaf osmotic adjustment also generally showed a similar response in roots. Tissue elasticity and ψπp of roots were higher than those of shoots, whereas ψπp of the two organs was similar for most sources. Because of greater elasticity, roots exhibited a more gradual decline in turgor and total water potential than did leaves as tissue relative water content decreased.  相似文献

Abstract. Osmotic adjustment, a mechanism whereby plants maintain positive turgor despite low water potential (ψ), was investigated in pearl millet ( Pennisetum americanum [L.] Leeke) in three types of field experiment at Hyderabad, India:
  • (1)

    Osmotic adjustment during the growing season was evaluated by comparing solute potential (ψs) of leaves taken at midday from irrigated and droughted plots and allowed to rehydrate in the laboratory. The degree of seasonal adjustment was also estimated by comparing observed values of ψs in the field with those expected if ψs decreased solely in proportion to water loss. Both types of assessment indicated the maximum seasonal adjustment to be about 0.2 MPa. The cultivars BJ 104 and Serere 39 differed in their capacity to adjust osmotically over the season; Serere 39 was least able to osmoregulate.

    Measurements of diurnal variations in ψ and ψs in BJ 104 revealed osmotic adjustment during the afternoon hours. At a given value of ψ, turgor (ψp) was about 0.1 MPa higher in irrigated, and over 0.2 MPa higher in droughted plants, in the afternoon, than in the morning.

    Osmotic adjustment of different leaves within the canopy was investigated. Upper leaves had lower ψ than basal leaves. Differences in ψ were matched by gradients in ψs, so that turgor was similar for all leaf layers.

Seasonal and diurnal measurements of leaf water potential (ψ1), relative water content (RWC) and stomatal conductance (gs) were made in the field on 19-year old Prunus salicina (L.) cv. Santa Rosa, a deciduous fruit tree species, irrigated with 3 different concentrations of saline water over a 3 year period (1985-1987). With the exception of stage III of fruit growth, little or no treatment difference in Φ1, leaf turgor potential (Φp), or RWC was noted during the day. Seasonal averages of morning (0700-0900) and afternoon (1500-1700) Φp did not decline with increasing salinity, indicating long-term osmotic adjustment in this species. Maintenance of leaf water status under saline conditions was in part a consequence of increased stomatal closure, with a subsequent reduction in leaf transpiration rate. However, during stage III of fruit growth, an increase in mean afternoon (1200-1700) stomatal conductance of 26-117%, independent of salinity treatment, was observed in 1985 and again in 1987. Higher conductance values during this period may be associated with rapid fruit expansion and greater assimilate demand. The observed increase in conductance resulted in greater leaf water loss and larger measured differences in midday ψ1 between salinity treatments. This research indicates that for Prunus salicina in the field, salinity stress resulted in leaf water deficits only during the final period of fruit expansion and ripening.  相似文献

Soybean ( Glycine max cv. Clark) was grown at both ambient (ca 350 μmol mol−1) and elevated (ca 700 μmol mol−1) CO2 concentration at 5 growth temperatures (constant day/night temperatures of 20, 25, 30, 35 and 40°C) for 17–22 days after sowing to determine the interaction between temperature and CO2 concentration on photosynthesis (measured as A, the rate of CO2 assimilation per unit leaf area) at both the single leaf and whole plant level. Single leaves of soybean demonstrated increasingly greater stimulation of A at elevated CO2 as temperature increased from 25 to 35°C (i.e. optimal growth rates). At 40°C, primary leaves failed to develop and plants eventually died. In contrast, for both whole plant A and total biomass production, increasing temperature resulted in less stimulation by elevated CO2 concentration. For whole plants, increased CO2 stimulated leaf area more as growth temperature increased. Differences between the response of A to elevated CO2 for single leaves and whole plants may be related to increased self-shading experienced by whole plants at elevated CO2 as temperature increased. Results from the present study suggest that self-shading could limit the response of CO2 assimilation rate and the growth response of soybean plants if temperature and CO2 increase concurrently, and illustrate that light may be an important consideration in predicting the relative stimulation of photosynthesis by elevated CO2 at the whole plant level.  相似文献

Abstract. A controlled-environment chamber constructed from a standard chest freezer was used to grow and measure the CO2 exchange of small stands of lettuce ( Lactuca sativa L. ev. Ambassador). The chamber, with horizontal air flow, provided good control of air temperature ( c. 6 to 16°C), irradiance (0–300 μmol PAR m 2S1 and CO2 (350–1000 μmol mol−1). The photosynthetic response to changes in these variables was measured using an inexpensive CO2 dosing system which recorded the input rate required to maintain a constant concentration of CO2 (to ± 2.5%). Characteristis of the growth environment and the changes in response to temperature and irradiance are described.  相似文献

Cultivated tomato Lycopersicon esculentum (L.) Mill. cv. P-73 and its wild salt-tolerant relative L. pennellii (Correll) D'Arcy accession PE-47 growing on silica sand in a growth chamber were exposed to 0, 70, 140 and 210 m M NaCl nutrient solutions 35 days after sowing. The saline treatments were imposed for 4 days, after which the plants were rinsed with distilled water. Salinity in L. esculentum reduced leaf area and leaf and shoot dry weights. The reductions were more pronounced when sodium chloride was removed from the root medium. Reduction in leaf area and weight in L. pennellii was only observed after the recovery period. In both genotypes salinity induced a progressive reduction in leaf water potential and leaf conductance. During the recovery period leaf water potential (ψ1) and leaf conductance (g1) reached levels similar to those of control plants in wild and cultivated species, respectively. Leaf osmotic potential at full turgor (ψos) decreased in the salt treated plants of both genotypes, whereas the bulk modulus of elasticity was not affected by salinity. Leaf water potential at turgor loss point (ψtlp) and relative water content at turgor loss point (RWCtlp) appeared to be controlled by leaf osmotic potential at full turgor (ψos) and by bulk modulus of elasticity, respectively. At lowest salinity, the wild species carried out the osmotic adjustment based almost exclusively on Cl and Na+, with a marked energy savings. Under highest salinity, this species accommodate the stress through a higher expenditure of energy due to the contribution of organic solutes to the osmotic adjustment. The domesticated species carried out the osmotic adjustment based always on an important contribution of organic solutes.  相似文献

The objective of the study was to investigate the interactive effects of elevated atmospheric carbon dioxide concentration, [CO2], and temperature on the wood properties of mature field-grown Norway spruce ( Picea abies (L.) Karst.) trees. Material for the study was obtained from an experiment in Flakaliden, northern Sweden, where trees were grown for 3 years in whole-tree chambers at ambient (365 μmol mol−1) or elevated [CO2] (700 μmol mol−1) and ambient or elevated air temperature (ambient +5.6 °C in winter and ambient +2.8 °C in summer). Elevated temperature affected both wood chemical composition and structure, but had no effect on stem radial growth. Elevated temperature decreased the concentrations of acetone-soluble extractives and soluble sugars, while mean and earlywood (EW) cell wall thickness and wood density were increased. Elevated [CO2] had no effect on stem wood chemistry or radial growth. In wood structure, elevated [CO2] decreased EW cell wall thickness and increased tracheid radial diameter in latewood (LW). Some significant interactions between elevated [CO2] and temperature were found in the anatomical and physical properties of stem wood (e.g. microfibril angle, and LW cell wall thickness and density). Our results show that the wood material properties of mature Norway spruce were altered under exposure to elevated [CO2] and temperature, although stem radial growth was not affected by the treatments.  相似文献

Vigna cutjang Endl. cv. Pusa Barsati seedlings, subjected to increasing degrees of water stress (−0.5, −1.0, −1,5 MPa), produced an approximately proportional increase in glycolate oxidase activity, hydrogen peroxide (H2O2) and proline content but a decrease in catalase activity, ascorbic acid and protein content. Leaf water potential (leaf ψ) and relative water content (RWC) were also lowered with increasing stress. Pretreatment with l -cysteine and reduced glutathione (10-3 M) decreased glycolate oxidase activity, H2O2 content, ascorbic acid oxidase activity, proline content and also slightly improved the water status of leaves stressed (−1.0 MPa) for 2 days. Pretreatment of non-stressed seedlings with these antioxidants had little or no effect. These studies indicate that treatment with antioxidants makes the plant tolerant against water stress by modulating the endogenous levels of H2O2 and ascorbic acid in stressed tissue.  相似文献

The temperature dependence of C3 photosynthesis may be altered by the growth environment. The effects of long-term growth in elevated CO2 on photosynthesis temperature response have been investigated in wheat ( Triticum aestivum L.) grown in controlled chambers with 370 or 700 μmol mol−1 CO2 from sowing through to anthesis. Gas exchange was measured in flag leaves at ear emergence, and the parameters of a biochemical photosynthesis model were determined along with their temperature responses. Elevated CO2 slightly decreased the CO2 compensation point and increased the rate of respiration in the light and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) Vcmax, although the latter effect was reversed at 15°C. With elevated CO2, Jmax decreased in the 15–25°C temperature range and increased at 30 and 35°C. The temperature response (activation energy) of Vcmax and Jmax increased with growth in elevated CO2. CO2 enrichment decreased the ribulose 1,5-bisphosphate (RuBP)-limited photosynthesis rates at lower temperatures and increased Rubisco- and RuBP-limited rates at higher temperatures. The results show that the photosynthesis temperature response is enhanced by growth in elevated CO2. We conclude that if temperature acclimation and factors such as nutrients or water availability do not modify or negate this enhancement, the effects of future increases in air CO2 on photosynthetic electron transport and Rubisco kinetics may improve the photosynthetic response of wheat to global warming.  相似文献

Abstract. The objective of this study was to investigate the effects of water stress in sweet potato ( Ipomoea batatas L. [Lam] 'Georgia Jet') on biomass production and plant-water relationships in an enriched CO2 atmosphere. Plants were grown in pots containing sandy loam soil (Typic Paleudult) at two concentrations of elevated CO2 and two water regimes in open-top field chambers. During the first 12 d of water stress, leaf xylem potentials were higher in plants grown in a CO2 concentration of 438 and 666 μmol mol−1 than in plants grown at 364 μmol mol−1. The 364 μmol mol−1 CO2 grown plants had to be rewatered 2 d earlier than the high CO2-grown plants in response to water stress. For plants grown under water stress, the yield of storage roots and root: shoot ratio were greater at high CO2 than at 364 μmol mol−1; the increase, however, was not linear with increasing CO2 concentrations. In well-watered plants, biomass production and storage root yield increased at elevated CO2, and these were greater as compared to water-stressed plants grown at the same CO2 concentration.  相似文献

Bifidobacterium adolescentis Int-57 isolated from human faeces produced extracellular amylase. The enzyme was purified from the culture supernatant fluids by ammonium sulphate precipitation, gel-filtration chromatography (Sephadex-G-75), ion-exchange chromatography (CM-cellulose) and FPLC. SDS-PAGE of the purified enzyme revealed a major band with an apparent molecular weight of 66 kDa. The pI was 5·2. Enzyme activity was optimal at 50°C, and at pH 5·5. The enzyme was stable at 20–40°C, and at pH 5–6 with a K m value of 2·4 g l−1 soluble starch. The activation energy was 42·3 kJ mol−1. The enzyme was significantly inhibited by maltose (10%), glucose (10%), Cu2+ (5 mmol l−1), Zn2+ (5 mmol l−1), N- bromosuccinimide (5 mmol l−1), EDTA (5 mmol l−1), I2 (1 mmol l−1) and activated by β-mercaptoethanol (10 mmol l−1).  相似文献

Pressure-volume curves and drought resistance in two wheat genotypes   总被引:1,自引:0,他引:1  
The water relations of two durum wheat cultivars ( Triticum durum Desf.) were studied throughout the growing season. Irrigated and unirrigated plants were compared from booting to milk stage; a period where water stress occurred naturally in the field. Modulus of elasticity (ε), turgid weight/dry weight ratio (TW/DW), relative water content at zero turgor (RWCo) and osmotic potential at full turgor (ε) declined throughout the season while average turgor (ψp) increased. Water stress induced a further decrease in ψπ100 and the TW/DW ratio. The elastic modulus varied greatly. During the first stages of growth, cv. Appulo (the more resistant cultivar) showed lower ε values than cv. Valforte. At the milk stage, ε was lower for the unirrigated than the irrigated plants. Correlation coefficients between the TW/DW ratio and the osmotic potential were significant for both cultivars. In cv. Valforte, TW/DW was also correlated with the average turgor and the bulk modulus of elasticity. Structural changes that affect the TW/DW ratio seem to be important factors influencing water relations and drought tolerance in durum wheat.  相似文献

Evidence is accumulating in favor of a linkage at the cellular level between various abiotic stresses. We conducted a study to evaluate the effect of water stress on the heat tolerance of zonal geraniums, Pelargonium × hortorum cv. Evening Glow. Water stress was imposed by withholding irrigation until pots reached 30% (by weight) of well‐watered controls, and by maintaining the pot weight by additions of water for another 7 days. Leaf xylem water potential (XWP, MPa), relative water content (RWC. %), and heat‐stress tolerance (HST; LT50, defined as the temperature causing half‐maximal % injury based on electrolyte leakage) were measured in control, stressed, and recovered plants. Proteins were extracted from the leaves following the above treatments, and SDS‐PAGE and immunoblotting were performed by using standard procedures. Immunoblots were probed with antibodies to dehydrin and 70‐kDa heat shock cognate (HSC70) proteins. Data indicate that XWP and RWC, respectively, were −0.378 MPa and 92.3% for control plants and −0.804 MPa and 78.6% for stressed plants. Water‐stressed plants exhibited a significant increase in HST compared to control (LT50 of 55°C vs 51°C). Water‐stress‐induced HST was not due to heat acclimation (leaf warming in stressed plants). Data also indicate that water‐stress treatment did not increase freezing tolerance of geranium leaves. Increased HST was associated with the accumulation of several heat‐stable, dehydrin proteins (25–60 kDa), and both cytosolic and ER luminal (BiP) HSC70 proteins. Leaf XWP, RWC, and HST reversed to control levels concomitant with the disappearance/reduction of dehydrins and HSC70 proteins in water‐stress‐relieved plants. The possibility of a cellular linkage between water stress and heat‐stress tolerance is discussed.  相似文献

Abstract Maintenance coefficients and rates of turnover of cell protein, cell walls and total cell phosphate were determined during the growth of Escherichia coli ML308 in fed-batch culture at different temperatures. The apparent Arrhenius activation energies ( E a) calculated for maintenance coefficients at temperatures over 30°C were 212–515 kJ· mol−1, indicative of a process thermodynamically similar to denaturation. The corresponding Arrhenius values for turnover were of a different magnitude, 44–115 kJ mol−1, more typical of enzyme-catalysed reactions. Addition of 0.5 M NaCl had a minor effect on maintenance at both temperatures. Turnover of cell material and salt homeostasis were therefore concluded to be minor components of the maintenance requirement of E. coli .  相似文献

Leaf Diffusive Conductance and Tap Root Cell Turgor Pressure of Sugarbeet   总被引:2,自引:2,他引:0  
Abstract. The interrelationships of leaf diffusive conductance, tap root cell turgor pressure and the diameter of the tap root of sugarbeet were studied. The study was conducted on well-watered plants growing in pots under artificial light in the glasshouse. In a typical experiment, on illumination (400 μmol m−2 s−1) leaf conductance increased from 0.6 to 7.4 mm s−1. Cell turgor pressure in the tap root decreased from 0.8 MPa to 0.45 MPa and the root diameter (9.0 cm) contracted by 145μm. Removal of light resulted in the reversal of each of the above parameters to their previous values. Quantitively similar results were obtained when sugar beet plants were uprooted and the response of each of the parameters was measured. The sequence of events however was different. On stimulation by light, changes in leaf diffusive conductance preceded the turgor and root diameter changes (which were simultaneous) by some 15–20min. In contrast, on uprooting the simultaneous changes in root turgor pressure and diameter preceded the changes in leaf conductance. The lag times between changes in diffusive conductance and turgor pressure in the root were between 20 and 30 min.
Tap root turgor pressure and diameter correlated strongly and permitted the calculation of an apparent whole root volumetric elastic modules (55–63 MPa). The small changes in tissue volume relative to the transpiration rate suggest that the tap root is not a significant source of transpirational water during the day.  相似文献

Rice ( Oryza sativa L. cv. IR72) was grown at three different CO2 concentrations (ambient, ambient + 200 μmol mol−1, ambient + 300 μmol mol−1) at two different growth temperatures (ambient, ambient + 4°C) from sowing to maturity to determine longterm photosynthetic acclimation to elevated CO2 with and without increasing temperature. Single leaves of rice showed a cooperative enhancement of photosynthetic rate with elevated CO2 and temperature during tillering, relative to the elevated CO2 condition alone. However, after flowering, the degree of photosynthetic stimulation by elevated CO2 was reduced for the ambient + 4°C treatment. This increasing insensitivity to CO2 appeared to be accompanied by a reduction in ribulose-1.5-bisphosphate carboxylase/oxygenase (Rubisco) activity and/or concentration as evidenced by the reduction in the assimilation (A) to internal CO2 (C1) response curve. The reproductive response (e.g. percent filled grains, panicle weight) was reduced at the higher growth temperature and presumably reflects a greater increase in floral sterility. Results indicate that while CO2 and temperature could act synergistically at the biochemical level, the direct effect of temperature on floral development with a subsequent reduction in carbon utilization may change sink strength so as to limit photosynthetic stimulation by elevated CO2 concentration.  相似文献

Three soybean ( Glycine max L. Merr.) cultivars (Maple Glen, Clark and CNS) were exposed to three CO2 concentrations (370, 555 and 740 μmol mol−1) and three growth temperatures (20/15°, 25/20° and 31/26°C, day/night) to determine intraspecific differences in single leaf/whole plant photosynthesis, growth and partitioning, phenology and final biomass. Based on known carboxylation kinetics, a synergistic effect between temperature and CO2 on growth and photosynthesis was predicted since elevated CO2 increases photosynthesis by reducing photorespiration and photorespiration increases with temperature. Increasing CO2 concentrations resulted in a stimulation of single leaf photosynthesis for 40–60 days after emergence (DAE) at 20/15°C in all cultivars and for Maple Glen and CNS at all temperatures. For Clark, however, the onset of flowering at warmer temperatures coincided with the loss of stimulation in single leaf photosynthesis at elevated CO2 concentrations. Despite the season-long stimulation of single leaf photosynthesis, elevated CO2 concentrations did not increase whole plant photosynthesis except at the highest growth temperature in Maple Glen and CNS, and there was no synergistic effect on final biomass. Instead, the stimulatory effect of CO2 on growth was delayed by higher temperatures. Data from this experiment suggest that: (1) intraspecific variation could be used to select for optimum soybean cultivars with future climate change; and (2) the relationship between temperature and CO2 concentration may be expressed differently at the leaf and whole plant levels and may not solely reflect known changes in carboxylation kinetics.  相似文献
