The induction of reciprocal translocations in mouse germ cells by combined treatments with chemicals and ionizing radiations has been studied. Male mice were intraperitoneally injected with doses of 5 or 10 mg/kg of adriamycin (ADR) and irradiated with doses of 5 or 9 Gy of gamma-rays 24 h later. Three types of response were found after analyzing diakinesis-metaphase I multivalent configurations: potentiation, with the dose of 5 mg/kg of ADR plus 9 Gy; subadditivity, with the dose of 5 mg/kg of ADR plus 5 Gy; and additivity, with the dose of 10 mg/kg of ADR plus 5 or 9 Gy. According to these results, the subadditive effect observed with the lower dose of ADR plus 5 Gy cannot be explained under the assumption that depletion of any kind of spermatogonia is sufficient for modifying the chromosomal response of stem cells to ionizing radiations. The role of DNA repair mechanisms modulating the response of spermatogonial cells to combined treatments is discussed under the assumption that some repair mechanisms can be triggered by treatment with a low dose of a chemical and these repair mechanisms can reduce cell mortality. Consequently, a higher frequency of more radioresistant cells can survive.  相似文献

The combined effects of mitomycin C (MMC) and thio-tepa (TT) with gamma-ray doses of 5 and 9 Gy on mouse stem cells were studied using the spermatocyte test. Both chemicals induced very low yields of translocations after single treatments. In combined treatments with a dose of 5 Gy, a subadditive effect of MMC and an additive effect of TT were found. Combined with a dose of 9 Gy the compounds potentiated the effect of radiations. Up to now, most of the chemicals tested have shown additive effects when combined with doses of the ascending part of the dose-response curve and potentiating effects when combined with doses of its descending part. This has been considered additional confirmation of the concept that depletion of any kind of spermatogonia is sufficient to modify the genetic response of stem cells. However, the subadditive and additive responses found could be considered evidence that common biological mechanisms can modulate the response to combined treatments of chemicals and ionizing radiations.  相似文献

The induction of reciprocal translocations in mouse spermatogonial stem cells, visualized in dividing primary spermatocytes, was studied after combined treatments with hydroxyurea (250 and 500 mg/kg) and X-rays (6, 8 and 9 Gy). The time intervals between the 2 treatments were 16 h (leading to extremely high cell killing) and 48 h (giving rise to less killing than irradiation alone). Comparison of the observed frequencies of translocations with reported data on stem cell killing (de Ruiter-Bootsma and Davids, 1981 show that the ratio between the probabilities that a radiation-induced basic lesion kills a cell or produces a translocation, theoretically calculated by Leenhouts and Chadwick (1981) to be about 10, can indeed be confirmed experimentally.  相似文献

The radioprotective drugs AET, serotonin, and ATP were tested for antimutagenic activity against induction by 4.0 Gy X-rays of reciprocal translocations in mouse spermatogonia. Single drugs administered in doses of 8, 24 and 360 mg/kg b.wt., respectively, had no effect on translocation yields recorded in diakinesis-metaphase I spermatocytes. Two-drug mixtures afforded insignificant protection. Three-drug mixtures, however, were found to reduce radiation damage considerably, and the extent of protection was dependent in part on the amount of ATP. The best effect was obtained with formulations of serotonin-AET-ATP at the following doses, respectively: 8 + 24 + 360 mg/kg, 16 + 24 + 336 mg/kg, and 16 + 32 + 264 mg/kg. Less effective were the serotonin-AET-ATP formulations: 16 + 32 + 120 mg/kg, and 8 + 24 + 480 mg/kg. Treatment with drugs omitting radiation exposure was observed to raise, though insignificantly, the level of spontaneous translocation frequency.  相似文献

3 adult monkeys, one Macaca fascicularis and two Macaca mulatta, were whole-body irradiated with 1 Gy gamma-rays (60 Co). Reciprocal translocations induced in spermatogonial stem cells were scored as translocation multivalents in primary spermatocytes from 7.5 to 27.5 months after exposure. The translocation yields ranged from 4.1% at the earliest to 1.8% at the latest sampling interval. No significant differences were observed in the responses of the individual animals. A decline in the translocation frequencies with time after treatment was found in all 3 animals. The present data are different from those reported for testicular X-irradiation of the rhesus monkey Macaca mulatta (van Buul, 1980; Lyon et al., 1976) in that the translocation yields are higher. They are consistent with the results reported for testicular gamma-irradiation of the crab-eating monkey Macaca fascicularis (Matsuda et al., 1984, 1985). In view of the present results it appears unlikely that a species difference exists within the genus Macaca in the sensitivity of spermatogonial stem cells to the induction of translocations by ionizing radiation.  相似文献

Male (101 × C3H)F1 hybrid mice, 10–12 weeks old, were injected i.p. with single doses of 2.5, 3.75 or 5.0 mg/kg of mitomycin C (MC). Spermatogonia were sample for mitotic chromosome analyses 6, 18 or 24 h after treatment. Spermatocytes were sample for meiotic chromosome analyses 50 or 60 days after treatment.The maximal yield of chromatid-type aberrations induced in spermatogonia was found 24 h after treatment with 5.0 mg/kg of MC. More than 50% of the cells carried at least one chromatid exchange. The majority (90%) of these were whole-arm exchanges derived from breaks in the centromeric heterochromatin.No translocation multivalents were found in spermatocytes analysed 50 or 60 days after treatment. The discrepancy between the presence of many symmetrical exchanges in spermatogonia and the absence of translocation multivalents in primary spermatocytes may be result of insensitivity of the stem cell spermatogonia against exchange induction by MC or of complete germinal selection against induced translocations before and/or during early meiosis. However, the possibility of missing translocations due to whole-arm exchanges in acrocentric chromosomes during the analysis of diakineses-metaphases I is also discussed.It is emphasized that comparisons of chromatid exchange frequencies in spermatogonia with the yield of translocation multivalents in spermatocytes descended from these spermatogonia as opposed to those from stem cells might provide an estimate of pre-diakinesis germinal selection against chromatid exchanges or the resulting translocations. This estimate is important for the quantitative evaluation of the genetic risk from environmental mutagens.  相似文献

Hybrid male mice were given 5 Gy + 5 Gy acute X-rays 24 h apart, with cytological examination of testes 16-19, 39-42 and 64-66 weeks later. Mean testis weights were significantly lower in the youngest group than in the other two. However, translocation frequencies in spermatocytes of the youngest group (mean of 0.57 per cell) were significantly higher than in either of the other two groups, which gave similar values averaging 0.36 translocations per cell. There was highly significant heterogeneity in translocation yields within the youngest group. The decline in translocation yield with time after irradiation is in line with that reported by Léonard and Deknudt (1970) in inbred strain C57BL males. Analysis of all available data suggests that high translocation yields are found during late stages in the process of germ-cell repopulation of the testis after high radiation doses and may be connected with changing frequencies of radiosensitive and radioresistant stem cell populations as repopulation proceeds.  相似文献

Heritable translocation and dominant lethal tests were conducted with random-bred Swiss albino male mice. The animals were provided drinking water containing triethylenemelamine (TEM) for 4 weeks, and were then mated for 3 successive weeks for analysis of dominant lethality and production of F1 progeny. Potential translocation carriers among F1 males were selected after two breedings and confirmed by cytogenetic analysis. Translocation heterozygotes were obtained in offspring of the TEM-treated groups, but not in the control groups. In F1 males produced from the first week of mating, the frequencies of translocations were 0, 1.78 6.2 and 10.0% for the control group and groups receiving TEM at 0.0125, 0.025 and 0.050 mg/kg/day, respectively, and in those produced from the third week of mating, the values were 0 and 2.1%, respectively, for the control group and the group receiving TEM at 0.050 mg/kg/day. F1 males from the second week of mating were not studied for the induction of heritable translocations. TEM-induced dominant lethality and heritable translocations were most prominent in the first week of mating after 4 weeks of treatment. In addition, heritable translocations appeared to be a more sensitive endpoint than dominant lethal mutations for the measurement of mutagenic effects of TEM.  相似文献

Cytological and fertility tests were performed in F1 male mice derived from different germ-cell stages of male parents treated with cyclophosphamide (350 mg/kg body weight). The objectives of the present experiment were: (I) to determine the sensitivity of the male germ-cell stages to the induction of translocations by the compound, and (2) to characterize translocation configurations in F1 and F2 males, in order to obtain information about the pattern of chromosome breakage induced and its transmission to subsequent generations. Of 508 F1 males studied, 39 were partially sterile. The group of males conceived 8-21 days after treatment contained by far the highest proportion of partially sterile animals (30%). It was also the only group in which totally sterile animals (11%) were found. Of 25 semisterile males from this group, 24 gave evidence of translocations when spermatocytes were scored at diakinesis. The translocation frequencies in F1 derived from treated spermatozoa and spermatocytes were 14 and 1%, respectively. No translocations were detected cytologically in 6 semisterile males derived from treated spermatogonial stages. These results indicate that spermatid stages are especially sensitive to the mutagenic action of cyclophosphamide. In 21 of the 31 semisterile translocation males (68%), the majority of the spermatocytes contained 18 bivalents plus a ring-of-four configuration, indicating that both breakpoints were relatively centrally located; and in several of these males, the frequency of cells with rings was close to 100%. In another 9 F1 males (29%) the predominant multivalent configuration was a chain-of-four, indicating one of the breakpoints to be relatively more terminally located; and in one male (3%), the majority of cells had two unequal bivalents, indicating both breakpoints to be fairly close to the ends of the chromosomes involved. Genetic damage by ethyl methanesulfonate (EMS) in male mice was measured at doses ranging from 50 to 300 mg/kg with dominant-lethal mutations and reciprocal translocations as endpoints. No appreciable increase in dominant-lethal mutations was detected following a dose of 100 mg/kg. Dominant lethals induced by EMS were convincingly detected only after a dose of 150 mg/kg, but in the translocation experiment an increase in the genetic effect was detectable at the 50 mg/kg dose. It is likely that dominant lethals had also been induced at the 50 and 100 mg/kg doses, but were not detected due to the relative insensitivity of the dominant..lethal procedure. Thus, for detection of low levels of EMS-induced chromosome breakage, translocations are a much more reliable endpoint than are dominant-lethal mutations. A procedure for large-scale screening of induced translocations is described.—The dominant-lethal dose-response curve, plotted on the basis of living embryos as a percentage of the control value, is clearly not linear as it is markedly concave downward. Similarly, the translocation dose-response curve showed a more rapid increase in the number of translocations with dose than would be expected on the basis of dose-square kinetics. It is clear for both of these endpoints that the effectiveness of EMS in inducing chromosome breakage is proportionately much lower at low doses.  相似文献

The clastogenic effect of the anticancer drug cis-diamminedichloroplatinum (II) (cisplatin) on meiotic prophase in primary spermatocytes and on spermatogonial stem cells of male (101/E1 x C3H/E1)F1 mice was studied. The intraperitoneal doses of cisplatin tested were 5.0, 7.5 and 10.0 mg/kg. Chromosomal aberrations were examined at diakinesis-metaphase 1 of meiosis 1-13 days after treatment, representing cells treated at diplotene, pachytene, zygotene, leptotene an preleptotene. Reciprocal translocations were evaluated 63-70 days after treatment, representing treated stem-cell spermatogonia. Cisplatin had a toxic effect in zygotene to preleptotene of meiosis, as indicated by the significant reduction in testicular weight. At diplotene, pachytene and zygotene no enhancement of aberrations was found. An increase in aberrant cells was observed during leptotene with preleptotene being the most sensitive stage. The dose-response relationship for aberrant cells was linear on day 13 after treatment. It is concluded that, like mitomycin C (Adler, 1976), cisplatin primarily caused aberrations during the premeiotic phase of DNA synthesis. No significant increase of translocation multivalents was found after treatment of stem-cell spermatogonia.  相似文献

目的: 观察小剂量辣椒素(Cap)抗小鼠肺纤维化的作用是否与其激动瞬时电位感受器香草酸受体1(TRPV1)有关。方法: 实验设对照(CON)组、博莱霉素(BLM)组、Cap(0.5、1、2 mg/kg)剂量组及Cap (2 mg/kg)+ TRPV1受体阻断剂SB-452533(2.5 mg/kg)剂量组,每组n=12。BLM(3.5 mg/kg)气管注射诱导肺纤维化小鼠模型。造模后连续皮下注射给药21 d。血浆降钙素基因相关肽(CGRP)浓度采用ELISA法快速测定。肺组织病理变化和胶原沉积分别采用HE染色、Masson染色和免疫组化进行检测。肺组织α-CGRP、β-CGRP、I 型胶原(collagen I)、III 型胶原(collagen III)、E钙粘蛋白(E-Cadherin)、紧密连接蛋白-1(ZO-1)、波形蛋白(Vimentin)、α-平滑肌肌动蛋白(α-SMA)、TRPV1、磷酸化ERK1/2(p-ERK1/2)和真核翻译起始因子3a(eIF3a)mRNA及蛋白表达分别采用qPCR和(或)Western blot法检测。结果: 与BLM组相比,小剂量Cap给药21 d后,病理检查发现小鼠肺纤维化明显减轻;肺组织胶原表达明显下降(P<0.05或P<0.01);肺泡上皮细胞上皮间质转分化(EMT)明显受到抑制(E-Cadherin和ZO-1的表达明显升高(P<0.05或P<0.01)而Vimentin和α-SMA的表达明显降低(P<0.05或P<0.01));TRPV1和CGRP表达水平明显升高(P<0.05或P<0.01)而ERK磷酸化水平和eIF3a表达水平明显降低(P<0.05或P<0.01)。但当使用TRPV1受体阻断剂阻断TRPV1后,小剂量Cap逆转肺泡EMT、改善肺纤维化的作用被显著取消,同时CGRP表达水平又明显降低(P<0.01)而p-ERK1/2和eIF3a的表达水平又明显升高(P<0.01)。结论: 小剂量Cap能够逆转肺泡上皮细胞EMT、缓解肺纤维化。其机制可能与其激动TRPV1、促进CGRP释放,进而抑制ERK磷酸化和下调eIF3a的表达有关。  相似文献

目的:探讨表皮生长因子受体(EGFR)在肺内的表达对博莱霉素(BLM)诱导小鼠肺纤维化中上皮-间质转分化的影响。方法:将40只4~6周龄C57BLB/c雄性小鼠随机分为正常对照组(气管滴入PBS),纤维化组(气管滴入BLM 3 mg/kg),EGFRRNAi组(气管滴入BLM 3 mg/kg+气管滴入siRNA 20μl)和RNAi阴性对照组(气管滴入BLM 3 mg/kg+气管滴入siRNA阴性对照20μl)。实验第10天处死小鼠,收获肺组织,检测羟脯氨酸含量;采用逆转录-聚合酶链反应(RT-PCR)法检测EGFR和α平滑肌肌动蛋白(α-SMA)mRNA的表达;肺组织切片行HE染色观察肺组织病理改变,免疫组化染色检测EGFR和α-SMA表达。结果:纤维化组EGFR和α-SMA两者的mRNA和蛋白表达均较正常对照组显著增加;RNAi组肺病理损伤较纤维化组减轻,气道上皮下胶原沉积及肺羟脯氨酸含量减少(P<0.05),肺组织EGFR和α-SMA两者的mRNA和蛋白表达均较纤维化组显著下降(P<0.05)。结论:在博来霉素诱导的肺纤维化中EGFR RNAi抑制EGFR活化,下调α-SMA的表达,减轻了博莱霉素诱导的肺纤维化病理改变。其抑制肺纤维化病理过程可能与其抑制上皮-间质转分化(EMT)有关。  相似文献

The experiments reported in this paper were designed to answer some questions relating to the augmenting effect of storage of sperm in the inseminated female, on the frequency of translocations in spermatozoa treated with the trifunctional alkylating agent trenimon. To see whether, upon storage, more chromosome breaks become available for interaction, sperm cells that had been treated with trenimon in the male were exposed to X-irradiation before or after 6 days storage in the female. The data of the first experiment indicated that in unstored sperm the translocation yield after treatment with both trenimon and X-rays, was lower than that expected on the basis of additivity of yields of the single treatments. The negative synergism between trenimon and X-irradiation has been confirmed in further experiments with both translocations and dominant lethals. The latter finding does not support an interpretation in terms of selective elimination of translocationsby cell death. Following storage, the translocation frequencies increase and after combination of trenimon and X-rays, yields corresponding to additivity of frequencies with single treatments are observed.To study whether changes in the maternal repair system contribute to the storage effect, trenimon-treated males were mated with aged females and the frequencies of translocations were determined. It was found that these frequencies were similar to those in young (unstored) females; this result suggests that the possibility raised above is unlikely.  相似文献

目的探讨博来霉素诱导小鼠肺纤维化最佳剂量和方法。方法 126只8周龄雄性ICR小鼠,随机分成一次性大剂量模型和多次小剂量模型。一次性大剂量模型分为200 mg/(kg.bw)BLM组、150 mg/(kg.bw)BLM组、100 mg/(kg.bw)BLM组及阴性对照组(DN组),每组18只,分别经尾静脉一次性注射BLM 200、150、100mg/(kg.bw)及生理盐水10 mL/(kg.bw),各组分别于第7、14、21天各处死6只。多次小剂量模型分为每日10 mg/(kg.bw)BLM组及阴性对照组(N组),分别经尾静脉注射BLM 10 mg/(kg.bw)及生理盐水10 mL/(kg.bw),每天1次,连续注射14 d,两组分别于第14、21、28天各处死6只。留取肺组织,观察肺组织病理改变,检测Ⅲ型胶原的含量,观察小鼠体重及生存率。结果①在一次性大剂量模型中,BLM各剂量组肺泡炎症评分及肺纤维化评分与正常组相比,除100 mg/(kg.bw)BLM组和150 mg/(kg.bw)BLM组在第7天的模型差异无显著性外(P>0.05),其余各组差异均有显著性(P<0.05);各个剂量组Ⅲ型胶原的表达面积与正常组相比,除100 mg/(kg.bw)BLM组在第7天的模型差异无显著性外(P>0.05),其余各组均较正常组高(P<0.05),各个剂量组分别在第21天达到高峰,以200 mg/(kg.bw)BLM组第21天组Ⅲ型胶原的表达面积最高;该模型小鼠各剂量组死亡率为0。②在多次小剂量模型中,各组的肺泡炎症与肺纤维化程度与正常组相比差异均有显著性(P<0.05);各组Ⅲ型胶原的表达也均高于正常组(P<0.05),且随着时间的延长呈进行性增加,在第28天达到高峰;该模型小鼠共死亡11只,死亡率为30.56%。结论在本实验中,以尾静脉一次性注射BLM 200 mg/(kg.bw)后第21天诱导建立的ICR小鼠肺纤维化模型成模最好,其小鼠死亡率低,操作简单,有效安全方便的特点使之有希望成为一种复制肺纤维化的理想模型。  相似文献

We studied engraftment in a murine model of allogeneic bone marrow (BM) transplantation. Recipient C57BL/6 (H-2b) mice were conditioned with single-dose (9 or 7.5 Gy) total body irradiation (TBI), fractionated (4 X 3.3 Gy) TBI, hyperfractionated (8 X 1.65 Gy) TBI, 2 X 120 mg/kg cyclophosphamide (CY) followed by 7.5 Gy TBI, or 300 mg/kg CY followed by 9 Gy total lymphoid irradiation (TLI). Conditioned mice were transplanted with BALB/c (H-2d) BM supplemented with splenocytes (BMS) to facilitate graft-vs-host disease (GVHD). Ex vivo T cell depletion of the BMS with anti-Thy-1.2 antibody and complement protected recipients from lethal GVHD. Engraftment was measured in transplanted animals by serotyping peripheral blood mononuclear cells with anti-H-2-specific antibodies and complement. Mice that were given a T cell-depleted BMS transplant after conditioning with 9 Gy TBI, fractionated TBI, or CY plus TBI showed a 99 to 100% incidence of engraftment. However, if the T cell-depleted graft was given to mice conditioned with hyperfractionated TBI, 7.5 Gy TBI, or CY plus TLI, only 3 to 32% of the animals engrafted. BM which was not T cell-depleted engrafted in 63 to 100% of the mice regardless of the conditioning used. Nonengrafted mice tested with anti-host type antibody demonstrated autologous recovery. We conclude that engraftment or failure/rejection of BM in transplanted mice is determined in part by a dynamic equilibrium between T cells present in the donor graft and the surviving hemopoietic cells in the conditioned recipient. More intensive conditioning of the recipient allows engraftment of T cell-depleted, mismatched BMS. Such conditioning is not limited to a single modality, but can be achieved with single-dose TBI, fractionated TBI, or with TBI combined with CY. These findings have timely and important implications for the current understanding of engraftment in human allogeneic BM transplantation following T cell depletion.  相似文献

Although WR-2721, S-2-(3-aminopropylamino)ethylphosphorothioic acid, is an effective radioprotector, its use is limited by its toxicity. Combining WR-2721 with other agents might decrease its toxicity and/or increase its effectiveness. The effect of selenium (Se) pretreatment on the acute toxicity and radioprotective effect of WR-2721 was studied in male CD2F1 mice. Injection of 1.6mg/kg Se 24 hr before WR-2721 (800-1200 mg/kg, IP) decreased the lethality of WR-2721 significantly. Lower doses of Se were also effective, but simultaneous administration was not effective. Se injection alone (1.6 mg/kg) 24 hr before cobalt-60 irradiation increased the survival (dose reduction factor, DRF = 1.1) significantly. A synergistic effect on post-irradiation survival was observed when Se was injected 24 hr before WR-2721 (200-600 mg/ kg IP before irradiation). For example, after exposure to 22 Gy (1 Gy/min), 30-day survival was 100% when mice were treated with both Se and 600mg/kg WR-2721, and was 13% with WR-2721 alone. The DRF after 400 mg/kg WR-2721 was 2.6 with Se compared to 2.2 without Se pretreatment. Alkaline phosphatase activity in bone marrow cells and serum was significantly depressed after treatment with 1.6 mg/kg Se, suggesting that a retardation of conversion of WR-2721 to its active free sulfhydryl form through the action of alkaline phosphatase might be partly responsible for the effects of Se. Other possible mechanisms related to the antioxidant properties of Se are under investigation.  相似文献

Reciprocal translocations were evaluated 63–70 days after treatment, representing treated stem-cell spermatogonia.Cisplatin had a toxic effect in zygotene to preleptotene of meiosis, as indicated by the significant reduction in testicular weight. At diplotene, pachytene and zygotene no enhancement of aberrations was found. An increase in aberrant cells was observed during leptotene with preleptotene being the most sensitive stage. The dose-response relationship for aberrant cells was linear on day 13 after treatment. It is concluded that, like mitomycin C (Alder, 1976), cisplatin primarily caused aberrations during the premeiotic phase of DNA synthesis. No significant increase of translocation multivalents was found after treatment of stem-cell spermatogonia.  相似文献

The radioprotective properties of indralin when it is used in combination with cystamine and mexamine are studied in inbred mice and rats. The mice and rats are irradiated with γ rays emitted by 60Co at doses of 9.0 and 9.5 Gy, respectively. A combined parenteral administration of indralin and cystamine in mice at doses of 25 mg/kg each is revealed to potentiate the radioprotective properties of indralin up to a level close to the ED50 effect, while the separate application of these drugs in doses of 25 mg/kg each has no or a very weak radioprotective effect. Indralin (50 mg/kg) and mexamine (12 mg/kg) injected intraperitoneally in rats are found to almost completely eliminate the animal mortality caused by gastrointestinal acute radiation syndrome; the mortality in the control radiation group reaches 60% on the seventh day after the animals have been exposed to radiation at a dose of 9.5 Gy. However, if bone-marrow acute radiation syndrome develops under the above condition of super-lethal dose, the radioprotectors have a low radioprotective effect. Under the this condition, the combined application of indralin and mexamine in the same doses has 50% of radioprotective effect reached by applying these radioprotectors separately in double doses.  相似文献
