Abstract

The first controlled excavation of a Clovis site in Iowa revealed a concentration of complete and fragmentary Clovis Fluted projectile points. The site, in eastern Iowa, is interpreted as a projectile point cache destroyed by agricultural implements. Abstract

Canonical analysis is used to examine intracemetery variation in cranial morphology in the Sully site Arikara skeletal collection. The site included four spatially distinct burial areas. lnterarea morphological differences in the crania are shown to relate to the multicomponent nature of the associated village. In 1970, a controlled surface collection was carried out at site 24 PW 320 in the Avon Valley of Montana. Lithic samples were obtained from two areas within the site. Clitopilus canariensis is described as a new species belonging to Clitopilus subgenus Rhodophana on the basis of a collection from La Palma, Canary Islands. BackgroundGuidelines recommend treatment with 4–5 days of granulocyte colony-stimulating factor (G-CSF) for optimal donor peripheral blood progenitor cell (PBPC) mobilization followed by day 5 collection. Given that some autologous transplant recipients achieve adequate collection by day 4 and the possibility that some allogeneic donors may maximally mobilize PBPC before day 5, a feasibility study was performed evaluating day 4 allogeneic PBPC collection.MethodsHLA-matched sibling donors underwent collection on day 4 of G-CSF for peripheral blood (PB) CD34+ counts ≥0.04 × 106/mL, otherwise they underwent collection on day 5. Those with inadequate collected CD34+ cells/kg recipient weight underwent repeat collection over 2 days. Transplant and PBPC characteristics and cost analysis were compared with a historical cohort collected on day 5 per our prior institutional algorithm.ResultsOf the 101 patient/donor pairs, 50 (49.5%) had adequate PBPC collection on day 4, with a median PB CD34+ cell count of 0.06 × 106/mL. Day 4 donors were more likely to develop bone pain and require analgesics. Median collected CD34+ count was significantly greater, whereas total nucleated, mononuclear and CD3+ cell counts were significantly lower, at time of transplant infusion for day 4 versus other collection cohorts. There were no significant differences in engraftment or graft-versus-host disease. Cost analysis revealed 6.7% direct cost savings for day 4 versus historical day 5 collection.DiscussionDay 4 PB CD34+ threshold of ≥0.04 × 106/mL identified donors with high likelihood of adequate PBPC collection. Day 4 may be the optimal day of collection for healthy donors, without adverse effect on recipient transplant outcomes and with expected cost savings.  相似文献

Background aimsThe ability to predict how many CD34+ cells a donor will collect on a given day is vital for efficient leukapheresis.MethodsWe validated a formula to predict daily CD34+ cell collections by leukapheresis, calculated as follows: (peripheral blood CD34+ cells/L) × (adjusted collection efficiency of 30%)/body weight (kg), multiplied by the number of liters processed. This validation was performed from 234 donors undergoing 30 L large volume leukapheresis (LVL) and 162 donors undergoing smaller collections (non-LVL). The LVL group consisted of 811 collection events (625 multiple myeloma, 186 non-myeloma). The non-LVL group consisted of 224 collection events (196 multiple myeloma, 28 non-myeloma). All predicted and observed CD34+ cell collection numbers were plotted (predicted versus observed) and assessed using linear regression analyses. Linear correlation coefficients (r-values), slopes and intercepts of the regression lines were evaluated.ResultsPredicted versus observed data points across all quantities of CD34+ cells/kg collected by both LVL and non-LVL had strong r-values of 0.947 and 0.913, respectively, demonstrating near perfect positive linear correlations. Data for LVL collections subgrouped by number of cells collected (poor, intermediate and good), mobilization regimen, collection day and diagnosis were analyzed the same way and showed consistent findings.ConclusionsWe have validated a formula with a strong ability to predict collection of CD34+ cells/kg that would allow for individualization of collection for any donor once the peripheral blood CD34+ cell count and optimal goal of collection were known; to date this has not been published by other groups.  相似文献

Background aimsUmbilical cord blood is an established source of stem cells in patients with hematologic malignancies who do not have HLA-compatible matched related or unrelated donors. The success of an umbilical cord blood transplant depends on the dose of total nucleated and CD34+ cells infused. Therefore, collecting, banking and listing high-quality cord blood units with high total nucleated and CD34+ cell dose are essential.MethodsHere the authors describe their cord blood bank's novel collection technique, which involves both in utero and ex utero collection of a single cord blood unit. The authors also evaluated maternal, neonatal and collection parameters that may impact the cell dose.ResultsMaternal gestational age and race, and neonatal weight and sex correlated with the total nucleated cell dose.ConclusionsThe optimized collection of umbilical cord blood is critical for its use as a source of stem cells for transplantation.  相似文献

Abstract

A small collection of obsidian and chalcedony microblades, from a site near Calgary, Alberta, is described and compared with other microblades from the Plains and from the Plateau. Although the age and cultural affiliations of the micro blades are not certain, there is some evidence to suggest that they are early in the Plains sequence. The microblades have some distinctive attributes including ridge flakes, for which a typology is suggested. A comparison of the High River microblades with a large sample from the Interior Plateau of British Columbia suggests that the stimulus for the Alberta micro blades did not derive from the Plateau.  相似文献

Abstract

In 1976 Alfred E. Johnson proposed a subsistencesettlement model for Kansas City Hopewell. The model consisted of a subsistence territory encompassirig a drainage upon which a large permanent village settlement was located near its mouth with a series of smaller specialized ancillary sites located up from the village. Limited corn and squash horticulture would be present in the latter half of the occupation too. A test of such a model would be actual limited activity sites. The Yeo site, a late kansas City Hopewell site, is a hickory nut/marsh elder seed collection/storage station. It is exactly the type of site necessary to substantiate such a model. Also, such a test implies the testing of the notion that there are “specialized, limited activity sites,” sites which are important to some of the suppositions of the New Archaeology. Additionally, the presence of the combination of hickory/cultivated marsh elder (Iva annua var. macrocarpa) utilization within the context of an upland oak-history forest may be interpreted as supporting Asch and Asch’s (1978) model of marsh elder domestication.  相似文献

Background

Metabolomics provides measurement of numerous metabolites in human samples, which can be a useful tool in clinical research. Blood and urine are regarded as preferred subjects of study because of their minimally invasive collection and simple preprocessing methods. Adhering to standard operating procedures is an essential factor in ensuring excellent sample quality and reliable results.

Aim of review

In this review, we summarize the studies about the impacts of various preprocessing factors on metabolomics studies involving clinical blood and urine samples in order to provide guidance for sample collection and preprocessing.

Key scientific concepts of review

Clinical information is important for sample grouping and data analysis which deserves attention before sample collection. Plasma and serum as well as urine samples are appropriate for metabolomics analysis. Collection tubes, hemolysis, delay at room temperature, and freeze–thaw cycles may affect metabolic profiles of blood samples. Collection time, time between sampling and examination, contamination, normalization strategies, and storage conditions may alter analysis results of urine samples. Taking these collection and preprocessing factors into account, this review provides suggestions of standard sample preprocessing.

Genetic diversity of Poa annua in western Oregon grass seed crops   总被引:3,自引:0,他引:3  
The genetic diversity of Poa annua L.populations collected from western Oregon grass-seed fields was surveyed using 18 randomly amplified polymorphic DNA (RAPD) markers. Markers from 1357 individual plants from 47 populations collected at three sampling dates (fall, winter, and spring) for 16 sites were used to measure genetic diversity within and among populations. Site histories varied from low to high herbicide selection pressure, and some sites were subdivided by 3 years of differing post-harvest residue management. Gene diversity statistics, simple frequency of haplotype occurrence, and analysis of molecular variance (AMOVA) revealed the presence of significant variability in P. annua among sites, among collection dates within sites, and within collection dates. Nei gene-diversity statistics and population-differentiation parameters indicated that P. annua populations were highly diverse. Mean Nei gene diversity (h) for all 47 populations was 0.241 and total diversity (HT) was 0.245. A greater proportion of this diversity, however, was within (HS=0.209) rather than among (GST=0.146) populations. When populations were grouped by season of collection, within-group diversity was HS=0.241, while among-group diversity was GST=0.017. When populations were grouped by site, within-group diversity was HS=0.224, while among-group diversity was GST=0.087. The diversity among populations within season for fall, winter, and spring collections was GST=0.121, 0.142, and 0.133, respectively. Populations collected from fields with histories of high herbicide selection pressure showed low differentiation among collection dates, with GST as low as 0.016, whereas those collected from fields with low herbicide selection pressure showed greater differentiation among collection dates, with GST as high as 0.125. At high selection-pressure sites, populations were also lower in gene diversity (as low as h=0.155), while at low selection-pressure sites there was higher gene diversity (as high as h=0.286). The site to site variability was greater for the high selection-pressure sites (GST=0.107 or 69% of the total among-population variance), while the season of germination variability was greater at sites of low herbicide-selection pressure (GST=0.067, or 70% of the total among-population variance). High initial diversity coupled with a long-term re-supply of genotypes from the seed bank must have been factors in maintaining the genetic diversity of this weed despite the intensive use of herbicides. Knowledge of the genetic diversity of Willamette Valley P. annua should help in formulating more effective strategies for managing this weed. Received: 24 July 1999 / 11 November 1999  相似文献

Abstract

The results of two years of collection in a small isle in the southern Sardinia are reported, consisting in a floristic list of 116 entities distributed in 96 genera and 41 families. The biological spectrum of this flora puts in evidence a typical Mediterranean environment, characterized by a marked summer dryness. The ratio: number of entities/surface of the studied site has been compared with that of other small southern Sardinian islands, resulting the highest value. This floristic aspect, as well as differences in the biological spectrum, is interpreted as the result of the presence in the area under study of a desultory link with the mainland, in form of a sandy isthmus. This seems important in breaking the biological balance of the small island.  相似文献

ObjectiveThis paper aims to determine the cost of establishing and sustaining a verbal-autopsy based mortality surveillance system in rural India.ResultsVerbal-autopsies were completed for 96.7% (5786) of all deaths (5895) recorded. The annual cost in year 1 was INR 1,133,491 (USD 24,943) and the total cost per death was INR 757 (USD 16.66). These costs included training of NPHWs and physician reviewers Rs 67,025 (USD 1474), data collection INR 248,400 (USD 5466), dual physician review for cause of death assignment INR 375,000 (USD 8252), and project management INR 341,724 (USD 7520). The average annual cost to run the system each year was INR 822,717 (USD18104) and the cost per death was INR 549 (USD 12) for the next 3 years. Costs were reduced by using single physician review and shortened re-training sessions. The annual cost of running a surveillance system was INR 900,410 (USD 19814).DiscussionThis study provides detailed empirical evidence of the costs involved in running a mortality surveillance site using verbal-autopsy.  相似文献

Background aimsLittle is known of the effect of anticoagulation on peripheral blood progenitor cell (PBPC) harvest during large-volume leukapheresis (LVL). Because of the interaction of heparin with stromal cell-derived factor (SDF)-1α, it has been proposed that a heparin-based anticoagulation may result in an increased PBPC collection efficiency compared with standard citrate-based anticoagulation.MethodsWe conducted a prospective randomized trial to address the effect of both anticoagulation regimes on safety, subjective comfort and CD34 + collection efficiency in 90 adult patients undergoing standardized LVL. Anticoagulation consisted of either citrate (group C) or a combination of heparin and low-dose citrate (group H).ResultsThe overall incidence of adverse reactions (AR) during LVL was 17%. AR consisted only of citrate-related AR; no bleeding complications were observed. Determination of parameters of the acid–base balance revealed a higher frequency of metabolic alkalosis in group C. Analysis of serum SDF-1α revealed no differences in SDF-1α plasma levels. There were no differences in the CD34 + cell collection efficiency, resulting in the harvest of equal CD34 + cell yields independent of the anticoagulation used.ConclusionsOur data show no clinical relevant effect of a heparin containing anticoagulation in terms of an increased overall CD34 + cell collection during LVL, although this regime shows some benefits in terms of the incidence and subjective tolerance towards AR. Based on our results the decision between a citrate- and heparin-substituted anticoagulation for LVL should be driven by patient-related factors, and should concern potential contraindications of both methods.  相似文献

Background aimsUmbilical cord blood (UCB) cells are a new alternative to bone marrow source for hematopoietic stem cell transplantation and their use has increased in the last decade. Thus efforts are being made to improve the umbilical cord blood unit's quality. Besides compatibility, other factors, such as the total nucleated cell (TNC) count and the percentage of CD34+ cells in the product, are very important for a successful transplant outcome. Our aim was to describe our donor population and assess the best cord blood collection technique at Hospital Israelita Albert Einstein's cord blood bank (São Paulo, Brazil).MethodsThis was a retrospective study in which all analyses were performed simultaneously. A Student's t-test was used for qualitative variables for non-matched samples. For qualitative analyses, we used either the chi-square test or the exact Fisher's test.ResultsThe stratification of the population characteristics allowed us to determine which ones had an impact on unit volume, TNC count and percentage of CD34+ cells. A significant correlation was observed between donor characteristics and the quality of UCB units as related to maternal and gestational age, type of pregnancy, route of delivery, cord blood collection technique and birth weight.ConclusionsWe found that cord blood collection technique and newborn weight were significantly correlated with the TNC content. The collection technique used at our institution significantly improved the UCB unit volume and consequently the TNC count. Some findings, such as the impact of maternal age and newborn weight, have led us to re-evaluate our protocol in order to achieve better results.  相似文献

PurposeLead free protective clothing can create a higher part of secondary radiation (SR) than products that are based on lead. Hence, the attenuation properties may be downgraded. The international measuring standard IEC 61331-1:2014 declares the “inverse broad beam geometry” (IBG) as standard method, which has recently been modified to IBG1 by the Physikalisch Technische Bundesanstalt (PTB). Because of the unspecific partial irradiation of the ionization chamber problems in the evaluation of lead equivalence values (LEVs) can occur. An alternative method proposed in this paper overcomes these problems.Materials and methodsThe alternative setup “modified broad beam geometry” (BBG1) was tested and compared to the IBG1 method by performing Monte Carlo simulations and radiation measurements including several lead-composite and lead-free protective materials.ResultsSimulations show a reduced collection efficiency of SR under IBG1 whereas BBG1 features a high degree of SR collection. Material samples with a high amount of SR can feature up to 8% higher LEVs compared to IBG1. For most of the currently salable materials the differences of BBG1 vs IBG1 amount to <3% (0.25 mm LEV) and <1% (0.50 mm LEV). In special cases the currently practiced method can lead to heavier protective clothings.ConclusionsThe proposed BBG1 setup meets the specifications of the IEC standard with respect to energy response and SR collection. The method should be implemented in the IEC standard.  相似文献

Background: Ecological surveys often aim to inform biodiversity conservation policy and practice. Survey types differ; all require clear aims and good design. A survey in Scotland was carried out to measure the distribution and health of juniper (Juniperus communis) and potential for its natural regeneration.

Aims: Explore if one-off national surveys can inform conservation action by examining, as an example, the Scottish juniper data set to: (1) determine site conditions favourable for juniper regeneration, (2) provide recommendations for site management for regeneration.

Methods: Associations between site conditions and juniper occurrence were investigated with principal component analysis (PCA) and using species distribution modelling (SDM). To identify changes in site conditions over time, we compared degree of site suitability for natural regeneration (PCA distance) and population age structure.

Results: Twenty-one per cent of the sites showed juniper regeneration. Compared to all juniper sites, conditions at regenerating sites were different (e.g. climatically wetter, lower site fertility). Change over time in conditions was suggested by patterns in population age structure.

Conclusions: Following the establishment of a juniper population, site conditions may change, becoming unsuitable for natural regeneration. Historical site information to confirm this is lacking. We cannot answer fundamental questions about the causal mechanisms of the natural regeneration of juniper and question whether one-off survey data can inform conservation management.  相似文献

BackgroundThe SecA DEAD (Asp-Glu-Ala-Asp) motor protein uses binding and hydrolysis of adenosine triphosphate (ATP) to push secretory proteins across the plasma membrane of bacteria. The reaction coordinate of nucleotide exchange is unclear at the atomic level of detail.MethodsWe performed multiple atomistic computations of the DEAD motor domain of SecA with different occupancies of the nucleotide and magnesium ion sites, for a total of ~1.7 μs simulation time. To characterize dynamics at the active site we analyzed hydrogen-bond networks.ResultsATP and ADP can bind spontaneously at the interface between the nucleotide binding domains, albeit at an intermediate binding site distinct from the native site. Binding of the nucleotide is facilitated by the presence of a magnesium ion close to the glutamic group of the conserved DEAD motif. In the absence of the magnesium ion, protein interactions of the ADP molecule are perturbed.ConclusionsA protein hydrogen-bond network whose dynamics couples to the occupancy of the magnesium ion site helps guide the nucleotide along the nucleotide exchange path. In SecA, release of magnesium might be required to destabilize the ADP binding site prior to release of the nucleotide.General significanceWe identified dynamic hydrogen-bond networks that help control nucleotide exchange in SecA, and stabilize ADP at an intermediate site that could explain slow release. The reaction coordinate of the protein motor involves complex rearrangements of a hydrogen-bond network at the active site, with perturbation of the magnesium ion site likely occurring prior to the release of ADP.  相似文献
