ABSTRACT

Introduction: The last decade has yielded significant developments in the field of proteomics, especially in mass spectrometry (MS) and data analysis tools. In particular, a shift from gel-based to MS-based proteomics has been observed, thereby providing a platform with which to construct proteome atlases for all life forms. Nevertheless, the analysis of plant proteomes, especially those of samples that contain high-abundance proteins (HAPs), such as soybean seeds, remains challenging.

Areas covered: Here, we review recent progress in soybean seed proteomics and highlight advances in HAPs depletion methods and peptide pre-fractionation, identification, and quantification methods. We also suggest a pipeline for future proteomic analysis, in order to increase the dynamic coverage of the soybean seed proteome.

Expert opinion: Because HAPs limit the dynamic resolution of the soybean seed proteome, the depletion of HAPs is a prerequisite of high-throughput proteome analysis, and owing to the use of two-dimensional gel electrophoresis-based proteomic approaches, few soybean seed proteins have been identified or characterized. Recent advances in proteomic technologies, which have significantly increased the proteome coverage of other plants, could be used to overcome the current complexity and limitation of soybean seed proteomics.  相似文献

Recent rescue excavations at Bercy (Paris), a site on the alluvial plain of the Seine valley, yielded plant remains which are associated with the recent occupation phase dating from the middle Neolithic II (Chasséen), when a village was established on the former channel of the river Seine. Various contexts (the channel, the flat lower part of the bank, and several archaeological features) have been studied and 84 taxa have been identified. Cultivated plants are represented by Triticum aestivum/durum, T. dicoccum, T. monococcum and Hordeum vulgare. Among the wild plants with potentially edible fruits or seeds, only very few satisfy the various criteria for association with human activities: Corylus avellana, Vitis sylvestris, Cornus sanguinea, Quercus sp. and Prunus spinosa of which carbonized fruits were also present, were found in archaeological features and were very abundant. Though not found in archaeological features, we consider that Crataegus monogyna (carbonized and well represented) and Rubus spp. (especially abundant) were not deposited there naturally and had also been intentionally collected. Finally, it is suggested that the allochthonous (varied) origin of these taxa is the reason why there are no concentrations of their fruits in the channel. The exploitation of wild seeds and fruits appears to have been very selective. All other wild taxa can be attributed to natural deposition.  相似文献

Capsule Seeds of plants from lower salt marsh communities are preferred, with insects less important.

Methods Droppings of Shorelarks Eremophila alpestris, Snow Buntings Plectrophenax nivalis and Twites Carduelis flavirostris sampled in the German Wadden Sea were analysed and compared with food abundance to assess preferences.

Results Shorelarks prefer seeds of Salicornia sp., Suaeda maritima, Atriplex sp., Halimione portulacoides and unidentified small grass seeds. Insects are eaten mainly in periods of seed shortage, but are consumed in smaller amounts during the winter. The food composition of Snow Buntings is very similar, but additionally Triglochin maritimum is commonly eaten. Twites are specialized on seeds of Salicornia sp. and Suaeda maritima and rarely ingest other seeds and insects. All seeds consumed were of plants from lower salt marsh communities. Many halophyte seeds, and especially those that birds feed on, are rich in energy. Large seeds and those which need a long handling time are avoided.

Conclusion Changes in the lower salt marshes of the Wadden Sea by embankments and intensified grazing might have been responsible for the rapid population decline from the 1960s to 1980s.  相似文献

Seed predation and dispersal by scatter-hoarding rodents are key processes that determine seed survival, and thus, plant regeneration within forests. For decades, there has been much debate on the important effects of seed size (one of the most important seed traits) on rodent foraging preference. Furthermore, the possible selective forces in the evolution of seed size may be influenced by primary selectivity and how rodents treat seeds after harvesting. In this study, different-sized seeds from four species (Pinus armandii, Pinus densata, Abies sp., and Viburnum sp.) harvested by scatter-hoarding rodents were studied in an alpine forest in Southwestern China for two consecutive years. Our results showed that seed size influenced rodent foraging preferences, with bigger seeds being preferred over smaller seeds, within and across species. Rodents only removed and cached the larger seeds of P. armandii, and ate the seeds of the other three species in situ. Rodents are purely seed predators for these three species. For the cached seeds of P. armandii, significantly positive correlations were observed between seed size and dispersal distance among both primary and secondary cached seeds in 2006, but not in 2005. Our results indicate that among many coexisting species with widely different-sized seeds, scatter-hoarding rodents played important roles in the seed dispersal of the big-seeded species alone. This caching behavior could offset the limited seed dispersal of large-seeded and wingless species (P. armandii), in comparison with that of small winged seed species (P. densata and Abies sp.) and frugivore-dispersed species (Viburnum sp.).  相似文献

Plant‐associated bacteria can have beneficial effects on the growth and health of their host. Nevertheless, the role of endophytic bacteria present in seeds has not been investigated in depth. In this study, the cultivable endophytic population of seeds from Arabidopsis thaliana exposed to 2 μm cadmium for several generations (Cd seeds) was compared with a population isolated from seeds of plants that were never exposed to Cd (control seeds). We observed obvious differences between the two types of seed concerning genera present and phenotypic characteristics of the different isolates. Sinorhizobium sp. and Micrococcus sp. were only found in control seeds, while Pseudomonas sp., Bosea sp. and Paenibacillus sp. were only found in Cd seeds. Sphingomonas sp., Rhizobium sp., Acidovorax sp., Variovorax sp., Methylobacterium sp., Bacillus sp. and Staphylococcus sp. occurred in varying numbers in both types of seed. Metal tolerance and 1‐aminocyclopropane‐1‐carboxylate deaminase activity were predominantly found in strains isolated from Cd seeds, while the production of siderophores, indole‐3‐acetic acid and organic acids was more prevalent in endophytes isolated from control seeds. These data support the hypothesis that certain endophytes are selected for transfer to the next generation and that their presence might be important for subsequent germination and early seedling development.  相似文献

Capsule Large buntings prefer cereal grains whilst sparrows also take oily seeds.

Aims To determine seed food preferences of Tree Sparrow Passer montanus and Corn Bunting Emberiza calandra.

Methods Forty seed choice experiments were conducted at two sites over two winters. In each experiment, two seed types were provided and the number of visits made by birds to each type was recorded over a set period. At one site, Tree Sparrows were colour-ringed, allowing choices made by individual birds to be recorded. Data were also collected for House Sparrow Passer domesticus, Yellowhammer Emberiza citrinella and Reed Bunting E. schoeniclus.

Results All five species fed intensively on cereal grain, and wheat and oats were consistently preferred to barley. Sparrows and Reed Buntings also took maize, which was avoided by Corn Bunting and Yellowhammer. Tree Sparrow exhibited a broad diet, selecting cereal grain and oily seeds including sunflower and oilseed rape, but rye-grass seed was almost completely avoided.

Conclusion Cereal grain should be a key component of over-winter provision of seed for farmland passerines, especially when targeted at Corn Bunting and Yellowhammer. Oily seeds such as brassicas and sunflower will benefit species with more generalist diets, including Tree Sparrows.  相似文献

Full analysis of eight seed samples collected in the 1960's excavations at Neolithic Çatalhöyük East, Turkey, is presented. Detailed investigation of the composition and context of the samples suggests that the Neolithic population collected, processed and stored seeds from Capsella sp. and Descurainia sp. (wild crucifers) for food use. In addition seeds of Vicia/Lathyrus sp. (wild vetch), Helianthemum spp. and Taeniatherum caput-medusae mixed with Eremopyrum type (grasses) were also found, some of which may have been used for food or other purposes. The analysis demonstrates that wild seed exploitation was a regular part of subsistence practice alongside the economic staple of crop production, and again demonstrates how diverse plant use practices were at the site.  相似文献

Five caesalpinioid legumes, Afzelia africana, Afzelia bella, Anthonotha macrophylla, Cryptosepalum tetraphylum and Paramacrolobium coeruleum, and one Euphorbiaceae species, Uapaca somon, with a considerable range in seed sizes, exhibited different responses to inoculation by four species of ectomycorrhizal (ECM) fungi, Scleroderma dictyosporum, S. verrucosum, Pisolithus sp. and one thelephoroid sp. in greenhouse conditions. Thelephoroid sp. efficiently colonized seedlings of all of the five caesalpinioid legumes except U. somon, but provided no more growth benefit than the other fungi. Thelephoroid sp. and S. dictyosporum colonized seedlings of U. somon poorly, but stimulated plant growth more than the other fungi. The relative mycorrhizal dependency (RMD) values of the caesalpinioid legumes were never higher than 50%, whilst U. somon had RMD values ranging from 84.6 to 88.6%, irrespective of the fungal species. The RMD values were negatively related to seed mass for all plant species. Potassium concentrations in leaves were more closely related than phosphorus to the stimulation of seedling biomass production by the ECM fungi. Our data support the hypothesis that African caesalpinioid legumes and euphorbe tree species with smaller seeds show higher RMD values than those with the larger seeds.  相似文献

Post‐dispersal seed predation and endozoochorous seed dispersal are two antagonistic processes in relation to plant recruitment, but rely on similar preconditions such as feeding behavior of seed consumers and seed traits. In agricultural landscapes, rodents are considered important seed predators, thereby potentially providing regulating ecosystem services in terms of biological weed control. However, their potential to disperse seeds endozoochorously is largely unknown. We exposed seeds of arable plant species with different seed traits (seed weight, nutrient content) and different Red List status in an experimental rye field and assessed seed removal by rodents. In a complementary laboratory experiment, consumption rates, feeding preferences, and potential endozoochory by two vole species (Microtus arvalis and Myodes glareolus) were tested. Seed consumption by rodents after 24 h was 35% in the field and 90% in the laboratory. Both vole species preferred nutrient‐rich over nutrient‐poor seeds and M. glareolus further preferred light over heavy seeds and seeds of common over those of endangered plants. Endozoochory by voles could be neglected for all tested plant species as no seeds germinated, and only few intact seeds could be retrieved from feces. Synthesis and applications. Our results suggest that voles can provide regulating services in agricultural landscapes by depleting the seed shadow of weeds, rather than facilitating plant recruitment by endozoochory. In the laboratory, endangered arable plants were less preferred by voles than noxious weeds, and thus, our results provide implications for seed choice in restoration approaches. However, other factors such as seed and predator densities need to be taken into account to reliably predict the impact of rodents on the seed fate of arable plants.  相似文献

Some of the most conspicuous fruit and seed remains from the Middle Eocene Princeton chert locality (Allenby Formation) are assignable to the Araceae. Several thousand campylotropous, reniform, spiny seeds 2.5–3.2 mm long x 1.8–2.3 mm wide have been found dispersed in the chert. Two fruits, each with a single locule and at least eight ovules, show three distinct wall layers and appear to have been fleshy. The two layers of seed integument have pitted isodiametric sclereids aligned in radial rows, the outermost being thinner walled. Spines are borne in three rows on the dorsal seed surface while the ventral side is flattened. Idioblasts that probably contained raphides are present in the outer integument. Nucellar tissue is attached to the integument for most of its length and is well-preserved in some seeds near the chalaza and in the large conical shaped area beneath the micropyle. Endosperm cells with dark contents and curved, linear, monocotyledonous embryos are present in some seeds. Sections of seeds of living araceous taxa of the subfamilies Monsteroideae (Epipremnum, Rhodospatha) and Lasioideae (Urospatha, Cyrtosperma) were made for comparison. The fossil seeds represent a new taxon most closely related to Cyrtosperma, Keratosperma allenbyensis Cevallos-Ferriz et Stockey gen. et sp. nov., Family: Araceae, Subfamily: Lasioideae, Tribe: Lasieae. The Princeton remains are the oldest described seeds of this tribe known to date and add to our knowledge of the subtropical elements of the Princeton flora.  相似文献

Paddock‐scale Acacia nilotica L. Willd. ex Del. (prickly acacia) spatial distribution, seed production and dispersal patterns were investigated in the Astrebla (Mitchell) grasslands of northern Australia as a step toward predicting future patterns of invasion. A number of hypotheses were tested based on what we know of this species in both Australia and regions where it is native, for example South Africa. It was hypothesized that most A. nilotica seeds would be produced by trees in riparian habitats with access to permanent water. In addition, we predicted that seeds would be dispersed throughout the Astrebla grassland landscape by cattle, following observations that cattle readily ingest and pass seeds and that cattle have access to all areas within paddock boundary fences. Tree density, seed production and seed dispersal by cattle were measured along a series of transects from permanent watering points to paddock boundary fences. Trees associated with permanent watering points produced more seeds per unit area and occurred at higher density than their non‐riparian counterparts. The importance of riparian trees decreased in years with high rainfall and in paddocks with only small areas of riparian habitat. Cattle spread dung and seeds throughout paddocks, with peaks of deposition adjacent to permanent watering points. These results suggest that invasion patterns are likely to be uneven across the landscape and may be reactive to climate. High seedling recruitment and possible thicket formation is expected adjacent to permanent watering points and wherever cattle congregate. Patterns of recruitment in non‐riparian areas are likely to be relatively sparse. The importance of post‐dispersal factors in determining recruitment patterns is discussed.  相似文献

A study of the seed morphology of Erica tetralix and the related species E. mackaiana and E. andevalensis was performed to evaluate the taxonomic importance of seed characters. Size and shape of seeds and surface cells were studied, and area, perimeter, main axis lengths, elongation and sinuosity were measured. Levels of intraspecific variation were considered by means of nested analysis of variance. The study of the intraspecific variation of the characters shows significant differences for lower levels, higher than variations between species for most characters. Levels of variation must be based upon the numerical analysis of seed characters for a correct approach to determining systematic value. E. mackaiana and E. andevalensis group together versus E. tetralix for sinuosity of testa cells. This is a good taxonomic character because it shows significant differences between species and no significant differences between populations. E. tetralix has strongly undulated testa cells while E. mackaiana and E. andevalensis have slightly undulated or curved testa cells. These two species can be discriminated by size of the seed, with seeds of 0.37-0.38 mm for E. andevalensis and 0.4 - 0.45 mm for E. mackaiana. Similarities are found in seed morphology for E. mackaiana and E. andevalensis, but we nevertheless consider the species status for the three taxa as the most natural taxonomic assignment. A key to the species is provided.  相似文献

Study on the dependence of orchids on fungi for seed germination and seedling development provides a mean for understanding the role of fungi in the orchid development process. The epiphytic orchid Coelogyne nervosa endemic to south India is exploited in an unsustainable manner for its therapeutic value. So a protocol for symbiotic seed germination was established for C. nervosa. We isolated a fungus by plating mycorrhizal root discs of the terrestrial orchid Eulophia epidendreae and identified it as Epulorhiza sp., by sequencing the internal transcribed spacer (ITS) regions of the ribosomal RNA gene. Germination of C. nervosa seeds was higher when inoculated with Epulorhiza sp. Uninoculated seeds of C. nervosa ceased to develop soon after the initiation of germination, and the embryo failed to rupture the seed testa. The isolated fungal hyphae entered the germinating seeds either through the pores in-between the integuments, or through the rhizoids. After the fungal establishment (peloton formation) in embryonic cells, the embryo transformed into a protocorm and after 45 days, 66% of the germinated seeds were transformed into protocorms. Nevertheless, promeristem formation occurred only after fungal association. Sixty-three percent of the protocorms developed their first leaf by 90 days and 62% of these produced a second leaf by 120 days after fungal inoculation. All the seedlings in green leaf stage produced roots and contained fungal pelotons. Our results suggest that the Epulorhiza sp. could be successfully used in the in vitro production of C. nervosa for their reintroduction into its natural environment.  相似文献

A review of palm remains recorded at archaeological sites throughout the New World is presented. Remains have been found at 130 sites from the southern United States to southern Uruguay. They are of four kinds: carbonized or dry endocarps or seeds, phytoliths, pollen, and implements. Twenty-nine genera and at least 50 species of palms (i.e., about 9% of all American species) have been recorded. The oldest record dates back to 14,700b.p. for carbonized endocarp fragments of an unidentified palm in Rondônia, Brazil. The use of palms, as recorded from remains, was particularly widespread after 9000b.p. The predominant remains are endocarps ofAcrocomia, Attalea s.l.,Astrocaryum, Bactris, Syagrus, Elaeis, andOenocarpus, all of which are important sources of edible oils or edible fruits and are still widely used by aboriginal peoples. The review supports the hypothesis that human groups have played an important role in the dispersal of some palm species in the neotropics. Humanaided dispersal ofAcrocomia aculeata from South America into Central America, and ofOenocarpus bataua from northwestern Amazonia to other areas, is postulated. Archaeological remains support the hypothesis that pejibaye (Bactris gasipaes) was domesticated in the inter-Andean valleys or on the adjacent Pacific lowlands of Colombia and later introduced into the Amazon Basin.  相似文献

Hard seeds of some legume species can germinate after seed-feeding insects bore through the seed coat and consequently break seed dormancy. Larvae of bruchine beetles are the main seed feeders attacking many legume species. Boring of the hard seed coat by bruchine beetle larvae enhances the germination percentage of legume species, but consuming too much of a single seed may reduce the chances the seed will survive. We hypothesise that the early mortality of bruchine larvae due to parasitism contributes positively to seed germination because larvae are killed before consuming too large a quantity of the seed. Here, we tested this hypothesis using Lathyrus japonicus seeds and Bruchus loti, the main seed feeder attacking this plant. B. loti larvae were mainly parasitised by two species of idiobiont parasitoids—Pteromalus sp. and Dinarmus sp. The seeds from which Pteromalus wasps emerged germinated more successfully than did the seeds from which B. loti adults emerged. B. loti larvae parasitised by the two wasp species consumed the seeds less intensively than did unparasitised larvae. Thus, the results of experiments supported our hypothesis. However, the germination success varied significantly between the seeds from which Pteromalus and Dinarmus wasps emerged. The difference in the size of seeds the two wasp species chose for parasitism may have influenced the germination percentage.  相似文献

BackgroundFenugreek and chromium are known to have anti-diabetic properties and this has been well demonstrated by multiple studies. Researches have been undertaken to study thebiofortification of chromium (Cr3+) in fenugreek. Some of the researchers have studied the biofortification from the soil irrigated with tannery waste water or from soil amended by tannery-sludge, with a view of enhancing the anti-diabetic effect of fenugreek plants. The present research work was also undertaken to increase the chromium content of fenugreek seeds, but through direct treatment of chromium picolinate solution to fenugreek seeds.MethodsFenugreek seeds were procured and divided in four groups having three batches of 10 g seeds- treatment1, treatment2, control1 and control2. Control1 group was kept raw and was given no treatment and control2 group was germinated using double distilled water. Treatment1 and treatment2, on the other hand, were given treatment of 0.02 g and 0.04 g chromium picolinate solution, on first day and 0.01 g and 0.02 g chromium picolinate solution on second day, respectively. The germinated samples were then completely dried, powdered, digested with di-acid mixture and assayed using Inductively Coupled Plasma optical emission spectrometry method for chromium content. The treatment1 sample was selected for further nutritional analysis along with control1 and control2 group to compare the nutritional composition of raw, germinated and chromium treated fenugreek seed flour. Fifteen sprouts from treatment1 group (treatment1A group) and fifteen sprouts from control2 group (control3 group) were sown in earthen pots for the analysis of chromium content in seeds of new plants.ResultsThe fenugreek seeds treated with two different concentrations of chromium picolinate viz. treatment1 and treatment2 group attained 55 and 80 times higher chromium content as compared to control2 group, respectively. All the estimated minerals and bioactive compounds were significantly high (p ≤ 0.01) in germinated fenugreek seed flour and chromium treated fenugreek seed flour compared to raw fenugreek seed flour. Germinated fenugreek seed flour and chromium treated fenugreek seed flour were statistically comparable to each other in respect of all the parameters analysed. Hence, it was evident that enriching fenugreek seeds with chromium, did not affect the nutritional content of fenugreek seed by any mean. Also, there was no significant difference between the chromium content in seeds of control3 group and T1A group.ConclusionsTreatment of fenugreek seeds with chromium solution seems to be an efficient and safe method for increasing their chromium concentration as compared to application of chromium to the soil for biofortification with minimal to no chance of chromium accumulation and inheritance in next generation plants. However, there is a need of more research to see how reliable these observations would be when different chromium salts and/or varied chromium concentration are used.  相似文献

ABSTRACT

The protein and oil contents in soybean seeds are major factors in seed quality. Seed proteins and oils are synthesized from sucrose and nitrogenous compounds transported into maturing seeds. In this study, we compared changes in the activity of phosphoenolpyruvate carboxylase (PEPC) and the accumulation profiles of protein and oil in maturing seeds of two soybean cultivars, which exhibit different protein and oil contents in seeds, to determine the interrelationships of them. A principal component analysis indicated a concordance of seed PEPC activity with the protein content, but did not with the oil content. PEPC activity per seed was highest in the late maturation stage, when the physiological status of the vegetative organs drastically changed. The high-protein cultivar had higher PEPC activity compared to the low-protein cultivar. These results highlight the biological role of PEPC in the synthesis of protein, therefore it was implied that PEPC could be a biomarker in soybean breeding.

Abstract

Different environments (initial forest, mature forest, Pinus and Eucalyptus stands) found in Seasonal Semideciduous Forest fragments affect the density of viable seeds, as well as the floristic similarity, diversity, and richness of tree species in soil seed banks. This hypothesis was tested in the current study. Soil seed bank samples were collected in the aforementioned environments during rainy and dry seasons, and taken to a shade house, where they remained under favorable seed germination conditions. Tree seedling emergence was measured, and sample species were identified every 15?days, for six months, in each sampling period. In total, 97 individuals m?2 and 23 species emerged in all environments and periods. The highest density of viable seeds of tree species in the soil seed bank was found in the initial forest stretch, mature forest stretch and abandoned Eucalyptus stand. Only the Pinus stand seed bank in the dry season had different floristic and lower viable seed density than the mature forest seed bank. Thus, all environments, except the abandoned Pinus stand, can preserved Seasonal Semideciduous Forest fragments.  相似文献

1. Most woody plant species in tropical habitats are primarily vertebrate‐dispersed, but interactions between ants and fallen seeds and fruits are frequent. This study assesses the species‐specific services provided by ants to fallen arillate seeds of Siparuna guianensis, a primarily bird‐dispersed tree in cerrado savanna. The questions of which species interact with fallen seeds, their relative contribution (versus vertebrates) to seed removal, and the potential effects on seedling establishment are investigated. 2. Seeds are removed in similar quantities in caged and control treatments, suggesting that ants are the main dispersers on the ground. Five ant species attended seeds. Pheidole megacephala (≈0.4 cm) cooperatively transported seeds, whereas the smaller Pheidole sp. removed the seed aril on spot. Large (> 1.0 cm) Odontomachus chelifer, Pachycondyla striata, and Ectatomma edentatum individually carried seeds up to 4 m. Bits of aril are fed to larvae and intact seeds are discarded near the nest entrance. 3. Overall, greater numbers of seedlings were recorded near ant nests than in control plots without nests. This effect, however, was only detected near P. megacephala and P. striata nests, where soil penetrability was greater compared with controls. Soil nutrients did not differ between paired plots. 4. This study confirms the prevalence of ant–seed interactions in cerrado and shows that ant‐derived benefits are species‐specific. Ant services range from seed cleaning on the spot to seed displacement promoting non‐random spatial seedling recruitment. Although seed dispersal distances by ants are likely to be shorter than those by birds, our study of S. guianensis shows that fine‐scale ant‐induced seed movements may ultimately enhance plant regeneration in cerrado.  相似文献
