Solid-phase synthesis has contributed to a significant portion of combinatorial libraries made to date. Among many advantages of solid-phase organic synthesis, one unique property is the pseudo-dilution effect. In reality, polymer support is a dynamic system and its structural mobility is controlled by many intrinsic and external factors such as cross-linking, load, solvent, temperature and incoming reagent concentrations. Dear Editor, Phosphoenolpyruvate carboxylase (PEPC; EC 4.1.1.31) islocated at an important branch point in the carbohydratemetabolism of plants. The enzyme is a homotetramer andcatalyzes the addition of bicarbonate to phosphoenolpyru-vate (PEP) to form oxaloacetate and phosphate. PEPC isregulated by metabolites and phosphorylation. AIIostericfeedback inhibition is mainly regulated by L-malate andL-aspartate which bind to a site separated from the activecenter (Kai et al., 1999; Paulus et al., 2013). Structure analy-sis of PEPC from Escherichia coli (Kai et al., 1999; Matsumuraet al., 2002), Zea rnays (Matsumura et al., 2002), Flaveria trin-ervia, and F. pringlei (Paulus et al., 2013) revealed that thesubstrate PEP and the feedback inhibitors bind to separatesites within each monomer.  相似文献

THE RNA of phage Qβ consists of about 3,500 nucleotides1 and comprises three known cistrons coding for maturation protein (a structural component of the viral particle also designated as A or A2 protein), coat protein and the β subunit of the viral replicase2. When ribosomes are bound to intact Qβ RNA they attach predominantly to one binding site, namely that at the beginning of the coat cistron3. THE single stranded RNA genome of bacteriophage Qβ has been variously estimated to consist of from 3,5001 to 4,5002 nucleotides. It contains three known cistrons3, which correspond to three of the four Qβ-specific proteins synthesized in vivo and in vitro4–6. These are: (1) the gene for the maturation or A protein (molecular weight 41,000 (refs. 4, 5)), (2) that for the major coat protein of the virus (molecular weight 14,000 (ref. 9)) and (3) the gene for the phage-specific subunit of the Qβ replicase (molecular weight 64,000 (ref. 10) or 69,000 (ref. 24)), listed in the probable order7,8 that they occur on the Qβ RNA. The fourth Qβ-specific protein, A1 or IIb (molecular weight 36,000 (refs. 4–6, 10)), has recently been shown by Weiner and Weber to have an N-terminal sequence which is identical (for eight amino-acids) to that of the coat protein7. Because increased amounts of A1 appear in virus particles grown in cells containing a UGA suppressor, Weiner and Weber postulate7 that this protein is the product of natural read-through at the UGA termination signal of the Qβ coat cistron. Such read-through (involving about 600 nucleotides) could occur entirely within a large “intercistronic” region between the coat and replicase genes, or could involve translation, either in or out of phase, of the replicase cistron. In hopes of distinguishing between these alternatives, I have isolated and examined the nucleotide sequence of the region surrounding the initiator codon of the Qβ replicase gene.  相似文献

The attachment site (attlambda) of bacteriophage lambda was examined in wild strains of Escherichia coli. Although the att region is non-coding, the DNA sequence was invariant in the 13 strains examined. Two other non-coding regions showed nine changes, all associated with a single strain. In four of 33 strains, sequences were inserted in or near the attlambda site and in two of these the insert was related to lambda. Among strains that can be lysogenized by lambda, integration was via the attlambda site in all cases. Some resistant strains can be lysogenized, and these have been termed "lenient." Most of these fail to give normal phage yield after induction. In some cases rare lysogens have been formed in cells that belong to a mutant subpopulation.  相似文献

In phage lambda, progeny particles bearing unreplicated chromosomes are recombinant by action of lambda's Red system only near the right end of the chromosome. These recombinants are frequently heterozygous (heteroduplex) for markers located there. In replication-blocked crosses involving two heavy-labeled parents we find that particles in the solitary peak, containing progeny with fully conserved DNA, vary in density. Those on the heavy side of this peak are more apt to be heterozygous than are those on the light side. The data fit a model in which a double chain cut at cos, lambda's packaging origin, is followed by partial exonucleolytic degradation of lambda's r chain from the right end leftward. The exposed l chain, which thereby constitutes a 3' overhang, invades an intact, circular homologue after itself suffering some degradation. Completion of the recombinant chromosome sometimes involves DNA synthesis primed by the invading chain.  相似文献

Background

Aging may be accompanied by a low grade chronic up-regulation of inflammatory mediators. A variety of endogenous locally released mediators as well as inflammatory cells have been reported in the human oral cavity. The aim of this investigation was to determine the presence of different classes of inflammatory mediators in human saliva and correlate the levels with age.

Methodology and Principal Findings

Unstimulated whole buccal salivary samples were obtained in the morning from 94 healthy volunteers within 30 minutes after waking. None of the participants had taken aspirin in the week prior to the saliva collection. Lysozyme activity, eicosanoid levels (prostaglandin E2 and leukotriene B4) and MMP-9 activity were measured. The antimicrobial activity (lysozyme activity) was not correlated with age whereas PGE2 levels were markedly correlated with age (r = 0.29; P<0.05; n = 56). Saliva from healthy subjects (≤40 years) compared with data derived from older volunteers (>40 years) demonstrated a significant increase in the mean values for PGE2 and MMP-9 activity with age. In addition, significant correlations were observed between LTB4 and PGE2 (r = 0.28; P<0.05; n = 56) and between LTB4 levels and MMP-9 activity in smokers (r = 0.78; P<0.001; n = 15).

Conclusions/Significance

The presence of significant levels and activity of inflammatory mediators in saliva suggests that the oral cavity of healthy subjects may be in a constant low state of inflammation associated with age.  相似文献

Surveys on the nesting sites of the crows Corvus splendens were conducted along different road-side trees and light-posts in different localities of greater Kolkata, India during the breeding seasons in 2009–2012. It is revealed that C. splendens prefer light-posts as nesting sites to trees under certain environmental conditions like availability of foods, assurance of nest, nest building materials and eggs and/or young. As there exists, in most cases only one spot for nesting in a light-post and the said spot is usually located at a height ranging from 5.5 to 6.0 m from the ground contrast to an average 11.9 m height of the tree nest. The height of the nest determines the time and energy to be required either for the collection of foods, or to return to the nest within a short time as and when needed in respect to the evolved circumstances. It is apparent that the crows did not care to face the unusual danger to ensure their survival by developing this sort of nesting habit. Thus, it seems that the crows have developed the strategy of nesting in the light-post by the road side or on the road only to maximize their benefit in respect to their survival and propagation.  相似文献

The incidence of testicular cancer, primarily seminoma, has been increasing in many countries, including the United States. The testis is often the site of residual cancer after adequate treatment with systemic chemotherapy. The blood-testis barrier is commonly cited as the explanation for residual tumor within the gonad after chemotherapy and as the indication for delayed orchiectomy. Conversely, complete eradication of viable tumor from the primary site is common and argues against the testis as a "tumor sanctuary." Residual tumor is also demonstrated within metastatic foci, and the disparity between the histopathologic response of the primary tumor and metastatic sites may be best explained by tumor heterogeneity and multiple tumor clones. Regardless of the scientific and academic arguments, delayed radical orchiectomy remains an important part of treatment for patients undergoing primary chemotherapy.  相似文献

Background Isolating Helicobacter pylori on culture media and performing antibiotic susceptibility testing is potentially the most useful tool for guiding antibiotic therapy, especially when antimicrobial resistance is suspected. The aim of this study was to determine whether the yield of H. pylori culture was related to the site from which the gastric specimen was obtained either before or after therapy.
Methods. Gastric mucosal biopsies from the antrum and the corpus of the stomach were cultured. H. pylori status was determined by histological assessment using the Genta stain.
Results. Fifty-two patients with documented H. pylori infection were studied: Twenty-three were tested before antibiotic therapy and 29 after therapy had failed. In 47 patients (90%), both antral and corpus culture specimens were positive. In 5 patients (10%), only one site was positive, with three false-negative antral and two false negative corpus cultures. The overall sensitivity of culture in detecting H. pylori infection was 95% (95% confidence interval = 89–98%) and was not significantly different for the antrum or corpus, either before or after therapy.
Conclusion. Culture of gastric biopsies from either the antrum or the corpus has an excellent diagnostic yield even in patients who failed antimicrobial therapy.  相似文献

Penicillin-binding proteins (PBPs), the main targets of β-lactam antibiotics, are membrane-associated enzymes that catalyze the two last steps in the biosynthesis of peptidoglycan. In Streptococcus pneumoniae, a major human pathogen, the surge in resistance to such antibiotics is a direct consequence of the proliferation of mosaic PBP-encoding genes, which give rise to proteins containing tens of mutations. PBP2b is a major drug resistance target, and its modification is essential for the development of high levels of resistance to piperacillin. In this work, we have solved the crystal structures of PBP2b from a wild-type pneumococcal strain, as well as from a highly drug-resistant clinical isolate displaying 58 mutations. Although mutations are present throughout the entire PBP structure, those surrounding the active site influence the total charge and the polar character of the region, while those in close proximity to the catalytic nucleophile impart flexibility onto the β3/β4 loop area, which encapsulates the cleft. The wealth of structural data on pneumococcal PBPs now underlines the importance of high malleability in active site regions of drug-resistant strains, suggesting that active site “breathing” could be a common mechanism employed by this pathogen to prevent targeting by β-lactams.  相似文献

Forty-nine amino acid substitutions were made at four positions in the Escherichia coli enzyme β-galactosidase; three of the four targeted amino acids are thought to be part of the active site. Many of the substitutions were made by converting the appropriate codon in lacZ to an amber codon, and using one of 12 suppressor strains to introduce the replacement amino acid. Glu-461 and Tyr-503 were replaced, independently, with 13 amino acids. All 26 of the strains containing mutant enzymes are Lac(-). Enzyme activity is reduced to less than 10% of wild type by substitutions at Glu-461 and to less than 1% of wild type by substitutions at Tyr-503. Many of the mutant enzymes have less than 0.1% wild-type activity. His-464 and Met-3 were replaced with 11 and 12 amino acids, respectively. Strains containing any one of these mutant proteins are Lac(+). The results support previous evidence that Glu-461 and Tyr-503 are essential for catalysis, and suggest that His-464 is not part of the active site. Site-directed mutagenesis was facilitated by construction of an f1 bacteriophage containing the complete lacZ gene on a single EcoRI fragment.  相似文献

Olfactory predators and scavengers rely on odors to locate food, and they forage primarily at night. We hypothesized that weather (e.g. wind speed, humidity, and temperature), vegetation, and landscape features affect the dissipation of odors in the atmosphere and, thus, impact the foraging efficiency of olfactory predators. We tested this hypothesis by assessing what conditions were correlated with the persistence of bait along the dike surrounding Willard Bay Reservoir, Utah. We distributed 124 chicken eggs and 199 dead European starlings (Sturnus vulgaris) as bait over 21 separate occasions spanning from May 21, 2009, to August 18, 2009. We used timers to record the time of consumption and cameras to identify which species ate the bait. Sixty‐six eggs and 87 dead European starlings were consumed. The bait was consumed mostly at night (83%) and by striped skunks (Mephitis mephitis), raccoons (Procyon lotor), and red foxes (Vulpes vulpes). We used nest survival models in program MARK to determine which covariates influenced bait persistence. We tested nine variables and found that bait persistence was influenced by section of the dike, time since study initiation, temperature, humidity, and wind speed but not bait type (egg or dead European starling), terrain, or visual cover. How quickly scavengers were able to locate food, which was inversely related to rates of bait persistence, decreased with higher temperatures and wind speeds and increased with higher humidity. Our results support the hypothesis that weather and micro‐habitat characteristics impact the ability of olfactory predators and scavengers to locate food.  相似文献
