目的了解Wistar大鼠的各种自发性肿瘤病变及其发生率。方法致癌实验中的对照组,采用4周龄SPF级Wistar大鼠,雌、雄性大鼠各60只,实验前观察1周,常规饲料喂饲104周后处死,进行组织病理学检查。结果报告了Wistar大鼠的各种自发性肿瘤病变及其发生率。雄性大鼠中发生肿瘤的动物占49.12%,发生良性肿瘤的动物占38.60%、发生恶性肿瘤的动物占17.54%;良性肿瘤主要有垂体腺瘤(19.30%)、睾丸间质细胞瘤(5.26%)和皮下纤维瘤(5.26%);恶性肿瘤主要有鳞状细胞癌(7.02%)和淋巴造血系统肿瘤(3.51%)。雌性大鼠中发生肿瘤的动物占60.34%;发生良性肿瘤的动物占50.00%、发生恶性肿瘤的动物占15.52%;良性肿瘤主要有乳腺纤维腺瘤(25.86%)和垂体腺瘤(24.14%);恶性肿瘤主要有腺癌(5.17%)和乳腺癌(3.45%)。结论本文报告的Wistar大鼠自发肿瘤及其发生率进一步丰富了现有SPF级Wistar大鼠自发性肿瘤的数据资料,可为有关技术人员提供一定的参考和借鉴。  相似文献

At many breeding facilities, rats are housed at relatively high densities until they are 5 weeks old, at which point they are either shipped for research or rehoused at standard cage densities according to weight. The authors carried out a pilot study in Long Evans and in Fischer 344 rats to investigate whether continuing to house rats at high densities (24 in(2) floor space per rat) past the age of 5 weeks, through puberty and into adulthood would alter behavioral or physiological parameters compared with raising rats at standard densities (about 72 in(2) floor space per rat). After rats reached puberty, the authors rehoused them with unfamiliar cagemates. The researchers evaluated clinical and behavioral signs of stress, weight, blood glucose concentration, white blood cell count and serum corticosterone concentration. Overall, cage density had little effect on the parameters measured, though gender seemed to affect stress in Long Evans rats. The results suggest that rats of these strains can be raised at the higher densities tested until any age and regrouped with unfamiliar cagemates without compromising rats' welfare or subsequent experimental data.  相似文献

Food hoarding was examined in male and female Long Evans rats. During a three-week period the animals were regularly submitted to the testing procedures, with food pellets in the home cage being either unrestricted (first week), or restricted (second and third week). When food pellets were present ad libitum, the hoarding scores were low, lower in females than in males. When food deprivation was imposed (second week), the animals responded by hoarding more pellets. This increase was more pronounced in males than in females. During the third week the hoarding scores became more or less stable, with males hoarding significantly more pellets than females.  相似文献

Rodents in search of food use visual environmental signals and complex spatial strategies and do not return to previously-visited locations, known as the win-shift strategy. The solution to the Olton Octagonal Maze (OOM) involves Working Memory (WM). A modified OOM was used that allows for measuring WM and Long Term Memory (LTM). The delayed spatial win-shift task consisted of a Training and Test phase separated by a delay. Prior to the Training phase, four arms were chosen at random and blocked, and food pellets were placed in the food cups of the four remaining open arms. Each rat was allowed to retrieve the pellets from the four open arms and then return to its home cage for the delay period (either 5 or 20 min). In the Test phase all 8 lanes were open, and the bait was placed in those blocked in the previous phase. Two experimental groups of rats, Long Evans and Norvegicus, and their corresponding control groups were trained. The experimental subjects performed Training-Delay-Test. The controls were only trained in the Test phase. Revisiting an arm previously explored in the 1st Phase was considered a LTM error. Revisiting an arm in the same trial constituted a WM error. It was concluded that the experimental groups do in fact possess LTM, with differences in favor of Norvegicus. There was no difference with respect to WM errors. The Norvegicus control group changes its strategy from allocentric to egocentric, which did not occur in the Long Evans control group.  相似文献

Spontaneous leukemia occurred in 36% of a group of 39 aged Slc: Wistar rats, which ranged in age at death from 99 to 123 (average age: 114) weeks old. The leukemic rats showed hemolytic anemia, jaundice and leukocytosis (21,000 to 360,000/mm3). Macroscopically, they were characterized by splenomegaly and hepatomegaly. The leukemia cells were rich in basophilic cytoplasm containing eosinophilic granules. Their nuclei were oval and occasionally revealed mitotic figures. These leukemia cells were consistently transplantable into both Slc: Wistar and F 344/NSlc rats. In the transplanted cases, the latency and survival periods were prolonged in proportion to decreases in the inoculated cell dose.  相似文献

The open field test (OFT) was carried out on Wistar and Sprague-Dawley rats between the ages of 3 to 20 weeks. At that time, the behavior of each naive rat was observed for two 3-minute periods separated by an interval of 24 h (Day). The OFT scores varied depending on the day and the age. Comparatively higher activity was observed in the extent of ambulation and rearing at 5 weeks old, rearing and preening at 7 weeks old, and preening and defecation at 11 weeks old in the Sprague-Dawley rats compared with the Wistar rats.  相似文献

Research has been carried out to investigate the effects of microwave exposure (7 GHz, surface energy density 10-50 mW/cm2, SAR 2.1-10.5 W/kg) on learned behaviors of rats in the paradigm of conditioned avoidance reflex. It was shown that transitory reductions in conditioned behavior after acute microwave exposure occurred at an SAR equal to the intensity of rat basal metabolism. It was found cumulative effects for intermittent exposures of rats at a power density of 10 mW/cm2.  相似文献

In the present study, we used closed colony-Wistar-Imamichi (WI), inbred WI and Long Evans (LE) adult male rats to examine the secretion of ACTH and corticosterone in response to restraint stress. Blood (0.3 ml) was withdrawn through a jugular cannula at 0, 15, 30, 60 and 120 min after the onset of restraint stress. Plasma concentrations of ACTH and corticosterone increased after stress in all groups, but the responses of ACTH and corticosterone secretion were higher in LE rats than in WI rats. Present data suggest that the LE rat might be a good model as a high-response strain and the closed colony or the inbred WI rat might be a good model as a low-response strain in restraint stress experiments.  相似文献

Background

The OLETF rat is an animal model of early onset hyperphagia induced obesity, presenting multiple pre-obese characteristics during the suckling period. In the present study, we used a cross-fostering strategy to assess whether interactions with obese dams in the postnatal environment contributed to the development of obesity.

Methodology

On postnatal Day (PND)-1 OLETF and control LETO pups were cross-fostered to same or opposite strain dams. An independent ingestion test was performed on PND11 and a nursing test on PND18. Rats were sacrificed at weaning or on PND90, and plasma leptin, insulin, cholesterol, triglycerides and alanine aminotransferase (ALT) were assayed. Fat pads were collected and weighed and adipocyte size and number were estimated. Body weight and intake, as well as the estrous cycle of the female offspring were monitored.

Principal Findings

During the suckling period, the pups'' phenotype was almost completely determined by the strain of the mother. However, pups independently ingested food according to their genotype, regardless of their actual phenotype. At adulthood, cross fostered males of both strains and LETO females were affected in regard of their adiposity levels in the direction of the foster dam. On the other hand, OLETF females showed almost no alterations in adiposity but were affected by the strain of the dams in parameters related to the metabolic syndrome. Thus, OLETF females showed reduced liver adiposity and circulating levels of ALT, while LETO females presented a disrupted estrous cycle and increased cholesterol and triglycerides in the long term.

Conclusions

The present study provides further support for the early postnatal environment playing a sex-divergent role in programming later life phenotype. In addition, it plays a more central role in determining the functioning of mechanisms involved in energy balance that may provide protection from or sensitivity to later life obesity and pathologies related to the metabolic syndrome.  相似文献

Administration of nitrofen (2,4-dichloro-4'-nitrodiphenyl ether) during organogenesis in rodents produces neonatal lethality accompanied by lung hypoplasia, diaphragmatic hernias, heart anomalies, and hydronephrosis. Different strains of rats, Long Evans Hooded (LEH) and Sprague-Dawley (SD), are reported to have different malformation responses to prenatal exposure, which could be due to true strain differences, to different levels and times of exposure, or to the use of different methods for detecting visceral malformations. In the present study, LEH, SD, and "virus-antibody-negative" SD (VAN-SD) rats were identically exposed to 0, 6.25, 12.5, or 25 mg/kg/day of nitrofen by gavage in corn oil on days 6 15 of gestation. At term, half of the litter was examined by the Wilson method of razorblade sectioning and the remainder by a modified Staples method of fresh visceral examination. The two methods were equally sensitive for detecting diaphragm, kidney, and lung anomalies, whereas heart malformations were more frequently identified with fresh visceral examination. The frequency of total malformations did not vary across strains at any dose, but there were substantial differences in the pattern of malformations in each strain. SD and VAN-SD rats responded similarly for all malformations, but had significantly higher incidences of diaphragm and lung anomalies than LEH rats. Conversely, LEH rats had significantly elevated levels of kidney anomalies compared to SD and VAN-SD rats, whereas frequency of heart malformations was low and comparable across strains. These results suggest that true strain differences exist in the pattern of malformation produced by prenatal exposure to nitrofen that may be based on genetic differences in embryonic susceptibility.  相似文献

Focal epithelial hyperplasia and metaplasia were observed in the intralobular ducts of the sublingual salivary glands of 302 out of 1142 Wistar rats. These rats (aged 16-145 weeks) served as controls in toxicological experiments. The ductal changes varied from small groups of irregular cylindrical cells with basophilic cytoplasm and vesicular nuclei, with often prominent nucleoli, to large areas of non-keratinized stratified squamous epithelium with marked mitotic activity and necrotic superficial cells being released in the lumen. Mononuclear inflammatory cells were often present in fibrous tissue surrounding altered ducts. Within certain limits the degree and incidence of the changes increased with increasing age; their aetiology is unknown.  相似文献

The rat is commonly employed in otological research, but spontaneous ear infections can confound the results of experimental procedures--wasting time, money, and animals. The authors focus on the incidence of spontaneous otitis media in Wistar rats. They compare disease incidence in animals housed in standard cages with those housed in barrier units, showing that 20% of their conventionally housed animals developed spontaneous otitis media, whereas only 5% of their animals housed in isolated units were infected. These results underscore the importance of strict control of the shipping, housing conditions, and manipulation of animals to be used in otological research.  相似文献

Dynamics of the conditioned behaviour of rats in a shuttle box was studied after combined exposure to Cs-gamma radiation (1 Gy, 1.32 Gy/min) and microwave radiation (2450 MHz, 1 mW/cm2, 3 h). The number of conditioned and interstimulus responses was found to decrease on day 5 after microwave + gamma irradiation and to increase on day 30 after gamma + microwave irradiation.  相似文献

Spontaneously hypertensive rats (SHRs) and the normotensive Wistar/Kyoto (WKY) rat were observed, along with Wistar rats (which represent the parent strain), on various open-field behaviors. All three strains were subsequently exposed to the activity-stress (A-S) ulcerogenic procedure. SHR and Wistar rats were very active in most open-field measures as compared with WKY rats, but only SHRs were active during the A-S treatment. WKY rats were very ulcer prone and had significantly more ulcers than SHRs, which in turn had more ulcers than Wistar rats. It was anticipated that Wistar rats would resemble the WKY rats, but in most measures the Wistars resembled the SHRs. The study suggests that although WKY rats function as an appropriate control for hypertension studies, these rats may be inappropriate as controls for other physiological and behavioral studies.  相似文献
