Some Physiological Aspects of Copper and Zinc Tolerance in Agrostis tenuis Sibth.: Cell Elongation and Membrane Damage 总被引:4,自引:0,他引:4
Three clones of Agrostis tenuis Sibth. were studied with respectto the effects of Zn and Cu on the growth of root segments excisedfrom the zone of cell elongation. Elongation growth in segmentsfrom a Cu-tolerant and a Zn-tolerant clone was inhibited toa lesser extent by Cu and Zn respectively than was the growthof a clone which was not tolerant to these metals. Concentrationsof Cu2+ which inhibited root growth also caused leakage of K+from the cells but toxic concentrations of Zn2+ did not induceK+ leakage. Copper induced a higher rate of K+ leakage at 25than at 0 °C. The impllcations of these results for thesite of the toxic effects of Zn and Cu and the nature of theresistance mechanisms are discussed. 相似文献

Investigations on the effects of low levels of Al on P adsorption,uptake and translocation in seedlings of the indigenous grassAgrostis capillaris were undertaken. Apparent uptake and transportof H2 32PO4 from nutrient solutions containing 10 or 100mmolm3 phosphate were characterized as functions of timeand concentration. Experiments on 32P uptake and transport insolutions containing no Al (control) or Al ranging from 3.7to 185 mmol m3 at pH ranging from 4.3 to 4.6, showedthat in 10 mmol m3 P, effects of Al at 3.7 and 37 mmolm3 on the size of the initial uptake shoulder were small,but some increase in subsequent P uptake to the roots was observed,though transport to the shoots was suppressed. With 37 mmolm3 Al in nutrient solution containing 100 mmol m3P, the uptake shoulder was much increased above the control.Subsequent root uptake was stimulated but transport was unaffected.Lack of toxicity of the Al concentrations used was indicatedby a lack of significant effect on plant fresh weight. AbsorbedAl was almost totally retained in the root in all treatments.Speciation calculations showed that the major species in Alamended nutrient solution at pH 4.4 were H2PO4, AI3+and AIHPO4+, together with substantial amounts of AISO4+ andsoluble aluminium hydroxy complexes (AIOH2+, AI(OH)2+), dependingon the relative concentrations of P and Al. The effects of Al,with 10 mmol m3 P, on adsorption of complexed P werepartly accounted for in terms of preferential cell wall adsorptionof Al complexes not containing P. Clonally propagated plants of Dactylis glomerata derived froma well-drained, heavily grazed cliff habitat (clone L) and froman undergrazed poorly-drained soil (clone A) were tested forwaterlogging tolerance in soil-culture. Water-logging did notaffect the two clones differentially, a result, which contrastedstrongly with that of a previous experiment in which simulatedgrazing (clipping to 20 cm) unexpectedly caused clone A to beless tolerant of waterlogging than clone L. Maximum leaf andleaf sheath length was reduced more by water-logging in cloneL than in clone A (P < 0.05). In solution-culture when providedwith factorial combinations of 0.5, 5 and 50 mg dm2 ofFe2+ and Mn2+ the shoot dry weight yield of the dry-soil clonewas reduced more than that of the wet-soil clone by 50 mg Fedm3 irrespective of Mn2+ concentration (P < 0.01)but the reduction of growth was less at higher Mn2+ concentrations.Fifty milligrams of Mn2+ dm3 reduced the growth of thedry soil clone but increased the growth of the wet soil clonewith Fe2+ at 5 mg dm2 (P < 0.05). Iron at 0.5 mg dm2was suboptimal for shoot growth of both clones at any levelof Mn2+ and caused more severe leaf chlorosis in the wet soilclone. Leaf tissue of clone L contained more iron than thatof clone A after waterlogging (P < 0.01) but in solutionculture, though increasing iron from 0.5 to 50 mg dm3almost doubled leaf iron content (P < 0.001), the interactionClones x Mn x Fe just failed to reach significance at P <0.05. The manganese content of leaf tissue from the two clonesvaried differently in response to solution manganese (Clonesx Mn P < 0.01), clone A showing a slightly greater increaseof manganese content at high solution concentration. Iron at50 mg dm3 suppressed Mn uptake (Mn x Fe, P < 0.001)in both clones. The two clones thus show marked environmentaladaptation to the chemistry of wet and dry soils. From compartmental analysis of 32P elution measurements, concentrationsand fluxes of orthophosphate were estimated for root corticalcells of intact seedlings of the indigenous grass Agrostis capillarisL. cv. Highland when in complete nutrient solution containing10 mmol m3 or 100 mmol m3 phosphate with or without3.7 and 37 mmol m3 Al, respectively, during loading andelution. When plotted as counts min1 remaining in thetissue as a function of time, the data failed to meet the criteriafor first order kinetics. Transformation of the data to meetthe kinetic criteria gave corrected values for compartmentalconcentrations and fluxes of phosphate, and estimates of theotherwise unresolved slowly exchanging compartment within thevacuole considered to be the cause of the discrepancy in fluxanalysis. In the control, the discrepancy was considered dueto sequestration of phosphate with Ca in the vacuole and a small,but not significant, increase in sequestered P occurred in thepresence of Al, the presence of which was confirmed by X-raymicroanalysis. A chemical speciation model was used to demonstrate,for various values of pH and carboxylic acid concentration,the possibility that phosphate was precipitated in root cellvacuoles as Ca and Al phosphates (hydroxy-apatite and variscite).The ecological significance of the ability of A. capillarisplants to make efficient use of scarce P resources by minimizingthe sequestration of P by Al in vacuoles, compared with Loliumperenne, was recognized. The effect of changes in nitrate-nitrogen supply on the absorptionand distribution of copper was examined in grasses grown inflowing solution culture with a maintained concentration ofcopper. Absorption by roots (µg Cu g1 dry root)decreased markedly when nitrogen had been depleted or was maintainedat 0.1 mg l1 N, but there was an immediate increase whennitrogen was maintained at 1.0 or 10.0 mg l1. There werealso large increases in the concentration of copper in the shootsof plants grown with 1.0 and 10.0 mg 11 N. The rootsof plants grown with 0.1 or 1.0 mg 11 N retained similarproportions of uptake, but a lower proportion was retained whenthe plants were grown with 10.0 mg 11. Although a lowerproportion of the copper was associated with cell walls in theplants grown at 10.0 mg 11 N this was the result of alower content of cell walls rather than an effect on copperitself. In a longer-term experiment in conventional solutionculture with a range of nitrogen concentration, the concentrationof copper in shoots was largely determined by shoot growth. Honeylocust (Gleditsia triacanthos L.) seedlings were grownin solution culture at pH 4.0) with 50, 150, 600 and 1500 mmolm3 aluminium. All levels of aiuminium reduced the sizeand weight of roots, shoots and leaves with the exception ofroot elongation at 50 mmol m3 Al. Aluminium content ofroots was 50 to 100 times that of shoots. With increasing concentrationof aluminium, aluminium content of leaves and roots increasedexponentially while a linear increase was observed for stems.The nutrient content of seedlings was improved in 50 mmol m3where increases in shoot calcium, magnesium, and phosphorusconcentrations were observed. Aluminium concentrations greaterthan 50 mmol m3 reduced shoot nutrient content. Presenceof aluminium increased the root phosphorus and calcium levelsbut had no effect on potassium and magnesium concentrations.Results show that honeylocust is an aluminium sensitive treespecies whose growth may be reduced by high soil Al levels. Chickpea cultivar ILC 482 was inoculated with salt-tolerantRhizobium strain Ch191 in solution culture with different saltconcentrations added either immediately with inoculation or5 d later. The inhibitory effect of salinity on nodulation ofchickpea occurred at 40 dS m1 (34.2 mol m3 NaCl)and nodulation was completely inhibited at 7 dS m1 (61.6mol m3 NaCl); the plants died at 8 dS m1 (71.8mol m3 NaCl). Chickpea cultivar ILC 482 inoculated with Rhizobium strain Ch191spcstrwas grown in two pot experiments and irrigated with saline water.Salinity (NaCl equivalent to 14 dS m1) significantlydecreased shoot and root dry weight, total nodule number perplant, nodule weight and average nodule weight. The resultsindicate that Rhizobium strain Ch191 forms an infective andeffective symbiosis with chickpea under saline and non-salineconditions; this legume was more salt-sensitive compared tothe rhizobia, the roots were more sensitive than the shoots,and N2 fixation was more sensitive to salinity than plant growth. Wheat, red clover and ryegrass were grown in flowing solutionculture with sufficient (+ Cu) and deficient (Cu) suppliesof copper. The rates of Cu absorption (µg g1 dryroot day1) did not differ greatly between species ineither treatment. Wheat plants, when transferred from the Cu to the +Cu treatment, absorbed Cu at a much slower rate thanthose which had remained throughout in the + Cu treatment. Inall plants considerable proportions of the absorbed Cu wereretained in the roots, even when the plants were Cu-deficient,and the concentration in roots usually exceeded that in anypart of the shoots in both treatments. Transferring wheat plantsfrom the +Cu to the Cu treatment decreased the concentrationin all plant parts except old leaves; similarly, transferringfrom the Cu to +Cu treatment increased the concentrationin all parts of the shoots, execept old leaves, and in the roots. In root tips of a salt-sensitive ecotype of Agrostis stolonifera,treatment with 100 mmol I1 NaCl for two weeks resultedin conspicuous structural damage to mitochondria, which becamedeficient in cristae, swelled and vacuolated. A salt-tolerantecotype maintained normal root tip mitochondrial structure underthe same salt treatment, in spite of accumulating higher levelsof sodium and chloride in its roots. Sugarbeet (Beta vulgaris L.) plants were grown in refined sandat graded levels of copper ranging from acute deficiency (0.000325µg Cu cm3) to excess (65 µg Cu cm3).Visible effects of copper deficiency appeared up to 0-00065µg Cu cm3and depression in growth up to 00065µCucm3. Copper deficiency decreased the concentrations ofDNA and RNA and the activities of polyphenol oxidase, cytochrome-coxidase, catalase and aldolase; and it increased the activitiesof peroxidase, ribonuclease and acid phosphatase in leaves.The maximum sucrose concentration in roots was obtained at 0-65µCucm3 Twenty four h after infiltration of a solution of 65µCucm3into copper deficient leaves, the activities of cytochrome-coxidase and peroxidase had increased even in the presence ofcycloheximide but that of polyphenol oxidase increased onlyin the absence of this inhibitor. The effects of various levels of copper on the uptake and distributionof copper in Chrysanthemum morifolium grown in solution cultureand peat-sand have been examined. Whole plants growing in shortdays were sampled at regular intervals, divided into roots,stem, leaves and lateral shoots, and analysed for copper. Thepartitioning of copper between these tissues showed that a relativelylarge proportion (3040 per cent) of the total plant copperwas accumulated in the roots of normal plants during the harvestingperiod, compared with approximately 10 per cent in the rootsof copper deficient plants. Whilst the copper content (ug g1) of leaves and stemfrom normal plants was negatively correlated with the amountof dry matter produced (P < 0·001), the correspondingcopper deficient tissues showed little variation in copper contentwith increases in tissue dry weight. A more detailed investigationof the copper content of leaves from normal plants showed thatgradients existed within the plant with respect to both leafposition and time of harvest which could be described by a singlecubic surface equation (P < 0·001). 相似文献

Nitrate ion uptake by the roots of hydroponically grown maizeseedlings was measured using the short-lived isotope 13N. Itis shown to be described by a four compartment model, recognizablynitrogen in the root bathing solution, nitrogen which is readilyexchangeable from the root, nitrogen bound in the root, andnitrogen transported from the root. Some of the absorbed activity leaks back into the root bathingsolution with the efflux from the root, as a fraction of theinflux, increasing with concentration to be greater than 08at external nitrate ion concentrations above about 1.0 mol m3.The capacity of the exchangeable root pool increases with externalnitrate ion concentration, approaching the expected cytoplasmicnitrate ion content at the highest external nitrate ion concentrationsstudied (70 mol m3). The investigation has highlighted the problems of interpretinguptake profiles in experiments for which the 10 min half-lifeof 13N dictates experimental times that are comparable withthe times for saturation of root pools. Several cell lines of Nicotiana plumbaginifolia resistant tomethotrexate (MTX) were isolated upon gradual elevation of theMTX concentration in the growth medium. One of the MTX-resistantcell lines, NP-19, acquired resistance to 10 mmol m3MTX, that is, at least 50-fold higher than the lethal dose forthe wild-type cell suspension. Its resistance was stably maintainedupon prolonged withdrawal of the drug. The acquisition of resistancewas accompanied by severe deterioration of regeneration potential.Regenerated shoots still manifested resistance to 50 mmol m3.MTX-resistant colonies and tiny shoots were recovered followingasymmetric fusion between -irradiated NP-19 protoplasts andN. plumbaginifolia leaf mesophyll-derived protoplasts on a mediumcontaining 10 mmol m3 MTX. The development of the shootsderived both from NP-19 calli and following somatic fusion wasarrested at the stage of 68 leaves. No difference wasfound in uptake of 3H-MTX between cell suspension of NP-19 andthe parental cell line. A 30-fold increase in binding of 3H-MTXto protein extract was found in cell line NP-19, suggestingdifferential capacity of MTX binding as a mechanism involvedin the MTX resistance of this cell line. Since differentiatedorgans seem more sensitive to MTX than undifferentiated tissues,this cell line is a promising source for a gene(s) conferringenhanced MTX-tolerance both in non-differentiated and differentiatedtissues. Rapid, Reversible Inhibition of Nitrate Influx in Barley by Ammonium 总被引:18,自引:2,他引:16
The rate of influx of nitrate into the roots of intact barleyplants was measured over a period of 35 min from externalnitrate concentrations of 1150 mmol m3, using13N-labelled nitrate as tracer. Ammonium at external concentrationsof 0.00550 mol m3 inhibited nitrate influx ina manner which did not conform to a simple kinetic model butincreased approximately as the logarithm of the ammonium concentration.At any particular ammonium concentration, inhibition of nitrateinflux reached its full extent within 3 min of the ammoniumbeing supplied and was not made more severe by up to 17 minpre-treatment with ammonium. On removing the external ammonium,nitrate influx returned to its original rate within about 3min. Potassium at 0.00550 mol m3 did not reproducethe rapid effect of ammonium on nitrate influx. Net uptake of nitrate also decreased when ammonium was supplied,over a similar timescale and to a similar extent as nitrateinflux. The decrease in nitrate influx caused by ammonium wassufficient to account for the observed reduction in net uptake,without necessitating any acceleration of nitrate efflux. Concentrationsof DCPA, propham and colchicine were selected to cause from0% to greater than 60% inhibition of oat (Avena sativa L. Victory)root growth after 24 h exposure. Root growth progressively declinedas concentrations were raised from 1·0 to 5·6mmol m3 DCPA, 1·05·0 mmol m3propham, and 50500 mmol m3 colchicine. In additionto inhibiting root growth each mitotic disrupter caused theroot tips to swell to an extent dependent upon concentration.All three compounds effectively disrupted mitosis at concentrationsthat caused less than maximal root growth inhibition. Mitoticdisruption was manifest as a reduction in the number of normalmitotic figures and an increase in the number of condensed prophase,multipolar and anaphase bridge division figures. The frequencyof each type of division figure was different for each of thethree compounds. DCPA disrupted mitosis more effectively whencompared with propham and colchicine at concentrations whichcaused the same amount of root growth inhibition. Each mitoticdisrupter also induced the formation of aberrant cell walls.DCPA was the most effective at disrupting cell plate formation,whereas colchicine was least effective. Protoplasts were successfully isolated from internodal callustissues of both Oxalis glaucifolia and O. rhombeo-ovata whenthey were digested in a solution containing 0.1% (w/v) MacerozymeR-10, 0.5% (w/v) cellulase Onozuka R-10 and 0.3 mmol m3sucrose. Protoplasts proliferated to give cell colonies on Gamborget al.'s B5 medium supplemented with 0.3 mmol m3 mannitol,0.5 mg dm32, 4-D, and 2.0 mg dm3 kinetin. Calluswas produced upon transfer of cell colonies to Murashige andSkoog medium containing 2.0 mg dm3 l-naphthaleneaceticacid (NAA) and 0.1 mg dm3 kinetin for O. glaucifolia,or with 5.0 mg dm3 NAA and 0.5 mg dm3 6-benzylaminopurine,for O. rhombeo-ovata. Plants were regenerated from O. glaucifoliaprotoplasts on a medium containing 0.1 mg dm3 NAA, 1.0mg dm3 kinetin and 1.0 mg dm3 gibberellic acid,but only vascular nodules were differentiated by O. rhombeo-ovataprotoplast-derived calli. Potato production in the tropical lowlands during the rainyseason is constrained by high temperature and low irradiance.This study examined the effect of these two variables on drymatter production and allocation, using plant growth, leaf anatomy,gas exchange and chlorophyll fluorescence measurements. Plantsof two clones, Solanum goniocalyx cv. Garhuash Huayro (GH) andDTO-33, a heat tolerant clone of S. tuberosum x S. phureja,were grown in growth chambers at 33/25 °C or 20/10 °Cday/night temperature. At each temperature, plants were grownin either 12 h high irradiance (430450 µmol m2s1 PAR) or 12 h low irradiance (250280 µmolm2 s1) both with a 6h photoperiod extensionof 6 µmol m2 s1. Plants were harvested after10 d (initial harvest) and after 20 d (final harvest). By theend of the study DTO-33 had produced more dry matter and hadtuberized, whereas GH had a greater leaf area ratio (LAR) andspecific leaf area (SLA). The highest relative growth rate (RGR)was at low temperature and low irradiance, possibly due to acombination of thin leaves with a large surface area. At thehigh temperature, low irradiance had the opposite effect, producingthe lowest net assimilation rate (NAR) and lowest RGR. Bothtuber number and weight were markedly reduced by high temperature.Low irradiance, in combination with high temperature, producedvirtually no tubers. Stomatal density, which was greater onGH than in DTO-33, was increased at high temperature. When measuredat 30 °C both clones, especially DTO-33, showed heat-adaptationin terms of ability to maintain a high rate of net photosynthesisat 30 °C. Plants grown at high irr-adiance and low temperaturehad the lowest net photosynthetic rate at 30 °C. Concurrentmeasurements of chlorophyll fluorescence indicated that onlythe initial (O) fluorescence parameter was affected. The absorption and distribution of Cu in red clover (Trifoliumpratense L.) were measured in plants grown in flowing solutionculture with Cu maintained throughout at 0.5 µg 11and N supplied either as nitrate or through symbiotic fixation.Although there was a decrease in Cu absorption, both with time,and with a depleted nitrate supply, it increased to its formerrate when nitrate was adjusted to 10 mg N 11 after aperiod of depletion. Differences in absorption between plantsdependent upon fixation and those supplied with nitrate wererelated to the slower initial growth of the plants fixing N.Considerable proportions (> 30 per cent) of the absorbedCu were retained by the roots. At the final harvest, and withthe exception of plants grown with nitrate adjusted to 0.1 mgN 11 after a period of depletion, the proportion of theCu retained was related to the concentration of N in the roots.The different N treatments produced differences in Cu concentrationin the shoots, and the effects were greater in the youngestfully expanded leaves than in older leaves. Callus cultures of 12 temperate grasses were established, somefor the first time, by incubating detached roots or whole seedlingson a Linsmaier and Skoog basal medium which contained 2, 4-dichlorophenoxyaceticacid (2, 4-D) at 1.0 mg 11 as the only growth hormone.The callus, which developed in the pericycle of the roots andon the embryo of the seeds, was subcultured on the same medium;further growth of the callus varied from good in the case ofDactylis glomeraia, Agrostis tennis, Cynosurus cristatus, andPoa trivialis, to poor in several Lolium species and varieties.Most cultures developed root primordia which sometimes grewinto visible roots, but shoot primordia, none of which grewinto shoots, were found only in the callus of Lolium multiflorumvar. westerwoldicum. Cell suspension cultures were also readily established and maintainedusing the same culture medium. Most cultures contained a highproportion of round or oval cells, which ranged from 18 to 77µm in diameter or length, while many also had a significantproportion of larger, more elongated cells which varied in lengthfrom 46 to 182 µm. The cells of Dactylis glomerata werecharacteristically larger and more convoluted than the cellsof other grass species that were examined. The addition of kinetinat 0.1 mg 11 to the 2, 4-D-containing culture mediumincreased the proportion of irregular-shaped cells and reducedthe dispersion of the cells, perhaps by improving cellular contactand adhesion; in some species, such as Agrostis tenuis and Phleumpratense, the presence of kinetin promoted the deposition ofstarch granules in cells. 相似文献

Perennial ryegrass (Lolium perenne L.) was grown from seed for29 d in flowing solution culture containing 0.1, 0.4 or 6.4mmol m3 P before the concentrations were changed (0.1and 0.4 raised to 6.4; 6.4 lowered to 0.4; controls unchanged)for an experimental period of two weeks to test the hypothesisthat after the seedling stage, the maximum rate of plant growthcould be sustained by a lower concentration of phosphate atthe root/solution interface than was necessary for the maximumrate of seedling growth. During the 29 d seedling period growthwas greatest on 6.4 mmol m3 P achieving 179 mg per plantdry weight compared with 122 and 26 mg on 0.4 and 0.1 mmol m3P respectively. During the experimental period growth on thetreatment 6.4 lowered to 0.4 mmol m3 P continued at thesame rate as the 6.4 control achieving 981 and 983 mg per plantdry weight respectively. Similarly growth of the treatment 0.4raised to 6.4 mol m3 P was unaffected by the change inconcentration and was comparable with the 0.4 control. Bothresults support the hypothesis for seedlings exceeding about100 mg per plant dry weight. In contrast the small plants ofthe treatment 0.1 raised to 6.4 mmol m3 P behaved similarlyto seedlings and responded rapidly to the increased concentrationof phosphate in solution, achieving high rates of phosphateuptake and increasing the growth of shoot more than the growthof root so that the ratio of root: shoot declined from 065 to0.34, a value similar to that for the seedlings grown on 6.4mmol m3 P.
