Numerous nucleoli can be observed in the macronucleus of the logarithmically growing ciliated protozoan Tetrahymena pyriformis; at late log phase the nucleoli aggregate and fuse. In stationary phase this fusion process continues, leaving a very few large vacuolated nuclear fusion bodies in the nucleus. When these stationary phase cells are placed into fresh enriched proteose peptone medium, the large fusion bodies begin to disaggregate during the 2.5-hour lag phase before cell division is initiated. By 3 to 6 hours after inoculation the appearance of the nucleoli in many cells returns to what it was in logarithmic cells. In view of the possible role of nucleoli in ribosome synthesis, attempts were made to correlate the morphological changes to changes in RNA and protein metabolism. The beginning of an increased RNA synthesis was concomitant with the beginning of disaggregation of the large fusion bodies into nucleoli, which was noticed in some cells by 1 hour after the return to fresh enriched proteose peptone medium. Increased protein synthesis then followed the increased RNA synthesis by 1 hour. The supply of RNA precursors (essential pyrimidines) were removed from cultures which were grown on a chemically defined synthetic medium, in order to study the relation between nucleolar fusion and synthesis of RNA and protein. Pyrimidine deprivation drastically curtailed RNA and protein synthesis, but did not cause fusion of nucleoli. When pyrimidines were added back to this culture medium, RNA synthesis was immediately stimulated and again preceded an increased protein synthesis by 1 hour. These studies suggest the involvement of unfused nucleoli in RNA and protein synthesis and demonstrate the extreme plasticity of nucleoli with respect to changes in their environment.  相似文献

During the growth cycle of Tetrahymena pyriformis the mitochondria undergo changes in position, number, and structure. Ciliates in the logarithmic growth phase possess elongated mitochondria which are aligned along the plasma membrane and are closely associated with the kinetosomes and kinetodesmata. Mitochondria appear to divide across the long axis at this time, resulting in two or more products. Throughout this phase of growth mitochondrial divisions keep pace with cytokinesis so that the population of mitochondria remains at essentially the minimal level. As the ciliates enter the stationary growth phase the mitochondria increase in number, become oval to spherical in shape, and some migrate into the cytoplasm. Intramitochondrial masses of various configurations appear at this time. Some of the mitochondria lying in the cytoplasm become incorporated into vacuoles. Within these vacuoles either a single mitochondrion appears or several mitochondria may be seen along with other cytoplasmic structures. Later in the stationary growth phase the contained mitochondria are dense and the tubules are more compact than normal. Various stages in disorganization of the mitochondria are observed in a single large vacuole. Cytochemical tests reveal the presence of acid phosphatase, suggesting that hydrolysis of the vacuolar contents occurs. Lipid droplets increase in number during the middle and late stationary phase of growth. These events are interpreted as being associated with the normal process of aging in T. pyriformis.  相似文献

The incorporation of 5-3H-uridine and 5-3H-cytidine into nucleolar and nonnucleolar RNA in the nucleus of monkey and pig kidney cells was measured in vitro during the cell life cycle. Time-lapse cinematographic records were made of cells during asynchronous exponential proliferation, in order to identify the temporal position of individual cells in relation to the preceding mitosis. Immediately following cinematography, cells were labeled with uridine-3H and cytidine-3H for a short period, fixed, and analyzed by radioautography. Since the data permit correlation of the rate of RNA labeling with the position of a cell within the cycle, curves could be constructed describing the rate of RNA synthesis over the average cell cycle. RNA synthesis was absent in early telophase, and rose very abruptly in rate in late telophase and in very early G1 in both the nucleus and the reconstituting nucleolus. Thereafter, through the G1 and S periods the rate of nuclear RNA synthesis rose gradually. When we used a 10-min pulse, there was no detectable change in the rate for nucleolar RNA labeling in monkey kidney cells during G1 or S. When we used a 30-min labeling time, the rate of nucleolar RNA labeling rose gradually in pig kidney cells. With increasing time after mitosis, the data became more variable, which may, in part, be related to the variation in generation times for individual cells.  相似文献

The mechanism of coordination between DNA replication and cell division was studied in Tetrahymena pyriformis GL-C by manipulation of the timing of these events with heat shocks and inhibition of DNA synthesis. Preliminary experiments showed that the inhibitor combination methotrexate and uridine (M + U) was an effective inhibitor of DNA synthesis. Inhibition of the progression of DNA synthesis with M + U in exponentially growing cells, in which one S period usually occurs between two successive divisions, or in heat-shocked cells, when successive S periods are known to occur between divisions, resulted in the complete suppression of the following division. In further experiments in which the division activities were reassociated with the DNA synthetic cycle by premature termination of the heat-shock treatment, it was shown that (a) the completion of one S period during the treatment was sufficient for cell division, (b) the beginning of division events suppressed the initiation of further S periods, and (c) if further S periods were initiated while the heat-shock treatment was continued, division preparations could not begin until the necessary portion of the S period was completed, even though DNA had previously been duplicated. It was concluded that a temporal incompatibility exists between DNA synthesis and division which may reflect a coupling mechanism which insures their coordination during the normal cell cycle.  相似文献

The mathematical and statistical problem of estimating the mean and variance of cell cycle times from cinemicrographically observed durations until mitosis (cytokinesis at late anaphase), disintegration, collision (cells superimposing each other) or emigration (moving out of the field of vision) of randomly chosen individual cells in a population in balanced exponential growth is treated. The resulting formulae are simple and considerably reduce the average cell observation time. They are applied to two normal human foetal cell lines and their SV40-transformed counterparts. One result is that the latter seem to have longer cycle times in spite of their shorter doubling times. Another result is that the cell mobility seems somewhat increased in the transformed populations. This corroborates earlier findings, indicating that the extent of cell loss, rather than the length of cycle times, may play a decisive role for the (net) doubling time of cultivated cell populations.  相似文献

运用Bernhard染色方法研究了小麦根端分生组织细胞核仁在细胞周期中的变化。结果显示,间期核仁染色很深,能够区分出纤维中心(FC)、致密纤维组分(DFC)和颗粒组分(G),而染色质被漂白,在染色质间可以观察到细小的RNP颗粒。进入前期,在染色质的边缘有小的RNP颗粒分布。中期,染色体周边分布着类似于间期核仁的深染的大RNP颗粒,形成一个不完全连续的“鞘”状结构;在染色体内部看不到类似核仁的深染颗粒。到了后期时,仍可见RNP“鞘”状结构的存在。进入末期,这些RNP植物逐渐由“鞘”脱离,最后参与新核仁的形成。这些结果表明,核仁解体后的物质直接转移到了中期染色的表面,并形成不连续的表层,没有进入染色体的内部。  相似文献

The rat hair cycle produces rapid anatomical and physiological changes in the entire skin. By the 4th day of the cycle, the epidermis has doubled in thickness. The present study shows that, during this proliferative period, there is a correlation between the biological state (growth) and the molecular composition (enzyme concentrations) of the epidermal cell. In proliferated 4th day epidermis there has occurred not only an increase in the number of viable cells but also an increase in succinic dehydrogenase and cytochrome oxidase activity and ribonucleic acid (RNA) content in the viable epidermal cell. Moreover, RNA content and enzyme activities appear to be correlated. The significance of these findings is discussed in the light of recent evidence supporting the relationship between RNA and enzyme synthesis and the role of hormones in the induction of enzyme adaptation.  相似文献

The fine structure of the nuclei of logarithmically growing Tetrahymena pyriformis, strain HSM, was studied at 30-minute intervals throughout the cell cycle. Organisms were selected at similar stages of cytokinesis by means of a braking pipette, incubated, fixed in OsO4, and embedded in agar to facilitate subsequent preparation for electron microscopy. Aggregates of micronuclear chromatin underwent a decrease in density and number with a concomitant increase in size throughout interphase. There were no impressive changes in macronuclear morphology. It was found possible to estimate a cell's progress through interphase by observation of micronuclear morphology, but attempts to correlate changes in fine structure with periods of DNA synthesis were unsuccessful.  相似文献

Synchronized populations of the chlorococcal alga Scenedesmus armatus (Chod.) Chod. were grown under five irradiance levels. During the cell cycles of these populations, reproductive processes such as DNA replication, nuclear division, protoplast fission, and daughter cell release and growth processes such as RNA and protein accumulation were followed. The amount of RNA and proteins increased stepwise with a short time interval between individual steps during which the rate of RNA and protein accumulation decreased. At each of the steps, the amount of RNA and protein approximately doubled and the number of steps increased with irradiance. At the end of each of the growth steps, a commitment to trigger the sequence of reproductive events (DNA replication, nuclear division, protoplast fission) was attained. After attaining the commitment point, the cells were able to trigger and terminate the whole reproductive sequence without any further growth, that is, even in the dark when the external supply of energy was cut off. With increasing irradiance, the number of commitment points attained during one cell cycle increased from one to four. Consequently, one to four sequences of the reproductive steps were triggered, and each of them ended by doubling the reproductive structures, which resulted in the formation of 2, 4, 8, or 16 daughter cells. The length of the precommitment periods shortened with increasing irradiance as the result of an increasing rate in growth. The length of postcommitment periods showed light independence and remained constant at the range of irradiances at which the number of growth steps and, consequently, the number of sequences of reproductive events did not change. At higher irradiances, the number of sequences of reproductive events increased, which caused a prolongation of postcommitment periods. The length of the cell cycle varied as a result of this distinct effect of irradiance on pre- and postcommitment periods.  相似文献

用放射免疫法测定了四膜虫在群体生长各阶段和在有性过程中cAMP的含量变化。在群体生长的延迟期(Lag phase)之末,即将进入对数期时,cAMP含量出现一个峰值。随着对数期的进展,cAMP含量陆续下降,在对数期之末,cAMP含量降至最低值。群体进入静止期后,细胞内cAMP含量略有回升。 在饥饿诱导接台的过程中,细胞内cAMP含量在开始饥饿的很短时间内即迅速降至一个最低点,并且在整个接合过程中一直保持较低的水平。接合抑制剂伴刀豆球蛋白A(CoaA)和精胺(spermine)等在抑制接合的同时,也阻止饥饿细胞cAMP含量的迅速下降,使之不能达到在正制情况下接合出现时所达到的低水平。 在饥俄细胞的培养液中检出了较高量的cAMP。这可能有助于说明何以饥饿细胞的cAMP含量能够在很短时间内迅速下降。至于排出的cAMP对四膜虫的接合有无促进或其他作用,有特继续研究。  相似文献

Addition of a 50 mm dosage of hydroxyurea (HU) to growing cultures of the cilitate Tetrahymena pyriformis allows cell growth to continue but inhibits DNA synthesis, and inhibits cell division after a 20–30% increase in cell number. Higher concentrations of HU cause cell death. T. pyriformis strain differences exist in sensitivity to HU. No cytotoxicity selective to a specific phase of the cell cycle could be demonstrated.
The inhibitory effects of HU are entirely reversed by removal of HU and this procedure can be used to induce cell cycle synchrony. Supplementing HU treated cells with deoxyribonucleosides relieves the inhibitory action of HU.
Carbamoyloxyurea (COU) is a reactive intermediate of HU and its action on Tetrahymena appears to be similar to that of HU. Because of this similarity and because COU is more potent than HU we postulate that COU is responsible for the unique properties previously ascribed to HU.  相似文献
