Two groups of proteins were isolated from the retina and pigment epithelium of eight-day-old chick embryos. Experiments with suspension cultures of retinal cells demonstrated that only the retinal extracts and the fraction of its acidic proteins can stimulate cell aggregation in vitro. Analysis by the method of high-performance liquid chromatography showed that fractions of acidic and basic retinal proteins, which markedly differ in their electric charge and biological activity, have similar composition. To study the effect of these proteins on the morphological and functional state of pigment epitheliumin vitro, a new experimental model is proposed, with the posterior segment of the newt (Pleurodeles waltl) eye used as a test tissue. The fraction of basic proteins isolated from the chick embryonic pigment epithelium stabilized cell differentiation in the newt pigment epithelium. The analyzed proteins proved to be biologically active at extremely low doses, corresponding to 10−12 M solutions.  相似文献

The chemical composition of chromatin from the livers of 12-, 15- and 19-day-old embryos, of 1-day-old chicks and of adult chickens was analysed. The process of embryonic development is accompanied by an increase in non-histone chromatin proteins and chromatin RNA, as well as in the phosphorus content of chromatin phosphoproteins. The amount of these components decreases in the livers of 1-day-old chicks and adults. Two-dimensional polyacrylamide gel electrophoresis of acid-soluble chromatin proteins showed an increase in the amount of the H1 histone in 19-day-old embryos and adult chickens. Non-histone proteins of embryo liver chromatin showed a high content of the fraction of Mr of about 40 000; this was not the case for adult chickens. The non-histone protein fraction of Mr of about 120 000, characteristic of adult chicken liver proteins, was not found in the livers of 12- and 15-day-old embryos. Non-histone chromatin proteins isolated from the livers of animals of different age exhibited also quantitative differences.  相似文献

The applicability of the audiocartridge system to simultaneous measurements of the ballistic movements from multiple points of the egg was examined in late chick embryos. Despite direct contact between the cartridges and the egg, minute movements of the egg attributable to cardiac contractions of the embryo (ballistocardiogram, BCG) could be simultaneously detected from at least three points. The results showed that the amplitude of BCG waves was dependent on the site of detection, indicating that the BCG was a directional wave. The cartridge system for BCG measurement was incorporated in the electrocardiography and arterial catheterization to acquire simultaneously electrocardiogram (ECG), BCG, and blood pressure from the embryo confined in the egg. Isochronal comparison of these waves indicated that the ballistic movements of the embryo might be caused not only by the ventricular contraction but also by the atrial excitation, i.e., the BCG comprised waves appearing in the ventricular presystolic, contraction, and ejection periods. For wide changes in the heart rate, the approximate isometric ventricular contraction period remained almost constant.  相似文献

We have determined the 679 nucleotide sequence of a cDNA clone which, by hybridization-translation experiments, corresponds to a 36K chick brain protein. Our studies provide a partial amino acid sequence for this protein, identifying it as chicken glyceraldehyde-3-phosphate dehydrogenase (GAPDH). Antisera raised against purified chicken GAPDH reacted with a 36K protein present in chick brain extracts and estimated to be the fourth most prevalent protein, as determined by either Coomassie Blue staining or by in vitro translation of chick brain mRNA. The amounts of GAPDH mRNA in chick brain, liver and muscle and adult chicken brain are similar, whereas the relative amount of adult chicken muscle GPDH mRNA is greatly elevated and that of adult liver lowered. The GAPDH protein levels showed a similar variation between tissues, suggesting that the levels of GAPDH protein are largely regulated by the amount of available GAPDH mRNA. The chicken GAPDH clone does not hybridize to rat mRNA, even though GAPDH is one of the most evolutionarily conserved proteins, indicating that selection pressures are heavier at the primary protein sequence level than at the nucleic acid sequence level for this gene, a situation contrasting to that of the tubulins.  相似文献

The ratio of concentrations Na+/K+ decreases in the brain and liver and increases in the cardiac muscle during the chick embryogenesis. The maximum concentration of Ca2+ was noted in the tissues under study on 14th day of chick embryonic development and its content decreased reliably at the subsequent stages. The concentration of Mg2+ in the tissues under study decreased during the embryogenesis as well.  相似文献

The frequency of triploidy was studied in early chicken embryos and hatchlings and in adult chickens which showed symptoms of intersexuality at an early age. The incidence of triploid embryos in the strain involved was not very high (0.6%), but it appeared to increase significantly when embryos were studied from eggs laid by very young hens (4.4%). Among the hatchlings screened only one single triploid individual was found (0.2%). The majority of the adults suspected of intersexuality proved to be triploid with a ZZW sex-chromosome triplet (66.1%). These data are briefly compared to those available from the literature.  相似文献
