Experiments were carried out to investigate the changes in CO2 assimilation, photon allocation, and photosynthetic electron flux in leaves of cucumber (Cucumis sativus L.) plants after chilling stress. Chilling significantly decreased CO2 assimilation, the energy flux via linear electron transport (J PS2) and non-constitutive thermal dissipation (J NPQ) but increased fluorescence and constitutive thermal dissipation (J f,D) in chilling-sensitive genotype Jinyan No. 4. In contrast, chilling had little effects on J NPQ and J f,D although CO2 assimilation and J PS2 were inhibited in chilling-tolerant genotype Jinchun No. 3. In parallel with the reduction in J PS2, electron flux to oxygenation and carboxylation by ribulose-1,5-bisphosphate carboxylase/oxygenase all significantly decreased while electron flux to O2 significantly increased, especially in chilling-sensitive genotype. Thermal and fluorescence dissipation were the main energy dissipation pathways whilst water-water cycle was an important electron sink when photosynthetic carbon reduction was suppressed after chilling. Chilling sensitivity of the photosynthetic apparatus was related to the operation of different photoprotection mechanisms.  相似文献

A gene encoding aminolevulinate synthase (ALA-S) in yeast (Saccharomyces cerevisiae YHem1) was introduced into the genome of tobacco (Nicoliana tabacum) under the control of Arabidopsis thaliana HemA1 gene promoter (AtHemA1 P). All transgenic lines transcribed the YHem1 gene, especially under light condition. The capacity to synthesize ALA and therefore chlorophyll was increased in transformed plants. Determination of gas exchange suggested that transgenic plants had significantly higher level of net photosynthetic rate (P n ), stomatal conductance (G s ) and transpiration rate (T r ), compared to the wild type (WT). Analysis with a modulated chlorophyll fluorometer demonstrated that the genetic transformation also caused a significant increase in photochemical efficiency of PSII (Fv /Fm F^{\prime}_{v} /F^{\prime}_{m} ), actual photochemical efficiency (Ф PSII ), photochemical quenching (qP), electron transfer rate (ETR) and the energy proportion in photochemistry (Pc), but decrease in proportion in heat dissipation (Hd). Chlorophyll-a fast fluorescence measurement and JIP-test indicated that photosynthetic performance index on cross section basis (PI CS ) and electron transport flux (ET o /CS) of transgenic tobacco were increased remarkably. And the probability that a trapped exciton can move a electron into the electron transport chain beyond Q A (Ψ o ) and the density of active reaction centers (RC/CS) were also increased obviously in transgenic tobacco. These results imply that transgenic tobacco plants expressing YHem1 gene had higher photosynthetic capacity and energy conversion efficiency than the WT plants.  相似文献

Brassinosteroids (BRs) are a new group of plant growth substances that promote plant growth and productivity. We showed in this study that improved growth of cucumber (Cucumis sativus) plants after treatment with 24-epibrassinolide (EBR), an active BR, was associated with increased CO2 assimilation and quantum yield of PSII (ΦPSII). Treatment of brassinazole (Brz), a specific inhibitor for BR biosynthesis, reduced plant growth and at the same time decreased CO2 assimilation and ΦPSII. Thus, the growth-promoting activity of BRs can be, at least partly, attributed to enhanced plant photosynthesis. To understand how BRs enhance photosynthesis, we have analyzed the effects of EBR and Brz on a number of photosynthetic parameters and their affecting factors, including the contents and activity of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco). Northern and Western blotting demonstrated that EBR upregulated, while Brz downregulated, the expressions of rbcL, rbcS and other photosynthetic genes. In addition, EBR had a positive effect on the activation of Rubisco based on increased maximum Rubisco carboxylation rates (V c,max), total Rubisco activity and, to a greater extent, initial Rubisco activity. The accumulation patterns of Rubisco activase (RCA) based on immunogold-labeling experiments suggested a role of RCA in BR-regulated activation state of Rubisco. Enhanced expression of genes encoding other Calvin cycle genes after EBR treatment may also play a positive role in RuBP regeneration (J max), thereby increasing maximum carboxylation rate of Rubisco (V c,max). Thus, BRs promote photosynthesis and growth by positively regulating synthesis and activation of a variety of photosynthetic enzymes including Rubisco in cucumber.  相似文献

In order to investigate the photoprotective function of photorespiration in grapevine under water stress, potted grapevines (Vitis vinifera L. cv. Cabernet Sauvignon) were randomly divided into three uniform groups for well-watered [watered every morning to keep the relative water content (RWC) of soil over 70 %], water-stress adapted (drought-adapted at 30 % relative soil water content for 30 days), and water stress without adaptation treatment (water-stressed to 30 % relative soil water content for 3 days). Net assimilation rate (A N), stomatal conductance (g s), substomatal CO2 concentration (C i), transpiration rate (E), actual photochemical efficiency of PSII (ΦPSII), and maximum photochemical efficiency of PSII (Fv/Fm) were recorded by combining measurements of gas exchange and chlorophyll fluorescence. Gross photorespiration (Pr), photosynthetic electron partitioning (JC/JT), photochemical quenching coefficient (qP), and non-photochemical quenching (NPQ) were also calculated. The ratio of net assimilation rate to transpiration rate (A N/E) was used as an indicator of water use efficiency (WUE). A N, apparent Pr, ΦPSII, Fv/Fm, qp, and g s decreased, NPQ increased, and gross Pr sustained at a high level under water stress. This suggests that both photorespiration and energy dissipation play important roles in protecting photosynthetic apparatus against photoinhibition. C i in water-stressed plants without adaptation treatment increased, which indicates the leaves suffered a non-stomatal limitation, while the water-stress adaped plants only suffered a stomatal limitation indicated by low C i.  相似文献

Chlorophyll (Chl) fluorescence of warm day/cool night temperature exposed Phalaenopsis plants was measured hourly during 48 h to study the simultaneous temperature and irradiance response of the photosynthetic physiology. The daily pattern of fluorescence kinetics showed abrupt changes of photochemical quenching (qP), non-photochemical quenching (NPQ) and quantum yield of photosystem II electron transport (ΦPSII) upon transition from day to night and vice versa. During the day, the course of ΦPSII and NPQ was related to the air temperature pattern, while maximum quantum efficiency of PSII photochemistry (Fv/Fm) revealed a rather light dependent response. Information on these daily dynamics in fluorescence kinetics is important with respect to meaningful data collection and interpretation.  相似文献

To investigate the effects of exogenously applied brassinosteroids on the thermotolerance of plants, leaf CO2 assimilation, chlorophyll fluorescence parameters, and antioxidant enzyme metabolism were examined in tomato (Lycopersicon esculentum Mill. cv. 9021) plants with or without 24-epibrassinolide (EBR) application. Tomato plants were exposed to 40/30°C for 8 days and then returned to optimal conditions for 4 days. High temperature significantly decreased the net photosynthetic rate (P n), stomatal conductance (G s), and maximum carboxylation rate of Rubisco (V cmax), the maximum potential rate of electron transport contributed to ribulose-1,5-bisphosphate (RuBP), as well as the relative quantum efficiency of PSII photochemistry (ФPSII), photochemical quenching (q P), and increased nonphotochemical quenching (NPQ). However, only slight reversible photoinhibition occurred during heat stress. Interestingly, EBR pretreatment significantly alleviated high-temperature-induced inhibition of photosynthesis. The activities of antioxidant enzymes such as superoxide dismutase (SOD), ascorbate peroxidase (APX), guaiacol peroxidase (GPOD), and catalase (CAT) increased during heat treatments, and these increases proved to be more significant in EBR-treated plants. EBR application also reduced total hydrogen peroxide (H2O2) and malonaldehyde (MDA) contents, while significantly increasing shoot weight following heat stress. It was concluded that EBR could alleviate the detrimental effects of high temperatures on plant growth by increasing carboxylation efficiency and enhancing antioxidant enzyme systems in leaves.  相似文献

Plant growth, chlorophyll (Chl) content, photosynthetic gas exchange, ribulose-1,5-bisphosphate carboxylase (RuBPCO) enzyme activity, and Chl fluorescence in radish (Raphanus sativus var. longipinnatus) plants were examined after turnip mosaic virus (TuMV) infection. Plant fresh mass, dry mass, Chl content, net photosynthetic rate (P N), transpiration rate (E), stomatal conductance (g s), and RuBPCO activity were significantly lower in infected plants after 5 weeks of virus infection as compared to healthy plants. The 5-week virus infection did not induce significant differences in intercellular CO2 concentration (C i, photochemical efficiency of photosystem 2, PS2 (Fv/Fm), excitation capture efficiency of open PS2 reaction centres (Fv'/Fm'), effective quantum efficiency of photosystem 2 (ΔF/Fm'), and photochemical quenching (qP), but non-photochemical quenching (qN) and alternative electron sink (AES) were significantly enhanced. Thus the decreased plant biomass of TuMV-infected plants might be associated with the decreased photosynthetic activity mainly due to reduced RuBPCO activity.  相似文献

The effect of salt concentration (NaCl) on growth, fluorescence, photosynthetic activities and pigment content of the cyanobacterium Arthrospira platensis has been investigated over 15 days. It has been observed that high NaCl concentration induces an increase of the growth, photosynthetic efficiency (α), phycobilin/chlorophyll ratio and a slight decrease of dark respiration and compensation points. Moreover, high NaCl concentration enhances photosystem II (PSII) activity compared to photosystem I (PSI). Results show that the phycobilin-PSII energy transfer compared to the chlorophyll-PSII (F695,600/F695,440) increases. However, data obtained about the maximal efficiency of PSII photochemistry are controversial. Indeed, the Fv/Fm ratio decreases in salt adapted cultures, while at the same time the trapping flux per PSII reaction center (TR0/RC) and the probability of electron transport beyond QA (0) remain unchanged at the level of the donor and the acceptor sites of PSII. This effect can be attributed to the interference of phycobilin fluorescence with Chl a when performing polyphasic transient measurements.  相似文献

Physiological and photosynthetic responses were investigated at three different depths of groundwater (DGW: 1.4, 2.4, and 3.4 m) in Elaeagnus angustifolia L., a locally adapted tree to the arid region in northwest China. Predawn leaf water potential and chlorophyll content declined gradually with the increasing DGW, whereas there was little effect on predawn variable-to-maximum chlorophyll fluorescence ratio F v/F m and leaf carotenoid compositions (xanthophyll cycle pool, neoxanthin, lutein, and β-carotene). Net photosynthetic rate (P n), quantum yield of PSII electron transport (ΦPSII), stomatal conductance (Gs), and intercellular CO2 concentration (Ci) declined obviously; however, P n decreased more than ΦPSII at deeper DGW. The photoinhibition of PSII at all three DGW occurred at midday in summer and increased as DGW increased. The ΔpH-dependent thermal dissipation and the level of de-epoxidation of the xanthophyll cycle at all three DGW reached their maxima at midday with the increase of light intensity. However, the fraction of functional PSII and light intensity at deeper DGW (2.4, 3.4 m) showed a negative correlation. This correlation suggested that most of violaxanthin was converted into zeaxanthin at midday, and the reversible inactivation of partial PSII reaction centers took place at deeper DGW. These results together suggest that both the xanthophyll cycle-dependent thermal dissipation and the reversible inactivation of partial PSII might have played important roles in avoiding the excess light-induced energy damage in leaves of this tree species at deeper DGW.  相似文献

Photosynthetic induction times and photoinhibition in relation to simulated sunflecks (sudden increase of irradiance from 20 to 1,500 μmol m−2 s−1) were examined in leaves of co-occurring Fagus lucida (a deciduous tree) and Castanopsis lamontii (an evergreen tree) saplings grown either in a beech forest understory or in an adjacent open site during a late rainy season. Two hypotheses were tested: (1) understory leaves would display faster photosynthetic induction times and greater photoinhibition than open-grown leaves; and (2) evergreen species would have slower photosynthetic induction times and lighter photoinhibition than deciduous species. Times to reach 90% of maximal CO2 assimilation rate (t 90%A ) and stomatal conductance did not differ between species, but showed faster by 3–5 min in open-grown leaves than understory leaves due to higher initial stomatal conductance (g s initial) and induction state 1 min into simulated sunflecks (IS1min) in the former. Our analysis across the published data on photosynthetic induction of 48 broad-leaved woody species again revealed the negative correlations between t 90%A and either g s initial or IS1min, and the similarity of t 90%A and between evergreen and deciduous species. Measurements of maximum PSII photochemical efficiency (F v/F m) indicated that photoinhibition occurred in saplings in any of the growth habitats during sunfleck-induced photosynthetic induction. Despite no interspecific differences in the degree of photoinhibition, understory leaves of both species suffered heavier photoinhibition than open-grown leaves, as indicated by a stronger decrease of F v/F m in the former. Dynamic changes in the quantum yields of PSII photochemistry and ΔpH- and xanthophyll-regulated thermal dissipation and adjustments in the partitioning of electron flow between assimilative and non-assimilative processes were functional to resist photoinhibition. However, such photoinhibition, together with stomatal and biochemical limitations, would decrease carbon gain during simulated sunflecks, particularly in understory leaves.  相似文献

Quantitative characteristics of photosynthetic electron transport were evaluated in vivo on the basis of the multi-exponential analysis of OJIP fluorescence transients induced by saturating actinic light. The OJIP fluorescence curve F(t), measured in Chlamydomonas reinhardtii cells, was transformed into the (1 − F O/F(t)) × (F V /F M)−1 transient, which is shown to relate to PS 2 closure. We assumed that kinetics of PS 2 closure during OJIP rise reflects time-separated processes related to the establishment of redox equilibrium at the PS 2 acceptor side (OJ), PQ pool (JI), and beyond Cyt b/f (IP). Three-exponential fitting was applied to (1 − F O/F(t)) × (F V /F M)−1 transient to obtain lifetimes and amplitudes of the OJ, JI, and IP components of PS 2 closure, which were used to calculate overall rates of reduction and re-oxidation of the PS 2 acceptor side, PQ pool, and intermediates beyond Cyt b/f complex. The results, obtained in the presence of inhibitors, oxidative reagents, and under different stress conditions prove the suggested model and characterize the introduced parameters as useful indicators of photosynthetic function.  相似文献

Summary Catenaria anguillulae parasitized and killed the eggs and second stage juveniles (J2) of Meloidogyne graminicola under natural conditions. The percentage of infection in eggs was higher than J2 of M.␣graminicola, which ranged between 0–50.3% and 0–18.9% in 2004 and 0–46.6% and 0–21.7% in 2005, respectively. The higher parasitism of eggs and J2 was recorded from those fields in which plants were severely infected with M. graminicola. The degree of parasitism of eggs and J2 by C. anguillulae varied with severity of root knot disease. The fields with a higher root gall index recorded a higher percentage of infection in eggs and J2 of M. graminicola. In general, old galls when teased and incubated, recorded higher parasitism of eggs and juveniles than young galls.  相似文献

To examine the role of acclimation versus adaptation on the temperature responses of CO2 assimilation, we measured dark respiration (R n) and the CO2 response of net photosynthesis (A) in Populus balsamifera collected from warm and cool habitats and grown at warm and cool temperatures. R n and the rate of photosynthetic electron transport (J) are significantly higher in plants grown at 19 versus 27°C; R n is not affected by the native thermal habitat. By contrast, both the maximum capacity of rubisco (V cmax) and A are relatively insensitive to growth temperature, but both parameters are slightly higher in plants from cool habitats. A is limited by rubisco capacity from 17–37°C regardless of growth temperature, and there is little evidence for an electron-transport limitation. Stomatal conductance (g s) is higher in warm-grown plants, but declines with increasing measurement temperature from 17 to 37°C, regardless of growth temperature. The mesophyll conductance (g m) is relatively temperature insensitive below 25°C, but g m declines at 37°C in cool-grown plants. Plants acclimated to cool temperatures have increased R n/A, but this response does not differ between warm- and cool-adapted populations. Primary carbon metabolism clearly acclimates to growth temperature in P. balsamifera, but the ecotypic differences in A suggest that global warming scenarios might affect populations at the northern and southern edges of the boreal forest in different ways.  相似文献

In a glasshouse, Bemisia tabaci infestation largely reduced response of photosynthesis to irradiance and CO2 concentration of Mikania micrantha compared with the non-infested control (C) ones. The maximum irradiance-saturated photosynthetic rate (P max) and saturation irradiance (SI) of the infested M. micrantha were only 21.3 % and 6.5 % of the C-plants, respectively. B. tabaci infestation led to the reduction of contents of chlorophyll and carotenoids in M. micrantha, which was accompanied with the decrease of actual photosystem 2 (PS2) efficiency (ΦPS2), efficiency of excitation energy capture by open PS2 reaction centres (Fv′/Fm′), electron transport rate (ETR), and photochemical quenching (qP). Moreover, superoxide dismutase and catalase activities significantly decreased while proline and glutathione contents significantly increased in infested M. micrantha. Hence B. tabaci infestation not only induced direct damage of photosynthetic apparatus but also altered the antioxidant enzymes activities in M. micrantha, which might as consequences accelerate senescence of this weed.  相似文献

Plants differ in how much the response of net photosynthetic rate (P N) to temperature (T) changes with the T during leaf development, and also in the biochemical basis of such changes in response. The amount of photosynthetic acclimation to T and the components of the photosynthetic system involved were compared in Arabidopsis thaliana and Brassica oleracea to determine how well A. thaliana might serve as a model organism to study the process of photosynthetic acclimation to T. Responses of single-leaf gas exchange and chlorophyll fluorescence to CO2 concentration measured over the range of 10–35 °C for both species grown at 15, 21, and 27 °C were used to determine the T dependencies of maximum rates of carboxylation (VCmax), photosynthetic electron transport (Jmax), triose phosphate utilization rate (TPU), and mesophyll conductance to carbon dioxide (gm). In A. thaliana, the optimum T of P N at air concentrations of CO2 was unaffected by this range of growth T, and the T dependencies of VCmax, Jmax, and gm were also unaffected by growth T. There was no evidence of TPU limitation of P N in this species over the range of measurement conditions. In contrast, the optimum T of P N increased with growth T in B. oleracea, and the T dependencies of VCmax, Jmax, and gm, as well as the T at which TPU limited P N all varied significantly with growth T. Thus B. oleracea had much a larger capacity to acclimate photosynthetically to moderate T than did A. thaliana.  相似文献

The aim of this study was to investigate acclimation of micropropagated plants of Rhododendron ponticum subsp. baeticum to different irradiances and recovery after exposure to high irradiance. Plants grown under high (HL) or intermediate (IL) irradiances displayed higher values of maximum electron transport rate (ETRmax) and light saturation coefficient (Ek) than plants grown under low irradiance (LL). The capacity of tolerance to photoinhibition (as assessed by the response of photochemical quenching, qp) varied as follows: HL > IL > LL. Thermal energy dissipation (qN) was also affected by growth irradiance, with higher saturating values being observed in HL plants. Light-response curves suggested a gradual replacement of qp by qN with increasing irradiance. Following exposure to irradiance higher than 1500 μmol m−2 s−1, a prolonged reduction of the maximal photochemical efficiency of PS 2 (Fv/Fm) was observed in LL plants, indicating the occurrence of chronic photoinhibition. In contrary, the decrease in Fv/Fm was quickly reverted in HL plants, pointing to a reversible photoinhibition.  相似文献

Photosynthetic characteristics were compared between plants of low altitude (LA) grown at LA (Palampur; 1 300 m) and at high altitude, HA (Kibber; 4 200 m), and plants naturally occurring at different altitudes (Palampur, 1 300 m; Palchan, 2 250 m; and Marhi, 3 250 m). Net photosynthetic rate (P N) was not significantly different between altitudes. However, the slopes of the curve relating P N to intercellular CO2 concentration (C i) were higher in plants at Palchan, Marhi, and Kibber compared to those at Palampur, indicating that plants had higher efficiency of carbon uptake (the initial slope of P N/C i curve is an indication) at HA. They had also higher stomatal conductance (g s), transpiration rate, and lower water use efficiency at HA. g s was insensitive to photosynthetic photon flux density (PPFD) for plants naturally occurring at Palampur, Palchan, and Marhi, whereas plants from LA grown at Palampur and Kibber responded linearly to increasing PPFD. Seeds of Suaeda salsa were cultured in dark for 3 d and betacyanin accumulation in seedlings was promoted significantly. Then the seedlings with accumulated betacyanin (C+B) were transferred to 14/10 h light/dark and used for chilling treatment 15 d later. Photosystem 2 (PS2) photochemistry, D1 protein content, and xanthophyll cycle during the chilling-induced photoinhibition (exposed to 5 °C at a moderate photon flux density of 500 μmol m−2 s−1 for 3 h) and the subsequent restoration were compared between the C+B seedlings and the control (C) ones. The maximal efficiency of PS2 photochemistry (Fv/Fm), the efficiency of excitation energy capture by open PS2 centres (Fv′/Fm′), and the yield of PS2 electron transport (ΦPS2) of the C+B and C leaves both decreased during photoinhibition. However, smaller decreases in Fv/Fm, Fv′/Fm′, and ΦPS2 were observed in the C+B leaves than in C ones. At the same time, the deepoxidation state of xanthophyll cycle, indicated by (A+Z)/(V+A+Z) ratio, increased rapidly but the D1 protein content decreased considerably during the photoinhibition. The increase in rate of (A+Z)/(V+A+Z) was higher but the D1 protein turnover was slower in C+B than C leaves. After photoinhibition treatment, the plants were transferred to a dim irradiation (10 μmol m−2 s−1) at 25 °C for restoration. During restoration, the chlorophyll (Chl) fluorescence parameters, D1 protein content, and xanthophyll cycle components relaxed gradually, but the rate and level of restoration in the C+B leaves was greater than those in the C leaves. The addition of betacyanins to the thylakoid solution in vitro resulted in similar changes of Fv/Fm, D1 protein content, and (A+Z)/(V+A+Z) ratio during the chilling process. Therefore, betacyanin accumulation in S. salsa seedlings may result in higher resistance to photoinhibition, larger slowing down of D1 protein turnover, and enhancement of non-radiative energy dissipation associated with xanthophyll cycle, as well as in greater restoration after photoinhibition than in the control when subjected to chilling at moderate irradiance.  相似文献

Two rice chlorophyll (Chl) b-less mutants (VG28-1, VG30-5) and the respective wild type (WT) plant (cv. Zhonghua No. 11) were analyzed for the changes in Chl fluorescence parameters, xanthophyll cycle pool, and its de-epoxidation state under exposure to strong irradiance, SI (1 700 μmol m−2 s−1). We also examined alterations in the chloroplast ultrastructure of the mutants induced by methyl viologen (MV) photooxidation. During HI (0–3.5 h), the photoinactivation of photosystem 2 (PS2) appeared earlier and more severely in Chl b-less mutants than in the WT. The decreases in maximal photochemical efficiency of PS2 in the dark (Fv/Fm), quantum efficiency of PS2 electron transport (ΦPS2), photochemical quenching (qP), as well as rate of photochemistry (Prate), and the increases in de-epoxidation state (DES) and rate of thermal dissipation of excitation energy (Drate) were significantly greater in Chl b-mutants compared with the WT plant. A relatively larger xanthophyll pool and 78–83 % conversion of violaxanthin into antheraxanthin and zeaxanthin in the mutants after 3.5 h of HI was accompanied with a high ratio of inactive/total PS2 (0.55–0.73) and high 1–qP (0.57–0.68) which showed that the activities of the xanthophyll cycle were probably insufficient to protect the photosynthetic apparatus against photoinhibition. No apparent difference of chloroplast ultrastructure was found between Chl b-less mutants and WT plants grown under low, LI (180 μmol m−2 s−1) and high, HI (700 μmol m−2 s−1) irradiance. However, swollen chloroplasts and slight dilation of thylakoids occurred in both mutants and the WT grown under LI followed by MV treatment. These typical symptoms of photooxidative damage were aggravated as plants were exposed to HI. Distorted and loose scattered thylakoids were observed in particular in the Chl b-less mutants. A greater extent of photoinhibition and photooxidation in these mutants indicated that the susceptibility to HI and oxidative stresses was enhanced in the photosynthetic apparatus without Chl b most likely as a consequence of a smaller antenna size.  相似文献
