Forms of zinc accumulated in the hyperaccumulator Arabidopsis halleri   总被引:16,自引:0,他引:16       下载免费PDF全文
The chemical forms of zinc (Zn) in the Zn-tolerant and hyperaccumulator Arabidopsis halleri and in the non-tolerant and nonaccumulator Arabidopsis lyrata subsp. petraea were determined at the molecular level by combining chemical analyses, extended x-ray absorption spectroscopy (EXAFS), synchrotron-based x-ray microfluorescence, and muEXAFS. Plants were grown in hydroponics with various Zn concentrations, and A. halleri specimens growing naturally in a contaminated site were also collected. Zn speciation in A. halleri was independent of the origin of the plants (contaminated or non-contaminated) and Zn exposure. In aerial parts, Zn was predominantly octahedrally coordinated and complexed to malate. A secondary organic species was identified in the bases of the trichomes, which contained elevated Zn concentrations, and in which Zn was tetrahedrally coordinated and complexed to carboxyl and/or hydroxyl functional groups. This species was detected thanks to the good resolution and sensitivity of synchrotron-based x-ray microfluorescence and muEXAFS. In the roots of A. halleri grown in hydroponics, Zn phosphate was the only species detected, and is believed to result from chemical precipitation on the root surface. In the roots of A. halleri grown on the contaminated soil, Zn was distributed in Zn malate, Zn citrate, and Zn phosphate. Zn phosphate was present in both the roots and aerial part of A. lyrata subsp. petraea. This study illustrates the complementarity of bulk and spatially resolved techniques, allowing the identification of: (a) the predominant chemical forms of the metal, and (b) the minor forms present in particular cells, both types of information being essential for a better understanding of the bioaccumulation processes.  相似文献

A previously published hydrothermal brine-river water mixing model driven by ocean crust production suggests that the molar Mg/Ca ratio of seawater (mMg/Ca(sw)) has varied significantly (approximately 1.0-5.2) over Precambrian time, resulting in six intervals of aragonite-favouring seas (mMg/Ca(sw) > 2) and five intervals of calcite-favouring seas (mMg/Ca(sw) < 2) since the Late Archaean. To evaluate the viability of microbial carbonates as mineralogical proxy for Precambrian calcite-aragonite seas, calcifying microbial marine biofilms were cultured in experimental seawaters formulated over the range of Mg/Ca ratios believed to have characterized Precambrian seawater. Biofilms cultured in experimental aragonite seawater (mMg/Ca(sw) = 5.2) precipitated primarily aragonite with lesser amounts of high-Mg calcite (mMg/Ca(calcite) = 0.16), while biofilms cultured in experimental calcite seawater (mMg/Ca(sw) = 1.5) precipitated exclusively lower magnesian calcite (mMg/Ca(calcite) = 0.06). Furthermore, Mg/Ca(calcite )varied proportionally with Mg/Ca(sw). This nearly abiotic mineralogical response of the biofilm CaCO3 to altered Mg/Ca(sw) is consistent with the assertion that biofilm calcification proceeds more through the elevation of , via metabolic removal of CO2 and/or H+, than through the elevation of Ca2+, which would alter the Mg/Ca ratio of the biofilm's calcifying fluid causing its pattern of CaCO3 polymorph precipitation (aragonite vs. calcite; Mg-incorporation in calcite) to deviate from that of abiotic calcification. If previous assertions are correct that the physicochemical properties of Precambrian seawater were such that Mg/Ca(sw) was the primary variable influencing CaCO3 polymorph mineralogy, then the observed response of the biofilms' CaCO3 polymorph mineralogy to variations in Mg/Ca(sw), combined with the ubiquity of such microbial carbonates in Precambrian strata, suggests that the original polymorph mineralogy and Mg/Ca(calcite )of well-preserved microbial carbonates may be an archive of calcite-aragonite seas throughout Precambrian time. These results invite a systematic evaluation of microbial carbonate primary mineralogy to empirically constrain Precambrian seawater Mg/Ca.  相似文献

When tobacco (Nicotiana tabacum) plants are exposed to toxic level of cadmium (0.2 mM Cd), their trichomes actively excrete crystals (Choi et al., 2001). In this study, we investigated the distribution of Cd and NaCI on trichomes and leaf surfaces. Energy dispersive x-ray (EDX) analysis revealed that, under toxic Cd stress, crystals exudated from the trichomes contained high amounts of Ca, Mg, and Cd, as well as low levels of P, S, and Mn. Electron spectroscopic imaging (ESI) from trichomes and attached crystals showed that these crystals emitted denser radiation energy for Ca and Cd than did the head cells of the trichomes. However, no Cd was detected on the trichome surface itself or within the leaf epidermis. In contrast, treatment with salt (NaCI) did not stimulate crystal formation; instead, it induced the abnormal expansion of trichome cells. Although Na was not accumulated within the crystals, a considerable amount of both Na and CI was sequestered within the stalk cells of the long trichomes. Therefore, we believe that tobacco trichomes play an important role in Cd crystal exudation through crystallization, but that, under NaCI stress, the long trichomes sequester those elements within their stalks.  相似文献

Formation of CaCO3 induced by fungal physiological activities is a potential way to sequestrate atmospheric CO2 in ecosystem. Alternaria sp. is a saprophytic fungus isolated from a forest soil. We examined the precipitation of CaCO3 induced by the fungus in response to different levels of Ca(NO3)2 or CaCl2 in agar media, and the biogenesis of CaCO3 was verified by low δ13C value. The formed CaCO3 was identified as calcite by X-ray diffraction analysis. Square, rectangular and rhombic CaCO3 crystals and amorphous calcium carbonate were observed around mycelia at higher levels of Ca(NO3)2. Acidification occurred in media at low concentrations (0 and 0.0002 M) of Ca(NO3)2, and no CaCO3 formed in these media. The quantities of CaCO3 formed in media increased with increasing concentrations of Ca(NO3)2 and were significantly correlated to fungal biomass, pH value and nitrite concentrations. No CaCO3 was formed in media with CaCl2 at all levels. These results collectively indicated that the formation of CaCO3 can be induced by the fungal assimilation of nitrate. The study also revealed that biogenic crystal of CaCO3 tended to grow on a silicon nucleus and the amorphous calcium carbonate (ACC) was the transient stage of CaCO3 crystal.  相似文献

The structure-directing influence of the organic dicarboxylates malonate, succinate, glutarate and adipate as templating species on the hydrothermal formation of CaCO(3) was investigated at different temperatures (60, 80, 90, 120, 150 and 190 degrees C) and with a range of molar ratios of [Ca(2+)]/[templating species] (20, 14.3, 10, 7.7, 5, 1, 0.5 and 0.33). In the presence of the dicarboxylates, one, two or three polymorphs of CaCO(3) - calcite, aragonite and vaterite - could be formed, depending on the reaction conditions. In addition changes in crystal morphology were observed for the CaCO(3) polymorphs depending on the concentration of the template. In contrast, synthesis under ambient conditions of temperature and pressure resulted only in calcite formation, although template-dependent morphological changes were again observed. Crystalline products were all characterized by powder X-ray patterns and SEM (Scanning Electron Microscopy) micrographs. The ambient reactions with the chelating, dinucleating carboxylato ligands H(3)heidi and H(5)hpdta produce more profound changes in calcite morphology. With H(3)heidi rounded calcite crystals with shapes similar to that of otoliths are formed and with H(5)hpdta the formation of microtrumpets of constructed from bundles of nanocrystals of calcite is observed. The possible mode of action of these ligands on calcite formation is discussed in the context of known coordination chemistry with other metal ions.  相似文献

The infrared spectroscopic investigation of biominerals isolated from different Cactaceae species belonging to the Opuntioideae subfamily shows the presence of a very complex mineral composition, including whewellite (monohydrated calcium oxalate), opal (SiO2) and calcite (CaCO3). The cellular compartmentation of elements was analysed in the Zn hyperaccumulator Arabidopsis halleri (L.) O'Kane & Al-Shehbaz (=Cardaminopsis halleri) using energy-dispersive X-ray microanalysis of frozen-hydrated tissues. Quantitative data were obtained using oxygen as an internal standard in the analyses of vacuoles, whereas a peak/background ratio method was used for quantification of elements in pollen and dehydrated trichomes. Arabidopsis halleri was found to hyperaccumulate not only Zn but also Cd in the shoot biomass. While large concentrations of Zn and Cd were found in the leaves and roots, flowers contained very little. In roots grown hydroponically, Zn and Cd accumulated in the cell wall of the rhizodermis (root epidermis), mainly due to precipitation of Zn/Cd phosphates. In leaves, the trichomes had by far the largest concentrations of Zn and Cd. Inside the trichomes there was a striking sub-cellular compartmentation, with almost all the Zn and Cd being accumulated in a narrow ring in the trichome base. This distribution pattern was very different from that for Ca and P. The epidermal cells other than trichomes were very small and contained lower concentrations of Zn and Cd than mesophyll cells. In particular, the concentrations of Cd and Zn in the mesophyll cells increased markedly in response to increasing Zn and Cd concentrations in the nutrient solution. This indicates that the mesophyll cells in the leaves of A. halleri are the major storage site for Zn and Cd, and play an important role in their hyperaccumulation. Growth was severely inhibited when tobacco plants were exposed to toxic levels of cadmium (0.2 mM). However, when this treatment was combined with a high concentration of calcium (30 mM), the Cd-induced damage was strongly alleviated. Under these enhanced conditions, i.e., 30 mM Ca and 0.2 mM Cd, Ca crystals not only were heavily deposited in the leaves but were also actively excreted from the trichomes. The X-ray spectrum from our Energy Dispersive analysis revealed that both intra-and extracellular Ca crystals contained detectable amounts of Cd. Moreover, intracellular Ca deposition in the leaves was stimulated only by a high Ca concentration (30 mM); moderate levels of Ca (3 mM) or a toxic amount of Cd (0.2 mM) alone resulted in crystal deposition that was undetectable under a light microscope. In contrast, extracellular crystal formation on the trichomes was stimulated by toxic Cd treatment but not by high Ca concentrations alone. Finally, Inductively Coupled Plasma Spectroscopy revealed that a high level of Ca (30 mM) suppressed Cd accumulation while also increasing the endogeneous Ca concentration in the leaves. These observations imply that the amelioration of Ca against toxic Cd in tobacco plants is a result of not only the inhibition of Cd uptake, but also the extra-and intracellular sequestration of cadmium via Ca crystallization.  相似文献

The Ca, Mg, K, and P content of dry barley (Hordeum vulgare) grains and seedlings was investigated using energy dispersive x-ray analysis and neutron activation analysis. Energy dispersive x-ray analysis of protein bodies in aleurone cells showed that these bodies contained very little Ca in relation to P, Mg, and K. Neutron activation analysis also showed that the endosperm contained very little Ca in relation to the other three elements. Surface sterilization and soaking treatments brought about slight loss of Ca but substantial loss of K from embryos. Over 6 days of growth the seedling plant gained minerals from the endosperm.  相似文献

Picris divaricata Vant., a plant species native to subtropical China, was recently identified as the first Cd/Zn hyperaccumulator from Asteraceae. P. divaricata was grown from wild collected seed for 4 months in a series of pH adjusted test soils with added Zn levels 0–7000 mg kg−1 and Cd levels 0–150 mg kg−1. Plants did not hyperaccumulate Zn (threshold >3000 μg g−1) and weakly hyperaccumulated Cd with little or no dose–response.P. divaricata has multicellular simple trichomes concentrated on the leaf margins and midrib. X-ray analysis showed that Zn was concentrated in larger trichomes and epidermal cells adjacent to the trichome but virtually absent in other leaf tissues. Within the trichomes, Zn was localized in ovate spots around the tips of individual cells. These tips and other locations in the trichome cell contained black electron dense material when examined with transmission electron microscopy, some of which was identified as SiO2. Silicon and Mn were concentrated in the same areas as Zn. Si has been previously associated with alleviating Zn, Mn and Cd toxicity. Our results support this observation and further investigation is warranted.Calcium and P were concentrated in the distal tips of trichomes, similar to patterns previously observed for calcicole plants grown in elevated Ca soils. Overall, nonsecretory trichomes from many plant families may have a common origin as tissues adapted to handle a variety of environmental metals.  相似文献

In tobacco (Nicotiana tabacum L.), long and short trichomes can be distinguished morphologically. The established function of long trichomes is to exude a sticky gum containing diterpenes, whereas that of short trichomes is not known. When tobacco seedlings were exposed to toxic levels of cadmium (Cd), growth was retarded, but trichome number was increased up to 2-fold in comparison with untreated samples. Observation by variable-pressure scanning electron microscopy (VP-SEM) indicated that large crystals of 150 μm in size were formed on head cells of both short and long trichomes. An energy-dispersive X-ray analysis system fitted with VP-SEM revealed the crystals to contain amounts of Cd and calcium (Ca) at much higher concentrations than in the head cells themselves. Transmission electron microscopy demonstrated crystal formation in amorphous osmiophilic deposits in vacuoles. When seedlings were treated with Cd in the presence of Ca, tolerance was increased in proportion to the increase in Ca concentration. These results indicate that tobacco plants actively exclude toxic Cd by forming and excreting Cd/Ca-containing crystals through the head cells of trichomes. Crop production in red soil areas may be limited by Al toxicity. A possible alternative to ameliorate Al toxicity is the application of such organic manure as crop straw and animal manure. A pot experiment was conducted to investigate the effects of organic materials on the alleviation of Al toxicity in acid red soil. Ground wheat straw, pig manure or CaCO3 were mixed with the soil and incubated, at 85% of water holding capacity and 25 degrees C, for 8 weeks. After the incubation, 14 seedlings of mung bean (Phaseolus aures Roxb) were allowed to grow for 12 days. Results showed that application of organic material or CaCO3 increased soil pH and decreased soil monomeric inorganic Al concentrations. Growth of mung bean seedling was improved sustantially by the application of organic material or CaCO3. Pig manure or wheat straw was more effective in ameliorating Al toxicity than was CaCO3. Mung bean plants receiving pig manure or wheat straw contained relatively high concentrations of P, Ca and K in their leaves. It is suggested that the beneficial effect of organic manure on mung bean is likely due to decreasing concentrations of monomeric inorganic Al concentrations in soil solution and improvement of mineral nutrition.  相似文献

Zinc can be toxic to plants growing on soils in areas of the Guadiamar River valley (southwestern Spain) affected by the spillage of pyritic sludge in April 1998. The shoots and the soil around the roots of two wild plants (viz. Amaranthus blitoides S. Wats., November 2000; and Xanthium strumarium L., June 2001) growing in the sludge-affected areas were sampled with the purpose of relating Zn phytoavailability to soil properties. The soils were calcareous and non-calcareous Entisols and Inceptisols which, after remediation, contained ploughed-in residual sludge and unevenly distributed industrial lime. Chemical extracts from the soils suggested that much of the sphalerite (ZnS) originally present in the sludge had weathered and Zn was partly bound to carbonates and Fe oxides, the total Zn concentration ranging from 37 to 2407 mg kg –1. To identify the soil properties that influenced Zn phytoavailability under controlled conditions, the soil samples (n=63) were homogenized and oilseed rape (Brassica napus var. Karola) was pot-grown on them in a growth chamber. The concentrations of Zn in oilseed rape shoots and roots were below phytotoxic levels, with mean ± standard deviation values of 142 ± 128 and 244 ± 328 mg kg –1 dry matter, respectively. Citrate/bicarbonate-extractable Zn in soil (Zn cb) was found to be the best predictor for the Zn concentration in both shoots and roots. Also, the Zn cb/Olsen P ratio exhibited a high predictive power for Zn in shoots as the likely result of the Zn-P interaction in soil. The shoot Zn concentration in the wild plants, generally lay below phytotoxic levels (the mean ± standard deviation values were 261 ± 255 and 200 ± 228 mg kg –1 dry matter for Amaranthus blitoides and Xanthium strumarium, respectively) and was not correlated with soil properties – by exception, there was slight correlation between the Zn concentration in Amaranthus blitoides and Zn cb/Olsen P. Such a lack of correlation can be ascribed to the local small-scale soil heterogeneity caused by remediation practices. The Zn concentration in wild plants growing on CaCO 3-poor soils was weakly correlated with Zn cb/Olsen P; no similar correlation was found in CaCO 3-rich soils, however. The wild plants growing on CaCO 3-poor and CaCO 3-rich soils differed little in Zn concentration; this suggests that further addition of lime to reduce Zn phytoavailability may be unjustified.  相似文献

A study on the types and distribution of trichomes, pollen morphology, chemical composition of essential oil (analyzed by GC-FID and GC/MS), and the content of macroelements (Na, K, Ca, and Mg) and trace elements (B, Fe, Cu, Mn, Zn, Al, Pb, Cr, Cd, Ni, Hg, and As) analyzed by ICP-AES (=inductively coupled plasma atomic emission spectroscopy) was conducted on Micromeria croatica (Pers.) Schott. Non-glandular trichomes, peltate trichomes (typical hairs of Lamiaceae), small capitate trichomes (composed of one basal epidermal cell and one head cell), and larger capitate trichomes (composed of one basal epidermal cell, two to three stalk cells, and one head cell with subcuticular space) were observed on leaves, stems, the calyx, and corolla. Pollen of M. croatica had six apertures (hexacolpate) which were set in the equatorial pollen belt (zonocolpate) and showed medium reticulate ornamentation (hetrobrochate type). The essential oil contained from 27 to 39?constituents, the most representative of which were caryophyllene oxide and E-caryophyllene. Among the investigated macroelements, the content of K was highest (8730-10080?mg/kg). The content of trace elements ranged from 0.12?mg/kg (Cr) to 78.00?mg/kg (Fe), while the content of Cd, Ni, Hg, and As were lower than the limit of quantification.  相似文献

Microbial aerobic methane oxidation (MOx) is intrinsically coupled to the production of carbon dioxide, favoring carbonate dissolution. Recently, microbial organic polymers were shown to be able to induce carbonate dissolution. To discriminate between different mechanisms causing calcite dissolution, experiments were conducted in the presence of solid calcite with (1) actively growing cells (2) starving cells, and (3) dead cells of the methanotrophic bacterium Methylosinus trichosporium under brackish conditions (salinity 10) near calcite saturation (saturation state (Ω) 1.76 to 2.22). Total alkalinity and the amount of dissolved calcium markedly increased in all experiments containing M. trichosporium cells. After initial system equilibration, similar calcite dissolution rates, ranging between 20.16 (dead cells) and 25.68 μmol L?1 d?1 (actively growing cells), were observed. Although concentrations of transparent exopolymer particles declined with time in the presence of actively growing and starving cells, they increased in experiments with dead cells. Scanning electron microscopy images of calcite crystals revealed visible surface corrosion after exposure to live and dead M. trichosporium cells. The results of this study indicate a strong potential for microbial MOx to affect calcite stability negatively, facilitating calcite dissolution. In addition to CO2 production by methanotrophically active cells, we suggest that the release of acidic or Ca2+-chelating organic carbon compounds from dead cells could also enhance calcite dissolution.  相似文献

Regulation of the concentration of Ca2+ in the apoplast of plants is essential in order to allow Ca(2+)-dependent processes, such as Ca(2+)-mediated signal transduction in stomatal guard cells, to function correctly. This is particularly important for plants growing with high levels of Ca2+ in the rhizosphere. Recently, we have shown that in two calcicoles, Leontodon hispidus L. and Centaurea scabiosa L., trichomes play a key role in this regulatory process. Ozone is known to have a marked effect on plant Ca2+ homeostasis. Therefore, we have examined the effect of this pollutant on the regulation by trichomes of apoplastic Ca2+ in the calcicole L. hispidus. Treatment with 100 nl l-1 ozone resulted in a reduction in stomatal conductance of approximately 25% in plants grown with 15 mM Ca2+ in the rhizosphere. Analysis of total Ca2+ levels revealed that these changes in stomatal behaviour reflect a decrease in the ability of trichomes to sequester Ca2+. The amount of Ca2+ present in the trichome tip cell was reduced by approximately 38%. This was accompanied by an increase in the levels of Ca2+ in the guard cells and other tissues of the leaf. These data suggest that ozone has a detrimental effect on the ability of trichomes to regulate the concentration of apoplastic Ca2+ in L. hispidus, resulting in altered stomatal behaviour, and hence gaseous exchange, possibly due to the disruption of guard-cell Ca(2+)-mediated signal transduction. This has important implications for the growth and survival of plants growing in Ca(2+)-rich soils.  相似文献

Satureja montana and S. subspicata are used as spice, pepper substitute, for preparing tea, juice, and as a medicine. Fourteen populations (seven per species) of Satureja montana L. and S. subspicata Vis . growing in Croatia were examined to determine the chemical composition of the essential oil (analyzed by GC‐FID and GC/MS), the content of macroelements (Na, K, Ca, Mg) and trace elements (B, Fe, Cu, Mn, Zn, Al, Pb, Cr, Cd, Ni, Hg, As) analyzed by ICP‐AES, antioxidant compounds (analyzed by UV/VIS spectrophotometer), and the types and distribution of trichomes (analyzed by scanning electron microscopy). The main constituents of the essential oil were carvacrol and thymol in S. montana (all populations belong to one phenol chemotype), while α‐eudesmol, β‐eudesmol, and spathulenol dominated in S. subspicata (three chemotypes could be distinguished). Both species possess considerably higher quantities of Ca and Mg, and moderate concentrations of K and Na, while Hg and As levels were below the limit of quantification. Non‐glandular trichomes, peltate trichomes, and three types of capitate trichomes were observed on leaves, stem, calyx, and corolla.  相似文献
