The distribution of nitrification potential (NP) with depth in sediment and season was investigated in a shallow sandy sediment (0.5 m water) and a deeper muddy sediment (17m water). In both sediments, nitrifying bacteria were present in the anoxic strata (oxygen penetration was 5 mm below the surface). The NP at 6–8 cm depth in the sediment was 50% and 10% of the surface NP at the sandy and muddy sediment, respectively. It is suggested that bioturbation and physical disturbance of the sediment were the most likely reasons for this distribution. The NP increased as sediment temperature decreased. This effect was less marked in the muddy sediment. It is concluded that during the summer, the numbers or specific activity of nitrifying bacteria diminished for the following reasons: There was decreased O2 penetration into the sediment and increased competition for O2 by heterotrophs; there was increased competition for NH4 + and there was inhibition by H2S. These effects counteracted the potentially higher growth rates and increased rates of NH4 + production at the elevated summer temperatures. The potential nitrification rates in the upper 1 cm, which were measured at 22°C, were converted to calculated rates at the in situ temperature (Q10=2.5) and in situ oxygen penetration. These calculated rates were shown to closely resemble the measured in situ rates of nitrification. The relationship between the in situ rates of nitrification and the nitrification potential is discussed.  相似文献

Summary The rate of nitrification by soil aggregates at pH 5.5 and 7.5 was examined by a perfusion technique. Nitrification occurred at both levels of H-ion concentration but at the higher pH the rate of nitrification was greater. The population estimates of nitrifying bacteria were correspondingly greater at the high pH. Once the pH was lowered from 7.5 to 5.5, the nitrification rate decreased slowly with a corresponding decrease in the numbers of nitrifying bacteria. The distribution of nitrifying bacteria throughout soil aggregates was homogenous. The lower limit of pH for nitrification was 4.3.  相似文献

Summary Mineralization of soil nitrogen studies with two acid sulfate soils under anaerobic and aerobic incubation at 30°C for 2 weeks showed that the mineral N was released and accumulated entirely as NH 4 + in both soils. Nitrification did not occur in either of the soils under conditions that stimulate nitrification. The acid sulfate soils studied release good amounts of mineralizable N, and, because of lack of nitrifying activity, denitrification may not be a serious problem in these soils.  相似文献

This study examined how sediment-sorbed PCBs and several large storms affected sediment nutrient dynamics based on potential nitrification rates and benthic flux measurements. PCBs were hypothesized to negatively affect potential nitrification rates due to the sensitivity of nitrifying bacteria. Sediment disturbance caused by the succession of storms, which can enhance nutrient inputs and phytoplankton production, was hypothesized to enhance both potential nitrification rates and benthic flux measurements as a result of higher nutrient and organic matter concentrations. Potential nitrification rates, benthic fluxes (NO3  + NO2 , NH4 +, and DIP), sediment PCB content, water content, organic content, salinity, bottom water dissolved oxygen, and sediment chlorophyll were measured at 13 different sites in Escambia Bay during the summer of 2005. Potential nitrification rates were highest at deep, organic-rich sites. Total PCB content did not have a direct effect on potential nitrification rates. An analysis of recent changes in benthic processes in relation to extreme meteorological events was performed by comparing the 2005 results with data from 2000, 2003, and 2004. Storm effects on sediment biogeochemistry were mixed with sediment nitrogen dynamics enhanced at some sites but not others. In addition, SOC and NH4 + fluxes increased in deeper channel sites after Hurricanes Ivan and Dennis, which could be attributed to the deposition of phytoplankton blooms. Sediment nutrient dynamics in Escambia Bay appear to be resilient to these extreme meteorological events since there were no significant effects on sediment processes in the Bay as a whole. The discovery that photosynthetic bacterial membrane-bound inorganic pyrophosphatase (PPase) catalyzed light-induced phosphorylation of orthophosphate (Pi) to pyrophosphate (PPi) and the capability of PPi to drive energy requiring dark reactions supported PPi as a possible early alternative to ATP. Like the proton-pumping ATPase, the corresponding membrane-bound PPase also is a H+-pump, and like the Na+-pumping ATPase, it can be a Na+-pump, both in archaeal and bacterial membranes. We suggest that PPi and Na+ transport preceded ATP and H+ transport in association with geochemistry of the Earth at the time of the origin and early evolution of life. Life may have started in connection with early plate tectonic processes coupled to alkaline hydrothermal activity. A hydrothermal environment in which Na+ is abundant exists in sediment-starved subduction zones, like the Mariana forearc in the W Pacific Ocean. It is considered to mimic the Archean Earth. The forearc pore fluids have a pH up to 12.6, a Na+-concentration of 0.7 mol/kg seawater. PPi could have been formed during early subduction of oceanic lithosphere by dehydration of protonated orthophosphates. A key to PPi formation in these geological environments is a low local activity of water.  相似文献

In this study, Na+ uptake and acid–base balance in the euryhaline pupfish Cyprinodon variegatus variegatus were characterized when fish were exposed to pH 4.5 freshwater (7 mM Na+). Similar to the related cyprinodont, Fundulus heteroclitus, Na+ uptake was significantly inhibited when exposed to low pH water. However, it initially appeared that C. v. variegatus increased apparent net acid excretion at low pH relative to circumneutral pH. This result is opposite to previous observations for F. heteroclitus under similar conditions where fish were observed to switch from apparent net H+ excretion at circumneutral pH to apparent net H+ uptake at low pH. Further investigation revealed disparate observations between these studies were the result of using double endpoint titrations to measure titratable alkalinity fluxes in the current study, while the earlier study utilized single endpoint titrations to measure these fluxes (i.e.,. Cyprinodon acid–base transport is qualitatively similar to Fundulus when characterized using single endpoint titrations). This led to a comparative investigation of these two methods. We hypothesized that either the single endpoint methodology was being influenced by a change in the buffer capacity of the water (e.g., mucus being released by the fish) at low pH, or the double endpoint methodology was not properly accounting for ammonia flux by the fish. A series of follow-up experiments indicated that buffer capacity of the water did not change significantly, that excretion of protein (a surrogate for mucus) was actually reduced at low pH, and that the double endpoint methodology does not properly account for NH3 excretion by fish under low pH conditions. As a result, it overestimates net H+ excretion during low pH exposure. After applying the maximum possible correction for this error (i.e., assuming that all ammonia is excreted as NH3), the double endpoint methodology indicates that net H+ transport was reduced to effectively zero in both species at pH 4.5. However, significant differences between the double endpoint (no net H+ transport at low pH) and single endpoint titrations (net H+ uptake at low pH) remain to be explained.  相似文献

Nitric oxide plays several roles in cellular physiology, including control of the vascular tone and defence against pathogen infection. Neuronal, inducible and endothelial nitric oxide synthase (NOS) isoforms synthesize nitric oxide. Cells generate acid and base equivalents, whose physiological intracellular concentrations are kept due to membrane transport systems, including Na+/H+ exchangers and Na+/HCO3? transporters, thus maintaining a physiological pH at the intracellular (~7.0) and extracellular (~7.4) medium. In several pathologies, including cancer, cells are exposed to an extracellular acidic microenvironment, and the role for these membrane transport mechanisms in this phenomenon is likely. As altered NOS expression and activity is seen in cancer cells and because this gas promotes a glycolytic phenotype leading to extracellular acidosis in gynaecological cancer cells, a pro‐inflammatory microenvironment increasing inducible NOS expression in this cell type is feasible. However, whether abnormal control of intracellular and extracellular pH by cancer cells regards with their ability to synthesize or respond to nitric oxide is unknown. We, here, discuss a potential link between pH alterations, pH controlling membrane transport systems and NOS function. We propose a potential association between inducible NOS induction and Na+/H+ exchanger expression and activity in human ovary cancer. A potentiation between nitric oxide generation and the maintenance of a low extracellular pH (i.e. acidic) is proposed to establish a sequence of events in ovarian cancer cells, thus preserving a pro‐proliferative acidic tumour extracellular microenvironment. We suggest that pharmacological therapeutic targeting of Na+/H+ exchangers and inducible NOS may have benefits in human epithelial ovarian cancer.  相似文献

In order to confirm that H+-ATPase plays an important role in the acid tolerance ofStreptococcus bovis , two mutants with low activities of H+-ATPase were isolated by use of ethyl methanesulfonate and neomycin resistance. The activity of H+-ATPase per cellular nitrogen was related to the lowest culture pH permitting growth. A mutant with little H+-ATPase activity (Mutant 2) was unable to grow below pH 5.5, which suggests that the intracellular pH should be maintained above 5.5 in S. bovis. Since lactate dehydrogenase activity, which is important for acid tolerance, was similar in parent and mutant strains, H+-ATPase activity is likely to affect acid tolerance. The amount of H+-ATPase protein as determined by Western-blot analysis with polyclonal antibody, was similar in Mutant 2 and its parent, indicating that H+-ATPase activity per enzyme protein is reduced by mutation. Probably, H+-ATPase synthesis was not changed by mutation. The gene encoding H+-ATPase of Mutant 2 had mutations at positions close to the ATP-binding motif A sequence in the β-subunit, which probably explains the reduced activity of H+-ATPase in this mutant. These results strongly support the assumption that H+-ATPase has a key role in the acid tolerance of S. bovis.  相似文献

The effect of low pH on net H+ release and root growth of corn (Zea mays L.) and broad bean (Vicia faba L.) seedlings was investigated in short-term experiments at constant pH. Broad bean was more sensitive to low pH than corn: the critical values (pH values below which net H+ release and root growth ceased) were pH 4.00 (broad bean) and pH 3.50 (corn) at 1 millimolar Ca2+. Both proton release and root growth were progressively inhibited as the medium pH declined. Additional Ca2+ in the root medium helped to overcome the limitations of low pH for net H+ release and root growth. Potassium (for corn) and abscisic acid (for broad bean) increased both net H+ release and root growth rate at the critical pH value. It is concluded that poor root growth at low pH is caused by a lack of net H+ release that may decrease cytoplasmic pH values. Inhibited net H+ release at high external H+ activity is not due to a shortage of energy supply to the H+ ATPase. Instead, a displacement of Ca2+ by H+ at the external side of the plasmalemma may enhance reentry of H+ into root cells.  相似文献

Rates of nitrification in well drained granitic soils from forest stands and grassland of differing successional status and from beneath isolated individuals of several tree species were compared in a series of laboratory experiments. Fresh samples were perfused with distilled water or nutrient solution for 10 to 14 weeks at 20°C. The following treatments were applied to the soils singly and in combination: 200 and 400 g N g–1 as (NH4)2SO4; 100 g P g–1 as KH2PO4; 4000 g CaCO3 g–1; inoculation of non-nitrifying soil with nitrifying soil; perfusion of nitrifying soil with leachate from non-nitrifying soil.Nitrification was absent or occurred at only a low rate in many soils; it generally increased as succession proceeded from nature grassland or eucalypt forest towards climax temperate rainforest, but decreased in mature climax forests. However, the influence of individual tree species was often paramount. Nitrification was stimulated by disturbance of a stand by disease. A possible inhibitor of nitrification in a rainforest soil could not be removed by leaching with water, nor transferred via the leachate to a nitrifying soil. Addition of P was without effect on either total amount of nitrate produced or on net mineralisation of soil N, but sometimes increased the rate of nitrification of added ammonium. Non-nitrifying rainforest soil of pH 4.3 was induced to nitrify only after addition of (NH4)2SO4, inoculation with a nitrifying soil, and addition of CaCO3 to raise pH by 3 units. However, once nitrification had commenced it could continue with little change in rate while pH decreased to a value of 3.4.It was concluded that rate of nitrification is dependent upon the presence of particular tree species in a stand, upon its history of disturbance, and hence in part upon the stand's successional status. It is not limited by pHper se within the range found in these soils, although an increase in pH may be necessary to initiate nitrification. In some soils the rate of nitrification may be limited by the level of ammonium substrate, and nitrifiers are virtually absent from others. Overall microbial activity is limited by lack of utilisable carbon substrate.  相似文献

Abstract No autotrophic nitrifying organisms were found in a podzolic brown earth forming nitrate. 350 fungi and aerobic heterotrophic bacteria were isolated from this soil and examined for their nitrifying abilities. About one quarter of the isolates produced 0.05–0.90 mg N·1−1 nitrite or nitrate in peptone solution, soil extract mixture or sterilised soil. The nitrification rate of the most active fungus, Verticillium lecanii , was highest at pH 3.5 in defined media. The results support the significance of heterotrophic nitrification in acid soils.  相似文献

Factors affecting heterotrophic nitrification by Alcaligenes faecalis OKK17, which was isolated from sewage sludge, were examined. Specific nitrifying activity increased as the pH increased up to 8.5. Most of the nitrogenous compounds (88%) in the culture supernatant were converted to hydroxylamine or nitrite at pH 9 but 87% of them remained as ammonium at pH 7. These results imply that the substrate for heterotrophic nitrification is ammonium and that the organism oxidizes ammonium to lower its toxic effect. Although the addition of acetate to a defined medium increased growth of the bacterium up to C/N = ca. 6, the accumulation of nitrification products almost paralleled the growth and the specific nitrifying activity decreased. Pyruvate and oxaloacetate increased the specific nitrifying activity six- to eightfold compared with the other organic acids examined, but the key enzyme activities in the glyoxylate cycle were not increased. Acetate, glyoxylate, and malonate did not increase the specific nitrifying activity, but they increased the enzyme activities. These results imply that the involvement of acetate metabolism in the heterotrophic nitrification is unlikely.  相似文献

Summary The conversion of substantial amounts of ammonia nitrogen into organic nitrogen as a direct result of nitrification at neutral pH-values, was monitored in soil suspensions amended with ammonium nitrogen. The influence of the chemolithotrophic nitrifying bacteria was verified by applying nitrapyrin as a selective inhibitor in control experiments. In addition, the role of phenolic compounds was examined by adding α naphthol. The factors influencing the nitrification processi.e. pH, NH 4 + −N, NO 2 −N, NO 3 −N were measured during a 60 days incubation period. Nitrification started to be active after 5 and 10 days in the normal and the naphthol spiked soil suspensions respectively; it was inhibited in the nitrapyrin controls. Parallel with nitrification, formation of organic nitrogen was observed. The humic matter fractions were extracted and analyzed by I.R. spectroscopy which revealed the valence vibration ranges of nitro and nitroso groups fixed in different positions on aromatic compounds, both for normal and naphthol spiked samples. High resolution gas chromatography combined with mass spectroscopic analysis indicated the formation of nitrosonaphtholes. In addition a novel organic nitro compound was identifiedi.e. an azido nitro benzene. No nitrogen was fixed in the samples treated with nitrification inhibitor. A mechanism for the fixation of nitrite nitrogen during nitrification is proposed.  相似文献

In this study, two laboratory scale sequencing batch reactors (SBRs) were conducted to study the stability of aerobic granules. The strategy was involved in stepwise increase in ammonium (NH4+-N) concentration in the influent. Results showed that the activity of nitrifying bacteria and diameter of the aerobic granules significantly increased with gradually increasing NH4+-N, which reached persistently new balances by homeostasis. As a result, the stability of aerobic granules was remarkably enhanced. The value of sludge volume index (SVI) was below 25 ml/g, the mean settling velocity was excellent up to 107 m/h. The NH4+-N removal efficiency averaged above 99% and total nitrogen (TN) removal was greatly enhanced and could reach 68%. Besides dissolved oxygen, the granules size was also a dominant factor to influence denitrification, which could gradually increase in variable conditions through homeostasis. Stable, dense and well-settling nitrifying granules can be developed for simultaneous nitrification and denitrification removal.  相似文献

Na+/H+ antiporters are integral membrane proteins that are present in almost every cell and in every kingdom of life. They are essential for the regulation of intracellular pH-value, Na+-concentration and cell volume. These secondary active transporters exchange sodium ions against protons via an alternating access mechanism, which is not understood in full detail. Na+/H+ antiporters show distinct species-specific transport characteristics and regulatory properties that correlate with respective physiological functions. Here we present the characterization of the Na+/H+ antiporter NhaA from Salmonella enterica serovar Thyphimurium LT2, the causing agent of food-born human gastroenteritis and typhoid like infections. The recombinant antiporter was functional in vivo and in vitro. Expression of its gene complemented the Na+-sensitive phenotype of an E. coli strain that lacks the main Na+/H+ antiporters. Purified to homogeneity, the antiporter was a dimer in solution as accurately determined by size-exclusion chromatography combined with multi-angle laser-light scattering and refractive index monitoring. The purified antiporter was fully capable of electrogenic Na+(Li+)/H+-antiport when reconstituted in proteoliposomes and assayed by solid-supported membrane-based electrophysiological measurements. Transport activity was inhibited by 2-aminoperimidine. The recorded negative currents were in agreement with a 1Na+(Li+)/2H+ stoichiometry. Transport activity was low at pH 7 and up-regulation above this pH value was accompanied by a nearly 10-fold decrease of Km Na (16 mM at pH 8.5) supporting a competitive substrate binding mechanism. K+ does not affect Na+ affinity or transport of substrate cations, indicating that selectivity of the antiport arises from the substrate binding step. In contrast to homologous E. coli NhaA, transport activity remains high at pH values above 8.5. The antiporter from S. Typhimurium is a promising candidate for combined structural and functional studies to contribute to the elucidation of the mechanism of pH-dependent Na+/H+ antiporters and to provide insights in the molecular basis of species-specific growth and survival strategies.  相似文献

Aims and background

The ability to suppress soil nitrification through the release of nitrification inhibitors from plant roots is termed ‘biological nitrification inhibition’ (BNI). Earlier, we reported that sorghum roots release higher BNI-activity when grown with NH 4 + , but not with NO 3 - as N source. Also for BNI release, rhizosphere pH of <5.0 is needed; beyond this, a negative effect on BNI release was observed with nearly 80% loss of BNI activity at pH >7.0. This study is aimed at understanding the inter-functional relationships associated with NH 4 + uptake, rhizosphere-pH and plasma membrane H+-ATPase (PM H+-ATPase) activity in regulating the release of BNIs (biological nitrification inhibitors) from sorghum roots.

Methods

Sorghum was grown hydroponically and root exudates were collected from intact plants using a pH-stat system to separate the secondary acidification effects by NH 4 + uptake on BNIs release. A recombinant luminescent Nitrosomonas europaea bioassay was used to determine BNI-activity. Root plasma membrane was isolated using a two-phase partitioning system. Hydrolytic H+-ATPase activity was determined. Split-root system setup was deployed to understand the localized responses to NH 4 + , H+-ATPase-stimulator (fusicoccin) or H+-ATPase-inhibitor (vanadates) on BNI release by sorghum.

Results

Presence of NH 4 + in the rhizosphere stimulated the expression of H+-ATPase activity and enhanced the release of BNIs from sorghum roots. Fusicoccin, which stimulates H+-ATPase activity, also stimulated BNIs release in the absence of NH 4 + ; vanadate, which suppresses H+-ATPase activity, also suppressed the release of BNIs. NH 4 + levels (in rhizosphere) positively influenced BNIs release and root H+-ATPase activity in the concentration range of 0-1.0 mM, indicating a close relationship between BNI release and root H+-ATPase activity with a possible involvement of carrier-mediated transport for the release of BNIs in sorghum.

Conclusion

Our results suggest that NH 4 + uptake, PM H+-ATPase activity, and rhizosphere acidification are functionally inter-connected with BNI release in sorghum. Such knowledge is critical to gain insights into why BNI function is more effective in light-textured, mildly acidic soils compared to other soil types.  相似文献

Up-flow oxygen-controlled biofilm reactors equipped with a non-woven fabric support were used as a single reactor system for autotrophic nitrogen removal based on a combined partial nitrification and anaerobic ammonium oxidation (anammox) reaction. The up-flow biofilm reactors were initiated as either a partial nitrifying reactor or an anammox reactor, respectively, and simultaneous partial nitrification and anammox was established by careful control of the aeration rate. The combined partial nitrification and anammox reaction was successfully developed in both biofilm reactors without additional biomass inoculation. The reactor initiated as the anammox reactor gave a slightly higher and more stable mean nitrogen removal rate of 0.35 (± 0.19) kg-N m−3 d−1 than the reactor initiated as the partial nitrifying reactor (0.23 (± 0.16) kg-N m−3 d−1). FISH analysis revealed that the biofilm in the reactor started as the anammox reactor were composed of anammox bacteria located in inner anoxic layers that were surrounded by surface aerobic AOB layers, whereas AOB and anammox bacteria were mixed without a distinguishable niche in the biofilm in the reactor started as the partial nitrifying reactor. However, it was difficult to efficiently maintain the stable partial nitrification owing to inefficient aeration in the reactor, which is a key to development of the combined partial nitrification and anammox reaction in a single biofilm reactor.  相似文献

Kinetic investigations on growth parameters of nitrifying and COD oxidizing bacteria were carried out with recourse to a three stage reciprocating jet bioreactor system using real life wastewater. The system employed in this investigation essentially consisted of separate aerobic oxidation stage along with nitrification stage and anaerobic denitrification stage with facility for biomass recirculation whenever necessary. Steady-state COD oxidation reactor performance was assessed for various values of residence time. Yield coefficient and decay coefficient of COD oxidizing biomass were obtained as 0.3329 kg BM/kg COD and 0.0032 (1/h) respectively.It was observed that COD oxidizing bacteria co-existed with nitrifying bacteria during nitrification process due to the nature of wastewater used. Steady-state nitrification reactor performance was also assessed for various residence time values. Exact concentration of nitrifying and COD oxidizing biomass in the nitrification reactor was then estimated with the help of kinetic growth parameters of COD oxidizing biomass and extent of COD oxidation achieved in nitrification reactor. This further enabled evaluation of corrected kinetic growth parameters estimated as 0.4272 kg BM/kg NH 4 + -N and 0.00626 (1/h) for nitrifier biomass yield coefficient and decay coefficient respectively.  相似文献

The efficiency of nitrification in a fixed bed reactor was compared to that found in an activated sludge reactor to determine their sensitivity to changing loads and lower temperatures. Two structurally identical lab‐scale systems, using the anaerobic/anoxic/aerobic (AAO) process to remove nitrogen and phosphorus simultaneously, were operated in parallel with secondary clarifiers and sludge return. The first aerobic system was operated as an activated sludge reactor, the second system as a fixed bed reactor. The aerobic fixed bed reactor was filled with porous ceramic materials for the immobilisation of predominantly nitrifying bacteria. The removal efficiencies of 99 % NH4+‐N, 90 % DOC, and 98 % PO43–‐P for normal loads of 0.11 g/L d DOC, 0.06 g/L d NH4+‐N, and 0.0054 g/L d PO43–‐P were achieved for both systems. However, the system with an aerobic fixed bed reactor was characterised by the following advantages over the system with an activated sludge reactor: a shorter time to reach almost complete nitrification, a higher nitrification rate at higher loads of NH4+‐N and a lower sensitivity of nitrification at lower temperatures down to 12 °C.  相似文献

The effects of H ion were studied by lowering the soil pH in a deciduous forest. Increased H+ burden significantly reduced the dry mass of herbaceous vegetation. Increased H+ significantly mobilized Ca (226%) and Mg (244%) to the deeper soil profile, thus reducing uptake of those nutrients by intact vegetation. Moreover, the low pH also had a drastic impact on nitrifiers and nitrification. The 70% reduction in NO-3-N was directly related to the reduction in numbers of Nitrosomonas (91%) and Nitrobacter (100%). Ammonium N also declined under H+ stress, indicating a reduction in N-mineralization process. It was hypothesized that increased H+ stress in deciduous forests may deplete major nutrients, mobilize micronutrients, reduce soil microbial populations and decrease plant biomass. Thus a deciduous forest may evolve into a nutrient-poor soil ecosystem and may shift toward a “semi-podzol ecosystem.”  相似文献
