The anterior cruciate ligament (ACL) is necessary for normal knee stability and movement. Unfortunately the ACL is also the most frequently injured ligament of the knee with severe disruptions requiring surgical intervention. In response to this, tissue engineering has emerged as an option for ACL replacement and repair. In this study we present a novel hydrogel-fibrous scaffold as a potential option for ACL replacement. The scaffold was composed of PLLA fibers, in a previously evaluated braid-twist structure, combined with a polyethylene glycol diacrylate (PEGDA) hydrogel to improve viscoelastic properties. Both hydrogel concentration (10%, 15%, and 20%) and amount of hydrogel (soaking the fibrous scaffold in hydrogel solution or encasing the scaffold in a block of hydrogel) were evaluated. It was found that the braid-twist scaffold had a greater porosity and larger number of pores above 100 μm than braided scaffolds with the same braiding angle. After testing for their effects on swelling, fiber degradation, and protein release, as well as viscoelastic and tensile testing (when combined with fibrous scaffolds), it was found that the composite scaffold soaked in 10% hydrogel had the best chemical release and mechanical properties. The optimized structure behaved similarly to natural ligament in tension with the addition of the hydrogel decreasing the ultimate tensile stress (UTS), but the UTS was still comparable to natural ACL. The current investigation was designed to further examine the reliability of the Robinson protocol, which is a run-to-exhaustion treadmill test. Robinson (10) originally examined this protocol with 5 subjects. The significance of the initial exploratory study was the impetus for expanding the study to examine the reliability of the protocol with a larger sample. Fifteen male subjects participated in 3 trial runs on the treadmill. The first trial was a modified McConnell (7) test to determine the aerobic capacity of each subject. The second and third trials were identical Robinson protocols (10). The first trial run mean, in seconds (262.04 +/- 74.50), was not significantly different from the second trial run mean (257.30 +/- 72.65), p = 0.526 (2 tailed). As expected, trial 1 and trial 2 were highly correlated (intraclass) (r = 0.927, p < 0.001). These results provide additional support for the hypothesis that the Robinson protocol with a greater subject pool is a reliable protocol that can be used in research studies interested in examining various physiological interventions or anaerobic training. 相似文献

We have developed a method to quantify the dynamic interaction between fingertip force magnitude (strength) and directional control (dexterity) during pinch with a novel strength-dexterity (S-D) test based on the principle of buckling of compression springs. The test consists of asking participants to use key and opposition pinch to attempt to fully compress springs, in random order, with a wide range of combinations of strength and dexterity requirements. The minimum force required to fully compress the spring and the propensity of the spring to buckle define the strength and dexterity requirements, respectively. The S-D score for each pinch style was the sum of the strength values of all springs successfully compressed fully. We tested 3 participant groups: 18 unimpaired young adults (40yr), and 14 adults diagnosed with carpo-metacarpal osteoarthritis (CMC OA) (>or = 36yr). We investigated the repeatability of the S-D test with 74 springs by testing 14 young adults twice on different days. The per-spring repeatability across subjects was >or = 94%. A minimum performance score for young adults was found as they all could compress a subset of 39 springs. Using this subset of springs, we compared the ability of the S-D score vs. maximal pinch force values to distinguish unimpaired hands from those with CMC OA of the thumb. The score for this 39-spring S-D test distinguished between CMC OA and asymptomatic older adults, whereas pinch meter readings did not (p<0.05). We conclude that the S-D test is repeatable and applicable to clinical research. We propose including the S-D test in studies aiming to quantify impairment and compare treatment outcomes in orthopaedic and neurological afflictions that degrade dynamic manipulation. 相似文献

The mouse spot test. Evaluation of its performance in identifying chemical mutagens and carcinogens 总被引:1,自引:0,他引:1
The published results on 60 chemicals and X-rays investigated in the mouse spot test were compared with data on the same chemicals tested in the bacterial mutation assay (Ames test) and lifetime rodent bioassays. The performance of the spot test as an in vivo complementary assay to the in vitro bacterial mutagenesis test reveals that of 60 agents, 38 were positive in both systems, 6 were positive only in the spot test, 10 were positive only in the bacterial test and 6 were negative in both assays. The spot test was also considered as a predictor of carcinogenesis; 45 chemicals were carcinogenic of which 35 were detected as positive by the spot test and 3 out of 6 non-carcinogens were correctly identified as negative. If the results are regarded in sequence, i.e. that a positive result in a bacterial mutagenicity test reveals potential that may or may not be realized in vivo, then 48 chemicals were mutagenic in the bacterial mutation assay of which 38 were active in the spot test and 31 were confirmed as carcinogens in bioassays. 12 chemicals were non-mutagenic to bacteria of which 6 gave positive responses in the spot test and 5 were confirmed as carcinogens. These results provide strong evidence that the mouse coat spot test is an effective complementary test to the bacterial mutagenesis assay for the detection of genotoxic chemicals and as a confirmatory test for the identification of carcinogens. The main deficiency at present is the paucity of data from the testing of non-carcinogens. With further development and improvement of the test it is probable that the predictive performance of the assay in identifying carcinogens should improve, since many of the false negative responses may be due to inadequate testing. 相似文献

A short term small volume14C-assimilation algal toxicity test usingSelenastrum capricomutum CCAP 278/4 as test organism was used to determine the potential toxicity of a few selected compounds. Generally the sensitivity of the method increased with increasing exposure time to the substances tested. It was found that the method complied with the prerequisites of an acute test, because in most instances toxicity could be detected within 0.5 h exposure. The steel companies are becoming increasingly aware about the sustainability challenges. In order to become a responsible corporate citizen, the industry has responded to these challenges through adoption of pillars of sustainability. The industry has made the beginning with identification of sustainability indicators. The indicators have been developed specifically for steel industry. Generally, it is quite difficult to evaluate the performance of company on the basis of large number of sustainability indicators. Integration of key sustainability indicators is quite essential for decision-making. Composite indicators are an innovative approach to evaluate sustainable performance. This paper presents a method for development of composite sustainability performance index (CSPI) that addresses the sustainable performance of steel industries along all the three pillars of sustainability—economic, environmental and societal. Organizational governance and technical aspects have also been considered fourth and fifth dimensions of sustainability.The objective of this paper is to introduce sustainability and to present a conceptual decision model, using analytical hierarchy process (AHP) to assist in evaluating the impact of an organization's sustainability performance. AHP has been used to determine the weights at various levels. Sub-indices have been evaluated and aggregated to form CSPI. Preference and performance of a gall-inducing sawfly: a test of the plant vigor hypothesis 总被引:2,自引:0,他引:2
To test the Plant Vigor Hypothesis, we determined female oviposition preference of Phyllocolpa leavitii (Hymenoptera: Tenthredinidae) for shoot lengths on 20 clones of Salix discolor (Salicaceae) and examined larval performance by determining larval survival based on clone, shoot length, and leaf length for each Phyllocolpa gall observed. Sawfly galls were found on significantly longer shoots compared to shoots without galls, and shoots with 2, 3, 4, and 5 galls were successively longer than shoots with fewer galls. Furthermore, a much higher proportion of longer shoots had galls that did shorter shoots. These data demonstrate strong sawfly oviposition preference for long shoots. However, when shoot length was adjusted for numbers of available leaves per shoot, the number of galls per available leaf showed a weaker, but still positive relationship with shoot size. Using a logistic regression on survival of Phyllocolpa larvae, we detected highly significant effects of clone, shoot length, leaf length, and the square of leaf length on survival. Additionally, much higher sawfly survival on the long shoots of one clone caused a significant clone by shoot-length interaction. Survival was positively correlated with shoot length, indicating directional selection favoring oviposition on longer shoots; still, shoot length explained only 11.8% of the variation in survival. The directional component of selection favored oviposition on smaller leaves as survival was inversely related to leaf length; however, disruptive selection for leaf length oviposition preference was also detected, with survival of Phyllocolpa galls lowest on intermediate leaf lengths. This study provided evidence supporting the preference prediction of the Plant Vigor Hypothesis. However, much of the data contradicted the performance prediction of the Plant Vigor Hypothesis, with only a modest amount of evidence supporting the performance prediction. 相似文献

An epidemiologic investigation was carried out in Modena (Italy) to evaluate the prevalence of faecal VEROtoxin (FVT) in diarrhoeal stool specimens. One thousand and sixty-six stool specimens, submitted to the Clinical Microbiology Laboratory of the University Hospital of Modena, were collected and faecal filtrates tested for neutralizable cytotoxin by a toxicity test on VERO cells. Cytopathic effect on VERO cells was produced by 301 stool specimens (28%); neutralizable VT was detected in 40 (13%) out of 301 positive samples (3.7% of 1066 specimens). The prevalent FVT type was VT2 (50%), followed by VT1 (32.5%) and VT1+2 (17.5%). We evaluated an assay that detects both VTs directly from stool specimens to demonstrate that enterohemorrhagic strains (EHEC) should be considerated a causative agent of sporadic non-bloody diarrhoea. Our results suggest that toxin neutralization assay is a sensitive and specific technique and may be used as an alternative method to diagnose diarrhoeal infections caused by EHEC. 相似文献

Exercise scientists frequently use criterion measure tests to assess levels of motor performance and physical fitness or to evaluate the success of their intervention programs. During this process, the same motor test might be administered multiple times. The aim of the present study was to examine whether familiarization with a psychomotor test would influence performance and/or learning. One hundred thirty (n = 130) primary school students participated in the study. Participants were randomly divided into two groups, experimental (n = 60, mean age = 10.74 years old, minimum = 9.08, maximum = 11.83) and control (n = 70, mean age = 10.68 years old, minimum = 9.5, maximum = 11.79). The flamingo balance test was used to examine the postulated hypothesis. Initially, both groups executed the balance test. Then, only the experimental group performed two additional trials, one trial every 3 days. A final trial was administered to all participants, followed by a retention trial 1 week later. The results revealed that both groups improved their performance. The experimental group outperformed the control group. The retention trial showed that performance levels for both groups continued to improve, suggesting that familiarization with the test influenced not only performance but learning as well. Exercise scientists who implement psychomotor tests, such as balance tests, for evaluative purposes should be aware that performance improvement does not necessary represent changes in individuals' performance, but it may reflect individuals' familiarization with the test. 相似文献

A test battery was constructed on a palmtop computer for ambulatory purposes. This study explored whether the test battery could assess circadian rhythmicity under constant routine conditions. Performance, body temperature, and subjective sleepiness of 12 healthy subjects were measured. The test battery consisted of a sleepiness questionnaire and three performance tests: a vigilance detection test, a working memory test, and a choice-reaction time test. The subjects were divided into early-start and late-start groups and were subjected to the constant-routine protocol. All tests showed a trough in performance in the early morning around 07:00h and a peak in the evening between 21:00 and 23:00h. In addition, an afternoon decrement in performance was observed between 15:00 and 17:00h. On average, the circadian (peak-to-trough) variation of the performance variables amounted to 16.9%±1.7 SEM of the maximum across subjects. The late starters showed a larger impairment in performance during the morning than the early starters. This could be attributed to prior wakefulness. The characteristics of the performance rhythms found in this study replicate findings in several other studies carried out under constant routine conditions. In conclusion, the present test battery appeared to be a good tool for future assessment of performance under natural conditions. 相似文献

Pregnant and nonpregnant ewes were injected with luteinizing hormone-releasing hormone (LHRH). Pituitary responsiveness, based on serum luteinizing hormone (LH), and follicle stimulating hormone (FSH) concentration, 2 hr after injection was then determined for each ewe, by radioimmunoassay (RIA) and correlated with the physiological reproductive state of each ewe. The serum LH release in pregnant ewes was significantly lower than that in nonpregnant ewes. Serum LH concentrations of pregnant ewes were further categorized according to whether the ewes were multiple (ML) or single lambing (SL). The responses by ML ewes were lower for LH than the SL responses. Follicle stimulating hormone responses were not different between pregnant or nonpregnant groups. Luteinizing hormone responses between pregnant ewes which were grouped according to 3 stages of pregnancy (1 to 5, 5 to 10 and 10 to 15 weeks pregnant) were not different from each other. Pregnancy diagnoses were made based on a fixed cut-off value, to which the LH response of each ewe to 5 mug LHRH was compared. Ewes whose response fell below this cut-off were diagnosed as pregnant. Accuracy of the diagnoses were determined by known lambing data. Diagnostic accuracy ranged from a low of 60% for nonpregnant, to a high of 95% for ML ewes. Accuracy for SL ewes (64%) was lower than for the overall pregnant group (79%), as well as that for ML ewes. Doses of LHRH, higher than 5 mug per ewe, generally produced LH release in pregnant ewes which was not significantly suppressed relative to responses of nonpregnant ewes. These results lead to the conclusion that gonadotropin response to exogenous LHRH injection is not an effective tool for pregnancy diagnosis. 相似文献

The objective of this study was to investigate the factors affecting ball velocity at the final instant of the impact phase (t1) in full instep soccer kicking. Five experienced male university soccer players performed maximal full instep kicks for various foot impact points using a one-step approach. The kicking motions were captured two dimensionally by a high-speed camera at 2,500 fps. The theoretical equation of the ball velocity at t1 given in the article was derived based on the impact dynamics theory. The validity of the theoretical equation was verified by comparing the theoretical relationship between the impact point and the ball velocity with the experimental one. Using this theoretical equation, the relationship between the impact point and the ball velocity was simulated. The simulation results indicated that the ball velocity is more strongly affected by the foot velocity at the initial instant of the impact phase than by other factors. The simulation results also indicated that decreasing the ankle joint reaction force during ball impact shifts the impact point that produces the greatest ball velocity to the toe side and decreasing the ankle joint torque during ball impact shifts the impact point that produces the greatest ball velocity to the ankle side. 相似文献
