We conducted a detail study of the photosynthetic apparatus in assimilating organs of three introduced evergreen conifer species: Taxus cuspidate S. et Z. ex E. (Far-Eastern yew), Thuja occidentalis L. (arbovitae “green”), and Th. occidentalis f. “Reingold” (arbovitae “yellow”) at various times in their life cycle. We studied the potential photosynthesis rate; composition and ratios of pigments, including primary carotenoids; the violaxanthin cycle (VC) activity, the synthesis of a secondary carotenoid, rhodoxanthin; and chloroplast ultrastructure. In winter and spring, β-carotene and lutein (primary carotenoids) contents were relatively constant in yew and arbovitae “yellow”. In December, the VC in yew was balanced and in arbovitae “yellow” unbalanced. In arbovitae “yellow”, the zeaxanthin pool was heterogeneous, and only part of it took part in the VC. It can be assumed that the other part of the pool can be oxidized to form a secondary carotenoid, rhodoxanthin. This secondary carotenoid was also accumulated in arbovitae “green”; its synthesis took place during the season, when the photosynthesis rate of plants was the lowest, and a significant chloroplast reorganization occurred (the number of thylakoids in grana decreased and plastoglobules appeared). We suppose that rhodoxanthin forms a filter for the light under the conditions of high insolation in winter. Thus, the evergreen conifer plants studied, which are adapted to growing at high latitudes where temperature is low and insolation is high in winter and spring, have a system for protecting the photosynthetic apparatus against photodestruction. In the basis of this system, the primary and secondary carotenoids lie, whose content changes during the year.  相似文献

We present here a bibliometric analysis of publications on photosynthesis research from 1992 to 2009 in the Science Citation Index Expanded (SCI-Expanded) Web of Science. This has allowed us to examine the growing trends and the key topics on this subject. We have assessed the document type, language of the publications, publication output, subject category, journal distribution, countries and territories of these publications, institutions involved, hot topics and highly cited papers. The top 30 countries/territories were ranked according to their total number of articles (TA), single country articles (SCA), internationally collaborative articles (ICA), first author articles (FAA) and corresponding author articles (CAA). Research directions on the subject of photosynthesis were also investigated and evaluated by statistically analyzing the distribution of author keywords in the database. Our analysis indicates that “water”, “stress”, “carbon dioxide”, “nitrogen” and “climate change” are hot topics of research on photosynthesis during this period.  相似文献

Analysis of new data and reinterpretation of published information for clay minerals found in temperate climate soil profiles indicates that there is often a gradient of “illite-like” clay minerals with depth. We used the term “illite-like” because these observations are based on X-Ray Diffractogram patterns and not on layer charge measurements which allow to define properly illite. It appears that “illite-like” layers are concentrated in the upper, organic - rich portion of the soil profile both under grassland and forest vegetation. “Illite-like” layer quantity seems directly related to soil potassium status. Indeed, intensive agriculture practises without potassium fertilization reduce “illite-like” content in surface soils, whereas several years of potassic fertilization without plant growth can increase “illite-like” content. The potassic soil clay mineral, illite, is particularly important in that it can be the major source of readily available potassium for plants. Spatial and temporal dynamics of clay minerals should be related to the potassium cycle. We propose that the frequently observed general trend of increasing exchangeable potassium in the top soil can be correlated with an increase in “illite-like” in the clays and that the decrease of potassium caused by intensive agricultural practices leads to “illite-like” layer destabilization. This vision of “illite-like” layer as a potassium reservoir refueled by plants and emptied by intensive cropping renews the concept of potassium availability and indicates a need to be discussed as well in natural ecosystems as in cultivated ecosystems.  相似文献

Genome projects are approaching completion and are saturating sequence databases. This paper discusses the role of the two-hybrid system as a generator of hypotheses. Apart from this rather exhaustive, financially and labour intensive procedure, more refined functional studies can be undertaken. Indeed, by making hybrids of two-hybrid systems, customised approaches can be developed in order to attack specific function-related problems. For example, one could set-up a “differential” screen by combining a forward and a reverse approach in a three-hybrid set-up. Another very interesting project is the use of peptide libraries in two-hybrid approaches. This could enable the identification of peptides with very high specificity comparable to “real” antibodies. With the technology available, the only limitation is imagination.  相似文献

Parent-specific, randomly amplified polymorphic DNA (RAPD) markers were obtained from total genomic DNA ofChlamydomonas reinhardtii. Such parent-specific RAPD bands (genomic fingerprints) segregated uniparentally (through mt+) in a cross between a pair of polymorphic interfertile strains ofChlamydomonas (C. reinhardtii andC. minnesotti), suggesting that they originated from the chloroplast genome. Southern analysis mapped the RAPD-markers to the chloroplast genome. One of the RAPD-markers, “P2” (1.6 kb) was cloned, sequenced and was fine mapped to the 3 kb region encompassing 3′ end of 23S, full 5S and intergenic region between 5S and psbA. This region seems divergent enough between the two parents, such that a specific PCR designed for a parental specific chloroplast sequence within this region, amplified a marker in that parent only and not in the other, indicating the utility of RAPD-scan for locating the genomic regions of sequence divergence. Remarkably, the RAPD-product, “P2” seems to have originated from a PCR-amplification of a much smaller (about 600 bp), but highly repeat-rich (direct and inverted) domain of the 3 kb region in a manner that yielded no linear sequence alignment with its own template sequence. The amplification yielded the same uniquely “sequence-scrambled” product, whether the template used for PCR was total cellular DNA, chloroplast DNA or a plasmid clone DNA corresponding to that region. The PCR product, a "unique" new sequence, had lost the repetitive organization of the template genome where it had originated from and perhaps represented a “complex path” of copy-choice replication.  相似文献

The author's theory of the adoption of certain types of behavior patterns (Rashevsky, N., 1957, “Contributions to the Theory Initiative Behavior”.Bull. Maths. Biophysics,19, 91–119; 1968,Looking at History through Mathematics, Cambridge, Massachusetts: M.I.T. Press) consisting of elementary behaviors for each of which there is an opposite one and the two are mutually exclusive, is applied to describe the changes in the general type of behavior of a society. The elementary acts of which the whole problem consists may be either overt activities or beliefs or opinions. The general behavior patternsadopted by the society are considered as the “proper” or “just” ones. Any deviation from it in either one or more of the component elementary behaviors is considered as “unjust” and is subject to some punitive action. The total number of possible mutually exclusive behavior patterns is very large but finite. Within this very large range of possible patterns, we find that this notion of justice is relative, because changes from any behavior pattern to any other may occur. It is further shown that the amount of punishment for the deviation from the accepted pattern in order to be effective as well as efficient must be applied in different ways to different individuals even for the same transgression.  相似文献

Light state transition is a physiological function of oxygenic organisms to balance the excitation of photosystem II (PSII) and photosystem I (PSI), hence a prerequisite of oxygen-evolving photosynthesis. For cyanobacteria, phycobilisome (PBS) movement during light state transition has long been expected, but never observed. Here the dynamic behavior of PBS movement during state transition in cyanobacterium Synechocystis PCC6803 is experimentally detected via time-dependent fluorescence fluctuation. Under continuous excitation of PBSs in the intact cells, time-dependent fluorescence fluctuations resemble “damped oscillation” mode, which indicates dynamic searching of a PBS in an “overcorrection” manner for the “balance” position where PSII and PSI are excited equally. Based on the parallel model, it is suggested that the “damped oscillation” fluorescence fluctuation is originated from a collective movement of all the PBSs to find the “balance” position. Based on the continuous fluorescence fluctuation during light state transition and also variety of solar spectra, it may be deduced that light state transition of oxygen-evolution organisms is a natural behavior that occurs daily rather than an artificial phenomenon at extreme light conditions in laboratory.  相似文献

Exploring the relationships between landscape pattern and ecological processes is the key topic of landscape ecology, for which, a large number of indices as well as landscape pattern analysis model were developed. However, one problem faced by landscape ecologists is that it is hard to link the landscape indices with a specific ecological process. Linking landscape pattern and ecological processes has become a challenge for landscape ecologists. “Source” and “sink” are common concepts used in air pollution research, by which the movement direction and pattern of different pollutants in air can be clearly identified. In fact, for any ecological process, the research can be considered as a balance between the source and the sink in space. Thus, the concepts of “source” and “sink” could be implemented to the research of landscape pattern and ecological processes. In this paper, a theory of sourcesink landscape was proposed, which include: (1) In the research of landscape pattern and ecological process, all landscape types can be divided into two groups, “source” landscape and “sink” landscape. “Source” landscape contributes positively to the ecological process, while “sink” landscape is unhelpful to the ecological process. (2) Both landscapes are recognized with regard to the specific ecological process. “Source” landscape in a target ecological process may change into a “sink” landscape as in another ecological process. Therefore, the ecological process should be determined before “source” or “sink” landscape were defined. (3) The key point to distinguish “source” landscape from “sink” landscape is to quantify the effect of landscape on ecological process. The positive effect is made by “source” landscape, and the negative effect by “sink” landscape. (4) For the same ecological process, the contribution of “source” landscapes may vary, and it is the same to the “sink” landscapes. It is required to determine the weight of each landscape type on ecological processes. (5) The sourcesink principle can be applied to non-point source pollution control, biologic diversity protection, urban heat island effect mitigation, etc. However, the landscape evaluation models need to be calibrated respectively, because different ecological processes correspond with different source-sink landscapes and evaluation models for the different study areas. On the occasion of the first international workshop on systems radiation biology we review the role of cell renewal systems in maintaining the integrity of the mammalian organism after irradiation. First, 11 radiation emergencies characterized by chronic or protracted exposure of the human beings to ionizing irradiation were “revisited”. The data provide evidence to suggest that at a daily exposure of about 10–100 mSv, humans are capable of coping with the excess cell loss for weeks or even many months without hematopoietic organ failure. Below 10 mSv/day, the organisms show some cellular or subcellular indicators of response. At dose rates above 100 mSv/day, a progressive shortening of the life span of the irradiated organism is observed. To elucidate the mechanisms relevant to tolerance or failure, the Megakaryocyte–thrombocyte cell renewal system was investigated. A biomathematical model of this system was developed to simulate the development of thrombocyte concentration as a function of time after onset of chronic radiation exposure. The hematological data were taken from experimental chronic irradiation studies with dogs at the Argonne National Laboratory, USA. The results of thrombocyte response patterns are compatible with the notion of an “excess cell loss” (compared to the steady-state) in all proliferative cell compartments, including the stem cell pool. The “excess cell loss” is a function of the daily irradiation dose rate. Once the stem cell pool is approaching an exhaustion level, a “turbulence region” is reached. Then it takes a very little additional stress for the system to fail. We conclude that in mammalian radiation biology (including radiation medicine), it is important to understand the physiology and pathophysiology of cell renewal systems in order to allow predicting the development of radiation induced lesions.  相似文献

The dikaryon #5026 (A2B2)+#5132 (A7B7) of the basidiomyceteCoprinus macrorhizus was treated with ultraviolet light (UV),N-methyl-N′-nitro-N-nitrosoguanidine (NG), and 5-bromouracil (BU) in order to induced developmental variations in fruiting. Among a total of 11,383 isolates, 742 were homokaryons. Fruiting test was made with the remaining 10,641 dikaryotic isolates, of which 1,594 showed abnormality in basidiocarp development. Both UV and NG were very efficient in inducing such variations, but BU was not. The induced variations were classified into seven basic types as follows: 1) “knotless”, in which hyphal knots, the first sign of fruiting, do not differentiate; 2) “primordiumless,” in which hyphal knots are formed, but no further development occurs; 3) “maturationles”, in which primordia are formed, but the maturation of primordia into adult fruit bodies does not occur; 4) “elongationless”, in which the elongation of stipe is blocked, and a basidiocarp with a very short stipe is produced; 5) “expansionless,” in which pilei do not expand normally; 6) “sporeless,” in which the formation of basidiospores is blocked and albinic pilei bearing none or only a small amount of spores are produced; and 7) “autolysisless,” in which the autolysis of pilei does not appear to occur. It has been noticed that the four steps of maturation, i.e. stipe elongation, pileus expansion, spore formation, and perhaps pileus autolysis, can proceed independently. Compound types of variations such as “elongation-expansionless,” “elongation-sporeless,” “expansion-sporeless,” and “elongation-expansion-sporeless” were also induced. UV treatment induced maturationless at the highest rate, while NG treatment sporeless. We first analyse a simple symmetric model of the idiotypic network. In the model idiotypic interactions regulate B cell proliferation. Three non-idiotypic processes are incorporated: (1) influx of newborn cells; (2) turnover of cells: (3) antigen. Antigen also regulates proliferation. A model of 2 B cell populations has 3 stable equilibria: one virgin, two immune. The twodimensional system thus remembers antigens, i.e. accounts for immunity. By contrast, if an idiotypic clone proliferates (in response to antigen), its anti-idiotypic partner is unable to control this. Symmetric idiotypic networks thus fail to account for proliferation regulation. In high-D networks we run into two problems. Firstly, if the network accounts for memory, idiotypic activation always propagates very deeply into the network. This is very unrealistic, but is an implication of the “realistic” assumption that it should be easier to activate all cells of a small virgin clone than to maintain the activation of all cells of a large (immune) clone. Secondly, graph theory teaches us that if the (random) network connectance exceeds a threshold level of one interaction per clone, most clones are interconnected. We show that this theory is also applicable to immune networks based on complementary matching idiotypes. The combination of the first “percolation” result with the “interconnectancr” result means that the first stimulation of the network with antigen should eventually affect most of the clones. We think this is unreasonable. Another threshold property of the network connectivity is the existence of a virgin state. A gradual increase in network connectance eliminates the virgin state and thus causes an abrupt change in network behaviour. In contrast to weakly connected systems, highly connected networks display autonomous activity and are unresponsive to external antigens. Similar differences between neonatal and adult networks have been described by experimentalists. The robustness of these results is tested with a network in which idiotypic inactivation of a clone occurs more generally than activation. Such “long-range inhibition” is known to promote pattern formation. However, in our model it fails to reduce the percolation, and additionally, generates semi-chaotic behaviour. In our network, the inhibition of a clone that is inhibiting can alter this clone into a clone that is activating. Hence “long-range inhibition” implies “long-range activation”, and idiotypic activation fails to remain localized. We next complicate this model by incorporating antibody production. Although this “antibody” model statically accounts for the same set of equilibrium points, it dynamically fails to account for state switching (i.e. memory). The switching behaviour is disturbed by the autonomous slow decay of the (long-lived) antibodies. After antigenic triggering the system now performs complex cyclic behaviour. Finally, it is suggested that (idiotypic) formation of antibody complexes can play only a secondary role in the network. In conclusion, our results cast doubt on the functional role of a profound idiotypic network. The network fails to account for proliferation regulation, and if it accounts for memory phenomena, it “explodes” upon the first encounter with antigen due to extensive percolation.  相似文献

The responses of photosynthetic gas exchange, chlorophyll fluorescence, content of pigments, main osmolytes, and malondialdehyde (MDA) to water-withholding for 15 days and re-hydration in seedlings of two mango cultivars (Mangifera indica L. var. “Choke Anand’ and var. “Khieo Sawoei”) under 50% sunlight and full sunlight were investigated. For both cultivars, the water-witholding resulted in progressively decreases in leaf relative water content, net photosynthesis (P n), stomatal conductance (g s), and increases in the conversion of xanthophyll cycle pigments estimated by an index of leaf spectral reflectance (ΔPRI), carotenoid to chlorophyll ratio, non-photochemical quenching (NPQ), the contents of malondialdehyde (MDA) and compatible solutes (total soluble sugar and proline). The effect of the water stress was more pronounced in full sunlight than 50% sunlight. The maximum photochemistry efficiency measured at dawn was fairly constant during the period of the treatment for both cultivars under both light regimes. The water stress caused less pronounced inhibition of photosynthesis in “Choke Anand” than in “Khieo Sawoei” cultivar under both light regimes. After re-hydration, the recovery was relatively quicker in “Choke Anand” than in “Khieo Sawoei” cultivar. Both cultivars in both 50% and full sunlight showed complete recovery in photochemistry after 5 days of re-watering but photosynthesis did not show a complete recovery as indicated by gas exchange rates. As the results of lower NPQ, ΔPRI and osmotic adjustment in the cultivar “Khieo Sawoei” compared to the cultivar “Choke Anand”, the former cultivar was less tolerant to drought than the latter. Our study further showed that partial shading (e.g., 50% of sunlight) significantly alleviated the harmful effect of drought stress on mango cultivars but in fact stomata of seedlings grown in partial shade was more responsive to water deficit than in full light.  相似文献

Binocular rivalry occurs when two very different images are presented to the two eyes, but a subject perceives only one image at a given time. A number of computational models for binocular rivalry have been proposed; most can be categorised as either “rate” models, containing a small number of variables, or as more biophysically-realistic “spiking neuron” models. However, a principled derivation of a reduced model from a spiking model is lacking. We present two such derivations, one heuristic and a second using recently-developed data-mining techniques to extract a small number of “macroscopic” variables from the results of a spiking neuron model simulation. We also consider bifurcations that can occur as parameters are varied, and the role of noise in such systems. Our methods are applicable to a number of other models of interest.  相似文献

In the rapid “quench” kientics of myosin, the “initial phosphate burst” is the excess inorganic phosphate that is produced during the early time-course of ATP hydrolysis by myosin subfragment-1 (S-1) or HMM. In general, the existence of a Pi burst implies a rapid (i.e., generally an order of magnitude faster than the steady-state hydrolysis rate) lysis of the phospho-anhydride bond within the ATP molecule, followed by one or more slower steps that are rate limiting for the process. Thus, the presence of a Pi burst can provide an important clue to the mechanism of the reaction. However, in the case of actomyosin, this clue as long been the subject of controversy and misunderstanding. To measure the (initial) Pi burst, myosin S-1 (or HMM) is rapidly mixed with ATP and then the mixture is acid quenched after a specific time period. The medium produced contains free Pi generated from hydrolysis of the ATP. The quantitative measure of the phosphate generated in this way has always been significantly greater than that expected by steady-state “release” of Pi alone, and it is that very difference between this measured Pi after the quench and that amount of Pi expected to be released by steady-state considerations in that same time period that has been referred to as the “initial Pi burst”. Recent investigations of the kinetics of Pi release have used an entirely new method that directly measures the release of Pi from the enzyme-product complex. These studies have made reference to the properties of the “initial Pi burst” in the presence of actin, as well as to a new kinetic entity: the “burst of Pi release”, and have been often vague concerning the true nature of the initial Pi burst, as well as the properties of Pi release as predicted by the current models of the actin activation of the myosin ATPase activity. The purpose of the current article is to correct this oversight, to discuss the “burst” in some detail, and to display the kinetics predicted by the current models for the actin activation of myosin. Furthermore, predictions for the kinetics of the new “burst of Pi release” are discussed in terms of its ability to discriminate between the two current competing models for actin activation of the myosin ATPase activity.  相似文献

One vocalization in the Japanese macaque (Macaca fuscata) system of communication is the “girney.” Previous studies indicated that the “girney” is used for short range communication and that it tends to occur when subordinate animals approach and groom dominant ones and when females without infants approach females who have infants. Data were collected on the social behavior of adult female Japanese monkeys of the Arashiyama-A troop in Texas in order to test those results. The study indicates that “girneys” are the most frequently occurring vocalizations of females during and following the birth season and that they occur primarily in two contexts. Those are the proximity of a female to another female with a new infant and the proximity of a lower ranking animal to a higher ranking one. The contexts are ones in which the risk of aggression is high, and the “girneys” appear to function as appeasement gestures to reduce the risk.  相似文献

It is a popularly held view that Darwin was the first author to draw a phylogenetic tree diagram. However, as is the case with most popular beliefs, this one also does not hold true. Firstly, Darwin never called his diagram of common descent a tree. Secondly, even before Darwin, tree diagrams were used by a variety of philosophical, religious, and secular scholars to depict phenomena such as “logical relationships,” “affiliations,” “genealogical descent,” “affinity,” and “historical relatedness” between the elements portrayed on the tree. Moreover, historically, tree diagrams themselves can be grouped into a larger class of diagrams that were drawn to depict natural and/or divine order in the world. In this paper, we trace the historical roots and cultural meanings of these tree diagrams. It will be demonstrated that tree diagrams as we know them are the outgrowth of ancient philosophical attempts to find the “true order” of the world, and to map the world “as it is” (ontologically), according to its true essence. This philosophical idea would begin a fascinating journey throughout Western European history. It lies at the foundation of the famous “scala naturae,” as well as religious and secular genealogical thinking, especially in regard to divine, familial (kinship), and linguistic pedigrees that were often depicted by tree images. These scala naturae would fuse with genealogical, pedigree thinking, and the trees that were the result of this blend would, from the nineteenth century onward, also include the element of time. The recognition of time would eventually lead to the recognition of evolution as a fact of nature, and subsequently, tree iconographies would come to represent exclusively the evolutionary descent of species.  相似文献

Summary Field data of a detailed vegetation survey, undertaken in a wet grassland in the nature reserve “De Kampina”, near Boxtel, Netherlands, were used to prepare aBraun-Blanquet phytosociological table. From this table two associations could be derived: Cirsio-Molinietum and Senecioni-Brometum racemosi. Association-analysis and principal component analysis of a species correlation coefficient matrix was carried out on the same data in order to test their usefulness in a further ecological interpretation of theBraun-Blanquet phytosociological table. It was found that association-analysis, although it was able to detect a similar pattern in vegetation, did not allow a further interpretation of the phytosociological table. Principal component analysis proved to be very useful in the ecological interpretation of details of the table and in the correct placing of less representative and intermediate stands.
Zusammenfassung Aufnahmen einer detaillierten Untersuchung einer sumpfigen Wiese im Naturschutzgebiet “De Kampina”, bei Boxtel, Niederlande, sind dargestellt worden in einer pflanzensoziologischen Tabelle. Zwei Assoziationen konnten herausgearbeitet werden: Cirsio-Molinietum und Senecioni-Brometum racemosi. Dieselben Daten wurden für eine “Assoziationsanalyse” und eine prinzipale Komponentenanalyse benutzt, um ihre Brauchbarkeit für eine weitere ?kologische Interpretation der pflanzensoziologischen Tabelle zu prüfen. Obwohl mit der “Assoziationsanalyse” ein ?hnliches Muster in der Vegetation zu entdecken war wie mit der pflanzensoziologischen Tabelle, erm?glichte sie keine weitere ?kologische Interpretation. Die prinzipale Komponentenanalyse zeigte sich sehr nützlich sowohl bei der ?kologischen Interpretation von Einzelheiten der Tabelle als auch bei der Einordnung nicht v?llig typischer bzw. intermedi?rer Aufnahmen.

Zearalenone is a mycotoxin mainly produced by severalFusarium species, which are known to colonize grains in temperate climates. The purpose of the study is to provide a reliable isotope dilution method for the quantification of this mycotoxin. A derivative of the analyte to be used as standard is obtained by reaction with acetic anhydride, which is available in two pure isotopic forms, a protonated (“light”) and a hexadeuterated (“heavy”). The derivatized standards are added to the matrix split intwo parts. Then, the derivatization procedure is repeated on both matrices derivatizing the part containing the “heavy” labelled standard with the “light” acetic anhydride and the part containing the “light” labelled standard with the “heavy” acetic anhydride. Both extracted mixtures are analyzed by LC/MS, monitoring the “light” and the “heavy” labelled analytes and using the former as standard for the latter in one case and viceversa in the other case. Comparative analysis reveals remarkahle homology between the sequences of bothpsbA gene nucleotides and the inferred amino acids of sorghum, a C4 plant, and those of rice, a C3 plant. The 5′-noncoding region of sorghumpsbA gene contains the conservative promoter elements, “—35” element and “—10” element, like the prokaryote and the promoter element, TATA box, like the eukaryote. As compared with that of the rice, an extra sequence of 7 bp is found in the leader sequence of the mRNA in the former. Using anin vitro system, it has been demonstrated that protein factor exists in sorghum chloroplast protein extract which specifically hinds to the 5′-noncoding region ofpsbA gene. Measurement of the expression of luciferase shows a 2–5 time greater reaction of the expression plasmids pALqs which contain leader region of sorghumpsbA gene than that of the expression plasmids pALqr which contain leader region of ricepsbA gene inE. coli.
