Summary Observations of aperture changes as sucrose is added to the solution bathing epidermal strips ofCommelina communis L. allow calculation of the osmotic changes required to open or close the stomatal pore, for comparison with changes in potassium content. With isolated guard cells, in strips in which all cells other than guard cells have been killed, the internal osmotic changes required are 83 mosmol kg–1 m–1 below 10m aperture, 129 mosmol kg–1 m–1 in the range 10–15 m, and 180 mosmol kg–1 m–1 above 15 m. For opening against subsidiary cell turgor in addition to guard cell turgor, in intact strips with live subsidiary and epidermal cells, these figures should each be increased by about 33 mosmol kg–1 m–1. A change in subsidiary cell turgor is magnified in its effects on the water relations of the guard cell by a factor greater than 3.7 for equal changes in the water potential of the two cells, or greater than 4.7 at constant volume of the guard cell.  相似文献

Summary Determination of paternity was attempted in the case of three children whose putative fathers are dead using DNA samples of the paternal grandparents. The DNA analyses were performed with both multilocus and single-locus probes which resolve highly polymorphic areas of human genome. The results were conclusive with both types of probes and facilitated, for example, the exclusion of the brother of the putative father. The evidence for true paternity obtained with DNA analyses can be considered reliable in this type of indirect paternity in which tests based on protein polymorphism are inconclusive.  相似文献

Summary We examined the proportions (or so called rates) of fetuses with 47,+21, 47,+18, or 47,+13 diagnosed prenatally in women at the upper extremes of age. Our analysis was prompted by results from a large scale European study of amniocentesis which indicated that after increasing exponentially from age 35 years, the proportions of the autosomal trisomies reached a peak at a specific age and then leveled off or declined at the upper end of the age range. We analyzed North American data on 56,075 fetuses studied because of no known cytogenetic risk factor (aside from maternal age). This is the largest series to data. For 47,+21, the data from amniocentesis studies provide no evidence for any drop in the rate of change of proportion with maternal age up to 49 years. There is, if anything, a trend in our data to a steepening in the exponential rate of change at the upper extreme of age (above 46 years). Data from livebirths on the Down syndrome phenotype are at least consistent with an exponential rate of increase in proportion affected up to age 49 years. For 47,+18 our data from prenatal diagnoses are more consistent with an exponential increase up to age 43 years and a level proportion (or rate) after that. For 47,+13 no cases were observed above age 42 years, consistent with the drop in proportion affected above this age observed in the European series. We emphasize the possible effect of sampling fluctuation and reporting error upon these apparent trends.  相似文献

Summary Transmission of paternal chloroplasts was observed in Nicotiana, considered to inherit organelles in a strictly maternal way. Plants carrying streptomycin resistant plastids were used as pollen donors. Cell lines with paternal plastids in the offspring were selected as green (resistant) sectors on calli induced from the seedlings on streptomycin-containing media. The presence of paternal plastids in the regenerated plants was confirmed by restriction analysis. In the Nicotiana plumbaginifolia xN. plumbaginifolia Np(SR1)3 and the N. plumbaginifolia Np(gos)29 xN. tabacum SR1 crosses 2.5% and 0.07% of the offspring were found to contain paternal (tabacum) plastids, respectively. These plants, however, carried maternal mitochondria exclusively. This sexual cybridization method offers a simple way to transfer chloroplasts solely, a goal not accessible by protoplast fusion.  相似文献

Histological examination of a large sample of the tsetse fly, Glossina morsitans orientalis, killed at timed intervals after emergence, showed that embryogenesis takes about 4 days at 25°C during the first intra-uterine cycle. This is considerably longer than is usually stated. Use of these data to obtain an average rate of development for the sample suggested a new method of age determination for the adult female fly which can be used in conjunction with ovarian determination of age.
Zusammenfassung Histologische Überprüfung einer großen Stichprobe von Tsetsefliegen, Glossina morsitans orientalis, die in bestimmten Abständen nach dem Schlüpfen getötet worden waren, zeigte, daß die Embryogenese während des ersten intrauterinen Zyklus bei 25° ungefähr 4 Tage dauert. Das ist beträchtlich länger, als gewöhnlich festgestellt wurde. Die Benutzung dieses Zeitabschnitts zur Ermittlung der durchschnittlichen Entwicklungsrate für eine Stichprobe bietet sich als neue Methode der Altersbestimmung erwachsener weiblicher Fliegen an, die in Verbindung mit der ovarialen Altersbestimmung verwendet werden kann.

The Sahara and Sahel regions of northern Africa have complex environmental histories punctuated by sudden and dramatic regime shifts in climate and ecological conditions. Here we review the current understanding of the causes and consequences of two environmental regime shifts in the Sahara and Sahel. The first regime shift is the sudden transition from vegetated to desert conditions in the Sahara about 5500 years ago. Geologic data show that wet environmental conditions in this region—giving rise to extensive vegetation, lakes, and wetlands—came to an abrupt end about 5500 years ago. Explanations for climatic changes in northern Africa during the Holocene have suggested that millennial-scale changes in the Earths orbit could have caused the wet conditions that prevailed in the early Holocene and the dry conditions prevalent today. However, the orbital hypothesis, by itself, does not explain the sudden regime shift 5500 years ago. Several modeling studies have proposed that strong, nonlinear feedbacks between vegetation and the atmosphere could amplify the effects of orbital variations and create two alternative stable states (or regimes) in the climate and ecosystems of the Sahara: a green Sahara and a desert Sahara. A recent coupled atmosphere-ocean-land model confirmed that there was a sudden shift from the green Sahara to the desert Sahara regime approximately 5500 years ago. The second regime shift is the onset of a major 30-year drought over the Sahel around 1969. Several lines of evidence have suggested that the interactions between atmosphere and vegetation act to reinforce either a wet Sahel or a dry Sahel climatic regime, which may persist for decades at a time. Recent modeling studies have indicated that the shift from a wet Sahel to a dry Sahel regime was caused by strong feedbacks between the climate and vegetation cover and may have been triggered by slow changes in either land degradation or sea-surface temperatures. Taken together, we conclude that the existence of alternative stable states (or regimes) in the climate and ecosystems of the Sahara and Sahel may be the result of strong, nonlinear interactions between vegetation and the atmosphere. Although the shifts between these regimes occur rapidly, they are made possible by slow, subtle changes in underlying environmental conditions, including slow changes in incoming solar radiation, sea-surface temperatures, or the degree of land degradation.  相似文献

Zusammenfassung In der Neurohypophyse fetaler und neugeborener Ratten entsteht die Mehrzahl der synaptischen Bläschen aus Erweiterungen der Neurotubuli. Ferner können pinocytotische Bläschen als synaptische Vesikel imponieren. Die Bläschenbildung aus Membranen von Elementargranula (vgl. Herlant, 1967) tritt dagegen in den Hintergrund. Ein Auftreten von Vesikeln im Innern von Elementargranula wurde nicht beobachtet.
The origin of synaptic vesicles in neurosecretory axons
Summary In the neurohypophysis of fetal and newborn rats the majority of synaptic vesicles originate from dilatations of neurotubuli. Moreover, pinocytotic invaginations give rise to synaptic vesicles. Evaginations of elementary granule membranes, as described by Herlant (1967), are seldom to be found and do not seem to play an important role in the formation of synaptic vesicles. The occurrence of vesicles within elementary granules was not observed.

Summary This study attempts to elucidate the mechanism through which lysosomal accumulation occurs with age in the epithelial cells of the thyroid gland and especially in the active follicles of the aging mouse thyroid. Thyroid morphology and function in old CBA (at least 24 months of age) male mice were compared with those in young (2 months of age) animals. The effects of different intake of iodine were tested and compared in both cohorts, each of which was divided into three groups: (i) low iodine group, (ii) moderate iodine group, and (iii) high iodine group. As expected, the present work confirmed the well-known accumulation with age of cold follicles coexisting with active follicles in the old mouse thyroid. Attention has been focused on the active follicles whose follicular cells contained in their cytoplasm a large number of pleomorphic dense bodies. The lysosomal nature of these bodies, referred to as secondary lysosomes, was confirmed by histochemistry; however, they displayed variability in acid phosphatase staining. In old animals, regardless of the type of iodine regimen, the ratio between relative follicular volume and relative colloid volume as determined by morphometry remained unchanged. Ultrastructurally, the relative volume occupied by secondary lysosomes in active follicles was always higher than in the young groups. Autoradiographic studies with 125I revealed that a large part of the radioactivity was located in secondary lysosomes of thyroid cells in active follicels of old mice when radioiodine was injected 3 weeks before death. Two different types of vacuoles were present in a non-negligible number of thyrocytes of the active follicles in aged cohorts. The first type was made up of grossly dilated rough endoplasmic cisternae, the second corresponded to intracytoplasmic microfollicular vacuoles. Both aspects have been described in conditions of chronic stimulation. It is concluded (1) that different intake of iodine for 6 weeks does not modulate the thyroid morphology in old mice; (2) that in the thyrocytes of the active follicles in old mice accumulation of secondary lysosomes occurs due to a slowdown of turnover; and (3) that the follicular cells of active follicles feature morphological aspects suggesting a hyperactive state compensating the lack of hormone production in the cold follicles.  相似文献

Summary Up to now Antirrhinum was classified as a typical example for a uniparentalmaternal inheritance of the plastids. However, the findings reported here prove that also the male gametophyte of Antirrhinum may occasionally transmit plastids into the egg. This conclusion is based on genetic experiments involving a form of the plastom mutant prasinizans which is described as gelbgrüne prasinizans. In contrast to all other plastid mutations known in Antirrhinum majus this mutant originated in Sippe 50 is completely viable. In plants containing plastids of this mutant type only, the mutant character is manifested during early growth stages. Cotyledons and first foliage leaves which are initially white or white yellow, slowly turn green and become indistinguishable from normal Sippe 50. Reciprocal crosses of green Sippe 50 with gelbgrüne prasinizans gave few variegated descendants; the others were exclusively plants identical with the maternal parent as far as leaf colour is concerned (Table). The variegated individuals cannot be gene mutants since selfing and crossing experiments showed non-mendelian inheritance. Furthermore it could be ruled out that in the cross Sippe 50 x gelbgrüne prasinizans the three variegated descendants represent spontaneous new plastom mutants because the pale tissue in these plants turned green in the same way as the paternal parent. Because of the typical greening of this mutant and since plastid mutations could be ruled out we have to conclude that plastids were transmitted by the pollen parent into the egg. There these plastids multiplied together with the maternal plastids giving rise to the chimeras after sorting-out of the two plastid types. This interpretation is supported by the observation of mixed cells in tissues where the leaf variegation is finely mosaiced. The results were possible only because the plastids of the pollen parent can be unequivocally recognised.  相似文献

This paper explores the usefulness of a concept of human behavioral breakdowns for studying psychiatric phenomena from a cross-cultural standpoint. The kinds of conceptualizations that are available in the English literature about human behavioral breakdowns across societies are reviewed. The way these breakdowns have been handled in the Western European Tradition of medicine is given special emphasis. Attention is focused on schizophrenia as one of the modern disease concepts of psychiatry that is a variant of human behavioral breakdowns. The essay then looks at schizophrenia using contemporary ideas about cultural relativism as these apply to the study of psychiatric phenomena in relation to society. Perspectives about the variable as versus the invariant in schizophrenia are reviewed.  相似文献

Lathyrism is a non-progressive motor neuron disease produced by consumption of the excitatory amino acid, 3-N-oxalyl-L-2,3-diaminopropanoic acid (-ODAP). To learn more about the mechanisms underlying Lathyrism three structural analogs of -ODAP were synthesized. Carboxymethyl-,-diaminopropanoic acid (CMDAP) evoked inward currents which were antagonized by APV (30 M), but not by CNQX (10 M). N-acetyl-,-diaminopropanoic acid (ADAP) evoked no detectable ionic currents but potentiated N-methyl-D-aspartate (NMDA)-activated currents. The potentiation of NMDA currents by ADAP was blocked by 7-chlorokynurenic acid. Carboxymethylcysteine (CMC) did not activate any detectable ionic currents. None of the three -ODAP analogs produced visible symptoms of toxicity in day old chicks when administered for 2–3 consecutive days. Ligand binding studies demonstrated that all the three compounds were effective to in displacing [3H]glutamate. The maximum inhibition was 92% for CMDAP, 61% for ADAP, 65% for CMC and 99% for -ODAP. These data indicate that analogs of -ODAP may interact with glutamate receptors without producing neurotoxicity.  相似文献

Summary Human liver alcohol dehydrogenase (ADH) variants were screened in random autopsy specimens from 53 North German and 34 Japanese individuals. Based on pH-activity profile and electrophoretic pattern, only ADH2 and ADH3 variants were detected. In relatively fresh specimens, an anodic band or -ADH band was also observed. The recently reported new molecular forms collectively called ADHIndianapolis (Bosron et al. 1980) could not be demonstrated and therefore may be confined hitherto only to the American black population.This paper is dedicated to Professor Dr. Dr. H. Summary Propionic acidemia is an autosomal recessive metabolic disease resulting from a deficiency of propionyl CoA carboxylase (PCC) activity. We have analyzed the molecular heterogeneity of Japanese propionic acidemia patients using anti-human PCC antiserum and cDNA clones coding for the two protein subunits ( and ) of the enzyme. The steady state levels of both and subunits of PCC from 15 Japanese patients were determined by Western blot. Three patients had neither nor subunits, and the amounts of both and subunits were low in 3 other patients. According to our previous data, we classified these 6 patients as having subunit deficiency. In the remaining 8 patients, subunits were normal, but the subunits were aberrant. Two patients had low levels of normal-sized subunits and 6 had subunits smaller than normal in size and greatly reduced in quantity. These 8 patients were assigned to the subunit deficiency category. One patient had apparently normal and subunits. We could not determine this patient's primary defect. These data reveal the genetic heterogeneity of molecular defects causing propionic acidemia in the Japanese. Southern blot analysis did not reveal any gross alteration in gene structure when DNA was digested withHindIII,EcoRI andTaqI. However, DNA from 3 -subunit-deficient patients, when digested withMspI and probed with PCC cDNA, revealed a unique 2.7-kb band not observed in blots of DNA from any other patient or 15 normal controls. We conclude that this alteredMspI restriction map is the result of a mutation in the subunit gene of these patients.  相似文献

Zusammenfassung 1974–1983 waren in einem mit 600 künstlichen Nisthöhlen ausgestatteten Lärchen-Versuchsforst (52.27 N, 7.15 E) bei Lingen/Emsland beim Trauerschnäpper 20,7% der nachweislich mit einem polygamen verpaart. Neben 51 Bigynie-Fällen konnte auch 4mal Trigynie nachgewiesen werden. mit Polygamie-Nachweis waren im Mittel älter als die übrigen (2,71 bzw. 2,18 Jahre). Polygame hatten im Durchschnitt längere Flügel und ein größeres Gewicht als ohne Polygamie-Nachweis, was als Folge der unterschiedlichen Altersstruktur zu deuten ist. Brutkästen der Bigamisten waren im Mittel 258 m voneinander entfernt (Maximum 750 m). Bei Bruten ohne Nachweis eines fütternden lag die Anzahl ausgeflogener Junge (im Mittel 2,9) gesichert unter dem Bruterfolg von zwei anderen Gruppen (fütterndes bei einer Brut festgestellt 4,7; fütterndes bei 2–3 Bruten festgestellt 4,5). Zwischen den Schlüpfterminen von Bigamisten-Bruten lagen 1–16 Tage. Das Durchschnittsalter der in den jeweils früheren Bruten betrug 2,67, in den jeweils späteren Bruten 1,67 Jahre.
Polygyny in the Pied Flycatcher (Ficedula hypoleuca) at the western border of its Central European range
Summary 1974–1983 in a study area of larch forest (52.27 N, 7.15 E), with 600 nestboxes, near Lingen/Emsland in western Lower Saxony, polygyny among Pied Flycatchers was identified as participation of in feeding at 2 or 3 nestboxes (bigyny or trigyny). 20.7% of were shown to be paired with a polygamous . Beside 51 cases of bigyny 4 cases of trigyny were identified. The incidence of polygyny was also considered in relation to the border situation of the study area. Proven polygamous were on average older than other (2.71 as compared to 2.18 years). On average polygamous had longer wings and greater bodyweight than without polygamous record, which is to be interpreted as a result of differing age structure. On average the mean distance between nestboxes of bigamists was 258 m (max. 750 m). In cases of broods without recorded feeding (generally this would occur in cases of secondary broods of polygamous in which the assisted only rarely or not at all) the number of reared young (on average 2.9) was significantly lower than the breeding success realised by 2 other groups ( recorded as assisting in rearing one brood only 4.7; recorded as assisting in rearing 2–3 broods 4.5). A period of 1–16 days (on average 7) elapsed between the hatching dates of broods of bigamists. The average age of in the earlier broods was 2.67 yrs, in the later broods 1.67 yrs.

Metal toxicity from sources such as orthopaedic implants was investigated in terms of immune system hyper-reactivity to metal implant alloy degradation products. Lymphocyte response to serum protein complexed with metal from implant alloy degradation was investigated in this in vitro study using primary human lymphocytes from healthy volunteers (n = 10). Cobalt chromium molybdenum alloy (CoCrMo, ASTM F75) and titanium alloy (Ti6Al4V, ASTM F136) beads (70 m) were incubated in agitated human serum at 37 degrees Celsius to simulate naturally occurring metal implant alloy degradation processes. Particulate free serum samples, which were incubated with metal, were then separated into molecular weight based fractions. The amounts of soluble Cr and Ti within each serum fraction were measured and correlated with lymphocyte proliferation response to the individual serum fractions. Lymphocytes from each subject were cultured with 11 autologous molecular weight based serum fractions either with or without added metal. Two molecular weight ranges of human serum proteins were associated with the binding of Cr and Ti from CoCrMo and Ti implant alloy degradation (at < 30 and 180–330 kDa). High molecular weight serum proteins ( 180 kDa) demonstrated greater lymphocyte reactivity when complexed with metal released from CoCrMo alloy and Ti alloy than with low (5–30 kDa) and midrange (30–77 kDa) serum proteins. When the amount of lymphocyte stimulation was normalized to both the moles of metal and the moles of protein within each fraction (MetalProtein Complex Reactivity Index, MPCRI), Cr from CoCrMo alloy degradation demonstrated approximately 10 fold greater reactivity than Ti in the higher molecular weight serum proteins ( 180–250 kDa). This in vitro study demonstrated a lymphocyte proliferative response to both CoCrMo and Ti alloy metalloprotein degradation products. This response was greatest when the metals were complexed with high molecular weight proteins, and with metalprotein complexes formed from CoCrMo alloy degradation.  相似文献

Summary In 683 leprosy patients from Chiang Mai, Thailand, the associations between ABO blood groups, type and clinical features of leprosy, and electrophoretically identifiable serumprotein fractions (albumins: 1-, 2-, - and -globulins) were examined. Besides, the blood group frequencies in 388 leprosy patients were compared with suitable controls. Blood groups A and AB turned out to be somewhat more frequent in patients than in controls. Combined analysis with 31 series from literature reports gave X=1.0776; 2=1)=12.232. In comparisons within our group of patients which contained almost exclusively lepromatous and dimorphous patients a certain tendency towards more severe involvement of blood group A was observed within the lepromatous group and a higher frequency of eye involvement in group A was (weakly) significant (2=1)=6.188).As to serum proteins 1- and 2-globulins were decreased (weakly) significantly in blood group A patients who were at least 40 years old. Furthermore, a number of relationships of serum protein fractions with age, sex, and state of the infection, most of which are known from the literature, could be confirmed.  相似文献

Summary Phytophthora infestans has three kinds of somatic nuclei: an oval shaped nucleus (approx. 3.1×2.7 ) which stains diffusely except for a crescent shaped Feulgen positive cap which stains intensely; a granular nucleus whose contents are organized into a fairly constant number of stained bodies, and, a deeply staining condensed nucleus. The capped nucleus is thought to be metabolic or resting and the granular nucleus is thought to be dividing as it is most commonly found in hyphal tips. Attenuated forms of all three kinds of nuclei are found.Nuclear division is mitotic and intranuclear. Eight—ten chromosomes are seen at metaphase.Sporangia have a mean of 6.3 nuclei which is constant for age and strain of culture. Sporangia become multinucleate as a result of nuclear migration and not by division in the developing sporangium. Zoospores are usually uninucleate.The nuclear cap is persistent throughout nuclear division when it also divides. It is associated with flagella production and nuclear migration and has some of the properties of a blepharoplast.  相似文献

To clarify the social functions of play panting in chimpanzees, I investigated when they emitted play panting in social play and how the interactions were affected by the occurrence of play panting. The subjects were the M-group chimpanzees living in Mahale, Tanzania. The following observations were made: (1) chimpanzees emitted play panting when they were tickled or chased but rarely did so when they tickled or chased others. Chimpanzee play panting does not have the function of a play signal communicating that these aggressive actions are performed not as aggression but as play. (2) Chimpanzees emitted play panting more often when they received aggressive actions that supposedly elicited higher arousal. (3) A chimpanzee tended to continue to perform aggressive actions when the target emitted play panting. Play panting activates the interaction of social play by encouraging the performer to continue tickling or chasing. These results can be summarized as showing that chimpanzee play panting serves as positive feedback to the play partner for continuing somewhat fragile interactions, which may contain the risk of excessive arousal and the risk of confusing defensive actions by the target of the aggressive actions with real efforts to escape the situation.  相似文献

Growth and oxygen uptake of potato callus is faster in an oxygen-enriched atmosphere (70% oxygen, v/v; oxygen-callus) than in air (20% oxygen, v/v; air-callus). Especially the non-mitochondrial, so-called residual respiration is increased in oxygen-callus. The capacities of the mitochondrial respiratory pathways (cytochrome pathway, Vcyt and alternative pathway, Valt) are also higher in this callus. In both callus types only a small part of the alternative pathway capacity is used in uninhibited respiration. The lower oxygen uptake of air-callus at normal air oxygen pressures is partially due to diffusional impedance. Measurement of the respiratory parameters of air-callus in oxygen-saturated medium leads to higher values than measurement in air-saturated medium, although these values are still lower than those of oxygen-callus.ATP-production was calculated from the oxygen-uptake data and compared with the dry weight production of the callus to give values of 10.0 and 10.8 g dry weight produced.-mol ATP-1, for air-callus and oxygen-callus respectively. Summary The histochemical activities of succinic dehydrogenase (SDH), myofibrillar Adenosine triphosphatase (ATPase) and alpha glycerophosphate dehydrogenase were studied in serial sections of rat vastus lateralis (red) (RVL), gastrocnemius and diaphragm muscles. Three main fibre-types were distinguished. The Type I fibres of RVL and gastrocnemius muscles fell into two distinct groups: one category-Type IA showed very low ATPase activity. The second category of Type IB fibres displayed moderate ATPase reaction. The Type IA fibres were divisible into two sub-groups when tested for SDH reaction. Type IA1 fibres possessed a homogenous distribution of diformazan·granules throughout the fibre: Type IA2 fibres displayed characteristic moth-eaten pattern of diformazan localization. The diaphragm muscle did not show either Type IB or Type IA2 varieties. The great majority of Type I fibres were sub-type IA1 in the three fast muscles studied. It is also demonstrated here that an inherent heterogeneity exists between Type I fibres of diaphragm and leg muscles in regard to -GPD localization. This histochemical data emphasizes the fact that subdivision of Type I striated muscle fibres of mammalian animals into two sub-types is only approximate and that a further subcategorization is possible.  相似文献
