Female remating is a widespread behaviour, reported in several insect species. This behaviour can affect the efficiency of sterile insect technique (SIT); however, little is known about the postcopulatory behaviour of some pest species considered as candidates to be controlled by this technique, such as Drosophila suzukii (Matsumura, 1931) (Diptera: Drosophilidae). In this study, we investigated the effects of male and female sterilization on mating and remating behaviour of D. suzukii. First, we tested the occurrence of multiple mating in different combinations between sterile and fertile males and females. Then, we tested the effects of male and female sterility on female propensity to mate and remate. We found an overall low remating rate by D. suzukii females. Male sterility did not influence mating and remating likelihood; however, copula duration of sterile males was shorter compared to fertile males. On the other hand, sterile females were less likely to mate. Our findings encourage further research regarding the use of SIT to control D. suzukii.  相似文献

The paper summarizes the researches conducted on male sterility in Mercurialis annua. Totally sterile individuals are very scarce in the dioecious species showing as the other Mercuries, unisexual flowers devoid of rudiments of the opposite sex. From one sterile male mutant, a ‘sterile series’ was conducted and genetics was studied. Sterile, semisterile, restored fertile male lines were constructed as well as female lines containing the inducer gene of male sterility, both fertility restorers and the sensitive cytoplasm. Morphology and ontogeny of these isogenic lines were presented. Male sterile anthers (empty) present a splitted tapetum and an abnormal meiotic end. Restored fertile male lines were normal. The relative abundance of auxin and cytokinins was studied. A specific cytokinin pathway measured as a background in fertile lines, the cis-oxidized pathway characterised the ‘sterile series’. Restoration of normal meiosis and tapetum appeared for the highest quantities of cis-zeatin (669 ng instead of 192 ng/100 g fresh weight in totally sterile). Auxin quantities were abundant compared with the normal males. Gene expression in the ‘sterile series’ was also compared with the fertile lines. t-RNAs specific for normal females were expressed in the male ‘sterile series’. Hybridization kinetics and in vitro translations pf poly(A)+RNAs demonstrate specific sequences for each line. Comparisons between identical organs (normal fertile male/restored fertile male or normal female/female of the ‘sterile series’) exhibited nearly 10% differences. The results suggest that for stamen development, a cascade of regulators probably exists: sex genes acting on the induction of stamen or pistil, then genes for sterility/restoration of fertility acting in anthers. Fertility-sterility regulators control the synthesis of a specific cytokinin pathway. The new hormonal signals are linked to several specific genes expressed in the floral morphology characterizing each line of the ‘sterile series’.  相似文献

Background  

Interspecific hybrids of frogs of the genus Xenopus result in sterile hybrid males and fertile hybrid females. Previous work has demonstrated a dramatic asymmetrical pattern of misexpression in hybrid males compared to the two parental species with relatively few genes misexpressed in comparisons of hybrids and the maternal species (X. laevis) and dramatically more genes misexpressed in hybrids compared to the paternal species (X. muelleri). In this work, we examine the gene expression pattern in hybrid females of X. laevis × X. muelleri to determine if this asymmetrical pattern of expression also occurs in hybrid females.  相似文献

Inbreeding occurs in numerous animal and plant species. In haplodiploid hymenopterans with the widespread single locus complementary sex determination, the frequency of diploid males, which are produced at the expense of females, is increased under inbreeding. Diploid males in species of bees, ants and wasps are typically either unviable or effectively sterile and thus impose a severe genetic load on populations. However, a recent study indicated that diploid males can be reproductive in the gregarious parasitoid wasp Cotesia glomerata, effectively reducing the diploid male load. To understand the role of inbreeding as a potential selective pressure towards the evolution of diploid male fertility, we genotyped specimens collected in the field at four locations using microsatellite markers to estimate the ratio of sibling matings under natural conditions. Results show that more than half of all matings involved siblings. We argue that the frequent occurrence of inbreeding has driven the evolution of diploid male fertility.  相似文献

Subspecies of Drosophila pseudoobscura, one occurring in the United States and the other in Bogota, Columbia, exhibit Haldane's Rule in one direction of the cross. Additionally, D. pseudoobscura produces two sperm types: short, sterile sperm and long, fertile, sperm. Here I examine the relationship between the production of short and long sperm and hybrid sterility. Fertile and sterile hybrid males produce a greater proportion of short sperm compared to parental males with sterile hybrids producing mainly short, immotile sperm. Sperm transfer and storage patterns were similar between fertile hybrid and parental strains; and unexpectedly, short, immotile sperm from sterile hybrids were stored. These findings raise the question of whether different genetic mechanisms disrupt both sperm heteromorphic production and sperm motility and whether this indicates that females exert some control over sperm storage.  相似文献

The attractiveness and the mating success of fertile (normal and brownie, br) and sterile (rudimentary testis, rt) Drosophila montana males were studied in female-choice experiments (one female + two males). The normal males courted the sterile rt males nearly as much as they courted the females, while they courted other males less frequently. The females required a long courtship with the rt males and accepted them as their mating partner less often than normal males. The courtship behaviour of normal, br and rt males was recorded in single pair situations with a normal female. The courtships of the males of all types proceeded in a similar manner, but the successful courtships of the rt males included more licking and touching than those of the normal and br males. The licking and touching periods are probably the critical phases at which the females are able to detect differences in male fertility.  相似文献

Our recent study found that 30% of young genes were essential for viability that determines development through stages from embryo to pupae in Drosophila melanogaster, revealing rapidly evolving genetic components involved in the evolution of development. Meanwhile, many young genes did not produce complete lethal phenotype upon constitutive knockdown, suggesting that they may not be essential for viability. These genes, nevertheless, were fixed by natural selection, and might play an important functional role in their adult stage. Here we present a detailed demonstration that a newly duplicated serine-type endopeptidase gene that originated in the common ancestor in the D. melanogaster subgroup 6~11 million years ago, named Slfc, revealing a strong effect in post-eclosion. Although animals survived constitutive knockdown of Slfc to adult stage, however, their life span reduced significantly by two-thirds compared to wildtype. Furthermore, the Slfc-RNAi males dropped their fertility to less than 10% of the wildtype level, with over 80% of these males being sterile. The Slfc-RNAi females, on the other hand, showed a slight reduction in fertility. This case study demonstrates that a young gene can contribute to fitness on the three important traits of life history in adults, including the life expectancy, male fertility and female fertility, suggesting that new genes can quickly evolve and impact multiple phenotypes.  相似文献

In the Drosophila melanogaster subgroup, the yakuba species complex, D. yakuba, D. santomea and D. teissieri have identical mitochondrial genomes in spite of nuclear differentiation. The first two species can be readily hybridized in the laboratory and produce fertile females and sterile males. They also form hybrids in natural conditions. Nonetheless, the third species, D. teissieri, was thought to be unable to produce hybrids with either D. yakuba or D. santomea. This in turn posed the conundrum of why the three species shared a single mitochondrial genome. In this report, we show that D. teissieri can indeed hybridize with both D. yakuba and D. santomea. The resulting female hybrids from both crosses are fertile, whereas the hybrid males are sterile. We also characterize six isolating mechanisms that might be involved in keeping the three species apart. Our results open the possibility of studying the history of introgression in the yakuba species complex and dissecting the genetic basis of interspecific differences between these three species by genetic mapping.  相似文献

Polyandry, female mating with multiple males, is widespread across many taxa and almost ubiquitous in insects. This conflicts with the traditional idea that females are constrained by their comparatively large investment in each offspring, and so should only need to mate once or a few times. Females may need to mate multiply to gain sufficient sperm supplies to maintain their fertility, especially in species in which male promiscuity results in division of their ejaculate among many females. Here, we take a novel approach, utilizing wild‐caught individuals to explore how natural variation among females and males influences fertility gains for females. We studied this in the Malaysian stalk‐eyed fly species Teleopsis dalmanni. After an additional mating, females benefit from greatly increased fertility (proportion fertile eggs). Gains from multiple mating are not uniform across females; they are greatest when females have high fecundity or low fertility. Fertility gains also vary spatially, as we find an additional strong effect of the stream from which females were collected. Responses were unaffected by male mating history (males kept with females or in male‐only groups). Recent male mating may be of lesser importance because males in many species, including T. dalmanni, partition their ejaculate to maintain their fertility over many matings. This study highlights the importance of complementing laboratory studies with data on wild‐caught populations, where there is considerable heterogeneity between individuals. Future research should focus on environmental, demographic and genetic factors that are likely to significantly influence variation in individual female fecundity and fertility.  相似文献

A new type of hybrid sterility was investigated in D. melanogaster . Matings between strain 27 males from Para Wirra, South Australia, and Canton-S females produce 70–80% fully sterile male and female progeny. Strain 27 males produce sterile progeny when crossed to females of other geographic origins, but produce fertile progeny when crossed to a second sympatric strain. The sterility is avoided by lower rearing temperatures. Heat shock and tetracycline produce no improvement in the fertility of the hybrids. Normal flies produce sterile progeny when injected with, or fed, homogenates of sterile flies. A combination of maternal and paternal factors may interact to produce sterile hybrids by inhibiting gonad development.  相似文献

The release of sterile males to suppress the Mexican fruit fly Anastrepha ludens (Loew) population was simulated in field cages to compare the sexual performance of sterile males with wild females in the presence and absence of sterile females. Using different release ratios of sterile and wild individuals, this study showed that the presence of sterile females negatively affected the sexual performance of sterile males in mating with wild females. We found that the sterility induced was proportional to the magnitude of the sterile/fertile ratio. These results indicate that the release of sterile males without sterile females can significantly improve the efficiency of the sterile insect technique for the control of A. ludens.  相似文献

A test for coadaptation of nuclear and mitochondrial genomes was performed using the sibling species, Drosophila pseudoobscura and D. persimilis. Two lines of flies with ``disrupted' cytonuclear genotypes were constructed by repeated backcrossing of males from one species to females carrying mitochondrial DNA (mtDNA) from the other species. Each ``disrupted' strain was competed in population cages with the original stock of each species from which the recurrent males were obtained during the backcrossing. As such, the two species' mitochondrial types were competed reciprocally in the nuclear genetic environments of each species. The trajectories of mtDNA haplotypes were followed in discrete-generation population cages using a PCR-four-cutter approach. A significant increase in the frequency of D. pseudoobscura mtDNA was observed in each of four replicate cages with a D. pseudoobscura nuclear background. In the D. persimilis nuclear background, one cage actually showed an increase in frequency of D. pseudoobscura mtDNA, although together the four replicate cages show little change in frequency. These results were repeated after frequency perturbations and reinitiation of each cage. An analysis of fitness components revealed that fertility selection greatly outweighed viability selection in these cytonuclear competition experiments. The asymmetry of the fitnesses of the mtDNA haplotypes on the two genetic backgrounds is consistent in direction with the previously reported asymmetry of female fertility in backcrosses between these two species. While our experiments do not allow us to identify mtDNA as the sole source of fitness variation, at a minimum the data indicate a fitness association between nuclear fertility factors and the D. pseudoobscura mtDNA on its own genetic background.  相似文献

We have attempted to estimate the number of genes involved in postzygotic reproductive isolation between two closely related species, Drosophila simulans and Drosophila sechellia, by a novel approach that involves the use of high resolution two-dimensional gel electrophoresis (2DE) to examine testis proteins in parents, hybrids and fertile and sterile backcross progenies. The important results that have emerged from this study are as follows: (1) about 8% of about 1000 proteins examined showed divergence (presence/absence) between the two species; (2) by tracing individual proteins in parental, hybrid and backcross males, we were able to associate the divergent proteins with different chromosomes and found that most divergent proteins are associated with autosomes and very few with X chromosome, Y chromosome and cytoplasm; (3) when proteins showing both quantitative and qualitative differences between the two species were examined in F(1) hybrid males, most (97.4%) proteins were expressed at levels between the two parents and no sign of large scale changes in spot density was observed. All the proteins observed in the two parental species were present in F(1) hybrid males except two species-specific proteins that may be encoded (or regulated) by sex chromosomes; (4) when different fertile and sterile backcross male testes were compared, a few D. sechellia-specific proteins were identified to be consistently associated with male sterility. These results along with the observation that a large proportion (23.6%) of first generation backcross males were fertile show that hybrid male sterility between D. simulans and D. sechellia involves a relatively small number of genes. Role of large scale genetic changes due to general genome incompatibility is not supported. The results also suggest that the large effect of X chromosome on hybrid male sterility is not due to higher divergence of X chromosome than autosomes.  相似文献

The Hawaiian picture-winged flies in the genus Drosophila are a spectacular example of rapid evolutionary diversification in which sexual selection is considered an important mechanism for reproductive isolation and speciation. We investigated the behavioral reproductive isolation of two closely related and sympatric Hawaiian picture-winged Drosophila species, D. silvestris and D. heteroneura, which are known to hybridize in nature and produce viable and fertile hybrids. We compared the mating success of parental, F1 and backcross males in pairings with D. heteroneura females. The F1 males were produced by mating D. heteroneura males with D. silvestris females, and the backcross males were produced by mating F1 females with D. heteroneura males. The mating success of backcross males paired with D. heteroneura females were significantly reduced relative to that of parental and F1 males. This reduced mating success occurred primarily at a late stage of courtship where female choice of mate may be important. Two- and three-gene models demonstrate that epistasis involving a few genes could account for the observed variation in male mating success. These results are consistent with negative epistasis in the backcross generation and support the importance of sexual selection and negative epistasis in the evolution and maintenance of these species.  相似文献

Summary The inheritance of a partial male fertile phenotype in somatic hybrid B. napus plants that carried novel mtDNA was investigated over five backcross generations to B. napus Triton. The recurrent parent and the original somatic hybrid both contained chloroplasts resistant to atrazine. The F1 population contained mainly plants that were partial fertile, and some of the plants differed in mtDNA. The partial fertility predominated in the progeny of each backcross generation, but fully male sterile and fertile plants were also obtained. However, the sterility/fertility of these latter plants was not stable; both the fully male sterile and the male fertile plants produced progeny that were again predominantly partial male fertile. This pattern of predominant partial fertility but occasional sterile and fertile plants persisted in different nuclear backgrounds. Neither the male sterility nor the male fertility could be fixed and made stable. Test crosses indicated that restorer genes were probably not associated with appearance of male fertile plants. The evidence indicates that the behavior of the partial male fertility is cytoplasmic, and probably controlled by the chondriome.  相似文献

Drosophila subobscura and D. madeirensis are closely related species, the first distributed over a large area and the latter restricted to the island of Madeira. These species can hybridize in laboratory conditions, yielding fertile females and sterile males. Hybrid offspring show several phenotypic anomalies, including sex combs on the second and third pairs of legs in males. The extra sex comb trait is a homeotic transformation of the mesothoracic and metathoracic legs into prothoracic legs. This anomaly is observed almost exclusively in F1 males with D. madeirensis mothers. Analysis of backcross males shows that D. subobscura and D. madeirensis have diverged at a minimum of four autosomal loci affecting the extra sex comb anomaly. In addition, some incompatibility involving the X chromosome and/or a maternal effect is also implicated.  相似文献

To study the genetic differences responsible for the sterility of their male hybrids, we introgressed small segments of an X chromosome from Drosophila simulans into a pure Drosophila mauritiana genetic background, then assessed the fertility of males carrying heterospecific introgressions of varying size. Although this analysis examined less than 20% of the X chromosome (roughly 5% of the euchromatic portion of the D. simulans genome), and the segments were introgressed in only one direction, a minimum of four factors that contribute to hybrid male sterility were revealed. At least two of the factors exhibited strong epistasis: males carrying either factor alone were consistently fertile, whereas males carrying both factors together were always sterile. Distinct spermatogenic phenotypes were observed for sterile introgressions of different lengths, and it appeared that an interaction between introgressed segments also influenced the stage of spermatogenic defect. Males with one category of introgression often produced large quantities of motile sperm and were observed copulating, but never inseminated females. Evidently these two species have diverged at a large number of loci which have varied effects on hybrid male fertility. By extrapolation, we estimate that there are at least 40 such loci on the X chromosome alone. Because these species exhibit little DNA-sequence divergence at arbitrarily chosen loci, it seems unlikely that the extensive functional divergence observed could be due mainly to random genetic drift. Significant epistasis between conspecific genes appears to be a common component of hybrid sterility between recently diverged species of Drosophila. The linkage relationships of interacting factors could shed light on the role played by epistatic selection in the dynamics of the allele substitutions responsible for reproductive barriers between species.  相似文献

We investigated the hypotheses that Liriomyza trifolii is a polyandrous species and that multiple mating is required to maximize female fecundity. In addition, the hypothesis that male copulating frequency reduces female fecundity was examined. Using sterile and fertile males, we found that L. trifolii females mate more than once during their life span, and that multiple mating is required to maximize female fecundity. A similar amount of sperm was transferred during five successive male copulations with an equal insemination probability. However, the probability of reproduction failure was strongly correlated with the male copulating sequence. Moreover, the failure rate of sperm transfer was found to be 6.8%. In light of these results, possible selection pressures driving for polyandry in L. trifolii are discussed.  相似文献

The sex‐ratio (SR), defined as the proportion of males, has been studied in three North American colonizing populations of Drosophila subobscura (Eureka, Davis and Gilroy). The proportion of sexes under laboratory conditions was studied using the one‐generation serial transfer technique in one‐ and two‐species populations, to infer whether biased SR affects the outcome when competing with Drosophila pseudoobscura, another member of the same group now in sympatry with D. subobscura in North America. The wild samples of D. subobscura yielded a significantly higher number of males than females during those months where the species is more abundant. However, there was no significant deviation in the 1 : 1 proportion of sexes in the descendants of D. subobscura at any of the experimental conditions. On the contrary, D. pseudoobscura produced a higher proportion of females which could be responsible for the exclusion of D. subobscura in laboratory competition experiments with overlapping generations. Thus, if sexes are equal at birth and survival is similar, the preponderance of males of D. subobscura in our wild collections could indicate greater activity and probably greater chance of dispersal of males versus females especially under favourable conditions.  相似文献

Chromosome numbers and fertility studies of 73 male and 30 female flowering germplasm accessions of Dioscorea alata L. were carried out. All males were tetraploids showing the same chromosome number (n=20 or 2n=40) and were pollen fertile (10.9–96.2%), most of them being highly fertile. Among the female the majority were higher ploids (hexa-and octoploids; 2n=60 and 80) and sexually completely sterile. There were only two tetraploid female accessions which were sexually fertile. Pollination studies revealed that seed sterility in D. alata was due to female sterility associated with the occurrence of higher levels of ploidy. The female sex-limited occurrence of higher polyploidy and sterility observed in D. alata is a curious situation among dioecious higher plants.  相似文献
