转基因烟草中Bt毒蛋白基因的表达行为   总被引:4,自引:0,他引:4  
Bt toxin genes were the insecticidal genes most widely used in genetic engineering of pest resistant plant, were of important significance to study their expression behavior in transgenic plants. In this work, a plant expression vector, pBinMoBc, was constructed. It contained the Cry IA(c) gene under control of chimeric OM promoter and the Ω factor. The vector was transferred into tobacco (Nicotiana tabacum L.) plant via Agrobacterium-mediated transformation. ELISA assay showed that the expression levels of the Cry IA(c) gene in transgenic tobacco plants were significantly higher than that in wild-type tobacco plants. The highest could be up to 0.255% of total soluble proteins; the expression level of CryIA(c) gene in transgenic tobacco plant was changeable during the development stages of tobacco plant. Bioassay showed that pBinMoBc transgenic tobacco plants had more notable insecticidal activity than the wild-type tobacco plants. The above results indicated that pBinMoBc was an effective pest-resistent plant expression vector. This study would be very helpful in screening transgenic cotton with high resistance to cotton bollworm (Heliothis armigeva Hubner).  相似文献

该研究以转高山离子芥的CbPLDα、CbPLDβ基因烟草为材料,研究了渗透调节物质和保护酶系对PEG6000溶液模拟干旱胁迫的响应机制.结果表明:渗透调节物质脯氨酸、可溶性糖、可溶性蛋白分别以各自不同的响应方式在干旱胁迫下增强转基因烟草的抗旱性,且在所有浓度PEG6000模拟的干旱胁迫下,转基因烟草的脯氨酸、可溶性糖、可溶性蛋白的含量始终显著高于野生型烟草(P<0.05).说明干旱胁迫下两种转基因烟草的渗透调节能力要强于野生型烟草.保护酶系中,超氧化物歧化酶(SOD)和过氧化物酶(POD)在减轻干旱胁迫下转基因烟草膜脂过氧化伤害中起到协同互补作用,而过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)在干旱胁迫下转基因烟草清除过氧化氢机制中发挥主要作用,说明保护酶系在抵制干旱胁迫和保护转基因烟草免受干旱伤害方面具有重要的生物学功能,这从生理角度揭示了高山离子芥CbPLDα、CbPLDβ响应干旱的生理生态机理.综上,高山离子芥CbPLDα、CbPLDβ基因参与了干旱胁迫下烟草的膜稳定性调节、渗透调节物质的积累和抗氧化酶系的调控.该研究结果为提高植物抗旱性研究及应用提供了新的基因资源,对于加强PLD功能研究、补充植物抗干旱理论及抗低温干旱育种种质资源的开发利用具有重要意义.  相似文献

The adoption of pest-resistant transgenic plants to reduce yield loss and pesticide utilization has been successful in the past three decades. Recently, transgenic plant expressing double-stranded RNA (dsRNA) targeting pest genes emerges as a promising strategy for improving pest resistance in crops. The steroid hormone, 20-hydroxyecdysone (20E), predominately controls insect molting via its nuclear receptor complex, EcR-USP. Here we report that pest resistance is improved in transgenic tobacco plants expressing dsRNA of EcR from the cotton bollworm, Helicoverpa armigera, a serious lepidopteran pest for a variety of crops. When H. armigera larvae were fed with the whole transgenic tobacco plants expressing EcR dsRNA, resistance to H. armigera was significantly improved in transgenic plants. Meanwhile, when H. armigera larvae were fed with leaves of transgenic tobacco plants expressing EcR dsRNA, its EcR mRNA level was dramatically decreased causing molting defects and larval lethality. In addition, the transgenic tobacco plants expressing H. armigera EcR dsRNA were also resistant to another lepidopteran pest, the beet armyworm, Spodoptera exigua, due to the high similarity in the nucleotide sequences of their EcR genes. This study provides additional evidence that transgenic plant expressing dsRNA targeting insect-associated genes is able to improve pest resistance.  相似文献

来源于昆虫病毒和动物的抗细胞凋亡基因能够诱导植物对生物或者非生物胁迫产生抗性.但其抗性机理有不同甚至相反的报道.本研究将来源于苜蓿银纹夜蛾核多角体病毒的p35基因转化烟草,T1代转化烟草Western blotting检测P35蛋白的表达,转化烟草接种烟草花叶病毒(Tobacco mosaic virus,TMV)抗病效果增强.进一步的抗病机理研究表明,转化和野生型烟草感染TMV后诱导过氧化氢积累无明显区别,野生型烟草感染24 h后出现DNA Laddering而转化烟草则没有;Western blotting结果显示PR-1蛋白表达没有显著差异.但接种另外一种病原真菌核盘茵(Sclerotiniasclerotiorum)后的RT-PCR分析结果表明,表达P35蛋白的烟草可增强感染核盘菌后PR-1基因的转录.而且表达时间提前.以上结果说明p35基因介导的广谱抗病反应的机理与接种的不同病原有关,对不同病原物的抗病机理存在差异,除抑制细胞凋亡外,还可能通过激活PR基因的表达提高对病原物的抗病能力.  相似文献

Hypersensitive response-assisting protein (HRAP) has been previously reported as an amphipathic plant protein isolated from sweet pepper that intensifies the harpin(Pss)-mediated hypersensitive response (HR). The hrap gene has no appreciable similarity to any other known sequences, and its activity can be rapidly induced by incompatible pathogen infection. To assess the function of the hrap gene in plant disease resistance, the CaMV 35S promoter was used to express sweet pepper hrap in transgenic tobacco. Compared with wild-type tobacco, transgenic tobacco plants exhibit more sensitivity to harpin(Pss) and show resistance to virulent pathogens (Pseudomonas syringae pv. tabaci and Erwinia carotovora subsp. carotovora). This disease resistance of transgenic tobacco does not originate from a constitutive HR, because endogenous level of salicylic acid and hsr203J mRNA showed similarities in transgenic and wildtype tobacco under noninfected conditions. However, following a virulent pathogen infection in hrap transgenic tobacco, hsr203J was rapidly induced and a micro-HR necrosis was visualized by trypan blue staining in the infiltration area. Consequently, we suggest that the disease resistance of transgenic plants may result from the induction of a HR by a virulent pathogen infection.  相似文献

为了对牡丹病程相关蛋白1(PsPRI)基因功能进行研究,构建了PsPRI基因超表达载体pBI121-PsPRI,通过农杆菌介导法将PsPR1基因转入普通烟草NC89中。经过抗性筛选和RT-PCR分子检测,获得含风PR1转基因植株。对转化烟草的T0代植株进行抗病性鉴定,发现转APRIJ基因烟草能轻微增强对烟草黑胫病的抗性,说明PsPRI基因具有抗烟草黑胫病原菌(Phytophthoraparasiticavar.nicotianae)的功能。  相似文献

将多肽抗生素apidaecin基因与病程相关蛋白的信号肽序列融合,构建了apidaecin的分泌型植物表达载体、apidaecin与另一多肽抗生素Shiva\|I的双价分泌型植物表达载体,以本实验室原来构建的Shiva-I分泌型植物表达载体做对照,转化了模式植物烟草。对3种转基因植物进行了分子检测,转化再生苗95%为PCR阳性,Southern杂交结果进一步证明外源基因已经整合到了烟草基因组中,RT-PCR检测表明外源基因可以在转基因烟草内正常转录。对T0代转基因烟草进行烟草野火病的抗病性实验,从3种转基因烟草中都得到了抗病植株,病情指数分析的初步结果显示,双价转基因烟草抗病性最好,apidaecin的次之,Shiva-I的最差。  相似文献

构建了含草甘膦抗性突变基因(aroAM12)和人工合成重组Bt抗虫基因(Bts1m)的植物表达载体pCM12_s1m。aroAM12基因的表达由CaMV35S启动子控制,Bts1m基因的表达由2E_CaMV35S启动子和Ω因子控制。通过农杆菌介导,将aroAM12和Bts1m基因转化到烟草中,转基因烟草通过在含草甘膦的MS培养基上筛选而获得。Southern blot分析表明所有经过草甘膦筛选出的转化植株都整合有aroAM12基因,约70%的转化植株同时整合有aroAM12和Bts1m基因。Northern blot、Immunodot blot分析进一步证明整合的两个基因在转录、翻译水平上均进行了表达,不同植株之间表达存在着差异。草甘膦抗性和虫试实验证明,获得的转基因烟草对草甘膦和烟青虫具有很强的抗性。  相似文献

In Arabidopsis , NPR1 ( AtNPR1 ) regulates salicylic acid (SA)-mediated activation of PR genes at the onset of systemic acquired resistance. AtNPR1 also modulates SA-induced suppression of jasmonic acid-responsive gene expression, and npr1 mutants manifest enhanced herbivore resistance. We have raised stable transgenic tobacco lines, expressing AtNPR1 constitutively, which showed elevated expression of PR1 and PR2 genes upon SA treatment. Herbivore bioassays with a generalist polyphagous pest, Spodoptera litura , revealed that the transgenic lines exhibited enhanced resistance compared to the wild-type plants, particularly with respect to younger larval populations. Insect-mediated injury induced several protease inhibitors (PIs), more significantly a 40-kDa serine PI in all the tobacco lines, but the induction was higher in the transgenic plants. We show in this communication that heterologous expression of AtNPR1 provides enhanced resistance to early larval populations of the herbivore, Spodoptera in transgenic tobacco plants.  相似文献

为了尽快地将抗病转基因烟草品种应用于生产,在选育抗病优良株系的同时,进行了转基因株系的大田抗病性鉴定。结果表明:(1)在田间自然发病情况下,转基因烟草的NC89各株系的发病率及病情指数显著低于对照NC89,对CMV的相对防治效果为55%-70%,表现较强的抗病性;同时对TMV也有一定的抗病力;(2)转基因烟草的产量、产值也明显高于对照。  相似文献

采用遗传转化技术获得了整合有拟南芥AtELHYPRP2(EARLI1-LIKE HYBRID PROLINE-RICH PROTEIN 2,AT4G12500)基因的转基因烟草株系,研究了该基因编码蛋白对真菌病原体赤霉菌的抗性及其亚细胞定位特征。以拟南芥Col-0生态型基因组DNA为模板,通过聚合酶链反应扩增AtELHYPRP2基因编码序列,经限制性酶切后连接至pCAMBIA1302载体,构建产生pCAMBIA1302-AtELHYPRP2-GFP融合表达载体。进一步采用农杆菌LBA4404转化烟草叶片外植体,筛选得到转基因烟草植株。RT-PCR、Western blotting印迹分析结果显示,AtELHYPRP2基因在转化体中可以有效表达。激光共聚焦显微观察发现AtELHYPRP2-GFP融合蛋白产生的绿色荧光与碘化丙啶染色后产生的红色荧光能够重合,说明AtELHYPRP2蛋白定位于细胞表面。真菌侵染实验结果显示,组成性表达AtELHYPRP2基因能够增强烟草对赤霉菌的抗性,被侵染部位有明显的H2O2积累。转基因烟草植株中PR1基因的本底表达水平比野生型高,PR1和PR5基因的系统表达水平比野生型高,说明AtELHYPRP2基因可能在SAR反应中具有一定的作用。  相似文献

This study generated transgenic tobacco plants expressing trehalose phosphorylase of Pleurotus sajor-caju (PsTP) constitutively under the control of the cauliflower mosaic virus (CaMV) 35S promoter. Sixteen transgenic lines were selected by genomic Southern blot analysis for further study. Unlike yeast TPS1-transformed or Escherichia coli TPS1-transformed tobacco or potato, all of the PsTP transgenic tobacco lines showed normal growth phenotypes both in the culture tubes and soil mixture. The study measured the trehalose contents of PsTP-transformed tobacco plants as well as the wild type and empty vector-transformed control plants. Results showed that the PsTP transformant contained 6.3molg–1 of plant tissues, while the wild type and the control plants had only minimal levels of trehalose. Because this study detected a significant amount of trehalose in PsTP transgenic tobacco plants, it decided to carry out a bioanalysis of the PsTP transgenic tobacco plants by drought treatment by not watering the plants for over 10days. A significant difference in drought resistance was observed from the second nonwatering day between the transgenic and the control tobacco plants. The transgenic tobacco plants had normal growth and did not wither, while the wild type and the only empty vector-transformed control plants withered severely. Among all the transgenic lines, line 10-4 showed the strongest resistance to drought stress. It did not wither even after 10days without watering. In addition, when the drought resistance of PsTP transgenic tobacco plants was tested using detached leaves, most transgenic plants, except one line, showed better capacity to retain water than the empty vector-transformed transgenic plant.  相似文献

Transgenic tobacco plants expressing the coat protein (CP) gene of tobacco mosaic virus were tested for resistance against infection by five other tobamoviruses sharing 45-82% homology in CP amino acid sequence with the CP of tobacco mosaic virus. The transgenic plants (CP+) showed significant delays in systemic disease development after inoculation with tomato mosaic virus or tobacco mild green mosaic virus compared to the control (CP-) plants, but showed no resistance against infection by ribgrass mosaic virus. On a transgenic local lesion host, the CP+ plants showed greatly reduced numbers of necrotic lesions compared to the CP- plants after inoculation with tomato mosaic virus, pepper mild mottle virus, tobacco mild green mosaic virus, and Odontoglossum ringspot virus but not ribgrass mosaic virus. The implications of these results are discussed in relation to the possible mechanism(s) of CP-mediated protection.  相似文献

The work was aimed at investigation of primary reactions of plant cell metabolism in response to salt stress influence. It was found that phospholipase D regulatory enzyme is activated in wild type tobacco plants and transgenic cax1 tobacco plants during the early stages of salt stress influence. We have shown that disturbance of intracellular calcium ions homeostasis and oppression of phospholipase D activity decrease resistance of tobacco plants under salinity influence and also indicate the implication of such systems to signaling during stress adaptation of plants.  相似文献

Tobacco (Nicotiana tabactum L. ) "NC89" plants were transformed with deletion mutant of cucumber mosaic virus (CMV) movement protein (MP) gene and full-length CMV MP gene, respectively. The transformed plants were analyzed with polymerase chain reaction (PCR), PCR-Southem, Southern and Western blots. R0 generation of the transgenic plants were inoculated with CMV. Five out of 10 lines of tobacco plants (BMPK) transformed with CMV MP deletion mutant gene showed high resistance to CMV infection and remained symptomless for up to 50 days post-inoculation. In contrast, tobacco plants (BMPR) transformed with full-length CMV MP gene did not show resistance to CMV infection. However, most of the infected full-length CMV MP gene transgenic plants recovered by showing none or very mild mosaic symptoms in 40 days post-inoculation. The results of R1 generation of the BMPK transgenic plants tested under field conditions showed that all 5 lines of transgenic plants could delay the virus disease development.  相似文献
