  • 1.1. The neuroendocrine caudodorsal cells (CDCs) of Lymnaea stagnalis are a network of about 100 electrotonically coupled neurones. The CDCs release multiple peptides, including an ovulation hormone, during a period of electrical activity, the CDC-discharge.
  • 2.2. In isolated brains, a similar period of electrical activity (the afterdischarge) can be induced in all CDCs by a period of intracellular repetitive suprathreshold stimulation of one CDC.
  • 3.3. In order to study the regulation of this electrical behaviour in the absence of electrical interactions and in a controlled environment, experiments were performed on CDCs in dissociated cell culture.
  • 4.4. Methods for isolation and cell culture are described. Cell cultures had long-term viability and outgrowth of neurities occurred under serum-free conditions.
  • 5.5. CDCs in cell culture maintained their capability of producing afterdischarges upon electrical stimulation. Cells in culture appeared more excitable than cells in the intact isolated brain.
  • 6.6. The characteristic responses of CDCs in intact isolated brains to acetylcholine and FMRFamide were preserved in cultured CDCs. Both agents induced a transient hyperpolarization of the membrane, inexcitability and inhibition of an ongoing discharge.
  • 7.7. In experiments where isolated CDCs were closely apposed, but physically separate, it was found that an afterdischarge in one CDC could induce a discharge in the other CDC.
  • 8.8. These results confirm previous results which showed that an excitatory factor is released from the brain during the afterdischarge (Ter Maat et al., 1988, Brain Res., 43, 77–82), and demonstrate that this excitatory factor is released from the CDCs themselves.

  • 1.1. A potentiometric method for the assay of cholinesterase has been proposed and compared with a colorimetric assay.
  • 2.2. Main kinetic parameters of cholinesterase from Hypostomus punctatus brain were determined indicating that true acetylcholinesterase is by far the predominant enzyme in the brain of this fish.
  • 3.3. We have compared our data with published results described from other fish species.
  • 4.4. The enzyme inhibition achieved after 3 hr incubation of brain homogenates with ethyl-parathion have indicated that this enzyme shows a characteristic organophosphorous sensitive behavior.

  • 1.1. A simple procedure for isolation of high molecular weight genomic DNA, and RNA, from Streptococcus sobrinus OMZ176 is described.
  • 2.2. Cell cultures were grown aerobically for 10 hr.
  • 3.3. Spheroplast formation and lysis was achieved by mutanolysin/lysozyme-dependent digestion of the cell wall, followed by N-lauroylsarcosinate-mediated lysis.
  • 4.4. Nucleic acids were isolated directly from cell-lysates using cesium-trifluoroacetate (CsTFA) densitygradient centrifugation.
  • 5.5. Three different centrifugation regimes were tested: self-generated density gradients in a fixed angle rotor; self-generated density-gradients in a swinging-bucket rotor; pre-formed density-gradients in a swinging-bucket rotor.
  • 6.6. Genomic DNA isolated by the CsTFA-procedure was found to have higher purity as compared to genomic DNA isolated in a conventional CsCl gradient.
  • 7.7. Isolated DNA was shown to be of a quality suitable for applications in molecular biology.

  • 1.1. Arginase, ornithine decarboxylase and S-adenosylmethionine decarboxylase are active in both retina and brain. Activity is higher in cerebellum than in the cerebral hemispheres and optical lobes.
  • 2.2. Arginase and ornithine decarboxylase are very active in the retina of very young chicks, while S-adenosylmethionine decarboxylase is poorly active. By contrast, S-adenosylmethionine decarboxylase is much more active in brain.
  • 3.3. The pattern of activity during development is different; only ornithine decarboxylase is very active during embryonal life; S-adenosylmethionine decarboxylase, at all events in brain, is more active in adult life.
  • 4.4. Ornithine decarboxylase is inhibited in vitro by α-difluoromethylornithine, but not in vivo. Diaminopropane inhibits brain ornithine decarboxylase, but does not induce an ornithine decarboxylase-antizyme.
  • 5.5. Methylglyoxal bis(guanylhydrazone) promotes an increase of S-adenosylmethionine decarboxylase activity in both the brain and the retina in vivo.

  • 1.The levels of water, Na, K, Ca and Mg in blood serum, brain and kidney and aldosterone level in blood of Naja haje haje were studied during the different phases of the annual cycle.
  • 2.The water content in the tissues studied displayed only minor changes as the animals passed from one phase to the other.
  • 3.A significant increase in Na was recorded in the brain during the different phases indicating a depressed sodium pump, whereas the blood Na level showed a significant decrease during hibernation.
  • 4.K increased in blood serum, brain and kidney during hibernation, while a nonsignificant decrease was found in blood serum during arousal. The brain may act as a potassium reservoir.
  • 5.An increase in Ca and Mg concentration was recorded in blood serum, brain and kidney during prehibernation and hibernation. The data suggested a homeostatic function in the transport and metabolism of these cations.
  • 6.Aldosterone exhibited a highly significant decrease especially during hibernation. The aldosterone regulation of ionic composition is discussed.
  • 7.Na/K and Ca/Mg ratios in the brain may explain the decreased excitability during winter torpor.

  • 1.1. Co-isolating proteins (Mr 170,000–220,000) from sodium channel preparations made from the electric organ of the electric eel (Electrophorus electricus) were detected on Western blots using monoclonal a antibodies.
  • 2.2. Similar protein patterns were seen on immunoblots containing immunoprecipitated protein from eel muscle and brain tissues but not heart.
  • 3.3. These co-isolating proteins could be separated from the mature TTX-sensitive channel protein (Mr 280,000) using a lentil lectin-Sepharose column.
  • 4.4. The 180 kDa proteins do not appear to be channel-related and can be detected as contaminants in electroplax sodium channel preparations using the monoclonal antibodies described here.

  • 1.1. The binding of [14C]cholesterol into rat brain mitochondrial membranes follows an exponential path described by the general formula y = a.ebx. [14C]cholesterol glucoside binding has a sigmoidal character where the “best-fit” curve of this type of binding is the one described by the Hill equation with Hill coefficient h = 2.06. These findings suggest a positive cooperativity in the binding of both compounds into rat brain mitochondrial membranes.
  • 2.2. The specific activity of the outer mitochondrial membrane enzyme monoamine oxidase was linearly decreased at different concentration of cholesterol or its glucoside.
  • 3.3. The specific activity of the inner mitochondrial membrane enzyme succinate-cytochrome c reductase was linearly decreased, while that of Rotenone-sensitive NADH-cytochrome c reductase was exponentially increased, at different concentrations of cholesterol.
  • 4.4. These results are discussed in terms of specific interactions of cholesterol with constituent mitochondrial membrane lipids and their implications for deviations from normal neuronal function.

  • 1.1. Treatment of isolated rat liver mitochondria with methyl methacrylate (MM) produced membrane disruption as evidenced by the release of citrate synthase, and changes in the ultrastructure of mitochondria.
  • 2.2. At concentration 0.1%, MM uncoupled oxidative phosphorylation as evidenced by stimulation of state 4 respiration supported either by pyruvate plus malate or succinate (+rotenone) and ATP-ase activity in intact mitochondria.
  • 3.3. At concentration 1% MM stimulated ATP-ase activity in intact mitochondria and succinate (+rotenone) oxidation at state 4 and was without effect on this substrate oxidation at state 3.
  • 4.4. MM inhibited pyruvate plus malate oxidation either at state 3 or in the presence of uncoupling agents.
  • 5.5. MM inhibited the NADH oxidase of electron transport particles at a concentration which failed to inhibit either succinic oxidase or the NADH-ferricyanide reductase activity.
  • 6.6. The data presented suggest that in the isolated mitochondria MM inhibits NADH oxidation in the vicinity of the rotenone sensitive site of complex I.
  • 7.7. The general conclusion is that MM may block an electron transport and to uncouple oxidative phosphorylation in rat liver mitochondria. The overall in vitro effect would be to prevent ATP synthesis which could result in cell death under in vivo conditions.

  • 1.1. The incorporation of myo-[2-3H]inositol into phosphatidylinositols was unmodified in brain cortex miniprisms from convulsant rats.

  • 3.3. The specific activity of brain trehalase was maximum when the insects were in active feeding periods, minimum activity appearing during moulting phases.
  • 4.4. During diapausing development, trehalosemia was very high at the beginning of pupal life, particularly when insects were exposed to wintering conditions.
  • 5.5. When diapause was broken, trehalosemia fell, announcing adult emergence.
  • 6.6. Brain trehalase activity showed the same qualitative variations as in non-diapausing larvae, but with rather lower values.
  • 7.7. During pupal life, brain trehalase activity decreased markedly during the long period necessary to obtain diapause breakdown.
  • 8.8. Wintering conditions allow a progressive increase of brain trehalase activity, which preceded the fall of trehalosemia.
