Background

Bacteria are associated with the gut, fat bodies and reproductive organs of stored product mites (Acari: Astigmata). The mites are pests due to the production of allergens. Addition of antibiotics to diets can help to characterize the association between mites and bacteria.

Methodology and Principal Findings

Ampicillin, neomycin and streptomycin were added to the diets of mites and the effects on mite population growth (Acarus siro, Lepidoglyphus destructor and Tyrophagus putrescentiae) and associated bacterial community structure were assessed. Mites were treated by antibiotic supplementation (1 mgg−1 of diet) for 21 days and numbers of mites and bacterial communities were analyzed and compared to the untreated control. Bacterial quantities, determined by real-time PCR, significantly decreased in antibiotic treated specimens from 5 to 30 times in A. siro and T. putrescentiae, while no decline was observed in L. destructor. Streptomycin treatment eliminated Bartonella-like bacteria in the both A. siro and T. putrescentiae and Cardinium in T. putrescentiae. Solitalea-like bacteria proportion increased in the communities of neomycin and streptomycin treated A. siro specimens. Kocuria proportion increased in the bacterial communities of ampicillin and streptomycin treated A. siro and neomycin and streptomycin treated L. destructor.

Conclusions/Significance

The work demonstrated the changes of mite associated bacterial community under antibiotic pressure in pests of medical importance. Pre-treatment of mites by 1 mgg−1 antibiotic diets improved mite fitness as indicated accelerated population growth of A. siro pretreated streptomycin and neomycin and L. destructor pretreated by neomycin. All tested antibiotics supplemented to diets caused the decrease of mite growth rate in comparison to the control diet.  相似文献

The acaricidal activity of 54 commercially available plant essential oils against Tyrophagus putrescentiae adults was compared with those of benzyl benzoate and N, N-diethyl-m-toluamide (deet). Responses varied with test oils. In a test with an impregnated fabric disc bioassay at 12.7 μg/cm2, over 80% mortality was observed with bay, citronella java, clove bud, clove leaf, lemongrass, nutmeg, oregano, pimento berry, thyme red, and thyme white oils. These oils were more toxic to T. putrescentiae adults than benzyl benzoate and deet. At 6.4 μg/cm2, lemongrass and thyme white oils exhibited 76 and 84% mortality, respectively, whereas the acaricidal activity of the other essential oils was significantly decreased. In a fumigation test with adult mites, lemongrass and pimento berry oils were much more effective in closed containers than in open ones, indicating that the effects of these oils were largely due to action in the vapor phase. Plant essential oils described merit further study as potential T. putrescentiae control agents.  相似文献

The effect of Tyrophagus putrescentiae on Fusarium poae transmission and fungal community composition was studied in nonsterile barley grain. The experiments included following treatments: control barley without mites; barley containing l0 or 50 mites without preincubation on E poae (Tpl0 and TpS0); barley containing 10 or 50 mites after preincubation on E poae (FTp 10 and FTp50). The number of mites, suc cessful transfer of E poae, and changes in the fungal communities were examined after 21 d of experiment. Increase of deoxynivalenol (DON) content in the barley was chosen as a criterion of successful Epoae transfer. The preincubation of T. putrescentiae on Epoae increased DON level approximately to 800 and 300μg/kg of grain for FTpl0 and FTpS0, respectively. T. putrescentiae population growth in FTpl0 was lower than in Tpl0, while no difference was found between FTp50 and Tp50. Fungal communities were compared by amplification, cloning and sequencing of ITS fragments, and operational taxonomic units (OTU) analysis. The OTU analysis did not support the transfer ofF. poae via mites. From the analyzed clones, only 13 cloned sequences clustered with E poae in an OTU defined at distance level 0.07. The related clones originated from FTpl0, Tpl 0, Tp50 and control treatments, but not from FTp50. However, the presence of E poae in FTp50 was confirmed by PCR amplification with specific primers. The observation may be explained by different effect of mite population density, that is, in the high density, (FTp50 treatment) the fungus was overgrazed, while the lower population density (FTp 10) supported E poae transfer.  相似文献

Generalist predatory mites are the common phytoseiid fauna in many agroecosystems, but little attention has been paid to their potential as biological control agents. In this study, we determined the functional responses of adult females of the generalist predator Neoseiulus barkeri Hughes on eggs, larvae, and adults of the two-spotted spider mite, Tetranychus urticae Koch, in the laboratory. Predation experiments were conducted on pepper leaf discs over a 24 h period at 25±1°C, 70–80% RH and 16L:8D photoperiod. Prey densities ranged 5 to 80 eggs, or 5 to 40 larvae, or 1 to 8 female adults of T. urticae per disc. The predation rate of N. barkeri adult females on T. urticae eggs was the same as on its larvae, but the predation rate on adult females was much lower. The role of generalist predatory mites in integrated and biological control of greenhouse pests was discussed.  相似文献

Mites of the genus Tyrophagus (Acari: Acaridae) are among the most widespread and common mites, inhabiting diverse natural and anthropogenic habitats. Some species are pests of agricultural products and stored food and/or live in house dust, causing allergies to humans. We sequenced 1.2 kb of the mitochondrial COI gene for 38 individuals belonging to seven species of Tyrophagus, including T. curvipenis, T. putrescentiae, T. fanetzhangorum, T. longior, T. perniciosus, and T. cf. similis. Molecular phylogenetic analyses (1) recovered two major clades corresponding to the presence or absence of eyespots, and (2) separated all included morphological species. Tyrophagus curvipenis and T. putrescentiae had the lowest between-species genetic distances (range, mean?±?SD): 14.20–16.30, 15.17?±?0.40 (K2P). The highest within-species variation was found in T. putrescentiae 0.00–4.33, 1.78?±?1.44 (K2P). In this species, we recovered two distinct groups; however, no geographical or ecological dissimilarities were observed between them. Based on our analyses, we document important morphological differences between T. curvipenis and T. putrescentiae. For the first time, we record the occurrence of T. curvipenis in the New World and suggest that it may be an emerging pest as it is currently spreading in agricultural produce.  相似文献

Background

Tyrophagus putrescentiae (Acari: Astigmata) and Fusarium sp. co-occur in poorly managed grain. In a laboratory experiment, mite grazing resulted in significant reduction of fungal mycelium on cultivation plates. The destruction of mycelium appeared to be a result of an interaction between the mites, fungi and associated bacteria.

Methodology and Principal Findings

A laboratory experiment was performed to simulate a situation of grain multiinfested by mites and Fusarium fungi. Changes of mite-associated bacterial community in T. putrescentiae were described in 3 habitats: (i) T. putrescentiae mites from a rearing diet prior to their transfer to fungal diet; (ii) fungal mycelium before mite introduction; (iii) mites after 7 day diet of each Fusarium avenaceum, F. culmorum, F. poae and F. verticillioides. Bacterial communities were characterized by 16 S rRNA gene sequencing. In total, 157 nearly full-length 16 S rRNA gene sequences from 9 samples representing selected habitats were analyzed. In the mites, the shift from rearing to fungal diet caused changes in mite associated bacterial community. A diverse bacterial community was associated with mites feeding on F. avenaceum, while feeding on the other three Fusarium spp. led to selection of a community dominated by Bacillaceae.

Conclusions/Significance

The work demonstrated changes of bacterial community associated with T. putrescentiae after shift to fungal diets suggesting selection for Bacillaceae species known as chitinase producers, which might participate in the fungal mycelium hydrolysis.  相似文献

Hypoaspis larvicolus (Acari: Laelapidae) (first report from Turkey) occurred together with Sancassania polyphyllae (Acari: Acaridae) on the larvae of the scarab beetle, Polyphylla fullo (Coleoptera: Scarabaeidae), that were feeding on the roots of strawberry in Aydin, Turkey. Laboratory studies were conducted to (1) observe whether H. larvicolus feeds and completes its life cycle on the various stages of S. polyphyllae or other astigmatid mites, such as Acarus siro, Carpoglyphus lactis and Tyrophagus putrescentiae (Acaridae), and to determine its population growth when feeding on these prey, and (2) to determine development periods, longevity and fecundity of H. larvicolus feeding on C. lactis. Hypoaspis larvicolus females did not feed on S. polyphyllae, but fed, developed and reproduced when A. siro, C. lactis or T. putrescentiae were provided as prey. Hypoaspis larvicolus is larviparous as well as oviparous. The female lays eggs or gives birth to larvae. If a female gives birth to a larva, it is attached under the female’s venter for 1–2 days, a phenomenon recorded for the first time in Hypoaspis; in fact, for the first time in mites. The results of the population growth experiments revealed that H. larvicolus feeding on C. lactis produced the highest number of eggs, juveniles and adults. The developmental periods of H. larvicolus feeding on C. lactis at life-cycle path I (larva to adult) and II (egg to adult) were 12.2?±?0.3 and 15.6?±?0.6 days (females) and 19.5?±?0.2 and 20.9?±?0.4 days (males), respectively. Longevity of females versus males of H. larvicolus was 120.6?±?7.2 versus 91.6?±?13.1 days (life cycle I) and 110.0?±?27.7 versus 118.3?±?10.9 days (life cycle II), respectively.  相似文献

Proteins and saccharides are the two most important nutrients of artificial insect diets. In this study, additional protein or saccharide sources were added to the diet, and their impact on the population increase of both the prey Tyrophagus putrescentiae Schrank (Acari: Acaridae) and the predator Neoseiulus barkeri Hughes (Acari: Phytoseiidae) was investigated. T. putrescentiae population increased by 319, 317 and 180 times within six weeks, when yeast powder, glucose or sugar was added to the basic wheat bran diet (diet mass: additive mass 10:3), respectively. However, T. putrescentiae population increased by only 70 times when reared on the basic diet. All three types of nutrients resulted in increased soluble saccharide level of mixed stages T. putrescentiae. Significant increase of soluble protein level was observed when yeast powder was added. When fed on T. putrescentiae reared on yeast powder, glucose or sugar added diets, the developmental duration of N. barkeri was shortened by 23, 23 and 33%, and the daily fecundity increased by 40, 20 and 27%, respectively. The proportion of N. barkeri female offspring was 64% when fed with T. putrescentiae reared on wheat bran, increased to 70% when yeast powder was added, and decreased to 59% and 58% when glucose and sugar was added, respectively. The commercial packaging requirement of N. barkeri is 80 mites per g. It generally takes 40 days from N. barkeri inoculation to reach this requirement, but this period was dramatically shortened to 20, 25 and 24 days when yeast powder, sugar and glucose were added to the diet of T. putrescentiae, respectively.  相似文献

The developmental rate of immature stages and the reproduction of adults of Tyrophagus putrescentiae (Schrank), T. neiswanderi Johnston and Bruce and Acarus farris (Oudemans) were examined at 70, 80 and 90% r.h. and a constant temperature of 25°C. At 70% r.h., T. putrescentiae and A. farris immature stages failed to reach the protonymph stage as 100% of the larvae died, whereas T. neiswanderi was able to complete development. The developmental time of all immature stages for the three species was significantly increased as relative humidity was reduced. The mobile stages were particularly susceptible, as the time needed to complete their development at lower relative humidities suffered greater increases than the egg stage. At 70% r.h., T. putrescentiae and A. farris were not able to lay eggs and only 24% of T. neiswanderi pairs were fertile. The reproductive parameters of the three species at the relative humidities at which they were able to lay eggs showed significant differences, except for the percentage of fertile mating at 80 and 90% r.h. As relative humidity increased, preoviposition period was reduced and fecundity and daily fecundity was increased, whereas the oviposition period showed different patterns for the three species. The intrinsic rate of increase (r m ) of T. neiswanderi at 70% r.h. was negative indicating that, at these conditions, mite populations of this species will diminish until they disappear. As relative humidity increased from 80 to 90% r.h. this parameter was almost twofold for both Tyrophagus species. The r m obtained for A. farris at 90% r.h. was similar to that of T. neiswanderi at the same humidity while at 80% r.h. it was very small so that the population doubling time was more than 84 days. The influence of relative humidity on biology of these mites and its practical application as control measure are discussed.  相似文献

Varying concentrations of powdered leaves of Eucalyptus and Mentha and rhizomes of Curcuma were evaluated in controlling Tyrophagus putrescentiae in wheat flour. At higher concentrations, all these plant materials significantly decreased the population build-up of the mite. Eucalyptus and Mentha leaf powders were effective at concentrations ranging from 100 to 5% in bringing about a decrease in the fecundity of the mite and reducing the egg numbers to 51.66 and 25.49 per female, respectively, as compared to 98.16 eggs per female in the controls. Rhizomes of Curcuma were effective even at a concentration of 0.1% reducing the egg laying to 7.66 eggs per female in wheat flour. These materials were more deleterious to immature stages (ova and larvae) than to the mature stages of T. putrescentiae. When tested on whole grains of wheat these plant materials showed more pronounced effects in controlling the mite population compared to that in wheat flour. Curcuma rhizomes were the most promising for possible use against T. putrescentiae.  相似文献

Eggs of 13 species of common, economically important stored-product pest mites (Acarus siro, A. gracilis, Tyrophagus putrescentiae, T. brevicrinatus, Tyroborus lini, Aleuroglyphus ovatus, Caloglyphus redikorzevi, C. oudemansi, Lepidoglyphus destructor, Glycyphagus domesticus, Aeroglyphus robustus, Chortoglyphus arcuatus and Carpoglyphus lactis) from four families (Acaridae, Glycyphagidae, Chortoglyphidae, Carpoglyphidae) were studied in order to build a diagnostic key. The morphological study dealt with shape, size and surface sculpturing of eggs. Morphological details were visualised using scanning electron microscopy. A key for distinguishing eggs at genus and species level was developed for the major stored-product mites.  相似文献

Little is known about the flow of chemical information fromhigher to lower levels within the animal food chain. However, this information may determine the behavior and distributionof many animals (e.g., that of potential prey) when exposedto direct and indirect cues of predation risk. We used herbivorousspider mites, Tetranychus urticae Koch (Tetranychidae) as amodel to examine the foraging and oviposition decisions thatprey make when exposed to these cues. We conducted laboratorytests to determine if the previous presence of predators (directcues) on leaf discs or the presence of injured conspecifics(indirect cues) alters the distribution of adults and eggs ofT. urticae. When given a choice, after 24 h, fewer adults and eggswere found on leaf discs that had previously contained specialistspider mite predators, Phytoseiulus persimilis Athias-Henriot (Phytoseiidae),than on discs unexposed to predators. Also, more T. urticaeemigrated from predator-exposed discs than from unexposed discs orfrom those that had previously contained nonpredatory mites(Tyrophagus putrescentiae, Acaridae). Finally, fewer T. urticaeforaged and laid eggs on predator-exposed discs or on thosewith artificially damaged conspecifics (eggs or dead adults)than on discs with intact conspecifics. Tetranychus urticaeprobably recognizes infochemicals (kairomones) from its predatorsor cues from injured spider mites and consequently avoids feedingor ovipositing in areas exposed to these cues. Recognition and avoidanceof kairomones from specialist predators by this prey are likelyto be hereditary, but avoidance of injured conspecifics maybe an adaptation to avoid predators that are not inherentlyrecognized. We discuss the behavioral and ecological implicationsof our findings.  相似文献

The pest potential of stored product mites depends on the reproduction rate that is affected by the environmental conditions. In this study we investigated the effect of temperature, ranging from 5 to 35°C, on the population growth of three important mite species, Acarus siro, Tyrophagus putrescentiae and Auleroglyphus ovatus at 85% r.h. Starting with 10 individuals the population increase of mites was observed after 3 weeks of cultivation, or after 6 weeks for those kept at low temperatures (5, 10, 12.5, and 15°C). The rate of increase was calculated for each temperature and species. The obtained data were fitted with polynomial models. The mite population growth rates increased with increasing moderate temperatures until 25°C, when r m -values were 0.179, 0.177 and 0.190 for A. siro, A. ovatus and T. putrescentiae, respectively. The lower development threshold was 10.2°C in all three species. Estimated upper temperature threshold was higher in T. putrescentiae (49°C) than in A. siro and A. ovatus (38°C). Simulation of the rate of population increase under ideal conditions, using real temperature records obtained from Czech grain stores, showed that the pest mite populations increase only during 3.5 months within a typical 9-month storage season in Central Europe. These results indicate that control of mites, be it chemical, physical or biological, is recommended during the months when allergens and pests are produced, i.e. from September to mid November and in May.  相似文献

Tyrophagus putrescentiae (Schrank) (Sarcoptiformes: Acaridae) is a serious pest of post-harvest durable stored food products for which traps are being developed as important tools for integrated pest management. This work addresses the effects of trap design, trap location, time of response, and light on the orientation of released groups of 7200 pedestrian T. putrescentiae to food-baited traps in experimental rooms maintained in full darkness at 28 °C and 70% RH. A standard trap made from a disposable Petri dish and four commercially produced stored-product pest traps of different designs were compared and evaluated for efficacy in trapping mites. Subsequent experiments evaluated the effect of trap placement, trapping duration, and light on mite capture. More mites were captured in the standard Petri dish trap when compared singly or side by side with four different commercial trap designs, so the standard trap was used in all other studies. Further experiments found that mites could be trapped 1 h after release at a distance of 2 m from the release point and at six meters from a mite source within 24 h. A greater number of mites were caught in traps placed along a wall than those away from a wall, demonstrating thigmotaxis to an edge. Traps at various heights on a shelf rack suggested that walking mites tended to be positively geotactic whether the source of mites was from the floor or from the top shelf at 1.8 m from the floor. More mites were caught in black-painted traps than in unpainted clear plastic traps in fully lighted rooms, suggesting that mites seek refuge from brightly lighted conditions. Light emitting diodes deployed with traps or assayed alone determined mites oriented positively to traps with violet or ultraviolet, with least preference to green, yellow, red and white lights. This research provides new information on orientation by T. putrescentiae that may help optimize trap designs and trap deployment for pest management decisions.  相似文献

Orius insidiosus is economically important for biological control of pests of vegetable and ornamental crops. To improve pest control with this predator, its densities in the crops can be enhanced by the provision of alternative foods, especially when prey are scarce. We therefore compared the performance of O. insidiosus on three alternative foods (Ricinus sp. pollen, bee pollen and the mixed stages of the astigmatid prey Tyrophagus putrescentiae) that are cheaper than frozen eggs of Ephestia kuehniella. Juvenile development was significantly shorter on E. kuehniella and T. putrescentiae than on Ricinus sp. pollen and on bee pollen and lowest without food. Female bugs had a higher oviposition rate when fed either E. kuehniella or T. putrescentiae, but produced fewer eggs when feeding on bee pollen and Ricinus sp. pollen. This shows that T. putrescentiae can possibly be used as cheap alternative food to boost predator populations in periods of low prey densities.  相似文献

Populations ofTyphlodromus doreenae Schicha on 18×14 cm arenas using pollen ofTypha orientalis Presl. as food, increased from an estimated 50–150 to 1,000–1,600 motiles per arena in the presence of mould mites,Tyrophagus putrescentiae (Schrank) and the fungusRhizopus stolonifer (Ehrenberg ex. Fr.) Lind.T. putrescentiae in association withR. stolonifer, developed pigmentation and were preyed on byT. doreenae which also became pigmented. UnpigmentedT. putrescentiae were not attacked byT. doreenae which remained pale coloured.T. doreenae died when supplied with the fungus alone. Bean plants dusted with pollen ofT. orientalis from a talcum powder dispenser supported a large population (7–12 stages/leaf) ofAmblyseius victoriensis (Womersley). These improvements to mass rearing ofT. doreenae andA. victoriensis are discussed with respect to their potential for commercialisation.  相似文献

Using incorporated devices, Tetranychus urticae spider mites were rinsed from hydroponically-grown lima bean plants, collected, separated and blow-dried. This yielded a reliable and large volume of eggs and larvae, which were fed to Amblyseius womersleyi rearings on 15×5cm2 polyethylene arenas. Of several feeding regimes tested, daily feeding of 10mg T. urticae eggs and larvae resulted in the highest predator population levels. The best harvest period was between 15 and 27 days, when predator density exceeded 600 mites per arena. A preliminary automatic mass-rearing device was tested for A. womersleyi. This incorporated both rearing and harvesting procedures. A micro-feeder was developed to supply the required volume of spider mites and maize pollen (1:1 mixture) to the predators. A Bakelite rearing arena reduced the space requirements of a polyethylene arena, was more durable and an essential component in the automatic mass-rearing and harvesting. Mite harvesting is carried out through the use of a vacuum-head harvester. Supplements of (sterilized) spider mites, pollen, vermiculite and wheat bran are automatically added to the predators. The devices for harvesting, filling and packing are incorporated and synchronized and the entire system is controlled by a single slide-switch. The design and system can be expanded without changing the basic processes and program, for example to adopt it for other species of predaceous mites.  相似文献

The mold mite, Tyrophagus putrescentiae (Shrank), frequently infests a variety of stored food products in ideal, but rather limited conditions. Major factors limiting survival of this mite are the temperature and humidity imposed on T. putrescentiae as it develops within and disperses among sites. However, since relative humidity is dependent upon air temperature, determining survivability in a habitat can be difficult in the presence of structural temperature variations. Vapor pressure deficit (VPD) provides a method of combining both relative humidity and temperature into a single number that can be used to determine conditions detrimental to mite survival. This study utilized a bioassay format to measure mortality of T. putrescentiae when exposed to a range of seven temperatures (5–35°C), 10 relative humidities (0–100% RH), 17 exposure times (0.5–240 h), with and without food. With these combinations of temperature and RH, mortality curves (mortality versus time) that displayed a sigmoidal relationship were used to calculate LT50 and LT90 estimates. These mortality estimates were then regressed on their associated VPD and the resulting regressions (LT50 and LT90) were significant at P < 0.0001, and provided acceptable R 2 values ≥0.83, regardless of whether food was present or not. At room temperature, threshold of VPD for T. putrescentiae development was below 8.2 mbar, this estimate being initially calculated from published values. For mites exposed to drier conditions, above 8.2 mbar, survival time was curtailed dependant on the magnitude of VPD. As the VPD exceeded 12 mbar, mites experienced substantial (>90%) mortality within 58 (33, 101) h; and further increasing VPD decreased the time of exposure to achieve mortality. This study demonstrates that making subtle changes in humidity or temperature to reach a target VPD may provide control of mite outbreaks and reduce areas inhabitable for T. putrescentiae. With the recent revision of the genus Tyrophagus (Fan and Zhang 2007), T. putrescentiae was split and the commonly encountered peridomestic mite was renamed T. communis. Voucher specimens of the species we used were identified as T. communis (B. OConnor, pers. comm.). However, there are current discussions as to which species name will be applied to the more common mite species (P. Klimov, pers. comm.). For the purposes of this paper we will continue to use T. putrescentiae.  相似文献

Several factitious foods were assessed for rearing the anthocorid predators Orius thripoborus (Hesse) and Orius naivashae (Poppius) (Hemiptera: Anthocoridae) in the laboratory. Developmental and reproductive traits of both Orius species were examined when offered frozen eggs of the Mediterranean flour moth, Ephestia kuehniella Zeller, frozen processed eggs of the medfly, Ceratitis capitata Wiedemann, or mixed motile stages of the astigmatid mites Tyrophagus putrescentiae (Schrank) or Carpoglyphus lactis (L). Whereas C. lactis and T. putresecentiae proved to be an inferior food for rearing O. thripoborus and O. naivashae, eggs of C. capitata fully supported development and reproduction of both predators. Results on medfly eggs were similar or slightly inferior to those on E. kuehniella eggs, which is the standard food for culturing these anthocorid bugs. O. thripoborus could be maintained for 4 consecutive generations on C. capitata eggs indicating that processed medfly eggs can be a suitable and cheaper alternative to E. kuehniella eggs for prolonged rearing of these Orius spp.  相似文献

Experiments examined the effects of no-tillage (NT) and conventional-tillage (CT) and increased soil moisture (irrigation) on southern corn rootworm (SCR), Diabrotica undecimpunctata howardi Barber (Coleoptera: Chrysomelidae), oviposition and survival and SCR below-ground predators in peanut (Arachis hypogeae L.) agroecosystems. No-tillage systems without irrigation had significantly greater soil moisture than CT systems without irrigation in 1987, but not in 1988. Both moist soil and the presence of an organic residue increased the number of southern corn rootworm eggs in a system. The same 17 species of predatory arthropods were found in all systems. However, their activity in the top 10-cm of soil which increased in NT systems, determined how effective they were as predators. Only 7 predators significantly reduced SCR immature stages, and of these 7, only one, Formicidae, preyed on all stages of SCR. Southern corn rootworm first instars and eggs had the greatest overall percent mortality, with second and third instars the lowest. Peanut yields and pod quality were equal between NT and CT systems. Overall return per ha showed NT, no-irrigation systems with the greatest return. Abiotic parameters dominated SCR survival in CT systems, while biotic parameters had the greatest influence on SCR survival in NT systems.
