To study nuclear events in fructifications of the Basidiomycetes, material was fixed 24 hr in a saturated aqueous solution of HgCl2 containing 1% glacial acetic acid, and embedded in Aquax (G. T. Gurr Ltd.). Following a 4 hr hydrolysis at 20 C in 60% H3PO4, sections were stained for 30 min in a mixture of 4 ml Giemsa R66 (G T. Gurr Ltd.) and 100 ml phosphate buffer at pH 6.5. Differentiation was carried out in sodium cacodylate-HCl buffer at pH 5.8 when required. Preparations were dehydrated in an acetone-xylene series prior to mounting in Euparal. The use of paraffin wax as the embedding medium and HCl as the hydrolysing agent yielded preparations of an inferior quality.  相似文献

Paraffin sections from tissue fixed 4-12 hr in 10% formalin containing 0.5% cetyl pyridinium chloride, and washed 2 hr, were stained as follows: (1) Hydrolyze in 5 N HCl at room temperature for 8.5-9 min, or use standard Feulgen hydrolysis at 60 C. (2) Stain in azure A-Schiff, 0.5% in bisulfite bleach (1 N HCl, 5; 10% Na2S2O5, 5; and distilled water 90—parts by volume) for 10 min. (3) Place in bisulfite bleach 2 changes, 2 min each; wash in water, 1-2 min. (4) Stain in Alcian blue (0.1% in 0.01 2V HCl, pH 2.0) for 10 min. (5) Place in 0.01 N HCl for 2-3 min; wash in water for 1-2 min. (6) Oxidize in 0.5% HIO4 for 5 min; wash in water, 1-2 min. (7) Stain in Schiff's leucofuchsiu, 10 min. (8) Treat with bisulfite bleach as in step 3; wash in running water, 10 min. (9) Stain in naphthol yellow S (0.01% in 1% acetic acid) for 1-2 min. (10) Place in 1% acetic acid for 2 min, dehydrate in tertiary butanol, clear and cover. Result: DNA is deep blue; acidic mucins are light blue; neutral polysaccharides, red to magenta; and proteins, yellow. Proper timing of the hydrolysis for the Feulgen reaction is the most critical step. Overhydrolysis results in green nuclei (staining by naphthol yellow S) whereas purplish nuclei are the results of insufficient hydrolysis.  相似文献

The lithiation of indole, using a slight excess of n-butyl lithium in THF, followed by methylation and reaction with [Cr(CO)6] in refluxing dibutyl ether, resulted in the formation of [Cr(η6-N-methylindole)(CO)3] (1a) and [Cr(η6-N-methyl-2-methylindole)(CO)3] (1b). In contrast, lithiation of quinoline in THF, silylation and the subsequent reaction with [Cr(CO)6] under similar reaction conditions, afforded [Cr(η6-N-trimethylsilyl-2-butyl-1,2-dihydroquinoline)(CO)3] (2) and [Cr(η6-{2-butyl-1,2,3,4-tetrahydroquinoline})(CO)3] (3). The formation of [Cr(η6-2,2′-bis{N-methylindolyl})(CO)3] (4) implied lithiation at the 2-position of 1a. However, metallation at the 7-position was also indicated during the same reaction. In the presence of [Mn(CO)5Br], product 4 and the transmetallation product [Cr(η6-{7-(N-methylindolyl)Mn(CO)5})(CO)3] (5) were isolated. Reaction with titanocene dichloride gave [Cr(η6-{2-(N-methylindolyl)TiCp2Cl})(CO)3] (6), which slowly converted into [TiCp2{Cr(η6-2-(N-methylindolyl)(CO)3}2] (7).  相似文献

It was observed that mitochondria are well-demonstrated by aniline-acid fuchsin staining after Zenker-formol fixation if the sections are not de-Zenkerized. Tests showed that after mordanting in HgCl2, K2Cr2O7, FeCl3, or FeSO4, mitochondria in sections from tissues fixed in neutral buffered formalin could be stained fairly intensely by the same method. Salts of Ag, Ba, Ca, Cd, Co, Cu, Mg, Mn, and Zn were ineffective. If the presence of occasional mercury crystals in the sections is not objectionable, demonstration of mitochondria in Zenkerformol fixed tissues offer speed and additional flexibility in the subsequent use of the blocks as advantages over usual methods.  相似文献

In this study we have compared the short-term effects of both NaCl and HgCl2 on aquaporins of Capsicum annuum L. plants, in order to determine whether or not they are similar. Stomatal conductance, turgor, root hydraulic conductance and water status were measured after 0.5, 2, 4 and 6 h of NaCl (60 mmol/L) or HgCl2 (50 μmol/L) treatment. When 60 mmol/L NaCl was added to the nutrient solution, a large decrease in stomatal conductance was observed after 2 h. However, when HgCl2 (50 μmol/L) was added, the decrease occurred after 4 h. The number of open stomata closed was always lower in plants treated with HgCl2 than in plants treated with NaCl. The water content of the Hg2+-treated plants was decreased, compared with controls and NaCl-treated. The root hydraulic conductance decreased after HgCl2 and NaCl treatment plants. Turgor of leaf epidermal cells was greatly reduced in plants treated with HgCl2, but remained constant in the NaCl treatment, compared with control plants. The fact that the stomatal conductance was reduced more rapidly after NaCl addition, followed by the stomatal closure, and that both water content and turgor did not differ from the control suggests that in NaCl-treated plants there must be a signal moving from root to shoot. Therefore, the control of plant homeostasis through a combined regulation of root and stomatal exchanges may be dependent on aquaporin regulation.  相似文献

Chlorous acid (HClO2), which is known to convert aldehyde groups to carboxylic groups, was found to be an effective blocking agent for tissue aldehydes. A 0.2 M solution of NaClO2 in 1 M acetic acid was found to block completely the Feulgen, PAS, ninhydrin-Schiff and plasmal reaction within 8 hr at 20 C. This blocking reaction appears to be irreversible.  相似文献

Na2S2O3 and HCl can be used as the reducing system to make Schiff reagent, the sulfur produced by the reaction acting as the adsorbent and consequently eliminating the need of decolorizing charcoal. To 100 ml of a 0.02% solution of basic fuchsin add 8 gm of dry Na2S2O3 shake for 3 min, add 0.5 ml of cone. HCl (final concentration 0.05 N). Do not heat. Store in a refrigerator (4° C) overnight, and filter. The filtrate is water-clear, and will remain stable for 24 hr at room temperature. Periodic acid-Schiff and Feulgen reactions can now be performed, preferably at refrigerator temperature, although the times required are notably prolonged (6-8 hr). Background color is slight, and specificity good.  相似文献

Selective lipase-catalyzed synthesis of glucose fatty acid esters in two-phase systems consisting of an ionic liquid (1-butyl-3-methyl imidazolium tetrafluoroborate [BMIM][BF4] or 1-butyl-3-methyl imidazolium hexafluorophosphate [BMIM][PF6]) and t-butanol as organic solvent was investigated. The best enzyme was commercially available lipase B from Candida antarctica (CAL-B), but also lipase from Thermomyces lanuginosa (TLL) gave good conversion. After thorough optimization of several reaction conditions (chain-length and type of acyl donor, temperature, reaction time, percentage of co-solvent) conversions up to 60% could be achieved using fatty acid vinyl ester as acyl donors in [BMIM][PF6] in the presence of 40% t-BuOH with CAL-B at 60 °C.  相似文献

Experimentation with the Papanicolaou stain in this laboratory led to the discovery that the eosin, combined with phosphotungstic acid, was responsible for differential staining of Negri bodies. Eosin prepared as in EA 36 was used, but without the light green and Bismarck brown. Paraffin sections of hippocampus from brains of animal affected with rabies were fixed in 10% formol or in a mixture of 2 volumes of saturated aqueous HgCl2 and 1 volume of absolute alcohol. They were stained first with hematoxylin and then with eosin. This procedure gave better results than staining with other types of eosin or by the original EA 36 mixture. The Negri bodies were well stained and their structure easily visible. The best results were obtained from material fixed with the HgCl2 solution.  相似文献

1,10-Phenanthroline-5,6-dione (C12H6N2O2 (1)) reacts with V(η6-mesitylene)2 and Ti(η6-toluene)2 affording coordination compounds of general formula M(O,O′---C12H6N2O2)3 (M=Ti (2); M=V (3)) which further react with TiCl4 or TiCp2(CO)2 yielding the tetrametallic species M(O,O′---C12H6N2O2---N,N′)3(M′Ln)3 (M=V, M′Ln=TiCl4 (4); M=Ti, M′Ln=TiCp2 (5); M=V, M′Ln=TiCp2 (6)). The complex salt [Fe(N,N′---C12H6N2O2)3][PF6]2 (7) has been obtained from iron(II) chloride tetrahydrate and 1 in the presence of NH4PF6. The reaction of 7 with TiCp2(CO)2 affords the tetrametallic derivative [Fe(N,N′---C12H6N2O2---O,O′)3(TiCp2)3][PF6]2 (8). TiCl2(THF)2 reacts with MCp2(O,O′---C12H6N2O2) to give MCp2(O,O′---C12H6N2O2---N,N′)TiCl2 (M=Ti (9); M=V (10)). By reaction of TiCp2(O,O′---C12H6N2O2---N,N′)TiCl2 (9) with C12H6N2O2, the bimetallic derivative TiCp2(O,O′---C12H6N2O2---N,N′)TiCl2(O,O′---C12H6N2O2) (11) has been prepared, which readily adds to TiCl4, to give the trimetallic titanium derivative TiCp2(O,O′---C12H6N2O2---N,N′)TiCl2(O,O′---C12H6N2O2---N,N′)TiCl4 (12). VCp2(O,O′---C12H6N2O2---N,N′)TiCl2 (10) reacts with the tris-chelate iron(II) cation 7 affording the heptametallic cationic complex [Fe(N,N′---C12H6N2O2---O,O′)TiCl2(N,N′---C12H6N2O2---O,O′)VCp2]3 +2 isolated as the hexafluorophosphate 13.  相似文献

The crystal structure, magnetic, redox and spectroscopic properties of a novel unsymmetrical dinuclear copper(II) complex, prepared by the reaction between copper(II) perchlorate, sodium acetate and the unsymmetrical, binucleating ligand HTPPNOL, where HTPPNOL is N,N,N′-tris-(2-pyridylmethyl)-1,3-diaminopropan-2-ol, is reported. HTPPNOL (1 equiv.) reacted with 1 equiv. of copper(II) ion, in methanol, and produced the mononuclear copper complex [Cu(TPPNOL)](ClO4)(BPh4) (1). On the other hand, the reaction of 1 equiv. of HTPPNOL with 2 equiv. each of copper (II) ion and acetate, in methanol, produced the dinuclear complex [Cu2(TPPNOL)(OOCCH3)](ClO4)2 (2), whose structure has been determined by X-ray diffraction. In complex 2, as a result of the inherent asymmetry of the ligand HTPPNOL, one copper ion is five-coordinated (distorted trigonal-bipyramidal) while the other copper is four-coordinated (distorted square-planar). Then, as a result of the presence of distinct geometries for the metal centres, complex 2 exhibits a ferromagnetic coupling (J=+25.41 cm−1). Titration experiments carried out on the dinuclear complex suggest a pKa=8.0, which was related to the aquo/hydroxo equilibrium. Complex 2 is able to oxidise 3,5-di-tert-butylcatechol to the respective o-quinone. The oxidation reaction was studied by following the appearance of the quinone spectrophotometrically, at pH 8.0 and 25 °C.  相似文献

The catalytic activity of papain in the synthesis of Z-Gly-Phe-NH2 in tert-butanol has been studied in the presence of solid-state acid–base buffers (acids and their sodium salts). All buffer pairs tested reduced the reaction rate compared with the control, particularly the most acidic and basic (assessed by either aqueous pKa or the response of an organic phase indicator). The highest rates, close to the control, were found with glutamic acid/glutamate-Na, PIPES/PIPES-Na and NaH2PO4/Na2HPO4. However, these pairs were unable to erase the pH memory phenomenon, or to overcome the effect of spiking with acetic acid. Hence, at least these buffers do not seem to be able to affect the protonation state and catalytic activity of papain. In the last aqueous solution before drying, the presence of activating agents (cysteine plus EDTA) was more important than buffer ions.  相似文献

Cyclizations of alkylhydrazines with N-acyl-S-methylisothioureas, readily synthesized from acyl chlorides, sodium thioisocyanate, dialkylamines then methyl iodide in a one-pot reaction, gave 1-alkyl-3-dialkylamino-5-phenyltriazoles 7 as major products. The regioisomers were assigned through the use of NOE NMR experiments. While bearing a N-bis(cyclopropyl)methyl-N-propylamino group, this series of compounds shows very good binding affinity on the human CRF1 receptor. Among them, 1-methyl-3-[N-bis(cyclopropyl)methyl-N-propylamino]-5-(2,4-dichlorophenyl)-1H-[1,2,4]triazole 7a had the best binding affinity for the CRF1 receptor (Ki=9 nM).  相似文献

Plasmalogen that contains choline was concentrated from bovine heart to 95% purity from an initial value of 45%, and ethanolamine-containing plasmalogen from bovine brain to 99% purity from initial 64% using phospholipase A1 from Serratia sp. MK1 in a two-phase system. In the two-phase system, Ca2+ was not required and Tris/HCl buffer and butyl acetate were used as an aqueous phase and a solvent phase of the two-phase system, respectively.  相似文献

A silver staining method for paraffin sections of material fixed in HgCl2, sat. aq., with 5% acetic acid is as follows. Process the sections through the usual sequence of reagents, and including I-KI in 70% alcohol, thiosulfate (5% aq.), washing and back to 70% alcohol containing 5% of NH4OH (conc. aq.). After 3 minutes in the ammoniated alcohol, wash through tap water and 2 changes of distilled water and silver 5-10 minutes at 25°C. in 15% AgNO3 aq. to which 0.02 ml. of pyridine per 100 ml. has been added. Blot the slide, but not the section and do not rinse. Reduce at 45°C. in 0.1% pyrogallol in 55% alcohol, then rinse in 55% alcohol and wash in water. The remainder of the process consists of gold toning, intensifying in oxalic acid, fixing in 5% Na2S2O3, washing, dehydrating, clearing and covering. When the specimen contains much smooth muscle, the I-KI solution is acidified before use by adding 2 ml. of 1N nitric acid per 100 ml., and the sections treated for 3 minutes instead of the usual 2 minutes. Formalin should not be added to sublimate-acetic, but specimens that do not contain strongly argyrophilic nonneural tissue may be fixed in formalin or, preferably, Bouin's fluid. Sections of tissue after the latter type of fixation will not require the I-KI and thiosulfate but can go from 95% alcohol to the ammoniated alcohol. The advantages of fixing in HgCl2-acetic acid are suppression of the staining of connective tissue and intensifying the staining of nerve fibers.  相似文献

The facile one step synthesis of 4-hydroxy-2(E)-nonenal and its dimethyl acetal via a cross-metathesis reaction between commercially available octen-3-ol and acrolein or its dimethyl acetal is reported. The method was extended to the synthesis of C6 and C12 4-hydroxy-2(E)-enals, their dimethyl acetal and of the 4-hydroxy-2(E)-nonenoic acid (4-HNA).  相似文献

Aldose diethyl dithioacetals react with 1.2 equivalents of 1-ethoxycyclohexene or 2-methoxypropene in N,N-dimethylformamide at 0° with p-toluenesulfonic acid as catalyst to give the five-membered ring acetal attached to the two terminal oxygen atoms as the major product in every case. In most instances, a small proportion of the terminal, six-membered ring acetal was also obtained, and in a few cases, terminal seven-membered ring acetals were also isolated. Cyclohexylidenation at room temperature gave the same products, but isopropyl-idenation at room temperature resulted in certain cases in partial rearrangement. Cyclohexylidenation reactions gave smaller proportions of the minor six- and seven-membered ring products. Structures were established from 13C-n.m.r. and mass spectra. The 13C-n.m.r. spectra of model cyclohexylidene derivatives were found very similar to those of isopropylidene derivatives previously studied. Two new features useful for structure determination were noted when the spectra of the precursor diols were compared with those of both types of derived acetals; the chemical shift of C-2 of a 1,3-propanediol derivative was shifted upfield by 6–9 p.p.m. on acetalation and the shifts of the diol carbon atoms attached to oxygen were affected according to the type of acetal and ring-size formed. [MnL](ClO4)2 (L = N,N′,N″-tris(2-hydroxypropyl)-1,4,7-triazacyclononane) has been tested for catalyzing sulfide oxidation. In the presence of this complex, ethyl phenyl sulfide, butyl sulfide and phenyl sulfide are completely oxidized to the corresponding sulfoxides and sulfones with H2O2 as the oxidant. 2-Chloroethyl phenyl sulfide oxidation yield 2-chloroethyl phenyl sulfone and phenyl vinyl sulfone. In ethyl phenyl sulfide oxidation, effects of complex and H2O2 concentration and temperature on the reaction rate have been discussed. Through controlling reaction conditions, ethyl phenyl sulfoxide and ethyl phenyl sulfone may be produced selectively. The UV–Vis and electron paramagnetic resonance (EPR) studies on catalyst solution indicate that metal centre of the complex is transformed from Mn(II) to Mn(IV) after the addition of H2O2. At 25 °C, rate constant for ethyl phenyl sulfide oxidation is 4.38 × 10−3 min−1.  相似文献

1. 1. Cyanide inhibits the catalytic activity of cytochrome aa3 in both polarographic and spectrophotometric assay systems with an apparent velocity constant of 4·103 M−1·s−1 and a Ki that varies from 0.1 to 1.0 μM at 22 °C, pH 7·3.

2. 2. When cyanide is added to the ascorbate-cytochrome c-cytochromeaa3−O2 system a biphasic reduction of cytochrome c occurs corresponding to an initial Ki of 0.8 μM and a final Ki of about 0.1 μM for the cytochrome aa3−cyanide reaction.

3. 3. The inhibited species (a2+a33+HCN) is formed when a2+a33+ reacts with HCN, when a2+a32+HCN reacts with oxygen, or when a3+a33+HCN (cyano-cytochrome aa3) is reduced. Cyanide dissociates from a2+a33+HCN at a rate of 2·10−3 s−1 at 22 °C, pH 7.3.

4. 4. The results are interpreted in terms of a scheme in which one mole of cyanide binds more tightly and more rapidly to a2+a33+ than to a3+a33+.
