A clonal propagation method has been developed for efficient multiplication ofVanilla planifolia. Multiple shoots were developed from axillary bud explants using semi-solid Murashige and Skoog (MS) medium supplemented with N6-benzyladenine (BA, 2 mg l–1) and -naphthaleneacetic acid (NAA, 1 mg l–1). The multiple shoots were transferred to agitated liquid MS medium with BA at 1 mg l–1 and NAA at 0.5 mg l–1 for 2–3 weeks, and subsequently cultured on semi-solid medium. Using this method, an average of 42 shoots were obtained from a single axillary bud explant over a period of 134 days. This report describes an efficient and reproducible protocol for large-scale multiplication of Hedychium coronarium plantlets. Axillary bud explants were cultured on Murashige and Skoog medium supplemented with 3 mg/l benzylaminopurine, 3 mg/l kinetin (KIN), and 0.2 mg/l thidiazuron, yielding a maximum of 13.2 ± 0.3 number of shoots. Sub-culturing of shoots every 4 weeks on fresh multiplication medium yielded a consistent proliferation rate. Shoot clusters containing three to five shoots were successfully rooted in KIN (3 mg/l) and indole acetic acid (0.5 mg/l), yielding a maximum of 6.3 ± 0.5 number of roots. Plantlets grown in vitro were acclimatized and subsequently transferred to the field for phenotypic evaluation. Random amplified polymorphic DNA and inter-simple sequence repeat analysis has confirmed the genetic uniformity of in vitro plantlets up to 2 years. After 2 years, these plantlets were transplanted to field, and evaluation of phenotypic characteristics was done. This study is of high significance as these could be commercially utilized for large-scale production of true-to-type plantlets.  相似文献

Summary Spilanthes mauritiana DC., (Compositae), a East African medicinal herb containing pharmaceutically promising secondary metabolites, has successfully been raised in vitro. We have developed a clonal propagation protocol that uses juvenile plants as starting material. The addition of benzylaminopurine (BA) (1.0 μM) and naphthaleneacetic acid (NAA) (0.1 μM) to the culture medium resulted in maximum shooting response with minimal callusing. Shoots rooted best in vitro in MS medium supplemented with indole-3-acetic acid (IAA; 0.2 μM), and plants that had already developed roots showed better growth, with maximum survival rate, in the greenhouse after an initial hardening.  相似文献

An efficient clonal multiplication system was developed for in vitro propagation of crocin — producing Gardenia jasminoides Ellis plants. Murashige and Skoog's (MS) medium containing 6-benzylaminopurine (BAP 1 mg l–1) and indole-3-butyric acid (IBA 1 mg l–1) resulted in multiple shoot initiation at the rate of 21 shoots per explant in 60 d of culture. Transfer of the microshoots into liquid MS medium supplemented with BAP (5 mg l–1) with two subcultures of 15 d duration in the same medium resulted in 400 ± 25 shoots per explant. Efficient rooting was achieved in MS medium supplemented with -naphthaleneacetic acid (5 mg l–1). The in vitro raised plants were hardened in a greenhouse and transplanted to the field successfully. Protocols for in vitro plant regeneration via axillary and adventitious shoot regeneration were established in an important medicinal plant, Aristolochia indica L. (Aristolochiaceae). Basal Murashige and Skoog's (MS) medium supplemented with 0.54 μM α-naphthaleneacetic acid (NAA) and 13.31 μM benzyladenine (BA) induced the maximum number of shoots (45-50) from shoot tip and nodal segment cultures. Phenolic accumulation in leaf and internodal stem derived callus cultured in MS medium containing NAA or 2,4-dichlorophenoxyacetic acid and BA or kinetin was controlled by the addition of 1.0 mg l-1 phloroglucinol (PG) to the callus induction medium. Basal medium supplemented with 2.69 μM NAA, 13.31 μM BA and 1.0 mg l-1 PG induced the best results in terms of shoot bud regeneration from leaf derived callus. Direct de novo development of shoots from leaf segments was achieved using 13.31 μM BA along with 50 mg l-1 activated charcoal. The microshoots were rooted in White's medium supplemented with 2.46 μM indolebutyric acid. More than 85% of rooted plants survived in the soil. Efficient protocols of axillary bud multiplication and indirect organogenesis were established for Holostemma ada-kodien Schult. (Asclepiadaceae). Murashige and Skoog (MS) medium supplemented with 2.0 mg l-1 N6-benzylaminopurine (BAP) and 0.5 mg l-1 indole-3-butyric acid (IBA) induced an average of eight shoots per node and was the best for axillary bud proliferation. Subsequent cultures enhanced the number of shoots. The explant source of callus and the growth regulator inducing the callus exhibited significant influence on organogenesis. Callus developed from the basal cut end of the node explants differentiated more than 15 shoots on MS medium fortified with 1.5 mg l-1BAP. Callus from internode explants developed fewer shoots than callus from the basal cut ends of node explants. Leaf-derived callus did not undergo organogenesis. The abscission of leaves and shoot tips of the developed shoots was prevented by the addition of AgNO3 or CoCl2, but with a concomitant significant reduction in the number of shoots. Half-strength solid MS or liquid medium with 0.05 mg l-1 IBA exhibited the best in vitro rooting. Ninety percent of the rooted shoots survived in the field.  相似文献

An axillary bud derived in vitro propagation procedure for Theobroma cacao is described. The method allows bud expansion and elongation, including leaf development, as early as 13 days after placing explants on establishment medium. Shoots have been maintained in culture vessels for six months with little evidence of decline. Propagules have been induced to form roots, hardened off, and moved to the greenhouse within six weeks of initial explant establishment. Some plants have been in the greenhouse for twelve months and are growing in an apparently normal manner. No major differences in response from UF-667 or EQX-100 derived seed-grown trees were found suggesting that the method may not be limited by genotype. This communication describes for the first time an efficient and reproducible protocol for large-scale multiplication of Bambusa nutans. Nodal segments collected from field-grown clumps and cultured on Murashige and Skoog (MS) medium supplemented with 4.4 μM benzylaminopurine (BA) and 2.32 μM kinetin (Kin) gelled with 0.2% gelrite yielded 80% aseptic cultures with 100% bud-break. The in vitro-formed shoots obtained after bud-break were successfully multiplied in MS liquid medium supplemented with 13.2 μM BA, 2.32 μM Kin, and 0.98 μM indole-3-butyric acid (IBA). Sub-culturing of shoots every 3 weeks on fresh multiplication medium yielded a consistent proliferation rate of 3.5-fold. Shoot clusters containing three to five shoots were successfully rooted with 100% success on half-strength MS liquid medium supplemented with 9.8 μM IBA, 2.85 μM indole-3-acetic acid (IAA), 2.68 μM naphthaleneacetic acid (NAA), and 3% sucrose. Plantlets grown in vitro were acclimatized and subsequently transferred to the field. Inter-simple sequence repeat analysis has confirmed the genetic uniformity of the tissue-cultured plants up to 27 passages.  相似文献

Guadua angustifolia Kunth was successfully propagated in vitro from axillary buds. Culture initiation, bud sprouting, shoot and plant multiplication, rooting and acclimatization, were evaluated. Best results were obtained using explants from greenhouse-cultivated plants, following a disinfection procedure that comprised the sequential use of an alkaline detergent, a mixture of the fungicide Benomyl and the bactericide Agri-mycin, followed by immersion in sodium hypochlorite (1.5% w/v) for 10 min, and culturing on Murashige and Skoog medium containing 2 ml l?1 of Plant Preservative Mixture®. Highest bud sprouting in original explants was observed when 3 mg l?1 N6-benzylaminopurine (BAP) was incorporated into the culture medium. Production of lateral shoots in in vitro growing plants increased with BAP concentration in culture medium, up to 5 mg l?1, the highest concentration assessed. After six subcultures, clumps of 8–12 axes were obtained, and their division in groups of 3–5 axes allowed multiplication of the plants. Rooting occurred in vitro spontaneously in 100% of the explants that produced lateral shoots. Successful acclimatization of well-rooted clumps of 5–6 axes was achieved in the greenhouse under mist watering in a mixture of soil, sand and rice hulls (1:1:1).  相似文献

Summary Cultures of two eranberry (Vaccinium macrocarpon Ait.) cultivars, ‘Ben Lear’ and ‘Pilgrim’, and three eranberry clones from natural stands in Newfoundland were established in a nutrient medium containing N6[2-isopentenyl]adenine (2iP) from nodal and/or shoot-tip explants obtained under aseptic conditions. The cultivars differed in shoot regeneration in terms of shoot number per explant with various concentrations of 2iP over two culture periods. Best total shoot production was obtained when nodal segments were cultured in the medium supplemented with 2.5–5.0 mg 2iP l−1 (12.3–24.6 μM). With higher 2iP levels, shoots did not expand and had a high mortality rate. Nodal explants of the three clones cultured in the same nutrient medium supplemented with 2.5 mg 2iP l−1 (12.3 μM) produced three to five healthy axillary shoots per explant. In another experiment, nodal explants were more productive than shoot tips. In all experiments with subculture, there was an increase in shoot multiplication rate for all genotypes. Shoots were rooted in vitro in the same media used for shoot proliferation, but without any growth regulators. After their transfer to potting medium, almost all of the rooted plants survived. Cranberry genotypes can be efficiently propagated and maintained through nodal culture in a nutrient medium without auxin that contains 2.5–5 mg 2iP l−1 (12–25 μM).  相似文献

A simple, rapid and efficient protocol for micropropagation of Cardiospermum halicacabum via axillary bud multiplication has been successfully developed. The organogenic competence of nodal segments was investigated on Murashige and Skoog (MS) medium supplemented with different concentrations of benzyladenine (BA), kinetin (Kn), thidiazuron (TDZ) and 2-isopentenyladenine (2-iP). Multiple shoots differentiated directly without callus mediation within 4 weeks when explants were cultured on a medium fortified with cytokinins. The maximum number of shoots (14.83 ± 0.52) was developed on a medium supplemented with 0.3 μM TDZ. Such proliferating shoots when subcultured onto MS media devoid of TDZ gave the highest rate of shoot multiplication (35.66 ± 1.00) by the end of fourth subculture passage. Elongated shoots were rooted on 1/3 MS medium augmented with 0.5 μM IAA. The plantlets thus obtained were successfully hardened and transferred to greenhouse.  相似文献

Summary Salvia valentina Vahl and Salvia blancoana Webb & Heldr subsp. mariolensis Figuerola, two endemic species of Salvia from the Mediterranean coastal region of Spain, were successfully gegenerated in vitro from adult plants using two explant types (apical and nodal segments). Maximum shoot proliferation for both species was obtained with nodal explants: for S. blancoana on Murashige and Skoog medium supplemented with 6-γ-γ-dimethylallylaminopurine at 1 mg 1−1 (4.9 μM). and for S. valentina on the same medium with kinetin at 1–2 mg 1−1 (4.6–9.3 μM). The influence of apical dominance, and the explant viability in culture were found to be the main differences between the two species during the shoot multiplication phase. Rooting of shoots was low, specially for S. valentina. For both species, rooting was achieved in Murashige and Skoog medium without growth regulators. In general the addition of the auxins indole 3 acetic acid or indole-3-butyric acid did not improve the rooting or, in the case of naphthaleneacetic acid, had an inhibitory effect. In the best rooting media, rooting shoots increased in length. The rooted plantlets were acclimated to ex vitro conditions and a survival percentage > 70% was obtained under greenhouse conditions. This work was carried out as an ex situ conservation method for these Spanish endemic species.  相似文献

A micropropagation protocol for Psiadia arguta, an endangered endemic plant from Mauritius is described using 15-day old in vitro seedling explants without the radicle. MS basal medium supplemented with TDZ (0.5–1 mg/l) proved to be the most effective medium for the induction of cotyledonary axillary buds as compared to MS medium containing NAA (0.5 mg/l) or both NAA (0.5 mg/l) and TDZ (0.5–1 mg/l). In fact, after transfer to hormone free MS medium, microshoots were obtained only from seedling explants cultured on media containing only TDZ. Regenerated shoots elongated and rooted when cultured on MS8900 containing IBA (0–1 mg/l). Hormone-free MS8900 was the best medium for rooting and development of plantlets for acclimatization.  相似文献

An efficient protocol was achieved for rapid propagation of Wedelia chinensis (Osbeck) Merr. through axillary bud proliferation and ex vitro rooting. Murashige and Skoog (MS) medium supplemented with benzyladenine (BA; 8.87 microM) and indole-3-butyric acid (IBA; 2.46 microM) was optimal for axillary bud proliferation, which developed a mean of 8.3 shoots/node. Excision and culture of node segments from in vitro shoots on medium supplemented with the same concentration of growth regulators developed more than 30 shoots within 40 days. Excision and culture of nodes in succession enhanced the number of shoots. Shoot multiplication did not exhibit decrease in the number of shoots even at 10th subculture. Nevertheless, the shoots exhibited a tendency towards stunted nature. But reduction of BA to 4.44 or 2.22 microM resumed normal growth of shoots. Half strength MS medium fortified with IBA (2.46 microM) induced the highest number of roots. All in vitro rooted shoots survived in field. Dipping of the basal end of shoots collected from multiplication medium in IBA (2.46 microM) solution for 7 days induced roots and its transfer to small pots facilitated the survival of all rooted shoots (100%). Rooting ex vitro by direct transfer of shoots from multiplication medium exhibited 89.2 per cent survival. Use of commercial sugar and tap water and also the omission of in vitro rooting reduce the propagation cost 50-70 per cent. The protocol enables to harvest more than 50,000 plantlets within 150 days starting from a single node explant.  相似文献

In vitro clonal multiplication of apple rootstock MM 111 using axillary buds and shoot apices were carried out. Vegetative axillary buds of the size of 0.2-2.0 cm and shoot apices measuring 4 mm in length were initiated to shoot proliferation on MS medium supplemented with BA (0.5 - 1.0 mgl(-1)), GA3(0.5 mgl(-1)), with or without IBA(0.05 - 0.1 mgl(-1)). Small size explants showed less phenol exudation and less contamination. Following establishment phase, the small shoots emerged from explants were subcultured on MS medium supplemented with different combinations and concentrations of growth regulators. BA (1.0 mgl(-1)) and GA3 (0.5 mgl(-1)) combination showed highest multiplication rate (1:5), andcl also produced longer shoots. Two step rooting was done by transferring microcuttings to auxin free solid medium after root initiation in dark on 1/2 strength MS liquid medium containing IBA (0.5 mgl(-1) ). Rooted plantlets were transferred to peat containing paper cups and resulting plants of MM 111 acclimated successfully for transfer to field.  相似文献

Clonal propagation of Camptotheca acuminata through shoot bud culture   总被引:5,自引:0,他引:5  
The chinese tree Camptotheca acuminata produces the anti-cancer and anti-retroviral drug camptothecin. Methods were developed for the clonal propagation of this important medicinal plant through shoot bud culture. Shoot buds were excised from 25 to 30 day old seedlings, presoaked for 48 h in three different liquid media containing either BA (2.22–17.4 M), kinetin (2.32–18.58 M), or thidiazuron (0.1–10 M) and were subsequently cultured on semi-solid medium of the same composition. Multiple shoots only developed from the 6-benzyladenine presoaked explants with the maximum number of shoots initiated from buds presoaked in and grown on B5 medium containing 17.4 M 6-benzyladenine. Individual shoots were removed from clusters and rooted on B5 supplemented with indole-3-butyric acid (4.9–19.6 M). The lowest concentration of indole-3-butyric acid (4.9 M) gave the highest percentage of rooting (82%) and the shortest root initiation period (18 d). Summary Ethylene effect on in vitro shoot proliferation of two apple rootstocks, MM111 and M9, was studied. Ethylene biosynthesis was proportionally stimulated by increasing concentrations of the precursor 1-aminocyclopropane-1-carboxylic acid (ACC). When 25 μM or more ACC was applied without any control of the headspace of culture vessels, shoot proliferation of both rootstocks was negatively affected. However, when shoot cultures were transferred to ACC-supplemented medium after the second week of culture, ACC had no effect. Supplementing the medium with aminoethoxyvinylglycine (AVG), an inhibitor of ethylene biosynthesis, together with the application of gas traps inside the flasks, significantly enhanced axillary shoot formation and elongation. Steady and high exogenous concentrations of ethylene in the culture flasks had negative effects on shoot proliferation. MM111 appeared to be more sensitive to ethylene than M9. For AVG a threshold dose was noticed, beyond which phytotoxic effects were induced.  相似文献

Summary An efficient, rapid and large-scale propagation of the woody, aromatic and medicinal shrub, Holarrhena antidysenterica, through in vitro culture of nodal segments with axillary buds, is described. N6-benzyladenine used at 15 μM was the most effective in inducing bud break and growth, and also in initiating multiple shoot proliferation at the rate of 43 microshoots per nodal explant with axillary buds, after 30 d of eulture. By repeated subculturing of nodal explants with axillary buds, a high-frequency multiplication rate was established. Efficient rooting was achieved with 35 μM indole-3-butyric acid which was the most effective in inducing roots, as 80% of the microshoots produced roots. Plantlets went through a bardening phase in a controlled plant growth chamber, prior to ex vitro transfer Micropropagated plants established in garden soil were uniform and identical to donor plants with respect to growth characteristics and vegetative morphology.  相似文献
